Abstract Retinoid X receptors (RXRs) are important transcriptional nuclear hormone receptors, acting as either homodimers or the binding partner for at least one fourth of all the known human nuclear receptors. Functional nongenomic effects of nuclear receptors are poorly understood; however, recently peroxisome proliferator-activated receptor (PPAR) \#947;, PPAR\#946;, and the glucocorticoid receptor have all been found active in human platelets. Human platelets express RXR\#945; and RXR\#946;. RXR ligands inhibit platelet aggregation and TXA2 release to ADP and the TXA2 receptors, but only weakly to collagen. ADP and TXA2 both signal via the G protein, Gq. RXR rapidly binds Gq but not Gi/z/o/t/gust in a ligand-dependent manner and inhibits Gq-induced Rac activation and intracellular calcium release. We propose that RXR ligands may have beneficial clinical actions through inhibition of platelet activation. Furthermore, our results demonstrate a novel nongenomic mode for nuclear receptor action and a functional cross-talk between G-protein and nuclear receptor signaling families.

Background: During last two decades, good progress has been made for the flavonoids in metabolic and infectious diseases. However, expectations have not been fulfilled when these compounds were extended to the in vivo studies, particularly in humans. This could due to low bioavailability and less absorption of flavonoids in the biological systems. A recent study revealed that methylation of flavonoids can bring about dramatic changes in pharmacokinetic and biochemical properties. Often, the partially methylated flavonoids show better activities by improving their metabolic stability, membrane transport capacity, facilitating absorption and for oral bio-availability. Though, partial methyl ethers occupy a large chemical space, their biological properties has not been well established. Azaleatin (quercetin-5-O-methyl ether) is one of such group of naturally occurring molecules. Methods: In the present study, we have utilized methoxymethyl (MOM) protecting groups for the preparation of azaleatin. Synthesized compounds and their derivatives were subjected for α-Amylase and DPPH activities. α-Amylase activity can be measured in vitro by hydrolysis of starch in presence of α-amylase enzyme. Antioxidant capacity was evaluated by measuring the scavenging activity of azaleatin and related compounds on the 2,2- diphenyl-l-1-picrylhydrazil (DPPH) radical. In order to identify the binding mode of the compound azaleatin, Auto Dock Tools (http://mgltools.scripps.edu) were used. Results: Quercetin, along with their derivatives, monomethyl ethers i.e. azaleatin, isorhamnetin, tamarixetin; dimethyl ether i.e. quercetin-3,7-dimethyl ether; quercetin-3,3',7-trimethyletherpachypodol; quercetin-3,3',4'7-tetramethyl ether and quercetin pentamethyl ether were taken for α- amylase inhibitory activity. The study showed that azaleatin was found to be comparable with the standard for the inhibition of α-amylase amongst the tested compounds. Since, azaleatin is a best for the inhibition of α-amylase, this compound was taken for the in-silico molecular modelling studies. Azaleatin, showed a good docking energy (-8.8 Kcalmol-1) with the α-amylase receptor. Similarly, other closely related derivatives were studied for their antioxidant capacity. It was found that amongst the compound tested quercetin was found to be best (EC50 of 30µg/mL) for their antioxidant capacity and second best compound was azaleatin; which showed EC50 of 36.1µg/mL. Conclusion: An efficient synthesis of azaleatin, a lesser known flavone has been achieved from quercetin. Amongst the compounds tested, azaleatin was found to inhibit α-amylase with the acceptable radical scavenging activity. Further, in-silico modelling studies indicated that azaleatin found to have very good affinity with the key residues i.e. Gln63, Asp197 and Arg195 of α-amylase receptor. Since, azaleatin has other free hydroxyls in their template, it can be effectively utilized for the activity improvement through further structural modifications.

AbstractLet G be a non-abelian finite simple group. In addition, let $$\Delta _G$$ Δ G be the intersection graph of G, whose vertices are the proper non-trivial subgroups of G, with distinct subgroups joined by an edge if and only if they intersect non-trivially. We prove that the diameter of $$\Delta _G$$ Δ G has a tight upper bound of 5, thereby resolving a question posed by Shen (Czechoslov Math J 60(4):945–950, 2010). Furthermore, a diameter of 5 is achieved only by the baby monster group and certain unitary groups of odd prime dimension.

A sonochemical route is used to prepare a few layers of MoSe2 and its 2D–2D nanohybrid is prepared with graphene by a solvothermal process. This nanohybrid exhibits a high specific capacitance of 945 F g−1. An ASC device MoSe2/G‖AC is fabricated, which delivers an energy density of 26.6 W h kg−1 and a power density of 0.8 kW kg−1.

Medicinal plants and their natural bioactive molecules, are evaluated as the foundation for health preservation and care of humanity. The licorice root, known as “Radix Glycyrrhizae”, is a perennial plant that comes from Mediterranean countries, central to southern Russia, Asia, Turkey, Iraq and Iran. The licorice root has been used in traditional Chinese medicines for centuries and has been defined as "the progenitor of herbs". The name 'Licorice' is derived from the ancient Greek word Glukurrhiza, meaning 'sweet root'. It consists of approximately 30 species, however, the most common ones consist of Glycyrrhiza glabra L., Glycyrrhiza uralensis Fisch and Glycyrrhiza Inflata. In addition, the licorice root contains chalcones, which are a part of an important class of natural products and are precursors of flavonoids. Chemically, chalcones are composed of two aromatic rings associated with α, β-unsaturated α-carbon ketone, representing the prima nucleus of the structure. They have been classified, according to chemical structures, in Licochalcone A, B, C, D, E, F and G. This review aims to highlight all the in vitro and in vivo studies that have been conducted on the licochalcones, extracted from Glycyrrhiza species. The main effects are as follows: anti-inflammatory, antioxidant, anticancer, antimicrobial, antiviral, antiallergic, antidiabetic, hepatotoxic and osteogenic. It is important to implement the introduction of biologically active natural molecules from the bench (research) to the bedside (clinical practice). However, in the future, it is required to conduct additional studies to validate these biological effects.

