The general notion of complete hydatidiform moles is that most of them consist entirely of paternal DNA; hence, they do not express p57, a paternally imprinted gene. This forms the basis for the diagnosis of hydatidiform moles. There are about 38 paternally imprinted genes. The aim of this study is to determine whether other paternally imprinted genes could also assist in the diagnostic approach of hydatidiform moles. This study comprised of 29 complete moles, 15 partial moles and 17 non-molar abortuses. Immunohistochemical study using the antibodies of paternal-imprinted (RB1, TSSC3 and DOG1) and maternal-imprinted (DNMT1 and GATA3) genes were performed. The antibodies’ immunoreactivity was evaluated on various placental cell types, namely cytotrophoblasts, syncytiotrophoblasts, villous stromal cells, extravillous intermediate trophoblasts and decidual cells. TSSC3 and RB1 expression were observed in all cases of partial moles and non-molar abortuses. In contrast, their expression in complete moles was identified in 31% (TSSC3) and 10.3% (RB1), respectively (p < 0.0001). DOG1 was consistently negative in all cell types in all cases. The expressions of maternally imprinted genes were seen in all cases, except for one case of complete mole where GATA3 was negative. Both TSSC3 and RB1 could serve as a useful adjunct to p57 for the discrimination of complete moles from partial moles and non-molar abortuses, especially in laboratories that lack comprehensive molecular service and in cases where p57 staining is equivocal.

This letter contributes how to calculate the anomalous behavior for the damping constant (halfwidth) of the ferroelectric Tris-Sarcosine Calcium Chloride (TSSC) and its brominated compounds TSSC1−xBrx (x = 0.13, 0.42 and 0.60) from the wavenumber data of the soft modes below the phase transition temperature of TC. The pseudospin-phonon coupled (PS) and the energy fluctuation (EF) models derived from the dynamical Ising model were used. Both PS and EF models have been used to contribute understanding the temperature dependence of the phase transition mechanism of TSSC1−xBrx. In addition, values of the activation energy for TSSC1−xBrx (x = 0, 0.13, 0.42 and 0.60) were extracted from the damping constant as calculated from both models (PS and EF). Our results indicate order-disorder type phase transition for TSSC1−xBrx. HIGHLIGHTS This work contributes how to calculating the critical behavior of the halfwidth for the soft Raman modes in ferroelectric TSSC1-xBrx (x = 0, 0.13, 0.42 and 0.60) liquid crystals from their observed wavenumbers data. Also, the values of the activation energy close to the ferroelectric-paraelectric phase transition of these crystals was deduced. This calculation of the halfwidth and the activation energy were performed using the pseudospin-phonon coupled (PS) and the energy fluctuation (EF) models in the ferroelectric phases (T < TC) of the crystals studied here. The Institute of Medicine (IOM) and the Commission on Cancer (CoC) recommend that survivors receive personalized Treatment Summaries and Survivorship Care Plans (TSSCP) in order to facilitate best practice in clinical surveillance and follow-up care. This TSSCP template development study joined advocacy, scientific and medical communities to comprise a diverse advisory council (AC, N=38) to inform the development of the TSSCP-AA (African American) and TSSCP-S (Latino) templates targeting BCA. This abstract reports on the development, and presents the TSSCP-AA and TSSCP-S. Methods: The overall study was guided by the Shared Care and Psychooncology Models, and Contextual Model of Health Related Quality of Life. The AC provided input into the culturally relevant modifications of the American Society of Cancer Oncology (ASCO) TSSCP template to create the TSSCP-AA and TSSCP-S. Health care professionals enlisted from cancer centers and community hospitals (n=51) evaluated the TSSCP-AA and TSSCP-S templates on the following domains: content, clarity, utility, and cultural and socioecological responsiveness. Results: AC input revealed that the clinical (i.e., comorbidities) and cultural contexts (i.e., language, spirituality) must inform TS and SCP, respectively. Preliminary evaluation analyses documented that health professionals rated our TSSCP-AA and TSSCP-S templates as excellent to outstanding on content, clarity, utility, cultural responsiveness, and socioecological responsiveness (p<0.01). Conclusions: The evaluation results imply that the TSSCP-AA and TSSCP-S templates achieved cultural responsiveness with high acceptability and utility among professionals and survivors. We created both paper and electronic versions of theTSSCP-AA andTSSCP-S (English and bilingual English-Spanish formats) for providers to create individualized TSSCPs. Further, our TSSCP development approach can be adopted for TSSCP template development targeting ethnic/linguistic minority cancer survivors.

AbstractWe investigated the role of the hematopoietic-specific tetraspanin superfamily member, TSSC6, in platelet function using wild-type mice and TSSC6-deficient mice. TSSC6 is expressed on the surface of murine platelets and is up-regulated by thrombin stimulation, indicating an intracellular pool of TSSC6. Immunoprecipitation/Western blot studies reveal a constitutive physical association of TSSC6 with the integrin αIIbβ3 complex under strong detergent conditions. In vivo evaluation of hemostasis by tail bleeding revealed increased bleeding time, volume of blood lost, and evidence of tail rebleeds in TSSC6 null mice, indicating unstable hemostasis. Using ex vivo techniques, we showed that TSSC6-deficient platelets exhibited impaired kinetics of clot retraction, platelet aggregation at lower doses of PAR-4, and collagen and platelet spreading on fibrinogen in the presence of normal integrin αIIbβ3 expression. TSSC6-deficient platelets showed normal alpha granule secretion, normal “insideout” integrin αIIbβ3 signaling (fluorescein isothiocyanate [FITC]–fibrinogen and JON/A binding), and normal platelet adhesion on fibrinogen. Furthermore, we show that absence of platelet TSSC6 affects the secondary stability of arterial thrombi in vivo upon vascular injury. These data demonstrate that TSSC6 appears to regulate integrin αIIbβ3 “outside-in” signaling events in platelets and is necessary for stability of arterial thrombi in vivo.

