Data recovery investigations conducted by Louis Berger & Associates, Inc. at the U.S. Coast Guard Support Center, Governors Island, New York, recovered evidence of a heretofore undocumented late-18th to early-19th century cemetery outside of Castle Williams. Subsurface investigations were conducted to address questions related to the nature, context, and extent of human remains encountered during the excavation of an electrical utility trench in the northwest section of the island. A total of six human burials was recovered from within burial shafts manifest through soil color differences. On the basis of the archaeological, osteological, and paleopathological analysis, we concluded that the individuals interred outside of Castle Williams represent late-18th or early-19th century Colonial-born, Caucasoid males probably stationed on the island as enlisted military personnel.

This paper presents the history and significance of Jagielnica Castle (more specifically, it is located in the village of Nagórzanka, near Jagielnica) mainly during the period when it served the Polish tobacco industry. The early days of the stronghold, which was built on a hill, date back to the 16th century. Throughout the centuries the fortunes of the castle changed. After restoration in the 18th century, it became the main residence of the Lanckoroński family. In 1817 the castle became a property of the Austrian government. A factory and a warehouse for tobacco and tobacco products were established there. During World War I the castle complex was destroyed. After war it was taken over by the Polish Tobacco Monopoly. After the old buildings were rebuilt and new ones were constructed, it became the seat of the Tobacco Cultivation Plant PTM in Jagielnica. After World War II the tobacco plant still operated, but it was in the Ukrainian Soviet Socialist Republic. In the early 21st century it was privatized. Today there are efforts to establish a tourist and leisure centre in the castle, taking into account the protection of cultural heritage

To sustain its exceptional financial performance, Tesla should promptly in-troduce new products to meet growing demand. This study emphasizes the need for Tesla to refine accounting policies to ensure transparency and compliance. Additionally, a careful analysis of financial indicators will enable the company to identify areas for improvement and make informed strategic decisions. Looking ahead, the paper underscores the im-portance of Tesla's future profitability, urging the company to capitalize on market trends and diversify its product offerings. In conclusion, this paper provides a comprehensive eval-uation of Tesla's competitive ability in the new energy industry. To maintain its leadership position, Tesla must adapt to the evolving market dynamics by refining accounting practices, monitoring financial indicators, and introducing new products to meet consumer demand.

Phytoremediation supported by bioaugmentation is a promising method considered for cleaning up areas polluted with petroleum hydrocarbons. In this study, phytoremediation was carried out using Echinacea purpurea as a phytoremediant on two types of soil: Soil DW—aged soil taken from an excavation pit, Soil OS—soil taken from an oil spill area. The tests for each soil were carried out in six test systems (non-inoculation, inoculation with the B1 microbial consortium, inoculation with the B2 microbial consortium, inoculation with the B1 microbial consortium with the addition of γ-PGA (γ-poly glutamic acid), inoculation with the B2 microbial consortium with the addition of γ-PGA and inoculation with the γ-PGA solution) for 6 months. The effectiveness of the remediation treatments used was assessed based on chromatographic analyses of soil and plant material (roots, shoots) and toxicological analyses using four types of toxicological tests (PhytotoxkitTM (MicroBioTests Inc., Gent, Belgium), OstracodtoxkitTM (MicroBioTests Inc., Gent, Belgium), Microtox® Solid Phase Test (Modern Water Inc., New Castle, DE, USA), MARA (NCIMB Ltd., Aberdeen, UK)). The research conducted showed that the most effective method of bioremediation of soils contaminated with petroleum hydrocarbons was phytoremediation supported by bioaugmentation with the microbial consortium B2 with γ-PGA, which allowed for reducing the concentration of total petroleum hydrocarbons (TPHs) and polycyclic aromatic hydrocarbons (PAHs) in the tested soils by 53.98% and 49.54% (Soil DW-5) and 60.47% and 37.55% (Soil OS-5), respectively. In turn, the lowest bioremediation efficiency was recorded in non-inoculated systems, for which the concentration of TPHs and PAHs at the end of the study decreased by 18.40% and 16.14% (Soil DW-1) and 21.87% and 18.20% (Soil OS-1), respectively. The results of toxicological analyses confirmed the relationship between the concentration of TPHs and PAHs in the soil and its toxicity level.

