Purpose The purpose of this paper is to investigate the quality of the frying oil used in restaurants, fast food establishments, and confectionary stores. The compliance of used frying oils with the quality standards as determined by the peroxide value (PV) and the total polar materials (TPMs) is investigated by analyzing 375 samples of oil. Design/methodology/approach The PV was measured according to the national standard procedure number 4179, while the TPM was determined using a Testo 270 cooking oil tester. Frying oils with a PV>5 mEq/kg and a TPM>25 percent were considered to be non-edible. For a comparison of groups, the Mann-Whitney and Spearman correlation tests were used, and p<0.05 was considered significant. Findings The maximum TPM and PV recorded for frying oils in fast food restaurants were 97.5 percent and 77.9 mEq/kg, respectively. The results also revealed that 60 percent of samples were non-edible according to the TPM, while 58.9 percent of the oil samples were non-edible because of the PV. TPM and PV correlated well with each other (r=0.99, p<0.001) and with oil replacement intervals (r=0.90, p<0.001). The relationship between the TPM and PV was stronger in the polynomial model than the linear model. The following equation was obtained: peroxide (mEq/kg oil)=0.0043 TPM2 (%)+0.1587 TPM (%)–0.6152. Originality/value Considering the current limitations in official supervision by health authority, on-site self-monitoring of the TPM using the Testo 270 cooking oil tester by sellers as a solution seems a new approach. Food stores, restaurants, and confectionary stores should be equipped with TPM analyzers to determine the quality of the frying oil and the timely replacement of non-edible oils.

Purpose The purpose of this paper is to present a new operational approach to equipment maintenance designed to exploit the complementary nature of total productive maintenance (TPM) and reliability-centered maintenance (RCM). Design/methodology/approach This paper focuses on the operational aspect of the maintenance practice, proposing an equipment maintenance methodology that can be suitably adopted to define a new maintenance plan or check and improve an existing one. It integrates principles and methods of TPM and RCM and uses different tools to support decision-makers and operators. It proposes several worksheets which facilitate planning and implementing maintenance activities, that range from autonomous maintenance tasks to maintainability improvement actions. Findings This paper exploits the complementary nature of TPM and RCM to design or improve maintenance plans for a generic system. The tools and worksheets presented can be helpful to practitioners and operators. Originality/value While the integration of TPM and RCM is evidently a central issue in the maintenance management context, it has received limited attention in literature. To the best of the authors’ knowledge, it lacks a structured practical equipment maintenance methodology that exploits the complementary nature of TPM and RCM. Therefore, the approach presented in this paper would fill the current gap.

Despite various reported analytical methods for topiramate (TPM) analysis, greener analytical approaches are scarce in literature. As a consequence, the objective of the current research is to design a normal-phase stability-indicating high-performance thin-layer chromatography (SI-HPTLC) methodology for TPM analysis in marketed tablet dosage forms that is rapid, sensitive, and greener. TPM was derivatized densitometrically and analyzed at 423 nm in visible mode with anisaldehyde-sulfuric acid as the derivatizing agent. The greener SI-HPTLC technique was linear in the 30–1200 ng band−1 range. In addition, the suggested SI-HPTLC methodology for TPM analysis was simple, rapid, cheaper, precise, robust, sensitive, and environmentally friendly. The greener SI-HPTLC method was able to detect TPM along with its degradation products under acid, base, and oxidative degradation conditions. However, no TPM degradation was recorded under thermal and photolytic stress conditions. TPM contents in commercial tablet dosage forms were recorded as 99.14%. Using 12 different principles of green analytical chemistry, the overall analytical GREEnness (AGREE) score for the greener SI-HPTLC method was calculated to be 0.76, confirming the proposed normal-phase SI-HPTLC method’s good greener nature. Overall, these results demonstrated that the suggested SI-HPTLC technique for TPM measurement in pharmaceutical products was reliable and selective.

