Streszczenie:
Abstract One of the most important tasks in China’s supply-side structural reform is to effectively clean up zombie firms. Using industrial firm-level data in China from 1998 to 2007, we identified zombie firms in Chinese industrial sectors. Based on the identified zombie firms, we measured the survival duration of zombie firms and empirically studied the effect of fiscal subsidies on the survival duration of zombie firms by using survival analysis. Results revealed that fiscal subsidies significantly lengthened the survival duration of zombie firms. Specifically, fiscal subsidies not only reduced the possibility of zombie firms exiting the market, but more importantly, they reduced the possibility of zombie firms’ resurrection. Thus, fiscal subsidies do not contribute to the “death” or “resurrection” of zombie firms. These results are robust despite changing the identification method and the survival analysis method, and considering the endogeneity of fiscal subsidies. Our findings show that we should adhere to market-oriented mechanism in disposing zombie firms and cut off the channels that government transfuses to zombie firms by fiscal subsidies.

Streszczenie:
Though frequently comprehended as a vehicle for social satire or post-cultural speculation, zombie fictions also demonstrably mobilize the climatic unease of the current Anthropocene. Focusing in particular upon Max Brooks’s 2006 novel World War Z: An Oral History of the Zombie War, this article considers the complex politics which have frequently underwritten a mythical origin for pandemics in the Othered East, and their contemporary reproduction in western concerns regarding unregulated surgery and the capitalism of human tissue. The article then proposes that the deterioration of human culture consequent upon the fictional zombie pandemic interrogates the contemporary understanding of integrated nationhood and problematizes the dichotomy structured between geographically stable and refugee populations. The sudden eclipse of the competitive Anthropocene by a mindless Zombicene brings not renewal for a planet no longer supporting agriculture and industry but rather a hastening of perceived environmental collapse, where unregulated hunting and the uncontrolled burning of natural resources accelerate climatic deterioration, imperilling further the survival of residual humanity. As a type of apocalyptic fiction, the zombie narrative thus poses questions with regard to the persistence of conventional human behaviours, even in a post-capitalist environment, where the political concepts structuring nationhood have come to function as little more than a memory.

Abstract Among everything else post-apocalyptic video games have come to stand for, notions of in-group versus out-group communication are paramount. The Last of Us (2014, Naughty Dog/Sony Computer Entertainment) is a case in point. I look into the game’s use of subtitles and didactic texts in order to find out to the extent speech acts shape the player’s understanding of what the video game is. As an understudied aspect of video games, HUD or menu elements, as well as characters’ exchanges and voice-over narration, disclose what it is like to be alive, dead or in-between. Essentially, they show the tensions between the avatar and the gamer: the hero makes all of the decisions by himself and the player has to abide or stop playing all together. The avatar’s identity comes alive through speech acts, while the player is left outside decision-making processes. Survival horror gaming, with a religious twist, gives insight into the in-game discussion on the representation of the zombie rather than on the zombie experience as such. On screen, the interplay between speech acts and written language amounts to a procedural language, which suggests that variability in language creates an environment conducive to learning. Particularly, language use is all about group values and communication styles that should help gamers tell apart friends from enemies, good from evil and, finally, people from zombies.

This report describes mutants of the zebrafish having phenotypes causing a general arrest in early morphogenesis. These mutants identify a group of loci making up about 20% of the loci identified by mutants with visible morphological phenotypes within the first day of development. There are 12 Class I mutants, which fall into 5 complementation groups and have cells that lyse before morphological defects are observed. Mutants at three loci, speed bump, ogre and zombie, display abnormal nuclei. The 8 Class II mutants, which fall into 6 complementation groups, arrest development before cell lysis is observed. These mutants seemingly stop development in the late segmentation stages, and maintain a body shape similar to a 20 hour embryo. Mutations in speed bump, ogre, zombie, specter, poltergeist and troll were tested for cell lethality by transplanting mutant cells into wild-type hosts. With poltergeist, transplanted mutant cells all survive. The remainder of the mutants tested were autonomously but conditionally lethal: mutant cells, most of which lyse, sometimes survive to become notochord, muscles, or, in rare cases, large neurons, all cell types which become postmitotic in the gastrula. Some of the genes of the early arrest group may be necessary for progression though the cell cycle; if so, the survival of early differentiating cells may be based on having their terminal mitosis before the zygotic requirement for these genes.

AbstractHost behavioural manipulation is a common strategy used by parasites to enhance their survival and/or transmission. Baculoviruses induce hyperactivity and tree-top disease (pre-death climbing behaviour) in their caterpillar hosts. However, little is known about the underlying mechanisms of this behavioural manipulation. A previous study showed that the baculovirus Spodoptera exigua multiple nucleopolyhedrovirus (SeMNPV) induced tree-top disease at 3 days post infection in third instar S. exigua larvae and that light plays a key role in triggering this behaviour. Here we investigated the temporal requirements for the presence of light to trigger this behaviour and found that light from above was needed between 43 and 50 h post infection to induce tree-top disease. Infected larvae that were not exposed to light from above in this period finally died at low positions. Exposure to light prior to this period did not affect the final positions where larvae died. Overall we conclude that light in a particular time frame is needed to trigger SeMNPV-induced tree-top disease in S. exigua larvae.

In order to recognize and calibrate the two parts to its structure, self-sacrifice in zombie cinema will be examined in terms of survival-based and emotion-based motivational frameworks. The interaction of these frameworks will be unpacked and their properties, differences and similarities will be appraised and questioned. Examinations of this kind require three different analytical methods that therefore determine the structure of this article. The first section will outline how the survival and emotional-based motivational frameworks exist within the same sequence in Train to Busan (2016). The implications of this will be addressed in relation to the organization of modern neo-liberalism and what Paul Verhaeghe coins the neoliberal meritocracy. The second section examines the temporal projections of the characters in the sequence (specifically how the sequence depicts a character’s understanding of the future and how their present situation fits into that). These projections are cross-referenced with the specific example of the neo-liberal South Korean economic climate to add credence to the proposition that the need (or fetishization) of survival is a neo-liberal symptom and a hangover from the pressures that are ceaselessly exerted to keep its hierarchies in place. The final section of this article examines abjection and identity in relation to the chosen sequence in Train to Busan. It explores the generation of identity in relation to self-sacrifice and concludes that self-sacrifice is a necessary enforcer of a specifically neo-liberal competition.

The parasitoid emerald jewel wasp (<i>Ampulex compressa</i>) subdues the American cockroach (<i>Periplaneta americana</i>) with a sting to the 1st thoracic ganglion, followed by a sting to the roach’s brain, causing long-term pacification. The wasp then leads the cockroach to a hole where it lays an egg on the roach middle leg before barricading the entrance and departing. Although many aspects of the wasp’s initial attack have been investigated, few studies have detailed the egg-laying process and the subsequent fate of the larvae. Here I show that larval survival depends on precise egg positioning on the cockroach by the female wasp. Ablation of sensory hairs on the wasp’s abdomen resulted in mislaid eggs, which seldom survived. In addition, the cockroach femur may block the oviposition site. The wasp contended with this challenge with a newly discovered suite of stings, 3 directed into the 2nd thoracic ganglion which resulted in extension of the femur, thus exposing the oviposition site and removing a potential barrier to the wasp’s successful reproduction. When the femur was glued in place, the wasp stung the cockroach over 100 times, in an apparent fixed action pattern triggered by the obscured oviposition target. These findings highlight the importance of proper egg placement by the wasp, and reveal sensors and new neural manipulations that facilitate the process.