TASER International, Inc. is one of the world's leading less-lethal weapons manufacturers and distributors. The case begins with a dramatic moment as the President and CEO of TASER International become aware of a highly critical article in Barron's. The article questions the legitimacy of their high stock price and casts doubt on their continued ability to grow. The case presents the company's counterarguments to the critical Barron's article, and asks for alternatives for TASER's next move into the relatively untapped consumer market with a new consumer-oriented product, the TASER X26C. The case resulted from lengthy in-person, email, and phone interviews with TASER's President, Tom Smith. In addition, the company and its products have been well publicized in the national business press and in the local newspapers. Further, product details and other information on TASERs and other less-lethal weapons has been published in numerous police and military sources. Finally, TASER International's website has been a rich source of supplemental information to support the writing of the case.

The paper deals with the the development of a specific company’s stock price time series. The aim of the paper is to use the time series method for a detailed analysis and evaluation of the development of Apple Inc. stock prices. Daily data from 2000 to 2020, daily data from the period of the economic crisis between 2007 and 2009 and daily data from the Covid-19 pandemic period from March 2020 to the end of the same year are used. The data, from the period of 2000 - 2020 show a gradual increase in Apple’s stock prices. The most common factor leading to the increase in stock prices is the launch of a new product or service on the global market. On the contrary, the reason for the decline in stock prices is customer dissatisfaction, the excess of demand over supply, or the political situation. The analysis of time series for the period of the economic crisis points to the fact that thanks to the development, innovation and constant introduction of new products into the market, the company was not significantly affected by the crisis and neither were stock prices. Naturally, there were some fluctuations in prices, but at the end of 2009, the company even reached the highest stock prices in its history to date. The analysis of time series during the global pandemic of Covid-19 shows a steady rise in stock prices. Currently, the company sells more and more products and introduces new services that help us work, study or entertain ourselves in these difficult times, in the safety of our homes.

Abstract We demonstrate the first use of an electrochemiluminescent (ECL) label, [4-(N-succimidyloxycarbonylpropyl)-4'-methyl-2,2'- bipyridine]ruthenium(II) dihexafluorophosphate (Origen label; IGEN Inc.), in DNA probe assays. This label allows rapid (less than 25 min) quantification and detection of polymerase chain reaction (PCR)-amplified products from oncogenes, viruses, and cloned genes. For the PCR, we used labeled oligonucleotide primers complementary to human papiloma virus and the Ha-ras oncogene. These samples were followed by ECL analysis or hybridization with specific, Origen-labeled oligonucleotide probes. These studies demonstrate the speed, specificity, and effectiveness of the new ECL labels, compared with 32P, for nucleic acid probe applications. We describe formats involving conventional methodologies and a new format that requires no wash step, allowing simple and rapid sample analysis. These rapid assays also reduce PCR contamination, by requiring less sample handling. Improvements in ECL detectability are currently under investigation for use in DNA probe assays without amplification.

Recently, companies such as Apple Inc, Fitbit Inc, and Garmin Ltd have released new wearable blood oxygenation measurement technologies. Although the release of these technologies has great potential for generating health-related information, it is important to acknowledge the repercussions of consumer-targeted biometric monitoring technologies (BioMeTs), which in practice, are often used for medical decision making. BioMeTs are bodily connected digital medicine products that process data captured by mobile sensors that use algorithms to generate measures of behavioral and physiological function. These BioMeTs span both general wellness products and medical devices, and consumer-targeted BioMeTs intended for general wellness purposes are not required to undergo a standardized and transparent evaluation process for ensuring their quality and accuracy. The combination of product functionality, marketing, and the circumstances of the global SARS-CoV-2 pandemic have inevitably led to the use of consumer-targeted BioMeTs for reporting health-related measurements to drive medical decision making. In this viewpoint, we urge consumer-targeted BioMeT manufacturers to go beyond the bare minimum requirements described in US Food and Drug Administration guidance when releasing information on wellness BioMeTs. We also explore new methods and incentive systems that may result in a clearer public understanding of the performance and intended use of consumer-targeted BioMeTs.

