Resumo:

These are exciting times in the world of computer animation. Individual artists can now produce animations which in the past required teams of artists and programmers to create, but the obstacles to an individual artist can still be daunting in terms of required manual input or technical skills. The goal of this thesis is to demonstrate a simple software tool (BRAT - Blender Rapid Animation Tool) which allows an artist to create cartoon-like 3D animations using the artist's 2D drawings, at the touch of a button. It could be useful for any artist producing 3D animations. The tool is a Python script addon to the Blender 3D modeling system. It is a high level program which links to Blender's Python API library of low level calls to automate the manual work normally required by 3D artists to create an animation. The output of BRAT is a 3D animation file showing a character traversing a rectangular matrix of buildings.

This thesis compares the manual way of creating such an animation with the BRAT automated process. The comparison explains the benefits of using BRAT. This thesis also discusses the design of BRAT and compares it to some other existing 3D software tools whcih also attempt to simplify the 3D animation production process. The conclusion discusses some future enhancements to make BRAT more useful.

L’avènement des nouvelles techniques de séquençage a permis d’augmenter de façon considérable le taux diagnostique des troubles du développement intellectuel (TDI) et plus de 2000 gènes impliqués sont aujourd’hui connus. Malgré ces progrès considérables, l’interprétation des variants identifiés par les techniques de séquençage reste parfois difficile et l’histoire naturelle des TDI nouvellement décrits est souvent méconnue. Notre travail a consisté à étudier quatre formes de TDI rares de modes de transmissionvariés sur les plans clinique et génétique afin de mieux comprendre ces affections et les mécanismes qui les sous-tendent. Nous avons d’une part précisé les tableaux cliniques associés aux variations de BRAT1, CNOT3 et MTOR et avons recherché l’existence d’éventuelles corrélations phénotype-génotype pour les variations de ces gènes. D’autre part, nous avons participé à la mise en place d’un test fonctionnel permettant de reclasser les variants de signification incertaine de PQBP1
The advent of new sequencing techniques has dramatically increased the diagnostic rate of intellectual disability (ID), and more than 2,000 genes are currently known to be involved. Despite these considerable progresses, interpreting the variants identified by sequencing methods remains challenging, and the natural history of newly described ID is often poorly understood. To better understand these disorders and their underlying mechanisms, we have studied four rare forms of ID with various inheritance patterns from both clinical and genetic perspectives. On one hand, we defined the clinical pictures associated with variations in BRAT1, CNOT3 and MTOR, and we investigated the existence of any phenotype-genotype correlations. On the other hand, we contributed to the design of a functional test to reclassify PQBP1 variants of uncertain significance

It could be argued that girl's play is witnessing a drastic transformation. This alteration is fostering much debate surrounding young girls and their notion of self identity. Neil Postman (1982) argues that childhood no longer exists as it has disappeared through the mass media. Likewise, Sharon Lamb (2001, 2006) argues that young girls are continually being sold the ideal attitude and a hyper-sexualized self identity through the media messages and products they consume. Such a problematic transformation raises several concerns with regards to girlhood studies. My research asks how MGA Entertainment's Bratz dolls place identity formation into question. By exploring the aforementioned notions, my research explores girl's play and identity and looks at how it contributes to the shaping of how a girl's choice in play impacts girlhood. I argue that such a claim would be best explored and answered through interviewing young girls and their mothers.

Denne oppgaven tar for seg forankring av nedgravde rørgater. Det er undersøkt interaksjonen mellom rør og jord, først i horisontalt terreng og deretter i bratt terreng med helning over 20 grader. I rapporten er det studert beregningsmodeller som benyttes for beregning av glidestabilitet, motstand mot oppløft og global knekking. Det er sett på både modeller som benyttes i vannkraftbransjen og modeller fra andre fagområder. For å undersøke hvorvidt de ulike beregningsmodellene gjengir virkeligheten ble det i atbeidet med oppgaven utført to typer forsøk. Modellforsøk med rør av polyvinylklorid (PVC) og sand og fullskala feltforsøk med rør av glassfiberarmert plast (GRP) med pukk som omfyllingsmasse. I modellforsøkene ble følgende undersøkt: - Aksiell friksjonskraft i horisontalt og bratt terreng - Motstand mot oppløft - Stivhet i jord og knekking Forsøksplanen for feltforsøkene ble utarbeidet på grunnlag av modellforsøkene, og i disse ble det sett nærmere på: - Aksiell friksjonskraft i horisontalt og bratt terreng - Motstand mot oppløft I feltforsøkene ble det også undersøkt i hvilken grad disse kreftene er avhengig av overdekningshøyde og komprimering. Beregning av knekking ble utført med elementmetodeprogrammene Focus Konstruksjon 2D og Abaqus. Dette for å se om programmene kan benyttes til å simulere knekking av rørgater i bratt terreng. Beregningene ble sammenlignet med modellforsøkene og viste at Abaqus er det mest egnede programmet. Verdiene fra 2D-modellen i Abaqus kan benyttes til gode overslagsberegninger, men for en fullverdig simulering av knekking av nedgravde rørgater bør det gjennomføres mer avanserte beregninger. Resultatene fra modellforsøkene virker til å følge samme trend som beregningsmodellene. Likevel er det noen avvik som kan komme av unøyaktigheter i målingene fra modellforsøkene. I feltforsøkene faller resulatene fra oppløftsforsøkene innforbi de beregnede verdiene. De aksielle friksjonskreftene avviker derrimot mye fra de beregnede verdiene. Sannsynligvis skyldes dette usikkerhet omkring friksjonsfaktoren mellom rør og omfyllingsmasse, og jordtrykket rundt røret. Det er anbefalt konkrete forslag for å studere temaet nærmere.