Purpose – The purpose of this paper is to review the literature on total productive maintenance (TPM) implementation practice to present an overview of TPM implementation practices adopted by various manufacturing organizations and suggest possible gaps from researchers and practitioner’s point of view. This study tries to identify the best strategy for improving competitiveness of small and medium enterprises (SMEs) in globalized market and evaluates TPM implementation practice in SMEs. Design/methodology/approach – The objective of this paper is to study the role of TPM program in context of Indian industries either from SMEs to large-scale industries. The approach has been directed toward justification of TPM implementation practice for its support to competitive manufacturing in the context of Indian manufacturing industries. Findings – TPM implementation improves productivity and working efficiency of employees and also improves equipment effectiveness and a positive inclination toward company is registered. Therefore, equipment maintenance is an indispensable function in a manufacturing enterprise. In this highly competitive environment, manufacturing organizations should consider maintenance function as a potential source for cost savings and competitive advantage. SMEs must be considered as an engine for economic growth all over the world (Singh et al., 2008). A total of 148 papers related to TPM implementation in large-scale industries and SMEs were collected, analyzed and classified on the basis of their applications, broadly into large industry and SMEs and further divided into Indian and Non-Indian, then case study, implementation, model, literature review, maintenance, service, etc. This classification has clearly shows that SMEs are in a need to adopt TPM implementation practice to compete in this global market and changing requirements of large industries. Maintenance is an indispensable function in a manufacturing enterprise. In this highly competitive environment, manufacturing organizations must be considered maintenance function as a potential source for cost savings and competitive advantage. Singh et al. (2008) have concluded that SMEs must be considered as an engine for economic growth of all over the world. Following are the objectives of this study: to suggest a classification of available literature on TPM implementation; to identify the need of TPM implementation in SMEs; to identify critical observations on each category of classification; to identify the potential of SMEs in India; to identify emerging trends of TPM implementation in India; to suggest directions for future researchers in the field of TPM implementation on the basis of above mention points; and to consolidate all available literature on TPM implementation practice. Research limitations/implications – The challenges of stiff competition and the drive for profits are forcing the organizations to implement various productivity improvement efforts to meet the challenges posed by ever-changing market demands. In the dynamic and highly challenging environment, reliable manufacturing equipment is regarded as the major contributor to the performance and profitability of manufacturing systems. Practical implications – In this dynamic world, importance of SMEs in the growth of the nation needs more attention of researchers and industrialists. After the globalization of market, SMEs have got many opportunities to work in integration with large-scale organizations. All the organizations from SMEs to large-scale industries can adopt effective and efficient maintenance strategies such as condition-based maintenance, reliability-centered maintenance and TPM over the traditional firefighting reactive maintenance approaches (Sharma et al., 2005). Social implications – In our view, this paper clearly identifies implications for research which will be useful for society. The gap discussed by authors needs to be addressed by future researchers. Originality/value – This implementation strategy can help to save huge amounts of time, money and other useful resources in dealing with reliability, availability, maintainability and performance issues. On the basis of available literature, it can be understood that SMEs should change their maintenance strategies to cope up with global competition so that a lot of resources can be utilized in a better direction.

Production management requires an effective and efficient maintenance management system. One approach to improve the performance of the maintenance system is through the implementation of Total Productive Maintenance (TPM). The key performance index to measure the effectiveness of TPM implementation is Overall Equipment Effectiveness (OEE). The TPM activities will eliminate equipment losses related to availability, performance rate, and quality rate. Hence, the TPM implementation will increase the OEE value. This paper outlines the theories of TPM and OEE. The paper also presents the studies carried out by different authors to show the improvement of OEE through implementation of TPM in manufacturing, such as electronic industry, steel manufacturer, as well as locomotive components manufacturer.

A precise approach for the tooth profile modification (TPM) of helical gear is presented in the paper based on the static contact finite element analysis (FEA). The high-precision finite element model of helical meshing gear pairs is established. The type and amount of TPM are accurately determined by the static contact FEA results. The dynamic contact simulations of helical gears with and without tooth modification are investigated to estimate the vibration reduction effect of the TPM. Moreover the numerical simulations are compared with the experimental results. Both results show that the proposed precise TPM of helical gears is effective on vibration reduction around the working load, and the dynamic contact simulation is effective on estimating the vibration reduction influence of the TPM.

