Human hydroxysteroid dehydrogenase-like protein 2 (HSDL2) is a member of the short-chain dehydrogenase/reductase (SDR) subfamily of oxidoreductases and contains an N-terminal catalytic domain and a C-termianl sterol carrier protein type 2 (SCP-2) domain. In this study, the C-terminal SCP-2 domain of human HSDL2, including residues Lys318–Arg416, was produced inEscherichia coli, purified and crystallized. X-ray diffraction data were collected to 2.10 Å resolution. The crystal belonged to the trigonal space groupP3121 (orP3221), with unit-cell parametersa=b= 70.4,c= 60.6 Å, α = β = 90, γ = 120°. Two protein molecules are present in the asymmetric unit, resulting in a Matthews coefficient of 2.16 Å3 Da−1and an approximate solvent content of 43%.

Abstract Relying on the high current temperature rise test, starting from three aspects of different materials, different structural strengths, and different contact conditions, the research on the current-carrying-temperature rise characteristics of the joint terminals of ±800kV UHVDC converter stations under different conditions is carried out. The results show that, for joint terminals of the same structure type and contact area, the difference in resistivity of joint terminals of different materials has a significant impact on the temperature rise level. When the material is the same, the temperature rise level can be reduced to a certain extent after the silver (tin) layer is added to the contact surface of the terminal. When the contact resistance of the joint terminal and the current density are the same, after increasing the contact area, the temperature rise and decrease level of the aluminum alloy plate lap type joint terminal is significantly better than other types of joint terminals. The contact resistance and temperature rise of the joint terminal decrease with the increase of the bolt tightening torque, and become stable when it reaches more than 80%. When the bolt tightening torque reaches more than 90%, the surface finish of the contact surface has no obvious effect on the contact resistance of the joint terminal. By controlling the material properties, structural strength, contact conditions, significantly reduces the temperature rise of the level of the joint terminal, effectively ensuring the safety ±800kV UHVDC converter station stable operation.

This study aims to describe and analyze (1) the Type B Passenger Terminal function in the Southeast Sulawesi Province and (2) the strategy for developing the function of the Type B Passenger Terminal in the Southeast Sulawesi Province. Data analysis, namely with qualitative descriptive analysis and SWOT analysis. The research was carried out in Kendari City in August - December 2022. The results of the study concluded that (1) the terminal function has not been running optimally and, (2) strategies that must be carried out, namely a) Utilizing the terminal's strategic position by encouraging public transport companies using large terminal land, b) Partnering with private parties in accordance with regulations to utilize large terminal land to build terminal supporting facilities, c) Make effective officers at each terminal to direct drivers and passengers to the terminal and control the shadow terminals that disrupt the function of existing terminals, d) Synergize with city/district government in the synergy of the city/regency route network with the location of the type B passenger terminal and e) Encouraging the improvement of the quality of Transportation Human Resources, especially terminal management so that the implementation and management of the terminal runs according to the specified SOP.

Many people are interested in using the shadow terminal area on Jalan Raya Cibeber as a place to wait for public transportation and an interaction point between modes and routes. The purpose of this study is to evaluate the existence of shadow terminals around Pasir Hayam Terminal, focusing on the factors that influence passenger preferences in choosing shadow terminals compared to official terminals. This research method uses a quantitative approach, where Importance Performance Analysis (IPA) is used to evaluate public transportation users' satisfaction with the performance of shadow terminals and main terminals. In addition, the SWOT Analysis Method is used as a tool to help structure the problem, especially in analyzing the strategic environment, which consists of two aspects, namely the internal and external environment. The results of calculations and analysis using the IPA method show that comfort and convenience are the main factors that influence passenger preferences in choosing the Shadow Terminal. Another significant factor is the availability of clean and complete dining and toilet facilities. Through SWOT analysis, Pasir Hayam Terminal is in quadrant 2 which indicates a diversification strategy. In this strategy, the focus will be on optimizing the strengths of the terminal, improving existing weaknesses, taking advantage of open opportunities, and overcoming threats that may be faced. In addition, this strategy includes efforts to increase the advantages of Pasir Hayam Terminal over Terminal Bayangan and utilize the potential of Terminal Bayangan as a strength.

