Background: The Portfolio-To-Impact version 2 (P2I v.2) financial forecasting tool estimates funding requirements for development of portfolios of candidate health products (drugs, biologics, vaccines or diagnostics). The assumptions and archetypes relating to diagnostics in P2I v.2 are based on limited data and may not accurately describe research and development costs, timelines and probability of success. This study aimed to revise the P2I v.2 tool by modifying the diagnostic assumptions to improve accuracy of predictions for diagnostic portfolios. Methods: Data from expert interviews and historical information on development of 26 existing diagnostics were used to determine approximate research and development costs, timelines and probability of success for development of diagnostics, and to revise diagnostic archetypes and development phases. To compare the revised tool with P2I v.2, data on 27 candidates from the Foundation for Innovative New Diagnostics (FIND) tuberculosis and pandemic preparedness portfolios were input into both versions. Results: The number of diagnostic archetypes increased from two in P2I v.2 to three in the revised tool. Total estimated costs to move the 27 candidates along the pipeline to launch were US$641.62 million with P2I v.2 and US$274.00 million with the revised model. The number of expected launches was 21.65 over five years with P2I v.2 and 11.48 over eight years with the revised model. Development timelines were extended and probability of success was lower with the revised model compared with P2I v.2. Conclusions: Outputs from the revised tool were in line with expert experience, suggesting that the proposed revisions improve the accuracy of the tool for estimating research and development costs, timelines and probability of success relating to diagnostic portfolios. Additional improvements to the tool could include further refinement of archetypes, incorporation of a measure of potential public health impact, and addition of a commercialization phase for diagnostics.

The article presents the results of two interrelated studies. The first is related to assessing the possibility of using a water-polymer ultrajet as a diagnostic tool for cutting tools for the disposal of plastics. The second is related to the development of a directly new tool design. As a result of research, it was shown that both the method of water-polymer diagnostics and the proposed design of the knife have prospects for practical application. A model of a composite tool for the disposal of composites is proposed, which potentially has high operational properties. The research results are confirmed by the data of a comparative analysis of various diagnostic methods and strength calculations of the tool.

The online community resource Phytophthora database (PD) was developed to support accurate and rapid identification of Phytophthora and to help characterize and catalog the diversity and evolutionary relationships within the genus. Since its release in 2008, the sequence database has grown to cover 1 to 12 loci for ≈2,600 isolates (representing 138 described and provisional species). Sequences of multiple mitochondrial loci were added to complement nuclear loci-based phylogenetic analyses and diagnostic tool development. Key characteristics of most newly described and provisional species have been summarized. Other additions to improve the PD functionality include: (i) geographic information system tools that enable users to visualize the geographic origins of chosen isolates on a global-scale map, (ii) a tool for comparing genetic similarity between isolates via microsatellite markers to support population genetic studies, (iii) a comprehensive review of molecular diagnostics tools and relevant references, (iv) sequence alignments used to develop polymerase chain reaction-based diagnostics tools to support their utilization and new diagnostic tool development, and (v) an online community forum for sharing and preserving experience and knowledge accumulated in the global Phytophthora community. Here we present how these improvements can support users and discuss the PD's future direction.

In Automotive Industries, to be confident regarding the success of a planned operation, performing accurate methods in order to detect abnormal operating conditions, known as faults, is crucial. An effective method for diagnosis and fault recognition ensures the safety of the operation, reduces manufacturing cost and any other potential impacts. In addition, mobile solutions have been widely adopted among automotive manufactures during recent years and they have taken full advantage of mobile strategies. Accordingly, it is necessary for there to be a future-proof plan to control the diagnostic operations in advance. In this thesis, the immediate objective has been to offer a future-proof and user-friendly solution to assist engineers and service technicians in the monitoring, detecting, and diagnosing of faults on Toyota/BT/CESAB branded trucks. A mobile cross-platform framework is used to develop the diagnostic mobile solution which is not only able to be deployed on Android and iOS mobile platforms, but also provides wireless communication between truck machines and mobile devices through Bluetooth and Wi-Fi ad hoc technologies. The diagnostic mobile tool is capable of processing real-time controller area network messages and visualizing the condition of different sensors in a more user-friendly way through rich hybrid and client-side web user interfaces. The experience of evaluating a cross-platform diagnostic tool on different mobile operating systems proved that cross-platform mobile development methodology can be a reliable technique for developing projects that essentially require real-time data processing. In addition, it indicates that Apple iOS offers a better runtime performance than Google Android for the current tool.

