MADS-box is a key transcription factor regulating the transition to flowering and flower development. Lagerstroemia indica ‘Xiang Yun’ is a new cultivar of crape myrtle characterized by its non-fruiting nature. To study the molecular mechanism underlying the non-fruiting characteristics of ‘Xiang Yun’, 82 MADS-box genes were identified from the genome of L. indica. The physicochemical properties of these genes were examined using bioinformatics methods, and their expression as well as endogenous hormone levels at various stages of flower development were analyzed. The results showed that LiMADS genes were primarily classified into two types: type I and type II, with the majority being type II that contained an abundance of cis-acting elements in their promoters. By screening nine core proteins by predicted protein interactions and performing qRT-PCR analysis as well as in combination with transcriptome data, we found that the expression levels of most MADS genes involved in flower development were significantly lower in ‘Xiang Yun’ than in the wild type ‘Hong Ye’. Hormonal analysis indicated that ‘Xiang Yun’ had higher levels of iP, IPR, TZR, and zeatin during its early stages of flower development than ‘Hong Ye’, whereas the MeJA content was substantially lower at the late stage of flower development of ‘Hong Ye’. Finally, correlation analysis showed that JA, IAA, SA, and TZR were positively correlated with the expression levels of most type II genes. Based on these analyses, a working model for the non-fruiting ‘Xiang Yun’ was proposed. During the course of flower development, plant hormone response pathways may affect the expression of MADS genes, resulting in their low expression in flower development, which led to the abnormal development of the stamen and embryo sac and ultimately affected the fruiting process of ‘Xiang Yun’.

Abstract Chemoradiation (CRT) is widely used to treat locally advanced non-small cell lung cancer (NSCLC) patients, but only around 30% of patients achieve pathological complete response. Here, we utilized an in vitro high-content clonogenic survival screening method to identify CRT sensitizers from the NCI Cancer Therapy Evaluation Program (CTEP) portfolio. Talazoparib (PARPi) was identified among the top candidates to show potent CRT sensitization effects in both KRAS mutant and TP53 mutant NCSLC cell lines. We validated the CRT sensitization effect of PARPi on lung tumors in vivo with cell line xenografts mouse models. To further investigate alterations in the tumor microenvironment (TME) induced by PARPi, we utilized syngeneic mouse models and showed that the combination of radiotherapy, PARPi, and PD-L1 blockade achieved superior tumor control effects. By applying single cell RNA sequencing technologies to the mouse tumors, we found that PARPi combined with CRT or immunotherapy significantly reprogrammed the tumor cells and TME, resulting in the increase of tumor-infiltrating T cells which elicit potent tumor control effects. Our data showed that PARPi sensitizes NSCLC tumors to CRT and immunotherapy through a combination of tumor-intrinsic and extrinsic phenomenon. Abstract Hepatocellular carcinoma (HCC) remains one of the most challenging tumors to treat. Cancer stem-like cells (CSCs) are believed to be important in HCC development, immune evasion, and treatment resistance. Dysadherin is a cell membrane glycoprotein linked to cancer progression by endowing HCC with CSC features. However, the underlying mechanism of increasing CSC characteristics in refractory advanced HCC is not entirely understood. A comprehensive bioinformatics study revealed that dysadherin-expressing HCC cells were associated with cancer stemness and the YAP signaling activation. Dysadherin knockdown or overexpression in liver cancer cell lines attenuated or increased cancer stemness and aggressiveness via YAP signaling, respectively. Correspondingly, dysadherin deficiency lowered cancer stemness and aggressiveness in the HCC mouse model by blocking the YAP pathway. Mechanistically, dysadherin promoted YAP nuclear localization, which activated stem cell transcription factors such as OCT4, KLF4, and MYC. Of note, our comprehensive gain and loss of function studies in CSCs demonstrated that targeting the dysadherin/YAP axis could boost sorafenib’s therapeutic effect and suppress PD-L1 expression, which helps tumor cells escape from immune surveillance. Thus, our results highlight that the dysadherin/YAP axis drives the aggressive phenotype of HCC, providing a potential therapeutic strategy for HCC. Lithium-metal electrodes are of particular interest for next-generation rechargeable batteries because of their high specific capacity and low redox potential. Therefore, Li-metal-based batteries may afford higher energy densities than commercially available Li-ion batteries with graphite anodes. However, various bottlenecks have hampered the commercial development of these batteries, including uncontrolled Li dendrite formation and huge volume changes during cycling. Consequently, Li-metal batteries suffer from low Coulombic efficiency and poor cycling stability. In recent years, there has been extensive research on the design and construction of three-dimensional (3D) porous electrodes that can host metallic Li. However, the low pore utilization and uneven Li plating remain crucial issues. This can be understood in terms of electronic and ionic transport through the framework electrode. The carbon frameworks exhibit high electronic conductance; however, large resistance to Li+ migration in the electrolytes of internal and interparticle pores inhibits the penetration of Li+ deep into the electrode. In this work, we demonstrate that a strong interaction between Li and a lithiophilic nanolayer on a substrate plays a critical role in enhancing pore utilization in carbon framework electrodes. As a model architecture, we examine a Li storage process in a framework electrode consisting of porous carbon derived from metal-organic frameworks (MOFs) and a galvanically displaced Ag layer on a Cu substrate (Cu@Ag). The electrochemical experiments combined with operando XRD measurements and microstructural characterizations suggest that a lithiophilic Ag on the Cu substrate preferentially reacts with Li+ to form Li x Ag during the initial stage of Li plating. This Li x Ag phase acts as a seed that can regulate the subsequent Li plating, promoting confined Li storage in the carbon framework electrode while suppressing top plating. Because of these advantages, the MOF-C framework electrode on Cu@Ag exhibits better cycling stability (>250 cycles) than the MOF-C framework electrode on Cu (140cycles). However, when the thickness of the MOF-C framework is increased to 90 μm, the diffraction peak for Ag remains dominant throughout Li plating-stripping, and the formation of Li x Ag alloys is not clearly detectable in the diffraction patterns, suggesting that only a limited amount of Ag is involved in the alloying reaction with Li+. Based on the computational studies, the efficacy of lithiophilic layers toward improving pore utilization is discussed in terms of the kinetic competition between Li+ transport through porous channels and the interfacial reaction of Li+ with the substrate. This study conveys an important message that the Li-substrate interaction plays a vital role in promoting the confined Li storage; hence, it should be considered a key design factor for porous carbon frameworks with high capacity and long cycle lifetime. Abstract Background: Treatment (tx) for LA SCCHN includes a combination of surgery, radiation and/or chemotherapy followed by monitoring for local recurrence/distant metastases as standard of care. Given poor outcomes, there remains a clear unmet need in this pt population. IMvoke010 (NCT03452137) evaluated the efficacy and safety of atezo in pts with LA SCCHN who are at high-risk for disease progression following multi-modal definitive tx. Methods: Eligible pts with LA SCCHN (Stage IVa or IVb involving the oral cavity, larynx, hypopharynx, HPV negative oropharynx or Stage III HPV positive oropharynx [per AJCC 8th edition]) with no disease progression after multi-modal definitive tx were randomized (1:1) to receive atezo 1200 mg or placebo. Tx was given every 3 weeks for 1 year or until disease progression, unacceptable toxicity or consent withdrawal. Primary endpoint: investigator-assessed event-free survival (INV-EFS). Other key endpoints: overall survival (OS, secondary for efficacy) and safety. Results: A total of 406 pts were randomized to receive either atezo (n=203) or placebo (n=203). 156 (38.4%) pts underwent surgery as part of definitive tx. At clinical cutoff (27 September 2023), median follow-up was 46.5 months (mos). Median INV-EFS was 59.5 mos with atezo vs 52.7 mos with placebo (HR, 0.94; 95% CI, 0.70-1.26). INV-EFS results were generally consistent across all subgroups. OS showed no difference between arms for atezo vs placebo. Safety data are reported (Table). Conclusion: This study did not meet the primary endpoint of INV-EFS, a numerical improvement in INV-EFS was observed for atezo in pts with LA SCCHN, but this was not statistically significant. The genus Haplotropis Saussure, 1888 belongs to family Pamphagidae, superfamily Acridioidea. It contains five species: Haplotropis brunneriana Saussure, 1888; Haplotropis xiai Ye, Shi & Li 2016; Haplotropis zhuoluensis Ye, Shi & Li 2016; Haplotropis aqiensis Zhang, Lin et Yin, 2018 and Haplotropis shandongensis Zhang, Yin et Liu, 2019 distributed mainly in Amur River region of Asia and China only (Saussure, 1888; Bey-Bienko et Mishchenko, 1951; Otte, 1994; Yin, Shi et Yin, 1996; Storozhenko & Paik, 2011; Yin et al, 2014; Dong et al, 2015; Ye et al, 2016; Zhang et al, 2017; Zhang et al, 2018; Zhang et al, 2019; Cigliano et al, 2019). A new species Haplotropis heiheensis sp. nov. is described in this paper. The new species is similar to Haplotropis aqiensis Zhang, Lin et Yin, 2018, but differs from latter by Krauss’ organ of male without ramified keel; tegmine narrower, length 2.4 times width; cercus of male gently tapering at apical half part; minimum width 1.4 times length in mesosternum and epiphallus with 36 spikes. Type specimens are deposited in the College of Plant Protection, Shandong Agricultural University, Taian, Shandong, China.

