This study is fulfilled to simulate numerically the interaction phenomenon of the bidimensional compressible and supersonic flow with M=2.28 of a turbulent boundary layer with an oblique choc wave. The oblique choc wave is released and oriented by a generator of choc in an angle of θw=8° on a plate plane maintained at a constant temperature. The interaction causes the detachment of the boundary layer and make it disturbed (The case of experimental study done by J. Deleuze (1995)). The contribution on the effects of turbulence and their characteristics is represented by k-w-SST model. This model is well adapted for this type of application. This study permits to underline the coherence of our analysis with existant numerical and experimental results. This simulation is effected by means of solver which based on the spatio-temporal discretization of the complex equations of Navier-Stockes with finite volumes method. We also use the Upwind scheme of second order.

Dans une certaine plage de nombre de Mach, l'interaction entre une onde de choc oblique et une couche limite s'effectue par une investigation numérique sur une plaque plane athermane, basée sur la résolution des équations de Navier-Stokes bidimensionnelles par le code Fluent. Cette interaction se matérialise sous la forme d'une onde de choc en 'lambda', décrite en premier par Mark en 1958. Elle peut être rencontrée dans les équipements des souffleries à haute enthalpie ou ceux relatifs à la distribution de gaz comprimés par exemple. Une bonne connaissance des phénomènes physiques intervenant dans de tels écoulements est nécessaire tant au point de vue des normes de sécurité qu'en terme d'accroissement des performances. Sous cette structure de choc, il se forme un bulbe de décollement massif engendrant une succession de structures tourbillonnaires en cas laminaire que turbulent due par l’effet du nombre de Mach amont qui est observé. Elle est suivie par un phénomène de transition de la réflexion régulière RR vers la réflexion de Mach MR pour le cas turbulent. Globalement les résultats numériques obtenus sont conformes avec ceux qui sont disponibles dans la littérature.

This article is a history of liquidity presented as interaction between metaphors and theoretical concepts in social contexts. While taking note of Zygmunt Bauman’s metaphor “liquid modernity,” the study instead surveys the wider conceptual field. The text turns around mercantile liquidity (liquidity as clarification) and liquidity in modern economics (characteristic of all assets), as well as older metaphors, notably the famous phrase of the Communist Manifesto, “all that is solid melts into air” (Alles Ständische und Stehende verdampft), which is revealed to have resonance in texts by poets, notably Heinrich Heine. The main result is the historical consistency of the field, where liquidity is a promise of knowledge and clarity.

Body-politic metaphors served historically as figurative vehicles to transmit assorted socio-political messages. Through an examination of the metaphors la mollesse (softness) and Adam Smith’s impartial spectator, this article will show that the language of eighteenth-century French and British writers was not simply heuristic or metaphorical. Contemporaries reacted to the growth of commerce and luxury, and the concomitant creation of new public spaces and forms of social interaction, by arguing that the corporeal mediated the social. I want to introduce the concept of corporeal sociability: cognitive physiology and the network of the senses, contemporaries argued, contained the information necessary to assess novel forms of commerce and revealed that sociability was congenitally embodied.

The debate between metaphor theorists and conceptual historians has been intensifying in recent years. This article takes this debate beyond the bias toward Blumenberg's metaphorology, and starts from the interaction view of metaphor as formulated by Max Black. The article opens with a theoretical framework that reformulates Black's notions of metaphorical resonance and emphasis. It adapts them to the requirements of Conceptual History, and adds a third, historical criterion for metaphoricity. It then applies these suggestions to the history of the metaphor play/game/Spiel/jeu within twentieth-century political thought. Here, the focus lies on the role this metaphor plays in the conceptual relations between the ideas of political order, conflict, and immanence.

This article introduces a collection of studies of biological concepts crossing over to other disciplines and nonscholarly discourses. The introduction discusses the notion of nomadic concepts as introduced by Isabelle Stengers and explores its usability for conceptual history. Compared to traveling (Mieke Bal) and interdisciplinary (Ernst Müller) concepts, the idea of nomadism shifts the attention from concepts themselves toward the mobility of a concept and its effects. The metaphor of nomadism, as outlined in the introduction, helps also to question the relation between concepts' movement and the production of boundaries. In this way conceptual history can profit from interaction with translation studies, where similar processes were recently discussed under the notion of cultural translation.

This article discusses possible dialogues between medical history and the history of concepts, suggesting that a “socio-conceptual-moral” history of medicine offers insightful elements for the historical analysis of conceptual change. Drawing mainly from Reinhart Koselleck’s Begriffsgeschichte and Ludwik Fleck’s theory of knowledge, I focus on three points of the “socio-conceptual-moral” perspective: the approach to medical statements as part of a semantic field, the interaction between a formulated concept and its practice, and negotiations about the meanings of medical concepts between different social arenas. I take the history of cancer prevention in Brazil as a case study to discuss these three aspects and emphasize the situated character of conceptual change. The article analyzes the period between the 1960s and the 1990s when substantial changes in the conceptual framework of cancer prevention confronted continuities in public health and medicine practices, policies, and institutions.