Abstract Bidirectional communication between the intestinal epithelial barrier and the immune cells in the underlying lamina propria is critical to human health. This study investigates the impact of human T cell subtypes on intestinal epithelial cell phenotype and survival. Blood-derived CD4+ or CD8+ T cells were isolated from healthy human participants using negative magnetic separation and co-cultured with healthy human colonic epithelial stem cell spheroids suspended in Matrigel. A subset of T cells was activated by anti-CD3/CD28 cross-linking for 24 h prior to co-culture. Only activated CD4+ T cells (ACD4) caused significant epithelial cell damage and death as measured by histological scoring of brightfield microscopic images and zombie violet+ epithelial cells detected by flow cytometry. The mechanism of cell damage and death was determined to be via soluble mediators, in that conditioned media from ACD4 but not activated CD8+ T cells (ACD8) significantly induced spheroid cell damage and death. A Luminex cytokine multiplex assay revealed that high levels of IFN-γ, IL-17, and TNF-α were produced by ACD4 as compared with ACD8. Neutralizing IFN-γ antibody completely protected spheroids from death induced by ACD4. In contrast, the addition of only IFN-γ protein did not induce spheroid cell death. This finding conflicts with currently published literature that indicates IFN-γ protein is able to induce spheroid cell death for both humans and mice. In addition, ACD8 also secrete IFN-γ, although to a lesser extent than ACD4, yet do not initiate spheroid cell death. These results indicate that there is a unique mixture of immune mediator released by ACD4s that mediate spheroid damage and death. DP1 DA-037997 T32 GM007250

Streszczenie:
This paper analyzes fantasy school dramas among the original Korean OTT dramas in the 2020s that deal with adolescents' entrance exams, competition, survival, and struggles. Exploring the “youth meritocracy” in these dramas. Originally, the meritocracy was considered the opposite of aristocracy, affirming that intelligence and hard work are the forces that create class mobility. However, over time, it has transformed into a meritocracy based on education and exams. Especially in Korea, it reinforces the obsession with prestigious universities and eventually becomes a way to justify endless competition and winner-takes-all thinking. However, in fantasy school dramas, young people don't just accept the distorted meritocracy as it is; they shape their own lives by exhibiting a variety of attitudes toward the meritocracy in schools which have become battlegrounds for life and death. Chapter 2 examines "High Cookie" a film about high school seniors at a prestigious school who are willing to use drugs to get to the top of the class, while Chapter 3 analyzes "After-School War Activities" a film about high school students who join the military as student soldiers to earn extra credit for the SAT and their unstable inner lives. Chapter 4 examines "All of Are Dead" explores the implications of teenagers choosing cooperation and solidarity over competition, albeit temporarily, in a zombie apocalypse. Chapter 5 focuses on the drama "Superior Shaman Gadusim" to examine the self-affirmation of adolescents who voluntarily sacrifice themselves to save their friends. In doing so, I argue that these dramas expose the absurdity of Korean society and youth meritocracy, but also reveal that they talk about the values that we need to recover beyond meritocracy, such as self-affirmation, friendship and love, cooperation and solidarity.

