Each September, the American Library Association (ALA) and libraries nationwide partner to promote Library Card Sign-Up Month. The goal is to remind community members, parents, students, teachers, and librarians about the importance of owning a library card, which is like a passport to vast book collections, digital resources, hands-on support, and much more.In 2019, staff at New Jersey State Library (NJSL) brainstormed ways to make the event extra special. We decided to create new partnerships and strengthen existing ones, so we collaborated with the New Jersey Department of Education (NJDOE), the New Jersey Library Association (NJLA), and the New Jersey Association of School Librarians (NJASL) to launch the Libraries = Success marketing campaign.

Purpose This research study aims to examine influences on the public library funding decision from the perspectives of New Jersey mayors and public library directors. Design/methodology/approach Content analysis was used to explore statements of 84 public library directors and 45 mayors about what was most/least effective in a recent library budget request. Findings Although they may differ on the relative importance of a specific concept, the public library directors and mayors in this study have high levels of agreement about what information is important in the budget decision process. Research limitations/implications The primary limitations of the findings of this study stem from the reliability of self-reported data and the low response rates. While the researcher believes that the findings and conclusions are valid for the survey respondents, it is less certain that they are valid for all public libraries in New Jersey, and it is not recommended that the conclusions be extended to public libraries outside of the state which differ in state mandates and other circumstances. Practical implications Understanding more about the factors that can influence library funding decisions provides insight into the very complex public library funding process and may help public library directors and mayors engage in mutually satisfactory budget negotiations. Originality/value This study contributes to the Library and Information Science research literature on library budgets by examining influences on the public library funding decision, specifically adding the perspectives of mayors and public library directors on a recent budget cycle.

A free public library is the cornerstone of most American communities. Libraries offer a variety of far-reaching services, ranging from books for self-education and leisure reading, to informational and cultural programs. Carnegie libraries constructed in the early 1900s through the philanthropy of Andrew Carnegie played a significant role in transforming the library movement from subscription libraries for special interest groups to free public libraries with services accessible by all. Of the 1,412 communities in the United States to build Carnegie libraries, twenty-nine communities in New Jersey benefitted from this program. This research project draws primarily on original correspondence between New Jersey communities seeking library building funds and Andrew Carnegie. The letters supporting funding applications create a unique demographic and economic snapshot of New Jersey communities during the early 1900s when the Carnegie libraries were erected in the state. This study offers historical insights and informs the role that the Carnegie libraries played in their respective communities.

Peptidomimetics are synthetic oligomers that resemble the activities of peptides. Their advantages over peptides include high stability towards proteolysis and enormous chemical diversity. Over the past two decades, there have been extensive efforts to develop peptide mimics, such as beta-peptides, peptoids, D-peptides, etc. The research on peptidomimetics have led to many important applications in both medicinal and material science. In order to explore new functions, the discovery of peptidomimetics with novel frameworks is essential. We reported the synthesis and evaluation of a new class of peptidomimetics, termed as gamma-AApeptides. Previous studies of gamma-AApeptides have revealed that gamma-AApeptides are highly resistant to proteolysis, and are highly amendable to chemical diversification. However, new biological activities and folding properties of gamma-AApeptides still need to be explored. In order to expand the potential of gamma-AApeptides in chemical biology and medicinal chemistry, I have been focusing on the development of new methods to synthesize linear and cyclic gamma-AApeptides, development of one-bead-one-compound (OBOC) gamma-AApeptide libraries for the discovery of inhibitors against beta-amyloid aggregation, exploring new helical foldamers for the rational design of protein-protein interaction (PPI) inhibitors, and studying cyclic gamma-AApeptides for antimicrobial development.

