Resumo:

This paper is the first publication of six Ancient Egyptian ostraca from the State Pushkin Museum of Fine Arts, Moscow (I,1b 334, 340, 344, 347, 348, 362) with fragments of The Teaching of Khety, also known as The Satire of the Trades. The ostraca used to belong to the collection of the Russian Egyptologist Vladimir S. Golenischeff. They date from the 19th– 20th Dynasty and probably originate from Deir el-Medina.

<b><i>Introduction:</i></b> Elaborated intrusion theory suggests that imagery is central to craving; however, the possibility that cue-elicited multisensory imagery produces such urges has not been studied enough in the literature. Thus, we investigated the role of cue-elicited multisensory imagery on alcohol craving in individuals who are hazardous and social drinkers compared to mental and neutral imagery conditions. <b><i>Methods:</i></b> In an online experiment, hazardous and social drinkers (<i>N</i> = 348) between 18 and 45 years old were randomised to multisensory, mental, and neutral imagery exposure. The level of craving intensity was measured before and after imagery exposure. Also, participants rated vividness and sensory features scales after the exposure. <b><i>Results:</i></b> The level of craving was significantly higher in multisensory imagery condition compared to neutral condition (<i>b</i> = 1.94, <i>p</i> < 0.001, SE = 0.30, <i>t</i>(344) = 6.52, standardised mean difference [SMD] = 0.89) and in mental imagery condition compared to neutral condition (<i>b</i> = 1.82, SE = 0.30, <i>t</i>(344) = 6.52, <i>p</i> < 0.001, SMD = 0.83). The difference between the level of craving intensity between the multisensory and mental was not significant (<i>b</i> = 0.12, SE = 0.22, <i>t</i>(344) = 0.53, <i>p</i> = 0.594, SMD = 0.06). Moreover, craving intensity in response to multisensory versus neutral imagery was significantly stronger among hazardous drinkers (<i>b</i> = −2.90, SE = 0.83, <i>t</i>(341) = −3.50, <i>p</i> < 0.001). The level of vividness was not significantly different between any conditions. The difference between levels of sensory features was higher in multisensory imagery condition compared to neutral (<i>b</i> = 0.95, SE = 0.30, <i>t</i>(345) = 3.17, <i>p</i> = 0.002, SMD = 0.49) and mental imagery condition (<i>b</i> = 0.67, SE = 0.23, <i>t</i>(345) = 2.36, <i>p</i> = 0.004, SMD = 0.35). <b><i>Conclusion:</i></b> Results suggest that cue-elicited multisensory imagery may be a useful tool for eliciting alcohol craving responses and provide an additional means for better understanding the multi-layered mechanism of craving.

