Ističem da je bit Linove filozofijske prakse umjetnost pisanja. Slično drugim filozofima umjetnosti života, Lin je umjetnik i filozof ujedno. Dakle, on sam središnja je tema njegovih eseja. U svojoj performativnoj umjetnosti, Lin konstruira svoje »sebstvo« i oblikuje model filozofijskog života duboko ukorijenjenog u tradiciju kineske kulture dokolice. Po opublikowaniu My Country My People (1935) oraz The Importance of Living (1937) Lin osiągnął wielką sławę jako „filozof chiński”, jednak jego własna myśl filozoficzna, rozproszona w wielu tekstach, rzadko bywała przedmiotem zainteresowania uczonych.

Abstract Interleukin-1 (IL-1) has been shown to stimulate hematopoietic progenitor cell growth both in vitro and in vivo. Although IL-1 alone lacks the ability to promote hematopoietic progenitor growth in vitro, it is a potent synergistic factor in combination with other colony- stimulating factors (CSFs). Because it was unknown whether type I (p80), type II (p68), or other IL-1-binding proteins mediated the synergistic effects of IL-1 on purified progenitor cells, we used the difference in immunoreactivity between type I and type II IL-1 receptor (IL-1R) to better assess the role of these receptors in hematopoietic progenitor growth. Therefore, the synergistic effects of IL-1 alpha on IL-3-, CSF-1-, and granulocyte macrophage (GM)-CSF-induced progenitor growth, both in CFU-c and single-cell assays, were determined in the presence of monoclonal antibodies (MoAbs) 35F5 and 4E2 that block the binding of IL-1 alpha to type I and type II IL-1R, respectively. The synergistic effect of IL-1 alpha on IL-3 responsive Lin- and Lin(-)-Thy- 1+ progenitors was indirectly mediated and could be inhibited by MoAb 35F5. In contrast, IL-1 alpha directly synergized with CSF-1 and GM-CSF to promote progenitor cell growth. The direct synergistic effect of IL- 1 alpha on CSF-1-induced progenitor growth was observed in all progenitor populations examined (Lin-, Lin-Thy-1+, and Lin-Thy-1-) and was inhibited by MoAb 35F5. However, the direct synergistic effect of IL-1 alpha on GM-CSF-responsive progenitors. Lin- and Lin-Thy-1+, was partially inhibited by MoAb 35F5. In contrast, the MoAb antitype II IL- 1R (MoAb 4E2) could not inhibit the direct synergistic effects of IL-1 alpha on CSF-1- or GM-CSF-induced progenitor growth. Thus, IL-1 alpha directly and indirectly stimulates the growth and differentiation of purified progenitors through the type I IL-1R but not the type II IL-1R.

In a recent paper, Francis Y. Lin proposes a “memory-challenge” to two main interpretations of Wittgenstein’s private language argument: the “no-criterion-of-correctness” interpretation and the “no-stage-setting” interpretation. According to Lin, both camps of interpretation fail to explain why a private language is impossible within a short time period. To answer the “memory-challenge”, Lin motivates a grammatical interpretation of the private language argument. In this paper, I provide a critical discussion of Lin’s objection to these interpretations and argue that Lin’s objection fails. In the case of the “no-stage-setting” interpretation, Lin suggests that the private language user can establish a stage within a short time period. However, I show that this stage is insufficient for a private language to be used correctly. In the case of the “no-criterion-of-correctness” interpretation, Lin believes that since memory is reliable within a short period, no criterion is needed for the correct use of a private language. However, I argue that his objection attacks a strawman, since the interpretation concerns the structure of justification, rather than the weakness of memory itself. I conclude with a critical discussion on memory and primitive expressions, the latter of which are crucial to Wittgenstein’s approach to public language. This discussion will help to draw a sharp line between private language and public language, and cast some doubt on Lin’s grammatical interpretation.