État et situation d’un système postérieurs à un choc, à un traumatisme, la transition « évoque l’idée d’un passage, d’un état à un autre, d’une position à une autre, que ce passage s’effectue graduellement, progressivement et enfin qu’il se passe quelque chose dans l’entre-deux ». C’est un processus qui engendre une interaction continue entre le processus amont et le processus aval, suivant la définition même du processus.À ce titre, elle établit la liaison entre le choc qui a initié le processus de résistance et le processus de résilience. Le changement qui en résulte affecte la durabilité du « système télescopé et traumatisé (STT) ».C’est à partir de ces divers éléments de définition que les enjeux et les conditions de vie (ou de sur-vie) de la transition sont précisés à l’aide de différents exemples et modèles en lien avec la systémique et ses applications dans le domaine du changement.Forgeant un système dynamique complexe, non-linéaire et chaotique, la transition avec la résilience (réactive et pro-active) induit le changement qui à son tour crée l’incertitude. Une adaptation s’impose pour comprendre le futur qui vient et qui est imprédictible dans un monde VICA ! Le processus du changement en U semble univoque et traduit paradoxalement une voie toute tracée, ce qui n’est pas le cas du processus en Wn ou en L ! Loi de Murphy oblige !La pensée systémique semble se présenter en panacée de par la durabilité des systèmes complexes, mais un concept plus fécond lui vient en aide, suivant la dialogique morinienne pour y adjoindre le concept de « système résilient et agile » ; marquant le passage de STT à SRA !Telle est la conviction de cet article qui essaie d’innover dans le domaine de la science politique, notamment en politiques publiques elles-mêmes systèmes complexes...

Introduction. Monolithic zirconia is able to achieve certain aesthetic, but its durability in resisting fracture has been questioned, as fractures often originate from margins of restoration. This study determined fracture resistance of highly translucent monolithic zirconia crowns with different margin designs in terms of marginal thickness and collar height. Materials and Methods. Zirconia blanks (Ceramill® Zolid HT+) were selected for the fabrication of zirconia crowns according to different designs, including varying margin thicknesses (light chamfer, CL; heavy chamfer, CH) and collar heights (no collar, NC; low collar, LC; high collar, HC), which resulted in CLNC, CLLC, CLHC, CHNC, CHLC, and CHHC groups (15 crowns each). The crowns were seated on a metal die and loaded vertically through round end punch (θ = 10 mm), contacting with inclined planes of cusp in a testing machine with crosshead speed of 0.2 mm/min until fracture. Videos with a rate of 50 frames/second were used to record fracture. Fracture load (N) and durable period (s) were compared for significant differences using ANOVA and Bonferroni test (α = 0.05). Results. The mean ± sd of fracture load (N) and durable time (s) were 3211 ± 778 and 212 ± 47 for CLNC; 3041 ± 1370 and 188 ± 53 for CLLC; 2913 ± 828 and 192 ± 27 for CLHC; 4226 ± 905 and 245 ± 35 for CHNC; 4486 ± 807 and 228 ± 29 for CHLC; and 4376 ± 1043 and 227 ± 37 for CHHC. This indicated that marginal thickness had a significant influence on load-bearing capacity and durable time (p<0.05). No significant impact of collar height was shown, either on load-bearing capacity or durable time (p>0.05). No interaction between two factors was presented (p>0.05). Conclusions. Heavy chamfer margin provided stronger zirconia crown than light chamfer, but both were capable of withstanding fracture load higher than maximum masticatory force. Neither presence nor absence of collar indicated any impact on strength. Fabrication of zirconia crowns with either heavy or light chamfer margin and either presence or absence of collar, with the consideration of emergence profile, should be considered.

In mammals, two TATA-less bidirectional promoters regulate expression of the divergently transcribed dihydrofolate reductase (dhfr) and rep3 genes. In CHOC 400 cells, dhfr mRNA levels increase about fourfold during the G1-to-S phase transition of the cell cycle, whereas the levels of rep3 transcripts vary less than twofold during this time. To assess the role of DNA-binding proteins in transcriptional regulation of the dhfr and rep3 genes, the major and minor dhfr-rep3 promoter regions were analyzed by high-resolution genomic footprinting during the cell cycle. At the major dhfr promoter, prominent DNase I footprints over four upstream Sp1 binding sites did not vary throughout G1 and entry into the S phase. Genomic footprinting revealed that a protein is constitutively bound to the overlapping E2F sites throughout the G1-to-S phase transition, an interaction that is most evident on the transcribed template strand. On the nontranscribed strand, multiple changes in the DNase I cleavage pattern are observed during transit through G1 and entry into the S phase. By using gel mobility shift assays and a series of sequence-specific probes, two different species of E2F were shown to interact with the dhfr promoter during the cell cycle. The DNA binding activity of one E2F species, which preferentially recognizes the sequence TTTGGCGC, did not vary significantly during the cell cycle. The DNA binding activity of the second E2F species, which preferentially recognizes the sequence TTTCGCGC, increased during the G1-to-S phase transition. Together, these results indicate that Sp1 and the species of E2F that binds TTTGGCGC participate in the formation of a basal transcription complex, while the species of E2F that binds TTTCGCGC regulates dhfr gene expression during the G1-to-S phase transition. At the minor promoter, DNase I footprints at a consensus c-Myc binding site and three Sp1 binding sites showed little variation during the G1-to-S phase transition. In addition to protein binding at sequences known to be involved in the regulation of transcription, genomic footprinting of the entire promoter region also showed that a protein factor is constitutively bound to the first intron of the rep3 gene.

La présente étude s’intéresse à la fois au développement de la couche limite turbulente en écoulement supersonique sur une plaque plane, à la distance requise pour qu’elle envahisse toute la section de la plaque, ainsi qu’aux effets de la taille de la zone d'interaction sur le développement de la même couche limite. L'augmentation de la force de l'interaction est une augmentation de la taille des zones d'interactions conduisant à la formation d'une bulle de re-circulation qui est une zone des pertes de charge importantes. Pour cette raison, on peut diminuer, puis augmenter le nombre de Mach pour voir une réflexion de Mach qui explique l'impact de l'onde de choc incident forte avec la couche limite. L'augmentation et la diminution du nombre de Mach a causé l'apparition d’une transition dans les interactions réflexion régulière (RR), réflexion de Mach (MR), qui est représenté par les iso contours des champs de la masse volumique. Ces études sont bien en accord par rapport à l'essai qui a été présenté par Délery et al. (2009). On a donné une autre contribution et des investigations du phénomène de l'interaction onde de choc/couche limite turbulente. Le modèle utilisé dans cette étude est le modèle kwSST, il est considéré comme étant le plus approprié pour ce genre de problème, avec un traitement spécial de la zone proche de la paroi. Des simulations numériques ont été faites à l’aide de logiciel FLUENT.