Denne masteroppgaven omhandler forankring av nedgravde rørgater i bratt terreng. Oppgaven er en fortsettelse av et samarbeidsprosjekt mellom NVE og Institutt for vann- og miljøteknikk ved NTNU. Det siste året er det skrevet en masteroppgave og tre prosjektoppgaver om samme tema.Teorien er basert på arbeidet i de nevnte oppgavene. Det er presentert fem ulike beregningsmodeller for å beregne stabiliteten for en rørgate forankret på friksjon. Modellene blir evaluert med to ulike typer av forsøk, modellforsøk utført i laboratorium og fullskala forsøk i felt. Forsøkene er utført med rørsegmenter på 3 ? 6 m, og med diameter, d = 100, 300 og 500 mm. I tillegg er det utført laboratorieforsøk (friksjonsforsøk) for å bestemme friksjonskoeffisienten (μ) mellom rør og omfyllingsmasser (pukk). Modellforsøk i laboratorium viser at friksjonskraften mellom rør og omfyllingsmasse reduseres som en funksjon av helningen på røret. Alle beregningsmodeller ser ut til å underestimere friksjonskraften mellom rør og omfyllingsmasse for forsøk utført med horisontalt rør. En av modellene gir imidlertid gode resultater for forsøk utført med rør i 35o helning. Dette viser at en justering av beregningsmodellene med √(〖tan〗^2 (φ)-〖tan〗^2 (α)), hvor φ er friksjonsvinkelen til omfyllingsmassen og α er helningen av røret, er en god justering for å kalkulere friksjonen internt i omfyllingsmassen.Fullskala forsøk viser at beregningsmodellene overestimerer friksjonskraften mellom rør og omfyllingsmasser.Resultater fra modellforsøkene utført med horisontalt rør viser godt samsvar med resultater fra feltforsøkene når de skaleres opp med en faktor, k=2*D_fullskala/D_modell , hvor D er diameteren til rørene benyttet i de ulike forsøkene.Resultater fra friksjonsforsøk viser at friksjonskoeffisienten (μ) mellom rør og pukk er, μ > 0.60, hvilket er en høyere verdi enn hva som er vanlig å benytte i bransjen. Det er vist at fullskala forsøk, med oppsettet som er benyttet under arbeidet med denne oppgaven, kan gi usikre resultat. Det er derfor foreslått at fullskala forsøk som foretas på senere tidspunkt, utføres med samme oppsett som er benyttet i modellforsøkene.Det viktigste videre arbeidet som kan utføres ved en videreføring av oppgaven er arbeid for å undersøke skala- og laboratorieeffekter, både for modellforsøk utført i laboratorium og fullskala forsøk. En god metode for å skalere modellforsøk vil være av stor verdi for å fortsette arbeidet med å kalibrere beregningsmodellene.