Abstract The isoflavone ME-344 is a potent anti-cancer agent with preclinical efficacy in solid tumors in vitro and in vivo. In a recently completed phase I clinical trial in patients with refractory solid tumors, ME-344 was well tolerated and clinical responses in refractory patients were observed. ME-344 has been shown to reduce mitochondrial ATP generation in a tumor selective manner, though other potential activities have not been fully defined. In addition, the preclinical efficacy of ME-344 in leukemia has not been established. Therefore, we investigated the anti-leukemic properties and the mechanism of action of ME-344. We treated a panel of 7 leukemia cell lines with increasing concentrations of ME-344, and measured cell growth and viability. ME-344 was cytotoxic to the 7 leukemia cell lines with an IC50 in the range of 70-260 nM. In addition, ME-344 induced preferential death in primary AML patient samples over normal hematopoietic cells. In an OCI-AML2 xenograft model, ME-344 reduced tumor growth by up to 95% of control without evidence of toxicity. Mechanistically, as reported in studies in solid tumors, ME-344 increased mitochondrial ROS generation in leukemic cells. However, antioxidant treatment did not rescue cell death, suggesting that ME-344 has additional targets beyond the mitochondria. To identify additional targets of ME-344, we conducted haplo-insufficiency profiling in S.cerevisiae to identify genes whose heterozygous deletion confers increased sensitivity to ME-344. The top hits from the screen were genes involved in cytoskeletal organization. Therefore, we tested the effects of ME-344 on actin and tubulin polymerization in cell free assays. While ME-344 did not inhibit actin polymerization, it inhibited tubulin polymerization by interacting near the colchicine-binding site and at a site distinct from vinca alkaloids. Furthermore, cells resistant to tubulin inhibitors due to tubulin point mutations were also resistant to ME-344. Finally, we showed that ME-344 synergizes with vinblastine in leukemia cells. Thus, our study demonstrates that ME-344 displays preclinical efficacy in leukemia through a mechanism at least partly related to targeting tubulin polymerization. Abstract Lack of clinically validated markers associated with the development of neutralizing antibodies (inhibitors) to the replacement protein-drug, Factor VIII (FVIII) is a major challenge in managing Hemophilia A (HA). By utilizing Machine Learning (ML) and Explainable AI (XAI) we identified and ranked relevant variables (biomarkers) that could be predictive for developing inhibitors to FVIII drugs in hemophilia A patients. The dataset used for the study was derived from the My Life Our Future (MLOF) repository and included variables such as age, race, ethnicity, mutations in the F8 gene and the Human Leucocyte Antigen Class II (HLA-II) type. We computed the number of foreign FVIII derived peptides, based on the alignment of the endogenous FVIII and infused drug sequences, and the foreign-peptide HLA-II molecule binding affinity (using the NetMHCIIpan algorithm). The complete dataset with the added variables was trained and validated using multiple ML classification models to identify the top performing model, which was chosen for further processing with XAI via SHAP (SHapley Additive exPlanations) to identify the variables critical for the prediction of FVIII inhibitor development in a hemophilia A patients. The top five variables for predicting inhibitor development based on SHAP values are: (i) the baseline activity of the FVIII protein, (ii) mean affinity of all foreign peptides for HLA DRB 3, 4, & 5 alleles, (iii) mean affinity of all foreign peptides for HLA DRB1 alleles, (iv) the maximum affinity among all foreign peptides for HLA DRB1 alleles, and (v) F8 mutation type. I enjoyed reading Kanavillil Rajagopalan's article "On the Theoretical Trappings of the Thesis of Anti-Theory; or, Why the Idea of Theory May Not, After All, Be All That Bad: A Response to Gary Thomas" in the Fall 1998 issue of the Harvard Educational Review, and I am grateful to him for his thoughtful and detailed response to my article, "What's the Use of Theory?" (HER, Spring 1997). I am glad that we can agree on the pretensions of some kinds of theory and on its inhibiting effects on original thinking. I am pleased that he believes that I examine certain theories "with great skill and perspicacity" (p. 337), even if I am also guilty of "smug skepticism" (p. 346), "crypto-scientism" (p. 344), making an "impassioned harangue" (p. 336), and making a "fatal mistake" (p. 347) in my reasoning. Rajagopalan's critique comes in two waves: in the first he tries to establish that theory's ambit is wide — indeed it is so wide that it encompasses anything to do with structured thought. A simple proposition follows: theory is any structured thought, ergo Thomas's structured thought is theory. Once this ground is established (that I'm a theorist), the way is easy for the second surge. Here, Rajagopalan claims that since I am a theorist I am guilty of grave contradictions and inconsistencies in my argument against theory.

На основу архивских истраживања, објављених историјских извора, периодике и релевантне литературе у раду је изанализирано једно од кључних питања спровођења културне политике у Краљевини Југославији: питање издавања јединствених уџбеника. Праћени су и напори Друштва народа да се установи посебна научна дисциплина која ће се бавити проучавањем, упоређивањем и ревизијом уџбеника. После проглашења диктатуре краља Александра 1929. у оквиру унификације економског и културног простора настојало се на државном издању јединствених уџбеника за целу земљу, како је то било дефинисано Законом о уџбеницима за народне, грађанске, учитељске и средње школе 27. септембра 1929. То је изазивало расправе у широј јавности, а посебно у стручној, поготово у вези са расписивањем конкурса за израду уџбеника у јануару и фебруару 1935. Покушаји да се уџбеници унифицирају су остали без успеха и ова политика је практично напуштена 1939. године.

