Resumo:

RÉSUMÉ Comment réfléchir aujourd'hui sur l'initiation en christianisme ? L'anthropologie renvoie à un sens de l'initiation plus précis que ne le fait le champ de l'initiation sacramentelle, très large. Et ce sens fondamental se rapporte notamment à la distinction entre rites d'initiation et rites de passage. Les premiers se fondent dans le processus d'incorporation proprement rituelle. Cela dit, il reste que l'approche de l'initiation dans nos sociétés modernes se présente plus généralement comme un chemin de sens évoquant de loin et par traces les antiques expériences initiatiques. L'enjeu est de saisir comment ces expériences continuent de travailler les quêtes religieuses et chrétiennes actuelles.

Introduction: Les patients dont l'arrêt cardiaque extrahospitalier (ACEH) n'a pas été témoigné sont généralement exclus des protocoles de réanimation par circulation extracorporelle puisque le délai avant l'initiation de leur réanimation est inconnu. Il a été proposé que la présence d'un rythme initial défibrillable (RD) est fortement suggestif une très courte période avant l'initiation des manœuvres de réanimation. La présente étude vise à décrire l'association entre la durée avant l'initiation de la réanimation et la présence d'un RD chez des patients souffrant d'un ACEH. Methods: Cette étude de cohorte a été réalisée à partir des bases de données collectées de la Corporation d'Urgences-santé dans la région de Montréal entre 2010 et 2015. Les patients dont l'arrêt était témoigné, mais dont les témoins n'ont pas entamé de manœuvres de réanimation, ont été inclus. Nous avons également inclus les patients dont l'arrêt était témoigné par les paramédics comme groupe contrôle (durée avant l'initiation de la réanimation = 0 minute). Les patients avec un retour de circulation spontanée avant l'arrivée des services préhospitaliers ont été exclus, tout comme ceux dont le rythme initial était inconnu. Nous avons décrit l’évolution de la proportion de chacun des rythmes et construit une régression logistique multivariée ajustant pour les variables sociodémographiques et cliniques pertinentes. Results: Un total de 1751 patients (1173 hommes et 578 femmes) d'un âge moyen de 69 ans (±16) ont été inclus dans l'analyse principale, parmi lesquels 603 (34%) avaient un RD. Un total de 663 autres patients ont vu leur ACEH témoigné directement par les paramédics. Un plus court délai avant l'initiation des manœuvres est associé à la présence d'un RD (rapport de cotes ajusté = 0,97 [intervalle de confiance à 95% 0,94-0,99], p = 0,016). Cependant, cette relation n'est pas linéaire et la proportion de RD ne diminue pas avant notablement jusqu’à ce que 15 minutes s’écoulent avant le début de la réanimation (0 min = 35%, 1-5 min = 37%, 5-10 min = 35%, 10-15 min = 34%, +de 15 min = 16%). Conclusion: Bien que la proportion de patients avec un RD diminue lorsque le délai augmente avant l'initiation des manœuvres, cette relation ne semble pas linéaire. La baisse principale de la proportion de patients avec RD semble se produire suite à la quinzième minute de délai avant le début de la réanimation.

Resumo:

Le mûrissement des tomates est un processus finement régulé avec de nombreux changements dynamiques qui concernent les voies du métabolisme primaire et secondaire, la respiration ou encore la signalisation hormonale. Comme les autres fruits climactérique, la tomate connait une transition développementale du stade « pré-maturation » à la phase de maturation qui est associée à une augmentation de la respiration au début de la maturation et à une augmentation de la production autocatalytique d'éthylène. Les facteur de réponse à l'éthylène, de la sous-clade E (ERF.E), ont un profil d'expression fortement associé au mûrissement du fruits de tomate. Les protéines ERF.E de la tomate sont notamment caractérisées par la présence d'un motif N-terminal conservé, qui a été identifié comme cible de la voie N-end rule en condition d’hypoxie chez leurs orthologues d'Arabidopsis. Cette étude dévoile pour la première le rôle positif de ERF.E1 dans la maturation des fruits de la tomate. La surexpression d'ERF.E1 favorise l'initiation du mûrissement, et est accompagnée d'une production avancée d'éthylène et d'une déplétion précoce de l'oxygène dans le gel, par rapport aux lignées WT et KO-erf.e1. ERF.E1 régule positivement les activités promotrices des gènes clés de la maturation, RIN et NOR, ainsi que les gènes de biosynthèse de l'éthylène du système 2. De plus, les résultats ont montré qu’en condition d’hypoxie, ERF.E1 est régulé aux niveaux transcriptionnel et post-traductionnel. En effet, les conditions hypoxiques induisent l’expression d’ERF.E1 et stabilise la protéine ERF.E1. Cette thèse se concentre principalement sur le rôle d'ERF.E1 dans l'initiation de la maturation, grâce à la caractérisation des lignées altéré dans l’expression de ERF.E1 et à l’étude des réseaux d'interaction protéiques potentiels d'ERF.E1
Tomato fruit ripening is a finely regulated process with many well-known dynamic changes, from metabolites, color, and respiration rate to hormone level. As a climacteric fruit, tomato fruit experiences a developmental transition characterized by a burst of respiration at the onset of ripening and a rise in autocatalytic ethylene production. Ethylene Response Factors (ERFs) are end mediators of ethylene responses and members of subclade E (ERF.E) of this transcription factor family display remarkable ripening-associated expression patterns in tomato fruit. Notably, tomato ERF.E proteins are characterized by the presence of a consensus N-terminal motif, which has been identified in their Arabidopsis orthologs as the target of the N-end rule pathway under hypoxia. The first outcome of our study is to reveal that initiation of tomato fruit ripening is preceded by a dramatic drop in oxygen levels in locular gel, thus giving a solid physiological marker of ripening initiation. The study also unveils the function of ERF.E1 in tomato fruit ripening showing that overexpression of ERF.E1 promotes the initiation of ripening, by advancing both ethylene production and oxygen depletion event in the gel, compared with the WT and KO-erf.e1 lines. ERF.E1 positively regulates the promoter activities of key ripening genes, RIN and NOR, as well as the ethylene biosynthesis genes involved in System 2 ethylene production. In addition, the data show that ERF.E1 is regulated by hypoxia at both transcriptional and post-translational levels. Hypoxia conditions induce transcription of ERF.E1 and promote ERF.E1 protein stability. Overall, while the study unraveled the role of ERF.E1 in ripening initiation based on the analysis of ERF.E1 impaired lines are altered at the early ripening stages it also uncovered potential proteins interacting with the ERF.E1 network components

Deux grandes théories sont au centre du discours sur l'initiation. Pour l'une, l'initiation est une expérience spirituelle; elle marque le passage du profane au sacré. Pour l'autre, l'initiation est un processus d'intégration sociale; un passage de l' "état naturel" à l' "état social". Ces théories constituent historiquement deux interprétations alternatives, et même exclusives du phénomène initiatique. Définir la nature de l'éducation initiatique, passe par la critique de ces conceptions. Du point de vue des objectifs, l'analyse montre qu'ils sont pluriels. Les enseignements ésotériques subsument les enseignements exotériques. Les moyens d'action et les techniques de l'effet sont imbriqués. Ces trois pôles de réflexion mettent en évidence la multi dimensionnalité de l'initiation. (première partie, chapitres 1,2,3. ) La deuxième partie, qui repose sur l'étude de l'initiation au Gabon, montre comment celle-ci participe à cette polyvalence; aussi bien à travers le rite de la circoncision (le "sakwe") qu'à travers la société initiatique masculine "bwete" (chapitres 4 et 5). Par ailleurs, l'étude de l'initiation sert de modèle d'approche à l'éducation traditionnelle. La comparaison de l'institution initiatique et de l'institution scolaire, autour des questions de la nature de l'enfant, des savoirs et des idéaux de chacune d'elles, révèle la présence d'un certain nombre de points de convergence et de divergence qui rendent problématique une éventuelle compatibilité fondamentale et structurelle. Ma thèse a porté sur la régulation de l'initiation de la réplication chez les vertébrés. J'ai participé à l'identification et la caractérisation d'origines de réplication (ori) sur 1% du génome humain. Ces ori ont été identifiées dans des cellules HeLa grâce à la quantification de petits brins naissants, spécifiques de l'initiation de la réplication. Nous avons pu montrer qu'un lien fort existe entre les ori et les îlots CpG et que l'association des ori avec des marques de chromatine ouverte comme l'acétylation des histones n'est pas indispensable pour l'initiation. De plus, il n'y a pas de lien clair entre densité en ori et moment de réplication d'une région génomique donnée. Je me suis aussi intéressée à l'étape de licensing c. -à-d. La fixation au niveau des sites d'initiation potentiels du complexe de Pré-réplication (Pré-RC) avec en premier lieu ORC (Origin récognition complex). Le licensing s'effectue en phase Gl du cycle cellulaire et seuls les sites de fixation du Pré-RC peuvent être utilisés comme site d'initiation. Nous avons choisi de travailler avec la lignée aviaire DT40 qui effectue la recombinaison homologue à forte fréquence. J'ai donc construit des lignées qui expriment une protéine Orcl ou Orc2 étiquetée à partir du locus endogène. J'ai réalisé des immunoprécipitations de chromatine pour identifier leurs sites de fixation. Nous arrivons à avoir des enrichissements faibles mais cohérents de Orcl au niveau des ori connues du poulet. Comme les faibles enrichissements obtenus compliquent des approches plus globales, nous envisageons de répéter ces expériences sur des cellules en phase Gl, obtenues par élutriation, afin d'augmenter les enrichissements obtenus
My PhD focused on the molecular mechanisms governing initiation of DNA replication in vertebrates. I participated in the identification and characterization of origins of replication (ori) on 1% of the human genome. These ori have been identified in HeLa cells by quantification of short nascent strands, which are specific of replication initiation. We showed that a strong link exists between ori and CpG islands and that the association of ori with open chromatin marks like histone acetylation is dispensable for origin specification. Furthermore, no clear link emerges between ori density and the moment at which a genomic region replicates. I have also been interested in the licensing step which consists in the binding on the initiation sites of the Pre-replication complex (Pre-RC), in first place ORC (Origin recognition complex). Licensing takes place in the Gl phase of the cell cycle and only Pre-RC binding sites can be used as initiation sites. We chose to work with the avian DT40 cell line, able to perform homologous recombination at high frequency. I thus constructed cell lines which express tagged versions of the Orcl and Orc2 proteins and from the endogenous locus. I realized chromatin immunoprecipitations (ChIP) to identify their binding sites. We obtain small but coherent enrichments of Orcl at well known chicken ori. As these small enrichments complicate genome-wide approaches, we plan to repeat these experiments with elutriated cells in Gl phase in order to increase the ChIP enrichments