ABSTRACT The tetraspanins are a family of integral membrane proteins with four transmembrane domains. These molecules form multimolecular networks on the surfaces of many different cell types. Gene-targeting studies have revealed a role for tetraspanins in B- and T-lymphocyte function. We have isolated and deleted a novel tetraspanin, Tssc6, which is expressed exclusively in hematopoietic and lymphoid organs. Using a gene-trapping strategy, we generated an embryonic stem (ES) cell line with an insertion in the Tssc6 locus. Mice were derived from these ES cells and, using RNase protection and reverse transcription-PCR, we demonstrated that the insertion resulted in a null mutation of the Tssc6 allele. Mice homozygous for the gene trap insertion (Tssc6 gt/gt mice) were viable and fertile, with normal steady-state hematopoiesis. Furthermore, responses to hemolysis and granulocyte colony-stimulating factor-induced granulopoiesis were equivalent to those of wild-type mice. Lymphoid development was normal in Tssc6 gt/gt mice. Whereas Tssc6 gt/gt B cells responded normally to lipopolysaccharide, anti-CD40, and anti-immunoglobulin M stimulation, Tssc6 gt/gt T cells showed enhanced responses to concanavalin A, anti-CD3, and anti-CD28. This increased proliferation by Tssc6-deleted T lymphocytes was due to increased interleukin 2 production following T-cell receptor stimulation. These results demonstrate that Tssc6 is not required for normal development of the hematopoietic system but may play a role in the negative regulation of peripheral T-lymphocyte proliferation.

Abstract Biogenesis of spliceosomal small nuclear ribonucleoproteins (snRNPs) and their recycling after splicing require numerous assembly/recycling factors whose modes of action are often poorly understood. The intrinsically disordered TSSC4 protein has been identified as a nuclear-localized U5 snRNP and U4/U6-U5 tri-snRNP assembly/recycling factor, but how TSSC4’s intrinsic disorder supports TSSC4 functions remains unknown. Using diverse interaction assays and cryogenic electron microscopy-based structural analysis, we show that TSSC4 employs four conserved, non-contiguous regions to bind the PRPF8 Jab1/MPN domain and the SNRNP200 helicase at functionally important sites. It thereby inhibits SNRNP200 helicase activity, spatially aligns the proteins, coordinates formation of a U5 sub-module and transiently blocks premature interaction of SNRNP200 with at least three other spliceosomal factors. Guided by the structure, we designed a TSSC4 variant that lacks stable binding to the PRPF8 Jab1/MPN domain or SNRNP200 in vitro. Comparative immunoprecipitation/mass spectrometry from HEK293 nuclear extract revealed distinct interaction profiles of wild type TSSC4 and the variant deficient in PRPF8/SNRNP200 binding with snRNP proteins, other spliceosomal proteins as well as snRNP assembly/recycling factors and chaperones. Our findings elucidate molecular strategies employed by an intrinsically disordered protein to promote snRNP assembly, and suggest multiple TSSC4-dependent stages during snRNP assembly/recycling.

1512 Background: Tobacco smoking profoundly impacts LC risk; however, data are limited as to what extent SC prior to diagnosis impacts LC overall survival (OS) and lung cancer specific survival (LCSS). LC screening offers a possible teachable moment, but there is uncertainty of SC benefits after a lifetime of smoking. We use the ILCCO database to answer if SC prior to LC dx is associated with better OS and LCSS, considering time since smoking cessation (TSSC). Methods: Using individual data, analysis was performed on 17 ILCCO studies with available TSSC to estimate survival using univariable analysis and models of stage-adjusted and cumulative smoking-adjusted multivariable analysis. Adjusted Hazard Ratios (aHR) from Cox models, cubic spline smooth curves and Kaplan-Meier curves were created. Sensitivity analysis was performed for TSSC and LCSS on 13 studies. Results: Of 34649 patients, 14322 (41%) were current smokers 14273 (41%) ex-smokers and 6054 (18%) never smokers at diagnosis. We confirmed that ex-smokers (aHR 0.88 CI 0.86-0.91) and never smokers (aHR 0.76 CI 0.73-0.8) improved OS compared to current smokers. Amongst ex-smokers, < 2y TSSC (aHR 0.88 CI 0.82-0.94), 2-5y TSSC (aHR 0.83 CI 0.77-0.90) and > 5y TSSC (aHR 0.8 CI 0.76-0.84) had improved OS compared to CS. Sensitivity analysis showed a trend towards improved LCSS survival for < 2y TSSC (aHR 0.95 CI 0.86-1.05) and 2-5y TSSC (aHR 0.93 CI 0.83-1.04), whereas > 5y TSSC significantly improved LCSS by 15% (aHR 0.85 CI 0.78-0.92). To mimic the LC screening participants, in analysis of > 30 pack-years individuals, associations were strikingly strong: < 2y TSSC had improved OS by 14% (aHR 0.86 CI 0.80-0.93); 2-5y TSSC by 17% (aHR 0.83 CI 0.76-0.90); and > 5 TSSC by 22% (aHR 0.78 CI 0.74-0.83), compared to current smokers; for < 30 packs-years, a trend towards better OS was observed for < 2y TSSC (aHR 0.95 CI 0.92-1.02) and 2-5y TSSC (aHR 0.86 CI 0.74-1.01), whereas > 5y TSSC improved OS by 23% (aHR 0.77 CI 0.72-0.82). Conclusions: Among ex-smokers, the risk of overall death was reduced by 12% on < 2y TSSC, 17% on 2-5y TSSC and 20% > 5y TSSC, whereas for LCSS, the benefit was significant only for > 5y TSCC, compared to current smokers at time of diagnosis. Here we demonstrate that convincing screening participants to quit smoking at any point of their trajectory, even just prior to dx such as < 2y TSSC, improved OS, and LCSS benefit was present beyond 5y of quitting. These relationships are independent of pack-years, age, across all stages and other prognostic variables.

