La presente investigación tiene como objetivo comprender las diferencias y semejanzas existentes entre los modelos de negocios de las empresas de taxi remisse y taxi por aplicativo en Lima Metropolitana. Para la realización de este análisis se utilizó una herramienta que integra tanto los elementos del Canvas descritos por Osterwalder & Pigneur (2010), así como los elementos de los modelos de negocios de economía colaborativa descritos por Siustaike, Pilinkiene y Zviadauskas (2019). De esta manera, se entrevistó a los colaboradores de las empresas de taxi de economía colaborativa -Beat y Cabify- y las empresas de taxi remisse -Directo, Satelital, Remi Taxi y Taxi Real-; para luego describir cada uno de los elementos de los modelos utilizados en ambos tipos de empresa. Finalmente, se presenta la manera en que dichos elementos se integran dentro de las empresas, creando así los modelos de negocios con los que operan actualmente. Los principales hallazgos de la investigación fueron los siguientes: en relación con la variable proveedores, ambos tipos de empresa tienen el mismo modelo de contratación de conductores, en el cual el conductor no forma parte de la planilla y genera ingresos en función a los viajes que realiza. A nivel de la variable recursos clave, ambos tipos de empresa usan un aplicativo para la gestión de solicitudes, pero el aplicativo de las empresas remisse requiere de una persona intermediaria, mientras que el de las empresas de economía colaborativa realiza la conexión entre pasajero y conductor directamente. Por último, a nivel de actividades clave, las empresas de remisse estudiadas tienen como principal actividad la gestión de la calidad del servicio, mientras que, las empresas de aplicativos, el soporte a la aplicación cuando hay un descalce entre oferta y demanda.

This thesis presents two aspects of air transportation systems, capacity and efficiency. The first study improved a runway capacity simulation model for estimating the capacity of airports under various conditions. The main contribution of this study was to develop a simulation model which is able to analyze different airports with individual aircraft types. Many air traffic regulations were added to the simulation model to give more realistic results to the potential users of the model. Analysis of different separation systems between aircraft pairs show that the capacity of airports can improve by using more efficient separation systems specially under tough weather conditions. One of the outputs of the study, is an upgraded user interface which can be used by airport authorities for estimating their facilities' available capacity under different scenarios. The second study represents a new method for estimating unimpeded taxi times for taxing airplanes at airports. This study focused specifically on 6 large airports in the United States and the real ground radar data for all of the movements at those facilities. By tracking the real trajectories of every operation, the taxing behavior of each flight was analyzed. A standalone application was designed to summarize the flights information at each airport and represent the taxing behavior. The results show that using the ground radar data at airports can represent the taxing behavior in the highest fidelity.

Taxis are a popular mode of travel in Singapore because of their convenience and high levels of comfort. Despite having approximately 28,000 taxis, Singapore’s taxi system is under-optimized. As a result, e-hailing applications have transformed the way consumers select ride services. Such apps have provided a faster and more convenient service. In fact, both consumers and taxi drivers have reaped significant benefits from e-hailing services. In our study, we use average wait times and prices to understand the impact e-hailing applications have had on the Singapore taxi industry. We predict that such services would have negatively impacted the existing taxi dispatch system. We found e-hailing applications reduced the average wait time for passengers. Prices were the cheapest for Uber. Due to limitations in the study, we cannot conclude if these are statistically significant results.

Creación de una empresa de servicio de taxi exclusivo para niños y adolescentes, el cual tiene como finalidad unir a dos clientes, por un lado los padres, quienes contratarán el servicio como tal para transportar a sus hijos y por otro lado los conductores, quienes realizarán la tarea de transportar a los menores. Los autos serán conducidos por damas para brindar una mayor confianza a los padres. El servicio cuenta con un aplicativo que permite al cliente monitorear el desplazamiento del vehículo mediante el sistema GPS y a su vez le permite visualizar las imágenes de sus hijos en el vehículo. Además el servicio cuenta con una central telefónica, para cualquier requerimiento o emergencia. Se ha determinado el tamaño del Mercado Total, padres con hijos entre 7 y 16 años del nivel socio económico A B de los distritos de La Molina, Miraflores, San Borja, San Isidro y Santiago de Surco Respecto a la participación del mercado, se ha considerado un 5%, con crecimiento anual del 3% cifra conservadora y bastante cercana a las estimaciones del PBI del año 2018. La estimación de la frecuencia de uso por cliente es de 3 veces por mes. Los precios se han estimado por zonas de traslado con un rango entre S/ 22 y S/ 27. Con los datos anteriores se ha determinado la demanda esperada y los niveles de ingreso con un escenario de 5 años. El costo del accionista y del proyecto con el método del CAPM y del WACC respectivamente, para determinar los indicadores de rentabilidad TIR y VAN los cuales arrojan cifras bastantes atractivas.

Taxon sampling is explicitly examined using simulations and probability analyses. The simulations focus on the ability of two different sampling strategies to correctly reconstruct relationships among subgroups of taxa. One sampling strategy chooses taxa randomly, the other strategy chooses taxa to increase the phylogenetic dispersal of the taxa. Also considered is whether the inclusion of a single basal taxon will increase the frequency of correctly reconstructing the relationships among subgroups. Inclusion of the basal taxon is found to decrease success (correct subgroup relationships). The strategies are found to differ, primarily due to differing tendencies to include the basal taxon (the random strategy includes the basal taxon less frequently and outperforms the dispersed strategy). The inclusion of a basal taxon is also found to decrease the probability of correctly reconstructing ancestral character states. The probability of randomly choosing a single taxon from one side of the root is developed.