The demand for functional foods has increased notably in recent years due to growing consumer interest in diet and health issues. Currently, the food industry is introducing many types of new food products with functional attributes. Consequently, cannibalization is a critical issue for firms that offer multiple products within a certain product category. The identification and assessment of cannibalization are integral factors when making strategic decisions about new product introductions. Using scanner data from Information Resources, Inc. (IRI), pertaining to a particular functional food, namely a phytosterol-enriched product for orange juice, we find that no cannibalization effects exist with respect to its introduction. We also provide estimates of own-price and cross-price elasticities of the orange juice category using a synthetic demand system.

Abstract BACKGROUND: The plasma protein C1-inhibitor (C1-inh), belongs to the serpin superfamily and is the major inhibitor of the proteases of the complement and contact phase pathways. Hereditary or acquired deficiency of functional C1-inh results in angioedema episodes in affected individuals due to uncontrolled contact pathway activation and therapeutic C1-inh products are effective treatment for these patients. Therapeutic C1-inh products have also been shown to attenuate neutrophil activation and infiltration in various inflammatory conditions. This 'novel' anti-inflammatory effect of C1-inh is attributed to its non-serpin N-terminal domain. This domain is thought to express the tetrasaccharide, sialyl Lewisx (SLeX), through which C1-inh can interact with selectins on inflamed endothelium and prevent neutrophil rolling. However, C1-inh products contain small but significant amounts of co-purified proteins, the major one being the glycoprotein α1- antichymotrypsin (ACT), which is also an anti-inflammatory serpin. The potential influence of the glycans of ACT on SLeX - selectin interactions is not clear. METHOD: We investigated the presence of SLeX -like epitopes on C1-inh and ACT from commercially available therapeutic C1-inh preparations using western blotting and mass-spectrometry. The influence of the products and separated C1-inh and ACT on SLeX -selectin interaction was investigated in an a model system where SLeX -beads were rolled on immobilized E-selectin molecules. RESULT: We do not find any evidence of SLeX on C1-inh using either western blotting with anti-SLeX antibodies or by mass spectrometric analysis of C1-inh N- glycans. C1-inh products show modest but significant interference in SLeX -selectin interaction but surprisingly this is not observed for 'pure C1-inh' obtained from gel-filtration of the commercial product. On the contrary, ACT, also from the C1-inh product, shows the presence of SLeX -like epitopes, as detected by the antibody HECA-452 on western blot. In addition, at concentrations present in C1-inh products (20 -150 μg ACT/ mg active C1-inh), ACT can interfere with SLeX -selectin interactions, in a sialic acid dependent manner. These concentrations of ACT can be achieved in vivo with a dose of as low as 2000 U of a C1-inh product, suggesting that ACT can contribute to the anti-inflammatory effects observed in studies with C1-inh products. CONCLUSION: We conclude that the 'novel' anti-inflammatory effects of C1-inh are unlikely due to SLeX and can in fact be partly due to ACT. This fresh evidence challenges a long held assumption and paves the way for development of ACT, alone or in combination with C1-inh, as a new anti-inflammatory therapeutic. Zeo, Inc. was one of the pioneer companies in the quantified self-industry offering a digital solution for individuals suffering from sleep problems. The company was founded by three undergraduate students who successfully raised capital, make strategic partnerships with major brands like the National Football League (NFL), and introduced their products in stores like Best Buy.The collegiate founders recognized their lack of knowledge and skills from the beginning. To address this gap, they developed an advisory board to guide them on scientific matters. Later, they hired a seasoned executive with industry experience to run and grow the company. Zeo, Inc. also was getting favorable attention among leading sleep experts with publications in scientific journals about the promise of this innovative technology. Zeo, Inc. was positioned to potentially disrupt the technological and business model of the sleep industry at the time.Suddenly, Zeo, Inc. announced it was shutting down the company. Technology pundits, the media and customers were surprised, “Why would a fast-rising company shut down?” Given the lack of board and senior leadership transparency, many speculated as to why Zeo, Inc. closed. The reasons ranged from a lack of a focus on the business model to not keeping up with new entrants into the digital health space such as FitBit. This case highlights for students the growth stages of startups with an emphasis of how a widely acclaimed startup can fail and the lessons to be learned from failure.