Protein translation initiation is a conserved process involving many proteins acting in concert. The 13 subunit eukaryotic initiation factor 3 (eIF3) complex is essential for assembly of the pre-initiation complex that scans mRNA and positions ribosome at the initiation codon. We previously reported that a gain-of-function (gf) mutation affecting the G subunit of theCaenorhabditis eleganseIF3 complex,eif-3.g(gf), selectively modulates protein translation in the ventral cord cholinergic motor neurons. Here, through unbiased genetic suppressor screening, we identified that the genelin-66mediateseif-3.g(gf)-dependent protein translation in motor neurons. LIN-66 is composed largely of low-complexity amino acid sequences with unknown functional domains. We combined bioinformatics analysis with in vivo functional dissection and identified a cold-shock domain in LIN-66 critical for its function. In cholinergic motor neurons, LIN-66 shows a close association with EIF-3.G in the cytoplasm. The low-complexity amino acid sequences of LIN-66 modulate its subcellular pattern. As cold-shock domains function broadly in RNA regulation, we propose that LIN-66 mediates stimulus-dependent protein translation by facilitating the interaction of mRNAs with EIF-3.G.

Abstract SPI1 belongs to the E26-transformation-specific (Ets) family of transcription factors and is exclusively expressed in hematopoietic cells. Originally identified as an oncogene that blocks erythroblast differentiation, SPI1 plays a critical role in the development and lineage commitment of both myeloid and lymphoid cells. Gene knockout studies show that spi1−/− mice exhibit a multi-lineage defect in the generation of monocytes, granulocytes, B, T, and dendritic cells. While SPI1 expression is required for normal hematopoietic cell development, the level of SPI1 expression influences specific lineage commitment. Using in-house generated species-specific clones, human peripheral blood mononuclear cells (PMBCs) and mouse splenocytes were evaluated for SPI1 expression by flow cytometric analysis. In human PMBCs, Lin− HLA-DR+ CD11c+ CD303− monocyte-derived dendritic cells (mDCs) exhibit high levels of expression while Lin− HLA-DR+ CD11cint CD303+ plasmacytoid dendritic cells (pDCs) do not express SPI1 protein. These data suggest SPI1 expression is required for the development and function of human mDCs, while SPI1 expression is not necessary for terminally differentiated human pDCs. Interestingly, SPI1 expression is detected in both murine DC subsets: Lin− I-A/I-E+ CD11c+ CD317− mDCs and Lin− I-A/I-E+ CD11cint CD317+ pDCs, although the expression level in pDCs is significantly lower than in mDCs. These observations indicate expression patterns differ between mouse and human cells, and demonstrate that DC subsets exhibit differential expression of SPI1 protein.

Introduction. Amyotrophic lateral sclerosis (ALS) is a fatal, neurodegenerative disease, leading to loss of muscle strength and motor control. Impaired speech and swallowing lower the quality of life and consequently may induce acute respiratory failure. Bone marrow-derived stem and progenitor cells (SPCs) may be a valuable source of trophic factors. In this study, we assessed whether adjuvant cellular therapy could affect the levels of selected neurotrophins and proinflammatory factors in the cerebrospinal fluid (CSF) and subsequently prevent the deterioration of articulation. Materials and Methods. The study group consisted of 32 patients with sporadic ALS who underwent autologous lineage-negative (Lin−) stem cell intrathecal administration to the spinal canal. Lin− cells were aspirated from the bone marrow and isolated using immunomagnetic beads and a lineage cell depletion kit. Patients were examined for articulatory functions by means of the Voice Handicap Index (VHI) questionnaire and Frenchay Dysarthria Assessment (FDA). In parallel, we carried out the analysis of selected trophic and proinflammatory factors in CSF utilizing multiplex fluorescent bead-based immunoassays. Results. Of the 32 patients who received the Lin− progenitor cell therapy, 6 (group I) showed improvement in articulatory functions, 23 remained stable (group II), and 3 deteriorated (group III) on the 28th day. The improvement was particularly noticeable in a better cough reflex, laryngeal time, and dribble reflex. A statistically significant lower level of brain-derived neurotrophic factor (BDNF) was observed on day 0 in group I compared to group II. The CSF concentrations of C-reactive protein (CRP) in group I significantly decreased 7 days after Lin− SPC transplantation. On the contrary, a significant increase in the tumor necrosis factor receptor (TNF-R) level was confirmed among patients from group I with improvement of dribble and coughing reflex, tongue movements, and respiration on the 7th day, as well as on day 28 including dribble reflex solely. Conclusions. An application of Lin− stem cells could potentate the beneficial humoral effect. The prevention of deterioration of articulatory functions in ALS patients after applying adjuvant Lin− stem cell therapy seems to be promising. Although the procedure is safe and feasible, it requires further in-depth studies.