En ny produkt, som utvecklats för att underlätta de många diagnoser och bedömningar avsjukdomen lymfödem, utvärderades och vidareutvecklades i denna studie. Tidiga mätningar avpatienter i riskzonen var viktigt för en tidig upptäckt av sjukdomen. Lymfödem yttrar sig somsvällande och förhårdnade kroppsdelar, ofta armar och ben, och uppkommer delvis till följd avcancerbehandling. De mätmetoder som användes inom vården när rapporten skrevs, bestod aven lång rad mätningar av kroppsdelars omkrets med ett vanlig måttband. Det fanns utrymme förförbättring av metoden som kunde göras mer konsekvent och effektiv. En risk fanns att klinikeranvände måttbandet på olika sätt när omkretsar på patienter mättes. Dessutom skulle denhårdhetsutvärdering av kroppsdelar, som genomfördes av klinikerna, kunna kvantifieras ochanalyseras om dessa utvärderingar sattes i siffror.Målen med detta projekt var att utvärdera den nya produktens noggrannhet och precision, samtutveckla och integrera en funktion för mätning av hudens hårdhet. Ett användargränssnittanpassad till klinikers krav och en konstruktion anpassad för testning och framtida tillverkning,var också projektmål. Med anledning av patientsäkerhetskrav gjordes en del av produktenutbytbar mellan mätningar av olika patienter.Under utvecklingen användes videoinspelningar av praktiska tester av prototypen ochundersökning som delades ut till kliniker som utförde lymfödembehandling. Mätningen avomkrets som utvecklats i denna studie var, enligt de tester som utförts, tillräckligt noggrannjämfört med den noggrannhet den vedertagna mätmetoden krävde. En automatisk beräkning aven bulkmodul K för vävnaden, utvecklades som en integrerad funktion för att kunna erhålla ettvärde på den hårdhet som kliniker kände med sina händer. K-värdet visade sig fungera som ettkoncept, genom mätningar av olika typer av skummaterial. Beträffande mätningar i vävnadvisade sig produktens bestämning av K-värden kunna detektera en ökning av muskelspänning,men resultaten var bara förutsägbara till en viss gräns i testet som presenteras i rapporten.Metoden kunde i denna studie, inte på detta sätt valideras för vävnad genom att jämförametoden med mätningar med en durometer. En av slutsatserna var dock att K-värdet fortfarandekunde vara en användbar indikator, som ett komplement till den bedömning som var gängse närrapporten skrevs. Utöver detta kunde produkten spara tid i de många mätningarna avlymfödempatienter. Denna sparade tid kunde användas för fler mätningar. Fler mätningar ökarchansen att upptäcka sjukdomen i ett tidigt stadium (Lawenda, Mondry, and Johnstone, 2009),vilket i sin tur är avgörande för en framgångsrik behandling.
To improve the frequently performed lymphedema measuring session – important for an earlydetection of the disease – a novel device was evaluated and further developed in this study.Lymphedema appears as limbs swelling, often arms and legs, partly because of cancertreatment. The current assessment technique includes several circumference measurements witha regular tape measure, and the method had room for improvement regarding consistency andeffectiveness. Different clinicians may pull the tape measure differently while measuring thecircumferences of a patient’s limb. An additional skin compliance evaluation, performed by theclinicians, could be quantified and analyzed if the assessments were put into numbers.The goals of this project were to evaluate and improve the accuracy and precision of this noveldevice, as well as develop and integrate a feature for measuring skin compliance. Furthermore,a user interface adapted to the clinicians’ demands and a design adapted to testing and futuremanufacturing was essential. Due to patient safety reasons, a part of the device was madereplaceable between patients.Developing the device, video recordings of hands-on testing were performed, as well as asurvey distributed to clinicians who provided lymphedema therapy. The circumferencemeasurement developed in this study was, according to the performed tests, accurate enoughcompared to the accuracy required the current measuring routine. An automatic calculation of askin bulk modulus K, was developed as a feature of the device to be able to obtain a value ofthe compliance clinicians felt for with their hands. The K-value was proven as a concept bymeasuring different foams. Measuring K-values of the tissue, the device was capable ofdetecting an increase in muscle tension, but results were only predictable to a certain limit inthe test presented in the report. The method could not yet be validated by comparing it to a skinhardness measurement with a durometer. A conclusion was however, that the K-value stillcould be a useful indicator in addition to the current assessment and that the device could savetime in the measuring session. This saved time could be used for more measurement sessions,beneficial in order to detect lymphedema early (Lawenda, Mondry, and Johnstone, 2009),which in turn was essential for a successful treatment.