Endosomes function as a hub for multiple protein-sorting events, including retrograde transport to the trans-Golgi network (TGN) and recycling to the plasma membrane. These processes are mediated by tubular-vesicular carriers that bud from early endosomes and fuse with a corresponding acceptor compartment. Two tethering complexes named GARP (composed of ANG2, VPS52, VPS53, and VPS54 subunits) and EARP (composed of ANG2, VPS52, VPS53, and Syndetin subunits) were previously shown to participate in SNARE-dependent fusion of endosome-derived carriers with the TGN and recycling endosomes, respectively. Little is known, however, about other proteins that function with GARP and EARP in these processes. Here we identify a protein named TSSC1 as a specific interactor of both GARP and EARP and as a novel component of the endosomal retrieval machinery. TSSC1 is a predicted WD40/β-propeller protein that coisolates with both GARP and EARP in affinity purification, immunoprecipitation, and gel filtration analyses. Confocal fluorescence microscopy shows colocalization of TSSC1 with both GARP and EARP. Silencing of TSSC1 impairs transport of internalized Shiga toxin B subunit to the TGN, as well as recycling of internalized transferrin to the plasma membrane. Fluorescence recovery after photobleaching shows that TSSC1 is required for efficient recruitment of GARP to the TGN. These studies thus demonstrate that TSSC1 plays a critical role in endosomal retrieval pathways as a regulator of both GARP and EARP function.

Decentralized treatment of stormwater runoff from heavily polluted surface can be a good solution for effective source control. Decentralized stormwater treatment systems (DS) and test procedures to monitor their performance, have been developed in recent years. At present in Germany, only lab-based tests are officially established to determine the removal efficiency of Total Suspended Solids (TSS), and in situ monitoring is still lacking. Furthermore, the fine fraction of TSS with particle sizes less than 63 µm (TSS63) have been established as a new design parameter in Germany, because of their substitute characteristics of adsorbing pollutant substances. For research and evaluation purposes continuous data of urban stormwater runoff quantity and quality at the in- and outflow of two different DS at two different sites were collected. Turbidity is used as a surrogate for TSS. Continuous turbidity data and time proportional sampling served to obtain (i) regression coefficients and (ii) to determine the TSS removal efficiency of DS. For a wide range of events the total removal efficiency of DS1 was 29% for TSS and 19% for TSS63 and for DS2 19% for TSS and 16% for TSS63. An event-based data analysis revealed a high variability of the efficiencies and its uncertainties. Moreover, outwash of still suspended or remobilization of already deposited material was observed at individual events. At both sites TSS63 dominates urban stormwater runoff as indicated by the mean ratios of TSS63 to TSS of 0.78 at the inflows and 0.89 at the outflows of both DS. A significant shift of TSS63 ratio from inflow to outflow demonstrates that TSS63 particles were removed less efficiently than coarser particles by DS1, for DS2 data was too heterogeny. It clarifies that common sedimentation methods can only contribute to a small extent to the reduction of solid emissions if the stormwater runoff contains mainly fine-particle solids. The findings suggest that treatment of urban stormwater runoff with high TSS63 pollution requires additional techniques such as a proper filtering to retain fine particles more effective.