Experimental and numerical study of transversal microjets injection influence on the supersonic underexpanded jet flow structure has been performed. Data of measurements and calculation have acceptable agreement. Interaction of microjets with main supersonic jet sets to a decrease of an initial gasdynamic region. Microjets lead to a longitudinal streamwise vortices generation and a mushroom-like flow structures create on an external jet mixing layer. Dissipation of longitudinal streamwise vortices was observed at the second jet cell. Complex gasdynamic flow structure of the supersonic underexpanded jet interacting with supersonic microjets has been studied for the first time. This structure contains system of complex chock waves and expansion waves spreading from the position of the impact microjets/main jet localization place. Future of interaction process a chock-wave structure of main jet with additional shock waves has been studied

Résumé Le problème des rapports entre l'épargne et l'investissement est un vieux débat dans le monde des économistes, mais son intensité est variable et obéit aux rythmes cycliques qui gouvernent la nature des préoccupations macro-économiques. Ce débat est revenu sur le devant de la scène depuis quelques années, parallèlement au redémarrage de la croissance intervenu dans les grands pays industrialisés sur la seconde moitié des années quatre-vingt. On pourrait donc voir dans ce remake un événement de nature principalement conjoncturelle : les besoins et désirs d'investissement associés à la phase d'expansion de la seconde moitié de la décennie quatre-vingt ont fini, classiquement, par trop tirer sur la corde des financements et fait ainsi ressentir un déficit d'épargne. De plus, à la fin de cette période d'expansion, le choc spécifique des changements affectant les pays est-européens et révélant brutalement l'ampleur de leurs besoins de financement, aurait aggravé ces tensions. On notera cependant d'emblée que la polarisation récente du débat macroéconomique sur le thème de la pénurie d'épargne est, au premier degré, quelque peu paradoxal, au terme d'une décennie qui, par réaction aux excès antérieurs de l'économie d'endettement administrée, prétendait rétablir des incitations saines et puissantes à une épargne volontaire et stable par le retour en force et l'approfondissement des marchés financiers. Si le thème de la pénurie d'épargne a une certaine vérité, il témoigne en ce cas d'une mise en échec des réformes financières intervenues au cours des années quatfë-vihgtV On peut aborder la question des rapports de l'épargne et de l'investissement comme un problème de nature conjoncturelle, lié au déroulement habituel du cycle économique de moyenne période, et commenter sous cet angle les indicateurs conjoncturels disponibles. Cette manière de voir est cependant insuffisante pour rendre compte des évolutions intervenues durant les années quatre-vingt : ces évolutions relèvent aussi de « changements de régime » qui infléchissent ou modifient les trends apparents et ces changements interfèrent avec le cycle conjoncturel. Pour saisir l'originalité de la configuration macro-économique présente, il faut bien évidemment prendre en compte ces interférences et expliciter leur contenu. Interpréter les enchaînements des décennies quatre-vingt et quatre-vingt-dix comme le moment d'une interaction forte entre cycles de différente périodicité, courts et longs, parait alors une hypothèse de travail stimulante. L 'article est articulé en deux parties. La première s 'attache à fournir un schéma général des interactions entre épargne et investissement au cours du cycle conjoncturel. Elle examine les données françaises sur l'ensemble de l'après-guerre, situées dans leur contexte international, à la lumière de ce schéma. La seconde partie se concentre sur le rôle joué par les facteurs patrimoniaux et financiers dans les dynamiques cycliques à l'œuvre durant les décennies quatre-vingt et quatre-vingt-dix.

Abstract Building upon the extended notion of conceptual history as a diachronic study of conceptual interactions, the article begins with deconstructing the paradoxical semantic core of incomparability statements that, it is claimed, endows them with a capacity of stabilizing social semantics. By declaring certain foundational values—positive (Shoah) or negative (God)—“incomparable” and thus immune to the challenges of cross-evaluation, the users of discourse uphold the boundaries of civilized society. On a smaller scale, this exclusion of competitive valuation is undergirded by the ascription of “incomparability” to the small pool of political and cultural figures, literary artifacts, social events, and representative allegories. The conclusion outlines the social contingency of conventions regulating the ascriptions of “incomparability” against the backdrop of their discursive stability across genres, epochs, and languages.

To disperse fatty acids in aqueous solution, choline, a quaternary ammonium ion, has been used recently. So far, only the self-assembly of myristic acid (MA) in the presence of choline hydroxide as a function of the molar ratio has been investigated, and, thus, the current understanding of these fatty acid systems is still limited. We investigated the self-assembly of palmitic acid (PA) in the presence of choline hydroxide (ChOH) as a function of the molar ratio (R) between ChOH and PA. The self-assemblies were characterized by phase contrast microscopy, cryo-TEM, small-angle X-ray scattering, and 2H NMR. The ionization state of PA was determined by pH, conductivity, and FT-IR measurements. With increase in R, various self-assembled structures, including vesicles, lamellar phase, rigid membranes (large sheets, tubules, cones, and polyhedrals), and micelles, form in the PA/ChOH system, different from those of the MA/ChOH system. The change in R induces pH variation and, consequently, a change in the PA ionization state, which, in turn, regulates the molecular interactions, including hydrogen bonding and electrostatic interaction, leading to various self-assemblies. Temperature is an important factor used to tune the self-assembly transitions. The fatty acid choline systems studied here potentially may be applicable in medicine, chemical engineering, and biotechnology.