Hydrogen sulfide (H2S) emerged recently as an anti-oxidative signaling molecule that contributes to gastrointestinal (GI) mucosal defense and repair. Indomethacin belongs to the class of non-steroidal anti-inflammatory drugs (NSAIDs) and is used as an effective intervention in the treatment of gout- or osteoarthritis-related inflammation. However, its clinical use is strongly limited since indomethacin inhibits gastric mucosal prostaglandin (PG) biosynthesis, predisposing to or even inducing ulcerogenesis. The H2S moiety was shown to decrease the GI toxicity of some NSAIDs. However, the GI safety and anti-oxidative effect of a novel H2S-releasing indomethacin derivative (ATB-344) remain unexplored. Thus, we aimed here to compare the impact of ATB-344 and classic indomethacin on gastric mucosal integrity and their ability to counteract the development of oxidative gastric mucosal injuries. Wistar rats were pretreated intragastrically (i.g.) with vehicle, ATB-344 (7–28 mg/kg i.g.), or indomethacin (5–20 mg/kg i.g.). Next, animals were exposed to microsurgical gastric ischemia-reperfusion (I/R). Gastric damage was assessed micro- and macroscopically. The volatile H2S level was assessed in the gastric mucosa using the modified methylene blue method. Serum and gastric mucosal PGE2 and 8-hydroxyguanozine (8-OHG) concentrations were evaluated by ELISA. Molecular alterations for gastric mucosal barrier-specific targets such as cyclooxygenase-1 (COX)-1, COX-2, heme oxygenase-1 (HMOX)-1, HMOX-2, superoxide dismutase-1 (SOD)-1, SOD-2, hypoxia inducible factor (HIF)-1α, xanthine oxidase (XDH), suppressor of cytokine signaling 3 (SOCS3), CCAAT enhancer binding protein (C/EBP), annexin A1 (ANXA1), interleukin 1 beta (IL-1β), interleukin 1 receptor type I (IL-1R1), interleukin 1 receptor type II (IL-1R2), inducible nitric oxide synthase (iNOS), tumor necrosis factor receptor 2 (TNFR2), or H2S-producing enzymes, cystathionine γ-lyase (CTH), cystathionine β-synthase (CBS), or 3-mercaptopyruvate sulfur transferase (MPST), were assessed at the mRNA level by real-time PCR. ATB-344 (7 mg/kg i.g.) reduced the area of gastric I/R injuries in contrast to an equimolar dose of indomethacin. ATB-344 increased gastric H2S production, did not affect gastric mucosal PGE2 content, prevented RNA oxidation, and maintained or enhanced the expression of oxidation-sensitive HMOX-1 and SOD-2 in line with decreased IL-1β and XDH. We conclude that due to the H2S-releasing ability, i.g., treatment with ATB-344 not only exerts dose-dependent GI safety but even enhances gastric mucosal barrier capacity to counteract acute oxidative injury development when applied at a low dose of 7 mg/kg, in contrast to classic indomethacin. ATB-344 (7 mg/kg) inhibited COX activity on a systemic level but did not affect cytoprotective PGE2 content in the gastric mucosa and, as a result, evoked gastroprotection against oxidative damage.