The first step of debugging a deployed application is to reproduce the reported bug. But bugs that cause unpredictable behaviors without crashing the applica- tion can be difficult to reproduce, because the developer has to rely on bug-re- ports issued by users. The goal of this thesis has been to develop a diagnostic tool which makes user-reported bugs easier to reproduce. More specifically, the goal has been to develop such a tool for React Native applications implement- ing Redux, taking into consideration both state-data stored in Redux stores and React Native components. Requirements for the diagnostic tool were laid out and partitioned into proto- types that were implemented separately in iterations and then merged into the resulting tool. Requirements were also drawn for three sample applications to test and evaluate the tool, as well as a back-end to prove the tool's capability in uploading and downloading state-data from a server. The thesis resulted in a diagnostic tool that's imported into an already existing code-base as a third-party library, and which collects the complete state of the reported application instance and delivers it to the developer as a single JSON document. The tool can then inject the state-data into a fresh instance of the ap- plication to make it identical to the malfunctioning instance reported by the user; a process referred to as "reviving". Redux stores demonstrated state-over- write protection which complicates state-injection. A study was performed to compare the impact this difference has on performance. As expected, the study revealed that the diagnostic tool takes marginally longer to revive state-data in Redux stores for this reason.

Rodon is a diagnostic tool developed by Sörman. SAAB’s interest in Rodon regards the possibility to use the tool for development and operations of aircraft systems. The main goal of this thesis was to evaluate the capacity of Rodon and determine how SAAB can use the diagnostic tool during development and operations.

The tool uses model based diagnosis with artificial intelligence for fault isolation which is a powerful approach. If Rodon is introduced at SAAB, then detailed models of systems will be necessary to create, including the nominal behavior of the system and different faulty behaviors. In order to achieve high quality fault isolation, it is necessary to have complete and consistent models. To be able to use all applications that Rodon feature for a modeled system, preferable characteristics are that the model should be static, have discrete control signals, and have well defined system behavioral modes.

During development of a system Rodon can be used to improve and easy the work for failure analysis, guidance of sensor placements, evaluation of tests, generation of decision structures, and fault isolation. Since design of tests during development is a desirable application that Rodon does not have, two different methods are presented that utilizes Rodon to generate all possible limit checking tests.

In conclusion, Rodon can be very useful in several different aspects if introduced, but benefits gained by using Rodon will have to be compared to the labor cost of creating good models.

Barrett's esophagus (BE) is a condition of the lower esophagus caused by gastroesophageal reflux disease. Patients with BE have an increased probability of developing dysplasia, an abnormal growth or development of cells. This dysplasia in BE is a precursor to cancer of the esophagus, but is currently difficult to detect and diagnose. If the dysplasia is allowed to progress to cancer, it is very difficult to treat successfully. Treatment for dysplasia itself, however, is very effective if done at an early stage. The goal of this thesis project will be to develop a real-time tool that uses spectroscopy to improve upon the methods of detecting dysplasia in BE. This will involve analyzing spectra acquired from patients with BE using models and extracting quantitative information on different aspects of tissue morphology and biochemistry. Using this information, diagnostic algorithms will be developed, optimized and displayed to the physician through a useful interface.