Estudios realizados a mediados del 2016 nos indican que las empresas de Taxi por aplicación movilizan alrededor de S/. 240 millones al año y que estos sólo representan un 10% de las transacciones de taxi en todo Lima, cifras que nos hablan de un gran potencial de mercado. Al ser encuestados los usuarios de las aplicaciones de Taxi, estos revelan tres inquietudes referentes al servicio: El primero de ellos es el alto costo de utilizar este servicio sobre todo en horas punta, el segundo se refiere al tiempo de espera y el último a la inseguridad. Nuestra idea de Negocio, busca participar de este importante mercado en crecimiento, brindando alternativas a los usuarios, que den solución a los inconvenientes que hoy manifiestan. Nuestro proyecto, Taxi Mix, es una aplicación que reúne a todas las aplicaciones de taxi en una misma pantalla permitiendo comparar precios, tiempo de espera y calificación de los conductores. Buscamos que el usuario pueda comparar según sus requerimientos entre todas las aplicaciones de taxi disponibles de una manera práctica y eficiente para poder tomar la mejor decisión. Pero, ¿quién es el usuario? Nuestro usuario son personas de ambos géneros residentes de la ciudad de Lima, cuyas edades oscilan en el rango de 18 a 55 años principalmente y que a su vez utilizan aplicaciones de taxi para movilizarse. La principal fuente de ingresos de nuestro negocio provendrá del cobro de una comisión de treinta (30) centavos de sol por cada transacción que se realice desde nuestra aplicación y sea derivada a la empresa de Taxi elegida. Las empresas de Taxis por aplicación serán las que nos paguen por cada transacción derivada. Este precio lo fijamos luego de revisar el mercado potencial al cual podemos acceder, la respuesta que estimamos obtener del mismo y los costos en los que tendremos que incurrir. No descartamos la posibilidad de cobrar en un futuro por venta de publicidad a empresas que deseen anunciar en nuestra aplicación, la cual no sólo ofrecerá un gran número de contactos, sino también la segmentación adecuada a cada marca para que su costo de conversión sea muy competitivo y puedan optimizar al máximo sus presupuestos. Siendo muy conservadores en nuestros supuestos, estimamos que el proyecto tendrá un índice de rentabilidad de 1.71, es decir por cada unidad monetaria que se invierta en él, obtendremos 1.71 veces lo invertido, en un período máximo de recuperación de la inversión de 2.23 años. La rentabilidad esperada es muy alta, haciendo de este, un proyecto totalmente viable. Por último tenemos una pregunta más por responder y es la siguiente ¿el modelo de negocio es escalable a otras plazas? La respuesta es un rotundo si y no solo eso, creemos que los resultados pueden ser mucho mejores debido a la porción de mercado con la que cuentan las aplicaciones de taxi en otros mercados, que son por mucho, superiores a las que tenemos en la ciudad de Lima.

La presente tesis tiene como propósito sustentar “Taxi de la Guarda” como una idea de negocio diferenciadora, novedosa, atractiva y, sobre todo, rentable, en el marco de una estrategia de desarrollo de una propuesta de valor. El Perú, al 2010, acumuló nueve años consecutivos de crecimiento económico y en lo que va del 2011 el crecimiento acumulado alcanza el 7.51%. Ello se ve reflejado directamente en la capacidad adquisitiva del peruano promedio: el PBI per cápita se duplicó en los últimos 10 años y se proyecta que la tendencia se mantenga en el mediano y largo plazo. Este hecho genera un espacio propicio para nuevas y mejores oportunidades de negocio y ello es así no sólo por una evidente mayor disponibilidad de gasto; sino también, porque el estilo del consumidor se vuelve más sofisticado, exige mayor calidad y, precisamente, por esa percepción de progreso se vuelve más exigente en tiempos y valor en todos los servicios que recibe. En esa línea, actualmente se viene invirtiendo millones de soles a nivel nacional en la implementación de mejoras en: infraestructura vial, educación, salud, telecomunicaciones, seguridad nacional y transporte de personas. En el caso de este último, el gobierno regional de Lima con el gobierno nacional, han destinado millones de dólares en implementar tanto el Metropolitano como el tren eléctrico. Actualmente, la seguridad en el transporte se ha vuelto un tema crítico, especialmente en el transporte público. La cantidad de personas que sufren asaltos o que se ven afectadas físicamente por accidentes vehiculares es numerosa. Incluso los taxis independientes dejaron de ser, hace ya muchos años, un medio de transporte seguro y confiable. La mayor proporción de personas afectadas por los problemas actuales en el sistema de transporte son las mujeres, quienes día a día reportan casos de vejaciones, faltas de respeto, robos, secuestros e incluso intentos de violación. Del estudio de mercado se desprende que las empresas actuales que cubren el servicio de taxi para mujeres son percibidas por éstas como inseguras y con un mal servicio. En este contexto surge “Taxi de la Guarda”, como una nueva empresa destinada a satisfacer una necesidad de transporte seguro, confiable, con altos estándares de calidad, confort y con una estructura de costos rentable, con el objetivo de hacer del viaje en taxi toda una experiencia de compra gratificante y segura. Según se podrá observar en el desarrollo de la presente tesis, el proyecto de negocio resulta altamente beneficioso en términos económicos, con una inversión inicial de S/. 299,000 aproximadamente y una tasa interna de retorno de 61%. En menester indicar que la proyección de ventas ha seguido una lógica conservadora en el sentido de estimar alcanzar, al término del proyecto, el 10% de la demanda total encontrada en el estudio de mercado. En esa misma línea, para los diez años del proyecto, el porcentaje del nivel de uso de la capacidad total que se ha utilizado en la presente tesis se encuentra 20% debajo del nivel de ocupación del sector. Producto de ello, los gastos fijos unitarios se ven incrementados y, con ello, el grado de apalancamiento también sufre un aumento. Sin embargo, aún cuando la proyección de ventas es conservadora y los costos fijos unitarios se ven incrementados, el proyecto resulta rentable.