AbstractDue to their unique properties, bone marrow-derived Lin− cells can be used to regenerate damaged tissues, including skin. The objective of our study was to determine the influence of the skin tissue-specific microenvironment on mouse Lin− cell proliferation and migration in vitro. Cells were analyzed for the expression of stem/progenitor surface markers by flow cytometry. Proliferation of MACS-purified cells in 3D cultures was investigated by WST-8 assay. Lin− cell migration was evaluated by in vitro scratch assay. The results obtained show that basement membrane matrix is more effective for Lin− cell proliferation in vitro. However, type I collagen matrix better enhances the re-epithelization process, that depends on the cell migratory properties. These studies are important for preparing cells to be used in transplantation.

Recently, Lin (2000) issued an erratum reporting a number of typographical errors in his seminal 1989 paper. Further, changes in Stata Version 7 required modification of concord. This note reports the effect of the errors and provides a corrected and updated program.

This paper studies the acceptance sampling for exponential distributions with type-I and type-II adaptive progressive hybrid censored samples. Algorithms are proposed for deriving Bayesian sampling plans. We compare the performance of the proposed sampling plans with the sampling plans of Lin and Huang (2012). The numerical results indicate that the proposed sampling plans outperform the sampling plans of Lin and Huang (2012).

Abstract We show the molecular and functional characterization of a novel population of lineage-negative CD34-negative (Lin−CD34−) hematopoietic stem cells from chronic myelogenous leukemia (CML) patients at diagnosis. Molecular caryotyping and quantitative analysis of BCR-ABL transcript demonstrated that about one third of CD34− cells are leukemic. CML Lin−CD34− cells showed kinetic quiescence and limited clonogenic capacity. However, stroma-dependent cultures induced CD34 expression on some cells, cell cycling, acquisition of clonogenic activity and increased expression of BCR-ABL transcript. Lin−CD34− cells showed hematopoietic cell engraftment rate in immunodeficient mice similar to Lin-CD34+ cells whereas endothelial cell engraftment was significantly higher. Gene expression profiling revealed the down-regulation of cell cycle arrest genes, genes involved in antigen presentation and processing, while the expression of genes related to tumor progression, such as angiogenic factors, was strongly up-regulated when compared to normal counterparts. Flow cytometry analysis confirmed the significant down-regulation of HLA class I and II molecules in CML Lin−CD34−cells. Imatinib mesilate did not reduce fusion transcript levels, BCR-ABL kinase activity and clonogenic efficiency of CML Lin− CD34− cells in vitro. Moreover, leukemic CD34− cells survived to BCR-ABL inhibitors in vivo. Thus, we identified a novel CD34− leukemic stem cell subset in CML with peculiar molecular and functional characteristics.

Umbilical cord blood (UCB)-derived stem/progenitor cells (SPCs) have demonstrated the potential to improve neurologic function in different experimental models. SPCs can survive after transplantation in the neural microenvironment and indu ce neuroprotection, endogenous neurogenesis by secreting a broad repertoire of trophic and immunomodulatory cytokines. In this study, the influence of brain-derived neurotrophic factor (BDNF) pre-treatment was comprehensively evaluated in a UCB-derived lineage-negative (Lin−) SPC population. UCB-derived Lin− cells were evaluated with respect to the expression of (i) neuronal markers using immunofluorescence staining and (ii) specific (TrkB) receptors for BDNF using flow cytometry. Next, after BDNF pre-treatment, Lin− cells were extensively assessed with respect to apoptosis using Western blotting and proliferation via BrdU incorporation. Furthermore, NT-3 expression levels in Lin− cells using RQ PCR and antioxidative enzyme activities were assessed. We demonstrated neuronal markers as well as TrkB expression in Lin− cells and the activation of the TrkB receptor by BDNF. BDNF pre-treatment diminished apoptosis in Lin− cells and influenced the proliferation of these cells. We observed significant changes in antioxidants as well as in the increased expression of NT-3 in Lin− cells following BDNF exposure. Complex global miRNA and mRNA profiling analyses using microarray technology and GSEA revealed the differential regulation of genes involved in the proliferation, gene expression, biosynthetic processes, translation, and protein targeting. Our results support the hypothesis that pre-treatment of stem/progenitor cells could be beneficial and may be used as an auxiliary strategy for improving the properties of SPCs.