The projection specificity of retinal ganglion cells and the morphology of their terminals were studied in the plethodontid salamander Plethodon jordani. In an in vitro approach, ganglion cells were stained with biocytin and reconstructed by means of light microscopy. Single retinal ganglion cells often have multiple terminal structures in the thalamus, pretectum, and tectum. The projection pattern in the diencephalic neuropils is related to the depth of the terminal arbor within the tectal fiber layer. Terminal arbors in the tectum differ in location, size, and branching pattern. The following types could be distinguished: The most superficial of the optic terminals in layer 1 are relatively small with a diameter of about 100 μm. With the exception of a few varicosities (beads) in the pretectal neuropils, their stem axons have no further collaterals or terminal arbors in the diencephalic neuropils. Intermediate terminals in layer 2 fan out to form a dense plexus with a medio-lateral extent of 180 μm on average. Some terminals in this layer show obvious antenna-like fibers reaching toward the surface of the tectum. The axons of layer 2 projecting neurons have additional collaterals and terminal arbors in the thalamus and pretectum. The deep layer 3 terminals spread out over a diameter of 400 μm on average and their degree of branching is moderate. The axons of layer 3 projecting ganglion cells have dense additional terminal arbors in the thalamus and pretectum. The deepest retinal terminals in the tectum are found within the predominantly efferent fiber layers. This type consists of an unbranched, but beaded axon which runs rostro-caudally with several bends and loops. The stem axon has an additional very dense terminal arborization in the neuropil of the nucleus Bellonci pars medialis and additional sparse collaterals in the pretectal area.

The design and crash test results of a median barrier version of the Flared Energy-Absorbing Terminal, known as FLEAT-MT, are presented. This energy-absorbing terminal is designed for use with a W-beam, strong-post median barrier. The FLEAT-MT terminal uses two standard FLEAT terminals, one for each of the two W-beam rail elements. The energy-absorbing capability of the FLEAT-MT terminal is based on the sequential kinking concept, similar to that used with the Sequential Kinking Terminal and FLEAT guardrail terminals. Three full-scale vehicle crash tests were conducted to evaluate the impact performance of the FLEAT-MT terminal in accordance with guidelines set forth in NCHRP Report 350: Test 3-35—pickup truck redirection test (Test No. FMT-1), Test 3-31—pickup truck head-on test (Test No. FMT-2), and Test 3-39—pickup truck reverse-direction test (Test No. FMT-3M). The terminal performed as designed. The FLEAT-MT terminal meets all evaluation criteria for a Test Level 3 median barrier terminal set forth in NCHRP Report 350. The FLEAT-MT terminal is being evaluated by FHWA for approval to be used on the National Highway System.

A Heterojunction Phototransitor (HPT) is an attractive high-speed photodetector for optical communications, as it can provide high gain and high power. It can also be used to generate millimetre-wave signals using optical heterodyne for radio astronomy and wireless broad-band communications. Edge illuminated 2-terminal HPTs (2T-HPTs) integrated with an optical waveguide are designed and characterised. A small signal model is developed and it simulates the 2T-HPT frequency responses with good accuracy and supplements the measurement to determine its gain and cut-off frequency. This overcomes the requirement to measure the input photocurrent to the 2T-HPT using an exact replica of its PIN structure. The high frequency responses for the longer devices, which give higher saturation current, are limited by the loss of the transmission line. This high loss is also observed by the 2-port characterisation of the back-to-back configured 2T-HPT and gives a trade-off between the cut-off frequency and saturation current in the design. Additional analysis shows that the cut-off frequency is also limited by the large device capacitances. Using scaling of the model, the cut-off frequency is shown to increase to 59.4GHz with a gain of 39dB for a 1mum x 10mum 2T-HPT. The above result shows that the 2T-HPT could be best utilised in a periodic structure (P-TWHPT), with small device area sections connected by short transmission lines. This increases the saturation current level without reducing the cut-off frequency and gain. A P-TWHPT model is developed to simulate and analyse its frequency response. It is shown that the effects of phase matching and reverse input termination are not critical for 2T-HPT structure, as its 3dB bandwidth is limited by the base discharge time constant. A four 5mum x 10mum 2T-HPT section P-TWHPT is simulated with the phase mismatch and without reverse termination. It has a gain of 49dB and a cut-off frequency of 67.5GHz.