The genus Haplotropis Saussure, 1888 belongs to family Pamphagidae, superfamily Acridioidea. It contains three species: Haplotropis brunneriana Saussure, 1888; Haplotropis xiai Ye, Shi & Li 2016 and Haplotropis zhuoluensis Ye, Shi & Li 2016 distributed mainly in Amur River region of Asia and China only (Saussure, 1888; Bey-Bienko et Mishchenko, 1951; Otte, 1994; Yin, Shi et Yin, 1996; Storozhenko & Paik, 2011; Yin et al, 2014, Dong et al, 2015; Ye et al, 2016; Zhang et al, 2017; Cigliano et al, 2018). A new species Haplotropis aqiensis sp. nov. is described in this paper. The new species is similar to H. xiai Ye et al., 2016, but differs from latter by tegmina of male extending over the hind margin of first abdominal tergite slightly, length of tegmina 1.7 times width, cover 2/3 tympanum; frontal ridge of male not widened at median ocellus; length interspace of mesosternum in male equal narrowest width; apical half part of cercus suddenly tapering in male and lower margin of epiphallus without projection in the middle. Objective: We aimed to show whether the antioxidant properties of pomegranate juice (PJ) show a protective effect against renal damage associated with oxidative stress caused by Extracorporeal Shock Wave Lithotripsy (ESWL) in patients who have undergone ESWL for kidney stones. Material and Methods: Ninety patients who were to undergo ESWL for kidney stones were included in the study and were divided into 3 groups with 30 patients each ESWL. Group 1 (control group) patients received only ESWL treatment. Group 2 (study group) patients were recommended to take 30 ml/kg/day pomegranate juice mixture orally for 2 days before ESWL. Group 3 (placebo group) patients were recommended to take 30 mg/kg/day water orally for 2 days before ESWL. All ESWL-treated patients received 2000 to 2500 shock waves at 18 to 24 in order to measure lipid peroxidation markers showing the DNA lesion associated with oxidative stress and to show tubular damage after ESWL, Results: There was a statistically significant mean increase of 52.39± 10.49 in 8-Hydroxy deoxyguanosine (8-OHdG) levels after treatment compared to pre-treatment levels in Group 1 (p<0.01). There was a statistically significant mean increase of 13.16±7.71 and 33.24±12.82 in Groups 2 and 3, respectively (p<0.01; p<0.01, respectively) There was a statistically significant mean increase of 19.20±5.21 in N-acetyl-beta-D-glucosaminidase (NAG) levels after treatment compared to pre-treatment levels in Group 1 (p<0.01). There was a statistically significant mean increase of 3.44±5.21 and 13.07±5.50 in Groups 2 and 3, respectively (p<0.01; p<0.01, respectively). Abstract Background: Colorectal cancer (CRC) remains one of the most commonly diagnosed cancers and a leading cause of cancer-related deaths worldwide. Between 3 and 6% of all CRC cases are attributed to hereditary colorectal cancer (HCRC). It is important to distinguish CRCs that arise hereditarily from those that arise sporadically, because patients with HCRC and their relatives can benefit from intensive clinical management and surveillance. The detection of germline variants using tumor tissue and paired non-cancerous tissue/whole blood cells (WBC) (conventional method) is considered as the gold standard for germline confirmation, but which could introduce high cost and long turnaround time. In this work, we aimed to investigate the feasibility of diagnosing HCRC using only tumor next-generation sequencing data without normal tissue/WBC data. Methods: One-hundred patients with suspected HCRC were prospectively enrolled according to the clinical diagnosis. Both tumor tissue and paired WBC were collected from each patient. Capture-based targeted sequencing using a panel consisting of 41 cancer-related genes was performed on both tumor tissue and WBC sample. The germline variants were identified using conventional and ColonCore method, respectively. The sensitivity and positive predictive value (PPV) of ColonCore method in identifying germline alterations were calculated, when conventional method was used as a reference. Results: A total of 636 alterations were identified using ColonCore method, including 606 somatic alterations and 30 germline alterations. Among the somatic alterations detected from this panel, 599 single-nucleotide variants and 7 copy number variants were observed. The most frequently mutated genes were APC, TP53, and KRAS, occurring in 76%, 66%, and 46% of patients, respectively. For ColonCore analysis, 17, 9, 2, and 2 patients harbored germline pathogenic variants in DNA mismatch repair (MMR) genes, APC, MUTYH, and ATM, respectively. Compared to the conventional method, the sensitivity of ColonCore method in the diagnosis of HCRC achieved 88.5% with a PPV of 82.1%. Next, the diagnostic performance of ColonCore method in Lynch syndrome (LS) and familial adenomatous polyposis (FAP) was subsequently investigated. We found its sensitivity in the diagnosis of LS achieved 100% (with a PPV of 98.8%), which was obviously higher than that in the diagnosis of FAP (70.0% with a PPV of 63.6%). Twenty-three patients identified as having a microsatellite instability (MSI)-high tumor using ColonCore method were also identified as having an MMR-deficient (dMMR) tumor using immunohistochemistry staining analysis. Conclusions: Our study suggests that ColorCore method is a one-stop approach for detecting germline/somatic alterations and MSI/dMMR status for suspected HCRC. Abstract Backgrounds: Cancer is the most common cause of mortality in Korea. According to International Agency for Research on Cancer (IARC), tobacco smoking is the Group I carcinogen, and the National Cancer Center Korea reported in 2009 that 20.9% and 2.3% of total cancer incidence were attributable to tobacco smoking in men and women, respectively. As of 2020, we estimated population attributable fraction (PAF) of cancer incidence due to tobacco smoking. Methods: We organized the Consensus Committee to develop strategy and methodology for PAF estimation. From the consensus, cancer sites for PAF estimation were selected based on the IARC list of classification with sufficient evidence in human. PAF was calculated by using Levin’s formula which includes exposure prevalence and relative risk (RR). Exposure prevalence was collected from the Korea National Health and Nutrition Examination Survey (KNHANES) in 2005, assuming that latent period from risk exposure to cancer development was 15 years. Tobacco smoking was defined as a categorical variable including non-smokers, past smokers, and current smokers. Pooled RRs were estimated from meta-analysis of RRs obtained from Korean Genome and Epidemiology Study, National Health Examination data provided by National Health Insurance Services, Korean Cancer Prevention Study-II, and KNHANES, each of which was linked with Korea Cancer Registry. Cox proportional hazards model were used to estimate RRs, while adjusted by age groups and alcohol consumption. Results: For men, 17.4% of all cancer cases were attributed to smoking, compared to 1.7% for women. Specifically, in men, 57.9% of lung cancer, 36.4% of bladder cancer, 31.9% of esophageal cancer, 39.1% of laryngeal cancer, 25.4% of oral cavity/pharyngeal cancer, 19.7% of liver cancer, and 19.5% of stomach cancer were attributed to tobacco smoking. In women, 51.7% of laryngeal cancer, 15.0% of esophageal cancer, 11.6% of lung cancer, 6.0% of bladder cancer, 5.1% of oral cavity/pharyngeal cancer, 5.0% of cervix cancer, and 4.6% of liver cancer were due to smoking. Conclusion: Tobacco smoking is an important cause of esophageal, laryngeal, and lung cancer in men, and esophageal and laryngeal cancer in women, and comprises a substantial proportion of PAF in cancer incidence. Thus, intervention measures are needed to reduce smoking prevalence for cancer prevention in Korea. Alcohol consumption was defined as non-drinking (&lt;0.1g/day), light (0.1~30g/day), moderate (30~60g/day), and heavy (60~588g/day) drinking according to the definition by World Health Organization. We estimated RRs from the cohort data established by Korean Genome and Epidemiology Study, National Health Examination data provided by National Health Insurance Services, Korean Cancer Prevention Study-II, and KNHANES, while each cohort was linked with Korea Cancer Registry. RRs were estimated by using Cox proportional hazards model adjusted by age group and smoking status. RRs were then meta-analyzed to yield pooled RR by using random-effects model. Results: A total of 4.1% and 1.2% of all cancer incidence were attributable to alcohol consumption in men and women. By cancer sites, 67.1% of esophageal cancer, 23.8% of oral cavity/pharyngeal cancer, 15.1% of laryngeal cancer, 12.0% of colorectal cancer, and 7.9% of liver cancer were attributable to alcohol consumption in men. In women, 54.1% of liver cancer, 10.3% of colorectal cancer, 7.8% of oral cavity/pharyngeal cancer, and 3.6% of breast cancer were due to alcohol consumption. Conclusion: PAF of cancer incidence due to alcohol consumption has increased in 2020 compared to PAF in 2009. As alcohol consumption is an important cause of oral cavity/pharyngeal, esophageal, and laryngeal cancer in men, and liver and breast cancer in women, prevention measures are required to reduce alcohol consumption for cancer prevention in Korea. Ye Yen Sun Cenozoic granitoid formations include biotite granite, fine- to medium-grained amphibol-biotite granite, and mostly undeformed fine- grained granite porphyry. Temperature calculation by the zircon saturation method gives the formation temperature ranging from about 680°C to 850°C; the pressure, determined on the basis of the major element composition, varies between 10 and 1 kbar (about 33 to 3.3 km deep). The granites viewed as "hot" or "intermediate" magmas were formed at the site of extension or transformation tectonics. Thus, the ability to form these granites due to the melting of the lower crust under the stretching region along the two wings of the Red River shear zone is a mechanism to be considered.

