The monomer 3-hexylthiophen-2,5-dicarboxaldehyde was synthetized from precursor 3-hexylthiophene by VilsmeierHaack reaction. Three p-type semiconductor polymers (PHT-P, PHT-B and PHT-H) were synthetized by aldol condensation from this monomer with three different ketones (propanone, butanone and hexanone). Three composites were created by adding PbS/ZnS nanoparticles to these polymers. Light absorption increased to the red from polymers to composites. Electrical conductivity increased from 10% S/cm in polymers to 10! S/cm in composites.

306 Background: S is the first systemic agent that has been shown to prolong survival in pts with CP A advanced HCC. However, its safety and efficacy have not been extensively evaluated in pts with CP B cirrhosis. Methods: We performed a descriptive analysis on pts with histologically documented advanced HCC and CP A/B cirrhosis, enrolled in a multicenter phase II randomized, open-label trial (data reported elsewhere), in order to assess the feasibility and efficacy of treatment with S in CP B pts. Written informed consent was obtained from all pts. Results: From April 2007 to July 2008, 297 pts were prospectively treated with S 400 mg bid, 234 (78.8%) CP A, 63 (21.2%) CP B. 232 pts were male (76%), median age was 68.3 yrs (range 19.8-89.2 yrs), 217 pts had no extra hepatic disease (73.1%). The two subgroups, according to CP class A or B, were homogeneous for all considered parameters. Median treatment duration was >3 months (mos) for 59.4% of CP A pts and for 27% of CP B pts (p <0.001). Median PFS for the total population was 3.9 mos, 4.3 mos for CP A pts and 2.1 mos for CP B pts (p<0.001). Median OS was 10 mos for CP A pts and 3.8 mos for CP B pts (p<0.001). Adverse events (all grades) were similar in type and incidence for CP A and B pts, with fatigue, stomatitis, diarrhea and weight loss having the highest incidence (44%, 41%, 38% and 38%, respectively). Specifically, no differences in terms of grade 3-4 events have been documented between the two groups. Conclusions: This study supports the feasibility of S also in advanced HCC pts with CP B status. Tolerability data suggest that pts with CP B status might potentially be treated safely with S for its potential survival benefits. Further prospective trials, specifically designed to investigate S in CP B pts, are eagerly advocated.

Fundamental absorption edge of b–Ga2O3 thin films, obtained by radio-frequency ion-plasmous sputtering, was investigated, using the method of optical spectroscopy. It was ascertained that the optical band gap Eg increases from 4.60 to 4.65 eV after the heat treatment films in argon atmosphere and to 5.20 eV after the reduction of annealed films in a hydrogen atmosphere. Consolidated effective mass of free charge carriers in b–Ga2O3 films after annealing and after reduction in hydrogen was estimated. It was found that the concentration of charge carriers after heat treatment in argon atmosphere is 7.30´1017 cm–3 and after reduction in hydrogen, is 2.62´1019 cm–3, which is typical for degenerated semiconductors. It was shown that the shift of fundamental absorption edge in thin films b–Ga2O3 after reduction in hydrogen is caused by Burstein-Moss effect.

The reaction of tin(II) fluoride with fluorosulfuric acid has been shown to produce tin(II) bisfluorosulfate. Crystals of Sn(OSO2F)2 are monoclinic, space group P21/c with a = 5.195(1), b = 9.709(1), c = 13.861(1) Å, β = 110.12(1)°, Z = 4, R = 0.029, and Rw = 0.030 for 1461 unique reflections. The structure consists of a three-dimensional framework of fluorosulfate groups linked by O—Sn—O bridges with the two crystallographically independent fluorosulfates acting as tridentate bridging ligands between tin atoms. There are four short bonds (Sn—O = 2.338(3), 2.350(3), 2.398(4), and 2.427(3) Å) and two longer ones (Sn—O = 2.695(3) and 2.702(3) Å) around tin. The four short bonds have a disphenoidal, AX4E, primary geometry with the lone pair occupying an equatorial site. If the two longer interactions are also considered the geometry is distorted octahedral, AX4Y2E. Two more non-bonding contacts of 3.420(4) and 3.319(4) Å complete an approximately dodecahedral arrangement of oxygen atoms around the tin. The fluorosulfate groups deviate significantly from C3υ symmetry and have the mean dimensions, S(1)—O = 1.419(4), S(1)—F = 1.541(4), S(2)—O = 1.435(3), S(2)—F = 1.539(3) Å, O—S(1)—O = 114.3(2)°, F—S(1)—O = 104.1(3)°, O—S(2)—O = 114.2(2)°, and F—S(2)—O = 104.1(2)°, respectively. The solid-state 119Sn nmr spectrum shows only one single resonance with a shift of −1534 ppm which is substantially different from that of a solution in HSO3F. The 119Sn Mössbauer spectrum shows an unresolved quadrupole splitting as a result of a distorted environment around tin, produced by the stereochemically active non-bonding electron pair. Key words: tin(II) bisfluorosulfate, 119Sn Mössbauer, solid-state nmr, X-ray crystallography

The present study was carried out in the period 2007- 2009 in the Experimental Farm in Bezek near the city of Chełm. The aim of the investigation was to compare weed infestation of the husked (Krezus) and naked (Cacko) oat cultivars cultivated under conditions of different weed control treatments. The following weed control levels were compared: A - control object; B - harrowing twice; C - application of the herbicide Mustang 306 SE; D - herbicide Mustang 306 SE + foliar fertilizer Insol 3; E - herbicide Mustang 306 SE + two foliar fertilizers: Insol 3 and FoliCare 18:18:18. The husked oat cv. Krezus was more competitive against weeds. Number of dicotyledonous weeds and of monocotyledonous weeds, total weeds, and air-dry weight of above-ground parts of weeds in the crop of this cultivar were all significantly lower compared to the naked cv. Cacko. From the group of dicotyledonous species, <i>Chenopodium album</i>, <i>Galinsoga parviflora</i>, <i>Matricaria maritima</i> ssp. <i>inodora</i>, <i>Polygonum aviculare</i>, and <i>Stellaria media</i> were the dominant species in the crops of the oat varieties under investigation, whereas among the monocotyledonous species <i>Echinochloa crus-galli</i>, <i>Setaria pumila</i>, <i>Apera spica-venti</i>, and <i>Elymus repens</i> occurred in greatest numbers. The application of the herbicide Mustang 306 SE decreased the number of dicotyledonous weeds and air-dry weight of weeds, compared to the treatment in which mechanical weed control had been used.