Streszczenie:
Abstract Despite recent advances, there is still a lack of treatment options for patients with high-risk multiple myeloma (MM), warranting the need for novel therapeutic targets. Monopolar spindle 1 (Mps1), also known as TTK protein kinase, is localized at the kinetochores and centromeres essential for the mitotic spindle checkpoint and centrosome duplication. In cancer cells, high levels of Mps1 help to support aneuploidy, a common malignant trait. In fact, high MPS1 expression correlates to unfavorable prognosis in colon and breast cancers. Here, we report the importance of Mps1 in MM and perform functional characterization of a novel Mps1 inhibitor, OSU-13. To assess the relevance of Mps1 in MM prognosis, we analyzed the CoMMpass database and examined the association of MPS1 expression with clinical outcome and genetic alterations in MM patients (n=769). Elevated MPS1 expression correlates with decreased overall survival (p-value = 0.0001) and decreased event-free survival (p-value &lt; 0.0001). In addition, MPS1 expression is higher in high-risk MM with specific genetic alterations, such as deletion of 17p (p-value = 0.0003), Myc translocation t(8;14) (p-value = 0.02), and gain of 1q21 (p-value = 0.0001). We therefore compared MPS1 expression in eight different MM cell lines and primary CD138 - and CD138 + cells isolated from bone marrow (BM) of four recently diagnosed, untreated MM patients. Gene expression levels assessed by quantitative qPCR were normalized to 18S mRNA internal control, and relative quantification was performed using the ΔΔCt method. MPS1 expression was higher in all MM cell lines compared to the average expression level in primary CD138 + BM plasma cells, with a minimum of 3.2-fold increase in L363 and a maximum of 16.2-fold increase in MM1S. In addition, MPS1 expression was 2-fold higher in CD138 + MM cells compared to CD138 - counterparts from the BM of the same patient, suggesting its selectivity as a target. Next, we characterized the effects of OSU-13, a novel Mps1 inhibitor. In an Mps1 target engagement assay (NanoBRET™) in HEK293 cells, we measured the relative levels of OSU-13-mediated inhibition of Mps1-NanoLuc binding to a fluorescent tracer in comparison to MM clinical candidates. OSU-13 showed EC 50=10 nM, almost 10-fold lower than other agents tested. Endpoint measurement by MTT (3-[4,5- dimethylthiazol- 2-yl]-2,5-diphenyltetrazolium bromide) assay was used to evaluate cytotoxicity of OSU-13. MM cell lines were treated with increasing doses of OSU-13 for 72h and cell viability was measured. OPM-2 was the most sensitive cell line (IC 50 = 610 nM), followed by H929 (IC 50 = 1440 nM), RPMI-8226 (IC 50 = 4196 nM), and U266 (IC 50 = 5071 nM). To investigate the kinetics and the mechanisms used by OSU-13 to cause cell death, we treated H929 and OPM-2 cell lines with 500 nM of OSU-13 for 24h, 48h, 72h, 96h, and 120h and assessed both Annexin V-FITC and Zombie-Aqua staining by flow cytometry. Treatment with OSU-13 caused an increase in Annexin V staining as early as 24h in H929 (2.1-fold) and OPM-2 (1.6-fold). Maximum increase of apoptotic cells (Annexin V single positive) was observed after 72h, with a 5-fold increase in OPM-2 and 8.6-fold increase in H929 cell line. Cell death peaked at 120h, with Zombie staining 74% of OPM-2 and 60% of H929 cells. The activity of caspases 3/7 was further assessed in H929 myeloma cells at five time points after treatment with different concentrations of OSU-13. Caspases 3/7 activation started after 48h with 500nM OSU-13, and peaked after 72h post-treatment with the same dose, in agreement with the Annexin staining kinetics results. Finally, we evaluated the in vivo effect of OSU-13 in tumor growth in a H929 MM subcutaneous xenograft model in CB.17 SCID mice. Mice were treated with 5 mg/kg or 10 mg/kg daily dose of OSU-13 per oral gavage for 21 days. Treatment with 10 mg/kg daily dose of OSU-13 produced a 22% tumor growth delay (p&lt;0.05). Collectively, we show here that elevated MPS1/TTK expression correlates with poorer prognosis in MM, and OSU-13, a novel Mps1/TTK inhibitor, induces apoptosis and cell death in MM cell lines, and decreases tumor growth in mice containing H929 cell xenografts. Together, our findings reveal that targeting the mitotic checkpoint using OSU-13 is a potential novel strategy, particularly for high risk MM. Abstract Constitutive genomic complexity, ongoing DNA damage, and accumulating mutations are observed with progression from monoclonal gammopathy of undetermined significance (MGUS) to active multiple myeloma (MM) to relapsed/refractory disease. Apolipoprotein B mRNA editing catalytic polypeptide-like 3B (APOBEC3B), a DNA cytosine deaminase, plays a prominent role in inducing mutations in multiple human cancers. In MM, APOBEC3B is linked to sub-clonal diversification, intra-tumor heterogeneity, and tumor evolution. Moreover, upregulation of APOBEC3B is associated with poor MM prognosis, suggesting that targeting MM cells with high APOBEC3B may represent a novel therapeutic approach. We here studied the upstream mechanisms of APOBEC3B dysregulation and further defined functional consequences of molecular manipulation of APOBEC3B in MM cells. We characterized its sequelae to identify novel strategies for cancer prevention or treatment by targeting this key driver gene of cancer mutagenesis. Since the expression of APOBEC3B is associated with replication stress in breast cancer, we first asked whether APOBEC3B levels are altered in MM cell lines upon treatments with Melphalan (Mel), an alkylating agent used to treat MM which is known to induce replication stress; or with ionizing radiation (IR). Using quantitative RT-PCR and Western blotting, we found that sub-lethal doses of Mel or IR induce APOBEC3B expression in a dose- and time-dependent manner in MM cell lines (n=7) associated with the phosphorylation of γH2AX. Interestingly bortezomib (btz), even at sub-lethal doses which triggers DNA damage signaling, also induced APOBEC3B expression in H929, MM1S, and U266 MM cells. Since DNA replication stress activates the ATR/ATM pathway, we next investigated whether these kinases mediate APOBEC3B induction following Mel- or IR- or btz-induced DNA replication stress. H929 and MM1S cells were treated with Mel or IR in the presence or absence of ATM or ATR inhibitors, and these cells were then lysed and assayed for APOBEC3B expression. Importantly, inhibition of ATR or ATM activation pathway significantly decreased Mel- or IR or btz-induced APOBEC3B, suggesting that replication stress induced by Mel, IR, or btz, activates transcription of APOBEC3B via an ATM/ATR dependent pathway in vitro. To test the effect of APOBEC3B on cell growth and survival, we used gene-specific CRISPR knock out (KO), shRNA knockdown (KD), and inducible-shRNA KD to study the functional impact of perturbation of APOBEC3B in MM cells. Both KO and KD of APOBEC3B decreased growth and survival in multiple MM cell lines sensitive or resistant to dexamethasone or lenalidomide. Using zombie aqua and annexin V-based flow cytometric analysis, we showed that APOBEC3B inhibition enhanced growth arrest, followed by apoptosis, in these MM cells. These data suggest an important role of increased APOBEC3B levels in MM cell survival. We next analyzed available data sources for MM cell lines from Cancer Cell Line Encyclopedia (CCLE) and the Genomics of Drug Sensitivity in Cancer (GDSC), which include microarray gene expression and drug sensitivity information. APOBEC3B expression negatively correlates with MM cell sensitivity to JQ1, a BET inhibitor which has been reported to inhibit MM cell growth and survival in vitro and in vivo. Importantly, in MM cell lines which are relatively resistant to pomalidomide and JQ1 than other cell lines, APOBEC3B KD by its shRNA enhances sensitivity to both drugs. Taken together, our findings provide new insights into the role of APOBEC3B in triggering cytidine deaminase-induced mutagenesis associated with progression of disease. Furthermore, we show that DNA replication stress triggered by Mel, IR, or btz upregulates APOBEC3B expression, which in turn confers drug resistance. The role of APOBEC in disease pathogenesis and progression, coupled with its role mediating drug resistance, suggest potential utility of targeting APOBEC in novel MM therapies. Streszczenie:
Background: Our RNA-Seq analysis of 249 untreated MYD88 mutated WM patients showed aberrant expression of stem cell, pre-B-cell, T-cell and myeloid genes in early WM B clones. This expression pattern was associated with inferior progression free survival with BTK inhibitor therapy and diminished with advancing disease. We therefore sought to validate this early signature at the protein level using flow cytometry and evaluate its potential as a clinical assay to monitor WM disease evolution. Methods: RNA-Seq predictions were validated on an independent cohort of treatment naïve MYD88 mutated WM patients and healthy (HD) samples. We designed a 13-color panel for flow cytometry detection of surface markers CD19, CD138, k and l immunoglobulin (Ig) light chains, CD34, CD117 (KIT), CD4, CD8A, CD33, FCGR3A (CD16), CEACAM8 (CD66b), pre-BCR (using an antibody that recognizes a unique conformational epitope of the BCR complex formed by the association of IGLL1 and VPREB1); Zombie Yellow was used to label dead cells. Peripheral B (PBMC) and/or BM mononuclear cells (BMMC) were isolated from WM and HD samples by density gradient centrifugation using Ficoll-Paque medium. Data acquisition was performed for at least 10^6 nucleated cells per tube in BDLSR Fortessa flow cytometer using the FACSDiva software. Results: RNA-Seq analysis showed high expression of genes atypical of mature B-cells that significantly diminished with increasing disease burden. For validation purposes, we selected genes reaching at least a 10-fold drop in their expression levels between the earliest and the most advanced disease cohort. In particular, the T-cell and myeloid genes including CD4, CD8A, FCGR3A, CD33, CEACAM8 as well as the prePCR surrogate light chain genes IGLL1 and VREPB1 respectively decreased from 67.6, 7.1, 80.5, 23.2, 8.3, 114, 38.3 to 2.4, 0.9, 1.7, 0.7, 0.4, 12.2 and 4.3 transcripts per million (TpM). A subclonal stem cell signature was detected, including CD34 and KIT, which dropped from 1.67 and 0.64 to 0.15 and 0.06 TpM, respectively. It is well established that WM represents a mature, post germinal center clone. Consistent with this notion, expression of memory B-cell and plasma blast genes in our RNASeq data set, including PRDM1, XBP1 and SPIB, was high in all patients regardless of disease progression and expression of the studied atypical genes. Our independent patient validation cohort had a median age of 68 (range 50-88 years), BM involvement by immunohistochemistry of 40% (range 10-95%), hemoglobin of 13.2 (range 8.1-13.4 g/dL) and serum IgM of 686 (range 536-1,151 mg/dL). An IgMk monoclonal spike was detected in all cases. All patients were mutated for MYD88. Within the CD19+k+-selected cells, we detected a subpopulation of cells co-expressing CD34, KIT, CD4, CD8A, FCGR3A, CD33, CEACAM8 and preBCR in all the patients, including in PBMC. As predicted, these markers were relatively enriched in samples with lower BM disease burden ( Figure 1, 2). Even in those with the highest BM involvement, the subclone was still detectable in both cellular compartments. Conversely, HD controls showed normal B cell populations and no evidence of the aberrant subclone. Conclusions: In WM primary cells, we observed the existence of a clonal B cell population that appears to re-activate stem cell programming early on and progressively is replaced by clones bearing more typical B cell markers with disease expansion. This study lays the foundation to develop a widely accessible methodology to accurately diagnose IgM MGUS/early-stage WM and monitor disease evolution. Flow-based sorting of these subclones will allow targeted multi-omic studies with insights into disease pathogenesis and targeted therapeutic applications.