Peptidomimetics can mimic hierarchical structures of peptides and proteins. Thus, they are extensively studied for therapeutic applications. To break the limitation of backbones and frameworks and expand the peptidomimetics family, a new class of peptidomimetics - “γ-AApeptides” was developed. Design of γ-AApeptides is based on the chiral peptide nucleic acids (PNAs) backbone. The World Health Organization estimates that one -third of all deaths in the world are on account of infectious diseases. AMPs are important because of their high activity against broad spectrum microbes, less susceptible to grow resistance and selectivity in binding to bacterial cells over human cells. γ-AApeptides as a new class of peptidomimetics have increased stability and enhanced chemical diversity. We have developed polymyxin mimic cyclic peptides, small linear molecules and hydantoin derivatives as potent antibiotic agents with γ-AApeptides. They have good bioactivity and selectivity. Combinatorial library is key technology for accelerating the discovery of novel therapeutic agents. One-bead-two-compound γ-AApeptides-based library was developed and screened against SMYD2 protein which is essential for tumor growing.

"Victorianism" is a highly controversial, sometimes ironic, term penned by historians throughout various works that has come to hold dramatic weight in both its meaning and its influence. Though the term is usually most closely associated with nineteenth century England, Victorianism was a highly influential movement in American culture simultaneously as well, specifically in the spheres of home, work, and play. Of those, "play," or leisure, is undoubtedly the least explored, especially before the latter decades of the twentieth century. Prior to this period, most literature about the Victorians, with the exception of a few works, only dealt with masculinity, religion, and the rigid dynamic of the nineteenth century household. Recently, historians like James Walvin, Pamela Horn, and Hugh Cunningham have attempted to draw attention to Victorian leisure with excellent works on pastimes and society during the nineteenth century, but they represent only a few. However, many works of this caliber focus on England, the "birthplace" of Victorianism. Thus, this work attempts to emphasize that the cultural phenomenon of Victorianism was just as present in the United States. Despite the recurring themes of the home and the workplace so often chosen by scholars, it is actually within the realm of leisure that the controversial issues of the Victorian period and its people can be best observed. Class, race, and gender were three major components of the Victorian culture that shaped the various forms of leisure and recreation, as well as the specific restrictions on those amusements.; All of these factors had a shared, tremendous influence on the progress (or lack thereof) towards a more modern era and society that occurred at the turn of the twentieth century. In the pages to follow, the numerous contradictions and paradoxes of Victorian leisure in America will be examined, ultimately demonstrating how pastimes and recreation (and their outlets) in the mid to late nineteenth century were neither truly Victorian nor truly progressive, but indeed a combination of both. This creates further irony during this controversial period. However, before exploring these outlets, the term "Victorian" will be examined while placing it into the context of mid to late nineteenth century Americans who belonged to all classes of travelers. It will become apparent that American Victorians had much invested in their values, but were also willing to break the rules regarding certain amusements and pleasures. Moreover, the "democratization" of leisure will be highlighted as the upper and lower classes began to enjoy the same recreations. Marked innovations of the period will also be discussed, as to highlight their importance on Victorian leisure and its development, which will also be referred to throughout the chapters. These topics will be addressed before examining the specific Victorian leisure culture of two of America's oldest seaside destinations: Cape May and Atlantic City, both in New Jersey. The guests, accommodations and transportation, and offerings at these resort towns will act as a mirror into mid to late nineteenth century culture. There, the contradictory ideals and rules of Victorianism are exhibited as the resorts rose to prominence. The decline of "elite-only" leisure and the rise of the "excursionist" will be examined throughout the progression of the towns' growth and boom periods.; Exploring the ironies of Victorian leisure through the proverbial lens of Cape May and Atlantic City proves effective, as the towns came to represent opposite ends of the "socially acceptable" spectrum after a short period, and were full of similar inconsistencies and paradoxes themselves. Additionally, their current fates remain a product of their polarized Victorian heydays, further proving the influence of seaside resort culture, the late Victorian period, and its ideals on the broader field of American leisure history.