The study examined predictors of male nurse turnover intention in Korea using data collected from Korean hospitals. The results were obtained based on a secondary analysis of data previously collected from 306 male nurses in 16 regions of Korea from December 2014 to February 2015. Our findings suggest that male nurse turnover intention is predicted by (1) individual factors: single (B = 0.93, p = 0.008); (2) organizational factors: organizational commitment (B = −0.36, p < 0.001), job satisfaction (B = −0.27, p = 0.001), and job stress (B = 0.24, p < 0.001); and (3) social factors: hospital location in medium-categorized cities (B = 0.70, p = 0.012) and kinship responsibility (B = 0.13, p = 0.026). These factors accounted for 56.9% of the total variance. To lower the rate of turnover intention among male nurses, strategic interventions should be implemented based on the factors identified in this study.

306 Background: CALGB 80303 evaluated gemcitabine (G) in combination with either B or placebo (P) in 602 patients (pts) with APC. No significant difference in overall survival (OS) between treatment arms was observed. Previously, plasma EDTA samples from CALGB 80303 were analyzed and potential predictive biomarkers for B were identified. This follow-up analysis evaluated citrated plasma samples for circulating proteins related to matrix remodeling and coagulation. Methods: Multiplex ELISA analysis was employed to assess the following circulating factors: E-selectin, matrix metalloprotease (MMP) 2, MMP9, von Willebrand factor (vWF), D-dimer, thrombospondin 1, and tissue factor (TF). Thrombin-antithrombin complex (TAT) was analyzed using a standard enzyme immunoassay. Prognostic characteristics were determined by associating baseline values with OS using a Cox model. Predictive markers were identified with inclusion of a treatment by marker interaction term. Results: Baseline citrated plasma samples from 109 pts were included in this analysis (59 pts G+P; 50 pts G+B). Negative prognostic factors for OS were identified, including E-selectin (p <0.01) for patients receiving G-based therapy, and E-selectin, TF, and vWF (p <0.05) in pts receiving G+P. Potential biomarkers predicting for OS benefit from B were also identified. Pts with low (< median) levels of TAT (interaction p=0.034) exhibited a median OS of 6.0 months (mo) vs. 8.3 mo (HR=0.49; 95%CI 0.28-0.88) when receiving G+P vs. G+B, respectively. Furthermore, pts with elevated levels of TF (>Q3) were found to benefit from B (interaction p=0.021), with median OS of 4.3 mo vs 4.8 mo (HR=0.44;95%CI 0.18-1.08), respectively. Longitudinal analysis of samples collected at baseline and at cycle 3 indicated that only vWF changed in pts receiving G+P, with an increase in vWF (p<0.05). Conclusions: In patients with APC, baseline levels of TAT and TF were identified as potential predictors of benefit from B. These data emphasize the importance of coagulation factors in modulating tumor angiogenesis. The potential predictive value of these factors warrants further validation.

The vacuolar ATPase (V-ATPase) is a multifunctional enzyme that consists of several subunits. Subunit B is a part of the catalytic domain of the enzyme. The result of the RTPCR suggested that the V-ATPase B subunit is a ubiquitous gene. 24 h after the larvae were infected with the Bombyx mori nucleopolyhedrovirus (BmNPV), the expression level of the V-ATPase B subunit in the midgut of the resistant strain NB was about 3 times higher than in the susceptible strain 306, and then the expression level of the V-ATPase B subunit decreased rapidly to a very low level. This indicated that the virus may cause a lot of changes of physiological conditions in the midgut. Localization of the V-ATPase B subunit was attempted in midgut cells of Bombyx mori by immunohistochemistry. The immunohistochemical localization with the antibody against the B subunit revealed a positive staining in goblet cell apical membranes of Bombyx mori midgut cells as well as in the midgut of Manduca sexta. This sequence has been registered in GenBank under the accession number EU727173.

This study investigated the microstructure and mechanical properties of a wide-gap region brazed with various powders mixing ratios of additive powder to filler metal powder. The IN738 and BNi-3 alloy powders were used as additive and filler metal powder for the brazing process. The wide-gap brazing process was carried out in a vacuum of 2×10-5 torr. It was observed that the region brazed with only filler metal had a microstructure consisting of fully eutectic and dendrite structures. However, the region brazed with 60wt.% IN738 additive powder consisted of IN738 additive, Ni3B-Ni eutectic and (Cr, W)B. The fracture strength of the wide-gap region (70 wt.% additive and 30 wt.% filler metal powder) was as high as 687 MPa at room temperature. The Cracks in the wide-gap brazed region initiated at the Ni3B-Ni eutectic and (Cr, W)B, and then propagated through them. It was found that the Ni3B-Ni eutectic and (Cr, W)B in the brazed region are important microstructural factors affecting the mechanical properties of the wide-gap brazed region.