Purpose The purpose of this paper is to comparatively assess the contributions of total quality management (TQM) and total productive maintenance (TPM) practices, exclusively and collectively, on manufacturing business performance. Design/methodology/approach Responses to survey questionnaires were collected using multiple modes of data collection. A total of 269 filled responses were collected and analyzed to ascertain the contribution made by exclusive and collective implementation of TQM and TPM practices on business performance. The respondent firms were grouped into three clusters based on early, in-transition and advanced stages of implementation of either or both TQM and TPM manufacturing practices. Comparative assessment of the impact of standalone TQM, standalone TPM and combined TQM–TPM manufacturing methods on business performance parameters have been evaluated using statistical tools within each cluster. Findings Evidences were found to support the supposition that integrated implementation of both TQM and TPM practices over a reasonable period of time contributes in delivering better business performance as compared to the standalone implementation strategy. Research limitations/implications The study has its own limitations because of its chosen research approach. As the data in this study were collected from production management personnel on the basis of their subjective evaluations, more comprehensive objective performance indicators shall be employed for comparative assessment of manufacturing practices in future studies. In-depth case studies are needed to further validate the findings of the research study. Practical implications The analytical results reveal that an integrated TQM–TPM strategic implementation can systematically contribute toward realization of significant improvement in manufacturing business performance parameters. The results of this study are vital for production managers and senior management personnel of manufacturing firms that have implemented TQM and are considering introducing TPM in their organization (or vice-versa). Originality/value This empirical study signifies the need for strategically aligning and managing various production improvement ideologies for realization of an organization’s growth and sustainability in a long run. In particular, this paper invigorates the discussion about improving the synergy between maintenance and quality functions in the organization for meeting the challenges of highly competitive market environment.

Purpose This study aims to introduce transformative place management – TPM – (defined as the deliberate efforts of place managers in commercial settings to provide a pool of restorative resources to improve the consumers’ emotional well-being) by merging the REPLACE framework and transformative service research. Additionally, this research analyzes the direct and indirect impacts of restorative resources as a form of TPM on consumers’ emotional well-being and place attachment, considering the moderating role of employee emotional labor. Design/methodology/approach A total of 240 customers were surveyed in an experience-based store in a developing country by using a questionnaire. Then, a moderated mediation model was applied to analyze the moderating role of employee emotional labor in the relationship between TPM and place attachment through consumers’ well-being. Findings TPM that provides restorative resources to consumers influences place attachment by improving consumer well-being. However, surface acting by employees reduces the ability of TPM to increase place attachment through the improvement of consumers’ emotional well-being. Deep acting, on the other hand, does not enhance the effect of TPM on place attachment through consumers’ emotional well-being. Originality/value This paper proposes new developments in the transformative service research (TSR) paradigm by introducing TPM. By showing how the place of consumption increases the well-being of customers, this paper helps TSR researchers to accomplish the purpose of transforming the lives of consumers through relevant research. Although marketing researchers and environmental psychologists have theoretically anticipated the positive effects on well-being from consumption settings, this paper explains how commercial places promote customer well-being through the provision of restorative resources. Also, this paper shows how the place of consumption transforms consumers’' lives and identifies some of the boundary conditions at which such a transformation occurs.

Quality has become one of the most crucial criteria in an institution’s success and survival as there is nothing more than what an era of globalization and intensity demands. Successful businesses recognize that consumer reliability may have a severe influence on their bottom lines. As a result, several competitive companies are constantly raising their quality requirements. Competitive companies think that improving quality is the best way to recover, and most authors have specified various procedures relevant to their processes. The majority of automobile assembly sectors are looking for high-quality requirements in their manufacturing techniques and are executing a quality system known as total productive maintenance (TPM). The study’s goal is to deploy the TPM program inside the metal forming industry to improve metal industry workstations. The overall equipment effectiveness (OEE) for various workstations such as rolling, bending, cutting, and die punching for the fiscal year 2018–2019 has been evaluated. In addition to the other reasons, inefficient resource utilization is a significant component that diminishes the factory’s OEE. In the financial year 2019–2020, the TPM approach was adopted in the enterprise. As a result, there has been an improvement in overall performance.

Purpose The purpose of this paper is to examine the multidimensionality of total productive maintenance (TPM) and its relationship with manufacturing performance improvement in the Malaysian manufacturing sector. Specifically, this study evaluates the contribution of each TPM success factors in improving manufacturing performance. Design/methodology/approach Data from 89 employees who participated in the survey were used to test the proposed research framework. A structured questionnaire adopted from Ahuja and Khamba (2006) was used to assess the Malaysian context. Findings The analytical results reveal that traditional maintenance initiatives and TPM implementation initiatives significantly affect manufacturing performance, but not top management leadership and maintenance organisation. Top management roles and commitment are critical in the early stage to determine the master plan and initiate the implementation of the whole programme. However, traditional maintenance and TPM implementation initiatives gradually enable engagement, proper planning, right execution and continuous improvement, ultimately improving the manufacturing performance indicators significantly. The findings further unveil that TPM is not sustainable in Malaysia’s manufacturing organisations in the long run. Practical implications This analysis is vital for senior managers of manufacturing organisations that have implemented TPM or are considering introducing TPM in their organisations. Originality/value This study contributes to the literature by examining beyond the introduction and stabilisation phase of TPM to provide an insight of whether TPM is sustainable in the long run.