Vários estudos envolvendo a avaliação da função endotelial em pacientes com insuficiência renal terminal (IRT) e em pacientes com diabetes tipo 2 (DM2) têm sido apresentados objetivando um melhor entendimento dos mecanismos fisiopatológicos e das condições que cercam essas patologias. As sessões de hemodiálise (HD) em pacientes com IRT e a microalbuminúria em pacientes com DM2 influenciam as respostas endoteliais e, diante disso, este trabalho constitui-se de dois estudos independentes com ambas as populações, sendo: o Estudo 1 objetivou avaliar a influência de uma sessão de HD na função endotelial venosa e no estresse oxidativo de nove pacientes com IRT comparados a um grupo controle (n=10). A função endotelial foi avaliada pela técnica de Dorsal Hand Vein e o estresse oxidativo pelas medidas de capacidade antioxidante total (TRAP) e de dano oxidativo à proteínas (Carbonilas). Antes da HD, os pacientes apresentaram reduzida venodilatação dependente do endotélio (VDE) na comparação com o controle (65,6 ± 10,5% vs. 109,6 ± 10,8%; P = 0,010). Após a HD houve um aumento na VDE (106,6 ± 15,7%; P = 0,045). A venodilatação independente do endotélio foi similar em todas as comparações realizadas. A HD reduziu o TRAP (402,0 ± 53,5 vs 157,1 ± 28,3 U of Trolox/μL plasma; P = 0,001) e não alterou as carbonilas (P = 0,389). O delta de VDE foi negativamente correlacionado com o delta de TRAP (r = -0,70; P = 0,037). Esses resultados sugerem que a sessão de HD melhora a função endotelial venosa associada a menor redução de antioxidantes. No Estudo 2 foram avaliados 28 pacientes com DM2, subdivididos em grupo normoalbuminúrico (n=16) e microalbuminúrico (n=12), com o objetivo de comparar entre os grupos a função endotelial no leito venoso e arterial. As funções endotelial venosa e arterial foram avaliadas pela técnica de Dorsal Hand Vein (venodilatação) e pela dilatação mediada pelo fluxo (FMD) da artéria braquial através de ultrasonografia de alta resolução, respectivamente. Pacientes microalbuminúricos apresentaram reduzida venodilatação (32,9 ± 17,4% vs. 59,3 ± 26,5%, respectivamente; P = 0,004) e FMD (1,8 ± 0,9% vs. 5,1 ± 2,4, respectivamente; P < 0,001), quando comparados a normoalbuminúricos. A venodilatação se correlacionou negativamente com albuminúria (r = -0,53; P = 0,004) e com HbA1c (r = -0,41; P = 0,032). O mesmo foi observado entre FMD e albuminúria (r = -0,49; P = 0,007) e HbA1c (r = -0,44; P = 0,019). Foi observada uma correlação positiva entre venodilatação e FMD (r = 0,50; P = 0,007). Assim, a função endotelial em ambos os leitos vasculares está prejudicada em pacientes com DM2 e microalbuminúria. Os dois métodos utilizados para avaliação da função endotelial se correlacionaram positivamente. Mesmo em um grupo de pacientes com bom controle metabólico, as respostas de vasodilatação se correlacionaram negativamente com HbA1c.
Several studies were shown, involving endothelial function assessment in patients with endstage renal disease (ESRD) and in patients with type 2 diabetes (T2D), aiming a better understanding of physiopathological mechanisms and conditions surrounding these pathologies. Hemodialysis (HD) sessions in patients with ESRD and microalbuminuria in patients with T2D influence the endothelial responses and, therefore, this work was based on two independent studies, namely: Study 1 aimed at evaluating the influence of an HD session in venous endothelial function and oxidative stress of nine patients with ESRD compared to a control group (n=10). Endothelial function was assessed by dorsal hand vein technique and oxidative stress by total radical trapping antioxidant potential (TRAP) and protein oxidative damage (carbonyls). Before HD, the patients showed a reduced endothelium-dependent venodilation (EDV) compared with controls (65.6 ± 10.5% vs. 109.6 ± 10.8%; P = 0.010). After HD there was an increase in EDV (106.6 ± 15.7%; P = 0.045). The endotheliumindependent venodilation was similar in all comparisons performed. HD decreased TRAP (402.0 ± 53.5 vs 157.1 ± 28.3 U of Trolox/μL plasma; P = 0.001) and did not change the carbonyls (P = 0.389). The delta in EDV was negatively correlated with the delta in TRAP (r = -0.70; P = 0.037). These results suggest that an HD session improves endothelial venous function, in association with an antioxidant effect. In Study 2 28 patients with T2D were evaluated, subdivided into normoalbuminuric (n=16) and microalbuminuric group (n=12), aiming to compare the endothelial function in venous and arterial bed between the groups. Venous and arterial endothelial function were assessed by the dorsal hand vein technique (venodilation) and brachial artery flow-mediated vasodilation (FMD) by high-resolution ultrasonography, respectively. Microalbuminuric patients presented impaired venodilation (32.9 ± 17.4% vs. 59.3 ± 26.5%, respectively; P = 0.004) and FMD (1.8 ± 0.9% vs. 5.1 ± 2.4, respectively; P < 0.001), as compared to normoalbuminuric patients. Venodilation was negatively correlated with microalbuminuria (r = -0.53; P = 0.004) and HbA1c (r = -0.41; P = 0.032). The same was observed between FMD and albuminuria (r = -0.49; P = 0.007) and HbA1c (r = -0.44; P = 0.019). Venodilation was positively correlated with FMD (r = 0.50; P = 0.007). Thus, both venous and arterial endothelial function are impaired in patients with T2D and microalbuminuria. The two methods used for endothelial function evaluation were positively correlated. Even though patients had good metabolic control as a group, vasodilation responses were negatively correlated with HbA1c.

