AbstractThe atmosphere parameters, spectral variability and the magnetic field geometry of the Bp-star θ Aur are evaluated from 30 coude-spectrograms. We adopt Te = 10 800K and lgg = 3.40 (with lower reliability of lg g ) . The equivalent widths of the metalic lines show a relatively great scattering which troubles the period search. The best fit to our data gives the period P = 3.620d, but the variability is clearly seen only for Sill lines. The uncertainties in the star parameters obtained bring to a great uncertainty in the magnetic dipole geometry: i = 86° + 39° and β = 18° + 80°. We adopt the most plausible star parameters to be: R = 3.8R⊙ M = 3.5R⊙, i = 80°, β = 20°, Bp = 1 KGs.

AbstractThe technique of aperture synthesis is well developed in radio astronomy. When applied to the optical regime, aperture synthesis allows one to partially overcome the blurring effects of the atmosphere and increase the angular resolution of large telescopes to the diffraction limit. MAPPIT (Masked APerture-Plane Interference Telescope) is a multi-element interferometer which operates at the coude focus of the 3.9 m Anglo-Australian Telescope. This instrument has recently been reconfigured to operate in a dispersed mode so that simultaneous observations in a band of wavelengths are possible. We will discuss this instrument’s new mode and present observations of the double star δ Sco and an angular diameter of the previously unresolved red giant β Gru.

β-thalassemia is an inherited blood disorder in which the body cannot produce hemoglobin normally. Since patients with this condition receive blood transfusions regularly, iron builds up primarily in organs such as the heart, liver and endocrine glands. Accumulation of iron in the organs necessitates chelation therapy. These patients must visit the hospital frequently to assess and follow up on their health condition. Physician intervention is required after each regular assessment to adjust the treatment. Lifelong healthcare support using a web-based expert system with a quick response code is designed for β-thalassemia management in order to deliver benefits to patients, physicians, and other healthcare providers. The aim of this study is to implement a web-based expert system for β-thalassemia management in order to provide treatment recommendations and support the lifelong healthcare of patients. The system provides patient-related details, such as medical history, medicines, and appointments, in real-time. It has been also tested in real-life cases and shown to enhance β-thalassemia management.

Spectral line shapes code in plasmas (SLSPs) code comparison workshops have been organized in the last decade with the aim of comparing the spectra obtained with independently developed analytical and numerical models. Here, we consider the simultaneous effect of a plasma microfield and a periodic electric field on the hydrogen lines Lyman-α, Lyman-β, Balmer-α, and Balmer-β for plasma conditions where the Stark effect usually dominates line broadening.

The angiotensin II (AngII) type 1 receptor (AT1R) is a member of the G protein–coupled receptor (GPCR) family and binds β-arrestins (β-arrs), which regulate AT1R signaling and trafficking. These processes can be biased by different ligands or mutations in the AGTR1 gene. As for many GPCRs, the exact details for AT1R–β-arr interactions driven by AngII or β-arr–biased ligands remain largely unknown. Here, we used the amber-suppression technology to site-specifically introduce the unnatural amino acid (UAA) p-azido-l-phenylalanine (azF) into the intracellular loops (ICLs) and the C-tail of AT1R. Our goal was to generate competent photoreactive receptors that can be cross-linked to β-arrs in cells. We performed UV-mediated photolysis of 25 different azF-labeled AT1Rs to cross-link β-arr1 to AngII-bound receptors, enabling us to map important contact sites in the C-tail and in the ICL2 and ICL3 of the receptor. The extent of AT1R–β-arr1 cross-linking among azF-labeled receptors differed, revealing variability in β-arr's contact mode with the different AT1R domains. Moreover, the signature of ligated AT1R–β-arr complexes from a subset of azF-labeled receptors also differed between AngII and β-arr–biased ligand stimulation of receptors and between azF-labeled AT1R bearing and that lacking a bias signaling mutation. These observations further implied distinct interaction modalities of the AT1R–β-arr1 complex in biased signaling conditions. Our findings demonstrate that this photocross-linking approach is useful for understanding GPCR–β-arr complexes in different activation states and could be extended to study other protein–protein interactions in cells.

Les microtubules sont des fibres du cytosquelette formées par l’assemblage d’hétérodimères d’α- et de β-tubuline. Ils contribuent à l’établissement de la forme des cellules et de leur polarité, ainsi qu’à leur mobilité. Ils jouent aussi un rôle important dans le transport intracellulaire et dans la division cellulaire. Le réseau microtubulaire s’adapte constamment aux besoins de la cellule. Il peut être constitué de microtubules très dynamiques ou d’autres plus stables. Pour moduler dans l’espace et le temps les différentes fonctions de ces fibres, de nombreuses modifications post-traductionnelles réversibles de la tubuline sont mises en jeu, à l’origine de ce qui est maintenant appelé le « code tubuline ». Dans cette revue, nous nous intéresserons au rôle de deux modifications caractéristiques des microtubules stables : l’acétylation et la détyrosination de l’α-tubuline. Nous discuterons également de l’implication de leur dérégulation dans certaines pathologies.