The present study was undertaken to determine the effect of Yi-Qi-Yang-Yin-Ye (Y-Q-Y-Y-Y), a compound of Traditional Chinese Herbal Medicine, on insulin resistance (IR) in the diet-induced obese rat model induced by intravenous injection with a low dose of streptozotocin and fed a high fat and high caloric diet. Y-Q-Y-Y-Y (2, 4, 8 g/kg) was administered via gavage daily for 4 weeks. The results showed that Y-Q-Y-Y-Y treatment decreased the levels of body weight, total cholesterol (TC), triglycerides (TG), low density lipoprotein-cholesterol (LDL-C), free fatty acid (FFA), insulin (INS) and fast blood glucose (FBG) and increased the level of high density lipoprotein-cholesterol (HDL-C) in the diet-induced obese rats. Glucose tolerance was improved in the diet-induced obese rats treated with Y-Q-Y-Y-Y as well as GIR (glucose infusion rate) in the hyperinsulinemic euglycemic clamp experiment compared to the model control rats (p < 0.01). Moreover, treatment with Y-Q-Y-Y-Y up-regulated glycogen contents in both liver and skeletal muscle and increased insulin receptor amounts on the erythrocytes surface as assessed by using 125I-labeled auto-antibodies against insulin receptors. Taken together, our data suggested that Yi-Qi-Yang-Yin-Ye ameliorates insulin resistance in the diet-induced obese rats.