In this work we propose a discrete predator-prey model with the purpose of analyzing the spatio-temporal dynamics of herbivorous and their predators. For that, we explicitly consider the dispersion of both species through Coupled Map Lattice models, that is, systems of difference equations coupled by dispersal. We suppose that the prey population presents density-dependent growth and predators, Holling type II functional response. In order to analyze the effects of prey-taxis showed by predators, which means, that predators direct their movement to regions of high prey densities, we computationally implemented our model for different movement schemes: difusion for both species, immobile prey and taxis for predators and, difusion for prey and taxis for predators. We conclude that the dispersal behavior of predators and preys plays a crucial role on their spatio-temporal dynamics and hence cannot be omitted when modeling predator-prey interactions.
Neste trabalho construímos um modelo presa-predador discreto com objetivo de analisar a dinâmica espaço-temporal de insetos herbívoros e seus predadores. Para tal, consideramos explicitamente a dispersão de ambas as espécies através de modelos de Rede de Mapas Acoplados, isto é, sistemas de equações a diferenças acopladas pela dispersão. Consideramos que as presas apresentam crescimento dependente da densidade e os predadores, resposta funcional Holling do tipo II. Para analisar os efeitos da movimentação de taxia dos predadores, isto é, o movimento direcionado às regiões de maiores densidades de presas, implementamos computacionalmente o modelo para diferentes esquemas de movimentação: difusão para ambas as espécies, presas imóveis e taxia para predadores e difusão para presas e taxia para predadores. Concluímos que o comportamento de movimentação de presas e predadores tem um papel relevante na sua dinâmica espaço-temporal e não pode ser ignorado na modelagem destes sistemas.

This research focuses on utilizing the typically overlooked taxi manifest data to analyze taxi operations with respect to transit, and also presents alternative uses for the data in transportation planning. Taxi travel characteristics are explored for Arlington, Virginia, a county containing both urban and suburban qualities. Previous research contends that manifest data can provide valuable quantitative descriptors of taxi travel. This thesis attempts to describe taxi travel by quantifying trip characteristics; the shortcomings of using manifest data are discussed and the results are reported. The taxi operations results are then compared for weekend and weekday travel and also for airport and non-airport bound travel. Several key differences between these groups of taxi trips are discussed. Next, an investigation of the relationship between mass transit facilities and taxi travel is conducted. Because taxis provide a complementary yet competing public transportation service to mass transit, it is hypothesized that examining the proximity to transit options and the timing of taxi trips can provide insight to the perceived gaps in mass transit services. However, the data show that simply examining geographic or temporal characteristics of taxi trips does not define clear relationships between transit facilities and taxi use. Instead, the results suggest that other variables such as land use and vehicle access may hold a greater influence over the generation of taxi trips. Despite the difficulty in using manifest data to determine gaps in transit, the data collected by taxi regulators could have numerous applications for planners. Possible applications for the type of taxi data used in this research are explored and a potential data flow for agencies is proposed.

Talin is a 270 kDa cytoskeletal protein implicated in integrin-mediated cellular interactions with the extracellular matrix. In the current study, the three vinculin-binding sites in talin have been further defined, using a yeast two-hybrid approach, and were found to be contained within short amphipathic -helices encompassing residues 607-636, 852-876 and 1945-1969. Each of these helices was found to compete for the same binding site within the vinculin head, and to bind with similar rate constants. The talin-binding site in vinculin was further defined to residues 1-167. Fluorescence assays have been used to demonstrate that a talin polypeptide encompassing residues 2270-2541 binds and bundles F-actin, and that native talin does not nucleate polymerisation of actin. In order to identify novel binding partners for talin, a polypeptide encompassing talin residues 1856-2301 was used to screen a cDNA expression library using a yeast two-hybrid approach. The library screen identified a single novel talin-binding protein that has been named TIP1 (talin-interacting protein-1). TIP1 binds specifically to talin in both the two-hybrid system and in vitro binding assays, and searches of genomic and EST databases have uncovered homologues in human, mouse and rat. The full-length human cDNA has been mapped and is predicted to encode a 300 kDa protein that does not bear similarity to known proteins. The size of the TIP1 protein has been confirmed by production of a polyclonal antibody followed by Western blotting of MEF protein extracts.