Microtubules are essential dynamic polymers composed of α/β-tubulin heterodimers. They support intracellular trafficking, cell division, cellular motility, and other essential cellular processes. In many species, both α-tubulin and β-tubulin are encoded by multiple genes with distinct expression profiles and functionality. Microtubules are further diversified through abundant posttranslational modifications, which are added and removed by a suite of enzymes to form complex, stereotyped cellular arrays. The genetic and chemical diversity of tubulin constitute a tubulin code that regulates intrinsic microtubule properties and is read by cellular effectors, such as molecular motors and microtubule-associated proteins, to provide spatial and temporal specificity to microtubules in cells. In this review, we synthesize the rapidly expanding tubulin code literature and highlight limitations and opportunities for the field. As complex microtubule arrays underlie essential physiological processes, a better understanding of how cells employ the tubulin code has important implications for human disease ranging from cancer to neurological disorders.

Departing from the original postulates that defined various neurodegenerative disorders, accumulating evidence supports a major role for soluble forms of amyloid proteins as initiator toxins in Alzheimer’s disease, Parkinson’s disease, frontotemporal dementias, and prion diseases. Soluble multimeric assemblies of amyloid-β, tau,α-synuclein, and the prion protein are generally englobed under the term oligomers. Due to their biophysical properties, soluble amyloid oligomers can adopt multiple conformations and sizes that potentially confer differential biological activities. Therein lies the problem: with sporadic knowledge and limited tools to identify, characterize, and study amyloid oligomers, how can we solve the enigma of their respective role(s) in the pathogenesis of neurodegenerative disorders? To further our understanding of these devastating diseases, the code of the amyloid oligomers must be broken.

Designers can use moment redistribution to reduce the design bending moment envelope. Code provisions for moment redistribution are not entirely rational. They neglect the effects of important parameters on permissible moment redistribution and can be very conservative. To establish a realistic limit on permissible moment redistribution, one needs a rational model for predicting the plastic rotation capacity of critical sections (plastic hinges). This paper presents a model for computing the plastic rotation capacity, θp, and permissible moment redistribution, β, in reinforced concrete beams. Important parameters, affecting θp and β, are identified and incorporated in the model. The model is validated against experimental results and shows good agreement. A comparison of the moment redistribution limits is made between the model and CSA A23.3-94. Although the code provides a reasonable estimate of β for unfavourable combinations of parameters, the code can be very conservative when conditions are favourable for moment redistribution. Deeper beams with closely spaced stirrups allow significantly more moment redistribution than that predicted by the code.Key words: moment redistribution, ductility, plastic rotation capacity, bond-slip, shear cracking, reinforced concrete beams, c/d, ultimate concrete strain.

The experiment was conducted to isolate and screen fungal strain and optimization of solid-state fermentation conditions for enhanced production of β-glucosidase. Different fungal cultures were isolated and screened for β-glucosidase production. The physicochemical and nutritional parameters were optimized for enhanced production of β-glucosidase from higher producer. Among all the isolates the isolate which exhibited highest β-glucosidase potential was identified and assigned the code as Aspergillus niger MBT-2. The optimum β-glucosidase production was obtained in M5 medium containing wheat bran after 72 hrs of incubation at 40°C, pH 6 and 20 ml of moisture contents. In addition to this 2% fructose and 2% yeast extract proved to be best carbon and nitrogen sources, respectively and gave maximal enzyme productivity. The exploitation of agricultural by products as a substrate reduced the production cost of enzyme and makes the process economical. The Aspergillus niger MBT-2 has promising potential of bioconversion of low-cost material into valuable product like β-glucosidase.

In this paper, we present new observations of β Cephei stars made with the CFHT coudé Reticon. Spectral sequences of the extreme star BW Vulpeculae show large velocity and line profile changes in Ha and in the C II doublet (λλ6578,6582) over 0.04 of a cycle, during three different cycles on three different nights. In fact, there are two such phases of line doubling where the velocities are nearly discontinuous before and after the so-called velocity “stillstand” (near maximum light). Also, the line doubling phases coincide with pronounced peaks in the Ha core residual intensity. The most straightforward interpretation of these observations is that there are two shock waves, separated by about 50 minutes, which generate double absorption components. For v Eridani and γ Pegasi, there are no discernible changes in the Ha profiles, despite rather regular velocity variations. This argues against the shock-wave interpretation for “classical” β Cephei stars.