Background: Cancer is a disease of high mortality. The therapeutic agents currently available are insufficient to cure it and are associated with serious side effects. 4-methylumbelliferone is a natural product containing benzo-α-pyrones as a central nucleus. Benzo-α-pyrone is a privileged moiety having multifarious biological activities including anticancer activity. A series of compounds were synthesized taking 4-methylumbelliferone as a core nucleus and screened for their anticancer activity against HeLa cancer cell line. Methods: The 4-methylumbelliferone was linked with aminoacids using chloroacetyl chloride or ethyl chloroacetate as linker. The N-methylmorpholine and isobutylchloroformate protocol was used for amidic coupling. The final compounds were tested against the HeLa cancer cell lines using SRB assay protocol. Results: Three compounds have shown significant anticancer activity viz 9a, 12f and 15l having GI50 (µg/ml) value of, 56.1, 30.9 and 50.9, respectively. Other compound 9f and 13 showed weak anticancer activity having GI50 (µg/ml) value of 97.2 and 71.1, respectively. Conclusion: It has been found that the synthesized derivatives have inhibitory effect on the growth of cancer cell line. Compound 12f has been found as the most active compound of the synthesized series.

O sistema olfatório de mamíferos é capaz de detectar milhares de substâncias químicas diferentes, mesmo em baixas concentrações. Um odorante disperso no ar pode se ligar a um receptor olfatório (OR) iniciando o processo de detecção. Os ORs são membros da super família de receptores acoplados a proteína G (GPCRs). Apesar de a via de transdução de sinal de odorantes estar bem descrita, pouco se sabe sobre os seus moduladores. Em 2005, nosso laboratório identificou RIC-8B como um possível fator de troca de nucleotídeos de guanina (GEF) que poderia amplificar a atividade da proteína G olfatória (Golf). No presente trabalho mostramos que RIC-8B é capaz de interagir com Gγ13. Procurando os outros componentes desse complexo identificamos Gβ1 como sendo a subunidade Gβ mais expressa no epitélio olfatório. Além disso, RIC-8B, Gαolf, Gβ1 e Gγ13 encontram-se concentrados nos cílios dos neurônios olfatórios e se co-localizam nesse compartimento celular. Nossos experimentos de co-imunoprecipitação mostram que RIC-8B interage mais fortemente com Gαolf, na presença de GDP do que na presença de GTP, como esperado para uma GEF. Curiosamente quando Gβ1 e Gβ13 estão presentes, RIC-8B e Gαolf co-imunoprecipitam igual, independente do nucleotídeo de guanina utilizado. Apesar de, na presença de Gβ1 e Gγ13, não observarmos dissociação física de RIC-8B e Gαolf com a mudança do estado de ativação, o efeito de RIC-8B na produção de AMPc não é afetada. Também mostramos que a quantidade de Gαolf, Gβ1 e Gγ13 presentes na membrana celular aumenta quando estas são co-transfectadas com RIC-8B em células de mamíferos. Nossos resultados apontam para um papel duplo da RIC-8B. Primeiro, RIC-8B poderia funcionar como uma chaperona auxiliando na formação e transporte do complexo heterotrimérico, Golf, em neurônios olfatórios. Em segundo lugar, RIC-8B também atuaria como uma GEF sobre Gαolf, aumentando a produção de AMPc e portanto amplificando a via de transdução de sinal de odorantes. Por fim, acreditamos que um possível papel para Gβ1 e Gγ13 nesse complexo seria funcionar como um andaime molecular, aproximando RIC-8B de seu alvo, Gαolf, potencializando ainda mais a atividade de GEF.
The mammalian olfactory system detects small amounts of thousands of different chemical compounds. Odorant perception starts when an odorant in the air binds to an olfactory receptor (OR). ORs belong to the super family of G-protein coupled receptors (GPCRs). Even though the odorant signaling pathway is well known, little is known about its modulators. In 2005, our lab identified RIC-8B as a putative guanine nucleotide exchange factor (GEF) that is able to interact with the olfactory G-protein (Golf) and amplify its activity. Here we show that RIC-8B also interacts with Gγ13. We also found that Gβ1 is the Gβ subunit that is predominantly expressed in the olfactory epithelium. Furthermore, RIC-8B, Gαolf, Gβ1 and Gγ13 are highly concentrated in the cilia of olfactory neurons and co-localize in this cellular compartment. We also show that RIC-8B interaction with Gαolf is stronger in the presence of GDP than GTP, as expected for a GEF. Curiously, in the presence of Gβ1 and Gγ13, RIC-8B and Gαolf remain associated, in the presence of both GDP or GTP, probably through an indirect interaction via Gβ1/Gγ13. We also showed that the amounts of Gαolf, Gβ1 and Gγ13 in the cell membrane increase if RIC-8B is cotransfected in the same cell. Our results suggest that RIC-8B plays two roles. First, it may act as a chaperone which assists in the assembly and trafficking of the G protein complex. Second, RIC-8B would also act as a GEF to increase Gαolf dependent cAMP production and thereby amplify odorant signal transduction. Lastly, we believe that Gβ1 and Gγ13 may act as a scaffold to position RIC-8B close to its target, Gαolf, further enhancing the GEF activity.

