The dimethyl sulfoxide hemisolvate of perindoprilat [systematic name: (1S)-2-((S)-{1-[(2S,3aS,7aS)-2-carboxyoctahydro-1H-indol-1-yl]-1-oxopropan-2-yl}azaniumyl)pentanoate dimethyl sulfoxide hemisolvate], C17H28N2O5·0.5C2H6OS, an active metabolite of perindopril, has been synthesized, structurally characterized by single-crystal X-ray diffraction and compared with its ethanol disolvate analogue [Pascardet al.(1991).J. Med. Chem.34, 663–669]. Both compounds crystallize in the orthorhombicP212121space group in the same zwitterionic form, with a protonated alanine N atom and an anionic carboxylate group at then-alkyl chain. The three structural units present in the unit cell (two zwitterions and the solvent molecule) are held together by a rich system of O—H...O, N—H...O and C—H...O hydrogen-bond contacts.

Anim. Sci. 91: 663–667. To better use distillers dried grains with solubles (DDGS) for non-ruminants, some of the fiber may be removed. The “elusieve” process removes approximately 10% of DDGS material, mostly fiber, yielding enhanced DDGS (E-DDGS). An experiment was conducted to compare the DE and ME in E-DDGS to those in DDGS when fed to growing and finishing pigs. Overall, E-DDGS has greater DE, ME and MEnthan DDGS.

Plant Sci. 95: 663–669. The antioxidant activities and the contents of total soluble phenolics, flavonoids, anthocyanins and proanthocyanidins were studied in wild and cultivated lingonberries (Vaccinium vitis-idaea L.). The antioxidant properties of wild clones were much higher than those of the cultivars. The levels of antioxidant compounds were positively correlated with latitude, altitude, reduced temperature and increased precipitation of the collected sites.

The mechanisms by which oscillatory shear stress (OS) induces, while high laminar shear stress (LS) prevents, atherosclerosis are still unclear. Here, we examined the hypothesis that OS induces inflammatory response, a critical atherogenic event, in endothelial cells by a microRNA (miRNA)-dependent mechanism. By miRNA microarray analysis using total RNA from human umbilical vein endothelial cells (HUVECs) that were exposed to OS or LS for 24 h, we identified 21 miRNAs that were differentially expressed. Of the 21 miRNAs, 13 were further examined by quantitative PCR, which validated the result for 10 miRNAs. Treatment of HUVECs with the miR-663 antagonist (miR-663-locked nucleic acids) blocked OS-induced monocyte adhesion, but not apoptosis. In contrast, overexpression of miR-663 increased monocyte adhesion in LS-exposed cells. Subsequent mRNA expression microarray study using HUVECs treated with miR-663-locked nucleic acids and OS revealed 32 up- and 3 downregulated genes, 6 of which are known to be involved in inflammatory response. In summary, we identified 10 OS-sensitive miRNAs, including miR-663, which plays a key role in OS-induced inflammatory responses by mediating the expression of inflammatory gene network in HUVECs. These OS-sensitive miRNAs may mediate atherosclerosis induced by disturbed flow.

Arbuscular mycorrhizal fungi (AMF) are important for plant nutrient uptake, and mycorrhizal dependence varies between wheat genotypes. Cultivar selection under fertilized conditions may have reduced mycorrhizal associations in modern cultivars. This study investigated AMF colonization, tissue P and grain yield of five modern (post 1990) and five older (pre 1970) spring wheat cultivars over 4 site-years in Manitoba. Older cultivars actually had significantly lower AMF colonization and grain yield than modern cultivars, but no tissue P concentration differences were observed. The two highest yielding lines were released after 2004, and one of these (FBC Dylan) was selected for suitability in organic farming systems.

The title compound, C5D6ClN2O+·Cl−, crystallizes in the orthorhombic space group, Pbcm, and consists of a 4-chloro-2-methyl-6-oxo-3,6-dihydropyrimidin-1-ium cation and a chloride anion where both moieties lie on a crystallographic mirror. The cation is disordered and was refined as two equivalent forms with occupancies of 0.750 (4)/0.250 (4), while the chloride anion is triply disordered with occupancies of 0.774 (12), 0.12 (2), and 0.11 (2). Unusually, the bond angles around the C=O unit range from 127.2 (6) to 115.2 (3)° and similar angles have been found in other structures containing a 6-oxo-3,6-dihydropyrimidin-1-ium cation, including the monclinic polymorph of the title compound, which crystallizes in the monoclinic space group P21/c [Kawai et al. (1973). Cryst. Struct. Comm. 2, 663–666]. The cations and anions pack into sheets in the ab plane linked by N—H...Cl hydrogen bonds as well as C—H...O and Cl...O interactions. In graph-set notation, these form R 3 3(11) and R 3 2(9) rings. Theoretical calculations seem to indicate that the reason for the unusual angles at the sp 2 C is the electrostatic interaction between the oxygen atom and the adjacent N—H hydrogen.

It crystallizes in the chiral orthorhombic space groupP212121, the same as the previously reported ethanol disolvate [Pascard, Guilhem, Vincent, Remond, Portevin & Laubie (1991).J. Med. Chem.34, 663–669] and dimethyl sulfoxide hemisolvate [Bojarska, Maniukiewicz, Sieroń, Fruziński, Kopczacki, Walczyński & Remko (2012).Acta Cryst.C68, o341–o343]. The asymmetric unit of (I)·H2O contains one independent perindoprilat zwitterion and one water molecule. These interactviastrong hydrogen bonds to give a cyclicR22(7) synthon, which provides a rigid molecular conformation. The geometric parameters of all three forms are similar. The conformations of the perhydroindole group are almost identical, but then-alkyl chain has conformational freedom. A three-dimensional hydrogen-bonding network of O—H...O and N—H...O interactions is observed in the crystal structure of (I)·H2O, similar to the other two solvates, but because of the presence of different solvents the three crystal structures have diverse packing motifs. All three solvatomorphs are additionally stabilized by nonclassical weak C—H...O contacts.

A module M is said to be H-supplemented if, for any submodule X of M, there exists a direct summand M′ of M such that M = X + Y if and only if M = M′ + Y for all Y ⊆ M (cf. [S. H. Mohamed and B. J. Müller, Continuous and Discrete Modules, London Mathematical Society Lecture Note Series, Vol. 147 (Cambridge University Press, 1999)]). We say that a module M is semi-lifting if any direct summand of M is H-supplemented. A H-supplemented module is a dual to a Goldie-extending module which was introduced by Akalan–Birkenmeier–Tercan [Goldie extending modules, Comm. Algebra37 (2009) 663–683]. In this paper, we give some characterizations of semi-lifting modules and H-supplemented modules. In addition, we consider generalizations of relative projectivities and apply them to the study of direct sums of semi-lifting modules.

This study investigated miRNA and cytokine expression changes in peritoneal fluid samples of patients with advanced ovarian cancer (OVCA) after receiving hyperthermic intraperitoneal chemotherapy (HIPEC) during cytoreduction surgery (CRS). We collected samples prior to HIPEC, immediately after HIPEC, and 24/48/72 h after CRS from a total of 6 patients. Cytokine levels were assessed using a multiplex cytokine array, and a miRNA PanelChip Analysis System was used for miRNA detection. Following HIPEC, miR-320a-3p, and miR-663-a were found to be immediately down-regulated but increased after 24 h. Further, significant upregulation post-HIPEC and sustained increases in expression were detected in six other miRNAs, including miR-1290, miR-1972, miR-1254, miR-483-5p, miR-574-3p, and miR-574-5p. We also found significantly increased expression of cytokines, including MCP-1, IL-6, IL-6sR, TIMP-1, RANTES, and G-CSF. The changing expression pattern throughout the study duration included a negative correlation in miR-320a-3p and miR-663-a to cytokines including RANTES, TIMP-1, and IL-6 but a positive correlation in miRNAs to cytokines including MCP-1, IL-6sR, and G-CSF. Our study found miRNAs and cytokines in the peritoneal fluid of OVCA patients demonstrated different expression characteristics following CRS and HIPEC. Both changes in expression demonstrated correlations, but the role of HIPEC remains unknown, prompting the need for research in the future.