Abstract Artificial neural networks (NNs) are trained, based on the numerical database, to predict the no-wall and ideal-wall β N limits, due to onset of the n = 1 (n is the toroidal mode number) ideal external kink instability, for the HL-2M tokamak. The database is constructed by toroidal computations utilizing both the equilibrium code CHEASE (Lütjens et al 1992 Comput. Phys. Commun. 69 287) and the stability code MARS-F (Liu et al 2000 Phys. Plasmas 7 3681). The stability results show that (1) the plasma elongation generally enhances both β N limits, for either positive or negative triangularity plasmas; (2) the effect is more pronounced for positive triangularity plasmas; (3) the computed no-wall β N limit linearly scales with the plasma internal inductance, with the proportionality coefficient ranging between 1 and 5 for HL-2M; (4) the no-wall limit substantially decreases with increasing pressure peaking factor. Furthermore, both the NN model and the convolutional neural network (CNN) model are trained and tested, producing consistent results. The trained NNs predict both the no-wall and ideal-wall limits with as high as 95% accuracy, compared to those directly computed by the stability code. Additional test cases, produced by the Tokamak Simulation Code (Jardin et al 1993 Nucl. Fusion 33 371), also show reasonable performance of the trained NNs, with the relative error being within 10%. The constructed database provides effective references for the future HL-2M operations. The trained NNs can be used as a real-time monitor for disruption prevention in the HL-2M experiments, or serve as part of the integrated modeling tools for ideal kink stability analysis.

Over the years, structural engineering codes and specifications in Canada and elsewhere have moved from an allowable stress design (ASD) approach to a load and resistance factor design (LRFD) philosophy. LRFD methodology takes better account of the inherent variability in both loading and resistance by providing different factors of safety for loads of distinct natures with regard to their probability of overload, frequency of occurrences and changes in point of application. The method also results in safer structures because it considers the behavior at collapse. While resistance factors for traditional construction materials based on LRFD in the National Building Code (NBC) of Canada are available, they cannot be used for non-conventional ones. This is because the resistance of such materials due to various load effects has unique bias factors (λR) and coefficients of variation (VR), which greatly impact their reliability index (β). In this study, relationships between the resistance factor ϕ and critical load effects from the NBC load combinations at ultimate limit states are developed for a wide range of resistance bias factors and coefficients of variation. The relationships are presented in the form of charts that are useful for researchers and code-writing professionals who have expertise in the various fields of structural engineering but lack proper background in reliability theory. The developed spectra showed that for the same ϕ, β increases with an increase in the live-to-dead load (L/D) ratio until it reaches 1; thereafter, the shape of the relationship will depend on the statistics of the resistance as well as on the magnitude of ϕ. For a small ϕ and VR, β will keep increasing with an increase in the L/D ratio from 1 until 3, albeit at a lesser rate. For L/D > 3, the relationship between the critical β and applied load is just about constant. This finding is also true for load combinations involving snow and wind. Application of the method is illustrated by a practical example involving the shear strength of a corrugated web steel beam.

Microtubules are cytoskeletal filaments that are dynamically assembled from α/β-tubulin heterodimers. The primary sequence and structure of the tubulin proteins and, consequently, the properties and architecture of microtubules are highly conserved in eukaryotes. Despite this conservation, tubulin is subject to heterogeneity that is generated in two ways: by the expression of different tubulin isotypes and by posttranslational modifications (PTMs). Identifying the mechanisms that generate and control tubulin heterogeneity and how this heterogeneity affects microtubule function are long-standing goals in the field. Recent work on tubulin PTMs has shed light on how these modifications could contribute to a “tubulin code” that coordinates the complex functions of microtubules in cells.

While the imparting of velocity ‘kicks’ to compact remnants from supernovae is widely accepted, the relationship of the ‘kick’ to the progenitor is not. We propose the ‘kick’ is predominantly a result of conservation of momentum between the ejected and compact remnant masses. We propose the ‘kick’ velocity is given by vkick = α(Mejecta/Mremnant)+β, where α and β are constants we wish to determine. To test this we use the BPASS v2 (Binary Population and Spectral Synthesis) code to create stellar populations from both single star and binary star evolutionary pathways. We then use our Remnant Ejecta and Progenitor Explosion Relationship (REAPER) code to apply ‘kicks’ to neutron stars from supernovae in these models using a grid of α and β values, (from 0 to 200 km s−1 in steps of 10 km s−1), in three different ‘kick’ orientations, (isotropic, spin-axis aligned and orthogonal to spin-axis) and weighted by three different Salpeter initial mass functions (IMF’s), with slopes of -2.0, -2.35 and -2.70. We compare our synthetic 2D and 3D velocity probability distributions to the distributions provided by Hobbs et al. (1995).

This paper presents a study on the seismic performance of rectangular hollow section (RHS) X-joints subjected to in-plane bending moment (IPBM). The study began by testing two RHS joint specimens with varying brace-to-chord width ratios (β) under quasi-static cyclic IPBM loading. The results showed that the final failure mode of the specimen with the large β value (β = 1.0) is the tearing of the weld near the brace root, while the specimen with the medium β value (β = 0.83) failed due to the tearing of both the weld and the adjacent chord face. The seismic performance of the X-joints depended considerably on the β value. The increase in β remarkably improved the strength of the X-joints but at the cost of energy dissipation capability, deformability and ductility. Our experimental results also demonstrated that the current code equations remarkably underestimate the flexural strength of RHS X-joints, while the modified equations that take the weld size into account can predict it well. In addition, the reason behind the experimental observation can be further explained by FE analysis and the proposed elastic-support plate analytical model.