The development of a clinical diagnostic scheme for ciguatera fish poisoning has been accomplished through the utilization of three assays, specifically a competitive enzyme-linked immunosorbent assay (ELISA) for quantification, and two procedures for separation of toxic potentials, i.e., a liquid-liquid partition method and an immunoaffinity column system. For the ELISA assay, quality control parameters were evaluated including the coating binding efficiency and the binding capability of the conjugate (okadaic acid-bovine serum albumin). A 100% recovery was obtained and reproducibility values were under 30% coefficient of variation (CV) for concentrations from 0.0001 to 100 ng okadaic acid/mL serum or g fish flesh. Additionally, the usefulness of the method was evaluated by determining sensitivity and specificity parameters (66.7 and 95%, respectively) from where the positive and negative predictive values of the test as a diagnostic instrument were assessed. A rapid extraction procedure utilizing a mixture of methanol:chloroform:water was adapted for extraction and isolation of toxins associated with ciguatera fish poisoning. The method has been optimized for the analytical evaluation of potentially ciguatoxic fish samples and human serum. Applications of the extraction procedure include the clinical diagnosis of ciguatera illness and, if leftover fish portions responsible for the intoxication event are available, analysis of the seafood. It can also be used as a tool for monitoring seafood products for food safety. Recovery values of the extraction method with and without an intermediary defatting step for spiked fish samples were of 105 and 86%, respectively. By attaching anti-okadaic acid monoclonal antibody onto an inert matrix (AH-Sepharose®), an immunoaffinity column specific to ciguatera-related toxins has been developed, also for serum analysis, but with the potential application in other body fluids. The binding efficiency was 82% and reproducibility values for spiked test portions at concentrations as low as 250 pg were below 10%.

Parkinson’s disease and Essential tremor are the two most prevalent movement disorders in the world, but due to overlapping clinical symptoms, accurate differential diagnosis is difficult. As a result, approximately 60% of patients with movement disorders symptoms will have their diagnosis changed at least once before death. By their subjective nature, clinical exams are inherently imprecise, leading to the desire to create an objective, quantifiable test for movement disorders; a test that currently is elusive. Eye movements have been studied for a century, and are widely appreciated to be quantifiably affected in those with neurological disease. Through a collaborative effort between the VA hospital and VCU, over 1,000 movement disorder subjects had their eye movements recorded, utilizing an SR Research Eyelink 2. Patients with Parkinson’s disease exhibited an ocular gaze tremor during fixation, normal reflexive saccades, and reduced blink rate. Subjects with Essential tremor exhibited slowed saccadic dynamics, with increased latencies, in addition to a larger number of square wave jerk interruptions of otherwise stable fixation. After diagnostic features of each disorder were identified, prospective data collection could occur in a blinded fashion, and oculomotor features used to predict clinical diagnoses. It was determined that measures of fixation stability were capable of almost perfectly differentiating subjects with PD, and a novel, combined parameter was capable of similar results in ET. As a group, it appears as if these symptoms do not progress as the disease does, but subanalyses show that individual patients on constant pharmaceutical doses tracked over time do slightly change and progress. The near perfect separation of disease states suggest the ability of oculomotor recording to be a powerful biomarker to be used for the differential diagnosis of movement disorders. This tool could potentially impact and improve the lives of millions of people the world over.