Ethical hacking and penetration testing are two methods often used when organizations and companies want to measure their level of information security, and find out if there are additional steps that can be taken in order to increase the security. This report describes a security test of the card terminal iZettle Reader 2, with the intention to examine its level of security based on the device’s frequent appearance in the society. The implementation is divided into three phases: prestudy and threat modelling, penetration testing and evaluation and conclusion of the security. The threat model was created using the two established models STRIDE & DREAD, which purpose is to identify the device’s various threats and attack vectors. From the threat model, a couple of attack vectors were selected to be penetration tested. By using conventional models and obtaining knowledge of common attacks such as Man-in-the-middle, Spoofing and Replay, the device and the payment solution could be tested with a systematical and reliable approach. A selection was made of the most prominent attack vectors, of which these were later tested; Man-in-the-middle of Bluetooth and HTTPS, and Reverse Engineering of the associated mobile application "iZettle Go". The result of the penetration tests indicated that the security around the device and surrounding systems is strong, but that it can be further supplemented with a couple of actions like certificate pinning and mutual authentication when communicating with TLS, as well as a more tamperproof software regarding the mobile application.
Etisk hackning och penetrationstestning är två metoder som ofta tillämpas i sammanhang när organisationer och företag vill mäta sin nivå av informationssäkerhet, samt även ta reda på om eventuella åtgärder kan tas för att stärka säkerheten. Denna rapport beskriver ett säkerhetstest av kortterminalen iZettle Reader 2, med syftet att undersöka dess nivå av säkerhet grundat på enhetens frekventa uppträdande i samhället. Genomförandet är uppdelat i tre faser: förstudie och hotmodellering, penetrationstestning samt utvärdering och avgörande av säkerheten. Hotmodellen skapades med hjälp av de två vedertagna modellerna STRIDE & DREAD, vars syfte är att identifiera enhetens olika hot och attackvektorer. Utifrån hotmodellen valdes några attackvektorer som sedan penetrationstestades. Genom att använda etablerade modeller samt erhålla kännedom om konventionella attacker såsom Man-in-the-middle, Spoofing och Replay, kunde man testa enheten och betallösningen med ett systematiskt och pålitligt tillvägagångssätt. Ett urval gjordes av de mest lovande attackvektorerna, varav dessa senare testades; Man-in-the-middle av Bluetooth och Wi-Fi samt Reverse Engineering av den tillhörande mobilapplikationen "iZettle Go". Resultatet av testerna påvisade en stark säkerhet gällande enheten och dess omgivande system, men att säkerheten kan kompletteras ytterligare med ett par olika åtgärder som certificate pinning och mutual authentication vid kommunikation med TLS, samt manipuleringssäker mjukvara med avseende på mobilapplikationen.