AbstractIn Chap. 10.1007/978-3-030-28561-6_1, the Japanese approach was explained as developing students who learn mathematics by and for themselves (Isoda, 2015), and also as trying to cultivate human character, mathematical values, attitudes, and thinking as well as knowledge and skills (Isoda, 2012; Rasmussen and Isoda, Research in Mathematics Education 21:43–59, 2019). To achieve these aims, the approach is planned under the curriculum sequence to enable students to use their previous knowledge and reorganize it in preparation for future learning. By using their learned knowledge and reorganizing it, the students are able to challenge mathematics by and for themselves. In relation to multiplication, the Japanese curriculum and textbooks provide a consistent sequence for preparing future learning on the principle of extension and integration by using previous knowledge, up to proportions. (The extension and integration principle (MED, 1968) corresponds to mathematization by Freudenthal (1973) which reorganizes the experience in the our life (Freudenthal, 1991). Exemplars of the Japanese approach on this principle are explained in Chaps. 10.1007/978-3-030-28561-6_6 and 10.1007/978-3-030-28561-6_7 of this book.) This chapter is an overview of the Japanese curriculum sequence with terminology which distinguish conceptual deferences to make clear the curriculum sequence in relation to multiplication. First, the teaching sequence used for the introduction of multiplication, and the foundation for understanding multiplication in the second grade, are explained. Based on these, further study of multiplication is done and extended in relation to division up to proportionality. The Japanese approach to multiplication is explained with Japanese notation and terminology as subject specific theories for school mathematics teaching (Herbst and Chazan, 2016). The Japanese approach was developed by teachers through long-term lesson study for exploring ways on how to develop students who learn mathematics by and for themselves (Isoda, Lesson study: Challenges in mathematics education. World Scientific, New Jersey, 2015a; Isoda, Selected regular lectures from the 12th International Congress on Mathematical Education. Springer, Cham, Switzerland, 2015b). This can be done only through deep understanding of the curriculum sequence which produces a reasonable task sequence and a concrete objective for every class in the shared curriculum, such as in the Japanese textbooks (Isoda, Mathematical thinking: How to develop it in the classroom. Hackensack: World Scientific, 2012; Isoda, Pensamiento matemático: Cómo desarrollarlo en la sala de clases. CIAE, Universidad de Chile, Santiago, Chile, 2016) (This is also illustrated in Chap. 10.1007/978-3-030-28561-6_7 of this book.).

Abstract In June 1956, Edmund Wilson, Class of 1916, returned to Princeton University to receive an honorary degree, in conjunction with the fortieth anniversary of his class’s graduation and the bicentennial of Nassau Hall. The occasion was slightly awkward. Wilson had great affection for his old college, and especially for the memory of his departed teachers Norman Kemp Smith and Christian Gauss. A few years earlier, when presiding over a set of the prestigious Princeton seminars named in Gauss’s honor-and presenting work that would eventually wind up as part of his monumental study, Patriotic Gore, edited by another Princeton graduate, Sheldon Meyer-Wilson one day took his friend Leon Edel on a private tour of his former stomping grounds, enthusiastically showing off the architectural highlights. But Wilson had never partaken of the rah-rah bonhomie for which Princeton graduates were, and are, so famous. And more than a few Princetonians regarded Wilson-the bookish, oft-married ex-radical and (thanks to Memom of Hecate County) reputed pornographer-as a disloyal odd duck.

This case study serves as a successful example of an external academic librarian-school librarian statewide partnership for other academic librarians in the United States to follow as they engage in outreach and marketing activities with high school students. A statewide partnership between a New Jersey academic librarian organization and a K-12 school librarian organization resulted in the formation of panels of New Jersey academic librarians who led virtual orientations to the academic library experience for New Jersey high school students. The virtual orientations to the academic library were developed to address students' feelings of anxiety when it comes to the academic library and to highlight the role that the academic library plays as a factor in the college choice of high school students. The success of the virtual library orientations held in October of 2021 will continue to influence future initiatives of the newly formed statewide partnership between academic librarians and school librarians in the state of New Jersey.