The last decade has seen a dramatic increase in media output aimed at “tweens” (preadolescent girls), and the expansion more broadly of tween popular culture. This exclusively female preadolescent consumer demographic is seen to emerge alongside a heightened visibility of girls within popular culture since the mid-1990s, and continuing anxieties about girlhood in this intensely mediated environment. However, this burgeoning field has yet to be matched in academic attention. This thesis offers a timely examination of mainstream tween films, television programmes, celebrities and extra-texts from 2004 onwards, including “princess” narratives The Prince & Me (2004, 2006, 2008, 2010) and A Cinderella Story (2004, 2008, 2011), and Disney Channel programming and Original Movies, Hannah Montana (2006-2011) and Camp Rock (2008, 2010). It forges a dialogue between postfeminism, film and television, celebrity, and the figure of the tween, in order to examine how the tween is both constructed and addressed by the films and television programmes that make up tween popular culture. Tweenhood is a discursive construction emerging in the mid-1990s and coming to cultural prominence in the early twenty-first century. The tween is understood to be defined by her transitional status, her “becoming”-woman; as such, the texts that make up tween popular culture can be seen to guide the tween in her development of an “appropriately” feminine and (post)feminist identity through a rhetoric of “choice”. Such identities are predicated on revealing and maintaining an “authentic” self. The need to develop an “appropriately” feminine and (post) feminist identity whilst remaining “authentic” requires the tween to be the ideal selfsurveilling, transforming subject of neoliberalism. Celebrity plays a central role in tween popular culture, articulating the parallels between “becoming”-woman and “becoming”-celebrity. Through a combination of textual analysis, and broader discursive and contextual analysis, this thesis highlights the centrality of femininity and celebrity to the tween as a figure constructed by the texts that make up tween popular culture. It analyses the ideal tween consumer as projected by the texts. This thesis draws attention to the culturally and academically devalued subject of the construction of tweenhood within a gendered, age-specific popular culture.

Il seguente elaborato si propone di fornire un esempio di traduzione del film Brat 2 di Aleksej Balabanov, che possa fungere da base per un eventuale sottotitolaggio. Il lavoro è suddiviso in quattro sezioni: in primo luogo, una breve nota alla traduzione, pensata per favorire una lettura diretta delle battute, in cui ci si soffermerà su alcuni aspetti della lingua e della cultura russe. Le linee orizzontali demarcano i cambi di scena, ovvero il passaggio da un ambiente ad un altro. La seconda sezione si concentra in parte sull'analisi di alcuni temi e caratteristiche del film in questione (anche in relazione con il suo antecedente Brat), in parte sul regista e sull'attore protagonista Sergej Badrov. La terza sezione fornirà invece una breve delucidazione riguardo la traduzione audiovisiva. Infine, la quarta sezione è composta da una tabella a due colonne, all'interno della quale sono inserite le battute tradotte con testo a fronte. La scelta è ricaduta sull'opera in questione data la sua rilevanza all'interno del panorama cinematografico russo, ma soprattutto, data la sua originalità. Brat 2 [Fratello 2] ha la capacità di mostrare, con poche ed efficaci pennellate, le immagini di un'America diversa da quella che si è soliti rappresentare sugli schermi occidentali.

Mutations in the N-terminal region of MCPH1/BRIT1 cause premature chromosome condensation (PCC), whereby cells enter mitosis before completing DNA replication. 792 chemical compounds (CC) were selected based on the crystal structure of the N-terminus of MCPH1/BRIT1 and assayed using high throughput- high content imaging to identify CC that induced PCC. Hit validation revealed 4 potential CC, 2 of which induced high PCC at low concentrations. A screen using a human protein kinase (hPK) siRNA sub-library was performed to identify genes that induced PCC. Four hits were selected for validation, however PCC induction was not confirmed. A complementary hPK siRNA screen combined with MCPH1/BRIT1 siRNA knockdown was performed. The cell number outputs from both hPK siRNA screens were analysed to identify synthetic lethal (SL) genes in MCPH1/BRIT1-deficient cells. CDK1/CDC2, STK39, VRK1 and TTK/MPS1 were subsequently validated as potential MCPH1/BRIT1 SL genes. The expression of MCPH1/BRIT1 was examined by immunostaining in breast cancer (BC) tissue pre and post neoadjuvant chemotherapy (NACT) to determine its effect on response and survival. MCPH1/BRIT1 expression increased in response to NACT with high expression in 51.4% (36/70) of cases pre-NACT compared to 81.4% (57/70) post-NACT (p = 0.0002). Reduced MCPH1/BRIT1 expression correlated with longer overall survival (OS) pre- but not post-NACT (p = 0.017). Change in MCPH1/BRIT1 expression (from low-high) post-NACT was significantly correlated with better OS (p = 0.010). MCPH1/BRIT1 has previously been found to regulate p53 stability in BC cell lines. Notably, in this study a significant increase in MCPH1/BRIT1 staining was accompanied by a decrease in p53 staining in post-NACT samples (p < 0.0001). In conclusion, these data support the idea that CC inhibitors targeting MCPH1/BRIT1 may sensitize BC cells to chemotherapy. Additionally, genes whose inhibition could promote cell death in MCPH1/BRIT1–deficient cells have been identified as potential therapeutic targets in tumours where MCPH1/BRIT1 expression or function has been compromised.