Compulsive smartphone users’ psyches, today, are increasingly directed away from their bodies and onto their devices. This phenomenon has now entered our global vocabulary as “smartphone zombies,” or what I will call “iZombies.” Given the importance of mind to virtually all conceptions of human identity, these compulsive users could thus be productively understood as a kind of human-machine hybrid entity, the cyborg. Assuming for the sake of argument that this hybridization is at worst axiologically neutral, I will construct a kind of phenomenological psychological profile of the type of cyborg which engages in these patterns of behavior. I follow Judith Butler in seeing this identity as the result of performance practices, which as such can be modified or replaced using other performances. Pursing one such alternative, I compose a dancing critique that “reverse engineers” the choreographies implied by these cyborgs’ survival practices. The upshot of this critique is that their movement patterns do indeed align closely to those of horror cinema’s zombies. I therefore conclude by suggesting a few possible choreographic imperatives to facilitate more enabling ways of being for iZombie cyborgs today.

Streszczenie:
Abstract Multiple myeloma (MM) is a disease derived from genetically abnormal clonal plasma cells. MM cells aberrantly express several surface antigens compared with normal plasma cells. Among others, CD56/NCAM1 is present at variable levels in approximately 70% of MM patients. Very little is known about its role in MM; however, CD56 positivity in MM correlates with greater osteolytic burden and lower frequency of good prognostic features, such as the presence of t(11;14). We first analyzed 569 patients with MM diagnosed between 1/1/2005 and 12/31/2014 at the Ohio State University Wexner Medical Center, stratifying them based on the percentage of CD56-expressing clonal MM cells. The mean percentage of CD56-expressing clonal MM was 26.5%, with range from 0 to 100%; the Mean Fluorescent Intensity values varied, with a quarter of patients expressing CD56 at high intensity. We then evaluated patient outcomes based on the percentage of CD56-expressing clonal MM cells. We noticed that MM patients with more than 30 or 50 percent of CD56-expressing MM clonal cells have inferior clinical outcomes than patients with less than 30 or 50 percent of CD56-expressing MM clonal cells, with median overall survival of 9.61 versus 7.64 years (log-rank p-value: 0.004) or 9.30 versus 6.47 years (log-rank p-value: 0.0009), respectively. We then demonstrated by conventional and real-time PCR analyses that the predominately expressed CD56 isoform in MM has signaling potential with a transmembrane portion and cytosolic tail. Therefore, we evaluated the functional role of CD56 in MM. By gain-of function studies in U266 and MM.1S MM cell lines, we showed that overexpression of CD56 promotes MM growth and viability; the opposite effect occurred with CD56 silencing in H929, OPM-2, and RPMI-8226 MM cell lines, which leads to reduced MM growth and increased apoptotic cell death. Overexpression of CD56 resulted in the phosphorylation and hence activation of ribosomal protein S6 kinase A3 (RSK2) and of the transcription factor, cAMP responsive element binding protein 1 (CREB1). This induced CREB1 binding to DNA consensus CRE elements, and promoted transcription of CREB1 targets, the anti-apoptotic genes BCL2 and MCL1. CD56 silencing in H929 and OPM-2 MM cell lines had opposite effects, with reduction of phospho-RSK2, phospho-CREB1, MCL1, and BCL2 levels. We then used shRNAs targeting RSK2 and CREB1 or specific inhibitors (BI-D1870 that is a RSK2 inhibitor, and 666-15 that is a CREB1 inhibitor) at 0.1-1 microM concentration. We evaluated viability by MTT assay or Zombie dye staining on CD138 positive MM cells and apoptosis by Annexin V-PI staining. We demonstrated that CD56 positive MM cell lines (H929, OPM-2, and RPMI-8226) or patients with high CD56 expression (&gt;30% of CD56-expressing clonal MM cells) are more sensitive to RSK2/CREB1 inhibition compared with CD56 negative MM cell lines (U266, L363, and MM.1S) or patients with low CD56 expression (&lt;30% of CD56-expressing clonal MM cells). Using similar strategies, we also proved that CREB1 is essential to CD56-protumoral phenotype, since CREB1 inhibition reduces cellular growth and viability in CD56 overexpressing U266 cells. RSK2 and CREB1 inhibition mimic CD56 silencing with decrease of BCL2 and MCL1 mRNA and protein levels. Furthermore, we observed that CD56 signaling by CREB1 activation decreases CRBN expression, reducing responses to lenalidomide. Conversely, CREB1/RSK2 blockade rescued CRBN levels in CD56 positive MM cells and increased lenalidomide response. These results support the hypothesis that targeting CREB1 is hence a so far unexplored but potentially effective synthetic lethal strategy for CD56-expressing MM patients. In conclusion, our study defines an effective threshold for therapeutic intervention in CD56-expressing MM patients. Moreover, our data pioneer the use of CREB1/RSK2 inhibition in CD56-expressing MM cells, either as single agents or in combination with lenalidomide, suggesting that CD56 can be a prognostic and predictive factor of response in MM. Abstract The Walking Dead is a popular TV series depicting a catastrophic and violent world. After a pandemic that turns humans into zombies, we witness the collapse of civilization with all its institutions, the depletion of the resources, and the struggle to build a new world in the middle of the wars between surviving groups. It illustrates a world of literal and metaphorical homo homini lupus. Some people choose sheer survival, and others try to build a moral, civil world. In this article, I propose a reading of this series from a Kantian perspective by employing his interrelated ideas on history, ethics, and politics. I claim that The Walking Dead represents the state of nature and the violence it contains, and illustrates the course of history toward a civil society as defined by Kant.