In the context of the Internet era, as a tech giant, Apple has a huge and lasting influence in the hearts of consumers. While Apple is constantly introducing new technical products, it is also enriching and updating its brand value in the process of development. Nowadays, brand culture is one of the hot research topics. However, there is still a research gap in analyzing brand culture from the perspective of semiotics. Based on the semiotic theories of Pierce and Barthes and other cultural theories, this essay analyzes Apples brand construction through a specific theoretical framework and discusses the cultural significance behind the brand. The research shows that Apples brand can be constructed in the symbolic sense from three aspects. Moreover, Apples long-term prosperity can be regarded as a cultural symbol, implying the symbol worship of consumers and the domination of modern technology. The article is an exploration of the application of semiotics and the symbolic meaning of the brand.

ABSTRACTCaldicellulosiruptor besciigrows optimally at 78°C and is able to decompose high concentrations of lignocellulosic plant biomass without the need for thermochemical pretreatment.C. besciiferments both C5and C6sugars primarily to hydrogen gas, lactate, acetate, and CO2and is of particular interest for metabolic engineering applications given the recent availability of a genetic system. Developing optimal strains for technological use requires a detailed understanding of primary metabolism, particularly when the goal is to divert all available reductant (electrons) toward highly reduced products such as biofuels. During an analysis of theC. besciigenome sequence for oxidoreductase-type enzymes, evidence was uncovered to suggest that the primary redox metabolism ofC. besciihas a completely uncharacterized aspect involving tungsten, a rarely used element in biology. An active tungsten utilization pathway inC. besciiwas demonstrated by the heterologous production of a tungsten-requiring, aldehyde-oxidizing enzyme (AOR) from the hyperthermophilic archaeonPyrococcus furiosus. Furthermore,C. besciialso contains a tungsten-based AOR-type enzyme, here termed XOR, which is phylogenetically unique, representing a completely new member of the AOR tungstoenzyme family. Moreover, inC. bescii, XOR represents ca. 2% of the cytoplasmic protein. XOR is proposed to play a key, but as yet undetermined, role in the primary redox metabolism of this cellulolytic microorganism.

This research aims to conduct an analysis of Five Forces and BCG Matrix for Apple Inc. The Five Forces analysis is used to evaluate the competitiveness of the industry in which Apple operates, while the BCG Matrix is employed to measure the company's product portfolio. Data for this study are sourced from reliable resources, including the company's financial reports, industry reports, and related publications. The Five Forces analysis reveals that the technology and software industry in which Apple operates is highly competitive. Threats from major competitors, the level of product substitution, and the bargaining power of buyers significantly impact the company's strategies and performance. However, Apple's strong brand equity and robust product ecosystem provide a competitive advantage in navigating the pressures from these five forces. In the BCG Matrix analysis, Apple's core products such as the iPhone, iPad, and Mac show a position of stars and question marks, indicating high growth potential. On the other hand, some older products and unproven innovations fall into the category of cash cows and dogs, with lower growth prospects. The results of the Five Forces and BCG Matrix analysis provide a comprehensive overview of Apple's market position and aid in formulating the company's long-term strategies. Apple's management can leverage these findings to optimize the product portfolio and address competitive challenges in the ever-evolving technology industry. While Apple holds a strong market position, new challenges will continually emerge, necessitating effective strategies to maintain competitiveness and ensure the company's growth in the future.

The important effect of dietary factors on health status has been recognized since antiquity. Since the discovery of the beneficial effects of dietary phytochemicals and bioactives, a new dimension of foods have emerged in the market. These “functional foods” are being developed by all major food companies and new ones are regularly brought into the market. While developing new functional foods and nutraceuticals, the association and identification of such foods and beverages with traditional foods and medicinal preparations, and/or popular forms of existing products are bound to bring long-standing consumer acceptance, which is an important desirable factor in sales and marketing. Following this concept, Zafi Beverages, Inc., Chicago, is developing a new line of functional products (new herbal teas and energy drinks). Zafiis also introducing a unique marketing and distribution system to create a new team of entrepreneurs, providing an excellent opportunity for growth in sales and marketing to new entrepreneurs. It targets ethnic entrepreneurs by offering an opportunity to use their networking abilities and be part of an exciting new partnership in the new host country, as well as a strategic business plan. The ethnic entrepreneurs are also constantly in contact with their consumers by virtue of the existing ethnic allegiance and cohesion and are able to identify the consumer needs and concerns directly. These small ethnic entrepreneurial networks can be identified as distinct “micro-marketing systems” within the national economy. The advertised market potential for Zafi is summarized to include an offer of immediate cash flow, more revenue and profits, marketing and financial education support, as well as a promising new line of products.