Dietary fucoxanthin has been reported to exert several physiological functions, and fucoxanthinol is considered to be the primary active metabolite of fucoxanthin. However, there is no information about the pharmacokinetics of fucoxanthinol in human subjects. In the present study, eighteen human volunteers were orally administered kombu extract containing 31 mg fucoxanthin, and their peripheral blood was collected 5 min before and 0·5, 1, 2, 4, 8 and 24 h after the treatment. Plasma fucoxanthinol concentrations were measured by HPLC, and the pharmacokinetics of fucoxanthinol were as follows: maximum concentration, 44·2 nmol/l; time at maximum concentration, 4 h; terminal half-time, 7·0 h; area under the curve (AUC) for 1–24 h, 578·7 nmol/l × h; AUC(∞), 663·7 nmol/l × h. In addition to fucoxanthinol, we also attempted to detect amarouciaxanthin A, a hepatic metabolite of fucoxanthinol, using HPLC, but it was not present in the volunteers' plasma. On the other hand, a peak that was suspected to represent the cis-isomer of fucoxanthinol was found in the HPLC chromatogram. By comparing the present results with those of a previous study using mice, we found that the bioavailability and metabolism of fucoxanthinol differ between human subjects and mice.

663 Background: Late-Stage Pancreatic Cancer (LS-PC) represents the 7th leading cause of cancer-related deaths worldwide. Hispanic (H) ethnicity is underrepresented in genomic studies and disparities have been demonstrated in overall treatment and outcomes. Germline, somatic tissue, and somatic liquid testing are effective tools that aid in identifying targeted genetic alterations. We comprehensively evaluated the differences in genomic testing among H and Non-Hispanic (NH) patients with LS-PC. Methods: This is a retrospective analysis of LS-PC with ductal adenocarcinoma from the University of Arizona Cancer Center from 2013 to 2021. Data collected included: patient demographics, clinical characteristics, treatment regimens, germline, somatic tissue, and somatic liquid genetic testing, and outcomes. LS was defined as unresectable and/or metastatic de novo or recurrence. Univariate comparisons used parametric and nonparametric tests as appropriate to evaluate for differences between H and NH groups. Fisher's exact tests were performed to evaluate the difference in frequency. Kaplan Meier and Cox regression analysis assessed progression-free and overall survival. Results: The analysis included 198 patients who had LS-PC with 20.7% H. 64.1%, 48.9%, and 48.9% of the patients had germline, somatic tissue, and somatic liquid testing, respectively. No significant differences were noted in baseline characteristics, treatments, and outcomes [PFS (HR 1.1, 95% CI 0.7-1.6, p=0.70) / OS (HR 1.2, 95% CI 0.7-1.9, p= 0.54)]. There was no statistically significant difference in common mutations or variants among groups. Conclusions: To our knowledge, this is the first and largest study to report LS-PC genomic and therapeutic implications in H patients. H were significantly less likely to receive germline screening and more likely to have potentially actionable pathogenic variants via somatic tissue and liquid biopsy testing. This highlights the importance of genetic testing to reveal mutations that can be targeted by clinical trials for LS-PC on minorities. Although our review revealed increased germline, somatic tissue, and somatic liquid biopsy genetic testing over time, there remains a critical need towards advancing genomic sequencing efforts to improve outcomes for LS-PC in the underrepresented H population.[Table: see text]

Different methods for the estimation of whole body protein synthesis (PS) in healthy adult humans were simultaneously compared in seven young adult subjects (6 males, 1 female) fed for 6 days a diet providing 1 g protein.kg-1.day-1 and approximately 188 kJ.kg-1.day-1. A 24-h intravenous tracer study with L-[1-13C]leucine was performed starting at 6 P.M. on day 6. During fasted (6 h) and fed (6 h) steady states, PS was estimated using an approach based on 13CO2 excretion (PSexcr) and on urinary nitrogen excretion data (corrected for changes in body urea pool). Simultaneously, we used the conventional two-pool model and plasma [13C]ketoisocaproate enrichment for estimating PS. The latter mean estimates of PS were significantly higher than PSexcr during fasting [861 +/- 58 (SD) vs. 663 +/- 160 mg protein.kg-1.6 h-1; P < 0.01] and feeding (985 +/- 63 vs. 779 +/- 127 mg protein.kg-1.6 h-1; P < 0.01) and were much less variable. In hourly small-meal feeding, urinary nitrogen excretion was not a reliable index of body protein oxidation when measured over short periods of 6 h, thereby introducing a lack of precision in PSexcr. We suggest that application of the 13CO2 technique to measure PS in humans is limited by the need for relatively prolonged experimental periods of urine collection and tracer infusion within a given physiological state.

ABSTRACT Background Driving is a common type of sedentary behaviour; an independent risk factor for poor health. The study explores whether driving is also associated with other unhealthy lifestyle factors. Methods In a cross-sectional study of UK Biobank participants, driving time was treated as an ordinal variable and other lifestyle factors dichotomized into low/high risk based on guidelines. The associations were explored using chi-square tests for trend and binary logistic regression. Results Of the 386 493 participants who drove, 153 717 (39.8%) drove &lt;1 h/day; 140 140 (36.3%) 1 h/day; 60 973 (15.8%) 2 h/day; and 31 663 (8.2%) ≥3 h/day. Following adjustment for potential confounders, driving ≥3 h/day was associated with being overweight/obese (OR = 1.74, 95% CI: 1.64–1.85), smoking (OR = 1.48, 95% CI: 1.37–1.63), insufficient sleep (1.70, 95% CI: 1.61–1.80), low fruit/vegetable intake (OR = 1.26, 95% CI: 1.18–1.35) and low physical activity (OR = 1.05, 95% CI: 1.00–1.11), with dose relationships for the first three, but was not associated with higher alcohol consumption (OR = 0.94, 95% CI: 0.87–1.02). Conclusions Sedentary behaviour, such as driving, is known to have an independent association with adverse health outcomes. It may have additional impact mediated through its effect on other aspects of lifestyle. People with long driving times are at higher risk and might benefit from targeted interventions.