Purpose: To assess the 3-year survival rates and the incidences of biological and technical complications of all-ceramic and metal-ceramic tooth-supported single crowns. Material and methods: An online research of literature on Pubmed was conducted independently by 3 reviewers to identify the clinical studies performed from 2000 up to 2017 and completed by a manual research. Keywords, inclusion and exclusion criteria were well-defined. Results: The research revealed 501 titles and led to a final analysis of 31 full text articles. Only 6 studies met the inclusion criteria and reported on 215 metal-ceramic and 501 all-ceramic TSSCs. The meta-analysis of the included studies indicated an estimated survival rate of metal-ceramic TSSCs of 90.14% (95% CI: 89.72%-91.35%) after 3 years. This was a little bit higher than the estimated 3-year survival rate of Lithium-disilicate (89.54%; 95% CI: 87.18%-91.89%). However the 3-year survival rate, of Zirconia TSSCs was statistically much higher (92.01%; 95% CI: 89.58-94.43%). When the outcomes in anterior and posterior regions were compared, Zirconia and Lithium-disilicate TSSCs exhibited survival rates of 91.64% and 91.12% respectively, in the posterior region, where metal-ceramic TSSCs exhibited only 90.14% as a 3-year survival rate. As for the anterior region, we could only compare the TSSCs made out of Lithium-disilicate and Zirconia as the metal-ceramic ones were all laid on the posterior regions. Zirconia performed the highest 3-year survival rate in the anterior region (93.54%; 95% CI: 91.08-96%) whereas Lithium-disilicate showed only 87.5%. This was due to the low level of the mechanical properties of such material, which clinically indicated by major chipping and tooth fracture. Conclusion: Survival rates of metal-ceramic crowns were nearly similar to those reported for most types of all-ceramic TSSCs in the posterior region. Weaker Lithium-disilicate-based ceramics should be limited to application in the anterior region, even if they expressed a higher survival rate in the anterior regions. Zirconia-based TSSCs should not be considered as a primary option due to their high incidence of technical problems. 72 Background: Multiple cancer organizations advocate for the use of treatment summaries and survivorship care plans (TSSCPs) in cancer patients. To better quantify the benefits of a nurse-navigated, culturally and linguistically responsive TSSCP in underserved breast cancer patients, we compared rates of compliance with treatment and follow up in 26 patients who were treated with TSSCPs to 38 similar controls who were treated without TSSCPs. Methods: We prospectively enrolled 26 consecutive, newly-diagnosed breast cancer patients who were given nurse-navigated TSSCPs under an IRB-approved protocol. At their first clinic visit, a trained nurse educated and assisted the patient in the use and completion of the TSSCP. Nurse-navigated TSSCPs were completed at each subsequent visit through 12 months of surveillance. Rates of compliance with treatment and follow up guidelines were compared to 38 similar control patients using a two group Fisher’s exact chi-square test. Statistical significance was set at p-value < 0.05. Results: All patients were treated under Medicaid insurance and 47% were racial and/or ethnic minorities. Time from diagnosis to treatment and time from initial clinic visit to treatment were similar across groups. The rate of compliance with first treatment recommendations was 96% (25/26) in the TSSCP group compared to 79% (30/38) in the non-TSSCP group (p = 0.07). The number of patients compliant with all follow up visits was similar: 22/25 (88%) of TSSCP patients and 22/30 (73%) in non-TSSCP patients (p = 0.31). Of the recommended total follow up appointments, 6/67 in the TSSCP group and 25/120 in the no TSSCP group were “no shows” (p = 0.04). Conclusions: Although the use of nurse-navigated TSSCPs may not improve time to treatment in medically underserved patients, adherence to first treatment recommendations and follow up appointment attendance shows some improvement in patients who participate in nurse-navigated TSSCPs. Further study is needed to fully assess the role of nurse-navigated TSSCPs in improving treatment and surveillance compliance rates and how these rates impact clinical outcomes. The debate about the necessary involvement of any form of self-consciousness in conscious experience has recently shifted its focus to the question of whether there are totally selfless states of consciousness (TSSC). The primary source of evidence for the existence of TSSC is the subjective reports from subjects who either are currently undergoing or have undergone altered experiences such as drug-induced ego dissolution. While the subjective reports are made largely after the occurrence of the experience, such reports have been challenged on the basis that one cannot coherently report about TSSC from one’s own, autobiographical memory. This paper addresses this issue regarding TSSC from the perspective of memory study. The aim is to examine whether and how it is possible for a subject to report a past TSSC based on her own memory without being considered as confabulating or misremembering. Such an examination can provide potential explanations for how a person reports a past experience of TSSC

CAD/CAM/CAE (3C) was effectively applied to twin-spirals scroll compressor (TSSC) from conceptual design to finished products in this paper. A virtual model and mathematical model of TSSC was developed. The swarm intelligence algorithm was used for TSSC optimization and the final optimization design variables were gotten. CAE was used to study the two key components of TSSC-orbiting and fixed scroll, which is useful for improving the quality and reliability of manufacturing and assembly accuracy at the later stages. CAD, CAPP, CAM, CAE and NC tool path code generation are included in 3C. The prototype of TSSC was manufactured based on 3C. The used results indicate that 3C method can well overcome the blindness and limitation of the traditional design. It offers the new method and idea for wide application of the scroll compressor.

ABSTRACT Many viral genes contain core promoters with two basal control elements, the TATA box and the pyrimidine-rich initiator (Inr). However, the molecular mechanisms involved in transcription initiation from composite core promoters (TATA+ Inr+) containing Inr elements are unclear. The Rous sarcoma virus (RSV) long terminal repeat (LTR) contains a transcriptionally potent enhancer and core promoter composed of a TATA box and an Inr-like sequence, termed the transcription start site core (TSSC). Previously we demonstrated that the TSSC binds the multifunctional Inr-binding protein YY1. Here we present evidence that the TSSC also binds the multifunctional transcription factor TFII-I and that both TFII-I and YY1 are required for RSV LTR transcriptional activity. Gel shift assays using anti-TFII-I antibody show that TFII-I is present in a protein complex that specifically binds to the TSSC. Mutations in the TSSC that reduce TFII-I binding also reduce RSV LTR enhancer and promoter activity. Transient-transfection assays demonstrate that TFII-I transactivates the RSV LTR from ca. fourfold (basal) to ca. sevenfold (enhanced) in both human and natural host cell lines. Importantly, the activity of the TSSC element can be attributed to the binding activity of TFII-I and the YY1 protein, since mutation of each of these binding sites within the TSSC element abolishes all viral expression as demonstrated by transient-transfection assays. Taken together, these data demonstrate that expression of RSV viral mRNA is dependent on both TFII-I and YY1.