Le mythe d'initiation, qui semble universel, apparait dans les litteratures francaise et africaine. Le schema initiatique nait probablement de l'intuition d'un paradis perdu qu'il est possible de retrouver symboliquement. Le motif du double dans la litterature peut traduire la dualite qu'il convient de depasser. Le chemin initiatique evoque un labyrinthe dont les entrelacs representent le chaos, la mort qu'il faut vaincre pour gagner le centre, espace de renaissance. Les heros ont a livrer un combat contre des monstres naturels ou humains. La transmutation de la personne est souvent rendue par des symboles de la femme et de la fecondite. Le cercle, la spirale, le serpent, les couleurs et les elements, de symbolisme ambivalent, s'integrent au processus resurrectionnel ; s'y ajoutent certains vegetaux, la musique et la danse accompagnant des fetes a caractere parfois dionysiaque. Un symbolisme alchimique peut exprimer l'evolution superieure de l'initie. Celui-ci possede des qualites morales exceptionnelles et des connaissances esoteriques. Son accomplissement est quelquefois symbolise par la figure de l'androgyne designant le depassement des contraires ainsi que la totalite existentielle. Les nombres sont associes a l'aventure initiatique, en particulier le un, le trois, le quatre et le sept. Les categories de l'espace et du temps transcendant sont symbolisees par des motifs verticaux tels que l'arbre, l'escalier, le pilier. . . , par les images du vol, de la fleche, et par des procedes d'ecriture. Le mythe d'initiation marque bon nombre d'oeuvres litteraires et peut se trouverdesacralise.

Influencé par l'initiation traditionnelle, le roman gabonais porte les germes profonds des rites et croyances du Gabon. On y rencontre des romans d'intégration sociale dans lesquels certains personnages, grâce aux rites sociaux de la circoncision et du " tchikumbi ", réussissent à passer du statut de l'enfance à celui des femmes et des hommes responsables. Ensuite, il y a des romans de purification spirituelle, dans lesquels les principaux personnages subissent une initiation leur permettant de percer les mystères du monde invisible et de maîtriser avec habilité les forces magiques. Enfin, les romans de sorcellerie où les sorciers se livrent à la métamorphose et à un véritable sabbat anthropophagique. Tous ces huit romans mettent en scène des personnages, un cadre spatio-temporel vertical et un langage initiatique qui dévoilent d'une part les rapports de l'homme gabonais avec le sacré clanique, et d'autre part, les relations entre les croyances traditionnelles et le christianisme, relations au départ conflictuelles et aujourd'hui constructives, grâce à l'inculturation religieuse basée sur la tolérance, le dialogue et l'acceptation de l'autre dans sa différence
Influenced by traditional initiation, gabonese novels embodies the basic ingredients of the rites and beliefs of the gabonese people. Gabonese novels evoke common themes which reflect different fundamental aspects of gabonese culture and tradition. In some novels, the subject of social integration is illustrated as certain characters undergo rites of circumcision or "tchikumbi" which marks the passage from childhood to adulthood. On novels, the subject of spiritual purification in other, principal characters undergo an initiation into the spiritual or invisible world and a mastery of the art of magic. And lastly, some novels situate on the theme of sorcery whereby sorcerers indulge in self metamorphosis and anthropophagi. These eight literary works which embody our literature highlight important symbolic characters, transcendental environment and an initiation parlance which unveils the one hand the link between the gabonese people and the sacred clan and on the other, the dynamic relationship between the traditional belief system and Christianity presumably the largest religion in Gabon, a relationship initially confrontational and later constructive based on inculturation, tolerance, dialogue and acceptance and recognition of mutual differences