The Activated Clotting Time (ACT) assumes an important role in documentation of adequacy of perfusion. Therefore, it is important to document the correlation and reproducibility of automated systems currently available to measure the ACT value. During surgical procedures of 37 patients requiring cardiopulmonary bypass a blood sample was drawn in a single syringe and ACT values obtained using four Hemotec ACTs (T-ACT), four Hemochron ACTs (M-ACT), and two Sonoclot ACTs (S-ACT). Sample points with the mean of two of the three sets of ACTs exceeding 650 were excluded and the values were divided into heparinized (ACTH) (n=44) and nonheparinized (ACTN) (n=58) groups. SummaryAim: The number of therapies with radiopharmaceuticals labelled with 188Re is increasing requiring the documentation of the beta radiation exposure Hp(0.07) of the staff at all working and production sites and during the application and follow-up of the patient according to the new German Radiation Protection Law (StrlSchV). However, data for β-radiation exposure are rare. Therefore, we determined the personal dose Hp(0.07) of the skin of the hands handling 188Re radiopharmaceuticals to identify steps of high radiation exposure and to optimize working conditions. Method: Thermoluminescence dosimeters (TLD 100) were fixed to the fingertips of the radiochemist, the physician and the nurse and compared to official ring dosimeters. In addition, to monitor radiation exposure continuously readable electronic beta- and gamma dosimeters EPD (Siemens) were used. At eight days in which therapies were performed these readings were evaluated. Results: Considering one therapy with a 188Re-labelled radiopharmaceutical the middle finger of the radiochemist (production) and the physician (application) showed a radiation burden of 894 and 664 μSv/GBq, respectively. The cumulative dose of the fingertips after eight days of therapy was 249 and 110 mSv for the radiochemist and physician, respectively. A cumulative finger dose after eight days of therapy of 17 and 38 μSv/GBq was found for physician and nurse leading to a Hp(0.07) of 3 and 6 mSv, respectively. Preparing the radiopharmaceutical labelled with 20GBq of 188Re the reading of the personal electronic dosimeter of the radiochemist showed a γ-dose rate H . p(10) of 55 μSv/h and a β-dose rate H . p(0.07) of 663 μSv/h which are obviously not representative for the true radiation dose to the skin of the fingertips. Conclusion: During therapy with 188Re-labelled radiopharmaceuticals the true radiation dose to the skin of the finger tips exceeds by far the readings of the official ring dosimeters as well as the continuously readable beta- and gamma dosimeters. This means a risk in exceeding the radiation limit of 500 mSv/a given in the German Radiation Protection Law (§55 StrSchV) primarily in the working field of the radiochemist and the administering physician.

Formula uniform design method was adopted to study the performance of iron-based matrix with high content of phosphorus and boron. Two forms of regression models on matrix hardness and wear resistance and the optimum matrix formulae were obtained respectively by regression analysis and constrained nonlinear programming using MATLAB. Regression analyses indicate that matrix hardness was mainly affected by the factor of Ni, Co and Fe; matrix hardness and brittleness increased with the increment of P-Fe alloy, but decreased with the increase of 663-Cu. These results can be used as a rough guide for design of highly adaptive diamond bits for different rock formations. After the indoor experiments, diamond bit based on 3# matrix formula was made to perform field drilling tests. The results were that the penetration rate was 1.91m/h and the average diamond bit life was 67.37m.

Ionizing radiation causes serious injury to the human body and has long-time impacts on health. It is important to find optimal biomarkers for the early quick screening of exposed individuals. A series of miRNAs signatures have been developed as the new biomarkers for diagnosis, survival, and prognostic prediction of cancers. Here, we have identified the ionizing radiation-inducible miRNAs profile through microarray analysis. The biological functions were predicted for the top six upregulated miRNAs by 4 Gy γ-rays: miR-1246, miR-1307-3p, miR-3197, miR-4267, miR-5096 and miR-7641. The miRNA-gene network and target gene-pathway network analyses revealed that DNAH3 is the target gene associated with all the six miRNAs. GOLGB1 is related to 4 miRNAs and other 26 genes targeted by 3 miRNAs. The upregulation of fifteen miRNAs were further verified at 4 h and 24 h after 0 to 10 Gy irradiation in the human lymphoblastoid AHH-1 cells, and some demonstrated a dose-dependent increased. Six miRNAs, including miR-145, miR-663, miR-1273g-3p, miR-6090, miR-6727-5p and miR-7641, were validated to be dose-dependently upregulated at 4 h or 24 h post-irradiation in both AHH-1 and human peripheral blood lymphocytes irradiated ex vivo. This six-miRNA signature displays the superiority as a radiation biomarker for the translational application of screening and assessment of radiation exposed individuals.

In this work we have obtained exact analytical formulae expressing the wave resistance of a two-dimensional body by the parameters of the downstream non-breaking waves. The body moves horizontally at a constant speed $c$ in a channel of finite depth $h$. We have analysed the relationships between the parameters of the upstream flow and the downstream waves. Making use of some results by Keady & Norbury (J. Fluid Mech., vol. 70, 1975, pp. 663–671) and Benjamin (J. Fluid Mech., vol. 295, 1995, pp. 337–356), we have rigorously proved that realistic steady free-surface flows with a positive wave resistance exist only if the upstream flow is subcritical, i.e. the Froude number $\mathit{Fr}=c/\sqrt{gh}<1$. For all solutions with downstream waves obtained by a perturbation of a supercritical upstream uniform flow the wave resistance is negative. Applying a numerical technique, we have calculated accurate values of the wave resistance depending on the wavelength, amplitude and mean depth.

A doença arterial obstrutiva periférica (DAOP) afeta 20% da população idosa e aumenta o risco de doenças cardiovasculares. O sintoma mais frequente da DAOP é a claudicação intermitente, que depende da discrepância entre suprimento e demanda de oxigênio exigidos pelos músculos durante caminhada. O objetivo deste relato de caso foi avaliar a eficácia de um programa de condicionamento em esteira ergométrica sobre a tolerância à caminhada (TC) de um indivíduo com DAOP. Indivíduo WCF, sexo masculino, 74 anos, apresentava obstrução total de artéria superficial da coxa e 95% de obstrução arterial em poplítea e tibial anterior, todas em membro inferior direito, identificadas por arteriografia. Foi submetido a um Teste de Tolerância Máxima à Caminhada (TTMC) em esteira ergométrica a 2,0 km/h e posteriormente a um programa de condicionamento realizado três vezes por semana em sete séries de 80% do TTMC, com um minuto de descanso, a 2,0 km/h durante quatro meses. Após esse período realizou-se novo TTMC. Foi observado que a TC aumentou 663%, ocorrendo também diminuição do número de paradas para descanso nos percursos habituais. Os resultados sugerem que o treinamento em esteira foi eficiente na melhora da TC nesse indivíduo.Palavras-chave: claudicação intermitente, reabilitação, terapia por exercício.

In the quality monitoring of 18 sunflower oil samples from the EU market, 14 were refined and 4 were cold-pressed. They demonstrated high quality of technological processing with low values of trans-unsaturated fatty acids, acid value, and peroxide value and also met the limits set by legislation in the content of process contaminants 3-monochloropropane-1,2-diol (3-MCPD) esters and glycidyl esters. Measurements of oxidative stability showed a difference in utility value. The average induction period of the oils from the traditional varieties was 2.6 h, predisposing them to cold cooking or short-term frying, while the 11.8 h of the four high oleic sunflower oils (HOSO) indicates the possibility of long-term heat stress. The nutritional benefit is the average vitamin E content of 663 mg/kg oil. The overall sensory quality of the samples was evaluated by a 12-member panel of trained assessors. On the seven-point category scale, the oils were of good to exceptional quality. The cold-pressed oils (CPOs) differed in having, on average, lower trans-unsaturated fatty acid content, process contaminants at unmeasurable levels, and, on average, higher vitamin E concentrations. The specific organoleptic properties of the CPOs were characterized by a pleasant nutty and sunflower seed flavor.