In recent years, the industrial requirements for rechargeable batteries in terms of energy density have continuously increased owing to the widespread adoption of portable electronic devices and electric vehicles. Li metal has been identified as a promising anode material to meet the ever-increasing demand for rechargeable batteries with high energy densities because of its low redox potential (−3.04 V vs. standard hydrogen electrode)and high specific capacity (3860 mAh g−1) [1]. Despite these advantages, the commercial use of Li metal anodes has been hindered by unresolved critical issues, such as the uncontrollable growth of Li dendrites and the severe volume changes during cycling. In practice, the dendritic growth of Li deposits causes the formation of dead Li species and triggers internal short-circuiting of batteries during cycling, leading to a significant loss of reversibility and raising safety concerns. Moreover, large volume changes in the Li metal anode during Li plating–stripping, which originate from the “hostless” character of the Li metal, impose severe stresses on the solid–electrolyte interphase (SEI) layers, and thus reducing the mechanical stability of SEIs. Once the SEI layers are cracked or fractured, bare Li metal is exposed to the electrolyte, leading to the continuous formation of thick SEI layers on the anode surface. This is mainly responsible for the continuous, irreversible loss of active Li, i.e., low Coulombic efficiency and short cycle lifetimes. 3D “host” architectures are beneficial for securing the dimensional stability of Li metal anodes by utilizing internal pores (i.e., free-spaces) as reservoirs for Li storage. Moreover, they provide large active surface areas and hence reduce the effective current densities, suppressing uneven Li plating (dendritic growth) during high-rate operations. Metal-organic framework (MOF)-derived carbon offers many advantages over conventional carbon materials in terms of specific surface area, pore volume, and structural tunability. Furthermore, the critical role of atomically dispersed heteroatoms has been verified in reducing the energy barriers for the nucleation and growth of Li [2]. Herein, we report MOF-derived porous carbon hosts with strong Li–host interactions achieved by galvanically displaced lithiophilic seeds. Owing to the distinctive chemical and structural nature of zeolitic imidazolate frameworks (ZIF-8), atomically dispersed Zn can be spontaneously replaced by Ag with a strong lithiophilicity via a galvanic displacement (GD) reaction. Based on a comparative study of carbon hosts with different sizes, morphologies, and distributions of Ag, in particular, we assess the efficacy of lithiophilic Ag in promoting reversible Li storage inside porous carbon hosts. The strong Li–host interactions enabled by lithiophilic Ag effectively reduce the overpotentials for nucleation and growth of Li, and more importantly, the role of lithiophilic Ag in governing the reversibility of Li plating–stripping is discussed in terms of the spatial distribution characteristics of Ag. The atomically dispersed Ag triggers the outward growth of Li from the internal pores of the carbon host, showing a stable cycling performance (>745 cycles). Contrarily, the surface-anchored Ag nanoparticles induce uneven Li plating on the outer surface of the carbon host, resulting in a rapid performance drop (~305 cycles) during cycling. Abstract Growth factor receptor-bound protein 2 (GRB2) is a cytoplasmic adapter for tyrosine kinase signaling and a nuclear adaptor for homology-directed-DNA repair. Here we find nuclear GRB2 protects DNA at stalled replication forks from MRE11-mediated degradation in the BRCA2 replication fork protection axis. Mechanistically, GRB2 binds and inhibits RAD51 ATPase activity to stabilize RAD51 on stalled replication forks. In GRB2-depleted cells, PARP inhibitor (PARPi) treatment releases DNA fragments from stalled forks into the cytoplasm that activate the cGAS-STING pathway to trigger pro-inflammatory cytokine production. Moreover in a syngeneic mouse metastatic ovarian cancer model, GRB2 depletion in the context of PARPi treatment reduced tumor burden and enabled high survival consistent with immune suppression of cancer growth. Collective findings unveil GRB2 function and mechanism for fork protection in the BRCA2-RAD51-MRE11 axis and suggest GRB2 as a potential therapeutic target and an enabling predictive biomarker for patient selection for PARPi and immunotherapy combination. In the paper we study relations of rigidity, equicontinuity and pointwise recurrence between an invertible topological dynamical system (t.d.s.) $(X,T)$ and the t.d.s. $(K(X),T_{K})$ induced on the hyperspace $K(X)$ of all compact subsets of $X$, and provide some characterizations. Among other examples, we construct a minimal, non-equicontinuous, distal and uniformly rigid t.d.s. and a weakly mixing t.d.s. which induces dense periodic points on the hyperspace $K(X)$ but itself does not have dense distal points, solving in that way a few open questions from earlier articles by Dong, and Li, Yan and Ye.

Abstract The use of mitochondrial inhibitors to target oxidative phosphorylation (OXPHOS) in cancer treatment presents a challenge due to dose-limiting toxicities. Moreover, while glycolysis-deficient cancers are vulnerable to OXPHOS inhibition in preclinical models, the full extent of phenotypical and mechanistic consequences of inhibiting OXPHOS in cancers capable of glycolysis is not yet well understood. Our results presented here offer promising insights into potential therapeutic gains from combining p53 restoration strategies with OXPHOS inhibitors, even when applied to glycolysis-competent CRC cells. Treatment with mitochondrial complex I inhibitors did not cause energy stress in CRC cells capable of glycolysis. It does, however, induce DNA replication stress, apparent through an observed cell cycle arrest at the S phase, enrichment of the G2/M DNA-damage checkpoint regulation pathway, and replication fork slowdown. Intriguingly, CRC cells harboring wildtype p53 exhibited more severe replication stress than those carrying mutant p53. Furthermore, siRNA knockdown of p53 attenuates replication stress and reduces cell cycle arrest, underlining the important role of p53 in CRC cell responses to OXPHOS inhibition. Our targeted metabolomics analysis reveals that OXPHOS inhibition results in reductions in the purine nucleotides, adenine monophosphate (AMP) and guanine monophosphate (GMP), as well as the pyrimidine nucleotide, uridine monophosphate (UMP), in CRC cells, regardless of their p53 mutational status. By supplementing cell culture mediums with the purine nucleobases adenine and guanine, and the pyrimidine nucleoside uridine, we observed a partial reversal of the replication fork slowdown and reductions in cell viability. This suggests nucleotide deficiencies are involved in the induction of DNA replication stress caused by OXPHOS inhibition. The nucleotide deficiencies were associated with a decrease in the nucleobase precursor aspartate. By adding aspartate at a supraphysiological concentration, to overcome the low expression of the excitatory amino acid transporter 1 required for cellular import of aspartate, we were able to restore the levels of nucleotides. Collectively, our findings suggest broader potential cancer treatment paradigms via OXPHOS targeting, extending beyond glycolysis-deficient cancers. Our data uncovers that CRC cells, which commonly exhibit the glycolytic phenotype, are susceptible to OXPHOS inhibition, with those carrying wildtype p53 showing heightened sensitivity. Therefore, p53 status could serve as a biomarker for predicting CRC responses to OXPHOS inhibitors. Moreover, our findings suggest that combined strategies of restoring p53 function, using small molecules such as APR-246, might enable reduced dosage of OXPHOS inhibitors in CRC treatment, thereby mitigating their dose-limiting toxicities. Leptobrachella (Anura: Megophryidae) is currently composed of seventy-one species distributed in China and Southeast Asia (Frost 2018). Fourteen species of Leptobrachella are currently known from southern China: L. alpina (Fei, Ye & Li, 1990), L. laui (Sung, Yang & Wang, 2014), L. liui (Fei & Ye, 1990), L. mangshanensis (Hou, Zhang, Hu, Li, Shi, Chen, Mo & Wang, 2018), L. maoershanensis (Yuan, Sun, Chen, Rowley & Che, 2017), L. oshanensis (Liu, 1950), L. purpura (Yang, Zeng & Wang, 2018), L. tengchongensis (Yang, Wang, Chen & Rao, 2016), L. wuhuangmontis (Wang, Yang & Wang, 2018), L. yingjiangensis (Yang, Zeng & Wang, 2018), L. yunkaiensis (Wang, Li, Lyu & Wang, 2018), L. ventripunctata (Fei, Ye & Li, 1990), L. pelodytoides (Boulenger, 1893) and L. sungi (Lathrop, Murphy, Orlov & Ho, 1998) and the first eleven species are endemic of this region (AmphibiaChina 2018). Many new species in this genus were discovered based on molecular, morphometric and bioacoustic data in recent years (Frost 2018). In comparison to molecular and morphological data, bioacoustics is especially useful in species identification of Leptobrachella in field surveys (Rowley et al. 2016). Although the advertisement calls of many Leptobrachella species were described along with the species descriptions (e.g., Rowley et al. 2016), the advertisement calls of only three Chinese endemic species (L. alpinus; L. laui and L. oshanensis) are known (Jiang et al. 2002; Matsui 2006; Xu et al. 2005). Here, we describe the advertisement calls of L. liui, which inhabits rocky streams in southeastern China (Fujian, Jiangxi and Zhejinag) and is morphologically diagnosed by the combination of a set of external color traits (Fig. 1).