Olanzapine is a second generation antipsychotic that show low incidence of extrapyramidal side effects and has been recommended as the first line drug for the treatment of schizophrenia and is also used in the treatment of bipolar disorder. But it has the weight gain as a side effect which is common in the chronic use of this medicine. A comprehensive review of the literature revealed that olanzapine induces more weight gain than most other antipsychotics, except clozapine. The incidence of weight gain induced by olanzapine and associated diseases such as diabetes and cardiovascular diseases is higher in this group of patients than in the general population. These unwanted side effects have decreased patients' adherence to treatment. Many clinical observations and studies have attempted to elucidate the possible mechanism involved. However, to date, the mechanism underlying the weight gain induced by olanzapine remains unclear. This present study retrospective evaluates 21 patients using olanzapine for a period of 20 to 119 months, compared within the sample, patients who lost weight or remained stable (<7% gain in relation to BMI) group which gained weight at a moderate or severe way during the use of olanzapine (> 7% gain in relation to BMI). We also evaluated the levels of glucose and plasma lipids of all patients. For the group of patients were also analyzed genetic polymorphisms of TaqIA DRD2 gene and G-308A TNF-α gene by PCR-RFL and ARMS-PCR, respectively. TaqIA of genetic polymorphism (C32806T) in the DRD2 gene was correlated with prolonged use of olanzapine with relevant statistical significance in relation to weight gain and biochemical changes observed in plasma of patients. Regarding the genetic variants of the SNP G-308A TNF-α gene, the findings of this study failed to corroborate or refute the findings of other studies.
A olanzapina é um antipsicótico de segunda geração que exibe uma baixa incidência de efeitos colaterais extrapiramidais e tem sido recomendada como fármaco de primeira linha para o tratamento da esquizofrenia e também é utilizada no tratamento do transtorno bipolar, mas tem o ganho de peso como efeito colateral comum no uso crônico deste medicamento. Uma análise abrangente da literatura revelou que a olanzapina induz maior ganho de peso do que a maioria dos outros antipsicóticos, com exceção da clozapina. A incidência de ganho de peso induzido pela olanzapina e doenças associadas, como diabetes e doenças cardiovasculares, é maior entre o grupo de pacientes do que a da população em geral. Estes efeitos secundários indesejados têm diminuído a adesão dos pacientes ao tratamento. Muitas observações clínicas e estudos têm tentado elucidar o possível mecanismo envolvido. No entanto, até o momento, o mecanismo subjacente ao ganho de peso induzido pela olanzapina permanece obscuro. No presente estudo foi realizada uma investigação retrospectiva, que avaliou 21 pacientes em uso de olanzapina por um período de 20 a 119 meses, comparando dentro da amostra, pacientes que perderam peso ou ficaram estáveis (< 7% ganho em relação ao IMC) ao grupo que ganhou peso de forma moderada ou grave durante o uso da olanzapina (>7% ganho em relação ao IMC). Também foram avaliados os níveis de glicose e lipídeos plasmáticos de todos os pacientes. Para o grupo de pacientes ainda foram analisados os polimorfismos genéticos de TaqIAno gene DRD2 e G-308A do gene TNF-α por PCR-RFL e ARMS-PCR, respectivamente. O polimorfismo genéticos da TaqIA (C32806T) no gene DRD2 apresentou relação com o uso prolongado de olanzapina com relevante significância estatística em relação ao ganho de peso e às alterações bioquímicas observadas no plasma dos pacientes. Em relação as variantes genética do SNP G-308A no gene TNF-α, os achados do presente estudo não permitiram corroborar ou refutar as conclusões de outros estudos.