Methylation of cytosine base in the CpG dinucleotide sequence (DNA methylation) is a major epigenetic modification of the genome and plays an important role in gene expression. Recently, global DNA methylation in genome was studied by using a restriction landmark genomic scanning (RLGS) method and/or a representational difference analysis (RDA) method. However, these methods are complicated and need to use restriction enzymes. Therefore, the information derived from those methods is restricted to the region of the DNA sequence which is able to be cleaved by restriction enzymes. In this study, to establish a simple method to estimate global DNA methylation level in bovine spermatozoa, we tried to develop the DNA methylation analyzing method by using immunostaining of 5-methylcytosine. The immunostaining method for 5-methylcytosine in this study was based on the method developed by Benchaib et al. (2003 Fertil. Steril. 80, 947-952) for human spermatozoa. Because of the species difference, we modified some treatments to apply to bovine spermatozoa. Frozen-thawed bovine spermatozoa were washed by using 30 and 45% Percoll gradient solutions. After washing, spermatozoa were treated with 0.25 dithicthreitol M (DTT) and 1% sodium dodecyl sulfate (SDS) at room temperature (RT). Then, treated spermatozoa were spread on a slide glass with Cytospin4 (30g, 5 104 cells/mL) and air-dried at RT. Air-dried bovine spermatozoa specimens were fixed in methanol: glacial acetic acid (3:1) solution at RT and treated with 1% Triton X and 1% SDS at RT; DNA was denatured with 6 N HCl at RT. After the denaturation, 5-methylcytosine in sperm DNA was analyzed by immunofluorescence technique with mouse anti 5-methylcytosine antibody and fluorescein isothiocyanate (FITC)-conjugated goat anti mouse IgG antibody. The total sperm DNA was counterstained with propidium iodide (PI). Stained samples were observed with a confocal laser scanning microscope and obtained images were analyzed with fluorescence image analysis software. The area that was clearly stained with PI in each sperm head was designated and measured as the area of total sperm DNA, and the number of the dots that showed FITC fluorescence within the total sperm DNA area was designated and measured as the area of 5-methylcytosine in total sperm DNA. The area measurement was performed with fixed light strength. Three bovine spermatozoa samples derived from different bulls, used daily for calf production by AI, were examined. The ratio of the mean total area of the 5-methylcytosine in sperm DNA to the mean total area of the sperm DNA was 34.1% in bull A (9.13 5.66 m2, 26.75 5.29 m2, n = 57), 45.2% in bull B (16.60 3.79 m2, 36.74 5.95 m2, n = 41) and 43.9% in bull C (14.66 4.27 m2, 33.45 7.13 m2, n = 22). There was significant difference in the ratio between bull A and bulls B and C (P < 0.01). More research is required to evaluate the meaning of this individual difference of DNA methylation between bulls. 306 Background: Combined therapy of an immune checkpoint inhibitor with a targeted antiangiogenic agent had been proved to be effective for the treatment of uHCC. Penpulimab was engineered to eliminate FcγR binding and ADCC/ADCP completely, where ADCC/ADCP effects could induce T-cell apoptosis and clearance and then compromise anti-tumor activity. Penpulimab demonstrated a slower PD-1 antigen binding off-rate, which resulted in better cellular activity and higher receptor occupancy. Penpulimab also showed numerous contacts with N58 glycosylation on the BC loop of PD-1 which could be an advantage to facilitate interaction of PD-1 antibody and might contribute to slower binding off-rate. These structural differentiations offer more robust biological effect and enhance anti-tumor activity of penpulimab. Anlotinib is a multi-targeted tyrosine kinase inhibitor selective for VEGF receptors 1/2/3, FGF receptors 1-4, PDGF receptors α and β, and c-kit. Methods: In this open-label, multicenter phase Ib/II study, patients (pts) without prior systemic treatment, and classified as BCLC stage B (not amenable for locoregional therapy) or C, Child-Pugh ≤7, and ECOG PS ≤ 1 received Penpulimab (200mg IV Q3W) and Anotinib (8 mg PO 2weeks on/1 week off). Primary endpoint was ORR (RECIST v1.1); secondary endpoints were safety, DCR, DoR, TTP, PFS and OS. Results: 31 pts (median age 56 years [23–74], ECOG 0/1 [64%/36%], BCLC B/C [23%/77%], HBV/HCV [61%/7%]) received combined therapy. As of August 31, 2020, median follow-up time was 11.9 mons (range 3.7-17.7). Median PFS was 7.6 mons with 6-mons PFS rate was 57.6% while median TTP was 8.5 mons with 6-mons TTP rate was 62.7%. Median overall survival had not been met and 6-mons OS rate was 93.2%. The ORR was 31.0% (9/29) and DCR was 82.8% (24/29). At data cutoff, 77.8% of responders remained ongoing and still on treatment. Treatment-related adverse events (TRAEs) occurred in 90.3% of pts (≥G3 in 16.1% [5/31], no G5, treatment discontinuation in 9.7% [3/31]). Most common TRAEs (≥15%) were increased AST (38.7%) and ALT (35.5%), increased blood bilirubin (22.6%),asthenia (22.6%),decreased platelet count (19.4%) and rash (16.1%). Conclusions: Penpulimab plus Anlotinib showed favorable antitumor efficacy and an acceptable safety profile in pts with uHCC. The further randomized, phase 3 study of Penpulimab in combination with Anlotinib at a higher dose (10 mg PO 2 weeks on/1 week off) in this setting is ongoing (NCT04344158).

Influence of a modified media on Hexaene H-85 and Azalomycine B production by Streptomyces hygroscopicus CH-7 was investigated. The amino acid L-tryptophan, as a nitrogen source, was replaced with isatin-3-thiosemicarbazone and its complexes with some divalent metal ions. Isatin-3-thiosemicarbazone was synthesized in crude glycerol obtained as a byproduct in biodiesel production from sunflower oil. The complexes were charac-terized by elemental microanalysis and magnetic susceptibility, as well as, by Atomic absorption(AA), Fourier-transform infrared (FTIR) and Ultraviolet?visible (UV/VIS) spectro-scopic methods. The spectral studies indicated an octahedral geometry for the Mn(II), Fe(II) and Ni(II) complexes and a tetrahedral one for the Zn(II) complex. Comparing to the basal medium, isatin-3-thiosemicarbazone (ITC) and its metal complexes in the concentration of 0.5 g dm-3 showed better results in the antibiotics production. Use of medium supple-mented with the Fe(II) complex resulted in the maximum Hexaene H-85 and Azalomycine B concentrations of 306 ?g cm-3 and 127 ?g cm-3, respectively. Addition of ITC and its complexes changed the morphology of S. hygroscopicus CH-7 from filaments to pellets as a dominant shape in media resulting in higher antibiotic production.

