Resumo: Estudos sugerem que a exposição pré-natal à testosterona (PT) está relacionada à inteligência ou à capacidade de aprendizado. A proporção entre os dedos indicador e anelar (proporção digital ou 2D:4D) correlaciona-se negativamente à exposição pré-natal à testosterona. Este estudo avaliou as correlações entre o 2D:4D e o sucesso acadêmico em disciplinas teóricas e práticas no currículo do curso de Odontologia da Faculdade de Odontologia de Piracicaba da Universidade Estadual de Campinas, Brasil. Ao todo, 80 indivíduos (40 homens e 40 mulheres) com idade entre 18 e 22 anos (média 19,5 ± 1,1 anos para os homens e 19,7 ± 1,2 para mulheres), alunos do 2º ao 4º ano de graduação tiveram sua mão direita fotografada por meio de câmera digital acoplada a dispositivo padronizador perpendicular à superfície. As imagens foram transferidas ao software Adobe Photoshop® e os dedos indicador e anelar foram medidos de forma linear, da depressão mais caudal ao ponto mais rostral da ponta do dedo. Os dados foram analisados usando os softwares Microsoft Excel, SPSS 16 e Statview. A proporção digital média foi de 0,980 ± 0,038 para homens e 0,991 ± 0,025 para mulheres. Correlacionou-se então esta proporção às notas teóricas e práticas obtidas pelos estudantes durante os primeiros quatro semestres letivos do curso. As notas teóricas e práticas foram negativamente correlacionadas à proporção digital em homens (especialmente após a remoção da influência das horas de estudo domésticas; p = 0,02 e 0,004, respectivamente), mas não em mulheres (p = 0,89 e 0,77, respectivamente). Este achado apóia a correlação entre a influência da testosterona pré-natal e o desenvolvimento da inteligência em homens. A falta de correlação entre a testosterona pré-natal e o sucesso acadêmico em mulheres sugere que a inteligência no sexo feminino deve ser influenciada por fatores alheios à testosterona pré-natal
Abstract: It has been suggested that prenatal testosterone (PT) is positively related to intelligence or learning-ability skills. Digit ratio (2D:4D) is a negative correlate of PT. This study considered the correlations between 2D:4D and academic success in practical and theoretical disciplines in the Dental School curriculum of the Piracicaba Dental School, State University of Campinas, Brazil. Overall, 80 subjects (40 males and 40 females) aged between 18 and 22 years (means 19.5 ± 1.1 and 19.7 ± 1.2, respectively) had their right hand palm photographed by a digital camera attached to a standardising device, perpendicular to the surface. The images were transferred to a computer and the index and ring fingers were measured from the ventralmost crease up to the tip of the finger using Adobe Photoshop. Data were analyzed using Microsoft Excel and SPSS 16. Mean digit ratio was 0.980 ± 0.038 for males and 0.991 ± 0.025 for females. Digit ratio was correlated to the grades obtained by the students through their first four semesters. Theoretical and practical grades were significantly negatively correlated to digit ratio in males (and this was particularly so after the influence of age and hours of study were removed, p = 0.02 and 0.004, respectively), but not in females (p = 0.89 and 0.77, respectively). This finding supports a link between high PT and intelligence in males. The lack of relationship between 2D:4D and examination marks in female students, suggests that intelligence in females may be influenced by factors other than prenatal testosterone

Resumo: Objetivo: avaliar a eficácia relativa do leuprolide 3,75 mg, leuprolide 7,5 mg e goserelina 3,6 mg quanto à redução da testosterona sérica, em níveis de castração. Método: foram avaliados 60 pacientes com carcinoma avançado de próstata, com indicação de bloqueio hormonal. Os pacientes foram divididos em 3 grupos de 20: Grupo 1) pacientes que receberam leuprolide na dose 3,75 mg; Grupo 2) receberam leuprolide 7,5 mg, Grupo 3) receberam goserelina 3,6 mg. Todos os grupos foram tratados por aplicação mensal das respectivas medicações. Os pacientes foram avaliados com dosagens de testosterona sérica em 2 momentos: pré-tratamento e após 3 meses de tratamento. Resultados: a idade dos pacientes foi semelhante nos três grupos, com mediana de 72, 70 e 70 anos, respectivamente aos grupos 1, 2 e 3. Dos pacientes que receberam leuprolide 3,75 mg, leuprolide 7,5 mg e goserelina 3,6 mg, 26,3%, 25% e 35%, respectivamente, não obtiveram nível de castração, considerando nível de corte de testosterona ? 50 ng/dl; e 68,4%, 30% e 45%, respectivamente, não obtiveram nível de castração, considerando nível de corte de testosterona ? 20 ng/dl. Conclusão: não houve diferença significativa na obtenção de níveis de castração com leuprolide 3,75 mg, leuprolide 7,5 mg e goserelina 3,6 mg, quando comparadas as três substâncias em conjunto. Quando comparados os grupos dois a dois, houve diferença estatisticamente significativa na análise do leuprolide 3,75 mg com o leuprolide 7,5 mg, havendo maior obtenção de nível de castração com 7,5 mg, quando se considera este nível como igual ou menor que 20 ng/dl. A importância dessa eventual diferença, no entanto, deve ser vista com cautela, já que a comparação dos três grupos simultaneamente, muito embora próximo, não atingiu o nível de significância estabelecido
Abstract: Purpose: to evaluate the relative efficiency of leuprolide 3.75 mg, leuprolide 7.5 mg, and goserelin 3.6 mg in relation to the reduction of serum testosterone, regarding the levels of castration. Methods: we evaluated prospectively 60 randomized patients with advanced prostate carcinoma, with indication for hormone blockade. The patients were divided into 3 groups of 20: Group 1) received leuprolide 3.75 mg; Group 2) received leuprolide 7.5 mg; group 3) received goserelin 3.6 mg. All groups were treated with monthly application of the respective drugs. Results: the patients' ages were similar within the three groups, with a median of 72, 70, and 70 in groups 1, 2, and 3, respectively. Of the patients that received leuprolide 3.75 mg, leuprolide 7.5 mg, and goserelin 3.6 mg, 26.3%, 25%, and 35%, respectively, did not reach castration levels, considering a testosterone cut of ?50 ng/dl. 68.4%, 30%, and 45%, respectively, did not reach castration levels, considering a testosterone cut of ?20 ng/dl. Conclusions: There were no statiscally significant differences in the levels of castration when comparing with leuprolide 3.75 mg, leuprolide 7.5 mg, and goserelin 3.6 mg, when comparing the three substances altogether. When compared in groups of two, there was a statiscally significant difference between leuprolide 3.75 mg and leuprolide 7.5 mg, in which the latter presented better results in reaching castration levels, when considered equal or less than 20ng/dl. The importance of this difference, however, must be measured with caution, since the comparison of the three groups simultaneously did not reach the established significance level, even though it came close

Pełny tekst źródłaThe aim of this dissertation was to investigate the effect of territoriality and the level of opponent on hormonal and behavioral responses in basketball players. Two studies were conducted. In the first one, in order to investigate the effect of territoriality on hormonal and behavioral responses from official basketball matches, eighteen male players, from two under-19 teams (ranked matches in first and second place in the competition) were investigated. The teams played against each other, so each team performed a game \"at home\" and \"away from home\" game. Saliva samples were collected before and after the two matches for hormone analysis [testosterone (T) and cortisol (C)]. Before warm-up, a pre-competitive anxiety questionnaire was answered (Competitive State Anxiety Inventory-2, CSAI-2), and 30 minutes after the matches, the athletes classified the magnitude of effort by means of session-RPE. It was observed a higher value for T PRE game during \"at home\" condition when compared to \"away from home\"; there was a rise in T and C concentrations from moment PRE to POST in both conditions (\"at home\" and \"away\"); significant correlations were observed between hormonal and behavioral responses. These results show the occurrence of territoriality phenomenon, demonstrated by the higher T concentration during \"at home\" PRE game moment, and that official competitive condition is a stressor event capable of altering hormonal concentration. In the second study, the purpose was to verify the influence of the level of the opponent on hormonal and behavioral responses in basketball players. Twelve athletes from an under-19 team were evaluated in three different official matches and one training session (TS). Each official match was played against an opponent with different level of quality: weak match (WM), medium match (MM) and strong match (SM). The classification of the level of the opponent was made by a member of the coaching staff at the beginning of the competitive season and monitored in accordance with the position of the teams during the championship. Saliva samples were collected before and after the four conditions (three games and a training session) for T and C analyzing. Before warm-up, players answered the CSAI-2 questionnaire and approximately 30 minutes after the game, answered session-RPE. It was demonstrated an increase in T concentration from PRE to POST moments in response to TS and SM conditions and T concentration during PRE-SM was higher when compared to PRE-TS condition. The concentration of C increased from PRE to POST moments during TS, MM, and SM and C concentration during PRE and POST-SM were higher compared to the same moments for TS condition. Athletes reported higher values of somatic and cognitive anxieties in MM and SM when compared to WM condition. Self-confidence values were greater during TS compared to MM and SM. Session-RPE presented higher scores in MM and SM compared to TS. The results of the present investigation indicate that official games against opponents of different levels may promote hormonal and perceptual responses of distinct magnitudes