O tráfego de leucócitos é um processo complexo, dependente da ação de inúmeras substâncias químicas, além da perfeita interação celular. Desta forma, este estudo teve como objetivo avaliar a ação dos GCe e da ANXA1 sobre o eixo SDF-1α/CXCR4 e IL-17/IL-23/G-CSF e sobre a expressão de moléculas de adesão CD18, CD49d e CD62L. Foram utilizados camundongos machos Balb/C selvagens (WT) ou ANXA1-/-. As avaliações foram realizadas em condições basais, na presença de altas concentrações de GCe e na vigência de processo inflamatório, induzidos pela administração de ACTH (5 µg/animal, i.p.) ou pela injeção de LPS (100 µg/kg, i.p.), respectivamente, ou na ausência da ação dos GCe, pela ação do RU 38486 (RU, 10 mg/kg, i.p.). A participação da ANXA1 e do receptor FPR2 foi avaliada pelo pré-tratamento com Ac2-26 (1 mg/Kg, i.p.) ou com BOC2 (10 µg/animal, i.p.) durante 4 dias, 1 vez ao dia. A quantificação total e diferencial das células foi realizada em câmara de Neubauer e em esfregaços corados por May-Grunwald ou citometria de fluxo. As quantificações de CXCR2, CXCR4, FPR2, CD18, CD49d, CD62L e maturação granulocítica (CD11b/Ly6G) em células da medula e da circulação foram realizadas por citometria de fluxo. A expressão de ANXA1 nos tecidos do estomago e do baço foi realizada por western blotting e nas células da medula óssea e sangue circulante foi realizada por imunofluorescência. As quantificações de IL-17, IL-23, G-CSF, SDF-1α e corticosterona foram realizadas por ELISA. A quimiotaxia de neutrófilos da medula óssea e sangue periférico foi ensaiada na placa de quimiotaxia com filtro de poro de 8 µm. A fagocitose de neutrófilos apoptóticos por macrófagos da medula óssea foi avaliada por ensaio in vitro. Para verificar os efeitos do ACTH na migração de neutrófilos no processo inflamatório, foi empregado o modelo de bolsa de ar (100 µg/mL; LPS); e o comportamento dos leucócitos circulantes de animais tratados com ACTH foi avaliado pela técnica de microscopia intravital. Os resultados obtidos, que estão apresentados em quatro temáticas, mostraram que: 1) neutrófilos da medula óssea e sangue periférico expressam ANXA1 no citoplasma e membrana, bem como o receptor FPR2, constitutivamente, e a expressão de ambos é regulada pelos GCe. A ANXA1, via receptor FPR2 expresso em células da medula óssea, controlam a maturação neutrofílica e o tráfego destas células da medula óssea para o sangue. A ANXA1, via interação ao FPR2, controla o clearance de neutrófilos do sangue para a medula óssea, modulando o eixo SDF-1α/CXCR4; 2) A administração do ACTH causa neutrofilia e os neutrófilos circulantes são ANXA1+, CD18+, CD49d+, CD62L+, mostrando que injeção do ACTH in vivo altera o fenótipo destas células na circulação. Estas modificações alteram o comportamento dos neutrófilos na circulação, bem como a migração para a bolsa de ar na vigência de inflamação e para os tecidos de clearance. Estes efeitos podem ser dependentes, pelo menos em parte, da inibição de migração orientada, já que quimiotaxia frente ao fMLP ou ao SDF-1α estavam reduzidas. Ainda, o clearance de neutrófilos é reduzido em animais tratados com o ACTH pela menor atividade fagocítica e secretora dos macrófagos medulares; 3) Animais tratados com RU 38486 e ANXA1-/- mobilizam granulócitos da medula óssea para o sangue circulante e, deste compartimento para o foco de inflamação com maior intensidade que o observado em animais controles. O eixo IL-17/IL-23/G-CSF parece estar envolvido na granulopoiese e na mobilização de neutrófilos para o sangue durante a inflamação, mas não é alvo de ação da ANXA1 e o GCe nesta etapa do processo inflamatório. Adicionalmente, foi observado que na vigência de peritonite, as moléculas de adesão, CD49d e CD62L estão envolvidas no processo de migração de neutrófilos da medula óssea para o sangue. Os resultados aqui obtidos permitem concluir que os GCe e a ANXA1 são relevantes para granulopoiese e tráfego dos neutrófilos da medula óssea em condições fisiológicas e na vigência de processo inflamatório. Ainda, em conjunto com os dados da literatura, os nossos resultados podem sugerir a participação da ANXA1 dos GCe na plasticidade fenotípica dos neutrófilos de acordo com os estímulos a que são submetidos, e podem auxiliar na compreensão dos novos conceitos sobre a produção, tempo de vida, localização e funções de neutrófilos.
The traffic leukocytes is a complex process dependent on the action of severals chemical mediators, in addition to perfect cell interaction. Therefore, this study aimed to evaluate the effect of GCe and ANXA1 on SDF-1α/CXCR4 and IL-17/IL-23/G-CSF and on the expression of adhesion molecules CD18, CD49d and CD62L. Balb/C wild type and ANXA1-/- male mice were employed. The analysis were performed at physiological conditions, in the presence of high concentrations of GCe and during of inflammatory process induced by ACTH administration (5 µg/animal, i.p.) or LPS injection (100 µg/kg, i.p.), respectively or in the absence of GCe action, by the action of RU 38486 (RU, 10 mg/kg , i.p.). The involvement of the receptor FPR2 and ANXA1 was assessed by pre-treatment with Ac2-26 (1 mg/kg, i.p.) or BOC2 (10 µg/animal, i.p.) for 4 days, once a day. The quantification of total and differential cell was performed in a Neubauer chamber and stained smears by May-Grunwald and flow cytometry. Quantification of expression of CXCR2, CXCR4, FPR2, CD18, CD49d, CD62L and granulocytic maturation (CD11b/Ly6G) in the bone marrow and circulation were performed by flow cytometry. The expression of ANXA1 on tissues was performed by western blotting and on cells from bone marrow and blood by immunocytochemistry. Quantification of IL-17, IL-23, G-CSF, SDF-1α and corticosterone were performed by ELISA. The chemotaxis of neutrophils from the bone marrow and blood was tested in the chemotaxis chamber with filter pore of 8 microns. The phagocytosis of apoptotic neutrophils by bone marrow macrophages was assessed by in vitro assay. To investigate the effects of ACTH in the migration of neutrophils in the inflammatory process, the model employed was air pouch (100 µg/ ml, LPS), and the behavior of circulating leukocytes from animals treated with ACTH were evaluated by intravital microscopy. The results obtained, which are presented in three sections, showed that: 1) neutrophils from the bone marrow and blood expressed ANXA1 in the cytoplasm and membrane, as well as FPR2, constitutively and the expression of both is regulated by GCe. The ANXA1 via FPR2 receptor expressed in bone marrow cells, controls the neutrophilic maturation and traffic of these cells from the bone marrow into the blood. The ANXA1 via interaction to FPR2 controls the clearance of neutrophils from the blood to the bone marrow by modulating the SDF-1α/CXCR4 axis; 2) the administration of ACTH induces neutrophilia and the circulating neutrophils are ANXA1+, CD18+, CD49d+ and CD62L+, showing that the injection of ACTH in vivo alters the phenotype of these cells in the blood. These modifications alter the behavior of neutrophils in the blood, as well as the migration to the air pouch in the presence of inflammation and to the tissue clearance, and these effects may be dependent, at least in part, on inhibition of migration oriented events, as chemotaxis in response to fMLP or SDF-1α were reduced. Further, the clearance of neutrophils is reduced in animals treated with ACTH due to the lower phagocytic and secretory activity of medullary macrophages; 3) Animals treated with RU 38486 and ANXA1-/- mobilize granulocytes from bone marrow into the blood, and from this compartment to the focus of inflammation with higher intensity than that observed in the control group. The axis IL-17/IL-23/G-CSF seems to be involved in granulopoiesis and mobilization of neutrophils into the blood during inflammation, but it is not the target of action of ANXA1 and GCe at this step of inflammatory process. Additionally, it was observed that in the presence of peritonitis, the adhesion molecules, CD49d and CD62L are involved in the migration of neutrophils from the bone marrow into the blood. The results obtained allow concluding that the GCe and ANXA1 are relevant to the granulopoiesis and the traffic of neutrophils from bone marrow under physiological conditions and in the presence of inflammation. Furthermore, together with literature data, the data presented here may suggest the involvement of ANXA1 the GCe in phenotypic plasticity of neutrophils according to the stimuli that are submitted, and may support to understand the new concepts of production, half-life, location and function of neutrophils.