H.-U. Wiemer opposes the image which Libanius gives of Constantine in his fifty-ninth oration (Panegyric of Constantius and Constans) to that which emerges from his later works, especially those of the Theodosian period when hostility is obvious, mirroring the opinion of pagan circles, who held this Emperor responsible for most of the calamities endured by the Empire in the fourth century. As the epideictic genre required, in 344/5 or in 348/9 the father had to be praised so that the sons could be praised too. However, Wiemer claims that ‘the panegyric of 344/5 already foreshadows the critical view’, by remaining silent about Christianity, the founding of Constantinople, and the tax policy on the one hand, and on the other hand by clearly declaring Constantine responsible for the war against Persia, which troubled the entire reign of Constantius. I wish to supplement these remarks with a few others and to suggest that the portrait of Constantine in Oration 59 presents us with a case of ‘disguised intention’—a rhetorical proceeding based on ὑποδήλωσις and παραψόγους: the orator tries to convey a message that is different from his apparent intentions, even opposite to them.

Abstract The 5-year survival rate for adult patients with acute myeloid leukemia (AML) treated with cytarabine-based chemotherapy remains less than 30%, due to drug resistance and disease relapse. Recently, a selective inhibitor of anti-apoptotic Bcl-2, venetoclax, was approved by the FDA in combination with low dose cytarabine or hypomethylating agents for treating newly diagnosed AML patients who are 75 years of age or older or for those who are unfit for standard chemotherapy, providing more treatment options for this group of patients. Although the response rate to these newly approved combination therapies is reported to be 70%, the median overall survival is only 10-18 months showing that the duration of response is limited. Therefore, novel therapeutic agents are in demand to enhance venetoclax activity against AML and to combat AML resistant to cytarabine-based chemotherapy. Cytarabine-resistant AML cells lead to relapse and rely on oxidative phosphorylation (OXPHOS) for survival. In addition, it has been reported that targeting OXPHOS can enhance venetoclax activity against preclinical models of AML. Thus, we hypothesize that OXPHOS suppressing agents would be good candidates to combine with and enhance venetoclax antileukemic activity against newly diagnosed AML and those with resistance to cytarabine. A novel isoflavone, ME-344, has been shown to suppress OXPHOS in cell lines derived from solid tumors by inhibiting Complex I of the electron transport chain. We hypothesized that combining ME-344 with venetoclax would result in synergistic antileukemic activity against AML. Consistent with our hypothesis, combining ME-344 with venetoclax resulted in synergistic induction of apoptosis in AML cell lines, including those with acquired cytarabine resistance. The combination of these two agents also resulted in synergistic antileukemic activity in one primary AML patient sample, as determined by MTT assay. The combination of ME-344 and venetoclax prolonged the median survival of MV4-11 leukemia- bearing NSGS mice by 37% (median survival of 48 days compared to 35 days for vehicle control treated mice, n=5 per arm, p<0.0001). This is in contrast to the venetoclax combination with cytarabine, which prolonged median survival of the same xenograft model by 7.5% (Luedtke et al., Signal Transduction and Targeted Therapy, 2020; 5:17). ME-344 alone (9-hour treatment) reduced basal mitochondrial respiration in AML cells by 10% prior to induction of apoptosis. When treated with ME-344 for 8-hours followed by combined ME-344 and venetoclax for an additional 1-hour, basal mitochondrial respiration was reduced by 18% (again prior to detection of apoptosis initiation). This sequential combination regimen also decreased the mitochondrial membrane potential (by JC-1 staining and flow cytometry analysis) when compared to untreated control and single treatment. Additionally, apoptosis induction by the combination of ME-344 and venetoclax or ME-344 alone was significantly enhanced when AML cells were forced to utilize OXPHOS by replacing glucose with galactose in the culture medium. Further investigation revealed that apoptosis induced by ME-344 was partially attenuated when Mcl-1 was overexpressed, Bak was knocked down, or caspase activation was inhibited. This suggests a mechanism that involves components of the intrinsic apoptosis pathway. Targeted metabolomics analyses of MV4-11 cells treated with ME-344 for 8 h revealed a significant reduction of essential metabolites involved in the de novo purine biosynthesis pathway, specifically AICAR (p=0.001) and IMP (p=0.004). Given the critical role of purine in cell proliferation and survival, suppression of purine biosynthesis by ME-344 may represent a novel mechanism underlying its enhancement on the antileukemic activity of venetoclax against AML. Interestingly, inhibition of this pathway by the purine biosynthesis inhibitor lometrexol, also synergistically enhanced apoptosis in AML cells induced by venetoclax. Taken together, these results suggest that ME-344 suppresses OXPHOS and the purine biosynthesis pathway to enhance the antileukemic activity of venetoclax against AML. Further in-depth mechanistic studies into the suppression of purine biosynthesis and OXPHOS, as well as studies of ME-344 and venetoclax against cytarabine-resistant AML in a mouse model are warranted. 1. Recent studies have demonstrated that the arterial baroreflex is imparied with aging and have implicated central components of the baroreflex arc in this autonomic dysfunction. Neurons in the medial portion of the nucleus tractus solitarius (mNTS) receive a major input from the arterial baroreceptors. The present study was undertaken to characterize the intrinsic membrane properties of mNTS neurons in young rats and to test the hypothesis that these properties are altered with aging. An in vitro brain stem slice preparation was used to record intracellularly from mNTS neurons; passive membrane properties, action potential characteristics, and repetitive firing properties were examined and compared. 2. Neurons in the mNTS of young (3-5 mo old) Fischer-344 rats (F-344; n = 35) had a resting membrane potential of -57 +/- 6.9 mV (mean +/- SD), a membrane time constant of 18 +/- 9.0 ms, and an input resistance of 110 +/- 60 m omega. Action potential amplitude was 81 +/- 7.5 mV with a duration at half-height of 0.83 +/- 0.15 ms. The spontaneous firing rate in 24 cells was 4.3 +/- 2.9 Hz. The amplitude and duration of the action potential afterhyperpolarization (AHP) were 6.6 +/- 3.0 mV and 64 +/- 34 ms, respectively. All neurons expressed spike frequency adaptation, action potential AHP, and posttetanic hyperpolarization. Delayed excitation and postinhibitory rebound were present in 34 and 14% of neurons tested, respectively. Neurons from adult (10-12 mo old) F-344 rats (n = 34) were similar to the young F-344 rats with respect to all of these variables. 3. Neurons from aged (21-24 mo old) F-344 (n = 32) were similar to those from young and adult rats, but there were two potentially important differences: the mean input resistance of the aged neurons was higher (170 +/- 150 M omega), with a larger proportion (46% of aged neurons vs. 20% of young neurons and 21% of adult neurons) having input resistances > 150 M omega; and there was a tendency for a smaller percentage of aged neurons (16% of aged neurons vs. 34% of young neurons and 29% of adult neurons) to express delayed excitation. 4. The potential significance of a high input resistance was tested by comparing the steady-state current-voltage (I-V) relationships and the frequency-current (f-I) relationships among low-resistance (1-100 M omega), medium-resistance (101-200 M omega).(ABSTRACT TRUNCATED AT 400 WORDS)