For decades the problems of access to and sustained use of water and sanitation (WATSAN) technologies in developing countries has dominated the political agendas of international organisations and governments. Despite the significant investments made and the apparent appropriateness of technologies transferred, the effective implementation and sustained use of WATSAN technologies remains a chimera. More importantly, improving access to water and sanitation does not necessarily guarantee the longevity of those systems transferred. Lessons from past interventions suggest that the success of WATSAN interventions depends on the ability of ensuring users‟ broad acceptance of the technologies and sustained used after donor assistance ends. Yet, in the academic literature users‟ feedback and experiences in the post-implementation stage of technologies has received scarce attention. Against this background, this thesis aims to contribute to understanding the dynamics involved in the process of WATSAN technology adoption and sustained use in developing countries by reporting the design and evaluation of a diagnostic post- implementation tool, called RECAP, to address and investigate the problem. This research employs a multiple case study approach to evaluate users‟ post-implementation experience of WATSAN technologies in South Africa and Indonesia. Semi-structured interviews with technology users as well as in depth interviews with local governments and health clinics were conducted in three case studies. By comparing and contrasting technology intended performance and users‟ experiences in the post-implementation stage this study aims to identify potential challenges to technology sustained used. Conclusions relate to the existence of discrepancies between performance and experience, manifested in the post-implementation stage, which suggest the necessity to develop evolving mechanisms to routinely assess users‟ feedbacks of the technologies and assist them with appropriate interventions. Further conclusions relate to the validity, reliability and flexibility of a post-implementation diagnostic tool in investigating user experiences, diagnosing emerging challenges and suggesting remedial intervention to contribute to sustained technology use.

The monograph presents an approach to the development of a digital tool for the professional training of students of speech pathologists — a virtual practice aimed at the formation of professional competencies in the field of pedagogical diagnostics of children with disabilities. The stages of virtual practice development are considered, the principles of its design are revealed. The place and functions of the created digital tool in the educational process of higher education are described. The results of an experimental study are presented, indicating the effectiveness of the use of virtual practice for the formation of professional competencies of students of speech pathologists. It is addressed to specialists of the education system, teachers, students of pedagogical universities, practitioners in the field of correctional pedagogy. It can be useful to all those who are interested in creating virtual, digital learning tools in the process of preparing students of various fields of study and specialties.

The textbook contains information on new technologies of radiation diagnostics and therapy of oncological diseases, new and promising therapeutic and diagnostic radiopharmaceuticals, new directions in nuclear medicine in Russia and in developed foreign countries, as well as on the coordination of efforts of international organizations to maintain and improve the health of the population of different countries. The active activity of the Rosatom State Corporation on the introduction and development of programs for the non-energy use of nuclear and radiation technologies to improve the health of the world's population, as well as training and training programs for high-tech medicine, combating widespread epidemic diseases, improving health systems in Russia and developing countries is shown. The basic nuclear-physical and therapeutic-diagnostic requirements for radionuclides and radiopharmaceutical drugs used in nuclear medicine are described. Information on the clinical application of new therapeutic and diagnostic tools developed in Russia for the needs of nuclear medicine is presented. In a comparative aspect, a brief description of educational programs of academic training of specialists for high-tech medicine in Russia and developed countries is given. Meets the requirements of the federal state educational standards of higher education of the latest generation 03.04.02 "Physics" (profile "Medical physics", educational programs "Medical physics of nuclear medicine" and "Medical physics of radiation diagnostics and therapy") and 30.05.02 "Medical biophysics". AbstractThe Authors developed a damage assessment system for mooring facilities. This system uses information on seismic motion observed by strong motion seismographs when an earthquake occurs in order to determine whether or not mooring facilities can be used based on the degree of damage suffered. The system creates maps estimating damage based on the results. Two methods are available for determining usability—immediate diagnoses and detailed diagnoses—and all calculations are performed automatically, from the acquisition of the seismic motion information to the determination of usability. The resulting maps estimating damage can be used to select surveyed facilities for on-site damage assessments, making this system an effective tool for supporting initial response systems after a disaster strikes.

