Acute kidney injury induced by renal ischemia-reperfusion (I/R) compromises microvascular density and predisposes to chronic kidney disease (CKD) and sodium-dependent hypertension. VEGF-121 was administered to rats fed a standard (0.4%) sodium diet at various times following recovery from I/R injury for up to 35 days. VEGF-121 had no effect on the initial loss of renal function, as indicated by serum creatinine levels measured 24 h after injury. Serum creatinine levels declined thereafter, indicative of renal repair. Rats were then switched to an elevated (4.0%) sodium diet for an additional 28 days to induce CKD. The 4.0% sodium diet enhanced renal hypertrophy, interstitial volume, albuminuria, and cardiac hypertrophy relative to postischemic animals maintained on the 0.4% sodium diet. Administration of VEGF-121 from day 0 to 14, day 0 to 35, or day 3 to 35 after I/R suppressed the effects of sodium diet on CKD development, while delayed administration of VEGF-121 from day 21 to 35 had no effect. Endothelial nitric oxide synthase protein levels were upregulated in postischemic animals, and this effect was significantly increased by the 4.0% sodium diet but was not influenced by prior treatment with VEGF. Conversely, microvascular density was preserved in postischemic animals treated with VEGF-121 relative to vehicle-treated postischemic animals. These data suggest that early, but not delayed, treatment with VEGF-121 can preserve vascular structure after ischemia and influence chronic renal function in response to elevated sodium intake.

The research focused to detect and identify genetic polymorphisms in exon 7 of the β-casein CSN2 gene in blood samples from Greek Holstein cows. For this purpose, DNA was extracted from 120 blood samples of cows. The desired region of exon 7 was amplified by PCR, resulting in a 121 bp product. The PCR product was digested by restriction fragment length polymorphism (RFLP) method. The results suggest that the A1A2 genotype prevails over the others. Specifically, of the 120 cattle, 72 showed triple bands of 121 bp, 86 bp, and 35 bp indicating the A1A2 genotype. The 42 cattle showed a single band at 121 bp, indicating that they carried the A1A1 genotype. The remaining 6 showed only two bands of 86 and 35 bps, indicating that they carried the A2A2 genotype. In the total population of heterozygotes A1A2-0.60 were the most frequent, while homozygotes A2A2-0.06 were the least frequent ones. This suggests a slight superiority of allele A-0.65.

ABSTRACT Lactobacillus reuteri LB 121 cells growing on sucrose synthesize large amounts of a glucan (d-glucose) and a fructan (d-fructose) with molecular masses of 3,500 and 150 kDa, respectively. Methylation studies and 13C or1H nuclear magnetic resonance analysis showed that the glucan has a unique structure consisting of terminal, 4-substituted, 6-substituted, and 4,6-disubstituted α-glucose in a molar ratio of 1.1:2.7:1.5:1.0. The fructan was identified as a (2→6)-β-d-fructofuranan or levan, the first example of levan synthesis by a Lactobacillus species. Strain LB 121 possesses glucansucrase and levansucrase enzymes that occur in a cell-associated and a cell-free state after growth on sucrose, raffinose, or maltose but remain cell associated during growth on glucose. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of sucrose culture supernatants, followed by staining of gels for polysaccharide synthesizing activity with sucrose as a substrate, revealed the presence of a single glucansucrase protein of 146 kDa. Growth of strain LB 121 in chemostat cultures resulted in rapid accumulation of spontaneous exopolysaccharide-negative mutants that had lost both glucansucrase and levansucrase (e.g., strain K-24). Mutants lacking all levansucrase activity specifically emerged following a pH shiftdown (e.g., strain 35-5). Strain 35-5 still possessed glucansucrase and synthesized wild-type glucan.