ObjectiveTo identify XK pathologic mutations in 6 patients with suspected McLeod syndrome (MLS) and a possible interaction between the chorea-acanthocytosis (ChAc)- and MLS-responsible proteins: chorein and XK protein.MethodsErythrocyte membrane proteins from patients with suspected MLS and patients with ChAc, ChAc mutant carriers, and normal controls were analyzed by XK and chorein immunoblotting. We performed mutation analysis and XK immunoblotting to molecularly diagnose the patients with suspected MLS. Lysates of cultured cells were co-immunoprecipitated with anti-XK and anti-chorein antibodies.ResultsAll suspected MLS cases were molecularly diagnosed with MLS, and novel mutations were identified. The average onset age was 46.8 ± 8 years, which was older than that of the patients with ChAc. The immunoblot analysis revealed remarkably reduced chorein immunoreactivity in all patients with MLS. The immunoprecipitation analysis indicated a direct or indirect chorein-XK interaction.ConclusionsIn this study, XK pathogenic mutations were identified in all 6 MLS cases, including novel mutations. Chorein immunoreactions were significantly reduced in MLS erythrocyte membranes. In addition, we demonstrated a possible interaction between the chorein and XK protein via molecular analysis. The reduction in chorein expression is similar to that between Kell antigens and XK protein, although the chorein-XK interaction is a possibly noncovalent binding unlike the covalent Kell-XK complex. Our results suggest that reduced chorein levels following lack of XK protein are possibly associated with molecular pathogenesis in MLS.

Forensic engineering field surveys of recent tsunamis (Saatcioglu et al. 2005, Chock et al. 2013) highlighted the importance of scour-related damage to structures located in coastal communities. To date, only a limited number of studies have investigated the interaction of extreme hydrodynamic flows and groups of structures, and none have studied the scour around multiple structures interacting with each other. One field example discussed by Yeh et al. (2013) documented flow concentration in between two tsunami-resistant buildings, leading to a deep scour hole between them and infrastructure failures onshore of the gap between the two buildings. This field example shows that multiple buildings, often crammed, lead to complex flow-structure interactions, leading to flow and scour either amplification or reduction depending on the relative position of the buildings. Nouri et al. (2010) and Thomas et al. (2015) investigated the flow velocity amplification caused by structures proximity, which concentrated the flow onto a downstream monitored structure. Their results informed the ASCE7 Ch.6 “Tsunami Loads and Effect” standard on flow velocity amplification caused by nearby structures. However, in this standard, there is currently no link between flow velocity amplification factors and their effects on scour around structures.

Two greenhouse studies were conducted to examine effects of nitrogen source on primary and secondary metabolism of pac choi (Brassica rapa L. subsp. chinensis cv. Mei Qing Choi) and diamondback moth (Plutella xylostella L.) consumption, development, survival, and body weight. Applications of a liquid organic source of nitrogen (fish hydrolysate fertilizer) were compared with a conventional fertilizer to determine whether nitrogen source directly impacts pac choi chemistry (elemental composition and phenolics) and biomass and indirectly affects diamondback moth fitness parameters. There was no significant effect of fertility treatment on pac choi chemistry or biomass with the exception of percent leaf phosphorus, which was significantly higher in the conventional fertility treatment, and p-coumarin, which was significantly higher in the organic fertility treatment. Diamondback moth also affected plant chemistry. Both calcium (Ca) and magnesium (Mg) were significantly higher in plants infested with larvae compared with uninfested plants. Fertilizer affected diamondback moth fitness with percent survival and cohort development significantly reduced on pac choi associated with the organic fertilizer. However, pac choi receiving the organic treatment was similar in regard to primary nutrients and secondary compounds compared with plants that received a conventional fertilizer.

Utilizing state-dependent projection operators and the Kang-Choi reduction identities, we derive the linear, first, and second-order nonlinear optical conductivities for an electron system interacting with phonons. The lineshape functions included in the conductivity tensors satisfy “the population criterion” saying that the Fermi distribution functions for electrons and Planck distribution functions for phonons should be combined in multiplicative forms. The results also contain energy denominator factors enforcing the energy conservation as well as interaction factors describing electron-phonon interaction properly. Therefore, the phonon absorption and emission processes as well as photon absorption and emission processes in all electron transition processes can be presented in an organized manner and the results can be represented in diagrams that can model the quantum dynamics of electrons in a solid.

Abstract. We present data on the distribution and thermophysical properties of snow collected sporadically over 4 decades along with recent data of ground surface temperature from Mont Jacques-Cartier (1268 m a.s.l.), the highest summit in the Appalachians of south-eastern Canada. We demonstrate that the occurrence of contemporary permafrost is necessarily associated with a very thin and wind-packed winter snow cover which brings local azonal topo-climatic conditions on the dome-shaped summit. The aims of this study were (i) to understand the snow distribution pattern and snow thermophysical properties on the Mont Jacques-Cartier summit and (ii) to investigate the impact of snow on the spatial distribution of the ground surface temperature (GST) using temperature sensors deployed over the summit. Results showed that above the local treeline, the summit is characterized by a snow cover typically less than 30 cm thick which is explained by the strong westerly winds interacting with the local surface roughness created by the physiography and surficial geomorphology of the site. The snowpack structure is fairly similar to that observed on windy Arctic tundra with a top dense wind slab (300 to 450 kg m−3) of high thermal conductivity, which facilitates heat transfer between the ground surface and the atmosphere. The mean annual ground surface temperature (MAGST) below this thin and wind-packed snow cover was about −1 °C in 2013 and 2014, for the higher, exposed, blockfield-covered sector of the summit characterized by a sporadic herbaceous cover. In contrast, for the gentle slopes covered with stunted spruce (krummholz), and for the steep leeward slope to the south-east of the summit, the MAGST was around 3 °C in 2013 and 2014. The study concludes that the permafrost on Mont Jacques-Cartier, most widely in the Chic-Choc Mountains and by extension in the southern highest summits of the Appalachians, is therefore likely limited to the barren wind-exposed surface of the summit where the low air temperature, the thin snowpack and the wind action bring local cold surface conditions favourable to permafrost development.