Iron(II) and nickel(II) bis(ligand) complexes of 6-(5-methyl-1,2,4-oxadiazol-3-yl)-2,2′-bipyridine (L) are described. The ligand field in the iron complex is close to that at the singlet ( 1 A1) ? quintet ( 5 T2) crossover and magnetic and Mössbauer spectral evidence indicates that a spin transition occurs in salts of the iron complex but is centred above room temperature. The structures of [FeL2] [CF3SO3]2.CH3CN and [NiL2] [BF4]2.CH3CN were determined and both are very similar to the structures of the corresponding terpyridine complexes. Spectroscopic data indicate that for the iron complex π-interaction between the metal and the ligand is less than that in the terpyridine system. [FeL2] [CF3SO3]2.CH3CN is monoclinic, space group P 21/c; a 8 . 232(5), b 25 . 273(10), c 17 . 306(10) Å, β 92 . 37(3)°, Z 4; [NiL2] [BF4]2.CH3CN is monoclinic, space group P 21/c; a 8 . 136(2), b 17 . 558(2), c 23 . 783(7) Å, β 109 . 32(1)°, Z 4.

The purpose of this study was to examine the validity of counting steps and computing indices of moderate-to-vigorous physical activity (MVPA) using miniature activity monitors with 3-D technology worn at various locations under controlled (CON) and free-living conditions (FL). Kenz e-style2, Tanita Calorism Smart, and Omron Calori Scan HJA-306 activity monitors were assessed. Nine and 31 young adult women were assigned to the CON and FL studies, respectively. While walking or jogging on a treadmill at 5 different speeds, the subjects simultaneously carried the 3 different monitors in a pants pocket (PP), a chest shirt pocket, and a shoulder bag (B). Under the FL condition, the 3 monitors were placed only at the PP and B locations for practical reasons. Significant effects of monitor location and walking/jogging speed on the step count measured by the 3 monitors were evaluated under the CON condition. Monitors placed at both PP and B tended to underestimate the number of steps; however, there were no significant differences between the values obtained with the Kenz monitor and those obtained with a criterion accelerometer under the FL condition. Moreover, strong correlations were observed between steps measured by monitors placed at PP and steps measured by the criterion accelerometer. The amount of MVPA for the PP location and the non-carrying duration of the bag for the B location were considered to be important determinants of the accuracy of step counting under the FL condition. In conclusion, monitors placed at the PP location, especially the Kenz monitor, showed acceptable accuracy for young adult women in real-life settings. In contrast, MVPA indices assessed using these monitors showed limited validity.

The SARS COVID-19 pandemic took the whole world by storm due to its high and rapid infectivity. As it spread, its patterns became clear, and it became evident that a few individuals were more predisposed to it than others. Showing a massive inflammatory response by the body, and a negative coagulative affect, doctors and scientists began to perceive the role of blood cells, and hence blood types in the disease process. This lead to the main objective of this research: to assess the link between COVID19 infection and blood group types, and follow disease outcome according to it. The study comprises data collected from patients within 3 hospital setting of Northern Pakistan, viz Military Hospital Rawalpindi, Benazir Bhutto Hospital Rawalpindi, and Pakistan institute of medical sciences Islamabad. The study was conducted during a period of 1 month i-e from 15th June 2020 to 15th July 2020 after approval by the worthy Ethical review committee of PEMH. The study design was Analytical- Case series. Purposive Sampling technique was used. The sample size was 306 based on WHO sample calculator. Patients who were PCR positive and symptomatic for COVID-19 were included whereas PCR negative and asymptomatic patients were excluded. SPSS version 25 was used to process the data and Graphs and Charts were used to explain it. Among 306 patients of COVID 19 the most commonly affected blood group was A+ve followed by B+ve. The least affected blood group was O of which both positive and negative contributed to 17.71%. A blood group contributed 36%, while B 35% and AB 10.8%. The study substantiates that blood group A Rh positive has more susceptibility for SARS COVID-19 infection than other blood group types. Blood group O might have a protective effect against this infection.

Lettuce (Lactuca sativa L.) belongs to the family Asteraceae and is known as a leafy salad vegetable. It is rich in vitamins like Vitamin A and C, and also a good source of minerals such as iron, sodium, calcium etc. It is more popular in temperate regions and also performing well under subtropical climatic conditions during the winter season. Therefore, the present investigation was carried out to identify the suitable variety of lettuce for higher yield and quality under subtropical climate. The experiment was conducted with five treatments replicated thrice. Seeds of these five lettuce varieties viz., Tango, Bingo, Summer Star, Grand Rapid and Black Rose were sown in portrays prefilled with a combination of media made of coco peat, vermiculite and perlite in the ratio 3:1:1. After thirty days plants were plugged out and transplanted under field condition to study the performance of lettuce varieties. Among the studied varieties, the maximum yield (191.44 q/ha) was recorded with the variety Summer Star which was at par (190.32 q/ha) with the Bingo variety, followed by (129.50 q/ha) in Tango and the least (85.51 q/ha) by Black Rose. However, vitamin C content and total carotenoid content were significantly found highest (15.98 mg/ 100 g) and 8.06 mg/ 100 g in the Black Rose variety, respectively. Tango variety observed maximum chlorophyll ‘a’ (5.85 mg/ g FW), ‘b’ (2.01 mg/ g FW) and total chlorophyll (7.85 mg/ g FW) content among studied lettuce varieties.