Trapped electron mode (TEM) is the main source of turbulence predicted for the unique operation regime of a flat temperature profile under low-recycling conditions in the LTX-β tokamak, while ion temperature gradient driven turbulence may also occur with gas fueling from the edge. To investigate mainly TEM scale density fluctuations, a high spatial and time resolution 2D beam emission spectroscopy (BES) diagnostic is being developed. Apart from spatially localized density turbulence measurement, BES can provide turbulence flow and flow shear dynamics. This BES system will be realized using an avalanche photodiode-based camera and narrow band interference filter. The system can acquire data at 2 MHz. Simulations with the Simulation of Spectra (SOS) code indicate that a high signal to noise ratio can be achieved with the proposed system. This will enable sampling the density fluctuations at this high time resolution. The design considerations and system optimization using the SOS code are presented.

In this paper we propose a method of fast neutron flux estimation from a pulsed D-T neutron generator with application of single CaF2 scintillation crystal. The analysis method relies on 19F(n, α)16N threshold activation reaction having neutron energy threshold at 1.6 MeV. As a result, the 16N undergo β− decay with half-life of 7.1 s, emitting β particles with endpoint up to 10.4 MeV in the scintillator medium. Integration of the β distribution curve, preceded by calculation of (n, α) rate on F with Monte Carlo N-Particle Transport Code v6 (MCNP6) for fixed geometry, allows to estimate the neutron flux in 4π per second within few minutes.

As one of the post-translational modifications, O-linked β-N-acetylglucosamine (O-GlcNAc) modification (O-GlcNAcylation) often occurs on serine (Ser) and threonine (Thr) residues of specific substrate cellular proteins via the addition of O-GlcNAc group by O-GlcNAc transferase (OGT). Maintenance of normal intracellular levels of O-GlcNAcylation is controlled by OGT and glycoside hydrolase O-GlcNAcase (OGA). Unbalanced O-GlcNAcylation levels have been involved in many diseases, including diabetes, cancer, and neurodegenerative disease. Recent research data reveal that O-GlcNAcylation at histones or non-histone proteins may provide recognition platforms for subsequent protein recruitment and further initiate intracellular biological processes. Here, we review the current understanding of the ‘O-GlcNAc code’ mediated intracellular biological functions of downstream proteins.

The “tubulin code” combines different α/β-tubulin isotypes with several post-translational modifications (PTMs) to generate microtubule diversity in cells. During cell division, specific microtubule populations in the mitotic spindle are differentially modified, but only recently, the functional significance of the tubulin code, with particular emphasis on the role specified by tubulin PTMs, started to be elucidated. This is the case of α-tubulin detyrosination, which was shown to guide chromosomes during congression to the metaphase plate and allow the discrimination of mitotic errors, whose correction is required to prevent chromosomal instability—a hallmark of human cancers implicated in tumor evolution and metastasis. Although alterations in the expression of certain tubulin isotypes and associated PTMs have been reported in human cancers, it remains unclear whether and how the tubulin code has any functional implications for cancer cell properties. Here, we review the role of the tubulin code in chromosome segregation during mitosis and how it impacts cancer cell properties. In this context, we discuss the existence of an emerging “cancer tubulin code” and the respective implications for diagnostic, prognostic and therapeutic purposes.

With few exceptions, all living organisms encode the same 20 canonical amino acids; however, it remains an open question whether organisms with additional amino acids beyond the common 20 might have an evolutionary advantage. Here, we begin to test that notion by making a large library of mutant enzymes in which 10 structurally distinct noncanonical amino acids were substituted at single sites randomly throughout TEM-1 β-lactamase. A screen for growth on the β-lactam antibiotic cephalexin afforded a unique p-acrylamido-phenylalanine (AcrF) mutation at Val-216 that leads to an increase in catalytic efficiency by increasing kcat, but not significantly affecting KM. To understand the structural basis for this enhanced activity, we solved the X-ray crystal structures of the ligand-free mutant enzyme and of the deacylation-defective wild-type and mutant cephalexin acyl-enzyme intermediates. These structures show that the Val-216–AcrF mutation leads to conformational changes in key active site residues—both in the free enzyme and upon formation of the acyl-enzyme intermediate—that lower the free energy of activation of the substrate transacylation reaction. The functional changes induced by this mutation could not be reproduced by substitution of any of the 20 canonical amino acids for Val-216, indicating that an expanded genetic code may offer novel solutions to proteins as they evolve new activities.