In order to examine the mixing properties of glycerol–water and diglycerol–water solutions, these solutions were measured using attenuated total reflection infrared spectroscopy. The absorbance spectra corrected for 1 µm thickness were subtracted by pure polyols for obtaining water spectra, and by pure water for polyol spectra. Both asymmetric and symmetric CH2 stretching vibration bands (around 2940, 2885 cm−1) shifted about 10 cm−1 to lower wavenumber side (redshifts) with increasing polyol concentrations, especially at higher concentrations. Redshifts of C–O–H rocking bands (around 1335 cm−1) with increasing polyol concentrations are slightly larger for diglycerol–water (10 > 6 cm−1) than glycerol–water solutions. C–O stretching bands of CHOH groups (1125 and 1112 cm−1) shift slightly but in opposite sides for glycerol and diglycerol at highest polyol concentrations (90–100 wt%). These shifts of CH2 stretching, COH rocking, and CO stretching of CHOH at higher polyol concentrations suggest interactions of outer CH2 with inner CHOH groups of surrounding polyols. The normalized band area changes with polyol concentrations could be fitted by quadratic polynomials possibly due to mixtures of different interactions between water–water, polyol–water, and polyol–polyol molecules. The OH stretching band for diglycerol 90 wt% shows three humps indicating at least three OH components: long, medium, and short H bond water molecules. Short H bond water molecules are the major component possibly between inner CHOH and outer side CH2OH groups, while the long H component might loosely bind to outer CH2OH groups.

The purpose of this paper is to understand how Clara’s prophetic ability and cosmic interest in the text translated by Choi Seung-ja, Isabel Allende’s first full-length novel, The House of the Spirits(1991), appeared in Choi Seung-ja’s 5th collection Lovers(1999) and 6th collection Lonely Far Away(2010) from the perspective of receptive aesthetics. As Choi Seung-ja, not only did she read The House of the Spirits as a reader and read it again by translating it, but she used it for her poetry creation. As such, poet Choi Seung-ja showed the interaction between the receptive aesthetics as a reader and the aesthetics of influence on her own beginning. Her ‘prediction and cosmic imagination’ has expanded the realm of Korean contemporary poetry indefinitely, and this attempt of ‘new method of reading’ is the core of this article.

TFIIIA is a zinc-finger transcription factor that is involved in post-transcriptional regulation during development. Here, the BcTFIIIA gene was isolated from pak choi. Sequence analysis showed that BcTFIIIA encodes 383 amino acids (aa) with an open reading frame (ORF) of 1152 base pairs (bp). We investigated the subcellular location of BcTFIIIA and found the localized protein in the nucleus. BcTFIIIA was suppressed when the pak choi was infected by the turnip mosaic virus (TuMV). The BcTFIIIA mRNA expression level in a resistant variety was higher than that in a sensitive variety, as determined by qRT-PCR analysis. Yeast two hybrid (Y2H) assay and bimolecular fluorescence complementation (BiFC) suggested that BcTFIIIA interacts with TuMV CP and VPg in vivo, respectively, and in vitro. A virus-induced gene silencing (VIGS) experiment showed that the silencing of BcTFIIIA gene expression in pak choi promoted the accumulation of TuMV. These results suggest that BcTFIIIA negatively regulates viral infection through the interaction with TuMV CP and VPg.