Pełny tekst źródłaLas guías clínicas del cáncer de próstata (CaP) recomiendan la medida de la concentración de testosterona para evaluar la eficacia de la castración y para diagnosticar la resistencia a la castración cuando se observa una progresión bioquímica o clínica. La respuesta a los análogos de la hormona liberadora de la hormona luteinizante (LH-RH) se considera eficaz cuando la concentración de testosterona se mantiene <50 ng/dL. Sin embargo, esta concentración llamada de castración fue definida en los años ochenta en pacientes sometidos a castración quirúrgica, siendo el límite inferior de sensibilidad del radioinmunoanálisis utilizados entonces. En el seguimiento de estos pacientes la concentración de testosterona es especialmente baja y es necesario utilizar procedimientos de medida exactos y reproducibles, para verificar la eficacia del tratamiento y poder definir la resistencia a la castración. Los inmunoanálisis han sido una herramienta fundamental para la medida de los esteroides sexuales. Sin embargo, los últimos años se ha demostrado que estos no tienen suficiente exactitud, precisión y reproducibilidad para medir concentraciones inferiores al límite inferior del intervalo de referencia. El desarrollo de procedimientos para la medida de la concentración de testosterona mediante técnicas de espectrometría de masas acoplada a técnicas de separación de cromatografía permite medir bajas concentraciones de testosterona con elevada sensibilidad y especificidad. Nuestra hipótesis inicial es que, en pacientes con CaP sometidos a castración médica, la medida de la testosterona con cromatografía líquida y detección por espectrometría de masas es más precisa que cuando se mide con los métodos de quimioluminiscencia para evaluar la eficacia del tratamiento y definir el estado de resistencia a la castración. Como hipótesis adicional nos hemos planteado que la medida de la concentración de LH es un método más específico para evaluar la actividad de la castración médica. Con la selección de pacientes y recogida prospectiva de datos entre los 2016 y 2018 se reclutaron 1510 pacientes con CaP en tratamiento con análogos de la LHRH y se seleccionó un grupo de pacientes control elegidos al azar de los hombres que tenían programada biopsia de próstata debido a la sospecha de CaP. En las bases de datos se recogieron datos referentes al diagnóstico y tratamiento, datos demográficos y resultados de diferentes pruebas analíticas: LH, FSH, testosterona, testosterona libre, PSA, SHBG y prolactina. Se hicieron diferentes subgrupos para estudiar la concentración de testosterona con diferentes procedimientos de medida y valorar su evolución al tratamiento. Se hicieron dos subgrupos de 249 y 143 pacientes respectivamente con resultado de testosterona medida con el equipo Advia Centaur®XP <100 ng/dL, y un subgrupo de 1091 pacientes con concentración de testosterona medida con el equipo Advia Centaur®XP < 150 ng/dL. Se seleccionaron 138 pacientes que presentaban concentración de LH <1,1 U/L. De las revisiones, comparaciones y valoraciones realizadas, se concluye que los diferentes métodos para medir la concentración de testosterona, los más utilizados son los que utilizan la quimioluminiscencia como principio de medida. El método de referencia para su medida en pacientes con CaP sometidos a supresión androgénica debe ser la LC-MSMS debido que los métodos quimioluminiscentes presentan diferencias de reproducibilidad, especificidad, sensibilidad y elevada variabilidad en concentraciones <50 ng/dL mientras que LC-MSMS presenta buenas características metrológicas. La concentración de castración con este método es <50 ng/dL. Y encontramos que la concentración de LH es mejor marcador que la testosterona para evaluar el correcto funcionamiento del tratamiento. Por lo tanto, proponemos la concentración de 1,1 U/L como concentración de castración y una concentración de LH de 0,12U/L la que nos permite diferenciar la castración óptima de la subóptima.
Prostate cancer (CaP) clinical guidelines recommend measuring testosterone concentration to evaluate the effectiveness of castration and detected observed. The response to luteinizing hormone-releasing hormone (LH-RH) analogs is considered effective when testosterone concentration is <50 ng/dL. However, this castration concentration was defined in the 1980s in patients undergoing surgical castration, a level established by using the lower limit of sensitivity of radioimmunoassay method. In these patients the testosterone concentration is especially low and it is necessary to use accurate and reproducible measurement procedures, to verify the effectiveness of the treatment and define the resistance to castration. Immunoassays have been a key tool in measuring sex steroids. However, several recent studies agree that these have low accuracy, precision and reproducibility to measure concentrations below the lower limit of the reference range. The development of procedures for the measurement of testosterone concentration using mass spectrometry techniques coupled with chromatography separation techniques allows us to measure low testosterone concentrations with high sensitivity and specificity. Our initial hypothesis is that in patients with CaP undergoing medical castration, the measurement of testosterone by liquid chromatography and detection by mass spectrometry is more accurate than when measured with chemiluminescence methods to evaluate adequate castration levels and define the state of resistance to castration. As an additional hypothesis, we have considered that the measurement of LH concentration is a more specific method for evaluating the activity of medical castration. With the selection of patients and prospective data collection between 2016 and 2018, 1510 patients with CaP in treatment with LHRH analogues were recruited and a group of randomly selected control patients from men who had scheduled biopsy of prostate to suspicion of CaP was selected. Data relating to diagnosis and treatment, demographic data, and results of different analytical tests were collected from the databases: LH, FSH, testosterone, free testosterone, PSA, SHBG, and prolactin. Different subgroups were made to study testosterone concentration with different measurement procedures and to assess its evolution to treatment. Two subgroups of 249 and 143 patients with testosterone results measured with the Advia Centaur®XP <100 ng/dL, and a subgroup of 1091 patients with testosterone levels measured with the Advia Centaur®XP <150 ng/dL were selected. 138 patients with a concentration of LH <1.1U/L were selected. From the reviews, comparisons and assessments performed, it is concluded that of the different methods for measuring testosterone, the most used useful are those that use chemiluminescence as a principle of measurement. The reference method for its measurement in patients with CaP undergoing castration should be LC-MSMS because chemiluminescent methods show differences in reproducibility, specificity, sensitivity and high variability at concentrations <50ng/dL while LC- MSMS presents good metrological characteristics. The castration concentration with this method is <50 ng/dL. Moreover, we find that LH concentration is more accurate than testosterone for assessing the LH-RH agonist efficacy. Therefore, we propose the concentration of 1.1 U/L as a castration concentration and an LH concentration of 0.12 U/L which allows us to differentiate the optimal castration from the suboptimal one.