Introduction: Milk is an important food in Egypt and most of it is sold as raw milk in informal markets. Methodology: This study was conducted to investigate the public health risks of Escherichia coli and Staphylococcus aureus in milk sold in informal markets in Egypt. A total of 121 milk samples were analyzed for occurrence, virulence genes and antibiotic resistance of E. coli and S. aureus. Results: A total of 35/121 (28.9%) of milk samples were contaminated with 16/121 (13.2%) E. coli, 22/121 (18.2%) S. aureus, and 3/121 (2.5%) both isolates. Shiga-toxin producing E. coli (STEC), Enterotoxigenic E. coli (ETEC) and Enterotoxigenic S. aureus were detected in 5/121 (4.1%), 2/121 (1.7%) and 8/121 (6.6%) of the examined milk samples, respectively. Multiple drug resistances (MDRs) were showed by 14/16 (87.5%) and 21/22 (95.5%) of E. coli and S. aureus isolates, respectively. E. coli isolates showed high resistance for cephalothin (87.5%), ampicillin (68.8%) and tetracycline (68.8%), but were sensitive for gentamicin and chloramphenicol. Resistance phenotypes of E. coli were diverse; however, STEC isolates were significantly associated with co-resistance to cephalothin, ampicillin and tetracycline (P< 0.05). Two (9.1%) of S. aureus isolates were methicillin-resistant (MRSA) but sensitive to gentamicin (GS-MRSA). Five (22.7%) of S. aureus isolates were gentamicin-resistant methicillin-sensitive S. aureus (GR-MSSA). S. aureus isolates also showed high resistance for ampicillin (100%), tetracycline (90.1%) and sulfamethoxazole-trimethoprim (90.1%). Conclusion: These findings highlighted the potential public health hazards of E. coli and S. aureus pathogens in raw milk sold in informal markets in Egypt.

Abstract Background Human bladder cancers were recently suggested to be grouped into different molecular subtypes. However, little is known about the difference in gene landscape between upper tract urothelial carcinoma (UTUC) and lower tract urothelial carcinoma (LTUC) of Chinese patients. Herein, we analyzed the genomic characteristics of primary UTUC and LTUC in China. Methods The formalin-fixed paraffin-embedded specimens of 250 LTUC and 121 UTUC patients have been consecutively collected and have underwent next-generation sequencing (NGS) from 2018 to 2021 in 3DMed Clinical Laboratory Inc. Gene mutation and TMB level were analyzed by NGS, detected PD-L1 expression by using Dako PD-L1 IHC 22C3 pharmDx, Tumor Proportion Score (TPS) was used to determine expression of PD-L1. Results NGS analysis showed that, in the samples of UTUC, the top ten gene mutations were CDKN2A/2B 6.6% (8/121), TP53 5.8% (7/121), FGF19 5.8% (7/121), TERT 3.3% (4/121), PREX2 3.3% (4/121), FRS2 3.3% (4/121), KDM6A 2.5% (3/121), PIK3CA 2.5% (3/121), EGFR 2.5% (3/121), MYCL 2.5% (3/121); while in the LTUC, the top gene mutations were TP53 44.4% (111/250), TERT 31.6% (79/250), CDKN2A/2B 25.7% (64/250), FGF3 18% (45/250), KDM6A 17.6% (44/250), PIK3CA 16.4% (41/250), RB1 14.0%(35/250), DGFR3 13.6% (34/250), ARID1A 12.4% (31/250). Furthermore, TMB level and PD-L1 expression in the UTUC and LTUC were also analyzed, and results showed that although the gene mutation was totally different, but no significant difference was found on TMB level (p>0.05). However, among these patients, higher PD-L1 expression level were found in UTUC than LTUC (p<0.05). Conclusion UTUC and LTUC in Chinese patients hold different gene landscape and PD-L1 expression, this suggested these patients may have different prognosis and response differently to targetable therapy and immunotherapy. Abstract dss4-1 suppresses the growth defect, the block in invertase secretion, and the accumulation of 100 nm secretory vesicles seen in sec4-8 cells at the restrictive temperature1. Suppression is seen at 35°C, but not at 37°C whether dss4-1 is present in single copy or overexpressed. dss4-1 can not suppress a deletion of SEC4 and can not suppress the growth defect of ypt1-1 (seep. 121). dss4-1 has no effect on wild-type SEC4 cells.