The β-glucanase enzyme is an enzyme protein that can hydrolyze β-glucan, one of the main components of the fungal cell wall. This enzyme protein is produced by several bacteria, one of which is B. subtilis. The three-dimensional (3D) structure of proteins is necessary to understand their properties and functions of proteins. Enzyme proteins can be analyzed for their structure and function using in silico method. This study aims to detect the β-glucanase gene from B. subtilis W3.15 and analyze it using the in silico method. The methods in this research are homology modeling and molecular docking analyses. Modeling was carried out using the SWISS-MODEL server and docking analysis using the PLANTS 1.1 program. Modeling the β-glucanase enzyme is based on the template of the β-glucanase enzyme protein model with PDB code 3o5s. The results of sequence alignment and model visualization were quite good as indicated by the model having a Ramachandran Plot value in the favored area of 91.10 %, a MolProbity score of 0.95, and a QMEAN value of 0.90 ± 0.06. The β-glucanase enzyme model was then docked using the PLANTS1.1 program with native ligand B3P, 1,4-β-D-Glucan, D-glucose, β-D-Glucan from oats, and N-Acetyl glucosamine. The results of docking analysis showed that the β-glucan ligand (β-D-glucan from oats) used as a substrate in the cultivation of isolate B. subtilis W3.15 had a better binding energy prediction value compared to the B3P ligand, which is a natural ligand in the template proteins. Histone cell cycle regulator (HIRA) is a histone chaperone and has been identified as an epigenetic regulator. Subsequent studies have provided evidence that HIRA plays key roles in embryonic development, but its function during early neurogenesis remains unknown. Here, we demonstrate that HIRA is enriched in neural progenitor cells, and HIRA knockdown reduces neural progenitor cell proliferation, increases terminal mitosis and cell cycle exit, and ultimately results in premature neuronal differentiation. Additionally, we demonstrate that HIRA enhances β-catenin expression by recruiting H3K4 trimethyltransferase Setd1A, which increases H3K4me3 levels and heightens the promoter activity of β-catenin. Significantly, overexpression of HIRA, HIRA N-terminal domain, or β-catenin can override neurogenesis abnormities caused by HIRA defects. Collectively, these data implicate that HIRA, cooperating with Setd1A, modulates β-catenin expression and then regulates neurogenesis. This finding represents a novel epigenetic mechanism underlying the histone code and has profound and lasting implications for diseases and neurobiology.

Precise timing is crucial for many behaviors ranging from conversational speech to athletic performance. The precision of motor timing has been suggested to result from the strength of phase–amplitude coupling (PAC) between the phase of alpha oscillations (α, 8–12 Hz) and the power of beta activity (β, 14–30 Hz), herein referred to as α–β PAC. The amplitude of β oscillations has been proposed to code for temporally relevant information and the locking of β power to the phase of α oscillations to maintain timing precision. Motor timing precision has at least two sources of variability: variability of timekeeping mechanism and variability of motor control. It is ambiguous to which of these two factors α–β PAC should be ascribed: α–β PAC could index precision of stopwatch-like internal timekeeping mechanisms, or α–β PAC could index motor control precision. To disentangle these two hypotheses, we tested how oscillatory coupling at different stages of a time reproduction task related to temporal precision. Human participants encoded and subsequently reproduced a time interval while magnetoencephalography was recorded. The data show a robust α–β PAC during both the encoding and reproduction of a temporal interval, a pattern that cannot be predicted by motor control accounts. Specifically, we found that timing precision resulted from the trade-off between the strength of α–β PAC during the encoding and during the reproduction of intervals. These results support the hypothesis that α–β PAC codes for the precision of temporal representations in the human brain.

Protein β-NGF berperan dalam kelangsungan hidup sel neuron dan diferensiasi embrio aves yang diekspresikan oleh gen NGF. Informasi protein β-NGF telah diidentifikasi pada ayam dan masih terbatas dilaporkan pada merpati. Tujuan penelitian adalah memprediksi struktur tiga dimensi protein β-NGF merpati untuk menambah informasi karakter proteinnya. Protein target diperoleh dari server UniProt dengan kode akses B4ZE95. Prediksi struktur tiga dimensi dan karakterisasi protein β-NGF menggunakan server SWISS-MODEL dan program Chimera. Hasil analisis menunjukkan bahwa protein β-NGF merpati dan template memiliki nilai identity sebesar 89,91%, QMEAN 0,51 serta GMQE sebesar 0,44. The analysis showed that pigeon β-NGF protein and template had identity of 89,91%, QMEAN 0,51 and GMQE of 0,44. The interconversion of glycerol 3-phosphate and dihydroxyacetone phosphate by glycerol-3-phosphate dehydrogenases provides a link between carbohydrate and lipid metabolism and providesSaccharomyces cerevisiaewith protection against osmotic and anoxic stress. The first structure of a glycerol-3-phosphate dehydrogenase fromS. cerevisiae, GPD1, is reported at 2.45 Å resolution. The asymmetric unit contains two monomers, each of which is organized with N- and C-terminal domains. The N-terminal domain contains a classic Rossmann fold with the (β-α-β-α-β)2motif typical of many NAD+-dependent enzymes, while the C-terminal domain is mainly α-helical. Structural and phylogenetic comparisons reveal four main structure types among the five families of glycerol-3-phosphate and glycerol-1-phosphate dehydrogenases and reveal that theClostridium acetobutylicanprotein with PDB code 3ce9 is a glycerol-1-phosphate dehydrogenase.