The cancer microenvironment plays a crucial role in promoting metastasis and malignancy even in normal cells. In the present study, the effect of acidic and conditioned media of cancer cells (MDA-MB-231), separately and in combination, was studied for the first time on the cell death mechanisms and DNA methylation of normal fibroblasts (NIH/3T3). Cell survival of conditioned media was rescued by the addition of acidic media to conditioned media, as shown by the results. Cell metabolic activity is deviated in a direction other than the Krebs cycle by acidic media The mitochondrial metabolic activity of all groups was enhanced over time, except for acidic media. Unlike the highest amount of ROS in conditioned media, its level decreased to the level of acidic media in the combination group. Furthermore, cells were deviated towards autophagy, rather than apoptosis, by the addition of acidic media to the conditioned media, unlike the conditioned media. Global DNA methylation analysis revealed significantly higher DNA hypomethylation in acidic media than in normal and combination media. Not only were cells treated with conditioned media rescued by acidic media, but also DNA hypomethylation and apoptosis in the combination group were decreased through epigenetic modifications. The acidic and conditioned media produced by cancer cells can remotely activate malignant signaling pathways, much like zombies, which can cause metabolic and epigenetic changes in normal cells.

In a horse’s belly: Interspecies relations as a critique of civilization Jenny Ingridsdotter & Kim Silow Kallenberg Keywords: natureculture, animals, popular culture, masculinity In this article, we analyse how the relationship between humans and other species is portrayed in contemporary films and series that have the rise and fall of civilization as their theme: Into the Wild (2007), The Revenant (2015), Into the Forest (2015) and The Walking Dead (2010-). The purpose is to understand how relationships between humans, animals and non-humans are portrayed. The films/series have been chosen on the basis of their portrayal of social downfall or social dissatisfaction and on the grounds that they are also widely recognised and popular portrayals. The analysis focuses on two male characters (The Revenant, Into the Wild) and two female characters (Into the Forest and The Walking Dead) to investigate relationships between humans and other species (animals and zombies) when it comes to survival, and how these relationships are possibly conditioned by gender. Methodologically, we approach these popular cultural depictions as ethnographers, with human meaning-making as the primary point of departure. Theoretically, we use concepts developed in the field of human-animal studies. The analysis shows that there are differences between the representations of different species and that these representations are also conditioned by gender in the human characters. Where men are alone in their struggle against nature, women are part of social relationships where they, together with others (human and non-human), struggle to survive. The analysis further shows how animals and other species constantly condition and enable human existence. However, lacking human language, the animals – who are absolutely vital for the actions and survival of the human characters – are rendered unimportant.

This paper is a study of transformations and mutations of a natural human desire, to be buried in one grave with one’s beloved. Most partners don’t die simultaneously, and burial-practices needed to provide flexibility for the dead and for the living. At the same time, religions had Views about the grave and the afterlife, and about the survival of the individual. Judaism and especially Christianity featured an astonishing doctrine, the Resurrection of the Flesh. Starting from Roman antiquity and in its epitaphic practices, the paper analyzes an intriguing early 4th C. Gallic poem, the Carmen de Laudibus Domini and its account of how the corpse of a dead woman was momentarily reanimated to greet her husband’s corpse. The poem reworks the resurrection of Lazarus with a little help from Juvencus. But a crucial (and unrecognized) source is (perhaps indirectly) Tertullian’s De Anima. These texts somehow generated a Late Antique urban legend about the mini-Resurrections of lovers’ bodies than can be traced into the central Middle Ages and beyond. It proved astonishingly lively and adaptable—to mariages blancs, to homosocial monastic situations, and to grave robbery, to name a few. This deeply sentimental legend needed to elbow aside darker phenomena, charnel (and also erotic) horrors from the pagan past, including zombies, vampires, and revenants, in order to preach its Christian message and help lovers who had been separated by death. Such resurrections were a down-payments on The Resurrection.

Posthuman Gothic is one of the recent emerging areas of research in the twenty-first century. It explores the different ways in which Posthuman thoughts and ideologies conflate with Gothicism in all its contemporary variations. Primarily, the posthuman gothic concerns itself with the human beings’ technological, biomedical, and supernatural experiments with the human body and consciousness that alters the human identity into the posthuman. The possibility and capability of humans to alter the human identity into something other than human or into the ‘posthuman other’ create anxiety among humans. The humans’ fear of becoming the posthuman other or encounters with the posthuman other over the course of evolution is the nucleus or the driving mechanics of the posthuman gothic genre. The Posthuman Gothic fiction deals with the scientific, technological, as well as supernatural developments on cyborgs, android robots, bio-engineered transhumans, vampires, zombies, and Frankenstein monsters in a gothic setting that opens up a dystopian posthuman future or condition. Ahmed Saadawi’s Frankenstein in Baghdad deals with the Frankenstein monster kind of posthuman that kills humans and poses a threat to human lives in a post-modern gothic setting. In this paper, the researchers try to highlight the dovetailing of the posthuman thoughts with the post-modern gothic setting and the posthuman monstrosity of the posthuman other, i.e a Frankenstein monster with multiple consciousness that threatens the human identity, lives, survival, and the very existence in Ahmed Saadawi’s Frankenstein in Baghdad through the posthuman gothic lens.

Critics have repeatedly focused on the political subtexts of the living dead films of George A. Romero, revealing, notably, how they reflect specific social concerns. In order to determine what makes the zombie movie and the figure of the zombie so productive of political readings, this article examines, first, the classic zombie movies influenced by voodoo lore, then Romero’s initial living dead trilogy (1968-1985), and finally some of the most successful films released in the 2000s. Resorting to a post-structuralist framework including Althusser’s notions of state apparatuses, Foucault’s distinction between subjection and subjectification, and Butler’s analyses of subversive resignification, the author argues that, while the classic zombie is entirely subjected to the master, and thus to the meanings the latter imposes, Romero’s living dead resist and sometimes create meaning, revealing both the contingency of the structures they disrupt and the constructiveness of the identities projected onto them. Resistance, then, is no longer directed at an “unnatural” order, as in the classic zombie movie, but at the “natural” order and is enabled by the contingency and multiplicity associated with the outbreak. The recent trend of zombie movies, whether survival horror or zombedies, confirms the diversity of complex, sometimes contradictory meanings enabled by the figure of the zombie.

Released in the West as Resident Evil, George A. Romero's model of the animated cadaver and Lucio Fulci's command of the atmospheric combined to provide the main inspiration for the game's visual and narrative aesthetic (Poole, 79): a complete and immersive reworking of the zombie genre by Tokyo based video game producer Shinji Mikami. Although not the first video game franchise to attain cult recognition or make a successful transfer to the big screen - Lara Croft and the Tomb Raider franchise come to mind - a determination, "to repackage the zombie as a mainstream monster and an icon of cool" (Russell, 171) clearly paid off: The Resident Evil games had succeeded...