Neste trabalho são apresentados estudos de Cu(α-AIB)2 utilizando-se a técnica de RPE à temperatura ambiente nas freqüências de 9,7 GHz e 34 GHz. Os espectros de R. P. E. mostram uma única ressonância tanto em banda X (9,7 GHz) quanto em banda Q (34 GHz), devido ao efeito de estreitamento por troca. Os valores das componentes do tensor g e da largura de linha foram determinados a partir dos espectros obtidos variando o ângulo entre H e os eixos do cristal a´, b e c em três planos a´b, a´c, e bc. O tensor g reflete as propriedades moleculares do complexo, com o íon Cu(II) em uma simetria axial e também a orientação destas moléculas dentro da cela unitária do cristal. A variação angular da largura de linha é analisada em termos da simetria do íon Cu(II) na rede cristalina e das contribuições das interações dipolar e Zeeman Residual. O parâmetro da interação de troca |J\'|, é obtido através da contribuição da interação Zeeman residual na largura de linha, |J\'| ~ 0,34K. É também observada uma característica magnética bidimensional no complexo Cu(α-AIB)2 concordando com os resultados cristalográficos.
In this work is presented a study of the complex Cu(α-AIB) 2 using EPR spectroscopy at room temperature, in two frequency bands (9.7 and 34 GHz). The EPR spectra, in both bands and any direction of the extremal magnetic field consist of a single resonance line. This fact can be understood considering the exchange narrowing between non-equivalent Cu(II) íons. The elements of the g tensor and line width were determined from the angular dependence of the EPR spectrum, in three ortogonal crystal planes a´b, a´c and ab (a´=b x c). The angular dependence of the g tensor reflects the molecular properties of the complex Cu(α-AIB)2 the axial symmetry of the molecule and the orientation on the crystal unit cell. The most important contributions to the line width were found to be: 2D dipolar interactions, the residual Zeeman effect and defects compatible to the symmetry of the crystal. The Exchange parameter, |J\'| ~ 0.34K, was obtained from the residual Zeeman contribution to the line width (Q band). The low dimension found for dipolar interations agrees with crystallographic results.