O presente trabalho apresenta uma nova perspectiva para as discussões jurídicas e bioéticas acerca dos direitos da personalidade dos pacientes terminais e encontra em nosso ordenamento jurídico, na afirmação de um direito geral da personalidade, a plenitude da tutela civil dos bens jurídicos personalíssimos do enfermo, sobretudo nas etapas finais da doença, ocasião em que a hipermedicalização do processo de morrer destaca-se como o principal fator gerador de lesões de diversas naturezas a esses bens. No caminho para chegarmos à proteção geral da personalidade, examinamos alguns direitos especiais que emergem ao final da existência humana, como o direito à morte em momento natural. No intuito de recuperar a centralidade da pessoa humana como fim único a que devem servir o Direito e a Medicina, construímos a trajetória do conceito de pessoa em seu desenvolvimento jusfilosófico para afirmar que todo ser humano é pessoa e sujeito de direito (ubi homo sapiens, ibi persona), ainda que não possua capacidade jurídica de fato, e, com isso, demonstrar a impossibilidade de pertença a uma classe de não pessoas independentemente de circunstâncias ou do desenvolvimento biopsíquico humano. Asseveramos, também, que o cuidar e o tratar em pacientes gravemente enfermos e terminais deve relacionar-se, antes, ao homem em sua dignidade e plenitude, em uma concepção biomédica, filosófica e metafísica conjugada da sua existência, e não se reduzir à simples obstinação prognóstica e terapêutica, visão reducionista que relaciona tratar a doença a um investimento no prolongamento estéril da vida humana. Nesse sentido, apresentamos perspectiva jurídica inovadora para a enfermidade e para a vivência dessa condição, do ponto de vista de pacientes terminais, cuidadores e equipes de saúde, à luz do pensamento de Emmanuel Lévinas e à centralidade que ele outorga à figura do Outro, que ilumina nossa hermenêutica do instituto dos direitos da personalidade. Concluímos que se faz necessária uma mudança do paradigma atual de cuidados de saúde em fim de vida também na esfera jurídica, com a aceitação, na escolha terapêutica, da naturalidade do evento morte ao final da existência: da busca da cura, para o cuidar; da quantidade para a qualidade da vida que resta.
The thesis presents a new perspective of the legal and bioethical discussions regarding individual rights of terminal ill patients and finds in our legal system, in the assertion of a general individual right, the plenitude of the protection of the legal rights of the ill, especially at the last stages of the disease, when hipermedicalization of the dying process asserts itself as the major source of the various damages caused to individual rights. On the pathway to achieve the general protection of the personhood, we highlight some special rights which emerge at the end of human existence, as, for instance, the right for a death at a natural moment. To recover the centrality of the human person as the single and only end to which Law and Medicine should serve, we present herein a path of the concept of personhood in its juridical and philosophical development to affirm that every human being is an individual (ubi homo sapiens, ibi persona), even if he/she doesnt have legal capacity and, therewith, demonstrate the impossibility of belonging to a class of non-persons independently of circumstances or the bio-psychic development. We also argue that treating and caring of seriously ill and terminal patients should be related with person in its dignity and fullness, in a biomedical, philosophical and metaphysical conception of existence, irreducible to mere obstinacy in prognosis and treatment, as a result of a reductionist perspective which relates treating a disease to a futile investment of a sterile extension of human life. In this sense, we present a innovative juridical perspective to illness and the experience of this condition, from the point of view of terminal ill patients, caregivers and health care teams in light of the thought of Emmanuel Lévinas and the centrality that he grants to the figure of the Other, which illuminates our interpretation of individual rights. We conclude that a change in the extant paradigm of the end-of-life care in Brazil is imperative also in the legal realm, with the acceptance, in the therapeutic choice, of the natural path of death at the end of our existence: from the search for cure, to care; from quantity to quality of the remaining life.

The C terminal binding proteins (CtBP) 1 and 2 are a family of transcriptional co-repressors overexpressed in a variety of cancers, and are frequently associated with poor prognosis and chemoresistance. CtBP has also been characterized in cell culture models as drivers of migration/invasion and epithelial-mesenchymal transition. CtBP mediates its transcriptional corepressor activity via its dehydrogenase domain, and inhibition of this domain interferes with CtBP oncogenic functions. The role of CtBP in APC mutant neoplasia remains obscure even though APC is responsible for degradation of both β-catenin and CtBP in suppressing colorectal tumorigenesis. Our prior work demonstrated that CtBP proteins can be effectively therapeutically targeted with substrate analogues of their intrinsic dehydrogenase domains. In addition, CtBP2 has been reported to play an important role in human colon cancer stem cells via its interaction with the transcription factor TCF4 on chromatin and promotes self-renewal of human colon tumor initiating cells (TICs) in vitro. To study CtBP2’s role in TIC activity in vivo, we studied TIC’s from Apc min/+ mice, which exhibit massive intestinal polyposis, in Ctbp2 wildtype or haploinsufficient backgrounds. Indeed, LGR5+, CD44+/CD24+, and CD133+ TIC populations were substantially decreased in Apc min/+ Ctbp2 +/- vs. Apc min/+ intestinal epithelia. Validating CtBP as a therapeutic target for TIC activity, we investigated intestinal TIC populations and polyposis in Apc Min/+ mice treated with the CtBP inhibitor 4-chloro-hydroxyimino phenylpyruvate (4-Cl-HIPP). Like Ctbp2 haploinsufficiency, 4-Cl-HIPP significantly decreased intestinal polyposis and downregulated LGR5+, CD44+/CD24+, and CD133+ TIC populations. To understand if CtBP2’s role in TIC activity and tumor progression extended to a cancer model, we studied the impact of Ctbp2 gene dosage in a mutant KRAS mouse pancreatic ductal adenocarcinoma (PDAC) model (CKP). Ctbp2 haploinsufficiency remarkably prolonged survival, abrogated peritoneal metastasis and ascites, and caused dramatic downregulation of c-Myc, a known critical dependency for TIC activity and tumor progression in PDAC. Overall, our data suggest a key role of CtBP2 in driving the TIC compartment and progression in both pre-malignant intestinal polyposis and aggressive murine PDAC models.