Speciation and gene duplication are fundamental evolutionary processes that enable biological innovation. For over a decade, biologists have endeavored to distinguish orthology (homology caused by speciation) from paralogy (homology caused by duplication). Disentangling orthology and paralogy is useful to diverse fields such as phylogenetics, protein engineering, and genome content comparison. A common step in ortholog detection is the computation of Bidirectional Best Hits (BBH). However, we found this computation impractical for more than 24 Eukaryotic proteomes. Attempting to retrieve orthologs in less time than previous methods require, we developed a novel algorithm and implemented it as a suite of Perl scripts. This software, Selectable Taxon Ortholog Retrieval Iteratively (STORI), retrieves orthologous protein sequences for a set of user-defined proteomes and query sequences. While the time complexity of the BBH method is O(#taxa^2), we found that the average CPU time used by STORI may increase linearly with the number of taxa. To demonstrate one aspect of STORI’s usefulness, we used this software to infer the orthologous sequences of 26 ribosomal proteins (rProteins) from the large ribosomal subunit (LSU), for a set of 115 Bacterial and 94 Archaeal proteomes. Next, we used established tree-search methods to seek the most probable evolutionary explanation of these data. Virginia Commonwealth University, 2009 Dr. William C. Bosher, Jr. Distinguished Professor of Public Policy and Education The purpose of this study is to determine to what degree local government officials in Virginia support fiscal autonomy for locally elected School Boards in the state. Currently School Boards in Virginia do not have the ability to raise their own revenues and must depend on the local City Council or Board of Supervisors to appropriate school funding each year. Many more states in the nation allow local School Boards to raise their own revenues than not, and some would argue that Virginia’s system is an inferior form of local government having a negative effect on K-12 education in the state. Others would argue that Virginia’s current system works quite well and to give taxing authority to local School Boards would degrade the quality of K-12 education in the state. To set the national context for this question a comparative survey was done of all fifty state systems in the nation to learn the differences between state systems for funding K-12 education. Second, a survey was conducted of 1,782 Virginia public officials whose professional lives would be affected by a change to allow local School Boards to raise their own revenue. These officials were asked a variety of questions the answers to which tell us whether they view fiscal autonomy for School Boards as progress. Finally, statistical analyses are performed on the responses to the survey using the public officials’ positions and their region of the state as variables. Through this statistical analysis we are able to determine whether position or region of the state have a significant affect on answers to the survey questions.

La idea de negocio parte de la necesidad de los autores por movilizarnos rápido, seguro y fácil por la ciudad, sin utilizar medios de transporte públicos que hacen que la experiencia de viajar sea incómoda, por la alta demanda del servicio y poca oferta. Esta situación, que se presenta a diario en miles de limeños, puede ser solucionado mediante una aplicación que simplifique el proceso de intercambio de información y nos evite caer en situaciones incómodas a las que los taxis colectivos tradicionales nos exponen. Asimismo, luego de la evaluación financiera preliminar del proyecto, notamos que el mismo tiene un potencial de rentabilidad altamente explotable, que constituye una oportunidad inmejorable para ser los primeros en este segmento, logrando lo que en su momento consiguió empresas como Uber o Globo. Además, se requiere una inversión relativamente pequeña frente a un negocio convencional y será totalmente accesible a través de las tiendas de Android y Apple. Finalmente, este proyecto será nuestra mejor carta de presentación e ingreso en el mundo de los negocios para nuestros futuros proyectos, ya que hemos recabado la información necesaria para entender la operación de ellos.
The project that we present in this document, which we decided to call "Habla, Vas", consists of a connection system through relevant information of demand and supply, between people who need to move in a group through the main routes of the city and groups who need to generate income on these routes with the highest traffic flow in Lima. The business idea stems from the authors' need to move quickly, safely and easily around the city, without using public means of transport that make traveling uncomfortable, due to the high demand for the service and low supply. This situation, which occurs daily in thousands of Lima, can be solved through by the application that simplifies the process of information exchange and prevents us from falling into uncomfortable situations to which traditional groups expose us. Likewise, after the preliminary financial evaluation of the project, we note that it has a highly exploitable profitability potential, which constitutes an unbeatable opportunity to be the first in the segment, achieving what companies such as Uber or Globo achieved at the time. Also, a relatively small investment is required compared to a conventional business and will be fully accessible through Android and Apple stores. Finally, this project will be our best letter of introduction and entry into the business world for our future projects, since we have collected the necessary information to understand their operation.

Events have been categorized, modeled and recorded by researchers and practitioners for many centuries. Life and events are always being the philosopher's topic of debate. For us, event is any happening worth of remembering. This thesis makes an in-depth philological query of the nature of events and their intricate relationship to other events in the tapestry of complex social structures. We tried to understand our life events from grainy to vast in nature and size. Causation and effects are investigated and a simplified model is proposed for a user centric personal event management system which is fundamentally different from any existing system. Facts as a priori and stories as a posteriori has been separated by formal definition. Novel visualization and interaction is proposed to meet every individual's needs. The concept of lifelines has been introduced for the organizational requirements of a single person's life events that made it possible to distinguish from being the part of an event and being the witness of an event. This visualization model made it easier to manage causal relationships between events. Rich and intuitive interaction has been developed and proposed through the user-centric design process.

Anställda behöver transporteras till och från sammankomster och för detta är taxi ett möjligt färdmedel. Claremont är ett Stockholmsbaserat IT-konsultföretag som alltid har behov att utveckla sina 220 konsulters kompetens, där taxi är ett vanligt transportmedel till sammankomster eller flygplatser. För att minska utgifter för transporter kan samåkning med taxi ske. I dagsläget sker logistikplaneringen för samåkning genom att företaget själva eller taxibolag manuellt utan hjälp av datorkraft försöker ta fram möjliga rutter för samåkning, rutter som i slutändan inte alltid blir bra. Kan denna process automatiseras och beräknas med datorkraft kan bra ruttförslag ges vilket resulterar i att planeringstid, restid och pengar sparas. Problemet samåkning med taxi från en plats till flera eller från flera platser till en har modellerats som en instans av Vehicle Routing Problem. Vehicle Routing Problem beskriver hur många och i vilken ordning ett eller flera fordon ska besöka olika destinationer. En systemarkitektur har tagits fram där adresser matas in och ett förslag på rutter returneras till användaren i form av en lista med adresser, där adresserna är grupperade i olika rutter. Visualisering sker även på en karta för kontroll av lösningen och ackumulerad tid samt uppskattat pris visas. Systemarkitekturen använder sig av avancerade algoritmer och heuristiker i Open Source Routing Machine och Optaplanner för att lösa problemet.
In order for companies’ staff to improve their knowledge employees may need to be sent to conferences and courses. Employees needs transport to and from gatherings and for this taxi is a possible mean of conveyance. Claremont is a Stockholm based IT-consulting firm that always have the need to improve their 220 consultants’ competence, where taxi is a common mode of conveyance to gatherings or airports. To reduce expenditures, it is possible to use ridesharing with taxis. Currently the logistics planning for ridesharing is performed by the company themselves or taxi companies manually without help from computer power, where they try to produce possible routes for ridesharing that in the end not always turns out good. If this process can be automatized and calculated with computer power it is possible to give satisfactory route suggestions which results in savings in planning time, travel time and costs. The problem ridesharing with taxis from many-to-one or one-to-many locations has been modelled as an instance of the Vehicle Routing Problem. The Vehicle Routing Problem describes how many and in which order one or multiple vehicles should visit multiple destinations. A system architecture has been developed where addresses are inputted and a suggestion for routes is returned to the user in the form of lists of addresses, where the addresses are grouped in different routes. Visualization is also performed on a map for checking the solution and accumulated time with approximated price is displayed. The system architecture uses advanced algorithms and heuristics in the Open Source Routing Machine and Optaplanner to solve the problem.