Abstract Designing mechanical parts using a feature vocabulary is a very effective and rich paradigm. Its expressive power, however, is severely limited if the set of feature types available in a feature library is fixed. It is, therefore, desirable to be able to extend and configure a feature library according to particular requirements, either of an end-user of a CAD system or of an application area. These requirements are not limited to topologic and parametric aspects of a generic feature definition, but include also validity conditions to be verified for each feature instance in a model. This paper proposes a new declarative scheme for the definition of feature classes. This scheme provides a unified description of the shape and validity issues of a feature class, as well as a flexible configuration of the feature class interface. In the definition process, the various constraint classes available play a central role, whereas an inheritance mechanism structures the feature library hierarchy. At the end of the process, validation of the class is performed, in order to avoid over- and underconstrained specifications. A graphical user interface supports the whole feature class definition process. Once defined, a feature class is automatically made available for use in a feature library of the modeling system.

This paper describes the creation of the Open Class 60 (BOC 60) racing yacht "Imagine". She was conceived to win the 1994 BOC singlehanded round the world race, an ambitious goal since the French sailors have dominated the race since its inception. This paper will examine the design of this complex racing machine, and the management of the project. The project produced a boat that was capable of attaining the goal of winning, but when the project management office failed to adhere to the project plan, the project unravelled and collapsed. In a squall at sea, at night, off Cape May, New Jersey, "Imagine's" boom failed, the main sail was dropped, and in the resultant short steep seas, "Imagine" slammed badly, dishing plating at both ends of the vessel, causing two forward frames to trip, and resulting in other structural damage to forward deck longitudinals and in the cockpit. "Imagine's" hull remained watertight, and she returned to Norfolk under short sail. Upon arrival she was inspected, and the necessary repair steps were outlined. These were discussed the following day with the project office and the ABS inspector. Within days drawings were provided to the project office for repair and to ABS for review. At that point matters stalled, no repair was started and within two week-; the project office announced their intent to abandon the project. The project plan will be reviewed, the basis for the design will be discussed, the incident at sea which precipitated the project's disbanding will be examined, and an analysis of the resultant damage given. Lessons learned from the project will be discussed. The actual design of the boat was interesting and rewarding, but it was a small part of the goal of the project. The goal was not attained, and when the BOC race started from Charleston in September 94, "Imagine" remained at the dock, her outfit and development incomplete. This paper will attempt to evaluate the reasons why. Two central themes repeat: lack of funds; and lack of sea trials. In our opinion it was this lack of funds, that led the project office to eliminate carefully planned steps in "Imagine's" development, specifically the sea trials. This also lead them to attempt an offshore voyage from Norfolk, Virginia to Newport, Rhode Island in November 1993, prior to sea trials, with a known defective boom, and a jury rigged boom vang.

College students are prone to a sedentary lifestyle while trying to balance daily challenges and school workload. Sedentariness is associated with increased risk of chronic diseases – obesity, cardiovascular disease, type II diabetes and even cognitive performance. Given that a full-time college student in the US spends more than 15 hours per week sitting in a college classroom, it may be an ideal setting for implementation of Sit-Stand Desk (SSD) to reduce college students’ sedentary time. Graduate and undergraduate students (N = 178) of New Jersey Institute of Technology were randomly recruited to complete an online assessment survey. Participants' mean(standard deviation) age was 22.4(4.7) years old, 63% identified as male, 33% identified as female while 4% were of the other gender class. Among the participants, 44.3% students self-reported to be overweight or obese according to their BMI (>25 kg/m2), 76% students led an inactive/sedentary lifestyle, and 63.5% students did not meet physical activity guidelines. Students’ perceived acceptability of SSD in the classroom was strongly favorable. If SSD were made available in a class that students are currently taking, only about 11% participants preferred not to stand at all during the class time, and 89% students preferred to stand for at least 10% of class time. Most of the students (85% - 99%) predicted either no change or positive change (get better) in all academic factors (focus, restlessness, attention, engagement and boredom) and health factors (physical health, fatigue and back pain), if SSD in introduced in the classroom. Collectively, the findings of this study strongly support students’ acceptability of introducing standing desks in college classrooms. Results of this study should be useful for policy makers regarding classroom designs. Future studies are needed to test the viability and efficacy of introducing sit-stand desks in college classrooms.