The purpose is the detection of changes in the design and operation of the storage part of blast furnaces, due to i mprovements in smelting technology, as well as increasing power and specific productivity of units. The methodology based on the use of a new approach to assessing the technological capabilities of the furnace and storage part of blast furnaces in the study of design and practical information on the profiling of furnaces over a period of time characterized by dynamic development of units and cast iron smelting technology. Findings The proposed characteristics of the technological capabilities of the furnace and the storage part of the furnace, as well as criteria for assessing these characteristics - the share of the storage part per air lance V_nch / n_f, the power of one air lance and the relative to the countercurrent part of the furnace / V_pr. It is shown that in the design and operation of blast furnaces it is desirable to maintain the criteria V_nch / n_f and V_nch / n_chl at the lowest possible level, and the criteria of air lance power and V_zum / V_pr at the maximum. There is a tendency to slow down the height of the furnace with increasing the useful volume of the furnaces. In modern furnaces, for every additional 1000 m3 of usable volume, the height of the furnace increased by an average of 9%. It is shown that the outstripping increase in the depth of the sump in comparison with other dimensions of the profile is due not only to its structural function, but also purely technological, namely the sump cast iron heat, which increases the inertia of the blast furnace process, and also participates in the cycle of accumulation and release of smelting products. Since the sump is part of the working space of the furnace, it is proposed to change the approach to determining the complete and useful height of the blast furnace. More specifically, the full height should be considered the distance from the ladder to the upper edge of the support ring (flange) of the grate cover, and useful - to the cone or tray of the backfill device in the lower working position. The study’s originality is to justify the new principles of the approach to assessing the technological capabilities of the furnace and storage part of blast furnaces, as well as to justify the need to consider the sump or dead layer as the sixth element of the profile. The practical value is to develop recommendations on the direction of changes in the design of the furnace and storage part of blast furnaces in the design of new and modernization of existing units, as well as a new approach to determining the useful and complete height of the furnace profile.

Purpose The purpose of this paper is to promote a system dynamics-discrete event simulation (SD-DES) hybrid modelling framework, one that is useful for investigating problems comprising multifaceted elements which interact and evolve over time, such as is found in TPM. Design/methodology/approach The hybrid modelling framework commences with system observation using field notes which culminate in model conceptualization to structure the problem. Thereafter, an SD-DEShybrid model is designed for the system, and simulated to proffer improvement programmes. The hybrid model emphasises the interactions between key constructs relating to the system, feedback structures and process flow concepts that are the hallmarks of many problems in production. The modelling framework is applied to the TPM operations of a bottling plant where sub-optimal TPM performance was affecting throughput performance. Findings Simulation results for the case study show that intangible human factors such as worker motivation do not significantly affect TPM performance. What is most critical is ensuring full compliance to routine and scheduled maintenance tasks and coordinating the latter to align with rate of machine defect creation. Research limitations/implications The framework was developed with completeness, generality and reuse in view. It remains to be applied to a wide variety of TPM and non-TPM-related problems. Practical implications The developed hybrid model is scalable and can fit into an existing discrete event simulation model of a production system. The case study findings indicate where TPM managers should focus their efforts. Originality/value The investigation of TPM using SD-DES hybrid modelling is a novelty.

Purpose – World-class total productive maintenance (TPM) status depends on critical success factors (CSF) to be properly implemented. The relevant literature has identified several CSF; however, researchers have not investigated the degree of importance of each phase of the process. The purpose of this paper is to prioritize the CSFs that influence TPM implementation during each phase of the process in order to aid decision making in companies. Design/methodology/approach – The main CSF are identified from a literature review. This paper focusses on the judgments of managers who are experts in TPM implementation in companies which have received TPM awards for excellence. The judgments are processed using the analytic hierarchy process (AHP) method. Three aggregation methods (geometric mean, Copeland and fuzzy) are employed for comparative analysis. Findings – Although the literature highlights some important CSF, the relevance of these factors is not the same in each phase of TPM implementation. Analysis shows that ranking using the Copeland method is most coherent. Research limitations/implications – This research was carried out in some Brazilian companies. Practical implications – If managers are aware of the CSF to be prioritized in each phase, this facilitates decision making, resulting in a more effective implementation of TPM. Social implications – If TPM implementation is effective, this stimulates the personnel involved and avoids the possibility of the program being abandoned. Originality/value – The originality of this paper consists in the proposed model to prioritize CSF during each phase of the implementation of TPM.