Abstract Abstract 2953 Poster Board II-929 Stem cells have been shown to play an important role in the pathogenesis and maintenance of a significant number of malignancies, including leukemias. Similar to normal hematopoiesis the AML cell population is thought to be hierarchically organized. According to this model, only a few stem cells (LSC) are able to initiate and maintain the disease. The inefficient targeting of the leukemic stem cells (LSC) is considered responsible for relapse after the induction of complete hematologic remission (CR) in AML. t(6;9)-positive AML is classified as a separate entity, because of its young age of onset and poor prognosis. The t(6;9) associated fusion protein is DEK/CAN. Assuming that in AML the genetic aberration, here the t(6;9) and the expression of DEK/CAN, represents the initiation event of the leukemogenic process we wanted i.) to disclose its effects on the biology of primitive hematopoietic stem cells (HSC) and its leukemogenic potential and ii.) to characterize the leukemia-initiating cell and the cell population able to maintain the disease in vivo. The model was based on a classical transduction/transplantation system of murine Sca1+/lin- HSC combined with a novel approach for the enrichment of transformed cells with long-term stem cell properties. We found that i.) DEK/CAN induced leukemia from the Sca1+/lin− HSC with a frequency of 20% and a long latency of 8-12 months. ii.) DEK/CAN did not efficiently block the differentiation of committed progenitors; iii.) DEK/CAN increased number of colony forming cells in Sca1+/lin− HSC which did not exhibit increased replating efficiency as compared to controls; iv.) DEK/CAN augmented ST-HSC potential but not LT-HSC of murine Sca1+/lin− HSCs, most likely due to its incapacity to up-regulate p21Cip1/Waf1 expression. Based on the hypothesis that DEK/CAN exerts its leukemogenic effects on only a small proportion of the Sca1+1/lin- population, we proceeded to select and to amplify rare DEK/CAN-positive cells with the leukemia-initiating potential, by a negative selection of cell populations with proliferation potential without long term stem cell-capacity (LT). Therefore we expressed DEK/CAN in Sca1+/lin− cells and enriched this population for LT- (lin−/Sca1+/c-Kit+/Flk2−) and ST-HSC (lin−/Sca1+/c-Kit+/Flk2+). After a passage first in semi-solid medium for 7 days and subsequent transplantation into lethally irradiated mice, cells from the ensuing CFU-S day12 were again transplanted into sublethally recipient mice. We here report that i.) after 4 to 42 weeks, 6/6 mice developed AML without signs of differentiation in the group transplanted with the lin−/Sca1+/c-Kit+/Flk2− population but not from that transplanted with lin−/Sca1+/c-Kit+/Flk2+ cells; ii.) the DEK/CAN-induced AML was efficiently transplanted into secondary recipients exhibiting a very aggressive clinical picture; iii.) the leukemic cell population gave origin to four different clearly distinct subpopulations defined by surface marker pattern as an expression of populations with distinct differentiation status, all able - after sorting - to give leukemia in sublethally irradiated recipients: lin−/Sca1+/c-Kit+/CD34− (LT) lin−/Sca1+/c-Kit+/CD34+ (ST), Sca1−/c-Kit+/Mac1+/Gr1+, Sca1−/c-Kit+/Mac1−/Gr1−. These findings strongly suggest that there is a difference between a leukemia-initiating (L-IC) and leukemia-maintaining (L-MC) cell population in the murine DEK/CAN leukemia model. In contrast to the L-IC, represented by a very rare subpopulation of primitive HSC, recalling a hierarchical stem cell model, the L-MC is represented by a larger cell population with a certain grade of phenotypical heterogeneity, but a high grade of functional homogeneity recalling a stochastic cancer induction model. Disclosures: No relevant conflicts of interest to declare.