Manual segmentation of brain tumours is a time consuming process, results often show high variability, and there is a call for automation in clinical practice. In this thesis the use of deep convolutional networks for automatic glioma segmentation in MRI is investigated. The implemented networks are evaluated on data used in the brain tumor segmentation challenge (BraTS). It is found that 3D convolutional networks generally outperform 2D convolutional networks, and that the best networks can produce segmentations that closely resemble human segmentations. Convolutional networks are also evaluated as feature extractors with linear SVM classifiers on top, and although the sensitivity is improved considerably, the segmentations are heavily oversegmented. The importance of the amount of data available is investigated as well by comparing results from networks trained on both 2013 and the greatly extended 2014 data set, but it is found that the method of producing ground-truth was also a contributing factor. The networks does not beat the previous high-scores on the BraTS data, but several simple improvement areas are identified to take the networks further.

Studien finner att 3D-nätverk har generellt bättre resultat än 2D-nätverk, och att de bästa nätverken har förmågan att ge segmenteringar som liknar mänskliga segmenteringar. ConvNets utvärderas också som feature extractors, med linjära SVM som klassificerare. Den här metoden ger segmenteringar med hög känslighet, men är också till hög grad översegmenterade. Vikten av att ha mer träningsdata undersöks också genom att träna på två olika stora dataset, men metoden för att få fram de riktiga segmenteringarna har troligen också stor påverkan på resultatet. Nätverken slår inte de tidigare rekorden på BraTS, men flera viktiga men enkla förbättringsområden är identifierade som potentiellt skulle förbättra resultaten.

Familial history is the strongest risk factor for developing ovarian cancer (OC), and a significant contributor to breast cancer risk. Most hereditary breast cancers and OCs are associated with mutation of the tumor suppressor Breast and Ovarian Cancer Susceptibility Gene 1 (BRCA1). Studying risk-associated BRCA1 truncation mutations, such as the founder mutation 185delAG, may reveal signaling pathways important in OC etiology. Recent studies have shown novel BRCA1 mutant functions that may contribute to breast and OC initiation and progression independent of the loss of wtBRCA1. Previously, we have found that normal human ovarian surface epithelial (HOSE) cells expressing the 185delAG mutant, BRAT ( BRCA1 185delAG Amino Terminal truncated protein), exhibit enhanced chemosensitivity and up-regulation of the OC-associated serpin, maspin. In the current study, I identify an additional target of the BRAT mutation, matrix metalloprotease 1 (MMP1), a key player in invasion and metastasis. BRAT-expressing HOSE cells exhibit increased MMP1 messenger RNA (mRNA) by real time PCR and protein by Western blotting. Pro-MMP1 levels are also higher in conditioned media of BRAT-expressing cells and HOSE cell lines derived from BRAT mutation carriers. c-Jun is critical for BRAT-mediated MMP1 up-regulation, as siRNA knockdown diminishes MMP1 levels. Luciferase reporter constructs reveal that activator Protein 1 (AP1) sites throughout the distal end of the promoter contribute to BRAT-mediated MMP1 expression, and basal activity is mediated in part by an AP1 site at (-72). Reporters containing a single nucleotide polymorphism (SNP) associated with OC risk and progression yield increased activity that is further enhanced in BRAT cells. Interestingly, BRAT-mediated changes in chemosensitivity and gene regulation are not recapitulated in a normal breast epithelial or breast cancer cell model. This suggests tissue-specific mutant BRCA1 functions may contribute to breast and ovarian tissue specificity of BRCA1 mutation-associated cancer risk and also to differential breast and ovarian cancer risk and penetrance associated with specific mutations. Early molecular and cellular changes such as MMP1 up-regulation in the ovarian surface epithelium of BRCA1 mutation carriers may promote OC initiation and progression and represent a step forward on the continuum of cellular malignancy. Further investigation is warranted, as elucidating these early changes will aid in identification of potential screening and treatment strategies.