Background: Hepatitis B is one of the most important blood borne infection, the health care workers may acquire while providing care to the patients. The overall aim of this study was to estimate the Hepatitis B vaccine coverage and its determinants among health care workers. Methods: This cross-sectional study was done in a rural tertiary care hospital in Palakkad district, of Kerala. We particularly looked for self-reported hepatitis B vaccination coverage and its determinants, using self-administered pretested questionnaire in 410 health workers. We calculated the proportions and looked for association between the various independent variables and dependent variables for vaccination coverage. Results: Out of 410 study participants, 119 (29%) of them were males and 291 (71%) of them were females. The mean age of the participant was 31.3 years and the range was between 19 to 70 years. 365 (89%) were vaccinated with atleast one dose of Hepatitis B Vaccine. Of these 306 (74.6%) had received all the 3 doses of vaccine, as per schedule. Gender and years of experience was not associated with the Hepatitis B vaccination status. However, variables like educational status, professional background was associated with hepatitis B vaccine coverage. Conclusions: The coverage of hepatitis B vaccine was high. Among those with incomplete vaccination few of them were waiting for completing the vaccination schedule in near future. Good hospital policies like vaccinating the new medical students and new employees were the main factors responsible for high vaccination coverage among the health care workers in our study.

4032 Background: RATIONALE-306 (NCT03783442) is the first global study to investigate anti-programmed cell death protein 1 therapy in combination with different chemotherapy (chemo) options in the first-line (1L) treatment of advanced/metastatic esophageal squamous cell carcinoma (ESCC). At interim analysis (IA), tislelizumab (TIS) plus chemo demonstrated a statistically significant, clinically meaningful improvement in overall survival (OS) versus placebo (PBO) plus chemo, with a manageable safety profile. Here, we report updated efficacy and safety data with minimum 3 years’ follow-up after study unblinding at IA. Methods: Adults with unresectable locally advanced recurrent/metastatic ESCC and no prior systemic treatment for advanced disease were enrolled and randomized (1:1; stratified by region, prior definitive therapy, and investigator [INV]-chosen chemo) to receive TIS 200 mg (Arm A) or PBO (Arm B) IV every 3 weeks plus chemo (platinum plus fluoropyrimidine or platinum plus paclitaxel), until disease progression or intolerable toxicity. The primary endpoint was OS in the intent-to-treat population. Secondary endpoints included progression-free survival (PFS), objective response rate (ORR), and duration of response (DoR), all per INV, and safety. Results: In total, 649 patients (pts) were randomized (Arm A, n=326; Arm B, n=323). At a minimum study follow-up of 36.0 months, improvements in OS, PFS, and DoR in Arm A versus B (Table) were maintained relative to the IA. The hazard ratio (HR) for OS with TIS plus chemo versus PBO plus chemo was 0.70 (95% confidence interval: 0.59, 0.83). Similar to the IA, incidences of any-grade (96.6% vs 96.3%) or grade ≥3 (67.0% vs 64.5%) treatment-related adverse events (TRAEs) were comparable between Arms A and B, respectively; treatment-emergent adverse events leading to treatment discontinuation were higher in Arm A (32.1%) versus B (22.1%). In Arm A versus B, respectively, serious TRAEs occurred in 29.9% vs 19.6% of pts; TRAEs leading to death occurred in 1.9% and 1.2%. Conclusions: After minimum 3 years’ follow-up, 1L TIS plus chemo continued to demonstrate clinically meaningful improvements in OS and PFS and durable antitumor response benefit versus PBO plus chemo in pts with advanced/metastatic ESCC, with no new safety signals. Drought is a major abiotic factor restricting rice yield; therefore, to cope with this stress, 2030 japonica rice accessions from China and other countries were evaluated in Beijing in 2017 and 2018. This was the first time six agronomic traits in the large-scale germplasm of rice under lowland and upland conditions with an augmented randomized complete block design (ARCBD) were analysed. The genotypes revealing drought resistant grade (DRG) scores of 1, 1–3, 3 and 3–5 were considered drought-tolerant and comprised 10% of the assessed germplasm. These findings were consistent with the agglomerative hierarchical cluster (AHC) analysis that classified germplasm in nine clusters. The generated clusters were further grouped in A, B, C and D classes based on the stress response. Approximately half of the genotypes with an upland ecotype were distributed in drought-resistant class A (cluster VII and VI) and moderately resistant class B (VIII and IX). The majority of the genotypes from China, Korea and Japan fall in drought-susceptible classes C and D. Genotypes of DRG 1, 1–3 and 3 belonged to the clusters VII and VI. Finally, we screened out 42 elite genotypes including seven improved upland rice lines (D78, LB37-13, NSU77, Handao 385, Handao 306, SF83 and HF6-65-119), three upland released varieties (Liaogeng 27, Hanfeng 8 and IRAT109) and three traditional lowland cultivars (Hongmaodao, Weiguo 7 and Xiaohongbandao). These genotypes might be used as priority parents in drought-tolerant rice breeding programmes and some of them could be recommended directly to farmers in water-deficient rice areas of China.

Understanding longitudinal associations between problematic peer relations and psychopathology is needed to inform public health. Three models have been proposed: Poor peer relations (a) lead or are a risk factor for psychopathology, (b) lag or are a consequence of psychopathology, or (c) both lead and lag psychopathology. Another model is that poor peer relations lead or lag psychopathology depending on the developmental period. To test these models, youths’ peer relations and clinical symptoms were assessed up to six times between ages 3 and 11 in 306 children. Bivariate latent-change-score models tested leading and lagging longitudinal relationships between children’s peer relations (peer victimization/rejection, peer-directed aggression, social withdrawal, prosocial behavior) and psychopathology (depression, anxiety, and externalizing symptoms). Peer victimization/rejection was a leading indicator of depression from early childhood into preadolescence. Peer-directed aggression was a leading indicator of externalizing symptoms (in late childhood).