Abstract Industrial relations continues to be a field which is dominated by men. The Annual British Universities Industrial Relations Association Conference (BUIRA), as one of the key events in the conference season for British industrial relations academics is a case in point. At the conference in 2001, held in Manchester, there were 121 named contributors in the published abstracts, of whom forty-two were women. The significance of such a comparison is that not only do women account for only 35 per cent of the industrial relations researchers at such a conference, but that (to borrow a phrase used with regard to collective bargaining in the context of a similarly male dominated trade union movement), ‘the absence of women at the table has to be part of the explanation for the absence of women on the table’ (Colling and Dickens 1998: 32 original emphasis). In other words, the lack of women researchers will mean a neglect of women as subject matter.

Freezing is an important thermal process with solar applications. The Phase Change Material (PCM) undergoes freezing process and the rate of freezing is influenced by natural convection in the melt if it is above the melting point. Experimental study was conducted on two PCMs namely Paraffin wax (m.p 63–65°C) and n-octadecane (m.p 27.7 °C). Steel and copper containers with insulation at both bottom and top by thermocol (1 cm thick) and acrylic plates (1 cm thick) are used to allow only radial heat flux. The PCMs were separately heated in a container to a known superheat and placed in an isothermal bath (± 1°C) containing the Heat Transfer Fluid (HTF). Water is used as HTF in this case. The temperature distributions in the PCM were measured using PT100 sensors with digital indicator to cover 10–16 points in radial and axial locations. In another separate set of experiments, the PCM under identical initial conditions of superheat is placed in the cylindrical metal container without any temperature sensors. Separating the frozen mass and unfrozen liquid at different time intervals and weighing, generate the frozen mass vs. time data following Sparrow et al (International Journal of Heat and Mass Transfer, 24, 273–284,1981). The range of parameters studied are given below: • Container L/D ratio: 0.6–5.4; • Initial Superheat of PCM: 5–35 °C; • Range of water temperature: 5–25 °C; • Volume of PCMs: 35–500 ml. From the experimental data of temperature distributions, frozen mass vs. time data were generated following El Dessouky et al (Journal of Solar Energy Engineering, Transactions of ASME, 121, 98–109, 1999), whose data pertains to melting outside tubes. The data of melt separation and weighing of n-octadecane and paraffin wax are compared with temperature-based data. A tentative correlation of entire data is presented.

Corn bran and wheat bran are coproducts of corn and wheat milling operations, respectively, and have lower commercial value mostly as animal feeds. They are rich sources of dietary fiber and phenolics. In this research, the effect of two physical modifications of wheat and corn brans 1) extrusion at two die temperature settings (140 and 120°C), and 2) grinding to < 500mm were evaluated on fermentation by Lactobacillus plantarum and Bacillus subtillis. Color measurement for extruded brans showed lower L* (lightness) and higher a* and b* (red/green and yellow/blue) when compared with the ground control brans. The diameter of the extruded brans at 25% moisture content was 3.3 mm and 3.49 mm for wheat and corn bran, respectively, compared to 3.0 mm 3.17 mm for wheat- and corn brans at 35% moisture. For fermentation performance, the extruded or ground brans were dispersed in water at 1:10 w/v, pH adjusted to 6.5 and pH 7, autoclaved at 121°C for 30 min, and inoculated (1% v/v) with L. plantarum and B. subtillis, respectively. The fermentation was carried out in 500 mL shake flasks at 37°C for 72 h with shaking at 200 rpm; 10 mL samples were taken out at 0, 6, 12, 24, 48, and 72 h time intervals for microbial viability and pH measurement. All the extruded or ground brans supported the growth of probiotic bacteria, which are expected to modify bran matrix favorably in making dietary/ nutrition components easily available for digestion.