Equation-Oriented (EO) modeling techniques have been gaining popularity as an alternative for simulating and optimizing process systems due to their flexibility and ability to leverage state-of-the-art solvers inaccessible to many procedural modeling approaches. Despite these advantages, adopting EO modeling tools remains challenging due to the significant learning curve and effort required to build and solve models. Many techniques are available to help diagnose problems with EO process models and reduce the effort required to create and use them. However, these techniques still need to be integrated into EO modeling environments, and many modelers are unaware of sophisticated EO diagnostic tools. To survey the availability of model diagnostic tools and common workflows, the U.S. Department of Energy�s Institute for the Design of Advanced Energy Systems (IDAES) has conducted user experience interviews of users of the IDAES Integrated Platform (IDAES-IP) for process modeling. The interviews reveal a gap between the availability and utilization of model diagnostic tools driven primarily by a lack of awareness of and lack of standard interfaces among different tools. To address this gap, the IDAES team has developed a recommended workflow for integrating diagnostics into the model development process and an IDAES Model Diagnostics Toolbox that provides a standard interface for many of these best practices. This paper identifies barriers to the widespread adoption of diagnostic tools for EO models and reduces these barriers by providing a standard, user-friendly interface for many different tools.

<div class="section abstract"><div class="htmlview paragraph">In the rapidly evolving automotive landscape, integrating cutting-edge off-board diagnostics tools has triggered a paradigm shift in diesel engine applications. Simultaneously, engineers are compelled to transform conventional mechanical engines into advanced common rail direct injection (CRDi) systems amidst India’s changing pollution norms for industries. Aligned with Bharat Stage Emission Standards, non-road vehicles face stringent emission limits, necessitating complex electronic control predominantly managed by the engine control unit (ECU). Government mandates require the ECU to detect NOx control malfunctions and emission-affecting faults, storing data for off-board analysis. A tool that can read engine data and monitor engine health is required to deal with this situation.</div><div class="htmlview paragraph">Network protocols such as CAN enable remote communication with specialized ECUs. This study examines implementing customized off-board tools, which helps easier coordination with protocols such as the unified diagnostic services (UDS) guided by ISO 14229, part 1 [<span class="xref">1</span>], keyword protocol (KWP), J1939, and the like. UDS protocol offers technicians access to vital engine parameters, helping in complex diagnostic procedures, and engine performance optimization. Moreover, the research focuses on an off-board tool that helps in diesel engine diagnostics.</div><div class="htmlview paragraph">This paper develops and validates a diagnostic tool for service engineers, accurately collecting data including faults, freezeframes, engine parameters, and emission data. It monitors engine health parameters such as rpm, oil pressure, coolant temperature, rail pressure, and the like, capturing part-specific data crucial for warranty considerations and does routine test validations. This diagnostic tool uses OTX to script diagnostic test procedures, while ODX is used for standardized ECU communication data. This method ensures accuracy and optimizes operations. Also, this paper emphasizes the pretest bench environment to avoid bugs during the initial stages of tool development using MATLAB environment and executes the RDBi feature.</div></div>

<div class="section abstract"><div class="htmlview paragraph">Due to increase in complexity of vehicle functionality and involvement of electronic components, the use of complex electronic control units is prevalent in today’s vehicles. This has led to increased amount of Electronic Control Unit (ECU) data, and in turn increased Diagnostic data. This Diagnostic data is described in the Automotive Open System Architecture Diagnostic Exchange Template (AUTOSAR DEXT), which is a standard diagnostic data format specified in AUTOSAR 4.2.1 for Unified Diagnostic Services and fault memory. It enables consistent exchange of Diagnostic information across Original Equipment manufacturer OEMs and between OEM and Suppliers, thereby aiding uniformity in configuration of basic software modules described in Automotive Open System Architecture (AUTOSAR) Layered Architecture across enterprise boundaries. DEXT provides the possibility to describe the data to be transported, using respective protocol, along with origin of data in ECU’s application software. When there is availability of both information and data, the configuration of diagnostic basic software becomes possible. A diagnostic tool (computer program) has been developed by the US Navy to provide the operators of LM2500 gas turbines (which are used as main propulsion on 6 ship classes in the USN) and the supporting technical staff, an assessment of the performance of this engine and the calibration of the engine control system. Typically, USN LM2500s do not mechanically degrade to the point where they will physically not be capable of making the required power. Instead, drift or lack of calibration of the engine sensors or its signal conditioning, mechanical wear and drift in the engine fuel scheduling control will cause the engine, after a period of time, either to not meet its performance expectations or report a performance level that is different from the actual condition. This can cause the engine to fail to meet the performance expectations of the propulsion control system, whether it is a real problem or a reporting problem (due to sensor mis-calibration), and in the worst case can cause an engine failure. To address these issues a performance diagnostic tool was developed that allowed the troubleshooter to compare engine performance, as reported by engine sensors etc., against what should be expected from the engine. Based on any differences, the tool provides the investigator guidance on the most likely source for the problem. To make such a tool, required integrating engine steady state performance models, with knowledge of the engine control loops as well as engine sensor, signal conditioning accuracy and calibration requirements. At present the diagnostic tool is being used by Fleet Technical Support Center Pacific to diagnose engine and controls problems. The goal is to provide ship’s force with the capability to diagnose most of their own problems and to provide a history of a particular engine’s performance.