COVID-19 has drastically affected the entire nation. This study involved collecting tweets and analyzing the COVID tweets for August 2020. The aim was to understand whether people have expressed sentiments related to COVID-19 across all the states of the United States and find any correlation between the sentiment tweets and the number of actual cases reported. Around 400000 COVID-19 Twitter data was collected for August 2020 from the primary Twitter database. A simple NLP-based unigram sentiment analyser, a novel approach different from the traditional machine learning approach, was adopted to identify twitter sentiments. The results indicate that tweets related to COVID demonstrate the two types of sentiments, one related to the deaths and the other about the COVID symptoms. Furthermore, the results show that the sentiments for each category vary from State to State. For example, states of New York, California, Texas are higher tweets sentiments regarding expressing death sentiment, and states of New York, California, Nevada, are higher regarding sentiments of expressing COVID-19 symptoms with an accuracy of 83%. As a part of the research, a new sentiment scorecard was created to provide a sentiment score based on the sentiments of the tweets expressed to the actual reported death cases. The sentiment scores for the ‘symptoms’ class are higher for Maryland, New Jersey, and Oregon, whereas sentiment scores for the 'death' class are higher for Virginia, Delaware, and Hawaii. These sentiment scores indicate that the Twitter users of these states are actively tweeting about symptoms and deaths even though the actual reported cases are less in these states. The analysis results also found no or little correlation between the COVID Tweets and the number of COVID death cases reported across all the states.

Abstract The small modular reactor (SMR) class core design concept of once-through SuperCritical light Water-cooled Reactor (SCWR) with fast neutron spectrum (Super FR) is being developed at Waseda University. The preceding studies revealed that SMR class Super FR core can be designed, whose performance is compatible to large Super FR core with respect to average power density and average coolant outlet temperature with small core pressure drop, which is favorable from the viewpoint of achieving natural circulation cooling after reactor shutdown. However, consequences of severe accident of such SMR class Super FR has never been considered. Two major issues need to be addressed. The first issue is the fundamental issue for a water-cooled reactor, which is the possibility of the whole core meltdown as the coolant boils off at low temperature under depressurized condition. Second issue is the fundamental issue for a fast spectrum core, which is to avoid re-criticality of molten core as the reactivity increases with core compaction. With the above described background, this study proposes In-Vessel Retention (IVR) of SMR class Super FR, which can avoid re-criticality even if the whole core relocates to the lower plenum of the Reactor Pressure Vessel (RPV). The debris coolability may be a subject for the future study. The core characteristics with a given set of design specifications and criteria are evaluated based on fully coupled neutronics and thermal-hydraulics core burnup calculations. The neutronics calculations are based on interpolation of macroscopic cross sections with neutron diffusion approximation (SRAC and COREBN codes with JENDL-4.0 nuclear data library). The thermal-hydraulics calculations are based on in-house code that adopts single channel model with Watts’ correlation for evaluating supercritical water heat transfer. The batch fuel shuffling pattern is considered to evaluate the equilibrium core characteristics. The debris criticality is evaluated based on Monte-Carlo based method (MVP) to consider the RPV lower plenum and debris configurations. The fuel debris are assumed to be homogeneous. Firstly, for a given discharge burnup, quantitative relationship between the core criticality and the debris criticality is evaluated. The results show that downsizing the core increases the debris criticality when relocated to the RPV lower plenum, because the Pu enrichment needs to be raised to attain the same discharge burnup with larger neutron leakage. The results also show that some measures are necessary to prevent re-criticality of the debris if the whole core is assumed to relocate to the lower plenum. Then, the new “debris dispersion IVR” concept is proposed. In this concept, heat resistant structure (tentatively ZrO2), which contains some neutron absorbing material (tentatively, B4C) is placed at the center of the lower plenum to disperse the debris and gain neutron leakage and absorption. Then, the trade-off relationships between the core discharge burnup and the size of the debris dispersion structure are evaluated with considerations of core design specifications such as average Pu enrichment, operation cycle length, and fuel shuffling patterns.