Receiving the Nobel Prize in 1971 for the invention and development of the holographic method, Dennis Gabor mentioned Mieczysław Wolfke as the person who proposed this method as early as 1920 (which he did not know, independently making the same discovery). 21016 Background: 5-y surv in surgically resected St I NSCLC is 60–70% whereas in cases with affected lymph nodes (LN) it is < 50%. The main risk factor for recurrence is nodal disease. Current histopathologic analysis can miss occult micrometastases in nodal tissues at initial diagnosis. Detection of micromets with a more sensitive technique would be useful to define a high-risk population selecting patients (p) for postop treatment. We assessed the role of several genes mRNA expression in pathological negative LN from resected St I NSCLC p as markers of occult micromets and correlated the results with relapse, and survival. Methods: Paired tumor and histological negative LN (n=344) obtained by systematic mediastinal lymphadenectomy from 38 surgically resected St I NSCLC p were analyzed for the presence of 12 genes mRNA expression using RT-Q-PCR in an ABIPRISM 7500. RNA was extracted using ABIPRISM 6100. Specifically designed primers and probes were purchased from Applied Biosystems as Assay-on-demand; GADPH was used as an endogenous control. Samples were also analyzed by ICH for LN staging. Results: 38 NSCLC p; 12 adenoca, 16 squamous cell, 10 undifferentiated . From the 12 tested genes CEA and PLUNC were found with high expression in lung tissue and low or null expression in normal LN. We consider molecular + LN those in which expression of CEA or PLUNC was detected. In the 344 pathological negative LN, 13% (44/344) were positive for CEA, 16% (54/344) for PLUNC. The expression patterns were similar for both markers. At a median follow-up of 24 mo (9–46) 11 p had died from NSCLC and 1 had died without recurrence. None of the living p had tumor recurrence. For the prognostic assessment, molecular + LN were classified as N1 and N2. Median DFS was 15 ± 11.74 mo in p with N2 molecular + nodes was and has not yet reached in cases of molecular (-) LN (p=0.028). Median survival of p with N2 molecular positive nodes was 17.3 ± 5.7 mo and has not yet reached (p= 0.0083) for molecular (-) LN. Conclusions: CEA and PLUNC mRNA expression could be used as molecular markers of occult micrometastases in mediastinal LN showing a prognostic effect . CEA and PLUNC expression provides a tool for selecting high-risk p considered for adjuvant therapies No significant financial relationships to disclose.