Telomeres are fundamental structures found at the end of all eukaryotic chromosomes that function to protect the end of chromosomes from end-to-end fusion, erosion and subsequent telomere dysfunction. Telomerase and alternative lengthening of telomere (ALT) mechanisms maintain the telomeres by compensating natural telomeric loss. ALT is found to be present in 15% of human tumours lines and it may be expressed at low levels in the normal mouse tissues. However, the exact mechanism behind ALT depression and/or activation in the mammalian cells is not fully understood. Previous studies have highlighted the role of BRCA1 in telomere dysfunction. Also, it has recently been shown that BRCA1 co-localises at telomeres in the ALT + human cells through BLM and Rad50. However, it is still unclear whether BRCA1 plays a direct role on telomere length maintenance and integrity. The aim of this project was to examine the role of BRCA1 in telomere maintenance associate with ALT in BRCA1 defective mammalian cells. Therefore to achieve this, we have set up series of experiments to look at, (a) hallmarks of ALT activity at the cytological level, (b) measuring of ALT activity using biochemical and immunocytochemistry techniques and (c) understanding the role of BRCA1 in DNA damage response mechanism and telomere dysfunction. Firstly, we found elevated levels of recombination at telomeres in the two human BRCA1 carrier cell lines and mouse embryonic stem cell with deficiency in Brca1-/-. Secondly, our data showed that human and mouse BRCA1 defective cells are significantly more sensitive to ionizing radiation in line with the DNA repair function of BRCA1. Moreover, we found persistent DNA damage at telomeres in the BRCA1 defective environment when after exposure of cells to ionizing radiation. Thirdly, we found evidence of ALT activity in some mouse cell lines, and elevated ALT in mouse cells defective in Brca1. Finally, we examined some other ALT markers using immunofluorescence. Our data indicate differences between human and mouse cells in regulating ALT. Taken together data presented in this thesis revealed that (i) BRCA1 plays a major role in telomere maintenance and defective BRCA1 mammalian cells show evidence of telomere dysfunction and telomere length shortening in line with previous publish data, (ii) BRCA1 defective mouse cells have elevated levels of ALT, (iii) the mouse lymphoblastoid LY-S cells have complete absence of ALT.

The dissertation deals with the Municipal Theatre of Göteborg with special reference to the period between 1967 and 1971, during which the group theatre developed and paved the way for a repertoire of social criticism. The theatre debate is viewed against the background of the socio-economic, ideo­logical, and cultural development of the 1960,s. After describing conditions at the Municipal Theatre of Göteborg under Mats Johansson's management from 1962 onwards, the study concentrates on analysing three group theatre plays produced by Lennart Hjulström: Flotten (spring 1967) by Kent Andersson, Hemmet (autumn 1967) by Kent Andersson and Bengt Bratt, and Sandlådan (autumn 1968) by Kent Andersson. The first play thematizes the ideological confusion of the middle-aged generation, the second the disappointment of the aged at the betrayal of socialist ideals, and the third the manipulative repression of children's education. A series of stagings with strong elements of social criticism caused a long and intense debate in the summer of 1969 about both the repertoire and the internal conditions of the Municipal Theatre. In spring, 1971, Kent Andersson and Bengt Bratt wrote Tillståndet (an ambiguous title meaning both 'condition' and 'permission'), by which the authors intended to ex­pose the repressive tolerance of bourgeois society. This play functions as an evaluation of both the group theatre period and the situation of the theatre as a whole, and it is therefore analysed in a metaperspective. It also functions as a summary of the criticism of the Swedish welfare state.

Brachial plexus Injury (BPI) occurs following trauma to the nerves as they exit the spinal cord in the neck. Dependent on the site and degree of the initial trauma, people with BPI display a board spectrum of ability to use their affected arm to perform day-to-day activities. BPI is a heterogeneous condition that has a significant impact on how the individuals use their arm. Some may only achieve simple stabilising or carrying tasks; others may undertake more complex movements that use the whole arm and hand. Historically, outcomes following adult traumatic BPI have been discussed in terms of impairments such as strength or range of motion. Despite evidence of good outcomes, people still report on-going dissatisfaction with the day-to-day use of their arm. Using the International Classification of Functioning Disability and Health (ICF) as a framework, a systematic review of the literature was undertaken to identify all patient-reported outcome measures that predominantly assessed activity. Two patient-reported outcome measures were identified, yet neither had been psychometrically evaluated for BPI. Further, they did not appear to include the range of activities required to assess the full spectrum of ability seen in adults with BPI, particularly for those with limited arm use. Seventeen additional outcome measures were identified that had been developed by experts solely for a research purpose, lending support for a new BPI outcome measure in the clinical setting.