Here we show that this uniform bound holds for a Zariski open and dense set in the space of all pairs, $\mathrm {Rat}_d \times \mathrm {Rat}_d$ , for each degree $d\geq 2$ . The proof involves a combination of arithmetic intersection theory and complex-dynamical results, especially as developed recently by Gauthier and Vigny, Yuan and Zhang, and Mavraki and Schmidt. In addition, we present alternate proofs of the main results of DeMarco, Krieger, and Ye [Uniform Manin-Mumford for a family of genus 2 curves, Ann. of Math. (2) 191 (2020), 949–1001; Common preperiodic points for quadratic polynomials, J. Mod. Abstract Modern Chinese confirmative shi (as in mei cuo, wo shi yao dusi ni (沒錯,我是要毒死你) ‘that’s right. I really wanted to poison you to death’) is not an auxiliary but an adverb. It derives from the adjective shi ‘true, real’ in Old Chinese (Yan zhi yan shi ye (偃之言是也) ‘what Yan said was true’). The grammaticalization pathway of the Modern Chinese confirmative shi is different from that of the copula shi (Laozhang shi huoche siji (老張是貨車司機) ‘Laozhang is a truck driver’) or the auxiliary shi (Laozhang shi kai huoche, wo shi kai keche (老張是開貨車,我是開客車) ‘Laozhang drives a truck and I drive a coach car’). Modern Chinese confirmative shi, copula shi, and auxiliary shi have the same morphological form because they all appear to derive from the adjective shi or demonstrative ( shi ke ren, shu bu ke ren ( 是可忍,孰不可忍) ‘if this could be endured, is there anything else that could not be endured’) in Old Chinese. Such a pattern of morphological sameness seems to be cross-linguistically rare, if not unique.

Abstract Background: There is an increasing demand for effective combinatory agents to improve upon PD-1/PD-L1-based therapeutics. One combinatory target axis is the adenosine metabolism pathway that consists of three major players, including CD39, CD73 and A2AR. Inhibition of any of these targets have shown enhanced preclinical efficacy in combination with PD-1/PD-L1 inhibitors. CD39 is an ectonucleotidase which degrades extracellular ATP to adenosine monophosphate (AMP). This is considered a rate-limiting step for the further degradation to adenosine by CD73. Adenosine is an immunosuppressive metabolite that can suppress NK and T cells. Blockade of CD39-mediated degradation of ATP to AMP may therefore recover anti-tumor immunity through preventing the enrichment of adenosine in the tumor microenvironment. Method: Two anti-CD39 VHH molecules were generated, named Ye-37 and Ye-46, which bind to two different epitopes on CD39. Binding experiments were carried out by bio-layer interferometry. Cell binding experiments were tested on CD39-overexpression cell lines by flow cytometry. Cellular CD39 enzymatic inhibition experiments were tested using an MOLP-8 cell line and PBMC via a luminescence-based assay. Soluble CD39 enzymatic tests were carried out on recombinant CD39 protein using a similar method. T cell proliferation assays were performed and observed on CD4+ or CD8+ T cell populations. In vivo efficacy studies were tested in B-NDG B2M-KO mice that were injected subcutaneously with A375-CD39+ tumor cells and human PBMC. An anti-PD1 x CD39 bispecific antibody was then generated by fusing the anti-CD39 biparatopic antibody to the C-terminus of an anti-PD1 IgG and tested using similar methods. Results: Two candidates, Ye-37 and Ye-46, were selected for their functional activity that recognize non-overlapping epitopes on CD39. The combination of Ye-37 and Ye-46 shows high potency in cell-based and soluble CD39 assays in blocking CD39 activity. Two biparatopic molecules were generated by fusing the biparatopic unit to the N- or C-termini of Fc (46-37-Fc and Fc-37-46) and showed similar activity to the combination. In vivo, we showed single-agent control of tumor growth and potentiation of tumor-growth inhibition when combined with anti-CD73 antibodies. An anti-PD1 x CD39 bispecific was generated and showed potent inhibition of PD-1/PD-L1 interactions by cell-based assays. Potent anti-tumor efficacy was shown, which was as effective as the combination of anti-PD1 plus anti-CD39 antibodies. Conclusion: Potent anti-CD39 and anti-PD1 x CD39 therapeutic candidates have been generated with promising activity as a combinatory or single agent, respectively. As such, we plan to file for clinical trial authorization of these programs by 2022. The analysis of the genetic diversity and historical dynamics of endemic endangered goose breeds structure has attracted great interest. Although various aspects of the goose breed structure have been elucidated, there is still insufficient research on the genetic basis of endemic endangered Chinese goose breeds. In this study, we collected blood samples from Lingxiang White (LX), Yan (YE), Yangjiang (YJ), Wuzong (WZ), Xupu (XP), and Baizi (BZ) geese (Anser cygnoides) and used Sanger sequencing to determine the partial sequence of the cytochrome b (CYTB) gene in a total of 180 geese. A total of 117 polymorphic sites were detected in the 707 bp sequence of the mtDNA CYTB gene after shearing and correction, accounting for approximately 16.55% of the entire sequence. The AT content (51.03%) of the processed sequence was slightly higher than the GC content (48.97%), indicating a preference for purine bases. The YJ, YE, and WZ breeds had the highest population genetic diversity, with a haplotype diversity greater than 0.9 (Hd > 0.9) and average population nucleotide difference of 8.01 (K > 8.01). A total of 81 haplotypes were detected and divided into six major branches. Among the six goose breeds, there were frequent genetic exchanges among LX, YJ, YE, and WZ geese (Nm > 15.00). We analyzed the distribution of base-mismatch differences in goose breeds and tested their historical dynamics for neutrality in Tajima’s D and Fu’s Fs. For YJ and WZ geese, Tajima’s D > 0, but the difference was not significant (p > 0.05). The actual values for the two breeds exhibited multimodal Poisson distributions. The population patterns of the WZ and YJ geese are purportedly relatively stable, and the breeds have not experienced population expansions or bottleneck effects, which is consistent with the neutrality test results. This study provides new insights into the diverse genetic origins and historical dynamics that sustain endemic endangered goose breeds.