This study introduces a diagnostic tool for determining the health of a public development bank. It defines in normative terms what good health looks like across various dimensions, which allows the PDB to delineate how it can improve its overall performance and achieve its financial and developmental goals. Due to the variety of mandates and business models used by PDBs, there is no one definition of what constitutes perfect health for a PDB; nonetheless there are common parameters and features to their overall systems that can be compared against under a flexible and comprehensive framework. The three common parameters of performance identified in the Health Diagnostic Tool are public policy and corporate governance, development impact and financial and operational performance.

The objectives of this research are to develop methods for performing ultrasonic flow meter (USM) diagnostic evaluation automatically and a software tool with all necessary attachments. USM-based diagnostics have been established and thirteen categories of knowledge rules of existing cases have been learned and integrated. A search engine for relevant standards, specifications, and other documents of the measurement system has been developed, which enables the free search of text content. Further, with the assistance of modern reasoning techniques, the authorized user only needs to configure an EXCEL file or scripts to activate the rules of the knowledge base by using Drools technology. Therefore, the integration of any potential knowledge rules is convenient and requires no professional skills or changing of the internal source code of the software. Secondly, a new flow meter diagnostic method is proposed based on multiple information methodologies and it is based on the real-time measurement data, operation data, and video data if applicable. The method is intended to identify abnormal states of the measurement system on a real-time basis with the assistance of the knowledge rules and to provide a strategy for mitigating the meter error of components within the measurement system. Thirdly, the applications of Gaussian quadrature diagnostics in daily acquisition nomination change situations and compressor-induced pulsating flow scenarios have been investigated and results are shown in the document. Based on the results, it can be concluded that the measurement uncertainty caused by compressor-induced pulsating flows is obvious, while the measurement uncertainty caused by daily acquisition nomination change is relatively smaller. The software is then developed based on the knowledge, the idea of multiple information methods, and applications of the Gaussian quadrature diagnostics method with all necessary attachments. The architecture, the algorithm, and a few examples are introduced.

The objectives of this proposal were to enhance animal health by 1) development of a BTV serogroup diagnostic assay using polymerase chain reaction (PCR) and 2) development of a BTV serotype specific diagnostic PCR assay. A PCR assay for diagnosis of bluetongue virus (BTV) serogroup from clinical samples meeting the criteria of objective 1 was developed. This PCR assay is more sensitive than virus isolation and has been adopted by both the U.S. and Israeli collaborating laboratories of this project, as well as at least one other U.S. laboratory for routine diagnosis of BTV infection in ruminants. The basic BTV PCR protocol has also become an essential tool in BTV molecular research in both collaborating laboratories. During development of the BTV serotype specific PCR we had the opportunity to investigate a nationwide outbreak of abortions and fatal disease in dogs in the U.S. purportedly due to BTV infection via a BTV contaminated canine vaccine. The BTV serogroup PCR was integral in confirming BTV in tissues from affected dogs and in lots of the suspect vaccine. This led to the first published report of BTV infection in dogs. We discovered that BTV can produce silent persistent infection in canine cell culture. This indicated a need for more stringent screening of biologics for occult BTV infection. A novel mixed cell culture method was developed to identify occult BTV and other occult viral infection cell cultures. Serotype specific primers for PCR detection of all U.S. BTV serotypes and two Israel serotypes (BTV-2 and 10) have been evaluated and are available. A subsequent collaboration would logically include sequencing of the L2 genes of Israel BTV-4, 6 and 16, allowing incorporation of these Israel BTV serotypes into a multiplex PCR assay.