Abstract Many GWAS loci occur in non-coding regions and often overlap with gene regulatory elements such as distant enhancers, making functional interpretation and target gene discovery challenging. The 3D chromatin organization brings enhancers in spatial proximity with a promoter to regulate target gene expression. Therefore, to map chromatin interactions between GWAS variants and target gene promoters we performed region-focused chromatin conformation capture assay (Capture-C) in primary human melanocytes. We baited the entire region of association for all 68 independent signals from the recent melanoma GWAS, and Capture-C interactions were called using CHiCAGO tool. Integrative analysis of Capture-C interactions with melanocyte- and melanoma-specific ATAC-sequencing, massive-parallel reporter assay (MPRA), ROADMAP chromatin imputed state model, and gene expression datasets helped prioritize target genes for functional follow-up. Capture-C assays identified physical chromatin interactions between fine-mapped risk variants and candidate causal gene (CCG) promoters at 90% of the GWAS loci; For 84% of the 68 loci, we observed at least one variant-to-gene promoter interaction longer than 100 kb, and for 20% of loci we found interactions beyond 1 Mb. For 76% of the 68 loci, the CCG-interacting variant was in annotated melanocyte or melanoma enhancer regions consistent with CCG regulation via an enhancer-promoter interaction. We observed at least one CCG-interacting variant in 63% and 51% of the 68 loci with distinct allele specific transcriptional activity in melanocyte and melanoma MPRA datasets respectively. A majority of the loci (60%) harbored CCG-linked risk variants in accessible chromatin regions in melanocytes and melanoma. Pathway enrichment analyses of Capture-C-nominated CCGs identified embryonic development, aryl hydrocarbon receptor signaling, and DNA repair pathways. Notably, we observed chromatin interactions between risk variants located near the 3’ of Rap Guanine Nucleotide Exchange Factor 1 (RAPGEF1) to the RAPGEF1 and UCK1 promoter regions. The risk allele of the lead variant (rs3780269) at this locus was associated with higher RAPGEF1 mRNA expression and was not associated with UCK1 expression in melanocytes. We performed a CRISPR knockout proliferation screen in immortalized melanocytes and identified RAPGEF1 as an essential gene for melanocyte growth or survival. Further, we validated the results of this screen by overexpressing RAPGEF1 in immortalized melanocytes and found that it leads to increased cellular growth. We are now characterizing its function in melanoma tumor incidence and progression in a zebrafish model. In summary, mapping GWAS loci chromatin interactions to target gene promoters and integrative analysis using cell-type specific datasets identified RAPGEF1 as a melanoma susceptibility gene. Chorea-acanthocytosis (ChAc), an autosomal recessive disorder, is associated with cognitive and behavioral abnormalities. Previous studies were focused around exploring the functional annotation of VPS13A gene in ChAc, whereas the genetic labyrinth underlying this disease and plausible drug targets were underexplored. In the present study, we have identified the pivotal genes and molecular pathways implicated in ChAc using comprehensive bioinformatics analysis. In our analysis we found 27 distinct genes in Homo sapiens linked to ChAc, out of which 15 were selected as candidate genes for enrichment analysis based on their Gene Ontology (GO) annotations and involvement in relevant molecular pathways. By constructing a Protein-Protein Interaction (PPI) network consisting of 26 nodes and 62 edges, we identified two gene modules. Subsequently, using the MCODE algorithm, we identified 6 hub genes—ATN1, JPH3, TBP, VPS13A, DMD, and HTT—as core candidates. These hub genes are primarily associated with processes such as neuron development and differentiation, the CAMKK-AMPK signaling cascade, ion transmembrane transport systems, and protein localization. Furthermore, using drug gene databank we identified 23 FDA-approved drugs that possess the propensity to target 3 out of the 6 identified hub genes. We believe that our findings could open promising avenues for potential therapeutic interventions in ChAc.

We investigated interactions between Typhoon Choi-wan (2009) and the Kuroshio Extension system with a coupled atmosphere-wave-ocean model in 14 numerical simulations performed with initial conditions obtained from daily oceanic reanalysis data for the northwestern Pacific Ocean from September 12 to September 25, 2009. Preexisting oceanic conditions affected the simulated central pressure and the inner-core axisymmetric structure of the simulated typhoon differently during the intensification, mature, and decaying phases, but they had little impact on the simulated track. Among the simulations, the simulated central pressure range during the mature phase was ~10 hPa. Simulated central pressure was highly correlated with the axisymmetric mean horizontal specific humidity flux between radii of 50 and 150 km and altitudes of 20 to ~2000 m, suggesting that this flux has potential as a new metric for predicting tropical cyclone intensity. Variations in preexisting oceanic conditions in the Kuroshio Extension region on the simulated atmospheric and oceanic horizontal fields affected the typhoon and a nearby stationary front differently. Around the stationary front, the impact on hourly precipitation was closely related to that on surface wind speed, whereas the impact on surface temperature was greatly affected by that on sea surface temperature through the latent heat flux.

A reaction between copper(II) nitrate and trans-1,4-cyclohexanedicarboxylic acid (H2chdc) carried out under hydrothermal conditions led to a new metal-organic coordination polymer [Cu2(Hchdc)2(chdc)]n. According to single-crystal XRD data, the compound is based on bi-nuclear paddlewheel-type carboxylate blocks that are joined with polymeric chains due to the (μ3-κ1:κ2) coordination of carboxylate groups. The chains are interconnected by chdc2− bridging ligands into layers containing free COOH groups of terminal Hchdc−. The neighboring layers adopt a RCOOH···OOCR hydrogen bond-assisted arrangement into a dense-packed structure. Magnetization measurements showed the presence of a strong antiferromagnetic exchange interaction (J/kB = −495 K) inside the bi-nuclear blocks. At the same time, no significant interaction was found between the {-Cu2(OOCR)4-} units in spite of their polymeric in-chain packing. Patterns of magnetic behavior of [Cu2(Hchdc)2(chdc)]n were thoroughly analyzed and explained from a structural point of view.

Language plays an important role as a mean of communication for cognitive development and social interaction. At inclusive early childhood education environments, Children with Autism Spectrum Disorder (children with ASD) must be familiar with and participate in all classroom activities. Because the defects of most children with ASD are limited in language, their cognitive and social interaction, participation in learning and integration are significantly affected. This research was conducted on the current situation of organizing shape activities for 3 or 4-years-old children with ASD at inclusive kindergartens. And the result of this research can contribute in helping children with ASD have closer access to the preschool education program as well as help teachers can better promote the effectiveness of special education interventions in inclusive kindergartens. Preliminary results on factors affecting shape activities and the correlation between educational activities which contribute to confirming the feasibility of organizing shape activities to develop language for 3 or 4-years-old children with ASD in the inclusive early childhood environments.