Os objetivos do presente estudo foram investigar as adaptações endócrinometabólicas em ratos submetidos ao exercício físico de natação associado com a administração de testosterona durante 8 semanas. Ratos jovens Wistar foram divididos em 4 grupos: controle sedentário (CS), controle treinado (CT), testosterona sedentário (TS), testosterona treinado (TT). O protocolo de natação consistiu de 1 hora/dia, 5 dias/semanas, durante 8 semanas, suportando durante a natação uma sobrecarga de 5% do seu peso corporal. A testosterona foi administrada 3 dias/semana, na concentração de 5 mg/kg de peso corporal do animal, por via subcutânea, também por 8 semanas. Ao final da sétima semana de experimento, os ratos foram submetidos aos testes de tolerância à glicose (GTT) e teste de tolerância à insulina (ITT). Após 10 semanas os animais foram sacrificados na condição de repouso. Amostras de sangue foram coletadas para a determinação do hematócrito, glicose, insulina, proteínas totais, colesterol total e triglicérides. Os testículos, a próstata e vesícula seminal, os tecidos adiposos foram analisados em balança analítica. O tecido muscular e hepático foram usados para determinação do glicogênio e da razão proteína/DNA. As adrenais foram pesadas e usadas para determinar o conteúdo de colesterol e ácido ascórbico. A análise estatística foi feita por ANOVA e aplicação do teste de post-hoc de Newman-Keuls, onde adequado, com nível de significância pré-fixado em 5%. Os resultados indicam que os ratos treinados tiveram maior tolerância à glicose comparados com os animais sedentários durante o GTT. Nossos dados mostram que a administração crônica da testosterona mais o treinamento físico foi associada ao aumento da sensibilidade à insulina.O grupo treinado administrado com testosterona mostrou uma maior taxa de desaparecimento...
The aims of this study were to investigate the endocrine-metabolic adaptations in rats submitted to chronic swimming exercise associate to administration of testosterone during 8 weeks. Young Wistar rats were divided into four groups: sedentary control (CS), trained control (CT), sedentary testosterone (TS), trained testosterone (TT). Training protocol consisted of swimming 1h/day, 5 days/week, during 8 weeks, supporting a load of 5% body weight. Testosterone was administered 3 times a week, in concentration of 5 mg/kg body weight, during 8 weeks. After 7 weeks rats were submitted to glucose (GTT) and insulin (ITT) tolerance tests. After 8 weeks rats were sacrificed in rest. Blood samples were collected to determine hematocrit, glucose, insulin, total protein, total cholesterol and triglyceride. Hepatic and muscular tissues were used to determine glycogen and protein/DNA ratio. Adrenals were weighed and used to determine cholesterol and ascorbic acid contents. The results were analyzed by ANOVA and Newman- Keuls post-hoc test where appropriate and the significance level was 5%. The results indicate that trained rats showed higher glucose tolerance compared to sedentary animals during GTT. Our data showed that chronic exposure to testosterone plus physical training was associated with increase insulin sensitivity. The animals trained and administered with testosterone, showed higher glucose disappearance rates when compared to other groups. Testosterone administration promoted decrease in testicular mass and increase in prostate and seminal vesicle. Training promoted increase in muscle when compared to the sedentary groups, taking us to presuppose that training stimulated glycogen syntheses. A crisina é um composto natural biologicamente ativo encontrado predominantemente em flores de maracujá azul, em mel, camomila, própolis e casca de frutas. Possui propriedades potentes como efeito anti-inflamatório, antioxidante e tem sido empregada para o tratamento de desordens reprodutivas. Essa flavona tem o potencial de aumentar os níveis de testosterona endógena, tanto por inibir a enzima aromatase, quanto por estimular a esteroidogênese testicular. Alterações no metabolismo hormonal mediadas por interferências exógenas podem interferir na fisiologia prostática resultando em modificações na atividade secretora da glândula e na regulação dos receptores hormonais. Assim, o objetivo deste estudo foi avaliar os efeitos da exposição à crisina sobre a próstata e gônadas de gerbilos adultos. Para avaliar o potencial androgênico da crisina, nós analisamos, comparativamente, os efeitos da testosterona sobre estes mesmos órgãos. Machos e fêmeas de gerbilos com 90 dias de idade foram tratados com crisina (50 mg/kg/dia) ou com cipionato de testosterona (1 mg/kg/semana) e divididos tem 3 subgrupos que foram eutanasiados após 3, 7 e 21 dias de tratamento. As próstatas foram coletadas para análises morfológicas, morfométrico-estereológicas, imuno-histoquímicas e ultraestruturais. Testículos e ovários foram submetidos a análises morfológicas e morfométrico-estereológicas. Os níveis séricos de testosterona e 17β-estradiol foram mensurados por ELISA. Machos e fêmeas tratados com crisina não apresentaram aumento dos níveis séricos de testosterona. No entanto, apenas as fêmeas tratadas com crisina durante 21 dias demonstraram aumento nos níveis séricos de estradiol. Foram observados tanto na próstata ventral masculina, quanto na próstata feminina em todos os tempos de tratamento com crisina, aumento da frequência de receptores androgênicos (AR), maior taxa de proliferação e hiperplasia glandular. Além disso, foi observada nas fêmeas em todos os tempos de exposição com crisina maior frequência imunomarcação para receptor estrogênico alfa (ERα). Ultraestruturalmente, machos e fêmeas tratados com crisina apresentaram intenso desenvolvimento das organelas da rota biossintética-secretora, acompanhada de intensa remodelação estromal, enquanto toxicidade celular foi observada apenas nas fêmeas. Nas gônadas masculinas o tratamento com crisina preservou a morfologia testicular. Em fêmeas, observou-se um maior número de folículos ovarianos em maturação. Comparativamente o tratamento com cipionato de testosterona foi capaz de causar lesões na próstata e gônadas, visto que focos de neoplasia intraepitelial prostática e degeneração gonadal foram observados em ambos os sexos. Esses resultados demonstram que a exposição à crisina, preservou a morfologia testicular normal, aumentou o número de folículos ovarianos em crescimento, aumentou a atividade secretória e alterou a regulação de receptores hormonais na próstata, potencializando a resposta desta glândula aos efeitos biológicos dos esteróides endógenos. Assim nas condições experimentais desse estudo embora a crisina e a testosterona evocassem alterações morfológicas semelhantes na próstata gerbilos masculina e feminina, a suplementação com crisina foi mais favorável à saúde da próstata. Contudo, a administração de crisina como agente fitoterápico deve ser considerada com precaução, visto que esse flavonóide parece exercer efeitos diferenciais em machos e fêmeas.
Chrysin is a naturally occurring biologically active compound found predominantly in blue passion flower, honey, chamomile, propolis and fruit peel. It has potent anti-inflammatory and antioxidant properties and has been used for the treatment of reproductive disorders. This flavone has the potential to increase endogenous testosterone levels both by inhibiting the aromatase enzyme and stimulating testicular steroidogenesis. Changes in hormone metabolism mediated by exogenous interferences may interfere with the prostatic physiology resulting in changes in the secretory activity of the gland and in the regulation of hormonal receptors. Thus, the objective of this study was to evaluate the effects of the exposure to chrysin on the prostate and gonads of adult gerbils. To evaluate the androgenic potential of chrysin, we comparatively analysed the effects of testosterone on these organs. Male and female gerbils aged 90 days were treated with either chrysin (50 mg/kg/day) or testosterone cypionate (1 mg/kg/week), subdivided into 3 subgroups and euthanized after 3, 7 and 21 days of treatment. The prostates and gonads were collected for morphological, morphometric-stereological, ultrastructural and immunehistochemical analyses. The testes and ovaries were subjected to morphological and morphometric-stereological analyses. Serum levels of testosterone and 17β-estradiol were determined using ELISA methods. Males and females treated with chrysin did not show increase in serum testosterone levels. However, only the females treated with chrysin for 21 days showed increase in the serum levels of estradiol. It was observed in both ventral prostate of males and in prostate of females in all the times of treatment with chrysin an increase in the frequency of androgen receptors (AR), a higher rate of proliferation and glandular hyperplasia. In addition, it was observed in all exposure times of females treated with chrysin a higher immunoreactivity frequency for alpha estrogen receptor (ERα). As to ultrastructure, males and females treated with chrysin showed intense development of the organelles of the biosynthetic-secretory route, accompanied by intense stromal remodeling. Cell toxicity was observed only in females. In the gonads of males the treatment with chrysin preserved testicular morphology. In females, we observed a larger number of ovarian follicles in maturation. Comparatively, the treatment with testosterone cypionate was able to cause lesions in the prostate and gonads, since the foci of prostatic intraepithelial neoplasia and gonadal degeneration were observed in both sexes. These results demonstrate that exposure to chrysin preserved normal testicular morphology, increased the number of ovarian follicles in growth, higher activity of secretory and changed the regulation of hormone receptors in the prostate potentiating the response of this gland to the biological effects of endogenous steroids. Therefore, under the experimental conditions of this study, although the chrysin and testosterone evoked similar morphological changes to those in the prostate of both sexes, supplementation with chrysin appeared to be more favorable to the prostate health. However, the chrysin intake as a phytotherapic agent should be considered with caution given that this flavonoid seems to exert distinct effects in males and females.

O potencial papel da testosterona na ativação do desejo e da sensibilização a estímulos sexuais tem recebido especial atenção, com resultados contraditórios. A predominância dos traços de personalidade Extroversão e Procura de Sensações tem sido associada a maiores níveis de excitação sexual e testosterona. No presente estudo, combinámos medidas de desejo sexual, testosterona livre salivar e traços de personalidade para explorar as interações destas variáveis no contexto da sexualidade feminina. Uma amostra de conveniência de mulheres portuguesas adultas e em idade fértil (N = 64) foi recrutada para responder a um questionário com duas escalas de desejo (domínios de Desejo do Female Sexual Function Index e Abbreviated Sexual Function Questionnaire) e duas escalas de personalidade (Sensation Seeking Scale – V e NEO – Five-Factor Inventory – 20). Também foram recolhidas amostras de saliva, para medição da testosterona livre. Os resultados para a totalidade da amostra revelam uma confirmação fraca e restrita das hipóteses colocadas. Detetaram-se correlações positivas significativas com o desejo para a Procura de Sensações e Neuroticismo, mas não para a Extroversão. Identificaram-se algumas associações entre desejo e testosterona. Não se detetou qualquer associação entre testosterona e personalidade. No entanto, ao dividir a amostra consoante a toma ou não de contraceptivos hormonais, identificaram-se correlações significativas mais consistentes entre os níveis de testosterona e as variáveis desejo e personalidade em mulheres que não tomam a pílula combinada.
Despite recent efforts to uncover psychobiological factors underlying women’s sexual response, the understanding of female sexual desire remains elusive. Among hormonal correlates, Testosterone’s potential role in the activation of desire and sensitisation to sexual stimuli has received particular attention, with mixed evidence. Regarding individual differences, the predominance of personality traits such as Extraversion and Sensation Seeking has been associated with a lower inhibition of sexual arousal and higher testosterone levels. In the current study we combined the measurement of reported sexual desire, basal salivary free testosterone levels and personality traits to better understand how these variables interact in the context of female sexuality. A convenience sample of adult Portuguese women of reproductive age (N = 64) was recruited to fill out a questionnaire including two sexual desire scales (Female Sexual Function Index’s and Abbreviated Sexual Function Questionnaire’s Desire dimensions) and two personality scales (Sensation Seeking Scale – V and NEO – Five-Factor Inventory – 20). Participants also provided saliva samples, from which free testosterone levels were assayed. The results for the whole sample provide very limited and weak support for the assumptions made. Positive and significant associations were detected between sexual desire and the traits Sensation Seeking and Neuroticism, but not Extraversion. Some associations were found between desire and testosterone. No association was found between testosterone and personality. However, dividing the sample according to hormonal contraceptive use allowed us to detect significant correlations between testosterone levels and the variables desire and personality in women not using the combined pill.

Pełny tekst źródłaFrom studies on the endocrine control of reproductive behavior in anurans, we know that many aspects of vocal behavior and agonistic interactions of these animals are related to changes in plasma levels of testosterone and corticosterone. Among the species of neotropical frogs of great interest for studies on the endocrine control of reproductive behavior stands out Hypsiboas faber, a hylid characterized by the presence of territorial males, whose agonistic interactions cover a variety of vocal patterns with aggressiveness scale, which may culminate in physical conflict. Males of H. faber also build nests for oviposition and presents optional guard of the nests with eggs. The aim of this study was to understand the interrelationships between plasma levels of corticosterone and testosterone with calling behavior and territorial defense in males of H. faber during reproductive season. These relationships were investigated by focal observations during the vocal activities, followed by blood sampling for hormonal assay. We additionally used an experimental approach, which consisted of using recorded advertisement calls (playback), simulating an invasion of another male in the territory of the focal subjects, followed by behavioral observation and blood sampling for hormonal assay. The experimental stimulus had no effect on plasma levels of corticosterone and testosterone, neither on the majority of behavioral variables. We can identify a pattern of seasonal variation in levels of corticosterone and testosterone, as well as a positive correlation between these hormones. Additionally, individuals who vocalize at higher rates have higher levels of corticosterone during the beginning of reproductive season, while higher levels of testosterone appear in individuals more responsive to stimuli from the social environment.