Bu çalışma, primer dismenorede telerehabilitasyon programının menstrual semptomlar üzerine etkisini incelemek amacıyla yapıldı. Yöntem: Çalışmaya primer dismenoresi olan 60 kadın dahil edildi. Bireyler müdahale grubu (n=30) ve kontrol grubu (n=30) olmak üzere iki gruba randomize edildi. Müdahale grubuna 8 hafta boyunca farklı egzersizleri içeren telerehabilitasyon programı uygulandı. Kontrol grubu ise analjezik ilaç kullandı ve lokal sıcak uyguladı. Bireylerin sosyodemografik bilgileri ve menstrüel özellikleri kaydedildi. Çalışma öncesi ve sonrasında menstrüasyon semptomları Menstrüasyon Semptom Ölçeği (MSÖ) ile değerlendirildi. Bulgular: Müdahale ve kontrol gruplarının MSÖ alt boyutları ve toplam puanı tedavi öncesi ve sonrası grup içerisinde karşılaştırıldığında her iki grupta da anlamlı fark bulundu (p&lt;0,05). MSÖ’ ye ait negatif etkiler/ somatik yakınmalar, ağrı belirtileri alt boyutları ve toplam puanında tedavi öncesi ve sonrasında müdahale ve kontrol grupları arasında anlamlı bir fark bulundu (p &lt;0,05). Ancak MSÖ’ ye ilişkin baş etme yöntemleri alt boyutunda tedavi öncesinde ve sonrasında gruplar arasında anlamlı bir fark bulunamadı (p&gt;0,05). Sonuç: Telerehabilitasyon programının primer dismenorede menstrual semptomlar üzerinde iyileştirici etkileri olduğu saptanmıştır. Telerehabilitasyon programı yan etkisi olmayan, uygulaması kolay, zaman tasarrufu sağlayan ve ucuz bir tedavi yöntemidir; menstrual semptomları azaltmak için güvenli bir şekilde uygulanabilir.

After reviewing the trace metals (Sn, W, Mo)-bearing granitoid deposits in the world, theauthors recognized that some deposits in Sweden (e.g. Nozzboten, Vasterbotten, Hotagen, Bergslagen, Bohus, Gosthenburg, Gotemar, Blekinge) contain not only trace metals (Sn, W, Mo), but also F, Li, U,Th, etc… which are similar to the Phia Oac granitoids. The correlation chart with the T. Carlsson (1982)’s criteria helps to determine the uranium-bearing granitoids. Initially, we constructed some correlation charts of granitoid fields in Northern Vietnam and concluded that some granitoid fields certainly related to uranium are Phia Oac, Xom Giau, Phu Sa Phin, and some granitoid fields possibly related to uranium are Ye Yen Sun, Muong Lat, Ban Chieng, and Song Chay (based on data from the Geology of Vietnam, Part II, Magma Section). This issue needs to be further studied.

Abstract Background: MET gene amplification is associated with poor prognosis in gastric cancer (GC) and gastroesophageal junction adenocarcinomas (GEJ). Savolitinib is a potent and highly selective oral MET tyrosine-kinase inhibitor. Here we reported the preliminary efficacy and safety from a phase 2 trial of savolitinib monotherapy in patients (pts) with MET-amplified advanced or metastatic GC/GEJ. (NCT04923932). Methods: Eligible pts had 2L+ GEJ or GC, with MET amplification and measurable lesions. Pts received savolitinib at 600 mg QD for body weight (BW) ≥50 kg, while 400 mg QD for BW &lt;50 kg in 21-day cycles until disease progression or meeting other criteria for end of treatment. Savolitinib BID regimen has also been additionally explored. The primary endpoint was objective overall response rate (ORR) evaluated by Independent Review Committee (IRC). One interim analysis (IA) was pre-defined at the first 20 QD pts who had at least 2 tumor assessments. Results: As of IA, 20 pts were enrolled for QD regimen. Demographics and clinical outcomes are shown in table 1. The mean relative dose intensity of 93.07%. Median duration of exposure was 2.09 months. Confirmed ORR by IRC was 45%, and reached 50% in 16 patients with MET GCN (high) while only 1 PR was observed in 4 patients with MET GCN (low). Duration of response rate at 4-month was 85.7% with median follow up time of 5.5 months. The most common Gr≥3 TRAE (≥5%) were platelet count decreased, hypersensitivity, anemia, neutropenia and hepatic function abnormal. In all pts, only 1 patient discontinued treatment due to grade 4 liver function abnormal (TRAE) and no patient died due to TRAE. Conclusion: Savolitinib monotherapy had manageable safety and showed promising efficacy in pts with MET-amplified GEJ or GC, particularly in pts with MET high GCN. BID regimen is being investigated to further evaluate the efficacy and safety of savolitinib in pts with MET high GCN. Table 1. Alkaline granites of the Muong Hum are distributed mainly in the NW Phan Si Pan zone. The granite closely has striped or clear gneissoid structures, coinciding with general NW-SE trends. It consists mainly of plagioclase (~20–30 %), alkaline feldspar (~30–50 %), quartz (~20–25 %), biotite (~1–5 %), aegirine (~1–3 %), and riebeckite (~1–2 %). It has 10,000×Ga/Al ratios of 4.70–4.93, A/CNK values of 0.87–0.90, and negative Eu-anomalies as well as apparent depletion of Ba, Sr, Ti, and P. The mineral assemblages and chemical characteristics show that it is typical of A-type granites. Compared with other adjacent Late Permian to Early Triassic A-type granitic plutons, geochemical characteristics of the Muong Hum granite are similar to the Phu Sa Phìn, Phan Si Pan, Ye Yen Sun, and Nam Xe-Tam Duong granites in NW Vietnam as well as the Taihe, and Panzhihua granites in SW China. Thus, the Phan Si Pan zone must have been a displaced portion of the Emeishan large igneous province. This might be a direct result of the left-lateral Cenozoic Red River shear zone.