Purpose – The purpose of this paper is to highlight the application of Interpretive Structural Modeling (ISM) to analyze the barriers in implementation of Total Productive Maintenance (TPM). TPM is explained in brief with emphasis on maintenance programs to improve quality of products, reliability of processes and reduction in cost. Barriers in implementation of TPM are also discussed. Concept of ISM and steps in developing ISM are described in detail. The authors then illustrate the research methodology which involves applying ISM to analyze barriers in TPM. Design/methodology/approach – The paper starts off by describing the concepts of TPM and ISM. Barriers in implementation of TPM are discussed. It explains ISM as a methodology to understand the underlying interrelationship among the inhibiting factors. The authors draw up an action plan to carry out research on the usage of ISM to study the TPM inhibitors, to develop an integrated model to establish the relationship among the different TPM inhibiting factors and to suggest action plan to mitigate these factors. Findings – Interpretive Structural Modeling (ISM) can be used to analyze the driving and dependence power of the variables inhibiting implementation of TPM. The barriers to implement TPM are described with detailed explanation. The complexity of the problem and the degree of interconnection among the variables can be found out. This will help Managers take action on mitigating the barriers. Practical implications – By analyzing the interrelationships among the barriers and their strengths, management can chalk out the strategy to implement TPM in an organization. Management will become aware of the barriers which have the maximum influence and then can act accordingly to mitigate these barriers. This will help in implementing TPM faster and in an organized manner. Originality/value – Many authors have used ISM to study various issues. A couple of authors have used ISM to determine barriers in implementation of TPM. The authors feel that most of the papers describe ISM in brief making it slightly difficult for readers to understand. This paper aims to explain elaborately step-by-step on how to develop an ISM making it easier for researchers to understand the ISM concept. Even though there are papers on TPM and difficulties in implementation of TPM, this paper explains the barriers in implementing TPM based on the experience of the corresponding author having worked in the refinery industry.

Streszczenie:
A systematic method of determining the design parameter values of two-point mooring (TPM) systems is presented. The conclusions of nonlinear stability analysis of horizontal plane slow motion dynamics of TPM systems with hydrodynamic memory are used to guide selection of the design variables. Specifically, within the stable regions of bifurcation diagram, a set of parameter values is chosen systematically to examine the corresponding TPM system. Then, each TPM system is simulated and the results are evaluated in terms of given criteria such as tension level of the mooring line or offset of the moored vessel. The hydrodynamic memory effect due to the oscillatory motions of moored vessel is included in the formulation. The memory effect influences the stability properties of TPM systems and alters the design chart/bifurcation diagram and particularly the static loss and dynamic loss of stability. The significance of hydrodynamic memory on several TPM systems is illustrated by comparing the statistics of the simulations. The design approach implemented in this work shows that nonlinear stability theory can be used effectively in the actual design process of mooring systems to reduce dramatically the number of simulations required. It is also shown that the often inconclusive nonlinear time simulations can be guided by theoretical nonlinear stability analysis of system dynamics to produce improved design values of system parameters of TPM systems.

Streszczenie:
Purpose: This paper develops a ‘light’ total productive maintenance (TPM) model suitable for small and medium-sized enterprises (SMEs). By design, the system is rudimentary, using a relatively small sum of capital investment and resources. The model recommends TPM implementation in three stages, namely plan, improve, and sustain.Design/methodology/approach: The literature review provides the inputs to the model development. Action research is used to demonstrate and verify the effectiveness and practicability of the framework, in an SME manufacturing hydraulic parts in China. Overall Equipment Effectiveness (OEE) and awareness of employees were studied before and after the implementation. Findings: The case study shows a significantly improved production efficiency of the equipment. The framework structuralizes TPM deployment and binding different levels of the organization into the program, from planning, implementation to sustaining the practices. To break the barrier of shop-floor resistance, the leader must drive many activities unassisted, it, therefore, necessitates an open endorsement of authority by the steering committee composed of top management. The Prudent pilot run of TPM helped to accelerate the implementation at critical equipment, in addition to cultivating experience and hence confidence among staff.Research limitations/implications: This study provides a pragmatic reference to other researchers and practitioners to promote a light TPM model in SMEs, without losing the essence of TPM. Being action research with the case study in a specific manufacturing industry, the resultant evidence, therefore, is anecdotal.Originality/value: The model adopts a phased method to implement TPM, without aggravating the financial and human resource burden of the enterprise. It promotes the cultivation of employees’ TPM awareness and active involvement, which can lay a solid foundation for the wide implementation of TPM in SMEs.