Estudos clássicos evidenciaram que para que se inicie o desenvolvimento prostático é necessário a presença de andrógeno produzido pelos testículos fetais. Porém, além da estimulação androgênica, estudos apontam para outros hormônios que também atuam na próstata como a Prolactina (PRL). PRL é um hormônio que é principalmente secretado por células lactotróficas da hipofise anterior e está envolvido em muitos processos biológicos, incluindo lactação e reprodução. Assim, investigamos se a modulação da sinalização PRL altera a morfofisiologia da próstata ventral (VP) em ratos castrados. Os ratos Sprague Dawley adultos (n = 6) foram castrados e após 21 dias divididos em 10 grupos experimentais: Castrado Controle (CC): animais castrados que não receberam tratamento; Castrado+testosterona (T): animais castrados que receberam T (4mg / kg); Castrado+PRL (PRL): animais castrados recebendo PRL (0,3 mg / kg); Castrado+T+PRL (TPRL): animais castrados que receberam associação de T e PRL; e Castrado+BR (BR): animais castrados que receberam Bromocriptina (BR) (0,4mg / kg). Pełny tekst źródłaOBJECTIVE: We evaluated the safety of testosterone treatment and its efficacy on bone mineral density and body composition in males with testosterone deficiency syndrome (TDS) over 24 months. MATERIAL AND METHODS: This prospective, non-randomized, open-label, long-term follow up study evaluated the safety and efficacy of testosterone treament in men with TDS. 50 males aged 50–65 years with TDS (Aging Males Symptoms Scale [AMS] >26 and calculated free testosterone [cFT] <250 pmol/l) were administered 50mg testosterone gel daily for one year. During the second year, patients received 1000mg of testosterone undecanoate every 2–3 months. Outcome measures were clinical chemistry values and total testosterone; SHBG and cFT, changes in AMS and IPSS; and changes in bone mineral density and body composition measured by dual-energy-x-ray absorptiometry. RESULTS: There were no clinically significant changes in clinical chemistry safety parameters. There were significant improvements in both total and cFT and in AMS scores after 3 months (p<0.001). Bone mineral density improved significantly in L2-L4 (2.90% and 4,5%), total femur (0,74% and 3%) and trochanter (1.09% and 3.2%) at 12 and 24 months. Lean mass increased 2.35% at 12 months and 4.5% at 24 months, but proportionally more muscle mass was gained in arms and legs than in the trunk. Fat mass decreased 4.2% at 12 months and 9.1% at 24 months. CONCLUSIONS: Testosterone treatment in males with TDS leads to significant improvement in lumbar spine and hip BMD. It leads to body changes affecting lean and fat mass with significant improvement in AMS scores.

Um dos importantes alvos das ações androgênicas é o sistema cardiovascular. Particularmente, existe uma importante lacuna de conhecimento em relação à ação da testosterona em leitos venosos. Observamos anteriormente que a orquidectomia promove aumento das ações vasomotoras da fenilefrina em veia porta de rato e que mecanismos relacionados à endotelina-1 poderiam estar envolvidos neste fenômeno. Assim, o objetivo do presente trabalho foi estudar a influência da orquidectomia, seguida ou não de reposição hormonal com testosterona, sobre as respostas vasomotoras de diferentes veias isoladas de rato a agonistas simpatomiméticos. Para isto, utilizaram-se ratos Wistar machos adultos controles e orquidectomizados, seguidos ou não de reposição hormonal (propionato de testosterona, 10mg/kg, i.p., por 3 semanas, com intervalo de 5 dias entre as doses). Destes animais, anéis (4–5 mm) de veias pulmonar, renal, femoral, mesentérica, assim como de artérias, pulmonar, femoral, mesentérica, aorta torácica e abdominal foram estudados em cubas de órgão isolado. Obtiveram-se curvas concentração-resposta para fenilefrina, noradrenalina, clonidina, metoxamina, assim como para endotelina-1, calculando-se pEC50 e resposta máxima (Rmax). Além disso, determinou-se a expressão do mRNA para endotelina-1 e para os receptores ETA e ETB. Os resultados mostraram que a orquidectomia seguida ou não de reposição hormonal não alterou a capacidade contrátil de veia porta frente à clonidina e metoxamina, tampouco frente à noradrenalina na presença de timolol...
The physiological repercussions of a long term reduction in testosterone levels have been systematically investigated, once it can be related with some clinical situations frequently seen in men such as hypogonadism and aging. One of the most important targets of androgen actions is the cardiovascular system. However, androgenic actions upon this system remain controversial especially in venous bed. We have previously observed that orchidectomy induces an increase in vasomotor responses of phenylephrine in rat portal vein. Probably mechanisms related to endothelin-1 could be involved in this phenomenon. Thus, the presented study aimed to investigate the influence of orchidectomy, followed or not by testosterone replacement, upon the response of different isolated veins to sympathomimetic agonists. In this manner, we used control and orchidectomized adult male Wistar rats, with the latter group followed or not by testosterone replacement (testosterone propionate, 10mg/kg, i.p., for 3 weeks, with 5-day intervals between the doses). Rings (3-4 mm) of pulmonary, renal, femoral and mesenteric veins and pulmonary, femoral and mesenteric arteries, as well as thoracic and abdominal aorta were studied in organ baths. Concentration-response curves were obtained to phenylephrine, norepinephrine, clonidine, methoxamine, and to endothelin-1. The maximum response (Rmax) and EC50 were calculated. Furthermore, we determined the mRNA expression of endothelin-1 and the ETA and ETB receptors. The results showed that orchiectomy, followed or not by testosterone replacement did not alter the contractile response of portal vein to clonidine and methoxamine, either face to noradrenaline in the presence of timolol (10-6M) or timolol (10-6M) + yohimbine (10-6M). Resumo: Os objetivos do presente estudo foram investigar as adaptações endócrinometabólicas em ratos submetidos ao exercício físico de natação associado com a administração de testosterona durante 8 semanas. Ratos jovens Wistar foram divididos em 4 grupos: controle sedentário (CS), controle treinado (CT), testosterona sedentário (TS), testosterona treinado (TT). O protocolo de natação consistiu de 1 hora/dia, 5 dias/semanas, durante 8 semanas, suportando durante a natação uma sobrecarga de 5% do seu peso corporal. A testosterona foi administrada 3 dias/semana, na concentração de 5 mg/kg de peso corporal do animal, por via subcutânea, também por 8 semanas. Ao final da sétima semana de experimento, os ratos foram submetidos aos testes de tolerância à glicose (GTT) e teste de tolerância à insulina (ITT). Após 10 semanas os animais foram sacrificados na condição de repouso. Amostras de sangue foram coletadas para a determinação do hematócrito, glicose, insulina, proteínas totais, colesterol total e triglicérides. Os testículos, a próstata e vesícula seminal, os tecidos adiposos foram analisados em balança analítica. O tecido muscular e hepático foram usados para determinação do glicogênio e da razão proteína/DNA. As adrenais foram pesadas e usadas para determinar o conteúdo de colesterol e ácido ascórbico. A análise estatística foi feita por ANOVA e aplicação do teste de post-hoc de Newman-Keuls, onde adequado, com nível de significância pré-fixado em 5%. Os resultados indicam que os ratos treinados tiveram maior tolerância à glicose comparados com os animais sedentários durante o GTT. Nossos dados mostram que a administração crônica da testosterona mais o treinamento físico foi associada ao aumento da sensibilidade à insulina.O grupo treinado administrado com testosterona mostrou uma maior taxa de desaparecimento... Abstract: The aims of this study were to investigate the endocrine-metabolic adaptations in rats submitted to chronic swimming exercise associate to administration of testosterone during 8 weeks. Young Wistar rats were divided into four groups: sedentary control (CS), trained control (CT), sedentary testosterone (TS), trained testosterone (TT). Training protocol consisted of swimming 1h/day, 5 days/week, during 8 weeks, supporting a load of 5% body weight. Testosterone was administered 3 times a week, in concentration of 5 mg/kg body weight, during 8 weeks. After 7 weeks rats were submitted to glucose (GTT) and insulin (ITT) tolerance tests. After 8 weeks rats were sacrificed in rest. Blood samples were collected to determine hematocrit, glucose, insulin, total protein, total cholesterol and triglyceride. Hepatic and muscular tissues were used to determine glycogen and protein/DNA ratio. Adrenals were weighed and used to determine cholesterol and ascorbic acid contents. The results were analyzed by ANOVA and Newman- Keuls post-hoc test where appropriate and the significance level was 5%. The results indicate that trained rats showed higher glucose tolerance compared to sedentary animals during GTT. Our data showed that chronic exposure to testosterone plus physical training was associated with increase insulin sensitivity. The animals trained and administered with testosterone, showed higher glucose disappearance rates when compared to other groups. Testosterone administration promoted decrease in testicular mass and increase in prostate and seminal vesicle. Training promoted increase in muscle when compared to the sedentary groups, taking us to presuppose that training stimulated glycogen syntheses. The training protocol, testosterone administration or combination of two treatments, resulted in decrease of epididimal and perirrenal adipose tissues weight... AIM: The aim of our study was to investigate the effects of T in women on metabolic parameters and body composition and effects of T on vaginal histology, proliferation and innervations. METHODS: 16 ovariectomized FtM subjects recived TU 1000 mg i.m. with placebo or dutasteride. At week 0 and 54 the following measurements were performed: body composition and metabolic parameters. 16 vaginal sampler from 16 FtM subjects treated with T and 16 PrM e 16 M subjects were collected. Morfhology, glycogen content, Ki-67 proliferation, estrogen receptor, innervations, androgen receptor were evaluated. RESULTS: The administration of T in FtM determines a non-significant increase in LDL cholesterol and significant decrease in HDL cholesterol. HOMA are significantly reduced in the group TU-alone and tend to increase in group TU + D. The hematocrit increases significantly. BMI, WHR and fat mass tend to decrease. Lean body mass tend to increase. No significant changes in bone metabolism have been reported. Vaginal samples from FtM showed a loss of normal architecture of the epithelium. T administration resulted in a strong proliferation reduction. Stromal and epithelial ERα and stromal PGP 9.5 exspression was significantly decrease in FtM. ARs were detected in mucosa and stroma. In mucosa AR density decrease with age but non change with T. In stroma, AR density increase con T. CONCLUSIONS: In conclusion, no major adverse effects were reported after T administration. T administration determines changes in histomorphology and reduces proliferation of vaginal epithelium. We found AR expression in epithelium and stroma. T increase AR expression in stroma.