The new coordination polymer, [Pb(L)(trans-1,4-chdc)] 1 (L = 1-(1H-imidazo[4,5-f][1,10]phenanthrolin-2-yl)- naphthalen-2-ol and trans-1,4-H2chdc = trans-1,4-cyclohexanedicarboxylic acid), has been hydrothermally synthesized and characterized by elemental analysis, IR spectroscopy and single-crystal X-ray diffraction. Crystal data: C31H24N4O5Pb, monoclinic, space group P21/c, a = 13.579(2), b = 9.7831(15), c = 19.958(3) Å, β = 105.058(2)°, V = 2560.3(7) Å3, Z = 4. The Pb(II) atoms are bridged by the flexible trans-1,4-chdc ligands to form a zigzag chain. The ligands L are located on both sides of the zigzag chains, where π-π interactions among neighboring zigzag chains give rise to a two-dimensional supramolecular network. In addition, O-H···O and N-H···O hydrogen bonding interactions further stabilize the supramolecular network structure of 1.

Abstract A new Cd(ii) coordination polymers, formulated as [Cd(1,4-chdc)(L)] n (1) (L = 1-(1H-imidazo[4,5-f][1,10]phenanthrolin-2-yl)naphthalen-2-ol ligands and 1,4-H2chdc = 1,4-cyclohexanedicarboxylate acid), were successfully synthesized by using L ligand and 1,4-H2chdc acid under hydrothermal conditions and characterized by single crystal X-ray elemental analyses, diffraction, and IR spectroscopy. Each Cd(ii) atom is a distorted pentagonal bipyramidal geometry connecting the deprotonated 1,4-chdc anion to form a 2D layer, and a 2D layer is formed into a 3D supramolecular structure by the π–π stacking interactions between neighboring layers.

RésuméNous introduisons le concept d'empreinte carbone dans un modèle d'offre globale et demande globale avec formation imparfaitement concurrentielle des prix et salaires et en examinons les propriétés de l'équilibre en présence d'une politique climatique. Nous étudions deux instruments possibles de cette politique, une taxe carbone ou un quota de permis de pollution. Nous montrons qu'à court terme, la politique climatique (ou son durcissement) constitue à la fois un choc d'offre globale négatif et, ceteris paribus, un choc de demande globale positif. Elle provoque donc des effets inflationnistes mais a un impact ambigu sur l'activité économique, l'emploi et le chômage. Ce n'est que dans une économie avec des ridigités nominales suffisantes que la politique climatique stimulera -sous certaines conditions- l'activité à court terme. Dans tous les cas de figure, elle pèsera négativement sur les salaires réels.Nous étudions encore les interactions entre la politique climatique et les politiques macroéconomiques traditionnelles de demande (stimulus budgétaire ou monétaire) et d'offre (baisse des cotisations sociales). Les effets multiplicateurs de ces politiques sont influencés par l'existence d'une politique climatique et diffèrent selon l'intrument choisi (taxe ou permis).Nous montrons les conditions sous lesquelles une réforme combinant durcissement de la politique climatique et baisse des cotisations sociales sur le travail peut atteindre le double objectif de réduire l'empreinte carbone de l'économie et le chômage, sans pénaliser les salaires réels des travailleurs. Une telle politique a toutefois des effets incertains sur le solde des finances publiques.

Abstract Mental or computer simulation of cause and effect of certain behaviors is a recognized approach to changing one’s attitude or triggering an action. Meanwhile, psychology research results suggest that frequency of simulation may affect the corresponding persuasiveness. This paper argues that with always-on sensing and data-driven visualization technologies, interactive tangible systems can be designed to simulate hypothetical outcomes of real-life behaviors in everyday contexts, which repeatedly stimulate users’ imagination of behavioral consequences and thereby behavioral intentions. To investigate the effect, a working prototype of Incingarette, including a smart ashtray in connection with a digital picture frame, was built. When the ashtray is used for smoking, the digital picture is incrementally covered by virtual dust. Field trials involved participants in five daily smoking sessions. Post-session surveys show increasingly stronger perceived causality between smoking and the simulated outcomes, increasingly more vivid mental imagery of consequences, and increasingly intense intention to reduce smoking. Results suggest that repeatedly presenting simulated outcomes cognitively linked to real-life behaviors can increase behavioral intentions.

Chitin nanocrystals (ChNCs) and cellulose nanocrystals (CNCs) have been recently used to stabilize emulsions; however, they generally require significant amounts of salt, limiting their applicability in food products. In this study, we developed nanoconjugates by mixing positively charged ChNCs and negatively charged CNCs at various ChNC:CNC mass ratios (2:1, 1:1, and 1:2), and utilized them in stabilizing soybean oil–water Pickering emulsions with minimal use of NaCl salt (20 mM) and nanoparticle (NP) concentrations below 1 wt%. The nanoconjugates stabilized the emulsions better than individual CNC or ChNC in terms of a reduced drop growth and less creaming. Oppositely charged CNC and ChNC neutralized each other when their mass ratio was 1:1, leading to significant flocculation in the absence of salt at pH 6. Raman spectroscopy provided evidence for electrostatic interactions between the ChNCs and CNCs, and generated maps suggesting an assembly of ChNC bundles of micron-scale lengths intercalated by similar-size areas predominantly composed of CNC. The previous measurements, in combination with contact angles on nanoparticle films, suggested that the conjugates preferentially exposed the hydrophobic crystalline planes of CNCs and ChNCs at a 1:1 mass ratio, which was also the best ratio at stabilizing soybean oil–water Pickering emulsions.