The short-term consumption of foods that differed in energy density (ED) and/or NaHCO3 supplementation, on subsequent food intake and diet selection in sheep were measured. Thirty sheep weighing 35·9 (SD 2·89) KG WERE USED. TWO FOODS WERE FORMULATED: H HAD 11 AND L HAD 8 MJ METABOLIZABLE ENERGY/KG FRESH MATTER. FOUR FURTHER FOODS WERE FORMULATED BY ADDING EITHER 40 G NAHCO3/KG OR 16·5 G NACL/KG TO FOODS H AND L. NACL WAS ADDED TO GIVE THE SAME NA CONCENTRATION AS WITH 40 G NAHCO3/KG TO CONTROL FOR ANY EFFECTS OF NA PER SE. IN A PRELIMINARY TEST, IT WAS FOUND THAT A 2 H CONSUMPTION OF FOOD H SUPPLEMENTED WITH NAHCO3 COULD BUFFER POTENTIAL IMPACT ON THE RUMEN ENVIRONMENT OF SUBSEQUENT CONSUMPTION OF FOOD H ALONE (AS JUDGED BY RUMEN PH AND ACID-BUFFERING CAPACITY); HOWEVER, IT WAS NOT AS EFFECTIVE AS THE CONSUMPTION OF FOOD L ALONE IN DOING SO. EACH FOOD TREATMENT WAS OFFERED TO ONE OF SIX GROUPS (N 5) FOR 2 H FOLLOWING 16 H OF FOOD DEPRIVATION. SHEEP WERE THEN OFFERED A CHOICE BETWEEN H AND L FOR A FURTHER 6 H. SUPPLEMENTING H OR L WITH EITHER NAHCO3 OR NACL HAD NO SIGNIFICANT EFFECT ON EITHER INTAKE OR DIET SELECTION. ED SIGNIFICANTLY (P<0·01) AFFECTED INTAKE DURING THE 2 H SINGLE FEEDING PERIOD, WITH SHEEP OFFERED H OR L CONSUMING 540 AND 663 (sed 37) g respectively, but had no effect on subsequent intake during the choice period. During the choice period all sheep showed a preference for food H, but sheep previously offered L selected significantly more H (0·873 g/g) than sheep previously offered H (0·544 (sed 0·028) g/g; P<0·001). It is concluded that short-term consumption of foods that differ in ED, and hence in their potential impact on the rumen environment, significantly affects subsequent diet selection. This is in agreement with the hypothesis that ruminant animals select a diet to help maintain the rumen environment within a certain physiological range. Food H with 40 g NaHCO3 added/kg may not have been sufficient to affect subsequent diet selection. It is suggested that larger, rather than smaller, changes in the rumen environment achieved through previous feeding should be expected to alter subsequent diet selection.

CoO nanocrystal is well-known photocatalyst for overall water splitting. However it suffers from a very short lifetime of only 1 h. The poor stability is derived from carrier recombination-induced thermal oxidation. This research will provide information about synthesis of CoO/ZnO nanocrystalline that can potentially enhance photocatalysts. CoO has been synthesized first under hydrothermal method, followed by calcination process. Thereafter, CoO has been used to produce CoO/ZnO under precipitation method. The samples were characterized using XRD (X-Ray Diffraction), FTIR (Fourier Transform Infrared Spectroscopy), and UV-VIS (UV–Visible Spectroscopy) to analyze their composition, chemical functional group, optical absorption, and band gap. The XRD spectrum showed that CoO/ZnO had cubic spinel and hexagonal phase structure with crystallite size of 69.0, 46.4, 32.8, and 32,4 nm. The bands in obtained FTIR spectrum at 586.36, 671.23, and 410-429 cm−1 were correlated with vibrations of the Co3+ in octahedral hole, the Co2+ in tetrahedral hole, and Zn-O, respectively. The band gap energy of CoO, CoO/ZnO with variation of 1:1 and 1:3 were 4.39, 4.14, and 3.65 eV, respectively. The photocatalytic activities of CoO/ZnO were confirmed by methylene blue dyes photodegradation of 663 nm under UV light irradiation in aqueous solution. The 22.4% methylene blue can be removed within 3 h. Overall, these findings reveal the potential of CoO/ZnO for practical application.

ABSTRACTOrientia tsutsugamushiis the etiological agent of scrub typhus, a mite-borne, febrile illness that occurs in the Asia-Pacific region. We conducted strain characterization ofO. tsutsugamushiisolates from chiggers obtained from rodents based the nucleotide sequence of the 56-kDa outer membrane protein gene. With the use of PCR, a total of 68 DNA sequences of 56-kDa antigen genes were amplified. Phylogenetic analysis revealed that there were at least six definable clusters among the 68 isolates: 37% Karp-related strains (25/68), 27% TA763 strains (18/68), 12% JG-related strains (8/68), 19% Kato-related strains (13/68), 4% divergent strains (3/68), and 1% representing a Gilliam prototype strain (1/68). Overall, theO. tsutsugamushigenotypes exhibited a high degree of diversity, similar to that seen in strains from the rest of the areas where scrub typhus is endemic. Moreover, the 56-kDa protein sequence similarity betweenO. tsutsugamushiisolates from mites and those from human patients (H. Y. Lu et al., Am. J. Trop. Med. Hyg. 83:658-663, 2010) were striking, thus highlighting potential risk factors for this emerging zoonotic disease.

Abstract Editor’s Perspective What We Already Know about This Topic What This Article Tells Us That Is New Background For cardiac surgery patients under chronic β-blocker therapy, guidelines recommend their early postoperative reintroduction to decrease the incidence of postoperative atrial fibrillation. The authors hypothesized that the timing of β-blocker reintroduction affects their effectiveness on the incidence of postoperative atrial fibrillation. Methods This multicenter prospective French cohort study included patients on β-blockers (more than 30 days before surgery) in sinus rhythm without a pacemaker. The primary outcome, time sequence of β-blocker reintroduction, was analyzed for 192 h after surgery. The secondary outcome, relationship between the occurrence of postoperative atrial fibrillation and timing of β-blocker reintroduction, was analyzed based on pre- and intraoperative predictors (full and selected sets) according to landmark times (patients in whom atrial fibrillation occurred before a given landmark time were not analyzed). Results Of 663 patients, β-blockers were reintroduced for 532 (80%) but for only 261 (39%) patients in the first 48 h after surgery. Median duration before reintroduction was 49.5 h (95% CI, 48 to 51.5 h). Postoperative atrial fibrillation or death (N = 4) occurred in 290 (44%) patients. After performing a landmark analysis to take into account the timing of β-blocker reintroduction, the adjusted odds ratios (95% CI) for predictor full and selected (increased age, history of paroxysmal atrial fibrillation, and duration of aortic cross clamping) sets for the occurrence of postoperative atrial fibrillation were: adjusted odds ratio (full) = 0.87 (0.58 to 1.32; P = 0.517) and adjusted odds ratio (selected) = 0.84 (0.58 to 1.21; P = 0.338) at 48 h; adjusted odds ratio (full) = 0.64 (0.39 to 1.05; P = 0.076) and adjusted odds ratio (selected) = 0.58 (0.38 to 0.89; P = 0.013) at 72 h; adjusted odds ratio (full) = 0.58 (0.31 to 1.07; P = 0.079) and adjusted odds ratio (selected) = 0.53 (0.31 to 0.91; P = 0.021) at 96 h. Conclusions β-Blockers were reintroduced early (after less than 48 h) in fewer than half of the cardiac surgery patients. Reintroduction decreased postoperative atrial fibrillation occurrence only at later time points and only in the predictor selected set model. These results are an incentive to optimize (timing, doses, or titration) β-blocker reintroduction after cardiac surgery.