Mobile agent technology is an active research topic and has found its uses in various diverse areas ranging from simple personal assistance to complex distributed big data systems. Its usage permits offline and autonomous execution as compared to classical distributed systems. The free roaming nature of agents makes it prone to several security threats during its transit state, with an added overhead in its interoperability among different types of platforms. To address these problems, both software and hardware based approaches have been proposed to ensure protection at various transit points. However, these approaches do not ensure interoperability and protection to agents during transit over a channel, simultaneously. In this regard, an agent requires a trustworthy, interoperable, and adaptive protocol for secure migration. In this paper, to answer these research issues, we first analyse security flaws in existing agent protection frameworks. Second, we implemented a novel migration architecture which is: 1) fully inter-operable compliance to the Foundation for Intelligent Physical Agents (FIPA) and 2) trustworthy based on Computing Trusted Platform Module (TPM). The proposed approach is validated by testing on software TPM of IBM, JSR321, and jTPMTools as TPM and Trusted Computing Software Stack (TSS) interfaces, JADE-agent framework and 7Mobility Service (JIPMS). Validation is also performed on systems bearing physical TPM-chips. Moreover, some packages of JIPMS are also modified by embedding our proposed approach into their functions. Our performance results show that our approach merely adds an execution overhead during the binding and unbinding phases

Purpose The purpose of this paper is to determine the relationship between total productive maintenance (TPM), kaizen event (KE) and innovation performance (IP) for Malaysian automotive industry using structural equation modeling (SEM). Design/methodology/approach The samples were selected from the list of Proton and Perodua automotive industry. The number of collected respondents was 238 respondents. An SEM technique was used in the study. In order to test the reliability and validity of the instrument, reliability analysis, exploratory and confirmatory factor analysis were conducted. Findings Based on the results, KE does not affect the relationship between TPM and IP. However, the impact of TPM on IP increases with mediating of KE for Malaysian automotive industry. Thus, this study has shown that empirical test results prove that the implementation of TPM and KE has improved the IP for Malaysian automotive industry. Research limitations/implications This study only focused on the Malaysian automotive industry. The other limitation in this research is the number of factors and limited measurement in this study. Only a few TPM, KE and IP measurements were considered. By using the SEM technique, four TPM constructs, three for KE constructs and three for IP measures were developed and verified. Therefore, this study can assist the researchers and practitioners to the practice of TPM, KE and IP for Malaysian automotive industry. Originality/value This research provides fundamental knowledge and direction for researchers in further research as well as practitioners to constantly improve IP through the implementation of TPM and KE for Malaysian automotive industry.

Streszczenie:
Recent proteomics studies of vertebrate striated muscle have identified lysine acetylation at several sites on actin. Acetylation is a reversible post-translational modification that neutralizes lysine's positive charge. Positively charged residues on actin, particularly Lys326 and Lys328, are predicted to form critical electrostatic interactions with tropomyosin (Tpm) that promote its binding to filamentous (F)-actin and bias Tpm to an azimuthal location where it impedes myosin attachment. The troponin (Tn) complex also influences Tpm's position along F-actin as a function of Ca2+ to regulate exposure of myosin-binding sites and, thus, myosin cross-bridge recruitment and force production. Interestingly, Lys326 and Lys328 are among the documented acetylated residues. Using an acetic anhydride-based labeling approach, we showed that excessive, nonspecific actin acetylation did not disrupt characteristic F-actin–Tpm binding. However, it significantly reduced Tpm-mediated inhibition of myosin attachment, as reflected by increased F-actin–Tpm motility that persisted in the presence of Tn and submaximal Ca2+. Furthermore, decreasing the extent of chemical acetylation, to presumptively target highly reactive Lys326 and Lys328, also resulted in less inhibited F-actin–Tpm, implying that modifying only these residues influences Tpm's location and, potentially, thin filament regulation. To unequivocally determine the residue-specific consequences of acetylation on Tn–Tpm–based regulation of actomyosin activity, we assessed the effects of K326Q and K328Q acetyl (Ac)-mimetic actin on Ca2+-dependent, in vitro motility parameters of reconstituted thin filaments (RTFs). Incorporation of K328Q actin significantly enhanced Ca2+ sensitivity of RTF activation relative to control. Together, our findings suggest that actin acetylation, especially Lys328, modulates muscle contraction via disrupting inhibitory Tpm positioning.