Le thème de l'initiation subit des modifications lorsqu'il est pris en charge par l'écriture. Certes, on peut repérer le scénario initiatique dans son integralité dans le recit. Cependant, il semble que la vocation de l'écriture veuille contredire celle de l'initiation. En effet, l'ordre du rite de passage de l'enfance à l'âge adulte est contrarié : l'invasion qu'il subit débouche sur une quête inavouée de l'enfance et du paradis perdu. C'est pourquoi l'ecriture tente de réhabiliter de facon dissimulée une figure féminine trop longtemps réprimée. La quête du mythe et de l'oralité n'est donc qu'une manière de rejoindre la quête de la mère. Ainsi, plus qu'une renaissance, c'est fondamentalement une "innaissance" que l'initiation revendique à travers la litterature africaine
The initiation theme is modified when it iq taken in charge by the writing. We certainly can recognize the initiatory scenario in its whole in the narration. Nevertheless, it seems that the writing vocation is at variance with the initiation one. In fact the ritual order of the transition between the childhood to the adult age is modified : the inversion wich affects it, leads to an unconfessed request of the childhood and the lost heaven. That is why the writing tries to rehabilitate in a hidden way a female face repressed for too long time. The myth request and the oral speech are only a meaning to refind the request of the mother. Therefore, more than a rebirth, it is basically an "inside birth" that the initiation claims throught the african literature

Le vegf est un facteur angiogenique majeur. Le taux d'expression de cette proteine s'avere important puisque l'invalidation d'un seul allele du gene vegf conduit a la mort embryonnaire. Au debut de cette these, il etait clair que l'expression du vegf etait regulee au niveau transcriptionnel et post-transcriptionnel. Par contre, tres peu de travaux decrivaient une regulation de la traduction du vegf. Pourtant, la region 5nt du messager presente des caracteristiques incompatibles avec le mecanisme classique de balayage des ribosomes. En effet, cette region est extremement longue et structuree et possede plusieurs codons d'initiation en phase avec le codon aug classique. Au cours de cette these, nous avons montre que l'initiation de la traduction du vegf depend d'une entree atypique cap-independante des ribosomes dans la region 5nt du messager au niveau de deux structures secondaires appelees ires pour internal ribosome entry site. L'ires aval (ires a) controle l'initiation de la traduction au codon classique aug. Par contre, l'ires amont (ires b) controle l'initiation d'une nouvelle isoforme, que nous avons appelee l-vegf, initiee au niveau d'un codon non canonique cug, situe entre les deux ires. Cette isoforme comporte une extension amino-terminale de 180 acides amines qui lui confere une localisation intracellulaire, contrairement aux formes classiques du vegf connues pour etre secretees. Cette proteine est clivee pour generer un fragment amino-terminal specifique et un fragment de taille similaire a la forme initiee au codon aug. Nous avons entrepris l'etude du role de cette nouvelle isoforme. Nous pensons qu'une regulation differentielle de l'utilisation des deux ires, qui lient des facteurs cellulaires differents, permettrait de favoriser l'initiation a l'un ou l'autre codon et donc la production d'isoformes possedant probablement des fonctions distinctes.

Dental enamel consists mainly of calcium-deficient hydroxyapatite (CDHA). The formation and evolution of enamel is a progressive and complex process the final stage of which is post-eruptive maturation (PEM), when mineralisation is completed following tooth eruption and exposure to oral fluids. Although PEM is directly correlated with decrease in caries susceptibility, a complete model to describe the whole process does not exist. Several reports have recently suggested that the previously observed caries decline, which started with the introduction of F- in drinking water and dental products, has stopped and is in some cases in reverse. New research approaches are therefore needed, which will focus on caries prevention and not treatment. This project monitored the in vitro effects of simulated PEM on the physicochemical properties of dental enamel and proposed a model which describes the whole process. For this purpose, primary and permanent bovine enamel was tested, using a suitably designed demineralisation/remineralisation laboratory protocol as well as characterisation techniques. The results were applied to the synthesis of enamel proxies, which could mimic the physicochemical properties of dental enamel; these proxies were evaluated for their potential to be used as enamel substitutes in dental research.

Resumo:

The disparate concepts of Pop Art and abstract painting heavily influence the scope of my work. Finding a link between these two concepts has been the focal point of my studio practices. The apex of my process is the focus on commercial imagery as abstract form. The merging of these two concepts presents a complex composition of balance, color and information. This thesis explores the various concepts as well as influences that have propelled the evolution of my work. It chronicles the steps I have taken in my quest to articulate my conceptual ideas. By describing the works and defining their characteristics, this analysis gives further insight to my perception as well as process.