For the individual tongue muscles, relatively little quantification of uniaxial force, fatigue, and fiber type-specific properties has been documented. Here, we assessed uniaxial-specific force generation, fatigability, and muscle fiber type-specific properties in the superior and inferior longitudinal muscles, the transversalis, and the genioglossus in Fischer 344 rats. The longitudinal muscles produced the greatest isometric tetanic-specific forces. The genioglossus was more fatigue resistant and comprised higher proportions of I and IIa fibers.

Recently, a set of proteins involved in the docking and fusion machinery of secretory organelles has been identified in anterior pituitary cells. In this study we analyzed, by Western blotting and immunocytochemistry, the expression of several proteins involved in exocytosis after long-term administration of 17beta-estradiol (E2) in Fischer 344 rats. No differences were observed in the amount of synaptosomal-associated protein of 25 kDa, synaptobrevin 2, syntaxin 1, synaptotagmin I and Rab3a in total brain homogenates from treated rats after E2 administration. In striking contrast, the levels of all of these exocytotic proteins, including cellubrevin, were notably decreased in pituitary glands of E2-treated rats. In addition, no differences were observed in the in vitro basal and 8-Br-cAMP-induced prolactin (PRL) release between pituitary cells from control and E2-treated rats, whereas TRH-induced PRL release in anterior pituitary cells from E2-treated animals was higher than in control donors. In conclusion, this study shows that protein components of the exocytotic machinery are specifically down-regulated in the pituitary gland of E2-treated Fischer 344 rats.

Abstract In the present paper we construct an example of a quaternion random variable such that Polya's type characterization theorem of Gaussian distributions does not hold. The matter is that in the linear form, consisting of the independent copies of quaternion random variables, a part of the quaternion coefficients is written on the right hand side and the other part on the left-hand side. This gives a negative answer to the question posed in [Vakhania and Chelidze, Teor. Veroyatnost. i Primenen. 54: 337–344, 2009].