The murine alpha B-crystallin gene (a member of the small heat shock protein family) is expressed constitutively at high levels in the lens and at lower levels in many other tissues, including skeletal muscle. We have previously used the herpes simplex virus thymidine kinase promoter fused to the human growth hormone gene to identify an alpha B-crystallin enhancer at positions -427 to -259 that has high activity in muscle and low activity in lens cell lines. In the study reported here, we performed DNase I footprinting, transfection, mutagenesis, and electrophoretic mobility shift experiments using the murine C2C12 muscle and alpha TN4-1 lens cell lines and the rabbit N/N1003A lens cell line to identify sequences responsible for activity of this enhancer. Enhancer activity in both the muscle and lens cells was dependent on novel elements called alpha BE-1 (-407 to -397), alpha BE-2 (-360 to -327), and alpha BE-3 (-317 to -306). These elements were also weakly occupied by nuclear proteins in L929 cells, which appear to express the alpha B-crystallin gene at a very low level (detectable only by the polymerase chain reaction). A fourth element containing a consensus muscle regulatory factor-binding site called MRF (-300 to -288) was occupied and used only by the C2C12 muscle cells. Cotransfection in NIH 3T3 cells and antibody-gel shift experiments using C2C12 nuclear extracts indicated that MyoD, myogen, or a similar member of this family can activate the alpha B-crystallin enhancer by interaction with the MRF site. Taken together, we conclude that the alpha BE-1, alpha BE-2, and alpha BE-3 elements are shared by both lens and muscle cells, but the MRF element is used only in muscle cells, providing the first example of a muscle-specific control element in a crystallin gene.

This study describes the length-weight relation (LWR) of small indigenous fish Pethia ticto from Gomti River, Aamghat, Haliyapur, Sultanpur district 260 29N and 810 44E, 751m (msl) Uttar Pradesh, India. A total of 306 specimens were caught by random sampling method using traditional fishing gear as gillnet, cast net and dragnet of various mesh sizes from June 2016 to May 2017, once in a month. Total Length (mm) was measured from anteriormost part of the snout to the posterior-most part of the caudal fin and body weight (g) were taken by digital electronic balance. The analysis of data shows that the allometric coefficient is close to isometric value (3.0). Allometric growth indicates a more rotund population of fish when the values of growth coefficient are higher than 3.0 (b>3). If the value of growth coefficient is less than 3.0, the population of fish is known to be less rotund. Isometric growth indicates that the shape of fish does not change as it grows (b=3). The coefficient of determination (R2 ) was also found significant (=0.97). Fulton’s condition factor further supports the results obtained ranged from 1.041 to 1.660 shows the degree of well being of a fish. This study is helpful in providing relevant information in the assessment of stock and estimation of the fish condition about its environment

306 Background: The combination of paclitaxel (PTX) plus ramucirumab (RAM) has been the standard regimen for advanced gastric cancer (GC) or gastro-esophageal junction cancer (GJC) patients (pts) who were treated with fluoropyrimidine-containing regimen. In a phase 2 trial, it was showen that nab-PTX plus RAM has promising activity (objective response rate [ORR], 55 %; median progression free survival [PFS], 7.6 months) [Bando H, et al. Eur J Cancer 2018] and is, therefore, a treatment option. However, high incidence of myelosuppression led to discontinuation of the nab-PTX. Therefore, we conducted this study to investigate whether schedule modification to biweekly nab-PTX plus RAM therapy is safe and effective. Methods: Pts with GC or GJC treated with fluoropyrimidine-containing regimen were enrolled and received nab-PTX (100 mg/m2) on days 1, 8, and 15 and RAM (8 mg/kg) on days 1 and 15 of a 28-day cycle. The treatment schedule of pts with severe myelosuppression during the first cycle was modified to biweekly therapy from the second cycle. The primary endpoint was PFS in modified pts. Secondary endpoints included ORR, disease control rate (DCR), overall survival (OS), and safety. The exploratory analysis evaluated PFS and OS in pts who continued standard-schedule therapy. Based on the hypothesis that biweekly nab-PTX plus RAM therapy would improve PFS from 4.5 months to 7.0 months, 40 pts were required for a power of 0.8 with a one-sided α of 0.05. Results: Of the 81 pts who were enrolled, 76 pts received standard nab-PTX plus RAM in the first cycle and 47 pts (58%) were modified to the bi-weekly therapy. Pts characteristics in the modified pts were as follows: median age, 70 years; male, 72%; ECOG PS 0, 81%; and poorly differentiated adenocarcinoma or signet ring cell carcinoma, 45%. The median relative dose intensities of the entire period and after modification were 66% and 60%, respectively. In the modified pts, median PFS was 4.7 months (95% Confidence Interval [CI]: 3.7–5.6), median OS was 13.9 months (95% CI: 9.1–16.9), ORR was 25% (95% CI: 11–39), and DCR was 75% (95% CI: 61–89). Tumor shrinkage was observed in 61% of the pts (n = 36) with measurable lesion. In pts who could continue standard-schedule therapy, median PFS was 2.7 months (95% CI: 1.8–4.0) and median OS was 8.0 months (95% CI: 5.6–14.7). Severe (Grade >3) adverse events after modification were neutropenia (55%), anemia (13%), hypertension (17%), and peripheral sensory neuropathy (13%). Conclusions: Our study could not meet the primary endpoint for PFS. However, PFS in pts with standard-schedule therapy was numerically lower than that in the modified pts. The modification to biweekly schedule might be considered to be a treatment option for GC/GJC pts with severe myelosuppression in second-line nab-PTX plus RAM therapy. (1) Background: It is not known whether different daily dosing schemes have different effects on colistin nephrotoxicity. We examined the effect of once- versus twice- or thrice-daily doses of colistin on renal function. (2) Methods: We performed a multicenter retrospective cohort study of hospitalized patients with a baseline glomerular filtration rate ≥ 50 mL/min who received intravenously the same colistin dose once (regimen A), twice (regimen B) or thrice daily (regimen C). The primary endpoint was acute kidney injury (AKI), defined as fulfilment of any of the RIFLE (Risk-Injury-Failure-Loss-End stage renal disease) criteria. (3) Results: We included 306 patients; 132 (43.1%) received regimen A, 151 (49.3%) regimen B, and 23 (7.5%) regimen C. Ninety-nine (32.4%) patients developed AKI; there was no difference between regimen A vs. B and C [45 (34.1%) vs. 54 (31.0%), p = 0.57]. In a propensity score–matched cohort, AKI was similar in patients receiving Regimen A, Regimen B, and Regimen C (31.6% vs. 33.3%, p = 0.78). On logistic regression analysis, diabetes was an independent predictor of AKI (OR = 4.59, 95% CI 2.03–10.39, p = 0.001) while eGFR > 80 mL/min (OR = 0.50, 95% CI 0.25–0.99, p = 0.048) was inversely associated with AKI. (4) Conclusions: Colistin once daily is not more nephrotoxic than the standard colistin regimens. The only independent predictor of nephrotoxicity was diabetes mellitus, while eGFR > 80 mL/min had a protective effect.