Recent data has indicated that the prolonged asymptomatic phase and freedom from clinical illness experienced by the majority of HIV-2 infected individuals results from a lower level of virus production during infection than that observed in either HIV-1 infection or in the small number of HIV-2 infected individuals displaying relatively rapid progression to disease. The rate at which viral gene products are transcribed is one mechanism by which the rate of virus production can be controlled and is mediated by the Long Terminal Repeat (LTR) regions of the HIV-2 genome. A limiting dilution sensitive nested PCR has been developed to amplify the HIV-2 LTR from clinically and phenotypically characterised infected sources, including a number of HIV-2 isolates and uncultured PBMC samples derived from Gambian HIV-2 infected long-term non-progressor (LTNP) and rapid progressor (RP) patients. Using a highly efficient cloning system LTR amplicons have been cloned into a firefly luciferase reporter vector. A dual luciferase reporter assay has been used to determine LTR-directed basal and Tat transactivated levels of transcription in physiologically relevant cell lines. LTRs derived from LTNP patients tended to direct lower levels of basal and Tat transactivated transcription when compared to LTRs derived from RP patients. The difference between the two groups was more pronounced in the T cell-like Jurkat cell-line. Nucleotide sequence analyses of the LTR clones has revealed that the rate of general and G-to-A mutations at single functional sites within the LTR is higher in sequences derived from LTNP patients compared to RP patients. Taken together our data implies a relationship between LTR activity, sequence variation, and overall levels of virus expression in HIV-2 as evidenced by higher levels of circulating peripheral HIV-2 RNA in the patients with progressive disease profiles. Therefore, different LTR responsiveness may relate to different rates of virus production and disease progression rates and hence be a determinant in viral pathogenesis.

The potential exists for stress corrosion cracking (SCC) of carbon steel pipelines transporting fuel grade ethanol (FGE) and FGE- gasoline blends. The objectives of SCC 4-4 were to: 1. Develop data necessary to make engineering assessments of the feasibility of transporting FGE and FGE blends in existing pipelines, 2. Identify ethanol blends that can be transported in existing pipelines without significant modification of the system and operations (Case 1); blends that require significant modifications (Case 2) and blends that cannot be transported in existing pipelines, but could be moved in specially designed systems (Case 3), and 3. Characterize the time to initiation of SCC in a range of potent ethanol environments and identify safe operating and or batching practices that prevent the initiation and growth of SCC. The results of the research (Phase 1 and Phase 2) demonstrated that: 1. Pipelines made of common line pipe steels (e.g., Grade B and X-42 to X-60) are likely to be susceptible to ethanol SCC and any differences in susceptibility are probably not relevant from an integrity perspective. 2. While differences in susceptibility were noted for some weld types, in general, the base metal, heat affected zone, and weld metal were all susceptible to SCC in SFGE. 3. For sharp cracks, SCC initiation times are short once the line pipe steel is exposed to FGE or FGE blends capable of promoting SCC. 4. Once cracks initiate, crack growth rates are high in comparison with other forms of pipeline SCC. 5. Batching does not appear to be a viable method for SCC mitigation. 6. The only blends that can be safely transported in existing pipelines without significant modification of the system or operations (Case 1) are those containing less than 15% ethanol. 7. All other blends require significant modifications of the system or operations (Case 2), or specially designed systems (Case 3). 8. Case 2 could include deaeration of the SFGE, or the addition of inhibitors. 9. Case 3 is the subject of ongoing research (SCC 4-5).