Untreated wheat straw (UWS) or ammoniated wheat straw without (AWS) or with (AWSP) a supplement of potato protein of a low rumen degradability was fed to three steers according to a 3 × 3 Latin square design. All rations were supplemented with sugar-beet pulp and minerals. Voluntary organic matter intake (OMI, g/kg0.75 per d) was 67.8, 76.0 and 80.1 for whole rations (51.1, 59.7 and 59.2 for straw) for UWS, AWS and AWSP respectively, which was significantly higher for AWS and AWSP than for UWS. Organic matter digestibility (OMD, g/kg) was 561, 596 and 625 for the respective rations UWS, AWS and AWSP, also significantly higher for AWS and AWSP than for UWS. The increased voluntary intake and digestion of ammoniated wheat-straw-based rations were associated with a significantly higher potentially degradable fraction (D) of neutral detergent fibre (NDF) in offered straw (556 and 661 g/kg for untreated and ammoniated wheat straw respectively) and in the rumen pool (469, 555 and 554 g/kg for UWS, AWS and AWSP respectively). Isolated small rumen particles (retained on sieves with a pore size < 1.25 and > 0.041 mm) had a significantly lower D of NDF (average 588 g/kg) than isolated large rumen particles (average 663 g/kg). Fractional rates of degradation of NDF did not differ significantly either between untreated and ammonia-treated wheat straw offered (2.9 and 2.6%/h respectively) or between rumen pools (1.8, 1.7 and 2.1 %/h for UWS, AWS and AWSP respectively). Rations based on ammoniated wheat straw had a significantly higher rumen NH3-N concentration than UWS. Although the rumen pool size of total contents differed significantly between treatments, those of dry and organic matter and of cell wall constituents were not significantly different. The proportion of rumen dry matter passing through a sieve with a pore size of 1.25 mm averaged 0.684 over rations (not significantly different between rations). Daily rumination (96 min) and eating (52 min) times/kg NDF ingested did not differ between rations. The rate of comminution of large particles estimated from the disappearance of indigestible NDF in large rumen particles from the rumen of animals without access to feed was 4.1, 6.3 and 7.1 %/h for UWS, AWS and AWSP respectively. These values were not significantly different. The fractional rate of passage estimated from the faecal excretion of Cr-NDF was 5.4, 6.1 and 6.3%/h for UWS, AWS and AWSP respectively (significantly higher for AWS and AWSP than for UWS) but the turnover rate of indigestible NDF did not differ between treatments.

Early warning scores (EWSs) help prevent and recognize and thereby act as the first signs of clinical and physiological deterioration. The objective of this study is to evaluate different EWSs (National Early Warning Score 2 (NEWS2), quick sequential organ failure assessment score (qSOFA), Modified Rapid Emergency Medicine Score (MREMS) and Rapid Acute Physiology Score (RAPS)) to predict mortality within the first 48 h in patients suspected to have Coronavirus disease 2019 (COVID-19). We conducted a retrospective observational study in patients over 18 years of age who were treated by the advanced life support units and transferred to the emergency departments between March and July of 2020. Each patient was followed for two days registering their final diagnosis and mortality data. A total of 663 patients were included in our study. Early mortality within the first 48 h affected 53 patients (8.3%). The scale with the best capacity to predict early mortality was the National Early Warning Score 2 (NEWS2), with an area under the curve of 0.825 (95% CI: 0.75–0.89). The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) positive patients presented an area under the curve (AUC) of 0.804 (95% CI: 0.71–0.89), and the negative ones with an AUC of 0.863 (95% CI: 0.76–0.95). Among the EWSs, NEWS2 presented the best predictive power, even when it was separately applied to patients who tested positive and negative for SARS-CoV-2.

ABSTRACT Evidence of triggered star formation at large spatial scales involving stellar clusters is scarce. We investigate a Galactic region (l = 130${_{.}^{\circ}}$0, b = 0${_{.}^{\circ}}$35) populated by several open stellar clusters that according to the last Gaia data release, are located at a distance of about 2.9 kpc. By analysing the interstellar medium (ISM) at infrared, centimeter, and millimeter wavelengths towards this group of clusters we discovered a shell of material of about 2° in size at the same distance. We suggest that the shell, mainly observed at 12 μm and in the H i emission at 21 cm, was generated by the action of massive stars belonging to clusters Berkeley 7 and UBC 414, which lie at its centre. Five clusters (MWSC0152, Czernik 6, Czernik 7, Berkeley 6, NGC 663, and NGC 654) lie at the border of this shell. From the comparison between the dynamical time of the discovered H i shell and the analysis of the ages of stellar populations in these clusters, we conclude that the expansion of the shell could have triggered in the past the formation of stars in some of them. We point out that in order to find physical evidence supporting a genetic connection between stellar clusters, it is necessary not only to study the individual clusters and their stellar populations, but also to investigate their surrounding ISM at a large spatial scale.

AbstractWe present CCD photometry in the Washington-system C and T1 passbands down to T1 ~ 22.5 mag in the fields of NGC 1697, SL 133, NGC 1997, SL 663, and OHSC 28, five mostly unstudied star clusters in the LMC. Cluster radii were estimated from star counts in appropriately sized boxes distributed throughout the entire observed fields. We perform a detailed analysis of field-star contamination and derive cluster colour–magnitude diagrams (CMDs). Based on the best fits of isochrones computed by the Padova group to the (C − T1, T1) CMDs, the δ(T1) index and the ‘standard giant-branch’ procedure, we derive metallicities and ages for the five clusters. With the exception of NGC 1697 (age = 0.7 Gyr, [Fe/H] = 0.0 dex), the remaining four clusters are of intermediate age (from 2.2 to 3.0 Gyr) and relatively metal poor ([Fe/H] = −0.7 dex). We combine our sample with clusters with ages and metallicities on a similar scale and examine relationships between position in the LMC, age and metallicity. We confirm previous results that clusters younger than ~ 1 Gyr were formed during an outside-in process; this occurred after a burst of cluster formation that took place mainly in the outer disk and peaked ~ 2 Gyr ago. Finally, the cluster and field age–metallicity relations (AMRs) show evidence for a metallicity offset but do overlap, particularly on the upper-envelope side of the cluster AMR.

AbstractThe objective of this work was to determine the rumen fermentation characteristics of maize land races used as forage in central Mexico. In vitro gas production (ml per 200 mg dry matter (DM)) incubations were carried out, and cumulative gas volumes were fitted to the Krishnamoorthyet al.(1991) model. The trial used a split-plot design with cultivation practices associated with maize colour (COL) as the main plot with three levels: white, yellow and black maize; growing periods (PER) were the split plots where PER1, PER2 and PER3 represented the first, second and third periods, respectively and two contrasting zones (Z1=valley and Z2=mountain) were used as blocking factors. The principal effects observed were associated with the maturity of the plants and potential gas production increased (P<0·05) in stems (PER1=51·8, PER2=56·3, PER3=58·4 ml per 200 mg DM) and in whole plant (PER1=60·9, PER2=60·8, PER3=70·9 ml per 200 mg DM). An inverse effect was observed with fermentation rates in leaves (P<0·01) with 0·061, 0·053 and 0·0509 (per h) and in whole plant (P<0·05) with 0·068, 0·057, 0·050 (per h) in PER1, PER2 and PER3 respectively. The digestibility of the neutral-detergent fibre (NDF) decreased with maturity especially in leaves (P<0·05) with values of 0·71, 0·67 and 0·66 g/kg; in rachis (P<0·01) 0·75, 0·72, and 0·65 in PER1, PER2 and PER3 respectively. The NDF content in leaves in leaves (668, 705 and 713 g/kg DM for PER1, PER2 and PER3, respectively), stems (580, 594 and 644 g/kg DM) and, husk (663, 774 and, 808 g/kg DM) increased (P<0·05) with increasing plant maturity, rachis were significantly different between periods (P<0·01). The structure with the best nutritive characteristics was the husk, because it had the lowest fibre contents, especially in acid-detergent lignin, with values of 22·6, 28·6 and 37·6 g/kg DM in PER1, PER2 and PER3, respectively.