L'objectif de cette thèse est d'étudier trois romans écrits par des auteurs anglophones du Pakistan ou de l'Inde, à savoir Bapsi Sidhwa, Bharati Mukherjee et Hanif Kureishi. On pourrait être tenté de placer les trois écrivains de cette étude dans la catégorie «littérature des immigrants». Ils écrivent tous à un moment de migration de masse lorsque l'idée de «choc culturel» parmi les peuples occidentaux commence à être plus évidente. Les trois écrivains sont affectés par des thèmes qui apparaissent seulement de manière marginale dans le débat évoqué ci-dessus, l'accent étant principalement mis sur les difficultés culturelles et sociales des femmes dans la société indo-pakistanaise. Quant à Kureishi, la polarisation mentionnée ci-dessus suppose un accent très différent, impliquant la situation d'un Asiatique né et élevé dans la société occidentale. Dans cette évaluation globale du contexte idéologique et historique commun aux trois écrivains, il sera important d'examiner les attitudes spécifiques adoptées par chaque écrivain par rapport à son expérience personnelle. L'objectif principal de cette étude sera donc thématique, en se concentrant sur les préoccupations spécifiques de ces écrivains et sur la manière dont cela se manifeste dans leur représentation particulière des tensions en jeu
The objective of this thesis is to study three novels written by English-speaking authors of Pakistan or India, namely Bapsi Sidhwa, Bharati Mukherjee and Hanif Kureishi. One might be tempted to place the three writers of this study in the category of "literature of immigrants." They all write at a time of mass migration when the idea of "cultural shock" among Western peoples begins to be more evident. All three writers are affected by themes which appear only marginally in the debate evoked above, much of the emphasis being on the cultural and social difficulties of women in Indo-Pakistani society. As for Kureishi, the polarization mentioned above assumes a very different emphasis, involving the situation of an Asian born and brought up inside Western society. Within this overall assessment of the ideological and historical context common to all three writers, it will thus be important to examine the specific attitudes adopted by each writer in relation to his or her own personal experience. The main focus of this study will therefore be thematic, centering on these writers’ specific preoccupations and the way this is seen in their peculiar depiction of the tensions at stake

The thesis follows one of the supporting characters from the New Testament, James, the brother of the Lord. The first chapter deals with the meaning and occurrence of the name James in both biblical and non-biblical sources. The following second chapter deals with the canonical biblical texts that depict fragments of James' life. However, many questions remain about the closer characterization of our wanted person James and the form of his earthly family. The thesis also examines whether James could or could not really be the brother of Jesus of Nazareth. Through the analysis of texts we look for the answer that canonical texts give us.Subsequently, in the third chapter attention is also directed to the apocryphal and non-biblical sources that govern us about our James. In connection with the wanted James is mentioned and the first church community in Jerusalem, in which James played a significant leadership role. The aim of the thesis is to describe the character of James, the brother of the Lord, by means of all analyzed sources. In conclusion, a summary possible solution of questions concerning James, the Lord's Brother is presented. Bone morphogenetic protein induces signals through the stimulation of serine/threonine kinase receptors which phosphorylated BMP signaling mediators, Smad1, Smad5 and Smad4. The phosphorylated Smad1/5/4 complex is then translocated to the nucleus and regulates the BMP target genes. BMP receptor associated molecule 1 (BRAM1) is a molecule that associates with BMP receptor IA. However its role in BMP signaling is still unclear. Thus, the specific aims of this study are examined (1) if BRAM1 affects BMP signaling; (2) If so, are the Smad proteins classical BMP signaling mediators involved? In this study, we demonstrate that BRAM1 interacts with BMP receptor by co-immunoprecipitaion, Western blot and confocal microscopy. The results show that BRAM1 associates with wild type BMP receptor as well as the constitutively activated BMPR. The Mynd domain of BRAM1 is the main region for BRAM1-BMPR interaction. Confocal microscopy study showed that BRAM1 co-localized with BMPR and Smad5, but not with Smad1. Furthermore, BRAM1 prevents the nucleus translocation of Smad4 in response to BMP treatment. Thus the results suggest that BRAM1 may be involved in BMP signaling. This study will provide more information regarding the function of BRAM1.

This case presents an overview of the California-based Micro Games of America (MGA) Entertainment, which is a consumer entertainment products company, engaged in innovative lines of proprietary and licensed products including toys and games, dolls, consumer electronics, home decor, stationery and sporting goods. It had more than 200 licences. In 2001, MGA launched a fashion doll called ‘Bratz’, and it sold 150 million Bratz dolls all over the world. Bratz line surpassed the legendary brand ‘Barbie’—Mattel, Inc.’s flagship brand—in a short span of time through many innovative marketing strategies and different product placement, roll-outs, tie-ins and other promotional tactics, despite facing many challenges, such as, fast-changing demographics, shorter product life cycle (PLC) and negative perceptions about brand. This case can be used to address two issues: first, structural change drivers and trends that shaped the toy industry in developed economies and, second, how to develop effective marketing strategies for product with shorter PLC in highly product-driven market?