Previous research and current efforts at control of babesiosis fall short of meeting the needs of countries where the disease is endemic, such as Israel, as well as the needs of exporting countries and countries bordering on endemic areas, such as the U.S. Our long-term goal is to develop improved methods of immunization against bovine babesiosis based on an understanding of the molecular mechanisms of immune protection and parasite targets of a protective immune response. In our previous BARD project, we established the basis for focusing on rhoptry antigens as components of a subunit vaccine against bovine babesiosis, and for additional research to better characterize rhoptry associated protein-1 (RAP-1) as a target of protective immunity. In this continuation BARD project, our objectives were to [1] optimize the immune response against RAP-1, and [2] identify additional rhoptry candidate vaccine antigens. The entire locus encoding B. bovis RAP-1 was sequenced, and the rap-1 open reading frame compared among several strains. Unlike B. bigemina, in which multiple gene copies with variant domains encode RAP-1, the B. bovis RAP-1 locus contains only two identical genes which are conserved among strains. Through testing of multiple truncated constructs of rRAP-1, one or more immunodominant T cell epitopes were mapped to the amino terminal half of RAP-1. At least one linear and one conformational B cell epitope have been demonstrated in the same amino terminal construct, which in B. bigemina RAP-1 also contains an epitope recognized by neutralizing antibody. The amine terminal half of the molecule represents the most highly conserved part of the gene family and contains motifs conserved broadly among the apicomplexa. In contrast, the carboxy terminal half of B. bovis RAP-1 is less well conserved and contains multiple repeats encoding a linear B cell epitope potentially capable of inducing an ineffective, T cell independent, type 2 immune response. Therefore, we are testing an amino terminal fragment of RAP-1 (RAP-1N) in an immunization trial in cattle. Cattle have beer immunized with RAP-1N or control antigen, and IL-12 with Ribi adjuvant. Evaluation of the immune response is ongoing, and challenge with virulent B. bovis will occur in the near future. While no new rhoptry antigens were identified, our studies did identify and characterize a new spherical body antigen (SBP3), and several heat shock proteins (HSP's). The SBP3 and HSP21 antigens stimulate T cells from immune cattle and are considered new vaccine candidates worthy of further testing. Overall, we conclude that a single RAP-1 vaccine construct representing the conserved amino terminal region of the molecule should be sufficient for immunization against all strains of B. bovis. While results of the ongoing immunization trial will direct our next research steps, results at this time are consistent with our long term goal of designing a subunit vaccine which contains only the epitopes relevant to induction of protective immunity. Parallel studies are defining the mechanisms of protective immunity. Apicomplexan protozoa, including babesiosis and malaria, cause persistent diseases for which control is inadequate. The apical organelles are defining features of these complex protozoa, and have been conserved through the evolutionary process, Past and current BARD projects on babesiosis have established the validity and potential of exploiting these conserved organelles in developing improved control methods applicable to all apicomplexan diseases.

The main objective of the proposed research was to study IQD1's mechanism of action and elucidate its role in plant protection. Preliminary experiments suggest that IQD1 binds CaM in a Ca²⁺-dependent manner and functions in general defense responses. We propose to identify proteins and genes that interact with IQD1, which may provide some clues to its mechanism of action. We also plan to dissect IQD1's integration in defense pathways and to study and modulate its binding affinity to CaM in order to enhance crop resistance. Our specific objectives were: (1) Analysis of IQD1's CaM-binding properties; (2) Identification of IQD1 targets;(3) Dissection of IQD1 integration into defense signaling pathways. Analysis of IQD1's CaM-binding properties defined four potential classes of sequences that should affect CaM binding: one is predicted to raise the affinity for Ca²⁺-dependent interaction but have no effect on Ca²⁺-independent binding; a second is predicted to act like the first mutation but eliminate Ca²⁺-independent binding; a third has no predicted effect on Ca²⁺-dependent binding but eliminates Ca²⁺-independent binding; and the fourth is predicted to eliminate or greatly reduce both Ca²⁺-dependent and Ca²⁺-independent binding. Following yeast two hybrid analysis we found that IQD1 interact with AtSR1 (Arabidopsis thalianaSIGNALRESPONSIVE1), a calcium/calmodulin-binding transcription factor, which has been shown to play an important role in biotic and abiotic stresses. We tested IQD1 interaction with both N-terminal or C-terminal half of SR1. These studies have uncovered that only the N-terminal half of the SR1 interacts with the IQD1. Since IQD1 has an important role in herbivory, its interaction with SR1 suggests that it might also be involved in plant responses to insect herbivory. Since AtSR1, like IQD1, is a calmodulin-binding protein and the mutant showed increased sensitivity to a herbivore, we analyzed WT, Atsr1 and the complemented line for the levels of GS to determine if the increased susceptibility of Atsr1 plants to T. ni feeding is associated with altered GS content. In general, Atsr1 showed a significant reduction in both aliphatic and aromatic GS levels as compared to WT. In order to study IQD1's molecular basis integration into hormone-signaling pathways we tested the epistatic relationships between IQD1 and hormone-signaling mutants. For that purpose we construct double mutants between IQD1ᴼXᴾ and mutants defective in plant-hormone signaling and GS accumulation. Epitasis with SA mutant NahG and npr1-1 and JA mutant jar1-1 suggested IQD1 function is dependent on both JA and SA as indicated by B. cinerea infection assays. We also verified the glucosinolate content in the crosses siblings and found that aliphatic GSL content is reduced in the double transgenic plants NahG:IQD1ᴼXᴾ as compare to parental lines while the aliphatic GSL content in the npr1-1:IQD1ᴼXᴾ and jar1-1: IQD1ᴼXᴾ double mutants was intimidated to the parental lines. This suggests that GSL content dependency on SA is downstream to IQD1. As a whole, this project should contribute to the development of new defense strategies that will improve crop protection and reduce yield losses and the amount of pesticides required; these will genuinely benefit farmers, consumers and the environment.