Resumo: O Diabetes mellitus geralmente se associa a algum tipo de disfunção sexual, provocando infertilidade, tanto em seres humanos, quanto em animais experimentais. Em trabalho anterior realizado em nosso laboratório, ratos machos que tiveram hiperglicemia induzida quimicamente pela administração de streptozotocin apresentaram diminuição da fertilidade através de múltiplos parâmetros analisados. O presente estudo teve como objetivo investigar os mecanismos envolvidos e o papel da testosterona no processo. Para tanto, ratos machos foram divididos em 3 grupos experimentais: normoglicêmico (controle), hiperglicêmico (streptozotocin), e hiperglicêmico com reposição hormonal (streptozotocin+testosterona) e foram avaliados parâmetros reprodutivos e espermáticos, níveis hormonais, a contratilidade do ducto deferente isolado in vitro, comportamento sexual e o número de espermatozóides ejaculados no útero. O ducto deferente de animais diabéticos apresentou um quadro de hipersensibilidade à metoxamina, um agonista sintético de adrenoceptores _1. Estes mesmos animais apresentaram os seguintes resultados: alteração de comportamento sexual e ausência de espermatozóides ejaculados, redução dos níveis plasmáticos de testosterona, perda de peso corpóreo e de órgãos como epidídimo, ducto deferente, vesículas seminais e próstata ventral, perda de células germinativas na luz e desorganização epitelial aparente em túbulos seminíferos, além da aceleração do tempo de trânsito dos espermatozóides pelo epidídimo. Os dados apresentados neste trabalho indicam que os mecanismos responsáveis pela perda de fertilidade natural de ratos diabéticos envolvem comprometimento do processo espermatogênico, assim como desregulação do eixo reprodutivo masculino, juntamente com evidências para problemas no processo de maturação espermática, tendo como fator complicante o prejuízo da função ejaculatória, dependente da contratilidade da musculatura lisa dos ductos deferentes. A reposição de andrógeno não foi totalmente capaz de reverter os danos causados pelo diabetes no sistema reprodutivo masculino de ratos adultos
Abstract: Diabetes mellitus is usually related with some kind of sexual dysfunction, promoting infertility in humans as well as in experimental models. In a prior work from our laboratory, male rats, which had a diabetic-induced state of hyperglycemia caused by streptozotocin administration, demonstrated reduced fertility through several parameters analyzed. The present study aimed at investigating the mechanisms involved and the role of testosterone in the process. Male rats were randomly allocated in 3 experimental groups: control, hyperglycemic (streptozotocin), and hyperglycemic with hormone replacement (streptozotocin+testosterone) and the following parameters were analyzed: reproductive and spermatic parameters, hormone levels, sexual behavior, contractility of vas deferens in vitro, sexual behavior parameters and the number of sperm ejaculated in utero. The vas deferens of diabetic animals was hypersensitive to methoxamine, a synthetic agonist of _1 adrenoceptors. The same animals showed the following results: alterations in sexual behavior and lack of sperm ejaculated, reduction in plasma testosterone levels, decreased body weight and epididymis, seminal vesicles, ventral prostate and vas deferens weights, loss of germ cells in the lumen and apparent epithelial disarrange in seminiferous tubules, and accelerated sperm transit time in the epididymis. The data presented herein indicate that the mechanisms underlying the reduced fertility through natural mating observed in diabetic rats involve impairment of the spermatogenic process, as well as a dysregulation of the male reproductive axis, together with evidence for problems in the sperm maturation process, which has as a complicant factor the impairment of the ejaculatory function, dependent on the vas deferens smooth muscle contractility. Androgen replacement was not totally capable of reversing the damage caused by diabetes on the male reproductive system of adult rats

Resumo: A próstata é uma glândula encontrada não somente em machos de mamíferos, podendo estar presente em fêmeas de alguns roedores e também em mulheres. Os andrógenos são essenciais durante o desenvolvimento e também para a manutenção da homeostasia prostática ao longo da vida. No entanto, sensíveis interferências, principalmente em momentos críticos do desenvolvimento, podem predispor este órgão a desenvolver lesões durante a vida adulta e senil. Pesquisas tem demonstrado que o programa de expressão gênica controlado pelo receptor de andrógeno durante a organogênese prostática é muito semelhante ao que é ativado durante a instalação do câncer de próstata. Aliado a isso, um grande número de estudos tem demonstrado que a predisposição às desordens prostáticas tem sua origem nos momentos iniciais da vida. Desta forma, o objetivo deste trabalho foi avaliar se a exposição androgênica intra-uterina e pós-natal pode interferir na morfofisiologia da próstata de roedores, podendo levar ao surgimento de desordens prostáticas durante a vida adulta. Para isto, este trabalho foi dividido em duas etapas. Primeiro, grupos de ratos Wistar jovens e adultos não tratados e de ratas adultas tratadas com testosterona (1 mg/Kg/48 horas) durante 7, 14 e 21 dias foram analisadas. As próstatas destes animais foram submetidas a análises morfométrico-estereológicas, imunohistoquímicas e ultra-estruturais. Em uma segunda etapa, grupos de gerbilos machos e fêmeas expostos à testosterona durante a vida embrionária (100 ?g ou 1 mg) e pós-natal (100 ?g) foram avaliados na vida adulta. As glândulas prostáticas destes animais foram submetidas a análises morfométrico-estereológicas, imunohistoquímicas, além da reconstrução tridimensional. Os resultados mostraram que as próstatas das ratas sujeitas a terapias androgênicas durante a vida adulta apresentaram um intenso desenvolvimento, tornando-se mais ativas nos processos de síntese e secreção. Além de sofrer um intenso desenvolvimento, a glândula prostática destes animais tornou-se, morfologicamente e fisiologicamente, muito semelhante à próstata ventral dos ratos. Entretanto, apesar das próstatas das ratas tratadas sofrerem um expressivo desenvolvimento induzido pela ação androgênica, não foram observadas a presença de lesões nestas glândulas. Já os machos e as fêmeas de gerbilos que sofreram exposição androgênica anormal durante o período embrionário, apresentaram uma alta incidência de lesões prostáticas na vida adulta, podendo-se evidenciar a presença frequente de focos inflamatórios e de neoplasias intra-epiteliais. Os resultados mostraram que o excesso de andrógeno interfere significantemente na homeostasia prostática, levando a um intenso desenvolvimento glandular ou mesmo predispondo esta glândula a desenvolver lesões durante a vida adulta. Estes aspectos são de grande interesse, pois é cada vez maior o número de relatos na literatura mostrando que a desbalanço hormonal durante os momentos iniciais do desenvolvimento prostático podem predispor este órgão a manifestar doenças durante a vida adulta e senil.
Abstract The prostate is a gland found not exclusively in male mammals, may also be present in female of some rodents and also in women. The androgens are essential for the development and maintenance of prostatic homeostasis throughout life. However, sensible interferences, especially during critical stages of development, can predispose this organ to develop lesions in adult and senile life. Researches have shown that the gene expression program controlled by androgen receptor during prostatic organogenesis is very similar to that activated during the installation of prostate cancer. Moreover, a large number of studies have demonstrated that the predisposition to prostate disorders has its origin in the early stages of life. Thus, the objective of this study was to evaluate whether intrauterine and postnatal androgen exposure alters the morphophysiology of rodents prostate leading to the development of prostatic disorders in adulthood. For this, the work was divided into two stages. At first, groups of untreated young and adult male Wistar rats and adult female rats treated with testosterone (1mg/Kg/48 hours) for 7, 14 and 21 were analyzed. The prostates of these animals were subjected to morphometric-stereological, immunohistochemical and ultrastructural analyses. In a second step, groups of male and female gerbils exposed to testosterone (100 ?g or 1 mg) during embryonic and postnatal (100 ?g) life were analyzed at adulthood. The prostatic glands of these animals were subjected to morphometric-stereological, immunohistochemical and also to three-dimensional reconstruction analyses. The results showed that prostates of female rats subjected to androgen therapy during adulthood had an intense development, presenting a more active synthesis and secretion status. In addition to this intense development, the prostate gland of these animals became morphologically and physiologically very similar to the ventral prostate of adult male rats. However, despite the prostates of treated rats suffered a significant development caused by the androgenic action, it was not observed the presence of lesions in these glands. In other hand, males and females gerbils submitted to abnormal androgen exposure during the embryonic period exhibited a high incidence of prostatic lesions in adulthood, being frequent the presence of inflammatory foci and intraepithelial neoplasia. The results showed that androgen excess interfere significantly in the prostate homeostasis, leading to an intense glandular development or even predisposes this gland to develop lesions during adulthood. These aspects are of great interest because a growing number of reports in the literature has shown that the hormonal imbalance during the early stages of prostate development may predispose this organ to manifest disease during adulthood and senescence.