Streszczenie:
The Walking Dead is an American post-apocalyptic horror drama television series based on the comic book series of the same name. In the opening episode, Sheriff’s Deputy Rick Grimes awakens after months in a coma in an abandoned hospital to find a post-apocalyptic world dominated by flesh eating zombies. The cause of the apocalypse is unknown, and Grimes does not know what has happened to his family. The start of the programme is situated around his quest to find his family, and the encounters he has with the many other survivors along the way. The plot of The Walking Dead centres on the survivors of the apocalypse as they search for safe haven away from the “walkers”, or “biters”; the first series focuses on how they cope with the immediate realities of life in the post-apocalyptic world. From the outset, the programme finds its way through the upheaval and destruction of everyday American life. Decay is persistent and inevitable in the threat of death from the “walkers”; this familiar Gothic trope is ever-present throughout the series. This paper uses a Gothic focus to examine The Walking Dead and considers the disintegration of society and family after the zombie Apocalypse. It focuses primarily on the first series of The Walking Dead, and examines Gothic tropes through a discussion of the decay of the walkers, the decay of the family, and the decay of society.Zombie Gothic It is important to examine the zombie narrative within a Gothic framework. Kyle Bishop argues that “zombies and the narratives that surround them function as part of the larger Gothic literary tradition, even as they change that tradition as well” (31). In contrast to other Gothic traditions that began in literature, the zombie is unique as it began in folklore, cinema and drama. Similarities to other Gothic traditions exist, such as ghosts and vampires, and the zombie narrative serves as a vehicle to examine cultural anxieties and prevailing attitudes. In fact, Bishop suggests that the zombie narrative has now proven itself to be as interesting and complex as more established Gothic traditions (31). Just as earlier Gothic traditions allowed people to explore themselves in the mirror that Frankenstein (1818) or Dracula (1897) provided, the zombie allows for a more modern examination: a world that is increasingly complex with its technological and cybernetic advances yet a world where humanity has still not resolved the great differences that exist. Bishop argues that “during the latter half of the twentieth century, for example, zombie movies repeatedly reacted to social and political unrest, graphically representing the inescapable realities of an untimely death…” (11). The zombie narrative’s ability to adapt to cultural anxieties make it part of the Gothic tradition. Bishop also argues that, in a post–9/11 climate, the zombie film works as an important example of:the contemporary Gothic, readdressing “the central concerns of the classical Gothic,” such as “the dynamics of family, the limits of rationality and passion, the definition of statehood and citizenship, the cultural effects of technology.” In addition to exposing such repressed cultural anxieties, Fred Botting emphasizes how Gothic narratives “retain a double function in simultaneously assuaging and intensifying the anxieties with which they engage.” (26) The Gothic situates itself from the 18th century as writing of excess and this tendency permeates much of the genre’s narratives, characters and settings. The mutability in Gothic texts provides a platform for many social issues and anxieties to be addressed; its ability to shift and adapt in order to reflect contemporaneous social trends is partly what has enabled it to remain popular (Botting Gothic). The zombie forces a confrontation with the fears of life and death, freedom and enslavement, and the destruction of modern society. These include “the exploitation of the masses in capitalist society, the soullessness of modern-day life, our fear of global apocalypse, our revulsion at the reality of war, and the inevitability of death” (Graves 9). The rise of the zombie narrative plays on humanity’s fear for the future: that modern civilisation has many fundamental weaknesses that may ultimately collapse through one form of global disaster or another. The rise of the zombie narrative plays on humanity’s fear for the future: that modern civilisation has many fundamental weaknesses that may ultimately collapse through one form of global disaster or another. I argue that the zombie is part of a new kind of Gothic with a new monster for a new age. This new monster facilitates the Gothic’s ability to remain relevant in a post-industrial, cyberspace era. Unnatural death is now more horrific, pervasive, and far-reaching than Walpole ever could have imagined when he wrote the now canonical Gothic novel The Castle of Otranto (1764), and the zombie works as a dramatic manifestation of this ever-present anxiety. The Gothic narratives of the zombie now represent a more modern reaction to the threats posed in the 21st-century, post-9/11 world which is potentially more traumatic than the threats posed in earlier Gothic novels of the 19th and 20th century. Global pandemics may manifest as a zombie apocalypse or disease that threaten complete annihilation of civilisation. Teresa Goddu emphasises how “the Gothic is not a trans-historical, static category but a dynamic mode that undergoes historical changes when specific agents adopt and transform its conventions” (32). Zombie narratives have updated Gothic conventions to then reflect modern day anxieties and fears. As Botting argues, “Gothic figures” represent anxieties associated with turning points in cultural historical progress (2002). Zombie narratives, then, serve to allow a confrontation with more modern terrors and threats that exist; the zombie narrative, it can be argued, confronts Gothic tropes and places them in a contemporary context. Cultural anxieties have been nurtured about the rise in terrorist activity around the world, witnessed with the collapse of the World Trade Centre towers, and the threats of pandemics that might eviscerate the human race in the form of SARS. The zombie narrative then represents a logical growth in Gothic monsters that have been used to explore modern day cultural anxieties. Post-Apocalyptic America and Gothic Zombies Apocalypse and zombie narratives represent the worst case scenario for both the people of USA and the world. Zombies, vampires and other apocalyptic monsters are used as faceless creatures to present either an unknown threat or pose them as social critique (Bo). The first signs of the Gothic are present from the start of The Walking Dead, where the familiar American suburban scene has altered into a mutilated disintegrating version of society. In the series, threat comes not just from the “walkers” but also from the fellow survivors who no longer obey the pre-apocalypse laws and way of life. Pre-apocalypse, Rick Grimes was a sheriff, and an upholder of the law. The series starts with Grimes in a police car having lunch with his best friend Shane Walsh while they discuss the differences between men and women. In a police shoot-out, Grimes is shot and awakens to find society as he knew it destroyed. After waking from his coma, he returns to his family home and meets two other survivors, Morgan Jones and his son Duane, who explain the implications of the zombie apocalypse to him. Grimes searches for his family and returns to the police station. He puts on his uniform which appears as a symbol of the normality he thinks he can return to. However, as he goes to get petrol, Grimes looks under a car and sees a young girl’s pink slippers. He calls to her saying “don’t worry little girl”, as the girl drags a pink rabbit. She turns around to face Grimes and runs at him: it is revealed she is a zombie. She is a grotesque parody of a little girl with her distorted and mutilated face; as Grimes draws out his gun and shoots her in the face, she is an uncanny reminder that humanity has permanently altered. The narrative of adults and police officers protecting innocent children is quickly subverted from the start of the series. From this moment, Rick Grimes is permanently fighting his own loss of morality and humanity in a society that has become a distortion of what it once was. The term “liminality” is employed by critics and theorists of the Gothic to refer to spaces or bodies situated:either on or at the recognized borders or boundaries of subjective existence. In eighteenth-century Gothic writing, these thresholds were mainly encountered through liminal spaces. These were often mountain ranges, secret rooms, and hidden passages. From the nineteenth century onward, the human body has increasingly become a liminal site where normative boundaries are challenged; the monster, vampire, and werewolf, are all liminal beings. (Hughes, Punter and Smith)The zombies continues the Gothic tradition of liminality and is perhaps more frightening as they represent the dissolution of death, yet are still in some form “alive”. In The Walking Dead, the survivors are also neither completely alive nor dead, as they are on the edge of losing life as they know it and becoming consumed by the undead. The “walkers” operate as terrifying prompts that what was once considered an incontrovertible fact—the difference between life and death—is not as final in the post-apocalyptic world of the series. The “walkers”, therefore, are walking manifestations of decay and liminality, a reminder that the fear of death has been transmuted into the fear if an even more dangerous entity, neither living nor dead. Gothic, Misha Kavka argues, is often about fear, localised in the shape of something monstrous that electrifies the collective mind (Kavka). In this case, the zombies are tangible displays of how a pandemic or global outbreak could alter humanity forever. Gothic is also about the paranoia around body manipulation, defined as a projection of the self on to the outside world where the boundaries blur between self and other (Kavka). In the zombie narrative of The Walking Dead, the boundaries between the living and dead collapse when decay reanimates