A Dictyostelium membrane fraction rich in vacuolar proton pumps, previously described by Nolta et al. (J. Biol. Chem. 266, 18,318-18,323, 1991), was used as the immunogen for production of monoclonal antibodies. We obtained antibodies that recognized polypeptides of 100 kDa and 68 kDa, corresponding to the two largest subunits of the vacuolar proton pump. In indirect immunofluorescence experiments, these two subunits were located on an interconnected collection of tubules and vacuoles. On frozen thin sections they were found principally on membranes of vacuoles and collections of small vesicles typically located just internal to the plasma membrane. These vesicles and vacuoles had electron-translucent lumens. No other structures in axenically grown Dictyostelium cells were labeled to a significant extent by these two antibodies. Using an affinity-purified antibody to calmodulin and a monospecific antibody to the B subunit of the chromaffin granule vacuolar ATPase, markers known to label the membranes of the contractile vacuole complex in Dictyostelium (Zhu and Clarke, J. Cell Biol. 118, 347–358, 1992; Heuser et al., J. Cell Biol. 121, 1311–1327, 1993), we showed that the 100 kDa and 68 kDa subunits had the same distribution as these two markers. Co-localization was seen in both interphase and mitotic cells. Thus, our results support the conclusion that vacuolar proton pumps are located principally on the membranes of the contractile vacuole complex in Dictyostelium. In addition, in indirect immunofluorescence experiments, these monoclonal antibodies provided improved images of the organization of the contractile vacuole system.

Abstract We recently identified a human minor histocompatibility (H) antigen encoded by the UDP glycosyltransferase 2 family, polypeptide B17 (UGT2B17) gene. UGT2B17 is a metabolic enzyme that is highly expressed in liver and colon, and at low levels in hematopoietic cells. UGT2B17 conjugates exogenous and endogenous compounds including steroid hormones and facilitates their elimination. The immunogenicity of the UGT2B17 was due to differential expression of the protein in donor and recipient cells as a consequence of a homozygous deletion of the UGT2B17 gene in the recipient. Deletion of this gene in a subset of normal individuals suggested two mechanisms by which UGT2B17 could affect the outcome of allogeneic BMT. First, the expression of UGT2B17 in recipient but not donor cells could provide minor H antigens and increase GVHD. Second, the presence of UGT2B17 in the donor or recipient could affect the metabolism, function, or toxicity related to endogenous hormones or drugs administered with BMT. The purposes of this study were to determine the frequency of homozygous UGT2B17 deletion in healthy individuals and patients with hematological disease and whether a deficiency of UGT2B17 in donor and/or recipient affected the outcome of allogeneic BMT. A total of 435 recipients were selected from the patients who received unrelated BMT through the Japanese Marrow Donor Program between January 1993 and March 2000. Selection criteria were (1) matched at HLA-A, B, C and DRB1 genotypes, (2) unmanipulated marrow graft, (3) cyclosporine A or tacrolimus as GVHD prophylaxis, and (4) available DNA was stored. DNA from a total of 358 of the donors was also available. Homozygous deletion of the UGT2B17 gene determined by SSP-PCR was found in 320 (84.8%) of 377 donors and 358 (82.3%) of 435 patients (P=0.32, χ2 test). These frequencies of UGT2B17 deletion were substantially higher than the 11% observed in the Caucasian population in a previous study. A multivariate analysis (Cox proportional hazard model) showed no significant association between UGT2B17-mismatch in the GVHD direction and the incidence of acute GVHD (grade II-IV or III-IV), chronic GVHD, relapse, or survival. However, a deletion of UGT2B17 in the donor was a favorable factor for transplant-related mortality (TRM) (relative risk, 0.530; 95% CI, 0.334 to 0.841; P=0.007), survival (0.567; 0.387 to 0.831; P=0.036) and disease-free survival (DFS) (0.647; 0.443 to 0.945; P=0.024). We also analyzed these outcomes in the subset of patients with standard-risk malignant disease by the Kaplan-Meier method. TRM, survival, and DFS at eight years after transplantation were 23.7% for patients transplanted from a UGT2B17-deleted donor vs 52.9% for patients with a UGT2B17-positive donor (P=0.001); 68.3% vs 37.5% (P=0.0009); and 66.9% vs 37.5% (P=0.0009), respectively. These data suggest that the UGT2B17 enzyme in donor-derived blood cells may affect the metabolism of exogenously administered or endogenous molecules and adversely influence the outcome of unrelated BMT. Analysis in another patient population will be important to confirm our results.