Intimina é uma adesina de natureza protéica das bactérias diarreiogênicas Escherichia coli enteropatogenica (EPEC) e enterohemorrágica (EHEC), capazes de induzir a lesão \'attaching e effacing\' em enterócitos. Os principais subtipos de intiminas de EPEC e EHEC prevalentes no Brasil são α, β e γ. Nosso objetivo foi investigar a transferência de anticorpos maternos anti-intiminas aos recém-nascidos de mães saudáveis de São Paulo, Brasil. Foram pesquisados anticorpos SIgA no colostro e IgG no soro de 50 mulheres saudáveis e no soro de cordão umbilical de seus recém-nascidos, por ELISA utilizando como antígeno proteínas recombinantes purificadas das regiões conservadas e variáveis de intiminas α, β e γ. As concentrações de anticorpos no colostro foram superiores quando comparadas com as concentrações do soro para todos os tipos de intiminas. Não se observaram diferenças estatísticas entre as concentrações de anticorpos reativos com as diferentes intiminas nas amostras de colostro. As concentrações de anticorpos reativos com a região conservada da intimina foram significativamente mais elevadas em comparação com as regiões variáveis no soro dos grupos de mães e de recém-nascidos. Houve alta correlação entre todos os anticorpos anti-intiminas nas amostras de colostro. Comparando-se as concentrações de anticorpos séricos, os coeficientes foram maiores entre anti-α e anti-β que entre os outros pares. Nossos resultados confirmam a transferência de anticorpos maternos para o recém-nascido pela placenta e pelo aleitamento materno e reforça o efeito protetor da amamentação contra infecção por EPEC.
Intimin is a proteic adhesin of enteropatogenic (EPEC) and enterohemorragic (EHEC) Escherichia coli, capable of inducing attachment and effacement lesion in enterocytes. The main subtypes of intimins of EPEC and EHEC prevalent in Brazil are α, β and γ. Our aim is to investigate the transference of maternal anti-intimin antibodies to the newborns of healthy mothers from Sao Paulo, Brazil. IgG and SIgA antibodies were determined in sera and colostrum from 50 healthy women and cord sera from their newborns, by ELISA using as antigens purified recombinant proteins, conserved and variable regions of α, β and γ intimins. The IgA antibody concentrations of colostrum are higher than IgG antibodies in serum for all intimins and there were no statistical differences between them in colostrum samples. The concentrations of antibodies reactive with the conserved region of intimin are significantly higher compared to the variable regions in the sera groups, mothers and newborns. There were high correlation coefficients between all the anti-intimins antibodies in colostrum samples. In the comparison of the seric antibody concentrations, the coefficients were higher between anti-α and anti-β than all the other pairs. Our results confirm the transference of maternal antibodies to the newborns by placenta and breastfeeding and reinforce the high protection effect of breastfeeding against EPEC infection.

Die Lotzdorfer Schule und der 2015 durch Brandanschlag zerstörte Lotzdorfer Gasthof nahmen im dörflichen Leben immer eine zentrale Stellung ein. Jubiläen der Schule waren Anlass zu Heimatfesten, Festumzügen, Volksfesten und waren auch für die Dorf-Entwicklung und Dorfgemeinschaft wichtig. Bereits die historischen Schulen als Vorläufer der heutigen Ludwig-Richter-Schule Lotzdorf waren immer Ausgangspunkt vielfältiger Aktionen. Die hiermit vorgestellte Chronik der Schulfeste und Ereignisse mit vielen Fotodokumenten, spiegelt eine Dorfgemeinschaft wider, die mit bewunderungswürdigen Aktionen immer wieder ihr Dorf auf hohem Niveau feierte, auch über die sogenannte „Wendezeit“ nach 1989 hinaus. Es ist Tradition geblieben, dass sich bis heute, zu gemeinsamen Klassentreffen der gesamten Schule, die Schüler aller Jahrgänge zu einem großen Fest vereinen und auch von überall her an ihre einstige Schule, in ihr Dorf kommen.

AbstractDisturbances of social and affective processes are at the core of psychiatric disorders. Together with genetic predisposing factors, deprivation of social contact and dysfunctional relationships during development are some of the most important contributors to psychiatric disorders over the lifetime, while some developmental disorders manifest as aberrant social behavior early in life. That the cause of mental illness is rooted in the brain was long held as a truism, yet finding the causes for and neurobiological correlates of these conditions in the brain has proven and continues to be difficult (Venkatasubramanian G, Keshavan MS, Ann Neurosci 23:3–5. https://doi.org/10.1159/000443549, 2016). In clinical practice, psychiatric disorders are diagnosed based on categorical manuals, such as the DSM and ICD, which form a useful guide for clinical diagnosis and interventions. Yet, understanding the specific neural mechanisms leading to or characterizing distinct psychiatric conditions through this categorical approach has been slow (see, for example, Lynch CJ, Gunning FM, Liston C, Biol Psychiatry 88:83–94. https://doi.org/10.1016/j.biopsych.2020.01.012, 2020). Findings in the brain often do not seem to lend support to common mechanisms for the defined disorder categories. This is not particularly surprising because, in these diagnostic manuals, multiple combinations of symptoms can often lead to the same diagnosis, which is reflected in highly variable phenotypes of psychiatric disorders.

Chem. Soc. 2014, 136, 1300) conditions for stepwise HAT, con­verting 1 to the thermodynamically-favored trans-fused ketone 2. Seth B. Herzon of Yale University devised (J. Am. Chem. Soc. 2014, 136, 6884) a protocol for the reduc­tion, mediated by 4, of the double bond of a haloalkene 3 to give the saturated halide 5. The Shenvi conditions also reduced a haloalkene to the saturated halide. Daniel J. Weix of the University of Rochester and Patrick L. Holland, also of Yale University, established (J. Am. Chem. Soc. 2014, 136, 945) conditions for the kinetic isomerization of a terminal alkene 6 to the Z internal alkene 7. Christoforos G. Kokotos of the University of Athens showed (J. Org. Chem. 2014, 79, 4270) that the ketone 9, used catalytically, markedly accelerated the Payne epoxidation of 8 to 10. Note that Helena M. C. Ferraz of the Universidade of São Paulo reported (Tetrahedron Lett. 2000, 41, 5021) several years ago that alkene epoxidation was also easily carried out with DMDO generated in situ from acetone and oxone. Theodore A. Betley of Harvard University prepared (Chem. Sci. 2014, 5, 1526) the allylic amine 12 by reacting the alkene 11 with 1-azidoadamantane in the presence of an iron catalyst. Rodney A. Yong-Qiang Wang of Northwest University oxidized (Org. Lett. 2014, 16, 1610) the alkene 6 to the enone 14. Peili Teo of the National University of Singapore devised (Chem. Commun. 2014, 50, 2608) conditions for the Markovnikov hydration of the alkene 6 to the alcohol 15. Internal alkenes were inert under these conditions, but Yoshikazo Kitano of the Tokyo University of Agriculture and Technology effected (Synthesis 2014, 46, 1455) the Markovnikov amination (not illustrated) of more highly substituted alkenes.