Personalized recommendation systems are becoming an essential feature of online shopping platforms, where an abundance of products and information can be overwhelming to users. These systems play a pivotal role in enriching user experiences by suggesting relevant products based on users' past behavior and preferences. However, accurate recommendations present several challenges, including data sparsity, cold start issues for new users or products and recognizing intricate patterns across large datasets. This research seeks to improve Amazon's e-commerce recommendation system using matrix factorization methods to overcome some of its challenges. Matrix factorization dissects user/item interactions into lower dimensional latent factors which represent user and item preferences. At Matrix Factorization Methods Inc, the author studies their effectiveness in increasing recommendation accuracy while managing data sparsity and cold start issues in real-world Amazon data collection, preprocessing and various matrix factorization algorithms implementation and evaluation will be carried out and compared against baseline approaches to demonstrate their potential in increasing recommendation accuracy while meeting challenges posed by e-commerce recommendation systems.

In June 2020, Massachusetts became the first state to implement a comprehensive flavored tobacco restriction. One concern was that Massachusetts residents would travel to New Hampshire to purchase restricted products. This article assesses tobacco sales in both states post–law implementation. Retail scanner data were obtained from the Nielsen Company and Information Resources, Inc (IRI), from 1 year pre–law implementation to 2 years post–law implementation. Data post–law implementation were compared with data from 1 year pre–law implementation (baseline). In Massachusetts, 2 years post–law implementation, flavored and menthol tobacco sales decreased by more than 90%. Total sales decreased by around 20%. In New Hampshire, menthol tobacco sales increased (25.1% in IRI and 18.2% in Nielsen), but total sales changed minimally (<5% increase in IRI, <5% decrease in Nielsen). When data from both states were combined, total sales decreased by around 10%. The net decrease in total tobacco sales across Massachusetts and New Hampshire indicates Massachusetts' flavored tobacco restriction resulted in a reduction in tobacco sales despite potential cross-border purchases.