According to U.S. government surveys, 12% of Americans used taxi service in the previous month' and spent about $3.7 billion a year for cab fare.2 Taxi service is one of the major modes of public transportation. Despite providing services 24 hours a day, driving relentlessly with an empty taxicab in search of passengers and answering dispatch calls instantaneously, taxi service is ranked the most unsatisfactory mode of transportation by the public. Charging higher fares than other major modes of transportation and averaging 10 to 12 hours work day, taxi drivers have a difficult time to earn a sustainable income.Approximately half of all the taxi mileage is paid mileage; this means a significant portion of a taxi's time and fuel is spent on non-revenue generating activities, i.e. without passengers. Current taxi allocation is inefficient. The number of taxis and the geographical service areas which they serve are heavily regulated in most cities. With limited competition and strict regulations, taxi service suffers with customers having to endure long wait times and inferior services. The current taxi systems in most U.S. cities may be greatly improved from their current state.
(cont.) This thesis investigates the factors of inefficiency in the current taxi system, reviews previous taxi efficiency studies, and suggests possible solutions. After extensive literature reviews and field research, a computer simulation model has been built in the MATLAB environment. This computer model tests various attributes that affect logistic optimizations for taxi services. In particular, the effect of taxi fleet size, the quantity of hotspots, and the concentrations of customers at hotspots are analyzed in detail using the model. The metric of interest includes the customers' wait time, taxi revenue, and costs of operations. Results from the computer simulation experiments, field research, and literature review are analyzed and synthesized. Possible solutions are proposed as part of this thesis.

The primary objective of this thesis is to provide a new approach to optimizing the supply of taxis as applied in UK cities. Taxi supply, both in the UK and elsewhere, is controlled by a series of regulations (defined in relation to controls affecting Quantity, Quality and Economics - QQE) that influence the ability of the market to respond structurally - in terms of access to the market, and operationally - affecting the ability to provide services within the market. This thesis identifies the existing structures in taxi regulation. It explores legislative disagreement and differences in both academic and practitioner literature and perspectives. The thesis provides both a new approach to, and a new model of, taxi regulation, which accommodates existing differences in regulatory structures. The thesis builds on existing methods applied in determining individual elements of control - which may be appropriate to some elements of control but fail to address a full cross section of impacts - and provides an enhanced approach and new modelling framework for taxi regulation. In constructing this approach use has been made of both survey and case study methodologies. Survey data has been collected from two surveys, of 52 cities and 21 licensing authorities respectively. The case studies used are of Glasgow! and West Dunbartonshire(2). The thesis concludes that existing approaches to regulation are conducted in separate regulatory domains without sufficient comprehension of the impacts of action in one regulatory domain on another. It also concludes that the instruments used in the assessment of the impacts of regulation on taxi supply are insufficiently specified and inadequately coordinated. It is possible to identify issues across regulatory domains that can be improved to better optimize supply appropriate in any given circumstance to the benefit of existing and potential passengers - this includes those with particular access needs. In optimizing supply, awareness of the needs of the taxi industry and its regulators has been an important element of consideration. The thesis makes recommendations for alterations in the application of standard methods of assessment of taxi supply. 1 Although central to the Strathclyde conurbation, the Glasgow licensing area relates to the central city area only (Clydebank, Paisley, etc. being included in separate council licensing areas). The city is thus defined as medium sized location. 2 Further focus city material bas been collected from Belfast and Edinburgh, which is included in comparison within the text, and summarised in the appendices of this document.

The Kompaï robot was partly developed for the purpose of assisting the elderly and people with special needs. The major related functionalities addressed in the thesis are navigating the Kompaï robot in a given environment and exhibiting the position taxis behavior.The overall objective of the project is to incorporate the Kompaï with the above mentioned features, thus supporting the idea of a robot companion to humans and also creating a platform by which future innovate ideas could be developed on the Kompaï robot.To achieve this, a cognitive system was developed which allows the Kompaï robot to display follow path behavior and position taxis behavior. The system encapsulates inter related activities such as representing the environment as a two dimensional grid map, developing a strategy to plan a path towards the destination, executing motor actions to follow the path, estimating the position of the robot as it follows the path in the map by acquiring and interpreting sensory information and developing a strategy that allows the Kompaï robot to reflexively move towards an object/RFID tag position in the environment.The results from the experiments show that the developed cognitive system, allows the Kompaï robot to navigate between two locations in the given environment and exhibit position taxis behavior.