Eugenol, used as bio-phenol, was designed to replace the hydrogen atom of hydrogenterminated siloxane by hydrosilylation reaction under the presence of alumina-supported platinum catalyst (Pt-Al2O3), silica-supported platinum catalyst (Pt-SiO2) and carbon nanotube-supported platinum catalyst (Pt-CNT), respectively. The catalytic activities of these three platinum catalysts were measured by nuclear magnetic resonance hydrogen spectrometer (1H NMR). The properties of bio-phenol siloxane were characterized by Fourier transform infrared spectrometer (FT–IR), UV-visible spectrophotometer (UV) and thermogravimeter (TGA), and its antibacterial property against Escherichia coli was also studied. The results showed that the catalytic activity of the catalyst Pt-CNT was preferable. When the catalyst concentration was 100 ppm, the reaction temperature was 80 °C and reaction time was 6 h, the reactant conversion rate reached 97%. After modification with bio-phenol, the thermal stability of the obtained bio-phenol siloxane was improved. For bio-phenol siloxane, when the ratio of weight loss reached 98%, the pyrolysis temperature was raised to 663 °C which was 60 °C higher than hydrogenterminated siloxane. Meanwhile, its autonomic antibacterial property against Escherichia coli was improved significantly.

The aim of this research is to analyze open government bibliometrics in the education sector. Education is one of the most popular fields in open government. This research uses a qualitative approach with bibliometric analysis methods. The data used is secondary, obtained from the Scopus database. Data was collected using Publish or Perish with the keyword education sector open government. The data obtained was then selected using OpenRefine and analyzed using VOSviewer to obtain a bibliometric map generated from keywords throughout the paper. Based on the results of this research, there have been 160 papers in the last 51 years. The first open government paper on the education sector was published in 1972. There are 17634 citations, 75 h-index, and 126 g-index, which shows this paper has high impact and productivity. In this paper, there are 153 keywords grouped into 13 clusters. From these keywords, 1878 total link strengths and 663 occurrences were obtained. The keywords that dominate this paper are open policy making, social auditing, open contracting, and open budgeting. This shows that open government studies in the education sector focus on these issues.

The study determined in vitro enteric methane (CH4) emission potential of whole-plant cereal (WPC) forages in relationship to nutrient composition, degradability, and rumen fermentation. Two varieties of each WPC (barley, oat, triticale, and wheat) were harvested from two field replications in each of two locations in central Alberta, Canada, and an in vitro batch culture technique was used to characterize gas production (GP), fermentation, and degradability. Starch concentration (g/kg dry matter (DM)) was least (p < 0.001) for oat (147), greatest for wheat (274) and barley (229), and intermediate for triticale (194). The aNDF concentration was greater for oat versus the other cereals (531 vs. 421 g/kg DM, p < 0.01). The 48 h DM and aNDF degradabilities (DMD and aNDFD) differed (p < 0.001) among the WPCs. The DMD was greatest for barley, intermediate for wheat and triticale, and least for oat (719, 677, 663, and 566 g/kg DM, respectively). Cumulative CH4 production (MP; mL) from 12 h to 48 h of incubation was less (p < 0.001) for oat than the other cereals, reflecting its lower DMD. However, CH4 yield (MY; mg of CH4/g DM degraded) of barley and oat grown at one location was less than that of wheat and triticale (28 vs. 31 mg CH4/g DM degraded). Chemical composition failed to explain variation in MY (p = 0.35), but it explained 45% of the variation in MP (p = 0.02). Variation in the CH4 emission potential of WPC was attributed to differences in DMD, aNDFD, and fermentation end-products (R2 ≥ 0.88; p < 001). The results indicate that feeding whole-plant oat forage to ruminants may decrease CH4 emissions, but animal performance may also be negatively affected due to lower degradability, whereas barley forage may ameliorate emissions without negative effects on animal performance.

Context:Resistance training is a common form of exercise for competitive and recreational athletes. Enhancing recovery from resistance training may improve the muscle-remodeling processes, stimulating a faster return to peak performance.Objective:To examine the effects of 2 different recovery modalities, neuromuscular electrical stimulation (NMES) and cold-water immersion (CWI), on performance and biochemical and ultrasonographic measures.Participants:Thirty resistance-trained men (23.1 ± 2.9 y, 175.2 ± 7.1 cm, 82.1 ± 8.4 kg) were randomly assigned to NMES, CWI, or control (CON).Design and Setting:All participants completed a high-volume lower-body resistance-training workout on d 1 and returned to the human performance laboratory 24 (24H) and 48 h (48H) postexercise for follow-up testing.Measures:Blood samples were obtained preexercise (PRE) and immediately (IP), 30 min (30P), 24 h (24H), and 48 h (48H) post. Subjects were examined for performance changes in the squat exercise (total repetitions and average power per repetition), biomarkers of inflammation, and changes in cross-sectional area and echo intensity (EI) of the rectus femoris (RF) and vastus lateralis muscles.Results:No differences between groups were observed in the number of repetitions (P = .250; power: P = .663). Inferential-based analysis indicated that increases in C-reactive protein concentrations were likely increased by a greater magnitude after CWI compared with CON, while NMES possibly decreased more than CON from IP to 24H. Increases in interleukin-10 concentrations between IP and 30P were likely greater in CWI than NMES but not different from CON. Inferential-based analysis of RF EI indicated a likely decrease for CWI between IP and 48H. No other differences between groups were noted in any other muscle-architecture measures.Conclusions:Results indicated that CWI induced greater increases in pro- and anti-inflammatory markers, while decreasing RF EI, suggesting that CWI may be effective in enhancing short-term muscle recovery after high-volume bouts of resistance exercise.

Antioksidan yang dihasilkan oleh tubuh berfungsi sebagai mekanisme perlindungan terhadap serangan radikal bebas memiliki jumlah yang terbatas. Antioksidan eksogen yang aman dan mudah diperoleh adalah antioksidan dari bahan alam seperti Halimeda macroloba yang mengandung senyawa bioaktif meliputi fenol, klorofil a dan b, serta karotenoid. Metode penelitian yang digunakan adalah deskriptif eksploratif. H. macroloba diperoleh dari Pantai Teluk Awur - Jepara dicuci bersih dengan air tawar dan diblender. Sampel dimaserasi pada pelarut metanol, etil asetat, dan n-heksan, serta diuapkan dengan rotary evaporator. Aktivitas antioksidan ditentukan dari nilai IC50. Nilai ini diperoleh dari persamaan kurva regresi linier pada nilai inhibisi yang terdeteksi dari nilai absorbansi DPPH murni terhadap pengaruh ekstrak H. macroloba yang diukur pada panjang gelombang 517 nm. Kadar total fenol diuji menggunakan larutan Folin-Ciocalteu dengan asam galat sebagai larutan standar yang diukur pada panjang gelombang 725 nm. Kadar klorofil a, b, dan karotenoid diukur pada panjang gelombang 663 nm, 646 nm, dan 470 nm. Hasil penelitian menunjukkan nilai IC50ekstrak metanol sebesar 38,57 ppm, etil asetat 1567,27 ppm dan n-heksan 259,34 ppm. Kadar total fenol pada masing-masing ekstrak adalah 87,10; 140,78 dan 102,24 (mg GAE/g berat basah sampel), kadar klorofil a sebesar 25,56; 28,44 dan 0,41 (mg/g berat basah sampel), kadar klorofil b sebesar 42,10; 41,94 dan 0,61 (mg/g berat basah sampel), dan kadar karotenoid sebesar 97,92; 85,68 dan 2,34 (µ mol/g berat basah sampel). Nilai IC50 dari ekstrak metanol termasuk kategori aktivitas antioksidan sangat kuat sedangkan ekstrak etil asetat dan n-heksan termasuk kategori aktivitas antioksidan sangat lemah.