The possibility of a chloro-bridged alkyl(aryl)chloroindium amide is also realized in [Mes(Cl)InN(SiMe3)2]2 (2), as shown by X-ray analysis. The reaction of a 1:1-mixture of i-Pr2InCl/i-PrInCl2 with LiN(H)t-Bu leads to all three possible compounds: [i-Pr(Cl)InN(H)t-Bu]2 (1), {i-Pr2In[N(H)t-Bu]2In(Cl)i-Pr} (5), and [i-Pr2InN(H)t-Bu]2 (6). Two isomers of the new compound 5 are detectable in solution. The crystal structure has been determined for the less sterically hindered isomer 5a, which forms hydrogen-bridged (Cl-H-N-bridge) infinite chains along the c-axis.

Lin Shu and Wang Shouchang's Bali chahua nü yishi (1899), a translation of La dame aux camélias (1848) by Alexandre Dumas fils, has usually been read with an eye to the idealized figure of the noble courtesan in the Chinese literary tradition. The resemblance of the lady of the camellias to that figure has served to explain both her popular appeal to late Qing readers and her transformation into a Chinese literary and cultural icon. Her iconic status, however, has overshadowed the complex ways in which Lin and Wang's translation retells Dumas fils's story in relation to the array of late Qing courtesan narratives that would have been known to contemporary readers. By closely reading the original story in light of these narratives, this article shows how Dumas fils's tale shares many elements with late Qing courtesan fiction, including negative images of courtesanship, which the translation omits or rewrites. I examine how the translation transforms the morally ambiguous heroine into a devoted lover and re-situates her suffering (passio) and recognition — central motifs in the original story's Christian mythos — within a nostalgic reflection on courtesan-literati culture, where the courtesan serves as the alter ego of the unrecognized literatus.

In this paper, I discuss some characteristics of Chinese quantified NPs that are distinct from those of standard quantifiers (Section 1), and argue for an analysis of these NPs as generalized quantifiers built up over plural individuals (Section 2). In effect, I am suggesting a compositional approach with quantifiers contributing quantificational force and distributive operators introducing distributivity. This proposal is, in many ways, similar to Lin' s (1998) account, but the crucial difference has to do with the status of the classifier. While Lin does not make any reference to the classifier, its semantic contribution is essential for me. This will become obvious when I discuss two major consequences of the proposed approach, one having to do with distributivity in Chinese universal quantifiers (Section 3) and the other with the semantics of definite NPs (Section 4).

AbstractLiterary back-translation, the practice of bringing a text back into the language of its source after it has passed through one or more translations, has long been overlooked. To recuperate its richly entangled global history, this essay looks to the representation of translation in Cervantes's Don Quixote and to the novel's recent back-translation based on 魔俠傳 (Moxia Zhuan; Story of the Enchanted Knight), a 1920s translation into classical Chinese by Lin Shu that itself was an indirect translation based on two eighteenth-century English editions of Cervantes's text. While foregrounding the generative labor of translators, both fictional and historical, the transnational and diachronic scope of this back-translational loop prompts us to rethink the domesticating/foreignizing binary. Taking a cue from Cervantes's metaphor of translation as the knotty back side of a tapestry, this study seeks to overturn front-facing notions of translation by approaching its history from the back.

In Caenorhabditis elegans, the fates of the six multipotent vulva precursor cells (VPCs) are specified by extracellular signals. One VPC expresses the primary (1 degrees) fate in response to a Ras-mediated inductive signal from the gonad. The two VPCs flanking the 1 degrees cell each express secondary (2 degrees) fates in response to lin-12-mediated lateral signaling. The remaining three VPCs each adopt the non-vulval tertiary (3 degrees) fate. Here I describe experiments examining how the selection of these vulval fates is affected by cell cycle arrest and cell cycle-restricted lin-12 activity. The results suggest that lin-12 participates in two developmental decisions separable by cell cycle phase: lin-12 must act prior to the end of VPC S phase to influence a 1 degrees versus 2 degrees cell fate choice, but must act after VPC S phase to influence a 3 degrees versus 2 degrees cell fate choice. Coupling developmental decisions to cell cycle transitions may provide a mechanism for prioritizing or ordering choices of cell fates for multipotential cells.