As part of ARL’s Research Library Impact Framework initiative, a team from the University of Pittsburgh Library System addressed how special collections support teaching, learning, and research. This project developed a toolkit that allows measurement of impact of engagement with primary sources. The University of Pittsburgh (Pitt) initially worked with University of California, Irvine (UCI) to develop student learning outcomes based on the ACRL/RBMS–SAA Guidelines for Primary Source Literacy and used a logic model framework to identify three types of outcomes: initial (attainment of new skills and knowledge), intermediate (behavioral change), and long-term (change in status). Pitt studied two successive cohorts of their Archival Scholars Research Awards (ASRA), a semester-long undergraduate scholarship program that matches each student with a University of Pittsburgh Library System (ULS) librarian/archivist and a Pitt faculty mentor to engage in an in-depth research project. Next, Pitt refined, expanded, and tested the toolkit for use in class visits to assess student learning and archivist/librarian performance in supporting instructor learning goals. The mapping and assessment toolkit Pitt developed may be applied to teaching with primary sources across different disciplines and institutional settings.

The BARD project was a continuation of a previous BARD funded research project. It was aimed at characterization of the 12kDa immunodominant protein and subsequently the cloning and expression of the gene in E. coli. Additional immunodominant proteins were sought among genomic B. abortus expression library clones using T-lymphocyte proliferation assay as a screening method. The 12kDa protein was identified as the L7/L12 ribosomal protein demonstrating in the first time the role a structural protein may play in the development of the host's immunity against the organism. The gene was cloned from B. abortus (USA) and B. melitensis (Israel) showing identity of the oligonucleotide sequence between the two species. Further subcloning allowed expression of the protein in E. coli. While the native protein was shown to have DTH antigenicity its recombinant analog lacked this activity. In contrast the two proteins elicited lymphocyte proliferation in experimental murine brucellosis. CD4+ cells of the Th1 subset predominantly responded to this protein demonstrating the development of protective immunity (g-IFN, and IL-2) in the host. Similar results were obtained with bovine Brucella primed lymphocytes. UvrA, GroE1 and GroEs were additional Brucella immunodominant proteins that demonstrated MHC class II antigenicity. The role cytotoxic cells are playing in the clearance of brucella cells was shown using knock out mice defective either in their CD4+ or CD8+ cells. CD4+ defective mice were able to clear brucella as fast as did normal mice. In contrast mice which were defective in their CD8+ cells could not clear the organisms effectively proving the importance of this subtype cell line in development of protective immunity. The understanding of the host's immune response and the expansion of the panel of Brucella immunodominant proteins opened new avenues in vaccine design. It is now feasible to selectively use immunodominant proteins either as subunit vaccine to fortify immunity of older animals or as diagnostic reagents for the serological survaillance.