Resumo: Introdução e Objetivo: A disfunção erétil (DE) é a incapacidade de ter e/ou manter uma ereção para um intercurso sexual satisfatório. Dentre os diversos fatores ligados a ela pode-se citar a queda dos níveis de testosterona na corrente sanguínea. Apesar da evolução representada pela descoberta dos inibidores da enzima PDE5 no tratamento da DE, muitos homens ainda procuram tratamentos alternativos através do uso dos fitoterápicos. Este estudo avaliou a eficácia do Tribulus terrestris no tratamento da disfunção erétil e nos níveis da testosterona sanguínea. Pacientes e Métodos: Trinta homens foram inscritos no estudo. Todos eles tinham 40 anos ou mais, não eram tabagistas, não haviam sido submetidos a nenhum tratamento para câncer de próstata, não apresentavam dislipidemia e podiam ter diabetes e/ou hipertensão controladas. Os pacientes responderam ao IIFE-5 e sua testosterona total foi medida. Os pacientes foram randomizados em dois grupos de quinze indivíduos. O grupo tratamento recebeu Tribulus na dose de 400 mg duas vezes ao dia por 30 dias. O grupo controle recebeu placebo com a mesma orientação de ingestão. Após este período, os pacientes responderam ao questionário e tiveram sua testosterona medida novamente. Resultados: Antes do tratamento os indivíduos do grupo Tribulus apresentaram IIFE-5 médio de 13,2 pontos (variação 5 -18) (dp 3,5) e testosterona total média de 417,1ng/dl (variação 270,7ng/dl - 404,7ng/dl) (dp 89,4). Após o tratamento o grupo Tribulus apresentou IIFE-5 médio de 15,3 pontos (variação 5 -21) (dp 4,3) p=0,2463 e testosterona total média de 409,3ng/dl (variação 216,9ng/dl e 760,8ng/dl) (dp 125,8) p=0,3551. Conclusões: O Tribulus terrestris na dose de 800mg dia durante 30 dias não foi superior ao placebo, estatisticamente, quando comparado o IIEF-5 e aumento da testosterona total sérica
Abstract: Introduction and Objective: Erectile dysfunction (ED) is the inability to have and/or maintain an erection sufficient for a sexual intercourse. It could be linked to many factors. Among them could be pointed low testosterone levels. Despite the huge revolution of PDE5 inhibitors in the treatment of ED, many men have been looking for alternative treatments with herbs. This study evaluated the efficacy of Tribulus terrestris in the treatment of erectile dysfunction and in the serum levels of testosterone. Patients and Methods: Thirty men were enrolled to the study. All of them had 40 years old or later, non-smoking, not have been submitted to any treatment for prostate cancer, without dyslipidemia and they could have controlled diabetes and/or hypertension. The patients answered to the IIEF-5 and the total testosterone was measured. All the patients were randomized in two groups of fifteen. The treatment group received 400mg of Tribulus twice a day during 30 days. The control group received placebo at the same way. After this period, the patients answered the questionnaire and total testosterone was measured again. Results: Both groups were statistically equivalent. Before the treatment, the patients from Tribulus group presented mean IIEF-5 of 13.2 points (variation 5 - 18) (sd 3,5) and mean total testosterone of 417.1 ng/dl (variation 270,7 - 404,7) (sd 89,4). After the treatment, the Tribulus group presented mean IIEF-5 of 15.3 points (variation 5 - 21) (sd 4,3) p=0,2463 and mean total testosterone of 409.3ng/dl (variation 216,9 - 760.8) (sd 125,8) p=0,3551. Conclusion: Daily 800 mg Tribulus terrestris during 30 days was not statistically superior to placebo when compared IIEF-5 and serum total testosterone

Pełny tekst źródłaGlucocorticoids modulate the immune response in complex ways in vertebrates exposed to different stressors. Given that natural populations have been exposed to a multitude of stressors, a better understanding of the functional association between duration and intensity of the stress response, the resulting changes in steroid hormone levels and their impact on different aspects of immunocompetence emerges as a cornerstone for vertebrate conservation strategies. We investigated the relationships between steroids levels and innate immunocompetence in anuran amphibians, incorporating the methodology of restraint challenge, experimental elevation of corticosterone levels by transdermal application, bacterial killing ability by spectrophotometry and the immune challenge with phytohemagglutinin. Our results demonstrate that the bacterial killing ability (BKA) measured by spectrophotometry is a reliable and accurate method to estimate the immunocompetence of anuran amphibians, additionally showed the existence of a large interspecific diversity in BKA from male anurans. When four different species of Bufonids were submitted to a restraint challenge, the general responses included increased in corticosterone plasma levels (CORT) and neutrophil/lymphocyte ratio (N:L) and decreased in testosterone plasma levels (T). The responses of BKA to restraint were much more variable, with R. icterica showing decreased and R. marina showing increased values. Additionally, CORT and N:L tended to increase more in response to restraint with movement restriction than to restraint without movement restriction, indicating that toads showed an increased stress response to the more intense stressor. All variables studied show interspecific variation. Rhinella ornate showed higher baseline CORT when compared to other species, while R. ornate and R. icterica showed the highest baseline BKA values. However, changes in N:L ratio, T levels and BKA, were not correlated to increased CORT within or between species. Transdermal application of corticosterone efficiently mimics repeated acute stress response events in Rhinella icterica, without changing the immune parameters even after thirteen days of treatment. Interestingly, long-term captivity did not mitigate the stress response, since the toads maintained three fold increased CORT even after three months under these conditions. Moreover, long-term captivity in the same condition increased total leukocyte count (TLC) and generated an even greater decrease in BKA, suggesting that consequences of the stress response can be aggravated by time in captivity. Based on our results, we consider that a careful evaluation is necessary in order to understand the modulation of the immune response by stress at intra and interspecific levels. The inclusion of different segments of the immune response is desirable, and a standardized data collection for all the species under the same period (e.g. inside or outside of breeding season) and same activity (e.g. calling or foraging) is mandatory

Pełny tekst źródłaIn response for several stressors, the amphibian hypothalamus release the corticotrophin release hormone (CRH), which stimulates pituitary to release the adenocorticotropic hormone (ACTH) in the bloodstream. The ACTH, in your turn, acts in inter renal glands stimulating the glicocorticoids releasing, being the corticosterone the main glicocorticoid in anphibians. While the modulation exercised by the glicocorticoids of diverses physiological responses of the stress is fundamental for de maintainance of homeostasis in short period, the frequent and/or prolongued activation of the hypothamlamus pituitary interenal axis (HHI) could inhibit important process in long periods, as the reproduction and immunocompetence and affect melatonin synthesis acting directly in pineal gland. The objective of the present study was evaluate possible alterations in plasmatic corticosterone levels, testosterone, eyes melatonin levels, hematocrit and immunity aspects (Total count leucocytes and antimicrobian plasmatic capacity - against Escherichia coli) from males of the treefrog Hypsiboas faber in response to ACTH injection, as well, the temporal course of these responses. Blood and plasma samples were obtained from the 30 males captured in the field, whitin 3 minutes from the capture. The animals were transferred to the laboratory and, after two months, divided in six groups to be sampled in three different times 1 h, 3 h and 6 h after treatment. For each time one control group and one experimental group were added, receiving one injection of 105 μL of saline or ACTH solution (0,446 μg de ACTH/g of body mass (experimental group)). The ACTH treatment increase significantly the plasmatic corticosterone levels and decrease the plasmatic testosterone levels, as well, the eye melatonin levels after one hour. After six hours of the ACTH injection, occurred the increase of the hematocrit percentage and the eye melatonin levels. The ACTH treatment did not affect the immunity aspects studied in the sampling interval. The results confirm the role of the activation on the axis HHI in the inhibition of the testosterone secretion and in the modulation of melatonin secretion, althougth the possible immunomodutatory effects could happen in temporal intervals later

Pełny tekst źródłaUsing up six stallions of various breeds during 60 days, with initial average weight of 472,67 ¬± 90,48 kg, fed twice a day with a diet consisting of Tifton hay (Cynodon dactylon) and commercial concentrate, supplemented with 300 mL. of a vegetable oil (rice bran or soybean), moreover mineralized salt and ad libitum water. The experiment evaluated the effect of the supplementation of diet with rice bran oil, with high level of gamma-oryzanol, about acceptability of the diets, weight gain, body score, levels of plasmatic lipids, apparent digestibility of nutrients of the diets (dry matter - DM, organic matter - OM, crude protein - CP, ether extract - EE, neutral detergent fiber - NDF and acid detergent fiber ADF), and spermatic quality and plasmatic testosterone. Blood samples were also held with 0, 15, 30, 45 and 60 days after starting treatment, for analysis of the values of testosterone, triglycerides, total cholesterol and its fractions (HDL-C, LDL-C and VLDL-C). To evaluation of the apparent digestibility of the nutrients of the diet, the animals had passed for three days of total fecal collection. To evaluation of the sperm quality, the sires had passed for seminal collection with 0, 15, 30, 45 and 60 days of the treatment. It was used completely randomized design for repeated measures design with repeated measures over time and the means were compared under 5 % significance level. The gotten average values for the apparent digestibility of the DM, OM, CP, EE, NDF and ADF were respectively 64,34; 68,03; 71,95; 83,37; 62,15 and 55.05 % for the treatment with soybean oil, and 58,97; 62,61; 66,96; 81,94; 54,85 and 45.87 % for the treatment with rice bran oil. There was no statistical difference (p < 0,05) from the apparent digestibility of nutrients of the diet. The average values for testosterone, total cholesterol, HDL-C, LDL-C, VLDL-C and triglycerides had been respectively 75,93 ng/dL; 92,73; 61,47; 26,99 and 4,28 mg/dL for the treatment with soy oil; and of 62,13 ng/dL; 110,20; 66,73; 38,44 and 5,02 mg/dL for the treatment with rice bran oil. In relation to the spermatic quality, also did not have difference (p < 0,05), and the average values for volume, motility, vigor, concentration, defects, lesser defects and total defects, had been respectively, of 71,87 mL; 69%; 2,63; 123 x 106 sptz/mL; 17.73%; 4.60% and 22.33% for the treatment with soybean oil, and 78,67 mL; 70.67%; 2,93; 115,67 x 106sptz/mL; 17.96%; 6.03% and 22,63% for the treatment with rice bran oil. The supplementation of diet with semi-refined rice bran oil, with high level of gamma-oryzanol, provided better weight gain and improves the body score, do not affect the sperm quality or plasmatic levels of testosterone, VLDL-C, HDL-C and triglycerides, however, it increase plasmatic levels of total cholesterol and LDL-C.