ABSTRACT The Rous sarcoma virus (RSV) long terminal repeat (LTR) contains a transcriptionally potent enhancer and promoter that functions in a variety of cell types. Previous studies have identified the viral sequences required for enhancer activity, and characterization of these elements has provided insight into the mechanism of RSV transcriptional activity. The objective of this study was to better define the RSV LTR promoter by examining the transcription start site core (TSSC) region. Deletion of the TSSC resulted in complete loss of transcriptional activity despite the presence of a functional TATA box, suggesting that the TSSC is required for viral expression. Homologies within the TSSC to the DNA binding motif of YY1 suggested that it might regulate promoter activity. YY1 has been shown to regulate transcription in some cellular genes and viral promoters by binding to sites overlapping the transcription start site. Gel shift assays using YY1 antibody identified YY1 as one of three complexes that bound to the TSSC. Mutation of the YY1 binding site reduced RSV transcriptional activity by more than 50%, suggesting that YY1, in addition to other TSSC-binding factors, regulates RSV transcription. Furthermore, in vitro transcription assays performed with Drosophila embryo extract (devoid of YY1 activity) showed decreased levels of RSV transcription, while transient transfection experiments overexpressing YY1 demonstrated that YY1 could transactivate the RSV LTR ∼6- to 7-fold. We propose that the TSSC plays a vital role in RSV transcription and that this function is partially carried out by the transcription factor YY1.

We observed a case of high-risk neuroblastoma (NB) carried by a 28-month-old girl, displaying metastatic disease and a rapid decline of clinical conditions. By array-CGH analysis of the tumor tissue and of the metastatic bone marrow aspirate cells, we found a high-grade amplification of six regions besides MYCN on bands 2p25.3–p24.3. The genes involved in these amplifications were MYT1L, TSSC1, CMPK2, RSAD2, RNF144A, GREB1, NTSR2, LPIN1, NBAS, and the two intergenic non-protein coding RNAs LOC730811 and LOC339788. We investigated if these DNA co-amplifications may have an effect on enhancing tumor aggressiveness. We evaluated the association between the high expression of the amplified genes and NB patient’s outcome using the integration of gene expression data of 786 NB samples profiled with different public platforms from patients with at least five-year follow-up. NB patients with high expression of the TSSC1 gene were associated with a reduced survival rate. Immunofluorescence staining on primary tumor tissues confirmed that the TSSC1 protein expression was high in the relapsed or dead stage 4 cases, but it was generally low in NB patients in complete remission. TSSC1 appears as a putative new oncogene in NB.

This paper compares by simulation the Thyristor Switched Series Capacitors (TSSC) Circuit with the Thyristor Switched Parallel Capacitors (TSPC) Circuit for wind turbines. The well-known TSSC circuit belongs to the Controlled Series Capacitor (CSC) circuits that have been used in power transmission lines in order to correct the power factor and improve the performance of the electrical system. Such a circuit can be used in wind power systems to improve and maximize the efficiency of a wind turbine. A typical direct-drive wind power system employs variable speed electric generators, but the downside is that systems like that suffer from high inductive reactance. A TSSC circuit, therefore, is able to counteract for any reactive losses, and improve the power factor as well as the efficiency. The main issue with the TSSC circuit is the use of a high number of capacitors that must be connected in series, which can increase the cost and the maintenance of the controller. This paper introduces a novel circuit with different control technique than the TSSC that employs capacitors in parallel configuration. The novel TSPC circuit was simulated in PSPICE and the benefits as well as the drawbacks are described

Abstract Particle accumulation and circulation in water distribution systems are significant in the development of good management practices to protect against discoloration events, which are a major cause of water customer complaints. Quantifying the amount of particles deposited in water pipes is usually done by obtaining total suspended solid measurements while performing flushing sequences, which requires time, skills, and equipment. Some authors explored the possibility of rapidly approximating total suspended solids concentration (TSSC) in water pipes by measuring water turbidity on site, but they obtained different results and coefficients of correlation. This paper presents the results of tests performed in the laboratory on a test loop. Unidirectional flushing (UDF) and air scouring sequences were performed under various hydraulic conditions and two different particle origins. Samples were obtained along each sequence, and the turbidity and TSSC were measured. The results illustrate that the ratio between turbidity and TSSC may vary greatly between samples, up to 10 times during UDF sequences and 20 times during air scouring sequences. Particle origin, flushing method, and sampling time are all factors impacting the turbidity/TSSC ratio. This is why TSSC should not be estimated from a single turbidity reading.