This report details a study investigating the environmental impacts associated with the development and manufacturing of carbon nanotube (CNT)–based ink (called CNTRENE 1030 material) and novel CNT temperature, flex, and moisture sensors. Undertaken by a private-public partnership involving Brewer Science (Rolla, Missouri), Jordan Valley Innovation Center of Missouri State University (Springfield, Missouri), and the US Army Engineer Research and Development Center (Vicksburg, Mississippi), this work demonstrates the environmental life cycle assessment (ELCA) methodology as a diagnostic tool to pinpoint the particular processes and materials posing the greatest environmental impact associated with the manufacture of the CNTRENE material and CNT-based sensor devices. Additionally, ELCA tracked the degree to which optimizing the device manufacturing process for full production also changed its predicted marginal environmental impacts.

Original objectives and revisions-The proposed research included these original objectives: field validation of diagnostic tests (PCR), the development and evaluation of new sensitive tools (LC-PCR/TaqManPCR, antibody detection by ELISA) including their use to study the ecology and the epidemiology of KHV (virus distribution in the environment and native cyprinids) and the carrier status of fish exposed experimentally or naturally to KHV (sites of virus replication and potential persistence or latency). In the course of the study we completed the genome sequence of KHV and developed a DNA array to study the expression of KHV genes in different conditions. Background to the topics-Mass mortality of koi or common carp has been observed in Israel, USA, Europe and Asia. These outbreaks have reduced exports of koi from Israel and have created fear about production, import, and movements of koi and have raised concerns about potential impacts on native cyprinid populations in the U.S.A. Major conclusions-A suite of new diagnostic tools was developed that included 3 PCR assays for detection of KHV DNA in cell culture and fish tissues and an ELISA assay capable of detecting anti-KHV antibodies in the serum of koi and common carp. The TKPCR assay developed during the grant has become an internationally accepted gold standard for detection of viral DNA. Additionally, the ELISA developed for detecting serum anti-KHV antibodies is now in wide use as a major nonlethal screening tool for evaluating virus status of koi and common carp populations. Real time PCR assays have been able to detect viral DNA in the internal organs of survivors of natural and wild type vaccine exposures at 1 and 10³ genome equivalents at 7 months after exposure. In addition, vaccinated fish were able to transmit the virus to naive fish. Potential control utilizing hybrids of goldfish and common carp for production demonstrated they were considerably more resistant than pure common carp or koi to both KHV (CyHV-3). There was no evidence that goldfish or other tested endemic cyprinids species were susceptible to KHV. The complete genomic sequencing of 3 strains from Japan, the USA, and Israel revealed a 295 kbp genome containing a 22 kbp terminal direct repeat encoding clear gene homologs to other fish herpesviruses in the family Herpesviridae. The genome encodes156 unique protein-coding genes, eight of which are duplicated in the terminal repeat. Four to seven genes are fragmented and the loss of these genes may be associated with the high virulence of the virus. Viral gene expression was studies by a newly developed chip which has allowed verification of transcription of most all hypothetical genes (ORFs) as well as their kinetics. Implications, both scientific and agricultural- The results from this study have immediate application for the control and management of KHV. The proposal provides elements key to disease management with improved diagnostic tools. Studies on the ecology of the virus also provide insights into management of the virus at the farms that farmers will be able to apply immediately to reduce risks of infections. Lastly, critical issues that surround present procedures used to create “resistant fish” must be be resolved (e.g. carriers, risks, etc.). Currently stamping out may be effective in eradicating the disease. The emerging disease caused by KHV continues to spread. With the economic importance of koi and carp and the vast international movements of koi for the hobby, this disease has the potential for even further spread. The results from our studies form a critical component of a comprehensive program to curtail this emerging pathogen at the local, regional and international levels.