BRAM1, bone morphogenetic protein receptor (BMPR) associated molecule 1, is a cytoplasmic protein consisting of 198 amino acids. BRAM1 shares 99% homology with the MYND domain-containing carboxy terminus of a tumor suppressor protein, BS69, which interacts with adenovirus E1A and Epstein-Barr virus-encoded nuclear protein EBNA2 through the MYND domain. Interaction of BS69 with E1A or EBNA2 inhibits viral oncoprotein-mediated transactivation. On the other hand, BRAM1 is a cytoplasmic protein and interacts with BMPR. To date, the biological role of BRAM1 remains unclear. In this thesis work, we evaluate the physiological role of BRAM1 in zebrafish model system. zebrafish bram 1 cDNA was generated by bioinformatic search of zebrafish EST database, followed by 5’ and 3’ RACE analysis. We demonstrate that zebrafish BRAM1 associates with C-terminal region of zebrafish BMP receptor through MYND domain in vitro and in vivo. Interaction between zebrafish BRAM1 and zebrafish BMP receptor blocked BMP signaling pathway. In zebrafish, bram1 is a maternal factor and its expression profile is coordinated with bmp4 and bmp receptor in early developmental stage. bram1 was expressed throughout the embryos before blastula stage as examined by whole mount in-situ hybridization analysis, and was expressed ubiquitously in different tissues such as brain, heart, muscle, ovary, and especially ovary as examined by northern blot assay. Using antisense morpholinos to knock down BRAM1 resulted in defects in heart looping and curly tails. This phenotype of bram1 morphants was similar to phenotype of mutants involved in Bmp signaling. Injection of constitutively active bmp receptor mRNA resulted in defects in heart looping. Overexpression of BRAM1 reversed the heart position from no looping (N-looping) to rightward looping (D-looping). Further analysis of zebrafish left-right asymmetry genes incluidng cyclops, southpaw, lefty1, lefty2, and pitx2c demonstrate that BRAM1 affects the zebrafish left-right asymmetry. As zebarfish midline is critical for development of left-right asymmetry, we hypothesize that BRAM1 may have earlier effect in regulating midline. Indeed, we show that expression of flh and gsc in bram1 morphants is reduced. These findings indicate that BRAM1 may function as a negative regulator of Bmp signaling to participate in the midline development during early cell fate decision. As Bmp signaling involves interaction of BMP and BMPR, and its downstream Smad mediators, BRAM1 may affect the BMP signaling by inhibiting Smad signaling. As tested in BALB/c3T3 and COS7 cells, mammalian BRAM1 (mBRAM1) inhibit Smad signalling pathway. mBRAM1 interacted with Bmp receptor type IA through MYND domain in the absence or presence of Bmp2 treatment. mBRAM1 interacted with Smads proteins, especially Smad4 and Smad5. Such interactions blocked the nuclear translocation of Smad4 or Smad5 in Bmp2-mediated manner. Thus, mBRAM1 inhibits Smad signaling through preventing Smad4 and Smad5 to translocate into nucleus. In conclusion, these results suggest that BRAM1 may function as a negative regulator of BMP-mediated biological functions.

The magazine is the most important printed periodical medium of the Evangelical Church of Czech Brethren which is the largest non-catholic church in the Czech Republic. The diploma thesis introduces the Evangelical Church of Czech Brethren as the second largest church and the largest prostestant church in the Czech Republic, and besides describing its contemporary work in the Czech society it handles also its history. Further it deals with the history and development of the Český bratr magazine since its creation in 1924 till today. Also other printed media given out by this church are described, and also the space is mentioned that Czech media of public service devote to religious topics. The thesis will treat the topic of the abolitionist movement from the perspective of social, cultural and literary history from the beginnings until the abolition of slavery in British colonies in the Caribbean in 1833 with the Slavery Abolition Act. The thesis will focus on the discourse of race and slavery. The chosen authors represent different opinions and perspectives as the discussion will focus on sentimental poetry, travel writings as well as slave narratives. The chief aim is to identify and define the strategies of abolitionist discourse and the rhetorical practices which it employed especially in shaping the image of Africans and how the hegemonic discourse of sentimentalism influenced their writing. Latent membrane protein 1 (LMP1), a major oncoprotein encoded by Epstein-Barr virus (EBV), has been shown to be involved in cellular signaling pathways. In this thesis, we tried to identify new targets of LMP1 signaling that may help to explain the role of LMP1 in non-lymphocyte cell transformation. Using yeast two-hybrid screening method with the C-terminal sequence of LMP1 as the bait, we found that BMP receptor-associated molecule 1 (BRAM1) associated with LMP1 in vitro and in vivo. This finding was supported by cotransfection experiments and examined by immunoassays and confocal microscopy. The association involves mainly the C-terminal half, the MYND domain, of BRAM1 and the CTAR2 region of LMP 1. The latter is critical for LMP1-mediated NF-kB and JNK signaling pathways, suggesting the role of BRAM1 in LMP1 signaling. To test this hypothesis, we examined the effect of BRAM1 on the induction of NF-kB and JNK activation by LMP1. We demonstrate that BRAM1 interferes with LMP1-mediated NF-kB activation in BALB/c3T3 cells. Since LMP1-induced NF-kB activation through the NIK-IKK-IkB pathway, we thus examined the protein level of total and phosphorylation form of IkBa in the presence and absence of BRAM1. The results indicated that BRAM1 decreased the phosphorylation of IkBa and delayed the degradation of total IkBa in the presence of LMP1. Since the CTAR2 region interacts with the TNF-a receptor-associated death domain protein (TRADD), it is interesting to find that BRAM1 also interferes with NF-kB activation mediated by TNF-a. Since the CTAR2 region of LMP1 is critical for LMP1-mediated JNK activation, we examined the effect of BRAM1 on the JNK activation mediated by LMP1, TNF-a treatment and overexpression of LTbR. We found that BRAM1 plays a role either in LTbR-mediated or in TNF-a mediated JNK activation but not in LMP1-mediated JNK activation. To further examine the role of BRAM1 in LMP1 signaling, we observed that BRAM1 disrupts the resistance of LMP1-expressing cells to TNF-a-induced cytotoxicity. Thus, we propose that BRAM1, a molecule uniquely associated with LMP1, functions as a negative regulator of LMP1-mediated biological functions.