The flavor of Pomelo is highly variable and difficult to determine without peeling the fruit. The quality of pomelo flavor is due largely to the total soluble solid content (TSSC) in the fruit and there is a commercial need for a quick but nondestructive TSSC detection method for the industrial grading of pomelo. Due to the large size and thick mesocarp of pomelo, determining the internal quality of a pomelo fruit in a nondestructive manner is difficult, and the detection accuracy is further complicated by the noise typically generated by the common methods for the internal quality detection of other fruits. Thus, the aim of this study was to determine the optimal method to accurately detect pomelo TSSC and find a de-noising model which reduces the influence of noise on the optimal method’s results. After developing a full-transmission visible/near infrared (VIS/NIR) spectroscopy sampling method, the confirming experimental results showed that the optimal pomelo TSSC detection model was Savitzky Golay + standard normal variate + competitive adaptive reweighted sampling + partial least squares regression. The R2 and RMSE of the calibration set for pomelo TSSC detection were 0.8097 and 0.8508, respectively, and the R2 and RMSE of the validation set for pomelo TSSC detection were 0.8053 and 0.8888, respectively. Both reference and dark de-noising are important for pomelo internal quality detection and should be calibrated frequently to compensate for time drift. This study found that large sensor response translation noise can be reduced with an artificial horizontal shift. Data supplementation is efficient for improving the adaption of the detection model for batch differences in pomelo samples. Using this optimized de-noising model to compensate for time drift, sensor response translation, and batch differences, the developed detection method is capable of satisfying the requirements of the industry (TSSC detection R2 was equal or larger than 0.9, RMSE was less than 1). These results indicate that full-transmission VIS/NIR spectroscopy can be exploited to realize the nondestructive detection of pomelo TSSC on an industrial scale, and that the methodologies used in this study can be immediately implemented in real-world production.

Abstract. This study conducts an analytical investigation of the dynamic response characteristics of a two-stage series composite system (TsSCS) with a planetary transmission consisting of dual-power branches. An improved incremental harmonic balance (IHB) method, which solves the closed solution of incremental parameters passing through the singularity point of the analytical path, based on the arc length extension technique, is proposed. The results are compared with those of the numerical integration method to verify the feasibility and effectiveness of the improved method. Following that, the multi-scale perturbation (MsP) method is applied to the TsSCS proposed in this subject to analyze the parameter excitation and gap nonlinear equations and then to obtain the analytical frequency response functions including the fundamental, subharmonic, and superharmonic resonance responses. The frequency response equations of the primary resonance, subharmonic resonance, and superharmonic resonance are solved to generate the frequency response characteristic curves of the planetary gear system (PGS) in this method. A comparison between the results obtained by the MsP method and the numerical integration method proves that the former is ideal and credible in most regions. Considering the parameters of TsSCS excitation frequency and damping, the nonlinear response characteristics of steady-state motion are mutually converted. The effects of the time-varying parameters and the nonlinear deenthing caused by the gear teeth clearance on the amplitude–frequency characteristics of TsSCS components are studied in this special topic.

This study evaluated the dissemination and implementation of a culturally tailored community-wide campaign (CWC), Tu Salud ¡Si Cuenta! (TSSC), to augment fruit and vegetable (FV) consumption and physical activity (PA) engagement among low-income Latinos of Mexican descent living along the U.S.-Mexico Border in Texas. TSSC used longitudinal community health worker (CHW) home visits as a core vehicle to enact positive change across all socioecological levels to induce behavioral change. TSSC’s reach, effectiveness, adoption, implementation, and maintenance (RE-AIM) was examined. A dietary questionnaire and the Godin-Shepherd Exercise Questionnaire measured program effectiveness on mean daily FV consumption and weekly PA engagement, respectively. Participants were classified based on CHW home visits into “low exposure” (2–3 visits) and “high exposure” (4–5 visits) groups. The TSSC program reached low-income Latinos (n = 5686) across twelve locations. TSSC demonstrated effectiveness as, compared to the low exposure group, the high exposure group had a greater FV intake (mean difference = +0.65 FV servings daily, 95% CI: 0.53–0.77) and an increased PA (mean difference = +185.6 MET-minutes weekly, 95% CI: 105.9–265.4) from baseline to the last follow-up on a multivariable linear regression analysis. Multivariable logistic regression revealed that the high exposure group had higher odds of meeting both FV guidelines (adjusted odds ratio (AOR) = 2.03, 95% CI: 1.65–2.47) and PA guidelines (AOR = 1.36, 95% CI: 1.10–1.68) at the last follow-up. The program had a 92.3% adoption rate, with 58.3% of adopting communities meeting implementation fidelity, and 91.7% of communities maintaining TSSC. TSSC improved FV consumption and PA engagement behaviors among low-income Latinos region wide. CHW delivery and implementation funding positively influenced reach, effectiveness, adoption, and maintenance, while lack of qualified CHWs negatively impacted fidelity.