Abstract Introduction: Hemophilia B is characterized by a functional deficiency in factor IX (FIX), which results in spontaneous or trauma-induced bleeding into soft tissue, muscles, and joints. Replacing deficient FIX with plasma-derived or recombinant FIX products, either prophylactically or on an on-demand basis, is a mainstay in the prevention and treatment of bleeding episodes in patients with hemophilia B. A previous study of patients with moderately severe to severe hemophilia B compared the efficacy and safety of 2 secondary prophylaxis regimens of a recombinant coagulation FIX product (nonacog alfa, 50 IU/kg twice weekly or 100 IU/kg once weekly) with on-demand treatment. The study found a significant reduction (P<.0001) in the annualized bleeding rate (ABR) for the prophylaxis regimens compared with on-demand therapy. No significant differences in the ABR were observed between the once-weekly and twice-weekly prophylaxis regimens, and safety and tolerability profiles were similar for both. Objective: To demonstrate that once-weekly prophylaxis with nonacog alfa reduces the ABR compared with on-demand treatment in patients with moderately severe to severe hemophilia B. Methods: This open-label, 2-period study enrolled males aged 12 to 65 years with moderately severe to severe hemophilia B (FIX plasma activity ≤2%), ≥100 prior exposure days to FIX products, and no history of or current FIX inhibitor. Patients received on-demand therapy with nonacog alfa for 6 months (dose selected at the investigator's discretion) followed by prophylactic treatment with nonacog alfa 100 IU/kg once weekly for 12 months. Recovery was assessed on day 1, week 26, and week 78, after a 72-hour washout from any FIX product. Plasma samples were collected before administration and at 30 minutes after nonacog alfa infusion. The primary efficacy end point was the ABR (number of bleeding events/[days on treatment period/365.25]). Secondary end points included the response to on-demand treatment of a bleeding event, incidence of less-than-expected therapeutic effect (LETE) in the prophylaxis setting; and FIX inhibitor development. Safety was also assessed. Results: In total, 25 patients were enrolled and received at least 1 dose of study medication; all patients completed the study. Mean ±SD age was 31.3 ±12.6 years; 5 patients were <18 years; 9 patients were Asian and 16 were white. ABR results are summarized in the Table; ABR for the prophylaxis period was significantly lower (P<.0001) than the ABR for the on-demand period. Of the 507 bleeding events treated over the study, 271 (53.5%) first infusions resulted in an “excellent” response and 177 (34.9%) resulted in a “good” response; the majority of bleeding events (416/507; 82.1%) resolved with 1 infusion. Of the 1254 prophylaxis infusions administered during the study, none were associated with an occurrence of LETE. No patient developed a FIX inhibitor or experienced a thrombotic event during the study. The most commonly reported treatment-emergent AEs (≥20%) were inappropriate schedule of drug administration (24%), arthralgia (20%), upper respiratory tract infection (20%), and toothache (20%) during the prophylaxis period, and headache (32%) and arthralgia (20%) during the on-demand period. Five patients (20%) experienced a total of 6 serious AEs; blood pressure decreased, pain resulting in hospitalization, chicken pox, lipoma, and nephrolithiasis (2 events in 1 patient). Only the serious AE of blood pressure decreased was considered related to study drug. No deaths occurred during the study. Conclusions: Prophylactic treatment with nonacog alfa 100 IU/kg once weekly significantly reduced the number of bleeding events compared with on-demand treatment in patients with moderately severe to severe hemophilia B. Nonacog alfa 100 IU/kg once weekly was well tolerated; no new safety issues were observed. Table. Walter Lorenz Surgical Inc., Jacksonville, FL, specializes in the medical devices known as rigid fixation implants. Lorenz Surgical was purchased by Biomet Inc. of Warsaw, Indiana in the year 1992, which resulted in Lorenzo owning two computer-aided design (CAD) systems. In 1996, with the completion of Lorenz Surgical's new manufacturing facility in Jacksonville, all Lorenz operations were transferred back to Florida, including all the manufacturing equipment and its three seats of Unigraphics, which by then were running on Windows NT workstations. The company's management feels it could have stayed with Unigraphics and accomplished its goals, but that adding Solid Edge was a good move. It gave Lorenz flexibility in hiring, allowed it to buy more CAD seats than it could have if it had stayed with Unigraphics alone, and provided a very productive tool. Lorenz's surgical instruments are currently designed exclusively in Solid Edge. Instruments can be modeled in either CAD system, but the job goes faster in Solid Edge. The creators of Solid Edge put a lot of effort into usability, and this shows in how few mouse clicks are needed for common operations. Products that have many standard features, such as a screwdriver consisting of m any cylinders, are very quickly modeled in Solid Edge.

With the rapid development of today's society and economy, more and more public investors are not satisfied with bank financial products, which have lower value-added, but turn their attention to the capital market. The quantitative technique of DCF model combined with qualitative analysis to assess the intrinsic value of enterprises provides an effective reference for public investors. Tesla, a company that produces new energy vehicles, has shown rapid growth in recent years and is suitable for valuation using DCF models. This paper predicts the growth rate of Tesla's operating income in the next five years based on its historical financial data, and uses the percentage of sales method combined with qualitative analysis to calculate the present value of the enterprise's free cash flow in the next five years, and also calculates the present value of the enterprise's future perpetuity based on the assumption of the enterprise's going concern, and the two are added together to obtain the intrinsic value of Tesla Inc. as $5160.95 billion. The total market value of Tesla Inc. on November 22, 2022 (valuation point in time) is $5365.34 billion (data source: https://www.msn.cn). The intrinsic value of the enterprise is lower than the market price, and investors are advised not to buy.