Integrins are large heterodimeric (αβ) cell surface receptors that play a key role in the formation of focal adhesion complexes and are involved in various signal transduction pathways. They are ‘activated’ to a high affinity state by the formation of an intracellular complex between the membrane, the integrin β-subunit tail and talin, a process known as ‘inside-out activation’. The head domain of talin, a FERM domain homologue, plays a vital role in the formation of this complex. Recent studies also suggest that kindlins act in synergy with talin to induce integrin activation. Despite much available structural and functional data, details of how talin activates integrins remain elusive. In this thesis a multiscale simulation approach (using a combination of coarse-grained and atomistic molecular dynamics simulations) together with NMR experiments were employed to study talin-mediated integrin inside-out activation. A number of novel insights emerged from these studies including: (i) the crucial role of negatively charged lipids in talin/membrane association; (ii) a novel V-shape conformation of the talin head domain which optimizes its interactions with negatively charged lipids; (iii) that interactions of talin with negatively charged moieties in the membrane orient talin to optimize interactions with the β cytoplasmic tail; (iv) that binding of talin to the β cytoplasmic tail promotes rearrangement of the integrin TM helices and weakens the integrin α/β association; and (v) that an increase in the tilt angle of the β integrin TM helix initiates a scissoring movement of the two integrin TM helices. These results, combined with experimental data, provide new insights into the mechanism of integrin inside-out activation. The same multiscale approach was used to demonstrate the crucial role of the Gx3G motif in the packing of the integrin transmembrane helices. Using recent structural data the integrin/talin complex was modelled and inserted in bilayers which resemble the biological plasma membrane. The results demonstrate the dynamic nature of the integrin receptor and suggest that the integrin/talin complex alters the lipid organization and motion in the outer and inner bilayer leaflets in an asymmetric way and that diffusion of lipids in the vicinity of the protein is slowed down. The work in this thesis demonstrates that multiscale simulations have considerable strengths when applied to investigations of structure/function relationships in membrane proteins.

Miniaturization of on-board actuation and powering engenders the proliferation of biohybrid microrobots, which integrate motile bacteria or cells with synthetic functional components to achieve micron-scale actuations. Flagellated bacteria like S. marcescens are among the leading candidates for the actuators of swimming microrobots. However, the high intrinsic stochasticity in bacteria-driven microrobots severely limits their potential applications, such as targeted drug delivery. Taxis behaviors (e.g., chemotaxis), which help free-swimming bacteria to navigate towards favorable environments and away from hazardous ones, may offer an elegant means to control the motion of bacteria-driven microrobots. Therefore, this thesis focuses on: (a) addressing the motion guiding of bacteria-driven microrobots using common bacterial taxis behaviors, specifically chemotaxis and pH-taxis, (b) explaining the physical mechanisms associated with the tactic motions in bacteria-driven microrobots, and (c) developing a biophysical model to describe the bacterial propulsion and the chemotaxis in bacteria-driven microrobots. In order to produce considerable chemotactic motion in bacteria-driven microrobots, an appropriate chemical concentration profile needs to be determined, which requires the knowledge of the chemotaxis response of the integrated bacterial species. Thus, we first propose an experimental and modeling framework to characterize bacterial chemotaxis. The chemotaxis response of a species against a chemoattractant is experimentally quantified under a linear concentration gradient of the attractant. A signaling pathway model is fitted to the experimental measurements over a series of gradients to determine the species-specific parameters in the model, thereby fulfilling an analytical characterization of the chemotaxis. Subsequently, in a multi-bacteria-driven microrobotic system, we quantify the chemotactic drift of the microrobotic swarms towards a potent chemoattractant L-serine and elucidate the physical mechanisms associated with the drift motion by statistical trajectory analysis. It shows that the microrobots have an apparent heading preference for moving up the gradient, which constitutes the major factor that produces the chemotactic drift. The apparent heading bias is caused by a higher persistence in the heading direction when a microrobot moves up the the L-serine gradient compared to traveling down the gradient. Besides chemotaxis, we explore the potential of utilizing ambient pH to guide the motion of the bacteria-driven microrobots. Under three different pH gradients, we demonstrate that the microrobots exhibit both unidirectional and bidirectional pH-tactic behaviors. Two factors, a swimming heading bias and a speed bias, are found to be responsible for the pH-tactic motion while the heading bias contributes more. Like in chemotaxis, the heading directions of the microrobots are also significantly more persistent when they move towards favored pH regions. Finally, a biophysical model is developed to describe the bacterial propulsion and the chemotaxis in an extensively adopted design of bacteria-driven microrobots. The model traces helical trajectories and chemotactic motion that resemble those observed from experiments, which validates the basic correctness of the model. The model simulation also suggests that the seemingly collective chemotaxis among the multiple bacteria attached to a microrobot could be explained by a synchronized signaling pathway response among these bacteria. La découverte de nouveaux fossiles au Pléistocène moyen a entrainé la révision de l’espèce Homo heidelbergensis. La morphologie particulière de la mandibule de Mauer (holotype de l’espèce) et l’intégration de nombreux fossiles ne présentant pas de mandibule dans le taxon ne permettent pas l’émergence d’un consensus: une espèce afro-européenne ancêtre des Homo sapiens et des Néandertaliens, ou un taxon européen ancêtre des Néandertaliens. Je teste la validité de l’espèce à l’aide de deux approches: une analyse morphologique (méthode usuelle d’identification d’espèces) et une analyse par morphométrie géométrique sur la mandibule, la face et la calvaria de 134 fossiles du Pléistocène et de 128 Hommes actuels. 139 caractères (mandibule: 39, face: 35, calvaria: 65) ont été analysés grâce à une analyse de morphologie comparée et à une analyse phénétique pour obtenir des groupes basés sur les similarités entre spécimens. Une seconde analyse par morphométrie géométrique basée sur 30 points repères (mandibule: 8, face: 8, calvaria: 14) me permet d’étudier la structure géométrique des spécimens. Les résultats des analyses morphologiques soutiennent Homo heidelbergensis en tant que taxon valide. Il peut être différencié des autres Homo sp. Du Pléistocène et défini par une liste de traits morphologiques. Leur présence chez des fossiles Africains et Européens supporte l’hypothèse d’un taxon afro-européen ancêtre des Néandertaliens et des Homo sapiens. L’analyse par morphométrie géométrique montre que la structure géométrique des spécimens du taxon est proche de celle de la plupart des autres Homo sp. Du Pléistocène. Je propose une révision de la diagnose d’Homo heidelbergensis Schoetensack, 1908.