Objectives: To evaluate in vitro the endogenous pH, titratable acidity and total soluble solid content (TSSC) of mouthwashes available in the Brazilian market.Methods: The study sample was composed of 10 commercial brands of mouthwashes based on different active ingredients: Cepacol®, Clinerize®, Equate®, Listerine Cool Citrus®, Oral-B®, Periogard ®, Peroxyl®, Plax Overnight®, Prevident 220® and Sanifill®. The experiments were performed in triplicate. The endogenous pH was evaluated by potentiometry, titratable acidity was evaluated by the addition of 0.1N KOH increments to the mouthwashes, and TSSC readings were performed by Brix refractometry using the Abbé refractometer.Results: pH values ranged from 3.56 (Peroxyl®) to 7.43 (Cepacol®) and three mouthwashes presented pHs below 5.5. The titratable acidity values ranged from 0.007 (Periograd®) to 0.530 (Prevident ®). Oral B® and Clinerize® presented the lowest (4.7%) and the highest (23.70%) TSSC, respectively.Conclusions: Some of the mouthwashes evaluated in this study presented low endogenous pH, even below the critical value for enamel dissolution (pH<5.5), high titratable acidity and high TSSC, and may be potentially erosive to the dental tissues if not properly used. AbstractObjectivesDrug resistance exists to all current and investigational antimalarial drug classes. Consequently, we have set out to develop chemically and mechanistically discrete antimalarials. Here we report on the development of thiosemicarbazone (TSC) antimalarials, with TSC3 as the most advanced lead.MethodsThiosemicarbazones were generated through simple condensation reactions of thiosemicarbazides and ketones. TSC3 was selected and tested for in vitro antimalarial activities against MDR Plasmodium falciparum lines using the [3H]hypoxanthine growth assay, in vitro cytotoxicity against mammalian cell lines using the alamarBlue fluorescence cell viability assay, in vivo potency in the mouse–Plasmodium berghei model and blood exposure in mice measured by LC-MS for pharmacokinetic analysis.ResultsTSC3 showed potent in vitro activity against atovaquone-, dihydroartemisinin-, chloroquine- and mefloquine-resistant P. falciparum lines (EC50 <15 nM). The selectivity index (EC50 cells/EC50Pf W2 line) of TSC3 was >500 in two of three mammalian cell lines. In P. berghei-infected mice, TSC3 showed potent activity in the Peters 4 day suppression test (ED50 1.2 mg/kg/day) and was as potent as artesunate and chloroquine in the curative modified Thompson test. A single oral dose of TSC3 at 16 mg/kg in healthy mice achieved a mean maximum blood concentration of 1883 ng/mL at 1 h after dosing and an elimination half-life of 48.7 h in groups of five mice.ConclusionsTSC3 shows promise as a persistent, potent and orally effective antimalarial. This, coupled with the extremely low cost of synthesis, suggests that the further development of antimalarial thiosemicarbazones is clearly warranted.

Regarding attempts to find de-escalation methods of treatment for patients with HPV16-positive squamous cell carcinoma of the oropharynx (OPSCC), there is an urgent need to identify new prognostic factors which allow physicians to differentiate the prognosis of these patients. The aim of the study is to compare the incidence of transcriptionally active HPV16 infection and its type as well as other epidemiological, clinical, and histopathological features between SCC of the base of the tongue (BOTSCC) and tonsils (TSSCC). The analysis was performed in a group of 63 patients with OPSCC, for which, in our earlier studies, we assessed transcriptionally active HPV16 infection and its type (viral load and viral genome status). Transcriptionally active HPV16 infection was significantly more common in TSSCC (96.3%) than in BOTSCC (3.7%). Patients with TSSCC had significantly higher disease-free survival rates (84.1%) than those with BTSCC (47.4%); the same was true in the subgroup with HPV16 positivity. The obtained results are an important indication for further research on the development of new prognostic and/or predictive factors for patients with HPV16-positive squamous cell carcinomas of the oropharynx.

AbstractIn this study, four tuber sizes (28S1 45mm, 45S255mm, 55S365 mm and S465 mm) and three potato cultivars (Jelly, Milva and Sante) were used to assess the variations of physico-mechanical, chemical and colour properties depending on tuber size and potato cultivars. Physico-mechanical properties (geometric mean diameter, sphericity, surface area, bulk and tuber densities, volume, static coefficient of friction, puncture force and deformation), chemical properties (pH, total acidity [TA] and total soluble solid content [TSSC]), and colour characteristics (L, a, b) were determined. The physico-mechanical (geometric properties and puncture force) and chemical properties of potatoes were significantly affected by tuber sizes and cultivars, respectively. pH, TA and TSSC of potato tuber were higher in Milva, TA was higher in Jelly and TSSC was higher in Sante than in those of the other cultivars, respectively.

Background Cetirizine has been shown to be effective for relief of seasonal allergic rhinitis (SAR) symptoms. Allergic rhinitis symptoms have been reported to have circadian variations, with symptoms tending to be most bothersome overnight and in the morning. Objective To evaluate the effects of different cetirizine dosing schedules in comparison to twice daily (BID) chlorpheniramine and placebo on SAR symptoms at 12 and 24 hours postdose. Methods Study 1 subjects received cetirizine 10-mg once daily in the morning (QAM), cetirizine 10-mg once daily at bedtime (QHS), cetirizine 5-mg twice daily, or placebo. Study 2 subjects received cetirizine 5-mg QAM, cetirizine 10-mg QHS, chlorpheniramine 8-mg BID, or placebo. The primary end point was total symptom severity complex (TSSC); TSSC was the sum of symptom severity ratings averaged over the 2-week study period. Post hoc analyses of reflective symptom severity assessed in the morning (TSSCAM) and in the evening (TSSCPM) were conducted to evaluate cetirizine’s effects at 12 and 24 hours postdose. Results In study 1, subject- and investigator-assessed TSSC was significantly lower in all cetirizine groups versus placebo ( P ≤ .003). In study 2, subject-assessed TSSC was significantly lower in all cetirizine groups versus placebo ( P ≤ .04) and was numerically lower for investigator-assessed TSSC. Post hoc analyses demonstrated that cetirizine significantly improved TSSCAM at 12 and 24 hours postdose versus placebo in both studies regardless of dosing schedule. TSSCPM significantly improved at 12 and 24 hours postdose in all study 1 cetirizine groups versus placebo. In study 2, versus placebo, TSSCPM significantly improved at 12 hours postdose in cetirizine 5-mg QAM group and numerically improved at 24 hours postdose in cetirizine 10-mg QHS group. Conclusion Regardless of dosing regimen, cetirizine demonstrates effective 24-hour relief of SAR symptoms, particularly on TSSCAM, which assesses overnight and early morning symptom control.