ImportanceHandheld phone use while driving is a major factor in vehicle crashes. Scalable interventions are needed to encourage drivers not to use their phones.ObjectiveTo test whether interventions involving social comparison feedback and/or financial incentives can reduce drivers’ handheld phone use.Design, Setting, and ParticipantsIn a randomized clinical trial, interventions were administered nationwide in the US via a mobile application in the context of a usage-based insurance program (Snapshot Mobile application). Customers were eligible to be invited to participate in the study if enrolled in the usage-based insurance program for 30 to 70 days. The study was conducted from May 13 to June 30, 2019. Analysis was completed December 22, 2023.InterventionsParticipants were randomly assigned to 1 of 6 trial arms for a 7-week intervention period: (1) control; (2) feedback, with weekly push notification about their handheld phone use compared with that of similar others; (3) standard incentive, with a maximum $50 award at the end of the intervention based on how their handheld phone use compared with similar others; (4) standard incentive plus feedback, combining interventions of arms 2 and 3; (5) reframed incentive plus feedback, with a maximum $7.15 award each week, framed as participant’s to lose; and (6) doubled reframed incentive plus feedback, a maximum $14.29 weekly loss-framed award.Main Outcome and MeasureProportion of drive time engaged in handheld phone use in seconds per hour (s/h) of driving. Analyses were conducted with the intention-to-treat approach.ResultsOf 17 663 customers invited by email to participate, 2109 opted in and were randomized. A total of 2020 drivers finished the intervention period (68.0% female; median age, 30 [IQR, 25-39] years). Median baseline handheld phone use was 216 (IQR, 72-480) s/h. Relative to control, feedback and standard incentive participants did not reduce their handheld phone use. Standard incentive plus feedback participants reduced their use by −38 (95% CI, −69 to −8) s/h (P = .045); reframed incentive plus feedback participants reduced their use by −56 (95% CI, −87 to −26) s/h (P &amp;lt; .001); and doubled reframed incentive plus feedback participants reduced their use by −42 s/h (95% CI, −72 to −13 s/h; P = .007). The 5 active treatment arms did not differ significantly from each other.Conclusions and RelevanceIn this randomized clinical trial, providing social comparison feedback plus incentives reduced handheld phone use while individuals were driving.Trial RegistrationClinicalTrials.gov Identifier: NCT03833219

Profound understanding of the local electronic and defect structure in semiconductors always plays a vital role in the further developing of applications of such materials. In the present work an investigation of the electronic structure in hydrogenated TiO2 (rutile) thin films is conducted by virtue of Time-Differential γ-γ Perturbed Angular Correlation spectroscopy (TDPAC or PAC) with 111mCd/Cd isotope, produced and implanted at ISOLDE/CERN. The measurements were performed at 581 K as a function of the temperature of the samples during hydrogenation. Despite the fact, that rutile single crystals usually show the presence of two local environments, when are studies with Cd/In isotopes, the current pristine thin films sample had a single electric field gradient. Upon various degrees of hydrogenation, Cd probe atoms showed underwent alterations, resulting in up to 3 different local surroundings, generally with high electric field gradients. Broad EFG distributions are likely due to randomly distributed point defects in the neighbourhood of Cd acceptors. Observed results suggest that hydrogenations performed at RT and 423 K are not able to promote unique defect configurations, while in the range of 473-573 K the formation of such configurations is observed. Therefore, one may assume that the formation of Cd-defect complexes (Cd-H pairs) is temperature enhanced. At higher levels of hydrogenation (663 K), the samples become partly amorphous that further hinders any atomistic studies with strong damped PAC spectra. Cd-H complexes seem to be stable up to annealing up to 581 K annealing. The obtained results give a deep insight into complex hydrogen defects, their interactions and bond formations with Cd acceptor.

Abstract The bleaching effluent discharged from a pulp and paper mill contains chlorinated organic compounds which are toxic to living matter. Physicochemical treatments such as coagulation and different advanced oxidation processes (AOPs) were employed for combined bleaching effluent generated from the first two stages (i.e. chlorination and alkali extraction) (pH = 3.5, chemical oxygen demand (COD) = 1,920 mg/L, and total organic carbon (TOC) = 663 mg/L). At optimum conditions (pH = 7.5, polyaluminium chloride (PAC) dose = 3.84 g/L and slow mixing time = 25 min), ∼68% removal in UV254 and ∼23% TOC removal was obtained during coagulation. Among various AOPs, UV/Fe2+/TiO2/H2O2 system showed the highest TOC and COD removals (∼78%) after 2 h duration (Fe2+:H2O2 molar ratio = 1:100). After the AOP process, chloride ion concentration and biodegradability of the treated wastewater was increased to 2,762 mg/L and 0.46 from an initial value of 2,131 mg/L and 0.29, respectively. The wastewater and sludge analysis showed oxidation and adsorption as the major mechanisms for organics removal. Upon reuse of the regenerated catalysts, TOC removal was reduced significantly. It was found that three times more sludge per unit TOC removal was generated after coagulation in comparison to that produced after UV/Fe2+/TiO2/H2O2 treatment.

DNA microarray technology was applied to gain insight into the role of the redox state of PQ pool as a retrograde factor mediating differential expression of Arabidopsis nuclear genes during the acclimation to changing irradiance. DNA microarray chips containing probes corresponding to 24,000 Arabidopsis nuclear genes were screened with cRNA samples prepared from leaves of plants exposed for 5 h to low irradiance (control) vs. medium, high and excessive irradiances (MI, HI and EI, respectively). Six hundred and sixty three genes were differentially expressed as a result of an exposure to at least one elevated irradiance. Among 663 differentially expressed genes a total of 50 were reverted by DCMU--24 ones modulated at medium irradiance, 32 ones modulated at high irradiance and a single one modulated at excessive irradiance. We postulate that their expression is regulated by redox state of plastoquinone (PQ) pool. Thus the PQ-mediated redox regulation of expression of Arabidopsis nuclear genes is probably limited to the irradiance window representing non-stressing conditions. We found that the promoter regions of the PQ-regulated genes contained conserved elements, suggesting transcriptional control by a shared set of trans-acting factors which participate in signal transduction from the redox state of the PQ pool.

Abstract Xenobiotic-metabolizing enzymes influence the susceptibility to malignancies as well as prognosis by causing chemotherapy resistance. We analyzed genetic polymorphisms of glutathione S-transferases T1 and M1 (del GSTT1 and del GSTM1) and NAD(P)H quinone-oxoreductase (NQO1 C609T) to evaluate the association and risk of acute myeloid leukemia (AML). A large-scale population-based, case-control study of 1700 controls and 663 cases was conducted in Chonnam National University Hwasun Hospital, Korea. GST T1 null genotype was associated with increased risk for AML [odds ratio (OR) = 1.20; 95% confidence interval (CI) = 1.003–1.45, p=0.047]. GST M1 null genotype was highly associated with a decreased risk for AML (OR= 0.38; 95%CI = 0.31–0.46, p&lt; 0.0001). Analysis of the NQO1 C609T polymorphism failed to show an association with AML, although a near significant increase was also found in NQO1 609GG genotype (OR= 1.30; 95%CI = 0.54–4.77, p=0.06) Our results suggest an association of extensive GST with an increased or decreased risk for AML, possibly by an increase in the metabolic activation of chemical carcinogens or linkage to another cancer-causing gene. Inconsistent result in other studies may be due to geographical differences in the type of environmental carcinogens to which different populations are exposed.