AbstractBackground and ObjectivesHealth Care Workers (HCWs) are at a high risk of needle stick injuries and HBV infection in Egypt; this problem is further aggravated by low Hepatitis B (HB) vaccination coverage. Limited data are available on the prevalence of HBV infection in Egyptian HCWs. In this study, we aimed to assess the HBV infection rate and genotypes among Egyptian HCWs.MethodsFive hundred and sixty-four (564) HCWs were included. Of them, 258 (45.74%) were health care providers and 306 (54.25%) were non-health care providers. All HCWs completed both the study questionnaires and provided a blood sample for HBV testing. Indeed, all HCWs were tested for Hepatitis B surface antigen (HBsAg) and antibody to Hepatitis B core antigen (anti-HBc), by enzyme-linked immunosorbent assay. HBVDNA was checked for HCWs who tested positive for HBsAg and/or anti-HBc, by nested Polymerase Chain Reaction (PCR). HBVDNA positive HCWs were further subjected to HBV genotyping.ResultsThe mean age of included HCWs was 33.0 ± 9.8 years, of whom 319 (56.56%) were males. The mean duration of health care work was 9.3 ± 6.7 years. The frequency of HBsAg and anti-HBc were 1.4%, and 24.5%, respectively. Old age and prolonged duration of health care work were significantly associated with anti-HBc seropositivity. Among 140 HCWs positive for HBsAg and/or anti-HBc, 14 (10 %) had positive HBVDNA by PCR. HBV/E (n= 7), HBV/D (n= 3) and co-infection with E and D (n= 4) genotypes were detected.ConclusionEgyptian HCWs have a significantly high rate of HBV exposure. The detection of HBV/E genotype among Egyptian HCWs suggests prevalent transmission of HBV/E among Egyptian populations.

The myc family of genes contains five functional members. We describe the cloning of a new member of the myc family from rat genomic and cDNA libraries, designated B-myc. A fragment of cloned B-myc was used to map the corresponding rat locus by Southern blotting of DNA prepared from rat X mouse somatic cell hybrids. B-myc mapped to rat chromosome 3. We have previously mapped the c-myc to rat chromosome 7 (J. Sümegi, J. Spira, H. Bazin, J. Szpirer, G. Levan, and G. Klein, Nature [London] 306:497-498, 1983) and N-myc and L-myc to rat chromosomes 6 and 5, respectively (S. Ingvarsson, C. Asker, Z. Wirschubsky, J. Szpirer, G. Levan, G. Klein, and J. Sümegi, Somat. Cell Mol. Genet. 13:335-339, 1987). A partial sequence of B-myc had extensive sequence homology to the c-myc protein-coding region, and the detection of intron homology further indicated that these two genes are closely related. The DNA regions conserved among the myc family members, designated myc boxes, were highly conserved between c-myc and B-myc. A lower degree of homology was detected in other parts of the coding region in c-myc and B-myc not present in N-myc and L-myc. A 1.3-kilobase B-myc-specific mRNA was detected in most rat tissues, with the highest expression in the brain. This resembled the expression pattern of c-myc, although at different relative levels, and was in contrast to the more tissue-specific expression of N-myc and L-myc. B-myc was expressed at uniformly high levels in all fetal tissues and during subsequent postnatal development, in contrast to the stage-specific expression of c-myc.

PURPOSE Total neoadjuvant therapy is a new paradigm for rectal cancer treatment. Optimal scheduling of preoperative chemoradiotherapy (CRT) and chemotherapy remains to be established. PATIENTS AND METHODS We conducted a multicenter, randomized, phase II trial using a pick-the-winner design on the basis of the hypothesis of an increased pathologic complete response (pCR) of 25% after total neoadjuvant therapy compared with standard 15% after preoperative CRT. Patients with stage II or III rectal cancer were assigned to group A for induction chemotherapy using three cycles of fluorouracil, leucovorin, and oxaliplatin before fluorouracil/oxaliplatin CRT (50.4 Gy) or to group B for consolidation chemotherapy after CRT. Secondary end points included toxicity, compliance, and surgical morbidity. RESULTS Of the 311 patients enrolled, 306 patients were evaluable (156 in group A and 150 in group B). CRT-related grade 3 or 4 toxicity was lower (37% v 27%) and compliance with CRT higher in group B (91%, 78%, and 76% v 97%, 87%, and 93% received full-dose radiotherapy, concomitant fluorouracil, and concomitant oxaliplatin in groups A and B, respectively); 92% versus 85% completed all induction/consolidation chemotherapy cycles, respectively. The longer interval between completion of CRT and surgery in group B (median 90 v 45 days in group A) did not increase surgical morbidity. A pCR in the intention-to-treat population was achieved in 17% in group A and in 25% in group B. Thus, only group B ( P < .001), but not group A ( P = .210), fulfilled the predefined statistical hypothesis. CONCLUSION Up-front CRT followed by chemotherapy resulted in better compliance with CRT but worse compliance with chemotherapy compared with group A. Long-term follow-up will assess whether improved pCR in group B translates to better oncologic outcome.