Эндофуллерены, содержащие один или несколько атомов металла внутри углеродного каркаса (металлофуллерены), представляют большой практический интерес в связи с возможностью создания на их основе эффективных контрастирующих агентов для магнитно-резонансной томографии (МРТ), антиоксидантных и противораковых средств. Данные соединения могут быть также использованы в спинтронике для создания наноразмерных электронных устройств. В настоящей работе в рамках теории функционала плотности произведен расчет структурных, электронных и термодинамических характеристик эндофуллеренов металлов подгруппы скандия с числом инкапсулированных атомов от одного до семи в газовой фазе. Описаны стабильные структуры с симметриямиCs, C2, C3 и Ci, соответствующие позициям, занимаемым атомами металла внутри каркаса фуллерена. Установлен теоретический предел числа атомов металла, при котором структура эндофуллерена сохраняет устойчивость – шесть атомов для скандия, четыре для иттрия и три для лантана. Расчет показывает, что наиболее устойчивыми являются структуры с двумя и тремя инкапсулированными атомами. Описана зависимость между числом инкапсулированных атомов металла и характером распределения электронной плотности. Общий заряд на инкапсулированном металлическом кластере положителен для соединений Me@C60 – Me3@C60, слабо положителен для Me4@C60(отдельные атомы имеют отрицательный заряд) и отрицателен для соединений Me5C60 – Me6@C60. Описан эффект спиновой утечки для структур с основным дублетным спиновым состоянием. Для соединений с тремя и более инкапсулированными атомами данный эффект незначителен, что указывает на нецелесообразность создания контрастирующих агентов для МРТ на их основе. ЛИТЕРАТУРА 1. Kroto H. Ammonia is an indispensable component of modern fertilizers and a key nitrogen source in chemical synthesis, making it one of the highest-volume commodity chemicals.1–3 However, its synthesis via the Haber-Bosch process contributes significantly to global carbon emissions, motivating the development of alternative pathways for ammonia synthesis.4–6 Of many alternative methods, an electrochemical lithium-mediated process is one of the most promising. Since its initial implementations,7–9 novel cell designs10–12 and electrolytes13–15 have greatly improved the selectivity and productivity of lithium-mediated electrochemical ammonia synthesis (LiMEAS), but questions persist regarding the mechanism of nitrogen fixation and the role of lithium in this process (Figure 1a). In particular, many in the literature have alluded to the possible importance of lithium passivation species, commonly known as the solid electrolyte interphase (SEI), but their small length-scale and reactivity make direct observation challenging.11,13,15 In this work, we leveraged a multiscale approach to reveal connections between device-scale performance and nanoscale passivation behavior in LiMEAS. We used four model systems, varying the presence of proton donor (no proton donor or 0.1 M ethanol), and the feed gas (Ar or N2), which allowed us to examine the influence of the two reactive components in LiMEAS which are not usually present for lithium metal electrodeposition in the literature: proton donor and nitrogen. For each model system, we quantified major reaction products (Figure 1b, f). Ammonia and lithium nitride were measured by the salicylate colorimetric assay,16 hydrogen by online gas chromatography (GC), and residual lithium by galvanostatic stripping. We used scanning electron microscopy to observe microscale morphology, and to gain deeper insights into nanoscale surface morphology, we implemented cryo-electron microscopy (cryo-EM) (Figure 1c-d, g-h). Cryo-EM is a powerful technique that enables high-resolution observation of beam-sensitive metallic lithium and SEI materials by holding them at cryogenic temperatures, which preserve their native state.17 In addition to nanoscale morphological information, cryo-EM provided chemical information in the form of selected-area electron diffraction (SAED), energy-dispersive X-ray spectroscopy (EDS), and electron energy loss spectroscopy (EELS), which were paired with X-ray photoelectron spectroscopy (XPS) to develop an understanding of the chemistry of the interphase. Our results indicate that the proton donor governs reactivity toward nitrogen reduction in LiMEAS via its influence on the SEI. In the absence of proton donor, the lithium deposit forms a passivating, mosaic-structured SEI that preserves the deposited lithium, regardless of whether the feed gas is argon or nitrogen (Figure 1e). With the addition of ethanol, the surface changes significantly. The resultant SEI does not passivate the lithium, allowing it to react with nitrogen and other electrolyte components (Figure 1i). This permeable SEI appears to be mosaic in structure, with organic phases dominated by ethanol breakdown products, and its formation is key to the functionality of lithium-mediated ammonia synthesis. These findings can help guide the development of optimal SEIs for selectivity and stability of LiMEAS, and beyond that, offer insights into the characteristics of the lithium SEI at a reactive rather than passivated interface. This paper aims at analyzing the use of scientific electronic papers and journals in the didactics material of the specialization Course on Educational Technologies from UFSCar. The bibliographical analysis focused on the development and legitimacy of scientific journals. The purpose of the documental analysis on the didactic material, composed by 48 volumes, was to verify the kinds of material cited and suggested in the multimedia study guides. The data show that books are the most cited material, whereas other electronic materials are the most suggested kind of complementary material. The international scientific community considers papers published in scientific journals as the main and most legitimate mean of scientific knowledge sharing. They are the third most cited and suggested kind of material by the author-docents. This result indicates dissociation between the course didactics material and its innovative proposal of hybrid, flexible and integrated education. It is also dissociated from the international trends of scientific knowledge legitimacy. Such dissociation, commonly found in human sciences, may jeopardize the quality of the Brazilian science and its integration in the international scientific community.ResumoEste artigo pretende analisar a utilização de artigos e periódicos científicos eletrônicos no material didático do curso de especialização em Educação e Tecnologias da UFSCar. Para tanto, foi feita análise bibliográfica sobre o desenvolvimento dos periódicos científicos e a questão da legitimidade conferida a esse tipo de publicação. A análise documental do material didático do curso, composto por 48 volumes, teve como objetivo avaliar o tipo de material citado e indicado nos guias de estudo multimídia. Os dados levantados mostram que os livros são o material mais citado pelos autores do material didático, enquanto outros tipos de material eletrônico compõem a maioria das indicações de material complementar. Os artigos publicados em periódicos científicos, considerados pela comunidade científica internacional o principal e mais legítimo meio de divulgação de ciência, aparecem como terceiro tipo de material mais citado e indicado pelos docentes autores. Esse resultado aponta uma dissociação do material didático do curso e a sua proposta inovadora de formação híbrida, flexível e integrada, bem como as tendências internacionais de legitimação do conhecimento científico.