Цель. Определить клинико-генетические факторы риска прогрессирования гипертрофии левого желудочка (ГЛЖ) с учетом полиморфизма генов ренин-ангиотензин-альдостероновой системы (РААС) по данным проспективного пятилетнего наблюдения пациентов с эссенциальной артериальной гипертензией (АГ). Материалы и методы. Клинико-инструментальное обследование выполнено у 155 пациентов с АГ, из них 82 мужчины и 73 женщины (средний возраст 54,5±9,4 года). Признаки ГЛЖ оценивались при эхокардиографии (ЭхоКГ) с расчетом индекса массы миокарда левого желудочка (ИММЛЖ). Генотипирование полиморфных локусов генов ренин-ангиотензин-альдостероновой системы (РААС), I/D ангиотензинпревращающего фермента (АСЕ), М235Т ангиотензиногена (AGT), G83A ренина (REN), C(-344)T альдостеронсинтазы (СYP11B2), A1166C рецепторов кангиотензину II 1-го типа (AGTR1) и 2-го типа C3123A (AGTR2) проводилось методом полимеразной цепной реакции. Результаты. Прогрессирование ГЛЖ по результатам повторной ЭхоКГ через 5 лет наблюдалось у 46 человек (29,7%), у 67 пациентов ИММЛЖ оставался в пределах нормативных значений и у 42 - отмечалось его снижение до нормативных величин. В многофакторной регрессионной модели прогнозирования предикторами прогрессирования ГЛЖ в течение 5 лет являлись: носительство мутантного ТТ генотипа гена AGT (ОШ=3,79; 95% ДИ 1,39-10,33; р=0,009), нерегулярное антигипертензивное лечение (ОШ=3,45; 95% ДИ 1,41-8,33; р=0,006), биаллельные сочетания полиморфизма A1166C гена AGTR1 (С аллель) и C(-344)Т) гена CYP11B2 (СТ/ТТ генотипы) (ОШ=1,53; 95% ДИ 1,05-2,23; р=0,028), более высокая степень АГ у носителей ААгенотипа гена AGTR2 (ОШ=1,58; 95% ДИ 1,07-2,33; р=0,022). Точность прогноза прогрессирования ГЛЖ у пациентов с АГ по разработанной многофакторной модели составила 73% при общей безошибочности модели 71%. Заключение. Прогрессирование ГЛЖ в течение 5 лет у пациентов с АГ обусловлено влиянием полиморфизма генов РААС - AGT (М235Т), AGTR1 (A1166C), CYP11B2 (C(-344)T), AGTR2 (С3123А) и их взаимодействиями в сочетании с нерегулярным лечением и степенью заболевания. Purpose. To determine the clinical and genetic risk factors of progression of left ventricular hypertrophy (LVH), taking into account the polymorphism of the genes of the renin-angiotensin- aldosterone system (RAAS), according to the data of a prospective five-year follow-up of patients with essential arterial hypertension (AH). Materials and methods. Clinical and instrumental examination was performed in 155 patients with hypertension, including 82 men and 73 women (average age - 54.5±9.4 years). The signs of LVH were assessed with the help of echocardiography with calculation of the left ventricular myocardial mass index (LVMI). Genotyping of polymorphic loci of the genes of the renin-angiotensin-aldosterone system (RAAS), I/D angiotensin converting enzyme (ACE), М235Т angiotensinogen (AGT), renin G83A (REN), C(-344)T aldosterone BC2 receptors) (CYP1166) II type 1 (AGTR1) and type 2 C3123A (AGTR2) was carried out with polymerase chain reaction. Results. The progression of LVH according to the results of repeated echocardiographic examination in 5 years was observed in 46 patients (29.7%); in 67 patients, LVMI remained within the normative values; in 42 patients, it decreased to the normative values. In the multivariate regression model, the predictors of LVH progression during 5 years were the following: mutant TT genotype of the AGT gene (OR=3.79; 95% CI 1.39-10.33; p=0.009), irregular antihypertensive treatment (OR=3.45; 95% CI 1.41-8.33; p=0.006), biallelic combinations of the polymorphism A1166C of the AGTR1 gene (C allele) and C (-344) T) of the CYP11B2 gene (CT/TT genotypes) (OR=1.53; 95% CI 1.05-2.23; p=0.028), a higher degree of AH in the carriers of the AA genotype of the AGTR2 gene (OR=1.58; 95% CI 1.07- 2.33; p=0.022). The accuracy of prediction of the LVH progression in patients with hypertension according to the multivariate model was 73%, while the infallibility was 71%. Conclusion. The progression of LVH within 5 years in patients with AH is caused by the influence of the polymorphism of the RAAS genes - AGT (M235T), AGTR1 (A1166C), CYP11B2 (C(-344)T), AGTR2 (C3123A) and their interactions in combination with irregular treatment and the degree of the disease.