The present study was designed to confirm the traditional use of the fruits of Ficus hispida Linn. (Moraceae) as an antioxidant agent. Fruits of the plant extracted with methanol and crude methanol extract (CME) were further fractionated with n-hexane, chloroform, and ethyl acetate. All the fractions, n-hexane (NHF), chloroform (CHF), ethyl acetate (EAF), aqueous (AQF) and CME were preliminary screened for in vitro antioxidant activity and total phenolic and total flavonoid content. In DPPH radical scavenging assay, CME exhibited highest scavenging activity (IC50 = 11.20 µg/mL) as compared to other fractions. In this assay, IC50 of reference standard BHT was 5.10 µg/mL. The reducing power of the samples was in the order as AQF > CME > CHF > EAF > NHF. The results for hydrogen peroxide scavenging activity indicated that CME, EAF and AQF had almost the same scavenging activity except NHF. Total antioxidant capacity of CME and other fractions were ranked as CHF > AQF > CME > EAF > NHF. In the assay of antioxidant constituents (total phenol and total flavonoids content), the CME had highest phenolic and flavonoids content. The results indicate that Ficus hispida fruits could be considered as a potential source of natural antioxidant.

This article explores the role that local elites played in the development of the Mazu cult, a local goddess cult in Putian district in late imperial China. I argue that local elites were central in the promotion and transmission of the cult. Through compiling and writing key Confucian texts featuring Mazu, they reshaped, manipulated, and represented certain aspects of her cult in accordance with their interests. As a result of the activities of local elites, Mazu became associated with the Lin lineage, an influential local lineage. In this manner, Mazu came to be seen as an expression of the lineage’s authority, as well as an imperial protector embodying local loyalty to the state and a daughter who was the paradigm filial piety. In addition to the literary production, local elites, in particular the descendants from the Lin lineage, established an ancestral hall of Lin in the port of Xianliang dedicated to Mazu, further sanctioning divinely the local elites’ authority and privilege in the community. I conclude that the locally promoted version of goddess worship operated at the intersection of state interests, Confucian ideology, the agency of local elites, and the dynamics of popular religiosity.

The genetic analysis of 28 lethal mutations has revealed that the mutator locus mut-4(st700)I causes both putative single-gene mutations and deficiencies. We have identified lethal mutations in three different genes, in addition to seven deficiencies. There is a mutational hot spot on linkage group V (left) around the lin-40 locus. Six mutations appear to be alleles of lin-40. In addition, 5 of 7 deficiencies have breakpoints at or very near lin-40. All seven deficiencies delete the left-most known gene on linkage group V (left) and thus appear to delete the tip of the chromosome. This is in contrast to gamma ray and formaldehyde induced deficiencies, which infrequently delete the closest known gene to the tip of a chromosome.Key words: Caenorhabditis elegans, mutator, deficiencies, lethal mutations.

AbstractIn his Research Note, “Shun 舜 and the Interpretation of Early Orthographical Variation,” in this issue of Early China, Jonathan Smith made several claims about the early evolution of the graph 舜, in particular suggesting that it should be identified with the graph lin 粦 that occurs in certain Western Zhou bronze inscriptions. While showing that these claims are ill-supported, I nevertheless concur in the identification with the bronze-inscriptional graph, but show that the word being written is 濬 ~ 浚 “deep, profound” and in no way connected with lin 粦.