The effect of water temperature on the duration of emergence and speed of movement of adult endeavour prawns, Metapenaeus endeavouri and M. ensis, was investigated in the laboratory under controlled conditions from 20˚ to 32˚C. The behaviour of 50 prawns was recorded on videotape over 48 h at five temperatures (20˚, 23˚, 26˚, 29˚ and 32˚C). In general, both species emerged from the substratum during dusk and remained on the substratum until dawn or just before dawn when they buried again. At 32˚C, however, some M. endeavouri emerged for much less time and started to bury shortly after midnight. The mean daily duration of emergence for M. endeavouri varied with temperature and was higher at 26˚ and 29˚C (653 40 min per day) than at other temperatures. In contrast, the mean duration of emergence for M. ensis was not significantly affected by temperature (range of times 663 45 to 693 9 min). The pattern of the mean hourly speed of movement for M. endeavouri varied with temperature, whereas that for M. ensis was bimodal at most temperatures: they were more active during dusk (1700 to 1800 hours) and dawn (0400 to 0500 hours) than at other times.

663 Background: Urachal cancer (UrC) is a rare genitourinary cancer with molecular drivers commonly found in colorectal cancer and harbors fewer actionable alterations. Although prior studies have identified genomic alterations associated with UrC, these lack comprehensive genomic profiling, often using targeted panels, and lack RNA interrogation. This study presents detailed characterization of molecular profiles and the tumor microenvironment of real-world UrC patient samples. Methods: UrC samples (n = 42) were profiled using next-generation sequencing (NGS) of DNA and RNA. Prevalence was calculated for pathogenic SNVs/Indels and copy number amplifications, dMMR/MSI status assessed by IHC and NGS, PD-L1 expression measured by IHC, and TMB-H defined as ≥ 10 mut/Mb. Immune cell fractions of TME were estimated using RNA deconvolution. Mann-Whitney U, chi-square, and Fisher exact tests were applied where appropriate, with p-values adjusted for multiple comparisons. Histological features were reviewed by a genitourinary pathologist. Results: UrC samples were collected from 19 males and 23 females with a median age at sample collection was 58.5 years (range: 24-86 years). Pathology review revealed the majority were adenocarcinomas (n=41/42, 97.6%), including mucinous (n=17/41, 41.5%) and enteric adenocarcinomas (n=10/41, 24.4%). Pathogenic variants were most commonly observed in TP53 (n = 35, 83.3%), KRAS (n=15, 36%), GNAS (n=5, 12%), SMAD4 (n=4, 10%), PIK3CA (n=4, 10%), STK11 (n=3, 7%), BRCA2 (n=3, 7%), ARID1A (n=2, 5%), PTEN (n=2, 5%), ERBB2 (n=2, 5%), BRAF (n=2, 5%), and APC (n=2, 5%). MAPK pathway alterations were present in 52.3% (n=22) overall, with increased MAPK pathway activation in MAPK-altered UrC (median MPAS: 0.77 vs -0.61, p = 0.066). Loss of heterozygosity was prevalent (n = 9, 29%). While no UrC tumors had dMMR/MSI, 5% (n=2) were TMB-H and 9% (n=3) were PD-L1+. The mean fraction of T cells – CD8 (0.4%) and monocytes (0.0%) represented a smaller fraction UrC TME composition compared to macrophages M1 (5.19%) and neutrophil (6.4%). The median NK cell fraction was significantly higher in MAPK-altered UrC patients (3.5 vs 2.7%, p = 0.022), yet a transcriptional signature of response to immunotherapy (IFN-g) was similar between MAPK-altered and MAPK-WT UrC patients (median IFN: -0.366 vs -0.384, p = 0.052). Conclusions: This study provides a comprehensive molecular characterization of UrC and the associated immune landscape. While MAPK and DNA repair gene alterations were common, UrC tumors rarely harbored predictive markers of response to immunotherapy, suggesting limited efficacy in this patient population. However, the recurrence of MAPK alterations and associated pathway activation in UrC warrants further investigation of MAPK-targeted therapies in prospective clinical trials.

Conventionally, we expect the transport of intercalated species in battery active particles is limited by diffusion. MgCr2O4 has been identified as a candidate active material for the Mg batteries based on its theoretical Mg-storing capacity, high open circuit potential, and acceptable diffusion barrier for Mg-ion transport (1,2). However, when operated electrochemically, its performance does not match the promised potential (3). Motivated by these observations, we examine charge transport in dense MgCr2O4 pellets in an electrolyte-free setup to conclusively probe bulk Mg transport. We find that the ion transport in this material cannot be explained by using the conventional understanding. We propose a new theory that extends the conventional picture and accounts for the unusual behavior in MgCr2O4 (4). 10027 Background: Sarcomas constitute a group of diverse diseases with many subtypes and varying prognoses. Hence, new approaches for therapy involving targeting of specific survival pathways are needed. Methods: We used 17-AAG to block HSP90 and rapamycin (Ra) to block mTOR, in 2 osteosarcoma cell lines, HOS and KHOS/NP. Cells were treated for 24 and 48 hours with 200 nM of 17- AAG or 5 micromolar of rapamycin. Gene profiling (GP) was performed using h-U133 2 Plus Array (Affymetrix). Results were pre-filtered using the GCOS pair-wise comparisons. ANOVA was filtered for >2-fold change between two conditions. We compared control HOS or KHOS vs. 24 H or 48 H treatment with Ra, or 17AAG. Gene identity, ontology and z-scores were derived. For micro RNAs (miRs), we used a miRNA chip (LC Sciences) and data were normalized, adjusted and clustered using t-test (p<0.01). Results and Conclusions: 17-AAG is a good inducer of apoptosis in HOS and KHOS cells (IC50 of ∼50nM) and blocked cells at mitosis. Ra is a weak inducer of apoptosis (IC20 at 5 micromolar) and blocked cells at G1. GP of 17AAG treated HOS/KHOS cells revealed >1,000 and 1,600 gene changes at 24 H and 48 H, respectively, with ∼700 genes affected at 24 and 48H. Ra affected ∼400 and ∼350 genes at 24 and 48H, respectively, with 20 genes affected at 24 and 48H. Of the genes upregulated by 17AAG, >450 were in cell and protein metabolism; >50 genes were in chromatin modifications and >40 in regulation of cell cycle. With 17AAG, changes were found in histones and deacetylases; cytoskeleton proteins; extracellular matrix proteins and MMPs; HSPs; MAP kinase, mTOR, IGF and VEGF pathways; cell cycle and apoptosis. Many targets were validated by by immunoblotting. MiRs were profiled following 24H treatment with 17AAG or Ra. 17AAG upregulated miR122a;145;143 and 663 (4–16 fold) and miR 542–5P was decreased (∼12 fold). Ra upregulated miR 498;145;143;452;663; 602 and miR224 (4–32 fold) and downregulated (4–32 fold) miR1;542–5P and miR335. Taken together, miR changes affect the expression of transcripts related to angiogenesis; cell cycle; cell signaling and apoptosis and are common to the changes observed by GP. In summary, new therapeutic targets were identified by the combination of GP and miRNA following 17AAG or rapamycin. No significant financial relationships to disclose.