340 Background: TIS, an anti-programmed cell death protein 1 (PD-1) antibody, + chemo as 1L therapy demonstrated statistically significant and clinically meaningful improvement in overall survival (OS) vs PBO + chemo in patients with advanced or metastatic ESCC (hazard ratio [HR] 0.66 [95% confidence interval (CI) 0.54, 0.80]; P< 0.0001), with a manageable safety profile, at interim analysis of the phase 3, double-blind RATIONALE-306 study (NCT03783442). Here, we report data from the non-Asia subgroup (Europe, Northern America, and Oceania). Methods: Adults with advanced or metastatic ESCC, with no prior systemic treatment for advanced disease were randomized 1:1, (stratified by region, prior definitive therapy, and investigator [INV]-chosen chemo) to receive TIS 200 mg intravenously (IV) once every 3 weeks (Q3W) (Arm A) or PBO IV Q3W (Arm B), with platinum + fluoropyrimidine, or platinum + paclitaxel until disease progression by INV per RECIST v1.1, intolerable toxicity, or withdrawal. The primary endpoint was OS in the intent-to-treat population. Secondary endpoints included: progression-free survival (PFS), objective response rate (ORR), and duration of response (DoR) by INV per RECIST v1.1; OS in the programmed death-ligand 1 score ≥10%; and safety. Results: Of 649 randomized patients, 163 (25.1%) were from the non-Asia subgroup (Arm A, n = 83; Arm B, n = 80). At data cutoff (Feb 28, 2022), the median study follow-up time in the non-Asia subgroup was 16.0 months (mo) in Arm A vs 8.4 mo in Arm B. OS (median 16.3 vs 9.0 mo; unstratified HR 0.66 [95% CI 0.45, 0.96]) and PFS (median 7.7 vs 5.5 mo; unstratified HR 0.59 [95% CI 0.41, 0.83]) were improved in Arm A vs Arm B, respectively. Arm A had higher ORR (61.4% vs 41.3%, odds ratio 2.27 [95% CI 1.21, 4.25]) and longer median DoR (7.1 mo [95% CI 5.6, 9.6] vs 5.7 mo [95% CI 3.8, 8.3]) than Arm B. More patients in Arm A vs Arm B experienced ≥1 treatment-related adverse event (TRAE; 94.0% vs 88.5%), serious TRAEs (25.3% vs 17.9%), and discontinuation due to treatment-emergent AEs (42.2% vs 35.9%, respectively). Similar proportions of patients in Arm A vs Arm B had ≥grade 3 TRAEs (56.6% vs 52.6%), and TRAEs leading to death (1.2% vs 1.3%), respectively. Conclusions: In the non-Asia subgroup, 1L TIS + chemo showed a clinically meaningful improvement in OS vs PBO + chemo in patients with advanced or metastatic ESCC, with a manageable safety profile, consistent with published results in the overall population. Abstract Colorectal cancer is a leading cause of cancer mortality and arises from genetic and epigenetic changes in colon epithelial cells. One of the mechanisms driving colon epithelial cell transformation is the bystander effect (BSE). BSE primarily arises from polarized macrophages (MØs) in stromal tissue. Herein, we report that alternatively activated (M2) MØs, as generated by interleukin (IL)-4, induce double-strand DNA breaks in target cells through BSE. Unlike M1 MØs that are activated by intestinal microbiota, BSE and DNA damage due to M2-like MØs occur through a distinct mechanism that differs from M1 MØs and does not involve pro-inflammatory mediators (e.g., cyclooxygenase-2 or tumor necrosis factor-α). Murine MØs (RAW264.7 cells) treated with IL-4 were polarized to an M2-like phenotype and found to strongly express arginase 1 (ARG1), a prototypical enzyme that converts L-arginine to L-ornithine and urea. L-Ornithine is a precursor in polyamine synthesis with H2O2 being a key oxidizing byproduct of polyamine catabolism. In a dual-chamber system using Young Adult Mouse Colon (YAMC) cells as targets for BSE generated by M2-like MØs, inhibition of ARG1 by N-hydroxy-nor-L-arginine abolished DNA damage (as measured by γH2AX). Direct treatment of YAMC cells with L-ornithine resulted in similar levels of DNA damage as M2-like MØs. YAMC cells exposed to M2-like MØs had increased intracellular H2O2. MDL-72527, an inhibitor polyamine oxidase and polyamine catabolism, reduced intracellular H2O2 to background levels and decreased DNA damage. YAMC cells sustaining DNA damage due to BSE from M2-like MØs showed specific phosphorylation of p53(ser15) and NF-κB(ser536) with no evidence for active caspase-3. These data along with an increased expression of cyclinD1 in YAMC cells indicated that DNA damage caused by M2-like MØs occurred via polyamine catabolism and was not due to cellular toxicity or apoptosis. Pathway analysis of RNAseq data for YAMC cells exposed to M2-like MØs identified multiple activated pathways including DNA repair, ubiquitination, Nrf2-mediated stress responses, endoplasmic reticulum overload responses, and lipid biosynthesis. These results provide additional evidence for global cellular responses to BSE generated by M2-like MØs. In conclusion, our findings show that M2-like MØs can generate BSE via a novel mechanism involving ARG1. Induction of BSE by M2-like MØs results in acute DNA damage in colon epithelial cells and activates multiple signaling pathways in target epithelial cells. These data imply a role for alternatively activated M2-like MØs in the malignant transformation of epithelial cells. 500-700 g betragen soll, existiert. Tragende und nicht tragende AS (B-A) bzw. JS und AS (B-B) wiesen jeweils ein ähnliches mittleres UG und zu ähnlichen Anteilen UG von < 400 / > 600 g (JS) oder < 500 / > 700 g (AS) auf. Sauen mit ET-Grad 4 wurden seltener (p ≤ 0,05) bzw. tendenziell seltener (p ≤ 0,1) tragend als Tiere mit den ET-Graden 1 und 2 (57,1% vs. 87,0% bzw. 82,2%).