Abstract MDSC’s are a heterogeneous population of myeloid cells at various differentiation stages. The human MDSC phenotypes, are controversial and subdivided into macrophage (M-), granulocytic (G-) and immature (i-) MDSC’s. Our studies used a single staining tube with antibodies to linage markers (CD3, CD19 and CD56), HLA-DR, CD11b, CD14, CD15, CD16, CD33, CD45, PD-L1 and LOX-1. Mobilized lymphoma patient apharesis (CA) products provided a product with a high frequency of MDSCs compared to normal donor blood samples (PB). SSCA and FSA identified lymphocytes, granulocytes and monocyte populations. A total population (CD45+), MDSCs (Lin-HLA-DR-CD11b+CD14+, Lin-HLA-DR-CD11b+CD14-CD33+ and Lin-HLA-DR−CD11b+ CD15+) and non MDSC myeloid populations (Lin-HLA-DR+CD14+, Lin-HLA-DR+CD33+ and Lin-HLA-DR+ CD15+Lin-HLA-DR-11b-) were exported from FlowJo for Spade analysis. This identified 11 nodes with 4 MDSC sub-populations. Impressively, cells from the non-MDSC populations, including monocytes and granulocytes did not occur within these nodes. SPADE identified an M-MDSC, G-MDSC and two iMDSC subsets with an increased frequency in the CA samples. Increased expression of LOX-1 and CD16 occurred in CA MDSCs vs. PB. Intracellular staining for expression of NOS-2 and Arg-1 were consistent with the M and G-MDSC subsets. Node 7 mainly expressed M-MDSC’s with a low frequency of CD15 dull G-MDSC’s. The cell frequency for G-MDSC’s in node 7 was 1,334 for CA and 0 for PB with M-MDSC’s 1,079 and 43 respectively. Node 8 had a frequency (3,319) of G-MDSC’s for CA and 2,286 for PB with M-MDSC’s 188 and 15 respectively. In summary SPADE analysis can be used to cleanly identify MDSCs from monocytes and granulocytes and provide insight into novel MDSC subsets.

Abstract Influenza viral infections have the ability to reprogram the immune system of the host and alter its functions to fight infections. However, the exact mechanisms and functions of virus-induced innate immunity in the context of leukemia are not yet fully understood. Our previous research has shown that a patient with refractory acute myeloid leukemia (AML) achieved complete remission (CR) after being infected with the Influenza A virus (IAV, H1N1 subtype), and this was functionally validated in two different animal disease relevant models. Our recent study demonstrates that mice pre-trained with the IAV promotes a long-term anti-leukemia effect. The anti-leukemia activity of IAV-induced trained immunity is due to the transcriptomic rewiring of hematopoietic stem and progenitor cells (HSPCs), resulting in a sustained enhancement of myeloid cells reprogramming toward an antitumor phenotype. This mechanism involves the upregulation of type I interferons (IFNs) and subsequent downregulation of inflammatory proteins. Our findings shed light on the dynamic changes in innate immune cells following viral infections and indicate a novel anti-leukemia facet of appropriate rewiring of granulopoiesis via upregulation of type I IFNs. Statement of significance The study profiles the dynamic changes in immune cells during exposure to the Influenza virus and sheds light on the role of Influenza trained innate immunity in anti-leukemia effect. The study also explores a novel anti-leukemia facet of appropriate rewiring of granulopoiesis via upregulation of the type I IFN pathway. These findings suggest that innate immune training could be a promising adjuvant therapy for cancer and warrant further investigation. Legend for Figure 5 . Abstract The purpose of this paper is to establish a necessary and sufficient condition for the boundedness of general product singular integral operators introduced by Han, Li and Lin [Y. Han, J. Li and C.-C. Lin, Criterion of the L 2 L^{2} boundedness and sharp endpoint estimates for singular integral operators on product spaces of homogeneous type, Ann. Sc. Norm. Super. Pisa Cl. Sci. (5) 16 2016, 3, 845–907] on the multiparameter Lipschitz spaces of homogeneous type M ~ = M 1 × ⋯ × M n {\tilde{M}=M_{1}\times\cdots\times M_{n}} . Each factor space M i {M_{i}} , 1 ≤ i ≤ n {1\leq i\leq n} , is a space of homogeneous type in the sense of Coifman and Weiss. These operators generalize those studied by Journé on the Euclidean space and include operators studied by Nagel and Stein on Carnot–Carathéodory spaces. The main tool used here is the discrete Littlewood–Paley–Stein theory and almost orthogonality together with a density argument for the product Lipschitz spaces in the weak sense.