Modern broadband seismic data provides new insight into the Eocene-Miocene syn- to early post-rift sections of the East Java Sea (North Madura) and South Makassar Basin. Previous subsurface imaging suffered from shallow imaging obstacles and “hard” Eocene-Miocene carbonates, which render the interpretation of basement and overlying early syn-rift sections difficult. Thickness and distribution of these syn-sift sections differ significantly when comparing the North Madura area and the South Makassar Basin. Late syn- and early post-rift carbonate development also differs significantly in that a carbonate inter-reef blanket interrupted by pinnacle reef formation on top of a syn-rift section is present in North Madura. This contrasts with carbonate sections including bioherms that are controlled by the geometry of the underlying basement substrate, and which occur over extensive parts of the western South Makassar Basin. Early syn-rift including source rocks appear limited, requiring petroleum charge generation and migration from the central South Makassar Basin or the Central deep depression.

A closed-loop two-phase microchannels cooling system using a micro-gear pump was built in this paper. The microchannels heat sink was made of oxygen-free copper, and 14 parallel microchannels with the dimension of 0.8mm(W)×1.5mm(D)×20mm(L) were formed by electric spark drilling followed by linear cutting which separated the channels from each other. The heat transfer performance was evaluated by the fluid temperature, the pressure drop across the micro-channels and the volumetric flow rate. Experiments were performed with refrigerant FC-72 which spanned the following conditions: initial pressure of Pin = 73 kPa, mass velocity of G = 94 – 333 kg/m2s, outlet quality of xe,out = 0 – superheat and heat flux of q″= 25–140 W/cm2. The result showed that, the maximum heat flux achieved 96 W/cm2, as the heating surface temperature was kept below 85 °C and critical heat flux occurred in the condition of low flow rate. Average two-phase heat transfer coefficients increased with the heat flux at low mass flux (G = 94 and 180 kg/m2s) and all heat fluxes, high mass flux (G = 333 kg/m2s) and all heat fluxes, and moderate mass fluxes (G = 224kg/m2s) under low and moderate heat fluxes (q″&lt;110 W/cm2 for G = 224 kg/m2s), which was a feature of nucleate boiling mechanism. Pressure drop through microchannels heat sink was found to be below 4kPa.

We studied the biochemical composition, nutritive value of the green mass and prepared hay from Ro-manian cultivars of perennial ryegrass, Lolium perenne L. „Mara” and „Măgura” grown in monoculture on the experimental plot of the National Botanical Garden (Institute) “Alexandru Ciubotaru”, Chisinau, R. Mol-dova. Results revealed that dry matter of harvested green mass contained 98-116 g/kg CP, 299-326 g/kg CF, 71-77 g/kg ash, 329-353 g/kg ADF, 546-593 g/kg NDF, 38-40 g/kg ADL, 291-313 g/kg Cel, 217-240 g/kg HC, with nutritive and energy value 60.9-65.3 % DMD, 60.1-64.8 % ODM, RFV=96-108, 12.12-12.45 MJ/kg DE, 9.95- 10.23 MJ/kg ME and 5.97-6.25 MJ/kg NEl. The biochemical composition, nutritive and energy va-lue of prepared hay: 94-110 g/kg CP, 351-385 g/kg CF, 79-82 g/kg ash, 390-423 g/kg ADF, 631-689 g/kg NDF, 46-50 g/kg ADL, 344-373g/kg Cel, 241-266 g/kg HC, RFV=76-86, 11.13-11.61 MJ/kg DE, 9.14- 9.53 MJ/kg ME and 5.17-5.55 MJ/kg NEl. The biochemical methane potential of green mass substrates reached 338-344 L/kg organic matter. The studied cultivars of Lolium perennec could be used in the Republic of Mol-dova for the restoration of degraded permanent grasslands, as a component of the mix of grasses and legumes for the creation of temporary grasslands, the harvested biomass can be used as feed for farm animals or as substrate in biogas generator for renewable energy production.

A previous PVP paper [1] identified suggested improvements to be made to ASME Code, Section XI, Nonmandatory Appendix G, “Fracture Toughness Criteria for Protection Against Failure” [2]. That paper also identified that the current version of Appendix G does not have any provisions for when the calculated operating stress (pseudo stress) exceeds the material yield strength. The treatment of stresses exceeding yield was included in earlier versions of Appendix G, but it was removed via Code Action ISI-94-40 in 1995. The specific reasons for removal of these provisions were not documented. In some Appendix G postulated flaw evaluations for pressure-temperature (P-T) limits, the calculated total linear-elastic (or pseudo) stress (i.e., including the primary stress due to pressure loading and thermal stress) may exceed the material yield stress. The ASME Section XI Working Group on Operating Plant Criteria (WGOPC) decided that this provision needed to be more fully considered, with appropriate benchmarking and possible adjustments to Appendix G made consistent with the current state of knowledge in elastic-plastic fracture mechanics (EPFM) methods. This is appropriate since the state of knowledge in EPFM has significantly advanced since the time the technical basis for Appendix G was established, as documented in Welding Research Council (WRC) Bulletin No. WRC-175, which was published in 1972. Furthermore, EPFM provides an improved method for evaluating the effects of high stresses. This paper describes the results of preliminary investigations of stresses exceeding the material yield stress in fracture toughness assessments associated with Appendix G. Also included in the technical evaluations presented are the temperature conditions for which upper shelf conditions are present and where EPFM methods are applicable.