SimPlate for Total Plate Count–Color Indicator (TPC-CI, IDEXX Laboratories, Inc., Westbrook, Me.) is a new medium that incorporates the redox dye resazurin to detect and quantify bacteria in food. Enumeration is achieved by the most probable number method using a SimPlate device. Viable bacteria are detected in each well of the SimPlate device by the biochemical reduction of resazurin, which is blue, to the pink resorufin or the clear dihydroresorufin indicators. Results after 24 h of incubation for TPC-CI are highly correlated with standard plate count agar after 48 h of incubation. Correlation coefficients from studies conducted at five laboratories ranged from 0.94 to 0.98 in side-by-side comparisons against standard plate count agar. Four additional test sites, using alternative methods for determining the aerobic plate count in food, reported similar results in comparison studies (r = 0.91 to 0.97). The slopes from linear regression analysis at all sites ranged from 0.91 to 0.98, with y intercepts ranging from 0.11 to 0.84. Samples used for the validation of TPC-CI included raw food products (i.e., liver and grains), which may contain natural enzymes that interfere with enzyme-based detection methods. No interference was seen from the foods tested. These results suggest that TPC-CI is a suitable alternative to existing plate count methods and has reduced incubation time.

Global programs targeting 5 preventive chemotherapy neglected tropical diseases (PC-NTDs) have scaled up rapidly in recent decades due, in large part, to the generous drug donations from 6 pharmaceutical companies—Eisai; Johnson & Johnson (J&J); GlaxoSmithKline (GSK); Merck & Co., Inc., Kenilworth, New Jersey, United States of America (MSD); Merck KgaA; and Pfizer. Today, the scale of the PC-NTD drug donation programs is staggering. Nearly 15 billion tablets have been manufactured, packaged, shipped, and distributed in order to reach the people in need. The supply chains established to support such massive operations are enormously complex. Here, we describe a unique public–private partnership that was formed to bring together supply chain expertise to overcome the critical challenges associated with such large-scale production and delivery of donated pharmaceutical products.

This article describes that fire researchers apply old and new tests to assure that materials meet safety requirements. Baltimore-based Hughes Associates Inc., a fire research firm, uses standard tests and computer modeling, and, in some cases, will develop tests to ensure that new building products satisfy the safety requirements of existing building codes. Hughes Associates also facilitates contact between its client and the appropriate code-making organizations, whether local, state, federal in the case of governmental agencies, or internationally through its offices in Singapore and in Milan, Italy. The data derived from the small-scale tests are also used in flame spread computer modeling testing. These tests use a series of proprietary computer modeling programs to predict the behavior of the product’s flames—for example, how high the flames would reach, and how quickly they would spread. The development of amusement park attractions is driving fire testing to prove that these attractions meet the stringent public assembly provision in fire codes.

A large number of higher rung companies have not been able to contribute effectively to correct India's balance of trade. In fact, a large proportion (over 50%) is involved in aggravating it. The performance of lower rung corporates is comparatively more satisfactory. The contribution by the higher rung corporates to the globalization of Indian business is even more unsatisfactory. With a few honourable exceptions, they are globalizing more actively inwards than outwards. Most of the corporates are not contributing to increasing the global competitiveness of India through technological capability building. There is increasing technological dependence of these companies, perhaps strategic dependence, on foreign companies as manifested by accelerated pace of foreign collaboration by corporate leaders in every field. Corporates are not able to withstand global competition. The performance of over 50 per cent of pre-reform corporate leaders has gone down both in terms of their sales rank and profit position. The performance of the neo-leader companies (entering the fray after 1992) is even worse. This downturn has started affecting the national economy and society. The deteriorating performance of corporate leaders has blocked a huge amount of country's savings in unutilized/underutilized fixed assets and created sizeable non- performing assets. A majority of corporates are still not considering internationalization of their business leaving the task to small companies, especially the software ventures, which are vulnerable to host country protests as well as increasing share of the MNCs in business process outsourcing. Considering all these factors, it is time the corporates realized that they also have a role to play in ensuring sound health of their industry and Indian economy without which their own survival may be at stake in the long run. The magnitude of fall in their performance suggests that the issue has to be taken up as a strategic challenge and new models and approaches for arresting the fall need to be developed. This study analyses the policy implications of this phenomenon and comes up with the following suggestions: The policy-makers need to critically evaluate the export-import and other policies and examine what policy support is needed to make more Indian corporates assume responsibility for increasing the net exports and for internationalization of Indian business. The creation of a large amount of non-performing assets due to failed expansion and diversification moves would entail major ongoing restructuring of the underutilized assets in the form of acquisitions, mergers, and divestitures. There is a need for proactive policies for facilitating such strategies with concern for protecting the interests of larger section of the society. The policy-makers also have to think and consciously decide as to what level of “exit” is to be tolerated as a price to be paid for better quality and increased supply of goods and services. Corporate leaders are not paying enough attention to develop and/or scale up native products that are embedded in the natural endowment and socio-cultural context of the country which could be leveraged as a strategic tool for meeting global competition on local strengths. New/additional policy measures seem to be neces- sary for the purpose. There is a need for large-scale, coordinated research by mobilizing and unifying the national efforts with active industry involvement at various stages of research.