CHO2 is a mammalian minus-end-directed kinesin-like motor protein present in interphase centrosomes/nuclei and mitotic spindle fibers/poles. Expression of HA- or GFP-tagged subfragments in transfected CHO cells revealed the presence of the nuclear localization site at the N-terminal tail. This domain becomes associated with spindle fibers during mitosis, indicating that the tail is capable of interaction with microtubules in vivo. While the central stalk diffusely distributes in the entire cytoplasm of cells, the motor domain co-localizes with microtubules throughout the cell cycle, which is eliminated by mutation of the ATP-binding consensus motif from GKT to AAA. Overexpression of the full-length CHO2 causes mitotic arrest and spindle abnormality. The effect of protein expression was first seen around the polar region where microtubule tended to be bundled together. A higher level of protein expression induces more elongated spindles which eventually become disorganized by loosing the structural integrity between microtubule bundles. Live cell observation demonstrated that GFP-labeled microtubule bundles underwent continuous changes in their relative position to one another through repeated attachment and detachment at one end; this results in the formation of irregular number of microtubule focal points in mitotic arrested cells. Thus the primary action of CHO2 appears to cross-link microtubules and move toward the minus-end direction to maintain association of the microtubule end at the pole. In contrast to the full-length of CHO2, overexpression of neither truncated nor mutant polypeptides resulted in significant effects on mitosis and mitotic spindles, suggesting that the function of CHO2 in mammalian cells may be redundant with other motor molecules during cell division.

The CHO1 antigen is a mitosis-specific kinesin-like motor located at the interzonal region of the spindle. The human cDNA coding for the antigen contains a domain with sequence similarity to the motor domain of kinesin-like protein (Nislow et al., Nature 359, 543, 1992). Here we cloned cDNAs encoding the CHO1 antigen by immunoscreening of a CHO Uni-Zap expression library, the same species in which the original monoclonal antibody was raised. cDNAs of CHO cells encode a 953 amino acid polypeptide with a calculated molecular mass of 109 kDa. The N-terminal 73% of the antigen was 87% identical to the human clone, whereas the remaining 27% of the coding region showed only 48% homology. Insect Sf9 cells infected with baculovirus containing the full-length insert produced 105 and 95 kDa polypeptides, the same doublet identified as the original antigen in CHO cells. Truncated polypeptides corresponding to the N-terminal motor and C-terminal tail produced a 56 and 54 kDa polypeptide in Sf9 cells, respectively. Full and N-terminal proteins co-sedimented with, and caused bundling of, brain microtubules in vitro, whereas the C-terminal polypeptide did not. Cells expressing the N terminus formed one or more cytoplasmic processes. Immunofluorescence as well as electron microscopic observations revealed the presence of thick bundles of microtubules, which were closely packed, forming a marginal ring just beneath the cell membrane and a core in the processes. The diffusion coefficient and sedimentation coefficient were determined for the native CHO1 antigen by gel filtration and sucrose density gradient centrifugation, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Numerous studies have been conducted to investigate the genomic characterization of bZIP genes and their involvement in the cellular response to endoplasmic reticulum (ER) stress. These studies have provided valuable insights into the coordinated cellular response to ER stress, which is mediated by bZIP transcription factors (TFs). However, a comprehensive and systematic investigations regarding the role of bZIP genes and their involvement in ER stress response in pak choi is currently lacking in the existing literature. To address this knowledge gap, the current study was initiated to elucidate the genomic characteristics of bZIP genes, gain insight into their expression patterns during ER stress in pak choi, and investigate the protein-to-protein interaction of bZIP genes with the ER chaperone BiP. In total, 112 members of the BcbZIP genes were identified through a comprehensive genome-wide analysis. Based on an analysis of sequence similarity, gene structure, conserved domains, and responsive motifs, the identified BcbZIP genes were categorized into 10 distinct subfamilies through phylogenetic analysis. Chromosomal location and duplication events provided insight into their genomic context and evolutionary history. Divergence analysis estimated their evolutionary history with a predicted divergence time ranging from 0.73 to 80.71 million years ago (MYA). Promoter regions of the BcbZIP genes were discovered to exhibit a wide variety of cis-elements, including light, hormone, and stress-responsive elements. GO enrichment analysis further confirmed their roles in the ER unfolded protein response (UPR), while co-expression network analysis showed a strong relationship of BcbZIP genes with ER-stress-responsive genes. Moreover, gene expression profiles and protein–protein interaction with ER chaperone BiP further confirmed their roles and capacity to respond to ER stress in pak choi.

This paper is concerned with the effect of a weak spanwise-variable mean-flow distortion on the growth of oblique instability waves in a Blasius boundary layer. The streamwise component of the distortion velocity initially grows linearly with increasing streamwise distance, reaches a maximum, and eventually decays through the action of viscosity. This decay occurs slowly and allows the distortion to destabilize the Blasius flow over a relatively large streamwise region. It is shown that even relatively weak distortions can cause certain oblique Rayleigh instability waves to grow much faster than the usual two-dimensional Tollmien–Schlichting waves that would be the dominant instability modes in the absence of the distortion. The oblique instability waves can then become large enough to interact nonlinearly within a common critical layer. It is shown that the common amplitude of the interacting oblique waves is governed by the amplitude evolution equation derived in Goldstein & Choi (1989). The implications of these results for Klebanoff-type transition are discussed.

This paper explores the relationship between content marketing and consumer brand loyalty to the ‘New-China-Chic’ brand. It constructs a structural equation model, Through questionnaires distributed to the fan groups of various brands and offline stores, results show that the four different dimensions of content marketing strategy, namely the culture, entertainment, emotion and interaction, are the main factors affecting consumers' loyalty to the ‘New-China-Chic’ brand. Among them, emotional content and cultural content??= 0.304??= 0.209) have a strong impact on brand loyalty. Brand identity also plays a strong intermediary role in the relationship between the four different dimensions of content marketing and brand loyalty. Moreover, the data shows that consumer ethnocentrism has a significant regulatory effect on brand identity and brand loyalty in cultural content and emotional content, while interactive content and entertainment content have a weak impact.