into the liminal form of the “walkers”. Decay of the Family The death of the family unit as a recurring trope is raised early on in the series, and the initial problem for Rick Grimes is locating his missing family. Grimes teams up with Morgan Jones and his son Duane. Morgan has his own dilemma when faced with the thought of killing his wife who has turned into a “walker”. She returns to their family house, and seems caught between life and death as if she has some memory of the life she had before. Rick Grimes’s family dilemma is further exacerbated when he finds his wife and son with other survivors who have formed a group. Thinking that her husband Rick was dead, Lori Grimes has started a relationship with Grimes’ best friend, fellow police officer Shane Walsh; a growing tension grows between the two men as the series continues. Ultimately this leads to a fight between the two men as the jealousy grows and each have different ideas on how to best keep the survivors safe. Where the men were once allies and friends, the apocalypse has turned them into enemies. Though the zombies are the most manifest threat to the survivors, there are other threats that come to the fore in The Walking Dead. These threats are in the form of the changes that occur between the characters (Bo). What were once everyday events turns into dangerous events: getting water, petrol and food, for example, become life-threatening activities, and the survivors must trust people who they meet up with on their travels. Rick Grimes’ pre-apocalypse ethics and humanity are tested by the new society. For example, he allows his son Carl Grimes to carry a handgun which Carl later uses to save Rick’s life. In contrast to the Grimes family that is at the centre of the narrative, another group of survivors live at a farm and are led by Hershel Greene, a farmer and religious leader. Their treatment of the “walkers” represents a different approach to the zombie apocalypse. Hershel keeps zombies in a barn and sees them as sick people, while Rick sees them as monsters. Many in the barn are Hershel’s family members; it is only later in the series that Hershel comes to see the zombie family as monsters intent on killing all human survivors. With the connections to family and love, the zombies act as a mirror to the human survivors of what they may potentially become. Societal Decay From the start The Walking Dead Rick Grimes needs to grapple with a world profoundly altered by the zombie apocalypse. The hospital is abandoned except for stray zombie corpses, and it is clear that the once secure place of the hospital is no longer a haven for the sick. One of the most obvious signs of decay is the streets littered with abandoned vehicles, and there are outward markers of chaos and apocalypse. There is much that is Gothic and uncanny in The Walking Dead, where cognitive dissonance is opened up when the familiar becomes strange. The world ostensibly looks the same but will never be normal again for Rick Grimes and the survivors. Here the “true horror lies in that which is most immediately at hand that the most proximal bears the capacity to contain the utterly unfamiliar” (Chopra). The decay of society is made both manifest and melancholic as it evokes the anxiety of being simultaneously normal and abnormal. The once known, or normal, world has become strange and unfamiliar. For example, in the first series of The Walking Dead there is a reference to the classic zombie horror film Dawn of the Dead (1978), where the survivors find themselves trapped in a department store, a famous scene commented upon by Bishop:This instinctual “drive to shop,” as it were, is repeatedly emphasized by Romero, who shows the mindless creatures pressed up against glass doors and windows, clamouring to get inside the shops, in a gross parody of early-morning-sale shoppers, to resume their earthly activities of gluttonous consumption—indeed, as Kim Paffenroth points out, their addiction for the place exists beyond death. (Bishop 41) As the maniacal governor in The Walking Dead later observes about the zombies: “The thing you have to realize is that they’re just us—they’re no different. They want what they want, they take what they want and after they get what they want—they’re only content for the briefest span of time. Then they want more” (Bishop 140). The zombies then serve as a mirror for the worst of humanity. Zombies further mirror other aspects of humanity that are hidden and ignored. Barbara Creed suggests that the popular horror film brings about a confrontation with the abject (the corpse, bodily wastes, and the monstrous-feminine), and by doing so re-draws the boundaries between the human and non-human (Creed). She argues:Firstly, the horror film abounds in images of abjection, foremost of which is the corpse, whole and mutilated, followed by an array of bodily wastes such as blood, vomit, saliva, tears and putrefying flesh. (Creed 253)The zombies/”walkers” in The Walking Dead are abject, mutilated walking corpses. Creed argues that the blurred boundaries between life and death, and the antinomies that humans like to ignore or pretend do not exist, are seen in creatures like zombies. Abjection is usually represented by bodily fluids such as pus or blood or a kind of in-betweenness, such as the zombies’ state between life and death. Mark Bould and Sherryl Vint argue that apocalyptic fiction uses the scenario of the end of the world as a way to rebuild and reorganise society (23). The very question of possible futures and society in a post-apocalyptic world is raised and questioned in The Walking Dead when everyday survival is at stake. This alters, however, when Rick Grimes has to focus on the long-term survival of the group and his family when his wife becomes pregnant. Lori Grimes argues that the world they find themselves in is no place to raise a child and to establish new lives. Rick does not agree with Lori and her pessimistic view of society as it stands. In an uncanny twist, the survivors do not know what has caused the apocalypse, but they later learn that everyone is infected and will re-animate when dead: they are the “walking dead”.Conclusion The Walking Dead is a modern Gothic text that uses many of the tropes of the Gothic to explore cultural anxieties present today. One such area is that of decay, chaos and lawlessness in the post-apocalyptic world of the zombie. This is a key area of the Gothic tradition played out in the modern afflicted world. At the start of The Walking Dead, society is seen as “normal”, two police officers are eating lunch in their vehicle and talking about life. After Rick Grimes awakens after his coma what was once “normal” has transformed into a site of uncanny horror, suspense and terror. Much of the first series is spent with Grimes and his survivors trying to contain and combat the zombie threat. Botting argues that early Gothic fiction articulated a shift from a feudal economy to a capitalistic one (2008). In similar vein, in The Walking Dead the future is one where capitalist society has totally collapsed. This could be a critique of the 2008 financial crash, or a fear of what could happen if a pandemic were to occur that ended consumer life and society as it is known (Bishop 41). It also demonstrates that what is seen as established norms quickly disintegrate in the new post-apocalyptic society. Social structures in the post-apocalyptic world no longer function as they once did. The normality of a pregnancy which should, under “normal” circumstances, herald hope for the future, sets off ambivalence in the Grimes family about the life circumstances the survivors find themselves in, and the future that is available to them or their offspring. Core institutions and structures have fallen; the hospital at the start of the series no longer functions and the police are no longer upholders of the law. Chaos and anarchy are now the everyday life that confronts the survivors. The survivors are frequently left with questions about what is the point is of their lives. At the centre of the chaos is the change in family and society. The structures of modern society are seen as flimsy and easily disturbed in the post-apocalyptic zombie future. As Botting says, “uncertainties about the nature of power, law, society, family and sexuality dominate Gothic fiction” (Gothic 3). The modern day Gothic then questions these key areas of society. The death in the Gothic post-apocalyptic zombie future is that of society as well as individuals. References Bishop, Kyle William. The survival of zombie firms severely crowds out healthy corporate investment and employment growth, eventually undermining economic growth potential. Hence, how to properly address the problem of corporate zombification is an essential part of achieving high‐quality economic development in China. In recent years, the phenomenon of common institutional ownership has become increasingly widespread in the capital markets and has a significant impact on the strategic decisions of companies. There are currently two views on the corporate governance role of common institutional ownership: the synergistic governance effect and the collusive fraud effect. Using data from Chinese‐listed firms from 2009 to 2021, this paper finds that common institutional ownership can significantly inhibit the formation of zombie firms. The higher the degree of their linkage and the greater the shareholding, the more pronounced the synergistic effect. The findings remained valid after considering the endogeneity issue and conducting robustness tests. Furthermore, the mechanism test suggests that common institutional ownership inhibits the formation of zombie firms by improving internal control quality and reducing agency costs. This paper contributes to the study of how to inhibit the formation of zombie firms by identifying common institutional ownership from the perspective of external governance mechanisms. In addition, this paper enriches the research on the economic consequences of common institutional ownership. Finally, various practical implications for policymakers may be realised, which may help curb the trend of corporate zombification through equity‐based instruments.