This paper summarizes the development of a probabilistic micromechanical code for treating fatigue life variability resulting from material variations. Dubbed MICROFAVA (micromechanical fatigue variability), the code is based on a set of physics-based fatigue models that predict fatigue crack initiation life, fatigue crack growth life, fatigue limit, fatigue crack growth threshold, crack size at initiation, and fracture toughness. Using microstructure information as material input, the code is capable of predicting the average behavior and the confidence limits of the crack initiation and crack growth lives of structural alloys under LCF or HCF loading. This paper presents a summary of the development of the code and highlights applications of the model to predicting the effects of microstructure on the fatigue crack growth response and life variability of the α+β Ti-alloy Ti-6Al-4V.

Moldavian Dragonhead (Dracocephalum moldavica L.) is an annual herb. In FGBNU VILAR a new variety of Moldavian Dragonhead «Nezhnost» has been developed. From the herb of the Moldavian Dragonhead, a technology has been developed for obtaining a dry purified extract, which has received the code name «Rozmatin». By NMR (1H and 13C) and HPLC-DAD-MS using standard samples and literature data, the qualitative and quantitative composition of the dry extract "Rozmatin" from the herb of the Dracocephalum moldavica was established. Rosmarinic, caffeic and ferulic acids, rosmarinic acid 3-O-glucoside, luteolin-7-O-β-D-glucuronide, acacetin-7-O-β-D-glucopyranoside (tilianin), apigenin-7-O-β-D-glucoside (Cosmosiin), apigenin-7-O-β-D-glucuronide, chrysoeryol-7-O-β-D-glucuronide, apigenin-7-O-β-D-(6ʺ-O-malonyl) glucoside, acacetin-7-O-β-D-glucuronide, acacetin-7-O-β-D-(6ʺ-malonyl) glucoside, schizotenuin F were identified. It was found that the content of rosmarinic acid in "Rosmatin" is 56.6%, the amount of flavonoids - about 27%. The study of anti-inflammatory and gastroprotective activity of "Rozmatin" in doses of 10 mg/kg, 100 mg/kg was carried out. A dose-dependent anti-inflammatory effect with suppression of the development of the exudative phase of inflammation, as well as a dose-dependent gastroprotective effect of "Rozmatin"", significantly exceeding the similar effect of the referent drug omeprazole, was revealed. Earlier it was found that "Rozmatin" has a tonic effect on the cardiovascular and nervous systems.

Abstract The equilibria of the HL-2M tokamak, designed for steady-state operation with reversed magnetic shear and double transport barriers, are calculated using TOQ code, and the n = 1 linear ideal magnetohydrodynamic (MHD) stability of the equilibria is investigated using GATO code. To improve the beta ( β N ) limit, the MHD stability of the equilibria with different heights of internal transport barrier (ITB) and edge transport barrier (ETB) is simulated. The simulation results indicate that the ITB drives low m globalized modes while the ETB drives high m edge localized modes, and thus a moderate barrier height ratio is beneficial to MHD stability. By adjusting the heights of both barriers with different plasma shapes, as well as different locations and widths of ITBs, optimal equilibria with improved MHD stability are achieved, which always have a moderate barrier height ratio unless the ITB is extremely wide or very close to the edge. In ideal-wall conditions, when the ITB is wide enough or very close to the edge, β N limits could exceed six, or even reach seven in some special cases. Increasing elongation and triangularity, broadening the ITB, as well as moving the ITB to a large minor radius, can contribute to the optimization of the β N limit, especially when combined with the stabilizing effect from an ideal wall.

Introduction: Microbial drug resistance generated serious problems in treating infectious diseases and decreased the efficiency and efficacy of antibiotics. Investigating for inhibitors of these pathogens is helpful to decrease microbial resistance against antibiotics and to decrease high mortality rates due to the resistance of microorganisms. In this paper, quercetin, which is a natural compound with significant medicinal effects, can be used to inhibit AmpC β-lactamase class C enzymes. Materials and Methods: Molecular docking and molecular dynamic simulation were performed to determine the binding pose, structural integrity, stability, and binding energy of class C β-lactamase with quercetin using Autodock 4.2.2 software and the GROMACS 2019.6 program applying the AMBER99SB force field, respectively. Results: Molecular docking results and interaction analysis of molecular dynamics simulations indicated favorable hydrogen bonds and van der Waals interactions of quercetin with AmpC. These findings suggest that targeting β -lactamase using quercetin inhibitor analogs could provide a novel approach to treating antimicrobial resistance and could be used as a guide for future experimental studies. Conclusion: By utilizing new molecular techniques, molecular docking, and molecular dynamics simulation, this paper suggests that quercetin, which has several medicinal effects, can be used to inhibit the AmpC- β-lactamase class C enzyme.