Abstract Background: SHR-A1811 is an ADC comprised of a humanized anti-HER2 monoclonal antibody (trastuzumab), a cleavable linker, and a DNA topoisomerase I inhibitor payload. Here we assessed SHR-A1811 in HER2-expressing/mutated unresectable, advanced, or metastatic solid tumors. Methods: Pts were eligible if they had HER2 positive breast cancer (BC), HER2 positive gastric/GEJ carcinoma, HER2 low-expressing BC, HER2-expressing/mutated NSCLC, or other HER2-expressing/mutated solid tumors, and were refractory or intolerant to standard therapy. SHR-A1811 at doses of 1.0-8.0 mg/kg was given Q3W (IV). The primary endpoints were DLT, safety, and the RP2D. Results: From Sep 7, 2020 to Sep 28, 2022, 250 pts who had undergone a median of 3 prior treatment lines in the metastatic setting received at least one dose of SHR-A1811 in dose escalation, PK expansion, and indication expansion part. As of data cutoff on Sep 28, 2022, 1 pt experienced DLT. Treatment-related adverse events (TRAEs) were reported in 243 (97.2%) pts. Grade ≥3 TRAEs, serious TRAEs, and treatment-related deaths were reported in 131 (52.4%), 31 (12.4%), and 3 (1.2%) pts, respectively. Interstitial lung disease (AESI) was reported in 8 (3.2%) pts. Exposures of SHR-A1811, total antibody, and the payload were generally proportional to dose from 3.2 to 8.0 mg/kg. ORR was 61.6% (154/250, 95% CI 55.3-67.7) in all pts. Objective responses were observed in pts with HER2 positive BC (88/108, ORR 81.5%, 95% CI 72.9-88.3), HER2-low BC (43/77, ORR 55.8%, 95% CI 44.1-67.2), urothelial carcinoma (7/11), colorectal cancer (3/10), gastric/GEJ carcinoma (5/9), biliary tract cancer (5/8), NSCLC (1/3), endometrial cancer (1/2), and H&N cancer (1/1). Subgroup analyses of ORR are shown in Table 1. The 6-month PFS rate was 73.9% in all pts. This essay is focused on the necessity of redefining the concepts of human beings to coexist in harmony with life-community in the Post-Corona age. The point is to explicate how the concepts of human beings in the Book of Changes provide implications for posthumanism from the viewpoint of beyond-borders through the Change-Penetrating view. Its mechanism connotatively implies the aspect of the reality of balance and changes in the world in the Book of Changes. In the Change-Penetrating view, the category of Yin and Yang, revealing the way of trans-boundaries through robustness and stillness, can verify new concepts of human beings. It has an axiological meaning on the direction in which human beings have to take in this fast changing world. This is reflected in the humanity of the sage, the best human image that appears in the protagonist, and the practicality of Jin-de-xiu-ye (廣德崇業). The attitude of consistent improvement of human beings and the axiology of beyond-borders in the book of change exhibit the new type of integrated viewpoints on the subject about the sustainable coexistence with non-human objects. It is what post-humanism pursues through the process of trans-boundaries.

Amaç: Bu çalışmada kliniğimizde toraks cerrahisi uygulanan hastalarda postoperatif analjezi amaçlı intratekal morfin kullanımı ile ilgili deneyimlerimizi sunmayı amaçladık. Gereç ve Yöntem: Bu çalışmada Ocak 2020-Haziran 2020 tarihleri arasında toraks cerrahisi geçiren ve postoperatif analjezi amaçlı intratekal morfin uygulanan toplam 18 erişkin hastanın dosyası ve anestezi formları retrospektif olarak incelendi. Grup 1;10 mcg/kg intratekal morfin, Grup 2; 7 mcg/kg intratekal morfin uygulanan grup olarak ikiye ayrıldı. Hastaların demografik verileri, uygulanan morfin dozları, anestezi ve cerrahi süreleri, intraoperatif anestezik ajan tüketimleri, postoperatif hemodinamik verileri, ağrı düzeyleri, ek analjezik ihtiyaçları ve yan etkiler incelendi. Bulgular: Hastaların 12’sine (% 66,7) ideal vücut ağırlığına göre 10 mcg/kg, 6’sına (%33,3) 7 mcg/kg intratekal morfin uygulandığı tespit edildi. İntraoperatif anestezik ajan tüketimi Grup 1’de Grup 2’ye kıyasla daha düşük bulundu, ancak aradaki fark istatistiksel olarak anlamlı değildi. Postoperatif 12, 24 ve 48. saatlerde dinlenme sırasında bakılan ağrı düzeyleri Grup 1’de istatistiksel olarak daha düşük. Postoperatif 12, 18, 24 ve 48. saatlerde efor sırasında bakılan ağrı düzeyleri de Grup 1’de istatistiksel olarak daha düşük bulundu. Sonuç: Toraks cerrahisi hastalarında uygulanan intratekal morfin güvenli bir postoperatif analjezi seçeneğidir. Bu amaçla 10 mcg/kg dozunda uygulanan intratekal morfinin 7 mcg/kg’e göre daha etkin analjezi sağladığı kanaatindeyiz.

Abstract HSP90 mediates the folding of many important cancer-associated proteins, but it can also direct its substrates towards degradation via the ubiquitin-proteasome system. Furthermore, in tumor tissues, HSP90 complexes are in an activated state relative to normal tissues, and small molecule HSP90 inhibitors display unique tumor-selective pharmacokinetics. To take advantage of these attributes, we have developed a novel targeted protein degradation technology, termed Chaperone-Mediated Protein Degradation (CHAMP), and present here an in-depth characterization of the CHAMP mechanism of action. Initially, from a chemical library of greater than 1000 linkered HSP90 binders, hetero-bifunctional CHAMPs were synthesized in which target protein binders and HSP90 binders were covalently coupled together by short linkers. The resulting compounds were screened for target protein degradation and cancer cell cytotoxicity to identify promising leads for further optimization. We found that CHAMPs can degrade a wide variety of target proteins. This included proteins that are known to be regulated by HSP90, such as transcription factor BRD4 or ERK5 kinase. However, proteins that are normally independent of HSP90 function can also be degraded, including mutated KRAS and SHP2 phosphatase. In vitro, CHAMP treatment of cells resulted in formation of a ternary complex between the target protein, CHAMP compound and HSP90. Moreover, an X-ray crystal structure was solved for a mKRAS-CHAMP-HSP90 ternary complex. CHAMP-mediated degradation required both the target- and HSP90-binding moieties to be covalently coupled and involved ubiquitination of the target protein. Multiple ubiquitin E3 ligases were present in ternary complexes, and depending on the target protein, NEDD8 inhibition or CRISPR knockouts of individual E3 ligases could suppress proteasome-dependent target degradation. In vivo, irrespective of target, CHAMPs displayed prolonged exposure in tumors relative to plasma and normal tissues, resulting in prolonged target degradation in tumors and strong tumor growth inhibition at well-tolerated doses. CHAMP technology can be applied to a diversity of cancer-associated targets and has potential advantages relative to other protein degradation approaches, including an improved safety margin due to preferential accumulation in tumor tissues. They reported rates of pharmacologic treatment failure as 50% one year after psychiatric admission and 75% after two years. Between many factors influencing, one is the relationship between the professional and the family. There are different models of family intervention that have been effective in improving relapse rate, symptoms, social and occupational functioning, adherence and knowledge of illness and family burden (Zhang, Yan, Yao, & Ye et al., 1993).ObjectivesTo assess the association and strength between the performance of family psycho education groups and adherence to treatment in a schizophrenic population.Material and methodsObservational non blinded descriptive study in people with schizophrenia treated in an outpatient service which has 75000 inhabitants of reference population. We divided the patients into two groups whether family psycho education was done or not in the past 5 years. In each group we randomly sampled and reached an n > 30 and then we practiced a 2-year prospective follow-up recording the following variables: diagnosis, disease evolution, comorbidity with affective disorder, GAF and CGI, type of drug treatment both at the beginning and end of follow up, number of therapeutic changes, compliance visits, number of hospital admissions, number of emergency visits and side effects tolerance. After the follow-up we will do a descriptive statistics (Student t, X2, ANOVA) through SPSS version 11.5.Results and conclusionsWill be sent at the end of the study.