15 populations of black medic (Medicago lupulina L.) of various ecological and geo-graphical origin were studied by the type of ontogenesis. It was found that 1 population is represented entirely by monocarp forms of plants, 7 populations have 20–60% of mono-carps. Wild alfalfa from the Moscow region and mutant forms created on its basis consist of 100% bicarpic plants. Bicarpic plants of varieties Georgia, Nordol, Rinata and selection number LH19-3, whose average dry matter productivity was 40.8–48.0 g/plant, are valuable as a source material for breeding of black medic. As sources of high seed prod-uctivity (10.0 and 10.3 g/plant), you can use bicarpic plants of the selection number VIK 256 and Georgia varieties, and high leafed (52.4 and 55.3%) — selection numbers DS-1 and VIK 61/94.

Rice (Oryza sativa L.) is the staple food for Sri Lankans, and it serves as a key source of essential mineral elements. The variation of grain nutrient concentrations as affected by genetic factors (variety, grain color and age group) are not known, and those were tested in the current study. Total of 200 rice grain samples were collected using a stratified random sampling approach, representing all agro-climatic zones in Sri Lanka. Grain nitrogen (N), phosphorus (P) and potassium (K) concentrations were measured using Kjeldahl, Colorimetric, and General methods, respectively. Grain N P and K concentrations were significantly different among rice varieties (P<0.1). Grain N concentration varied between 4-19 mg g−1 . The highest grain N concentration was recorded in Bg 307 (14 mg g−1 ) while the lowest (i.e., < 10 mg g−1 ) in Bg 367, Bg 374, Bg 358, Bg 310 and Bg 379-2. Grain P concentration varied in the range 0.6- 1.7 mg g−1 . Grain P concentration in At 406 was the highest (1.6 mg g-1), followed by Bg 307, Bg 94-1, Bg 367 i.e., >1.3 mg g-1 and the lowest in Bg 403, Ld 365 and Bg 310 i.e., < 1 mg g−1 . Bg 357, Ld 365, Bg 406, Ld 368 and Bg 310 rice varieties showed significantly lower P concentration than other varieties (P<0.1) Grain N, P and K concentrations were similar among different age classes of rice varieties (P>0.05). Moreover, grain N and P concentrations between the red and white grain varieties were similar (P>0.05). However, varieties with white color grains had higher K concentration than in red rice varieties. There was a significant correlation between grain P and K concentrations (r =0.496, P<0.001). This information would be useful when selecting rice varieties with high and low nutritional qualities and implementing sustainable nutrient management practices in rice-based cropping systems in Sri Lanka. Švinas yra sunkusis metalas, pasižymintis stipriomis toksinėmis savybėmis. Šio elemento randama nuotekose, o kartais ir geriamajame vandenyje. Straipsnyje nagrinėjamas švino šalinimas iš vandens, naudojant sorbcijos procesą, norint nustatyti efektyviausiai šviną šalinantį sorbentą. Tyrime naudoti trys sorbentai: molis, sapropelis ir geležies paplavos. Nustatyta, kad molis šviną pašalina 47–91 % efektyvumu, naudojant sapropelį pasiekiamas 94–99 % švino pašalinimas, o naudojant geležies paplavas švino pašalinimo efektyvumas pasiekiamas – 72–99 %. Atlikti tyrimai parodė, kad efektyviausiai šviną iš tiriamojo vandens šalina sapropelis. Naudojant skirtingas sapropelio kiekius ir esant skirtingai kontako trukmei, švino koncentracija tiriamajame vandenyje po valymo neviršija leistinų geriamojo vandens normų (0,01 mg/l), todėl sorbcijai galima naudoti mažiausią sapropelio kiekį – 1 g/l. Efektyviausias švino išvalymas pasiekiamas, kai kontakto trukmė – 150 min.

Abstract We present a proof of concept demonstration of solar thermochemical energy storage on a multiple year time scale. We designed and fabricated a 4-kW solar rotary drum reactor to carry out the solar-driven charging step of solar thermochemical storage via metal oxide reduction-oxidation cycles. During the summer of 2019, the solar reactor was operated in the Valparaiso University solar furnace to effect the reduction of sub-millimeter cobalt oxide particles in air at approximately 1000°C. A particle collection system cooled the reduced particles rapidly enough to maintain conversions of 84–94% for feed rates of 2.9–60.8 g min−1. The solar to chemical storage efficiency, defined as the enthalpy of the reduction reaction at 1000°C divided by the solar energy input, reached 20% for an input solar flux of 1400 kW m−2 and a cobalt oxide feed rate of 60.8 g min−1. Samples of the reduced cobalt oxide particles were stored in vials in air at room temperature for more than three years. The stored solar energy was released by reoxidizing samples in air in a benchtop reactor. Measurements of the oxygen uptake by the reduced metal oxide confirm its promise as a medium to store and dispatch solar energy over long durations.