This paper seeks to historicize and demystify “Voodoo” religion in Africa and Haiti while also drawing comparisons and contrasts to concepts and themes related to “the gothic”. What is assumed to be “supernatural” or “paranormal” in Western and Gothic circles has long been a part of everyday reality for many peoples of African descent and devotees of Vodun in Western Africa and Vodou in Haiti. Tropes that are essential to realms of the gothic (supernatural characters, mystery, the macabre, spirits, and paranormal entities) — are also central to the cosmology and liturgy of so-called “Voodoo”. As “the gothic” undergoes a resurgence in academic and popular cultures, so too does “Voodoo” religion. And yet, both terms continue to be conflated by popular culture, and by equating “voodoo” with “the gothic”, the true spirt of both concepts become confounded. A certain racialized Eurocentric hegemony devalues one of the world’s least understood religions (“Voodoo”) by equating it with equally distorted concepts of “the gothic”. As globalization transforms society, and the neo-liberal order creates more uncertainty, the continued distortion of both terms continues. Vodun does more than just speak to the unknown, it is an ancient organizing principle and way of life for millions of followers. Vodou/Vodun are not cognates of the “American Zombie gothic”, but rather, are a mode of survival and offer a way of seeing and being in an unpredictable world.

The post-apocalyptic environment has been popular in video games for many decades—from nuclear fallout to alien invasions, there is a fascination with the decline of mankind. This article looks, in light of the Covid-19 outbreak, at the depiction of failing and inhuman worlds in video games about pandemics. The environments in games such as Bioshock (2007), Left 4 Dead (2008, 2009), and The Last of Us (2013), are unflinchingly cruel, often depicting humans (and the player) as selfish entities seeking only self-survival. These environments are destructively violent, and, although the player is often surrounded by ‘mutants', these worlds are also oppressively lonely. This article looks at the fears reflected in pre-Covid-19 pandemic video games and what they say about our world, offering a retrospective view now we are living through a global pandemic perhaps not so different from those found in these games.

We explore the notion that consciousness may be more of a belief than an empirically verifiable fact, shaped by the subjective nature of our experiences and the inherent limitations of reporting them. Contemporary science has yet to provide definitive proof of subjective experience beyond self-reports, which even sophisticated machines could mimic under specific conditions. Philosophical concepts such as phenomenological zombies—creatures identical to us but lacking consciousness—highlight the difficulty of distinguishing between actual conscious experience and mere behavioral reports. Current experimental designs often conflate metacognition (beliefs about perception) with consciousness, as seen in Higher-Order Thought theories and the Perceptual Reality Monitoring theory. These frameworks suggest that our sense of being conscious arises from metacognitive processes, potentially leading to cognitive biases. Studies involving disruptions to brain regions responsible for metacognitive accuracy further complicate our understanding of consciousness. Additionally, phenomena like delusional misidentification syndromes challenge the assumption of a coherent self that reliably perceives and reports reality. By questioning these assumptions, we propose that consciousness might be an elaborate belief system, a convenient construct for survival rather than an intrinsic quality of our being. This perspective urges a reconsideration of the fundamental nature of consciousness and our approach to studying it.

Walters, Eric. The Rule of Three. Toronto: Penguin Canada Books Inc., 2014. PrintWith all the dystopian novels written over the last ten, years Eric Walters has composed a refreshing and believable look at how a community survives after the world has been changed by a computer virus which has immobilized any vehicle, utility, or device that requires computer technology. The villains the community members have to face are not zombies or other fantasy creatures but humans. How long will this disaster last? They don’t know. The “rule of three” is an idea that we can live for three seconds without air, three days without water, and three weeks without food. Keeping this in mind, the protagonist, sixteen-year-old Adam Daley, his mom, the police chief, and neighbour Herb, an ex-spy, must come up with ways to keep their community of Eden Mills safe and self-reliant. Three days after the outage, people begin to panic because the basics of life – water and food – become either inaccessible or depleted. The protection of one’s own family becomes the priority and mob mentality and the survival of the fittest become paramount obstacles that Eden Mills residents must endure. There is a fight for power both figuratively and literally. They must get energy to survive and also have the physical, mental, and social power to live.In some ways, The Rule of Three takes an idealistic view of how a community could band together to survive after a disastrous event. I would hope that if this happened in any community that citizens would have the same attitude and skills. Following Eric Walter’s style, the characters are realistic, relatable, and believable. Like We All Fall Down and many of Walters’ other books, reluctant readers would find the book both readable and interesting. Readers looking for high vocabulary and intricate plot may find this read disappointing. The Rule of Three has suspense, drama, likable characters, and even a little romance; ages 13 and up would enjoy this novel. I am looking forward to the sequel The Rule of Three: The Fight for Power.Recommended: 3 out of 4 starsReviewer: Marylou DicksonTeaching Division III English Language Arts and impacting students’ abilities to improve their literacy practices are passions of Marylou Dickson.