Pełny tekst źródłaLittle is known about reproductive physiology in Ranfastidae and this knowledge is needed for the development of conservation programs and for successful captive breeding. Non-invasive methods allow long-term analysis in small animals, facilitate studies with free-living animals and minimize the handling stress. The purpose of this work was to measure the testosterone (males and females), progesterone and oestrogen (females) metabolites concentrations in the droppings of Toco-toucan (Ramphastos toco) and Red-billed-toucan (Ramphastos dicolorus), inside and outside the reproductive season. Three captive couples of each species were studied. Collected behavioural data showed that the animals were more active during reproductive season, but social behaviours and the couple proximity were rare in the two phases during the observation sessions. Progesterone metabolites concentrations could be obtained in the Reproductive Phase for both species, but weren`t detected in the Nonreproductive Phase. Testosterone metabolites could be measured for both species in the two phases, in males and females; and the results showed higher concentrations in most of individuals in the Reproductive Phase. Concentrations of testosterone metabolites above basal levels were associated to egg laying, which occurred during Reproductive Phase in one of the R. toco females. Results of oestrogen metabolites concentrations could not be obtained in both species, probably because the fail of the antibody in detecting these metabolites. However, this work showed that the utilized technique was efficient to measure testosterone and progesterone metabolites in those species droppings, evidencing seasonal variations in hormonal concentrations and indicating a relationship between testosterone concentrations and oviposition.

Pełny tekst źródłaThe objective of the present experiment was to evaluate the possible effect of diabetes mellitus, on the reproductive function in male rats. Towards this end, 27 male Wistar rats (98 days old), housed at the FMVZ-USP animal holding facility, were randomly assigned into three groups: Group A (GA) consisting of 10 healthy animals; Group B(GB) consisting of eight Wistar rats, with diabetes mellitus chemically induced through the intraperitoneal administration of estreptozotocin (65mg/Kg); and C group (GC) constituted by nine animals with diabetes mellitus chemically induced by the intraperitoneal administration of estreptozotocin (65mg/Kg), associate to an insulin treatment (3IU/rat per day for 42 days) that begun 4 days after the streptozotocin administration. Animals were euthanized 46 days after the administration of the estreptozotocin and the following results were obtained: diabetes mellitus led to an increase of the glicemia, reduction of the corporeal weight, decreased testicular wheight, serum concentrations of testosterone and FSH. No effect of diabeted were found for the serum levels of inhibin B. Results suggested that the exogen replacement of insulin was capable of hindering the reduction on corporal weight, reduction of testicular weight and reduction of the testosterone levels and FSH. On the other hand despite the fact that insulin treatament was capable of avoiding the reduction on bofy weight, it was not capable to provide similar body development observed in the normal animals.

Resumo: O objetivo deste estudo foi avaliar a influência dos hormônios sexuais na nocicepção da articulação temporomandibular (ATM) de ratos e os possíveis mecanismos envolvidos. A injeção de formalina na ATM em uma concentração (0,5%) que não induziu nocicepção em machos intactos, induziu em machos gonadectomizados e em fêmeas intactas, o que sugere que os níveis fisiológicos de testosterona diminuem o risco de machos desenvolverem nocicepção da ATM. A resposta nociceptiva induzida pela injeção de uma alta concentração de formalina (1,5%) na ATM de machos é significativamente menor que àquela induzida na ATM de fêmeas em diestro, fase do ciclo estral com baixos níveis de estrógeno, mas semelhante àquela induzida na ATM de fêmeas em proestro, faze do ciclo estral com altos níveis de estrógeno. Esse resultado sugere que a nocicepção da ATM, em fêmeas, é exacerbada durante a fase do ciclo estral em que os níveis de estrógeno estão baixos. A administração sistêmica de estrógeno ou progesterona em fêmeas gonadectomizadas e de testosterona em machos gonadectomizados reduz a resposta nociceptiva induzida pela injeção de formalina na ATM. A influência do sexo e dos hormônios ovarianos na nocicepção induzida pela injeção de formalina ou de glutamato na ATM foi exatamente a mesma, o que demonstra que o efeito antinociceptivo dos hormônios ovarianos na ATM não é estritamente relacionado a nocicepção induzida pela formalina. A semelhança entre estudos clínicos e os resultados obtidos utilizando estes dois agentes nociceptivos sugere que o modelo comportamental de nocicepção da ATM pode ser útil e confiável para estudar os mecanismos envolvidos no efeito antinociceptivo dos hormônios sexuais na ATM de ratos. A administração de drogas no líquido cefalorraquidiano da região de complexo sensorial trigeminal também é útil para o estudo desses mecanismos, mas o procedimento cirúrgico realizado para a implantação do cateter usado para a injeção pode afetar a expressão dos comportamentos relacionados a nocicepção orofacial. Portanto, a técnica que permite a injeção direta de drogas nessa região, sem a necessidade de procedimentos cirúrgicos contribui para o estudo dos mecanismos envolvidos no efeito antinociceptivo dos hormônios sexuais na ATM de ratos. A administração, por meio dessa técnica, do antagonista de receptores opióides naloxona no espaço subaracnóide da região do complexo sensorial trigeminal bloqueou o efeito antinociceptivo induzido pelos níveis fisiológicos de estrógeno em fêmeas em proestro e pela administração sistêmica de estrógeno ou progesterona em fêmeas gonadectomizadas e de testosterona em machos o gonadectomizados. No entanto, a co-administração de naloxona e formalina na ATM bloqueou o efeito antinociceptivo da progesterona e da testosterona, mas não do estrógeno. Esses dados sugerem que mecanismos opióides centrais medeiam o efeito antinociceptivo do estrógeno, da progesterona e da testosterona, enquanto mecanismos opióides periféricos também medeiam o efeito antinociceptivo da progesterona e da testosterona. A administração local de estrógeno, conjugado ou não com a albumina plasmática, na ATM de fêmeas reduziu significativamente a nocicepção induzida pela formalina. Como o estrógeno conjugado com a albumina tem ação restrita a receptores de membrana, esse dado sugere que o estrógeno reduz a nocicepção através de uma ação periférica não genômica. O efeito antinociceptivo do estrógeno foi bloqueado pelo antagonista de receptores estrogênicos ICI 182 780 e pelos inibidores da óxido nítrico sintase, L-NNA, e da guanilato ciclase, ODQ, mas não pelo antagonista de receptores opióides, naloxona. Esse dado sugere que o efeito antinociceptivo periférico do estrógeno é mediado pela ativação da via do óxido nítrico/GMP cíclico. Juntos, os resultados desse estudo demonstram que os níveis fisiológicos de testosterona diminuem o risco de ratos desenvolverem nocicepção da ATM e os de estrógeno diminuem a nocicepção da ATM em ratas. Além disso, a nocicepção da ATM também é diminuída pela administração sistêmica de estrógeno ou progesterona em ratas e de testosterona em ratos. O efeito antinociceptivo dos hormônios sexuais é mediado por mecanismos opióides centrais, enquanto mecanismos opióides periféricos medeiam o efeito da progesterona e da testosterona, mas não do estrógeno. De fato, a administração de estrógeno na ATM reduz a nocicepção através de um mecanismo periférico não genômico, mediado pela ativação da via do óxido nítrico-GMPc, mas não pela ativação do sistema opióide periférico
Abstract: The aim of this study was to evaluate the effect of sex hormones on temporomandibular joint (TMJ) nociception in rats and the possible mechanisms underlying their effect. The TMJ injection of 0.5% formalin induced nociception in intact females and gonadectomized males, but not in intact males, suggesting that the physiological level of testosterone protect males by decreasing their probability to develop TMJ pain. A higher dose of formalin (1.5%) induced a nociceptive behavior response significantly higher in female rats during diestrus phase of the estrous cycle than in those during proestrus phase and male rats. Since estradiol serum level was higher in proestrus than in diestrus females, this finding suggests that during low estradiol level of the estrous cycle the TMJ nociception is increased in female rats. Systemic administration of estradiol or progesterone in gonadectomized females and of testosterone in gonadectomized males significantly decreased 1.5% formalin-induced TMJ nociception. The role of sex and ovarian hormones in formalin and glutamate-induced TMJ nociception was virtually the same, showing that the antinociceptive effect of ovarian hormones was not exclusively related to the nociception induced by formalin. The similarity between clinical studies and the present results, obtained by using two different nociceptive agents, suggests that the TMJ behavior model may be useful and reliable to study the mechanisms underling the antinociceptive effect of sex hormones in the TMJ. Drug delivery to the medullary cerebrospinal fluid is also useful to study these mechanisms, however, the surgical procedure for implantation of the catheter used for drug delivery may affect the expression of the nociceptive behaviors related to orofacial nociception. Therefore, the technique for direct drug delivery to the medullary cerebrospinal fluid, without catheter implantation, will contribute for the study of the mechanisms underling the antinociceptive effect of sex hormones in the TMJ. The administration, though this technique, of the opioid receptor antagonist naloxone in the medullary region blocked the antinociceptive effect of estradiol, progesterone and testosterone. However, the co-administration of naloxone with formalin into the TMJ blocked the antinociceptive effect of progesterone and testosterone, but not of estradiol. These findings suggest that central opioid mechanisms mediate the antinociceptive effect of estradiol, progesterone and testosterone, while peripheral opioid mechanisms also mediated the antinociceptive effect of progesterone and testosterone. The local administration of estradiol, conjugated or not with the bovine serum albumin, significantly decreased formalin-induced TMJ nociception in female rats. Given that estradiol conjugated with bovine serum albumin is a membrane impermeable compound, these findings suggest that estradiol decreases TMJ nociception by a peripheral nongenomic mechanism. The antinociceptive effect of estradiol was blocked by an estrogen receptor antagonist and by a nitric oxide synthase and a guanilato cyclase inhibitors, but not by a opioid receptor antagonist. These findings suggest that estradiol decreases TMJ nociception in female rats through a peripheral activation of NO-cGMP signaling pathway. Taken together, the findings of this study suggest that the high physiological level of testosterone decreases the risk of male rats develop TMJ pain and that of estradiol decreases TMJ nociception in female rats. Furthermore, TMJ nociception was also decreased by systemic administration of estradiol or progesterone in female and of testosterone in male rats. The antinociceptive effect of sex hormones is mediated by central opioid mechanisms, while peripheral opioid mechanisms mediate the antinociceptive effect of progesterone and testosterone, but not of estradiol. In fact, the administration of estradiol in the TMJ decreases nociception by a peripheral non-genomic mechanism mediated by activation of the nitric oxide-cGMP signaling pathway, but not by opioid receptors