Background: To explore the prescription rules of famous ancient physicians in the treatment of threatened miscarriage. Methods: Traditional Chinese Medicine (TCM) prescriptions for threatened miscarriage were screened out of Fu Ren Da Quan Liang Fang by Ziming Chen, Yi Zong Jin Jian by Qian Wu, and Fu Qing Zhu Nv Ke by Qingzhu Fu. Data were standardized and analyzed through the TCM inheritance auxiliary platform. Results: A total of 29 prescriptions for threatened miscarriage were screened. Dang Gui, E Jiao, Gan Cao, Chuan Xiong, Bai Shao were the top five frequently prescribed Chinese herbs. The common herb–herb combinations used by Ziming Chen contained E Jiao, Dang Gui, Chuan Xiong, Ai Ye, Cong Bai, and Sang Ji Sheng. Ren Shen, Gan Cao, and Bai Zhu were the common herbal groups used by Qingzhu Fu. Huang Qi, Shu Di Huang, Bai Shao, Dang Gui, and Gan Cao were one of Qian Wu’s core prescriptions, with Dang Gui and Chuan Xiong being the others. According to the analysis of four Qi, five flavors, and meridian tropism of the prescriptions, herbs with the warm nature, or with the sweet, pungent, bitter flavors topped the list of application. The top six meridian tropisms of high-frequency herbs were: liver, spleen, lung, kidney, heart, and stomach meridian. Conclusion: Based on the principle of restoring the balance within the organs and enriching Qi and blood, clinical treatment of threatened miscarriage involves invigorating the Chong and Ren channels, nourishing Yin, dispelling cold and wind, generating and activating blood, regulating and harmonizing Qi.

Abstract Purpose: Cancer has become the second leading cause of death in the world. In 2020, there were 19.29 million new cancer patients and a total of 10 million deaths. Drug are the most popular ways to control the development of cancer, but the effectiveness of anti-tumor drugs is only about 25%. Drug efficacy is affected by many factors, among which the most critical is genetic variants. Genomic analysis based on large-scale population sequencing was performed to calculate the mutation frequency of antineoplastic drug-related genetic variations and systematically analyze general descriptions and population differences. Methods: Information of antineoplastic drugs is available from FDA and CFDA, and gene list comes from DrugBank, PharmGKB, FDA biomarker list and clinical guidelines. The maximum likelihood method was used to estimate the population mutation frequency of a single locus, and the calculation process was completed on Ali Cloud platform. GATK, ANNOVAR, PROVEAN, SIFT and other software were used for quality control, annotation and function prediction of mutations. Perl, Python, R and other computer languages were used for statistical analysis. Results: we collect 206,604 samples from mainland China for analysis, and identified 104 antineoplastic drugs and 517 drug-related genes at the same time. There were 201,774 genetic variations, most of which were intron mutations, exon mutations account for about 1% of all mutations. We found 41,955 novel mutations, which unrecorded in dbSNP, accounting for about 20.8%. Most of these mutations were rare mutations with frequencies less than 1%, subsequent prediction of mutation function also indicated that a large proportion of these mutations were deleterious. These results suggest that rare mutations may also play an important role in the use of antineoplastic drugs, which has previously been underestimated. The mutation frequency distribution of different types of genes suggests that transporters have a greater impact on personalized medicine during the use of antineoplastic drugs in China. By analyzing the genetic variation of important metabolic enzymes, we found that mutations such as UGT1A1*6, DPYD*9A, CYP2D6*2 and *10 have a high frequency and high detection probability in the Chinese population. Therefore, when patients use drugs metabolized by related metabolic enzymes, genetic testing should be done in advance to prevent the occurrence of adverse reactions. Regional analysis revealed that the overall differences among provinces in China were small, but the differences were large compared with other ethnic groups. Conclusion: Results of large-scale population sequencing reveal distribution and discipline of genetic variation of antineoplastic drug-related genes in Chinese population, further confirm the importance of precision medicine in cancer, and illustrate that the use of antineoplastic drugs can also be affected by ethnic differences. The lithium metal anode is highly valued in the development of high-energy density storage devices owing to its high specific capacity. However, the growth of lithium dendrites and high-volume expansion during the charging process creates significant issues for commercial application. To regulate the lithium deposition behavior various methods have been proposed among which carbon-based materials are being widely investigated because of their unique advantages including high conductivity, along with the material availability and ease of operation. However, current literature on lithium deposition in carbonaceous hosts have contradictory conclusions and further investigation is needed1–4. Herein, pure hollow core-carbon spheres (PHCCSs), deposited on copper foil (PHCCS@Cu), are used to study the lithium deposition behavior with respect to this type of structure. It is demonstrated that lithium shows some initial and limited intercalation into the PHCCSs and then plates on its external walls and the top-surface of the hollow core carbon structures during the charging process. Figure 1 shows initial lithium insertion followed by lithium deposition with a greater amount of charging. A possible mechanism of lithium deposition inside the PHCCSs is discussed from the aspect of lithium-ion transport and lithium deposition site preference. ArgumentRecent studies in Sinology have shown that Qing dynasty editors acted as philologists. This paper argues that the identification of their philological methods and editorial choices suggests that their choices were not totally neutral and may have significantly shaped the way modern historians interpreted specific works edited by mathematicians of that dynasty. A case study of the re-edition in 1798 of a Song dynasty treatise, theYigu yanduan(1259), by a Qing dynasty mathematician will illustrate this point. At the end of the eighteenth century, Li Rui (1773–1817) was asked to prepare an edition of the mathematical works written by Li Ye (1192–1279) for a private collection. Li Rui was a talented mathematician, but he was also a meticulous editor and trained philologist. He adopted his editorial model from the preparation of the imperial encyclopaedia, theSiku quanshu, but Li Rui also made some corrections to the text in an effort to restore an older version of Li Ye's treatises that had been lost. Convinced of the Chinese origin of algebra, Li Rui used philological techniques to recover the lost materials and to restore the roots of “Chinese mathematics.” TheYigu yanduancontains two algebraic procedures to set up quadratic equations, one from the procedure ofCelestial Source(tian yuan shu) and the other from theSection of Pieces[of Areas] (tiao duan). Curiously, the second procedure has not yet attracted the attention of scholars so far, although Li Rui's edition is the one typically used by twentieth-century historians of mathematics. Today, theCelestial Sourcecharacterizes “Chinese algebra.” However, the specific concerns of Li Rui about the procedure ofCelestial Source, combined with his editorial methods, contributed to this perspective.

Abstract Androgen receptor- (AR-) indifference is a mechanism of resistance to hormonal therapy in prostate cancer (PC). Here we demonstrate that ONECUT2 (OC2) activates resistance through multiple drivers associated with adenocarcinoma, stem-like and neuroendocrine (NE) variants. Direct OC2 targets include the glucocorticoid receptor (GR) and the NE splicing factor SRRM4, which are key drivers of lineage plasticity. Thus, OC2, despite its previously described NEPC driver function, can indirectly activate a portion of the AR cistrome through epigenetic activation of GR. Mechanisms by which OC2 regulates gene expression include promoter binding, enhancement of genome-wide chromatin accessibility, and super-enhancer reprogramming. Pharmacologic inhibition of OC2 suppresses lineage plasticity reprogramming induced by the AR signaling inhibitor enzalutamide. These results demonstrate that OC2 activation promotes a range of drug resistance mechanisms associated with treatment-emergent lineage variation in PC and support enhanced efforts to therapeutically target OC2 as a means of suppressing treatment-resistant disease.