Abstract Highly energetic stellar tidal disruption events (TDEs) provide a way to study black hole characteristics and their environment. We simulate TDEs with the Phantom code in a general relativistic and Newtonian description of a supermassive black hole’s gravity. Stars, which are placed in parabolic orbits with different β parameters, are constructed with the stellar evolution code MESA and therefore have realistic stellar density profiles. We study the mass fallback rate of the debris M ̇ and its dependence on β, stellar mass, and age as well as the supermassive black hole’s spin and the choice of the gravity description. We calculate the peak value M ̇ peak , time to peak t peak, duration of the super-Eddington phase t Edd, time t > 0.5 M ̇ peak during which M ̇ > 0.5 M ̇ peak , early rise-time τ rise, and late-time slope n ∞. We recover the trends of M ̇ peak , t peak, τ rise, and n ∞ with β, stellar mass, and age which were obtained in previous studies. We find that t Edd, at a fixed β, scales primarily with the stellar mass, while t > 0.5 M ̇ peak scales with the compactness of stars. The effect of the SMBH’s rotation depends on the orientation of its rotational axis relative to the direction of the stellar motion in the initial orbit. Encounters in prograde orbits result in narrower M ̇ curves with higher M ̇ peak , while the opposite occurs for retrograde orbits. We find that disruptions, at the same pericenter distance, are stronger in a relativistic tidal field than in a Newtonian one. Therefore, relativistic M ̇ curves have higher M ̇ peak , and shorter t peak and t Edd.

Objectives This study aimed to lay the groundwork for enhancing the ethical decision-making confidence of nurs-ing students by identifying factors influencing this competence. Methods The study participants were 180 second-year nursing students from two universities in J Province. They were surveryed from January 5 to 15, 2024 using questionnaire. The data were analyzed using descriptive sta-tistics in the SPSS/23.0 program, in addition to an Indepent t-test, Pearson correlation coefficients, and stepwise multiple regression. Results The results are as follows. First, nursing students’ ethical decision-making confidence had a significant positive correlation with ethical values (r=.597, p<.001), ethical sensitivity (r=.589, p<.001), and nursing pro-fessionalism (r=.528, p<.001). Second, the factors influencing participants’ ethical decision-making confidence were ethical values (β=.332, p<.001), ethical sensitivity (β=.220, p=.009), nursing professionalism (β=.215, p=.003), and awareness of the Korean Nurses Code of Ethics (known) (β=.177, p=.040), in that order. These varia-bles explained 45.1% of nursing students’ ethical decision-making confidence. Conclusions This study is meaningful as it identified factors that affect the ethical decision-making confidence of lower-grade nursing students. To improve lower-grade nursing students’ confidence in ethical decision-mak-ing, it is necessary to impart appropriate ethical values, improve ethical sensitivity and nursing professional in-tuition, and develop curriculum and programs for accurate recognition of the Korean Nurses Code of Ethics, tail-ored to the level of the target audience. Various learning methods that take these factors into account will be needed.

Abstract Background Recently, we described a collection of ST298 Pseudomonas aeruginosa (PA) isolates that caused a prolonged epidemic of XDR infections. Many of these contain derivatives of a new plasmid, pPABL048, that harbors an MDR integron, in1697. In1697 contains a series of antimicrobial resistance (AMR) genes, one of which is the class D β-lactamase blaOXA-10. Variants of blaOXA-10 have been described that confer both extended-spectrum β-lactamase (ESBL) and carbapenemase activity. Methods Of all ST298 isolates, three were resistant to ceftazidime (CTZ). Genomic comparison of in1697 in CTZ-resistant and CTZ-sensitive strains revealed that all three strains harbored a blaOXA-10 allele with two single nucleotide variations resulting in amino acid changes at positions 153 (F153S) and 157 (G157D). Using the NCBI database, we identified this allele as unique and defined this β-lactamase as OXA-935. OXA-935 shares the G157D variation with OXA-14 which is known to confer resistance to ceftazidime. We sought to characterize the function of OXA-935 and to determine the crystal structures of OXA-14 and OXA-935. Results Deletion of blaOXA-935 phenotypically converted all three strains to CTZ-susceptible. Expression of blaOXA-14 and blaOXA-935 conferred CTZ-resistance to laboratory PA strains PA01 and PA14. Determination of the crystal structures of OXA-14 (PDB code 7L5R) and OXA-935 (PDB code 7L5V) revealed that the F153S variant resulted in increased flexibility in the enzyme’s Ω loop. Conformational changes in the Ω loop likely contributed to the lack of carbamylation at lysine-70 (K70) observed in OXA-935. Carbamylation of K70 is known to be critical for enzymatic activity of class D β-lactamases. Conclusion OXA-935 is very similar to OXA-14; however, comparison revealed that the F153S variant has unique structural features and is functionally distinct. Despite these differences, both enzymes confer high-level CTZ resistance. As we increasingly rely on β-lactam antimicrobial therapy (e.g. ceftazidime, cefepime) and combination (e.g. ceftazidime-avibactam) therapy to treat MDR PA infections, it is critical that we continue to explore the mechanistic basis of β-lactam AMR in an effort to preserve existing treatments and design novel ones. The three-dimensional structures of two hexapeptide repeat motifs (PGQGQQ and SGQGQQ, in one letter code) in the repetitive central domain of HMW glutenin subunits are investigated by using the multicanonical simulation procedure. Ramachandran plots were prepared and analyzed to predict the relative occurrence probabilities of β-turn and γ-turn structures and helical state. Structural predictions of PGQGQQ repeat motif indicated the presence of high level of β-turns and considerable level of γ-turns. Simulations of the repeat motifs in the repetitive central domain of HMW glutenin subunits indicated that these structures take important part in the three-dimensional structures of repeat motifs.