Introduction: Risk stratification of patients with early endometrial cancer for recurrence is inadequate. Objectives: To study factors that influence recurrence in uterus-confined, early stage endometrial cancer (UCD). Patients and Methods: We studied 140 consecutive patients with endometrial cancer, operated at Action Cancer Hospital, Delhi, from August 2010 to September 2015. All patients underwent staging laparotomy, TAH + BSO + BLPND + para-aortic LN sampling, and omental biopsy. Adjuvant treatment was given as per the NCCN guidelines. They were followed up 3 monthly for 2 years, and 6 monthly thereafter. 121 patients (86.4%) had UCD (FIGO stages IA, IB, II). Excluding one post-operative mortality, and 4 who were lost to follow up, we included 116 patients in this study. Results: The median age of these patients was 60.5 years (range: 35-81 years), with median BMI of 31.2 kg/m2 (range=19.8-57.5). Diabetes or hypertension was present in either or both of 76 (65.5%) patients. The median pelvic LN harvest was 17 (range: 4-42). Eight (6.9%) patients had non-endometroid histology, and 5 (4.3%) patients had LVSI. Grade 1, 2, and 3 tumor was found in 74 (63.8%), 30 (25.9%), and 12 (10.3%) patients, respectively. The median follow up was 28 months (range 5-61 months), and recurrence was seen in 13 (11.2%) patients. On univariate analysis we found that age, co-morbidities (DM and HT), LVSI, and non-endometroid histology were related to recurrence. The tumor grade and adjuvant treatment did not influence recurrence rates. On multivariate analysis, presence of comorbidities and non-endometroid histology were independently related to disease recurrence (p=0.044, and 0.011, respectively). Conclusions: Disease recurrence was seen in one in ten patients with UCD, despite stage-appropriate treatment. Presence of co-morbidities and non-endometroid histology were independently related to recurrence.

Background. In the practice of clinical research, it is traditionally accepted to evaluate parameters that characterize the degree of impairment or loss of the function of an organ or system, and not their preservation. A fundamental change in the approach distinguishes a new trend in modern medicine, which makes it possible to assess the degree of preservation of functional resources, which was the basis of this study.Study purpose: To develop a method for dynamic quantitative assessment of the severity of arterial hypertension and evaluate the relationship between a qualitative indicator of arterial hypertension and a quantitative indicator.Instruments and Data Collection Procedure. Index of adaptive aptitude (IAA) (patent for invention No. 2342900 "Method for eavaluation of the functional reserves of the body", author - Kurnikova I. A.). using automated assessment (certificates of official registration No. 2007614560 and No. 2007613898).IAA=0.011(P-P*)+0.014(S-S*)+0.008(D-D*)+0.009(W-W*),Where: P - actual heart rate (b/min.); P* - ideal pulse rate (beats/ min.); S - systolic blood pressure, actual average per day (mm Hg); S* – ideal systolic blood pressure (mm Hg); D - diastolic blood pressure, actual average per day (mm Hg); D* - ideal diastolic blood pressure (mm/Hg); W - body weight at the time of examination (kg); W* - ideal body weight (kg); H - patient's height at the time of examination (cm).Assessment of daily heart rate variability on the Valenta system, equipped with a program for computer processing of spectral analysis indicators.Results:- 143 patients with arterial hypertension (AH) were examined. The relationship between a qualitative indicator - the severity of AH and quantitative indicators - rehabilitation potential, the numerical value of which is PAS, the circadian index (CI) as an indicator of increased sensitivity of the heart rate to sympathetic stimulation, and the LF/HF vago sympathetic balance coefficient, which increases with hypersympathatic tone, was considered. Statistical analysis of the surface plot using the weighted least squares distance allowed this relationship to be clearly demonstrated. Quantitative analogue of the Framingham arterial hypertension severity scale (AHSS) in a specific patient (certificate for invention №. 201152181)AHSS = 119,31 + 2,22× (CI) - 2,03× (IAA) - 1,33×(2×(CI)) + 2,72×(CI)× (IAA) + 7,06×(IAA)where: AHSS less than 120 - normal. Mild AHSS = 121 – 130 points; average severity of AHSS = 131 – 140 points; severe AHSS - 141 points or more. The effectiveness of rehabilitation is good with AHSS less than 120, satisfactory - with 121 - 130; unsatisfactory - more than 131.For illustration, the data of 54 patients are presented. Observation group "1" - patients with AH (BMI<25), mean age - 49±1.9 years (19 people). Observation group "2" - patients with metabolic syndrome (BMI>25; HOMA index> 2.5), mean age - 50±1.7 (35 people). All patients were assessed for the severity of hypertension according to the WHO criteria and the AHSS index at the beginning of the study. Normal value was achieved in 41.8% in 1st group during treatment, in 47.1% there was a decrease in the severity of AH according to AHSS. In group 2, in patients with metabolic syndrome during treatment, it was possible to normalize blood pressure in 55.8%, and to obtain satisfactory results in 35.9%.Conclusion. Developed mathematical modeling methods, a quantitative analogue indicator of the severity of arterial hypertension - AHSS allows you to dynamically monitor the effectiveness of treatment and evaluate the achieved result based on a quantitative assessment within the severity indicated by the classification. The effectiveness of the rehabilitation of patients with hypertension is considered sufficient if the AHSS decreases below the value of 120 units or one level from the baseline