Resumo:

Five peptides of general structure X-Ser-Pro-Thr-Ser-Pro-Ser-Y (X = Tyr, quinoxaline- 2-carbonyl, acridine-9-carbonyl, Y = Tyr, (quinoxalin-2-yl)amino) were prepared using standard solid-phase peptide synthesis technique. Their interaction with DNA (calf thymus DNA and plasmids pUC9, p∆NS and pGEMEX) was studied using UV and CD spectroscopy, sedimentation analysis and agarose gel electrophoresis after treatment with topoisomerase I. In contrast to earlier findings (Suzuki M.: Nature 1990, 344, 562) intercalation into DNA structure has not been proved for any compound studied.

Problem To determine if epidermal cell proliferation in colony-raised Fischer 344 rats changes with age and diet. Methods Fischer 344 rats fed ad libitum and calorie-restricted (CR) diets were obtained from the NIA colonies, and young, young adult, and old animals from both groups were used for this study (six in each group). Tissue sections from the dorsal skin (DS) and foot plate (FP) were used for immunohistochemical staining of proliferating cell nuclear antigen (PCNA). The proliferation index (PCNA-I) was computed from counts of stained and total number of keratinocytes. Simultaneous measurements of epidermal thickness were obtained from same sections. Data were analyzed with Excel and SPSS 14.0 software for statistics. Results Two-way analysis of variance (ANOVA) was applied to the data to probe the effect of age, diet, and age-diet interaction. A significant effect of age was noticed in the two parameters i.e., DS PCNA (F 3.96, P .011), FP epidermal width (F 3.37, P .021) and FP PCNA-I (F 9.0, P .000). A significant correlation between DS width and PCNA values was also noted (r 0.5, P .01). Conclusion There is a trend of reduction of PCNA positive cells with increasing age irrespective of thickness of epidermis, and this trend is more apparent in CR rats. Significance This cell proliferation study has implications in relation to CR effect on age-related disease conditions, and biogerontology. ABSTRACT Aims Ramadan is a great opportunity for scientific research due to its peculiar nature. This study was designed to investigate whether morning saliva cortisol pick can change during and after Ramadan compared to before it. Materials and methods A total of 30 healthy volunteer men (aged between 30 and 76 years) were asked to participate in a cross-sectional study between August 12 and September 10, 2010. Saliva was collected 2 weeks before the beginning of Ramadan (BR), during the first week (R1), middle (R2), the last week (R3) of Ramadan and 3 weeks after Ramadan (AR). Cortisol concentration was analyzed by ELISA. Statistical analysis of one-way repeated measures analysis of variance (ANOVA) was used. Results The mean unstimulated saliva cortisol concentration and its output were significantly higher in the BR than during and after Ramadan. Conclusion Mean saliva cortisol concentration and its output tended to be lower during and 3 weeks after Ramadan. Clinical significance Cortisol as a stress hormone seems to be low during Ramadan. Koorie High School is a Government school. It is not the school's real name but it is the name I will be using for this article.Koorie High School is an innercity comprehensive high school from Years 7-12 in Sydney. It has a total population of 344 students, 186 at the Intensive English Unit on a different site and 158 in the mainstream high school campus. Fifty-five percent of the 158 students are Aboriginal. There are also 32 students in the Back-To-School Program, an annex of the main high school. All of these students are Aboriginal. I will mainly be referring to the main high school and no reference will be made to the Intensive English Unit as they are not part of this study.