The present work focuses on the development of computer code related to the lateral stability analysis on a jet engine aircraft model. The computer code designed in such a way, so for a given aircraft data will produce the result in term of: 1) Lateral directional dimensional stability derivatives, 2) Lateral directional responses following rudder maneuver with respect to the time, 3) The lateral direction transfer function for the rudder maneuvers, 4) The relationship between rudder deflection and lateral angle of attack (sideslip angle) β, the Euler roll angle ɸ, and Euler yaw angle ψ with respect to the time maneuvers. Through this developed computer code can make an assessment the lateral behavior for the turbojet driven light aircraft [1].

AbstractObjective:To describe the distribution of carotenoid intakes and important food sources of carotenoids in the diet of a representative population of older Australians.Design:Population-based cohort study.Setting:Two post-code areas in the Blue Mountains, west of Sydney, Australia.Subjects:We studied 2012 (86%) of the 2334 participants aged 55 + years attending the 5-year follow-up of the cross-sectional Blue Mountains Eye Study (BMES), who completed a detailed semi-quantitative food-frequency questionnaire. The intakes for five carotenoids were studied: α-carotene, β-carotene, β-cryptoxanthin, lutein and zeaxanthin combined, and lycopene.Results:The mean intake per day for each carotenoid was: α-carotene, 2675 μg; β-carotene equivalents, 7301 μg; β-cryptoxanthin, 299 μg; lutein and zeaxanthin, 914 μg; lycopene, 3741 μg; retinol, 653 μg; total vitamin A, 1872 μg retinol equivalents. β-Carotene equivalents contribute a substantial proportion of total vitamin A intake (65%) in this population. Women had slightly higher intakes than men for α-carotene, β-carotene equivalents, and lutein and zeaxanthin (P<0.05) . Carrots and pumpkin were the main contributors to α-carotene and β-carotene equivalent intakes. Orange juice, oranges and papaw were the main contributors to β-cryptoxanthin intake. Broccoli, green beans and oranges contributed substantially to lutein and zeaxanthin intake. The main contributors to lycopene intake were tomatoes and bolognaise sauce.Conclusions:Vitamin A intake in this population is high relative to the Australian Recommended Dietary Intake. Carotenoid intakes, particularly β-carotene, make a substantial contribution, particularly from fruit and vegetables. This study provides important information as a basis for examining associations between dietary carotenoid intake and eye disease in the BMES.

Objective.To determine the association of socioeconomic status [SES; education and zip code-based annual household income (Z-AHI)] and ethnicity with health-related quality of life (HRQOL) among patients with systemic lupus erythematosus (SLE).Methods.Data on 211 subjects from a cross-sectional study (LupusPRO©) using the Medical Outcomes Study Short Form-36 questionnaire to evaluate physical health scores (PCS) and mental health scores were used to obtain education and zip code. The 2000 US Census was used to obtain each zip code’s median annual household income.Results.Education and Z-AHI correlated with PCS (education standardized β = 0.17, 95% CI 0.47, 3.65, p = 0.01, r2 = 0.03; Z-AHI standardized β = 0.15, 95% CI 0.57, 8.30, p = 0.02, r2 = 0.02) on regression analysis. Z-AHI was linked to PCS, independent of education. Ethnicity was associated with PCS through disease activity and SES.Conclusion.SES is associated with HRQOL in SLE. Z-AHI and education are equally predictive surrogates of SES; however, Z-AHI, independent of education, was predictive of HRQOL. Z-AHI has less subject bias and is easily obtainable, therefore its use for future HRQOL studies is suggested.