Feline sporotrichosis is a zoonosis transmitted by Sporothrix brasiliensis that causes cutaneous and disseminated lesions. The study aimed to evaluate the efficiency of the clinical routine, diagnosis and treatment of feline sporotrichosis. 96 cases suggestive of sporotrichosis were analyzed, through clinical and laboratory examinations. A protocol defined by the Sporotrichosis Service of the Zoonosis Control area of the City of Vitória – ES was recommended. The material was collected by Swabs or by the imprint method , from an exudative lesion, identified, packaged and examined in the laboratory. Of the 95 (100%) animals were diagnosed with sporotrichosis, 69 (73%) submitted to the proposed treatment (itraconazole or itraconazole + potassium iodide) were cured and 26 (27%) were unsuccessful in the treatment. Therefore, it is concluded that felines between 23 months and 35 weeks, classified in stages I and II of treatment, achieved satisfactory cure rates and demonstrated that the therapeutic protocol used was effective.

In ASME 1997 International Gas Turbine and Aeroengine Congress and Exhibition. American Society of Mechanical Engineers, 1997. http://dx.doi.org/10.1115/97-gt-121.

Resumo:
Dispersion of coolant jets in a film cooling flow field is the result of a highly complex interaction between the film cooling jets and the mainstream. Understanding this complex interaction, particularly near the injection location, is critical for improving the predictive capabilities of existing film cooling models, especially when very high free-stream turbulence levels exist. This study uses a high frequency response temperature sensor to investigate the mean and fluctuating thermal field of a film cooling flow for two vastly different free-stream turbulence levels (0.5% and 20%). The high frequency response temperature sensor provides new information about the film cooling flow in terms of actual rms levels (Θ′), probability density functions (pdf’s), and frequency spectra of the thermal field. Results are presented for both free-stream conditions using round holes inclined at 35°, at a momentum flux ratio of I = 0.156 and density ratio of DR = 1.05. The mean thermal field results show severe degradation of the film cooling jet occurs with very high free-stream turbulence levels. Temperature rms results indicate levels as high as Θ′ = 0.25 exist at the jet-mainstream interface. More information is provided by the temperature pdf’s which are able to identify differences in the jet-mainstream interaction for the two free-stream conditions. With small free-stream turbulence, strong intermittent flow structures generated at the jet-mainstream interface disperse the jet by moving hot mainstream fluid into the coolant core, and ejecting coolant fluid into the mainstream. When the free-stream has large scales and very high turbulence levels, the jet-mainstream interface is obliterated by large scale turbulent structures originating from the free-stream which completely penetrate the coolant jet causing very rapid dispersion of the film cooling jet.