It is widely accepted that taxi fare regulation in Hong Kong has been adopted for a long time without posing serious problems. The government and most of the taxi operators and passengers are used to such a regulatory measure. Nevertheless, a series of economic downturn and fierce competition among various public transport modes and within the taxi trade have emerged in the recent decades. These have gradually changed this mindset. Some people start to question whether fare regulation is the best option. Even Transport Advisory Committee supports that Hong Kong should, in the long run, adopt a more flexible fare system which should allow taxi operators to set their own fare. Against this background, this dissertation is conducted with a view to answering a question: “to what extent should the taxi fare system in Hong Kong be regulated or de-regulated”. With a view to providing recommendations on the future taxi fare policy, this dissertation examines thoroughly the theoretical principles and foreign experiences in taxi fare regulation and de-regulation. Local market structure and public perception (including the government, taxi operators and passengers) of fare regulation / de-regulation are also explored to see if the theories and overseas experiences are applicable to Hong Kong. This is a pioneering study on the feasibility of fare de-regulation in Hong Kong. The study results should be significant and should point a direction to the future of how to tackle the above question. Overall, the dissertation shows that the concept of fare regulation is deep-rooted in the mind of the general public. The government and taxi operators are very resistant to fare semi-regulation (i.e. maximum and/or minimum fare) and de-regulation on the grounds of potential cut-throat competition and adverse social impacts, e.g. overcharging and consumer’s confusion. While taxi passengers are generally worried about the loss of legal protection and fairness of charging secured by the regulation. However, theoretical principles and foreign experiences do prove some fruitful results that can be achieved by fare semi-regulation / de-regulation. For instance, the availability of more price options and service improvements are beneficial to customer welfare while the possible rise of fare level is favourable to the business of operators. To strike a balance between different interests, this dissertation recommends that Hong Kong should adopt a taxi fare regulation in the form of a combination of maximum and minimum fare. That is, a price ceiling and floor are prescribed by the government and taxi operators are allowed to set their fare level within this range. The only requirement for the operators is to file the fare to the government for record purpose as and when the fares are changed. These fares should be widely publicized before implementation. To save the effort by customers to ascertain and compare the prices, it is suggested that all taxi operators are required to be affiliated to radio-dispatched centers (DCs). DCs will take up the role of fare setting and all subordinated taxis must follow suit. The government should closely monitor the level of maximum and minimum fares and make necessary adjustments in response to market conditions.

Nedávné studie předpovídají nárůst pasažérů využívajících leteckou dopravu. Tento trend bude vyžadovat zavedení nových leteckých linek, důsledkem čeho bude zhuštěn letový provoz s dopadem hlavně na nápor letišť v metropolitních oblastech. Automatizovaně řízení pojíždení letounu umožní menší rozestupy mezi jednotlivými linkami a zvýšení příletové a odletové kapacity letišť. Tato práce se zabývá návrhem modelu pohybu dopravního letounu po zemi s ohledem na různé provozní podmínky jako např.: stav povrchu vzletové a přistávací dráhy za různého počasí a lišící se provozní parametry letounu (tlak v pneumatikách, zatížení podvozků a pod.). Validace modelu byla založena na sledování poloměru zatáčky pro různe uhly natočení přední podvozkové nohy. Výsledky simulace byly validovany vzhledem k analytickému modelu Ackermanovy geometrie a na specifikační dokument od Boeingu určený pro plánovaní pohybu letounu na letišti. Výsledky prokázaly přesnost modelu a potvrdily jeho možné nasazení pro simulace v reálnem čase.

Svarbiausias iš jų yra mokestinių pajamų didinimas skatinant savanorišką mokesčių mokėjimą. Tai galima pasiekti tik tuomet, kai mokesčių mokėtojas pasitiki valstybe. Todėl svarbu užtikrinti tinkamą mokesčių mokėtojo teisių apsaugą. Mokestinių ginčų teisena yra vienas iš teisinių mechanizmų, skirtų realizuoti mokesčių mokėtojo teisę į gynybą. Magistriniame darbe buvo iškelta hipotezė – mokestinių ginčų teisenos reglamentavimas sudaro sąlygas pažeisti mokesčių mokėtojo teises. Magistrinį darbą sudaro įvadas, keturi skyriai, išvados ir pasiūlymai. Pirmame skyriuje nagrinėjamos pagrindinės sąvokos. Antras skyrius skirtas mokestinių teisės aktų analizei, siekiant nustatyti pagrindines mokesčių mokėtojo teises mokesčių administravime. There is analysis of regulations of taxing disputes’ legal proceedings in Lithuania and in foreign countries performed in this master’s study. The goal of the study is to find out if regulations of taxing disputes’ legal proceedings in Lithuania allow breach of the taxpayer’s rights, which are given him in administration of taxes. The suggestions how to improve the legal system are given as the conclusion of analysis. The Lithuanian administrative system of taxes has to improve constantly to be modern enough to satisfy the needs of changing reality. One of the most important challenges it faces is the motivation of taxpayers to pay taxes honestly. Only by achieving this destination it can ensure steady grow of taxing incomes of the national budget. That’s why the safeguard of the taxpayer’s rights’ is so important. Citizens are cooperative with the state when they trust it. There are couples of legal mechanisms which allow taxpayer to defend his rights. Taxing disputes’ legal proceedings is one of them.