Abstract Spray trials were conducted in Kent and New Castle Counties, DE, and Salem and Cumberland Counties, NJ. Treatments consisted of oil-based isolates of Bacillus thuringiensis formulated as Condor OF and Dipel 8 L diluted with water, Condor OF applied undiluted and an untreated control. Treatments were replicated five times in a randomized complete block. Woodlots were 10-84 acreas in size and consisted of isolated stands. Fifteen 0.025-acre fixed radius plots were established within each woodlot where treatment effects were measured. A Grumman Ag-Cat 600 aircraft equipped with eight AU-5000 Micronair rotary atomizers on a standard spray boom was used to apply the treatments. All B. thuringiensis formulations were applied at 20 billion international units (BIU)/acre (40 oz formulation/acre). For the diluted applications, Condor and Dipel were applied at a volumetric rate of 128 oz/acre including 2% (v/v) Bond (Loveland Industries, Inc.) as a sticker. The aircraft was operated at an airspeed of 90 mph on a 75 ft swath width, about 50 ft above the forest canopy. The pump pressure was 40 psi and the variable restrictor unit on the nozzles was set at 9 and 5 for the diluted and undiluted applications, respectively. The blade angle on the nozzles nearest the fuselage was set at 65 and 35°, and 70 and 35° for the other six nozzles on the boom, for the diluted and undiluted applications, respectively. Woodlots were treated on 22 and 27 May in Delaware and New Jersey, respectively, when GM was predominately 3rd instar. Pre- (Spring) and postspray (Fall) egg mass counts were made within each of the 15 subplots. GM larvae were counted under burlap bands placed on one dominant preferred tree 25 ft outside each 0.025- acre plot. Estimates of feeding damage were based on examination of each burlapped tree and were rated in one of seven classes: 1, 0%; 2, 1-5%; 3, 6-10%; 4, 11-25%; 5, 26-50%; 6, 51-75%; and 7, >75.

Abstract Factor VIII (FVIII) replacement products enable comprehensive care in hemophilia A. Treatment goals in severe hemophilia A are expanding beyond low annualized bleed rates to include long-term outcomes associated with high sustained FVIII levels. Endogenous von Willebrand factor (VWF) stabilizes and protects FVIII from degradation and clearance, but it also subjects FVIII to a half-life ceiling of ∼15 to 19 hours. Increasing recombinant FVIII (rFVIII) half-life further is ultimately dependent upon uncoupling rFVIII from endogenous VWF. We have developed a new class of FVIII replacement, rFVIIIFc-VWF-XTEN (BIVV001), that is physically decoupled from endogenous VWF and has enhanced pharmacokinetic properties compared with all previous FVIII products. BIVV001 was bioengineered as a unique fusion protein consisting of a VWF-DʹD3 domain fused to rFVIII via immunoglobulin-G1 Fc domains and 2 XTEN polypeptides (Amunix Pharmaceuticals, Inc, Mountain View, CA). Plasma FVIII half-life after BIVV001 administration in mice and monkeys was 25 to 31 hours and 33 to 34 hours, respectively, representing a three- to fourfold increase in FVIII half-life. Our results showed that multifaceted protein engineering, far beyond a few amino acid substitutions, could significantly improve rFVIII pharmacokinetic properties while maintaining hemostatic function. BIVV001 is the first rFVIII with the potential to significantly change the treatment paradigm for severe hemophilia A by providing optimal protection against all bleed types, with less frequent doses. The protein engineering methods described herein can also be applied to other complex proteins.