The objective of this study was to find an efficient method for measuring the total soluble solid content (TSSC) and water content of “Luogang” orange. Quick, accurate, and nondestructive detection tools (VIS/NIR spectroscopy, NIR spectroscopy, machine vision, and electronic nose), four data processing methods (Savitzky–Golay (SG), genetic algorithm (GA), multi-source information fusion (MIF), convolutional neural network (CNN) as the deep learning method, and a partial least squares regression (PLSR) modeling method) were compared and investigated. The results showed that the optimal TSSC detection method was based on VIS/NIR and machine vision data fusion and processing and modeling by SG + GA + CNN + PLSR. The R2 and RMSE of the TSSC detection results were 0.8580 and 0.4276, respectively. The optimal water content detection result was based on VIS/NIR data and processing and modeling by SG + GA + CNN + PLSR. The R2 and RMSE of the water content detection results were 0.7013 and 0.0063, respectively. This optimized method largely improved the internal quality detection accuracy of “Luogang” orange when compared to the data from a single detection tool with traditional data processing method, and provides a reference for the accuracy improvement of internal quality detection of other fruits.

The effect of storage condition (room temperature and sunlight exposure) of eight carbonated beverages sold in Nigeria were assessed over a period of 15 weeks of storage time. Properties such as antimony (Sb) leaching, pH, acidity, specific gravity (S.G.) and total soluble solid content (TSSC) were analyzed using appropriate instrument and methodology at three weeks interval respectively. The concentration of Sb determined ranged from 0.001-0.011 mg/L and 0.001-0.0015 mg/L for room temperature and sunlight exposure. pH was between 2.82-4.81 and 2.82-4.82. TSSC were 0-14 O brix and 0-14.96 O brix. Acidity were 0.025-0.19 and 0.025-0.34. Specific gravity was 0.9921-1.052 and 0.9921-1.0577. The result shows that pH decreased with time as Sb, TSSC, acidity and S.G. increased with time thereby influencing significance (p<0.05). Difference in Sb amounts shows that sunlight exposure had more impacts compared to room temperature as storage time increased. TSSC values increased steadily leading to hydrolysis of sugar and other chemical ingredients, thus affecting the specific gravity. Acidity impacted by increased reactivity from carbon dioxide present, which reduces pH of the drink. Chemometric assessment such as contamination factor and pollution load index indicate low concentration and no pollution associated. Factor analysis conducted showed that all parameters and storage time were positively interrelated except for pH due to side reaction. Cumulative variance showed high variance (>50). Health risk assessment conducted for adults and children showed that Tolerable Dose Index and Hazard Index were below one, thus indicates no adverse health impact as the values were relatively higher in children compared to adults. Prolong consumption of carbonated drinks stored longer than expected can cause fatigue and headaches on a short-term basis, and weight loss and diabetes on a long run especially in children. Regular parental monitoring is advised to mitigate health impact for children.

This paper has analyzed the functioning conditions for the technological system of secondary condensation (TSSC) in a typical ammonia synthesis unit of the AM-1360 series with the use of a system-control approach. The coordinates of control vectors and external disturbances have been determined. An algorithm has been developed for predicting the coordinates of the control vector for the subsystem of decision support under the conditions of external disturbances for such a complex inertial object with high metal consumption as TSSC. The method of mathematical modeling was used to determine, based on the developed algorithm, the patterns and quantitative dependences of the influence of external disturbances such as the temperature of primary condensation and the flow rate of circulation gas on the efficiency of TSSC heat exchange processes. The regularity of increase in the heat flows and coordinates of control vector with an increase in the temperature of primary condensation has been established. The parametric sensitivity of the coordinates of the control vector under the conditions of change in the temperature of the primary condensation has been determined, which, compared with the circulation gas flow rate, exceeds it by more than six times. The executed software implementation of the algorithm employing the MATLAB programming environment makes it possible, owing to the embedded client part (ORC client), free software access to the current data on the technological process. The functional structure of computer-integrated TSSC technology with the proposed correction subsystem under a supervisory control mode has been designed. Correction solutions involving the additional hardware and software based on the programmable logic controller VIPA and SCADA-system Zenon have been practically implemented. The implementation of the developed system ensures the stabilization of the secondary condensation temperature at the regulatory level of −5 °C, which reduces the consumption of natural gas by almost 1 million nm3 per year.

Sweat glands (SGs) are spread over almost the entire surface of the human body and are essential for thermoregulation. Theoretically, tissue-specific stem cells (TSSCs) are excellent candidate cells for the regeneration of SGs due to their genetic stability and differentiation ability. Herein, we attempted to isolate TSSCs derived from adult human sweat glands (ahSGs). ahSGs were localized and identified by H&E staining, double immunofluorescence staining, transmission electron microscope (TEM), and immuno-TEM. We found a population of cells with stem cell characteristics (SGSCs), located in basal myoepithelial cells of the secretory portion of the solenoid bulb. The SGSCs expressed alpha-smooth muscle actin (α-SMA) and showed the typical characteristics of mesenchymal stem cells (MSCs), with a positive antigen profile for CD44, CD73, CD90, and CD105, and had the multilineage differentiation potential to osteoblasts and adipocytes. In addition, the isolated α-SMA positive cells remained stably phenotypic and proliferative cycles at passage 12. This is the first report of successful isolation of MSC-like cells from ahSGs, which may contribute to wound repair and SG regeneration.