The role of vinculin in cell adhesion was investigated by isolating NIH3T3 and Balb/c 3T3 cells containing a plasmid vector expressing a vinculin antisense RNA under the control of the MMTV promoter. Stable cell lines of NIH3T3 cells were cultured with dexamethasone but none showed any reduction in vinculin protein levels. Two Balb/c 3T3 cell lines, when cultured with dexamethasone displayed a marked reduction in vinculin levels and displayed an altered cell shape from a spread to a more spindle-shaped morphology. These phenotypic changes were reversible upon removal of the dexamethasone. The reduced adhesion corresponded with a reduction in the growth rate of the clones. The cells were also unable to spread on fibronectin and the ususal increase in the phosphotyrosine content of two signalling proteins, paxillin and pp125FAK, normally associated with cell adhesion, was not observed. The results establish that vinculin is a key component of integrin-mediated cell adhesion. To explore the structure-functional relationship of talin, six anti-talin monoclonal antibodies were microinjected into human fibroblasts and the effect upon the actin cytoskeleton was assessed. Two of the antibodies, TA205 and TD77 resulted in the disintegration of actin stress fibres, and migration of CEF was also severely impaired following microinjection of either antibody. The epitopes recognised by TA205 and TD77 had been mapped to talin residues 102-497 and 2269-2541 respectively. Microinjection of CEF with a polypeptide containing residues 102-497 demonstrated that they were mainly associated with the detergent-soluble cytoplasmic portion of the cell and were also able to disrupt stress fibre and focal adhesion integrity. Residues 2269-2541 were associated with the detergent-insoluble cytoskeletal fraction of the cell but did not affect stress fibre or focal adhesion integrity. Attempts to identify proteins that interacted with residues 102-497 of talin were unsuccessful but an actin-binding site was identified within residues 2269-2541. The results indicate the presence of domains within the N- and C-terminus of talin that are essential to the involvement of talin in the formation of focal adhesions.

This thesis attempts to quantify the impact of kombi-taxis on the conventional modes of public transport, in particular the bus, in the Cape Town Metropolitan Area. The impact is quantified in terms of the resultant modal shift of commuters from the buses, trains, cars and walking, in favour of the kombi-taxi. The approach adopted involved a study of the kombi-taxi and bus operations and characteristics on the different kombi-taxi routes in the study area. Five representative routes were selected for a detailed study, involving an Observation survey and an Interview survey directed at the bus and kombi-taxi users on these routes. On a further 66 routes, a bus-taxi modal split survey was conducted. The findings of the study show• that the majority of present kombi-taxi users are former bus users. Conservatively, an estimated 30.6% of all the daily bus passenger trips have been lost to the kombi-taxi. The effect on trains has not been insignificant with an estimated 4.4% of all commuter train trips having been converted to kombi-taxi trips.

At the nascent focal adhesion, GTPase Rap1A recruits the adaptor protein RIAM (Rap1 Interacting Adaptor Molecule), and RIAM recruits the scaffold protein talin facilitating integrin activation (Lee et al., 2009). As the nascent adhesion matures, actin linkages are strengthened via recruitment of the talin-actin bridging protein vinculin. In vivo data shows that RIAM is not found in mature focal adhesions; furthermore RIAM and vinculin occupy the same binding sites within talin (Goult et al., 2013). This thesis shows that RIAM-talin interaction is synergistic, with multiple domains simultaneously contributing to the binding. Vinculin and RIAM compete for talin, providing a mechanism of RIAM displacement by vinculin as adhesions mature. The interactions are thus characterised by a range biophysical techniques. SHARPIN (Shank associated RH-domain interacting Protein) potentially competes with talin for integrin binding (Rantala et al., 2011). SHARPIN interaction with the synaptic protein SHANK3 (SH3 and multiple ankyrin repeat domains protein 3) may provide a mechanism for SHANK3 recruitment to the adhesion (Lim et al., 2001). This thesis solves the structure of the SHANK N-terminal domain that contains the SHARPIN binding region. The structure shows the presence of a previously uncharacterised RA (Ras-association) domain that packs against the following ankyrin repeat domain. We show the RA domain binds with high affinity to the GTPase-Rap1A. Rap1A-SHANK could provide a negative feedback pathway for integrin regulation in adhesion. In the course of adhesion maturation multiple proteins are recruited to the adhesion complex; these proteins facilitate linkages to the actomyosin contractile machinery. One such protein is DLC1 (Deleted in Liver Cancer 1); DLC1 contains an LD-like motif that is homologues to the LD domains of the adhesion protein paxillin (Brown et al., 1996). This LD-like motif is responsible for tumour suppressor function and binding to talin (Li et al., 2011; Cao et al., 2012). In this thesis we characterise the talin-DLC1 interaction using X-ray crystallography, and suggest mutations to disrupt the interaction in cell experiments.