The key equations of BiCGStab are summarised to show its connections with Pade and vector-Pade approximation. These considerations lead naturally to stabilised vector-Pade approximation of a vector-valued function (VPAStab), and an algorithm for the acceleration of convergence of a linearly generated sequence of vectors. A generalisation of this algorithm for the acceleration of convergence of a nonlinearly generated system is proposed here, and comparative numerical results are given.

abstract: American families have been coming to South Korea accompanying active duty service members or Embassy employees since before the Korean War. While their numbers were originally smaller, they continue to increase as South Korean assignments undergo "tour normalization", a transition from a location intended for service members to come alone for one year to a location where service members come accompanied by their family and stay for longer periods of times. The U.S. maintains a large presence in South Korea as a deterrence against possible threats from North Korea. Despite establishment of an armistice at the conclusion of the Korean War, a constant state of potential threat was created. This paper will examine what affect the recent rise in tension between North Korea and South Korea has on the American children living in South Korea with their active duty service member parent(s).

Chromosomal instability (CIN) has been established as one of the hallmarks of cancer, which is prevalent in most of the solid and advanced tumours. CIN enhances genetic heterogeneity in cancer cells. This heterogeneity provides selective advantages to cancer cells against the drugs and the therapies, which are linked to poor prognosis and relapse of cancer. Altered metabolism is another hallmark of cancer, which is being targeted for cancer therapy. In this thesis, I have discussed the therapeutic effect of targeting metabolism in CIN cells and CIN tumours. Chapter 1 is my introduction in which I have reviewed cancer, its therapy, CIN, its types, mechanisms, causes, and therapeutic targeting of CIN. I also review cancer metabolism, its targeting for the treatment, and targeting metabolism in CIN cells. Chapter 2 is a published review article about Drosophila being a model for CIN. In this article I have discussed different CIN models and their limitations, then I described Drosophila as a model for CIN studies. I later discussed different Drosophila CIN model systems which have been studied to understand CIN and cancer. As Drosophila has been extensively studied for CIN and cancer therapy, our lab has focused on targeting CIN cells in Drosophila. In an earlier study (Shaukat et al, 2012) it was found metabolic candidates such as Pas kinase and phosphofructokinase could be crucial for CIN cell survival. Chapter 3 is a further screening of metabolic candidates. We found few potential targets from all the major metabolic pathways whose knock down can specifically kill CIN cells. It was found, mitochondrial activity and oxidative stress was high which induced DNA damage and apoptosis in CIN cells targeted by these metabolic alterations. In chapter 4, I discuss the application of the selected candidates on CIN tumours. We further explain how one of my metabolic candidates stopped the tumour growth. This chapter also discusses the mechanism of ROS (reactive oxygen species) production and implications of high NADH levels in CIN cells, which was deficient in our earlier studies. Chapter 5 is my discussion in which I have collectively discussed my results, the significant of my work, my current model, and future directions. In appendix 1 I have presented a published review article on the role of JNK in response to oxidative DNA damage. This chapter encompasses activation of JNK by ROS, outcomes of JNK in response to ROS. Appendix 2 has figures for SOX drug and ovary numbers of the hosts.