A doen?a renal cr?nica consiste em les?o renal com perda progressiva e irrevers?vel da fun??o dos rins. Nesta fase, os pacientes recebem indica??o de terapia renal substitutiva sendo a Hemodi?lise o recurso mais utilizado. Evid?ncias indicam que a pr?tica de exerc?cio f?sico intradial?tico ? ben?fica. Entretanto, os benef?cios desta modalidade de exerc?cio sobre par?metros end?crino-metab?licos e de estresse oxidativo ainda s?o poucos conhecidos. Objetivo: Avaliar o efeito de uma sess?o de exerc?cio f?sico de intensidade leve a moderada, sobre par?metros end?crino-metab?licos e de estresse oxidativo de pacientes com doen?a renal cr?nica submetidos ? hemodi?lise. M?todos: Doze volunt?rios, alocados de forma randomizada, do Servi?o de Hemodi?lise da Santa Casa de Caridade de Diamantina/MG participaram do estudo. Todos foram cross-over durante o experimento, a fim de comparar os resultados entre os dias de hemodi?lise sem exerc?cio (HD s/ exc) e hemodi?lise com exerc?cio (HD c/exc), e os dias posterior ? sess?o de hemodi?lise sem exerc?cio (P-HD s/ exc) e posterior ? sess?o de hemodi?lise com exerc?cio (P-HD c/exc), para cada volunt?rio. O exerc?cio f?sico intradial?tico ocorreu de maneira supervisionada, e por meio de um cicloerg?metro port?til. A coleta de saliva para avalia??o dos par?metros end?crino-meteb?licos cortisol e testosterona ocorreu durante todos os dias do protocolo experimental, em seis momentos ao longo do dia, a saber: ao acordar (T1) e trinta minutos ap?s (T2), 12:00h (T3), 14:00h (T4), 16:00h (T5) e ?s 19h (T6). As amostras de sangue para avalia??o dos par?metros de estresse oxidativo foram coletadas apenas em dias de hemodi?lise, em dois momentos: antes (T3) e ap?s (T4) a sess?o de hemodi?lise. Resultados: As concentra??es de cortisol e testosterona para os dias de hemodi?lise com exerc?cio f?sico (HD c/exc) n?o se elevaram significativamente em rela??o ?s concentra??es apresentadas no dia de hemodi?lise sem exerc?cio f?sico (HD s/exc), o que tamb?m ocorreu para os dias posteriores ? aplica??o do protocolo de exerc?cio intradial?tico (PHD c/ exc e P-HD s/ exc). Entretanto, a raz?o testosterona/cortisol aumentou significativamente no dia posterior ? sess?o de hemodi?lise com exerc?cio (P-HD c/ exc) quando comparada ao dia posterior ? sess?o de hemodi?lise sem exerc?cio (P-HD s/ exc). Os par?metros de estresse oxidativo n?o apresentaram altera??es induzidas pelo protocolo de exerc?cio f?sico. Chronic kidney disease is a kidney injury with progressive and irreversible loss of kidney function. At the severe stage, patients undergo renal replacement therapy and the hemodialysis being the most used medical resource. Evidence indicates that the practical of physical exercise during the hemodialysis sessions is beneficial. However, the benefits of this exercise modality on endocrine-metabolic parameters and on oxidative stress remains unknown. Objective: To evaluate the effect of an exercise session mild to moderate intensity, on endocrine-metabolic parameters and oxidative stress in patients with chronic renal failure undergoing hemodialysis. Methods: Twelve users of the Hemodialysis Service of Santa Casa de Diamantina Charity / MG participated, were assigned randomly as volunteers in the study and were cross-over during all the experiment in order to compare the results between the days of hemodialysis session without exercise (HD s/ exc) and hemodialysis session with exercise (HD c/ exc), and the subsequent days of hemodialysis session without exercise (P-HD s/ exc) and after the hemodialysis session with exercise (P-HD w / exc) for each volunteer. The exercise protocol was performed in a portable bicycle ergometer and its intensity was previously determined for each volunteer as mild to moderate. Saliva samples were collected during all days of the experimental protocol, six times throughout the day, namely on waking (T1) and thirty minutes after (T2), 12: 00 (T3), 14 : 00 (T4), 16: 00h (T5) and 19h (T6) to determine the salivary cortisol concentrations and salivary testosterone concentrations. Blood samples were collected in days of hemodialysis sessions, in two times: pre (T3) and after (T4) hemodialysis session to determine the REDOX status parameters. Results: The concentrations of cortisol and testosterone on the days of hemodialysis with exercise (HD c / exc ) did not increase significantly in relation to the concentrations shown on hemodialysis day without exercise (HD s / exc ), which also happened after the exercise protocol application days (P-HD c/ exc and PHD s/exc). However, the reason testosterone/cortisol increased significantly on the day after the hemodialysis session with exercise (P-HD w / exc) when compared to the day after the hemodialysis session without exercise (P-HD s / exc). The oxidative stress parameters did not show induced changes by the physical exercise protocol. Conclusion: It was shown that the exercise protocol induced changes in the anabolic status 24 hours after its realization and did not induce changes in oxidative stress parameters.

Pełny tekst źródłaLa detecció d’esteroides androgènics anabolitzants endògens (EAAE) és un dels reptes analítics més difícils en la lluita contra el dopatge. El problema més important per a la seva detecció és distingir entre concentracions endògenes i aquelles que s’observen després de l’administració exògena d’un EAAE. Els mètodes de cribatge per a la detecció d’EAAE estan basats en la determinació del perfil esteroïdal i la introducció d’aquest en el passaport biològic de l’atleta. La inclusió de nous metabòlits d’esteroides pot ajudar a millorar les capacitats de cribatge del perfil esteroïdal. Per tant, l’objectiu d’aquesta tesis és detectar i caracteritzar nous metabòlits d’EAAE que puguin implementar-se en l’actual perfil esteroïdal i l’avaluació de la seva utilitat en la lluita contra el dopatge. Quatre metabòlits desconeguts de la testosterona van ser detectats i caracteritzats mitjançant la utilització de la cromatografia líquida acoblada a l’espectrometria de masses en tàndem. L’origen d’aquests compostos es va demostrar que provenia de la degradació de conjugats amb cisteïna. La formació d’aquests conjugats implica l’addició d’un doble enllaç com a reacció metabòlica de fase I acompanyat per la conjugació amb glutationa i la subseqüent degradació d’aquesta a cisteïna en orina. Per tal de poder veure la seva aplicació en el camp del dopatge, es va desenvolupar i validar un mètode per la quantificació indirecta d’aquests compostos en orina. Utilitzant aquest mètode es van establir límits de referència basats en l’anàlisi de 174 mostres de orina. Addicionalment, diferents factors descrits que poden afectar l’excreció en orina d’aquests compostos també van ser estudiats en detall. Finalment, es va avaluar la utilitat d’aquests metabòlits conjugats amb cisteïna per a la detecció de l’abús d’EAAE mitjançant l’ anàlisis de mostres després de l’administració de diferents EAAE. L’ús d’aquests metabòlits va millorar (en alguns casos) els temps de detecció comparant-los amb els actuals marcadors utilitzats en rutina.

Pełny tekst źródłaSignificantly lower peak ACTH and B responses to 10 min of restraint stress were found in intact animals with high basal levels of T (4.0 $ pm$ 0.6 ng/ml) compared to animals with low levels of T (1.2 $ pm$ 0.3). No differences were found in prestress, basal levels of ACTH and B as a function of T. To assess the importance of these naturally occurring variations in T levels on HPA activity, animals were gonadectomized (GDX) and replaced with subcutaneous steroid implants supplying physiological levels of T (1 to 5 ng/ml). Significantly lower ACTH and B responses to restraint stress were observed in high (5.1 $ pm$ 0.6 ng/ml) compared to low (1.7 $ pm$ 0.3 ng/ml) T-replaced animals. Plasma and intra-pituitary CBG levels were also reduced in GDX animals replaced with high T levels. Although T had no effect on median eminence CRH levels, arginine vasopressin (AVP) content was significantly lower in GDX animals replaced with elevated T levels. A strong negative correlation (r = $-$0.9) was found between pre-stress levels of T and AVP content in the median eminence. We then characterized corticosteroid receptor binding as a function of circulating T levels. The results revealed a dose-dependent relationship between plasma T levels and glucocorticoid type II binding that was apparent only in the medial preoptic area (MPOA) of the brain. No differences were found in GC type I receptor binding in the brain, pituitary, and liver as a function of T replacement. Glucocorticoid type II binding was significantly higher in the MPOA of the brain and liver in high T-replaced animals. Implants of crystalline B or T into the MPOA caused a significant decrease in ACTH and B responses to restraint. These implants also caused a decrease in median eminence AVP content, but not CRH. Lesions of the MPOA blocked the inhibitory effects of high peripheral T levels on ACTH responses to stress.