The general objective of this study was to develop rationally designed mimetic antagonists (and agonists) of the PK/PBAN Np class with enhanced bio-stability and bioavailability as prototypes for effective and environmentally friendly pest insect management agents. The PK/PBAN family is a multifunctional group of Nps that mediates key functions in insects (sex pheromone biosynthesis, cuticular melanization, myotropic activity, diapause and pupal development) and is, therefore, of high scientific and applied interest. The objectives of the current study were: (i) to identify an antagonist biophores (ii) to develop an arsenal of amphiphilic topically active PK/PBAN antagonists with an array of different time-release profiles based on the previously developed prototype analog; (iii) to develop rationally designed non-peptide SMLs based on the antagonist biophore determined in (i) and evaluate them in cloned receptor microplate binding assays and by pheromonotropic, melanotropic and pupariation in vivo assays. (iv) to clone PK/PBAN receptors (PK/PBAN-Rs) for further understanding of receptor-ligand interactions; (v) to develop microplate binding assays for screening the above SMLs. In the course of the granting period A series of amphiphilic PK/PBAN analogs based on a linear lead antagonist from the previous BARD grant was synthesized that incorporated a diverse array of hydrophobic groups (HR-Suc-A[dF]PRLa). Others were synthesized via the attachment of polyethylene glycol (PEG) polymers. A hydrophobic, biostablePK/PBAN/DH analog DH-2Abf-K prevented the onset of the protective state of diapause in H. zea pupae [EC50=7 pmol/larva] following injection into the preceding larval stage. It effectively induces the crop pest to commit a form of ‘ecological suicide’. Evaluation of a set of amphiphilic PK analogs with a diverse array of hydrophobic groups of the formula HR-Suc-FTPRLa led to the identification of analog T-63 (HR=Decyl) that increased the extent of diapause termination by a factor of 70% when applied topically to newly emerged pupae. Another biostablePK analog PK-Oic-1 featured anti-feedant and aphicidal properties that matched the potency of some commercial aphicides. Native PK showed no significant activity. The aphicidal effects were blocked by a new PEGylated PK antagonist analog PK-dF-PEG4, suggesting that the activity is mediated by a PK/PBAN receptor and therefore indicative of a novel and selective mode-of-action. Using a novel transPro mimetic motif (dihydroimidazole; ‘Jones’) developed in previous BARD-sponsored work, the first antagonist for the diapause hormone (DH), DH-Jo, was developed and shown to block over 50% of H. zea pupal diapause termination activity of native DH. This novel antagonist development strategy may be applicable to other invertebrate and vertebrate hormones that feature a transPro in the active core. The research identifies a critical component of the antagonist biophore for this PK/PBAN receptor subtype, i.e. a trans-oriented Pro. Additional work led to the molecular cloning and functional characterization of the DH receptor from H. zea, allowing for the discovery of three other DH antagonist analogs: Drosophila ETH, a β-AA analog, and a dF analog. The receptor experiments identified an agonist (DH-2Abf-dA) with a maximal response greater than native DH. ‘Deconvolution’ of a rationally-designed nonpeptide heterocyclic combinatorial library with a cyclic bis-guanidino (BG) scaffold led to discovery of several members that elicited activity in a pupariation acceleration assay, and one that also showed activity in an H. zea diapause termination assay, eliciting a maximal response of 90%. Molecular cloning and functional characterization of a CAP2b antidiuretic receptor from the kissing bug (R. prolixus) as well as the first CAP2b and PK receptors from a tick was also achieved. Notably, the PK/PBAN-like receptor from the cattle fever tick is unique among known PK/PBAN and CAP2b receptors in that it can interact with both ligand types, providing further evidence for an evolutionary relationship between these two NP families. In the course of the granting period we also managed to clone the PK/PBAN-R of H. peltigera, to express it and the S. littoralis-R Sf-9 cells and to evaluate their interaction with a variety of PK/PBAN ligands. In addition, three functional microplate assays in a HTS format have been developed: a cell-membrane competitive ligand binding assay; a Ca flux assay and a whole cell cAMP ELISA. The Ca flux assay has been used for receptor characterization due to its extremely high sensitivity. Computer homology studies were carried out to predict both receptor’s SAR and based on this analysis 8 mutants have been generated. The bioavailability of small linear antagonistic peptides has been evaluated and was found to be highly effective as sex pheromone biosynthesis inhibitors. The activity of 11 new amphiphilic analogs has also been evaluated. Unfortunately, due to a problem with the Heliothis moth colony we were unable to select those with pheromonotropic antagonistic activity and further check their bioavailability. Six peptides exhibited some melanotropic antagonistic activity but due to the low inhibitory effect the peptides were not further tested for bioavailability in S. littoralis larvae. Despite the fact that no new antagonistic peptides were discovered in the course of this granting period the results contribute to a better understanding of the interaction of the PK/PBAN family of Nps with their receptors, provided several HT assays for screening of libraries of various origin for presence of PK/PBAN-Ragonists and antagonists and provided important practical information for the further design of new, peptide-based insecticide prototypes aimed at the disruption of key neuroendocrine physiological functions in pest insects.