We have studied the regulation of the maize Ac and MuDR transposable elements activities during plant development. Ac was studied in an heterologous system (transgenic tobacco plants and cell suspensions) while MuDR was studied in the native maize background. The focus of this study was on the transcriptional regulation of Ac and MuDR. For Ac, the major achievements were to show that 1-It is autoregulated in a way that the Ac-encoded transposase can repress the activity of its own promoter; 2-It is expressed at low basal level in all the plant organs that were studied, and its activity is stronger in dividing tissues -- a behaviour reminiscent of housekeeping genes; 3- the activity of Ac promoter is cell cycle regulated -- induced at early S-phase and increasing until mitosis; 4- host factor binding sites were identified at both extremities of Ac and may be important for transposition. For MuDR, It was shown that it encodes two genes, mudrA and mudrB, convergently transcribed from near-identical promoters in the terminal inverted repeats. Distinct 5' start sites, alternative splicing, production of antisense RNA and tissue specificity were all shown to be involved in the regulation of MuDR.

In the recent past years there has been a considerable amount of effort devoted to developing methods and instruments to correct measured pipe-to-soil potentials for IR drops that may occur from currents (from the cathodic protection system or stray sources) in the soil to obtain the polarized potential. However, many of the methods or instruments available are either time-consuming, cumbersome to use in the field, applicable to only certain types of cathodic protection systems and under particular circumstances, subject to influences from stray current sources or not fully developed as of yet. Thus, there is a need to develop a practical method of determining the polarized pipe potential free of IR drop errors. This report examines and compares seven available methods for measuring polarized potential and describes the advantages and disadvantages of each. The following methods for measuring polarized potential were evaluated: (1) pipe-to-soil measurements with cathodic protection; (2) pipe-to-soil measurements with interrupted cathodic protection (also called instant-off); (3) measurements based on rectifier AC waveform analysis; (4) stepped current reduction; (5) a four-terminal network; (6) extrapolation to zero resistance (IR drop); and (7) a combination of DC/AC transverse measurements.

This study was motivated by the need to develop new means and approaches to the design of future, environmentally-safe, insecticides. Utilization of anti-insect selective toxins from scorpion venoms and clarification of the molecular basis for their specificity, are a major focus in this project and may have an applicative value. Our study concentrated on the highly insecticidal toxin, LqhaIT, and was devoted to: (I) Characterization of the neuropharmacological and electrophysiological features of this toxin. (II) Establishment of a genetic system for studying structure/activity relationships of the toxin. (III) Analysis of the insecticidal efficacy of an entomopathogenic baculovirus engineered and expressing LqhaIT. The results obtained in this project suggest that: 1) The receptor binding site of LqhaIT on insect sodium channels differs most likely from its analogous receptor site 3 on vertebrate sodium channels. 2) The effects of LqhaIT are presynaptic. Hyperexcitation at the neuromuscular results from dramatic slowing of sodium channel inactivation and enhanced peak sodium currents causes by LqhaIT. 3) The putative toxic surface of LqhaIT involves aromatic and charged amino acid residues located around the C-terminal region and five-residue-turn of the toxin (unpublished). 4) The anti-insect/anti-mammalian toxicity ratio can be altered by site-directed mutagenesis (publication 8). This effect was partly shown at the level of sodium channel function. 5) The insecticidal efficacy of AcNPV baculovirus increased to a great extent when infection was accompanied by expression of LqhaIT (publication 5).

Por consiguiente, esta nota de clase relaciona los diferentes esquemas de aprendizaje para que el estudiante de Ingeniería adquiera estructuras cognitivas y las pueda relacionar con las otras áreas del conocimiento. El documento trata de los elementos básicos, conceptos, reglas y técnicas para determinar si un argumento matemático es válido. Es fundamental conocer y manejar los conceptos de conjuntos numéricos, operaciones algebraicas, ecuaciones e inecuaciones de primer grado que pueden ser recordados de manera simultánea. Esta nota busca ser transversal y aplicarlo en todas las ingenierías. Los conceptos que se van a desarrollar empiezan por conjuntos, lógica matemática, inferencia lógica y álgebra de Boole, para terminar con compuertas lógicas. Asimismo, esta nota ayuda a determinar la validez de razonamientos lógicos tanto deductivos como inductivos, presentando ejercicios simples pero ilustrativos que permiten llegar de manera gradual hasta los que tienen un mayor grado de dificultad.
