Tauopathies: A Distinct Class of Neurodegenerative DiseasesNeurodegenerative diseases are characterized by neuronal loss and intraneuronal accumulation of fibrillary materials, of which, neurofibrillary tangles (NFT) are the most common. Neurofibrillary tangles also occur in normal aging and contain the hyperphosphorylated microtubule-associated protein tau. A detailed presentation is made of the molecular bases of Alzheimer's disease (AD), postencephalitic parkinsonism, amyotrophic lateral sclerosis/parkinsonism-dementia complex (ALS/PDC) of Guam, progressive supranuclear palsy (PSP), corticobasal degeneration (CBD), Pick's disease, frontotemporal dementia (FTD), Down's syndrome, myotonic dystrophy (DM) and Niemann-Pick Type C (NPC) disease, which are considered to be common tauopathies. The unique human tau gene extends over 100 kb of the long arm of chromosome 17 and contains 16 exons. The human brain contains six tau isoforms that contain from 352 to 441 amino acids. To date, 34 pathogenic tau mutations have been described among 101 families affected by FTD with parkinsonism linked to chromosome 17 (FTDP-17). These mutations may involve alternative splicing of exon 10 that lead to changes in the proportion of 4-repeat- and 3-repeat-tau isoforms, or may modify tau interactions with microtubules. Tau aggregates differ in degree of phosphorylation and in content of tau isoforms. Five classes of tauopathies have been defined depending on the type of tau aggregates. The key event in tauopathies is the disorganization of the cytoskeleton, which is based on mutations/polymorphisms in the tau gene and lead to nerve cell degeneration. In this review, tauopathies as a distinct class of neurodegenerative diseases are discussed with emphasis on their molecular pathology and genetics.

A mechanism that describes the progression of traumatic brain injury (TBI) to end-stage chronic traumatic encephalopathy (CTE) is offered in this article. This mechanism is based upon the observed increase in the concentration of both tau protein and of human leukocyte antigen (HLA) class I proteins; the HLA increase is expressed on the cell membrane of neural cells. These events follow the inflammatory responses caused by the repetitive TBI. Associated inflammatory changes include macrophage entry into the brain parenchyma from increased permeability of the blood-brain barrier (BBB) and microglial activation at the base of the sulci. The release of interferon gamma from the microglia and macrophages induces the marked increased expression of HLA class I proteins by the neural cells and subsequent redistribution of the tau proteins to the glial and neuronal surface. In those individuals with highly expressed HLA class I C, the high level of HLA binds tau protein electrostatically. The ionic region of HLA class I C (amino acid positions 50-90) binds to the oppositely charged ionic region of tau (amino acid positions 93-133). These interactions thereby shift the cellular localization of the tau and orient the tau spatially so that the cross-linking sites of tau (275-280 and 306-311) are aligned. This alignment facilitates the cross-linking of tau to form the intracellular and extracellular microfibrils of tau, the primary physiological characteristic of tauopathy. Following endocytosis of the membrane HLA/tau complex, these microfibrils accumulate and produce a tau-storage-like disease. Therefore, tauopathy is the secondary collateral process of brain injury, resulting from the substantial increase in tau and HLA expression on neural cells. This proposed mechanism suggests several potential targets for mitigating the clinical progression of TBI to CTE.

In neurons, the microtubule associated protein, tau, is found in the axons. Tau stabilizes the microtubules required for neurotransmitter transport to the axonal terminal. Since tau has been found in both Alzheimer neurofibrillary tangles (NFT) and in paired helical filaments (PHF), the study of tau's normal structure had to preceed TEM studies of NFT and PHF. The structure of tau was first studied by ultracentrifugation. This work suggested that it was a rod shaped molecule with an axial ratio of 20:1. More recently, paraciystals of phosphorylated and nonphosphoiylated tau have been reported. Phosphorylated tau was 90-95 nm in length and 3-6 nm in diameter where as nonphosphorylated tau was 69-75 nm in length. A shorter length of 30 nm was reported for undamaged tau indicating that it is an extremely flexible molecule. Tau was also studied in relation to microtubules, and its length was found to be 56.1±14.1 nm.

Tauopathies are a class of neurodegenerative diseases characterized by the presence of hyperphosphorylated and aggregated tau pathology in neuronal and glial cells. Though the ratio of neuronal and glial tau aggregates varies across diseases, glial tau aggregates can populate the same degenerating brain regions as neuronal tau aggregates. While much is known about the deleterious consequences of tau pathology in neurons, the relative contribution of glial tau pathology to these diseases is less clear. Recent studies using a number of model systems implicate glial tau pathology in contributing to tauopathy pathogenesis. This review aims to highlight the functional consequences of tau overexpression in glial cells and explore the potential contribution of glial tau pathology in the pathogenesis of neurodegenerative tauopathies.

Parkinson's disease (PD) is a chronic and degenerative condition that affects millions of Americans. Current approach of PD evaluation, diagnosis, and treatment is mainly qualitative using the Unified Parkinson's Disease Rating Scale (UPDRS) or the Hoehn and Yahr scale. Assessment of the efficacy of the drugs used is difficult and subjective. A long-term monitoring device that can collect movement data in assisting quantitative analysis proves to be useful and needed. This thesis discusses a discrete prototype of a passive EPC Class 1 Gen 2 UHF RFID sensor tag which is a preliminary step in realizing such a monitoring device. The prototype is capable of collecting 8-bit sensor (temperature, inertial, etc) data and transmits it in real-time through a RFID backscatter link to an UHF reader. It is shown that the device can achieve a read distance up to 3 meters at 5 reads/s and a max data rate of about 640 Kbps.

It has become common practice for cities to use neoliberal planning to improve the region’s economic growth and old industrial cities are using an additional cultural strategy to reinvigorate their city image. This is the case for Malmö which tries to renew itself from an old industrial city to a knowledge-based city by changing the built environment and using place branding. The change is made to attract innovation, companies and the desirable creative class. Malmö’s latest endeavor is to redevelop Rosengård, a part of the city notorious for its large immigrant population, its social inequality and its bad image.This study aims to compare culture worker’s perception of Rosengård to the image set by the urban planners in their planning documents. The culture workers are part of the so- called creative class and are favored by cities that believe this group will help to promote economic growth and contribute to the cities’ renewal.Malmö, like other cities, wants to change its image to be more alluring for desirable possible citizens, but if the city changes its image to something that is unrecognizable to its existing inhabitants Malmö risks not only to displace those inhabitants, but also to start an uprising.

For the past 50 years, cultural economics has developed as an independent research specialism. At its core are the creative industries and the peculiar economics associated with them, central to which is a tension that arises from the notion that creative goods need to be experienced before an assessment can be made about the utility they deliver to the consumer. In this they differ from the standard private good that forms the basis of demand theory in economic textbooks, in which utility is known ex ante. Furthermore, creative goods are typically complex in composition and subject to heterogeneous and shifting consumer preferences. In response to this, models of linear optimization, rational addiction and Bayesian learning have been applied to better understand consumer decision- making, belief formation and revision. While valuable, these approaches do not lend themselves to forming verifiable hypothesis for the critical reason that they by-pass an essential aspect of creative products: namely, that of novelty. In contrast, computer sciences, and more specifically recommender theory, embrace creative products as a study object. Being items of online transactions, users of creative products share opinions on a massive scale and in doing so generate a flow of data driven research. Not limited by the multiple assumptions made in economic theory, data analysts deal with this type of commodity in a less constrained way, incorporating the variety of item characteristics, as well as their co-use by agents. They apply statistical techniques supporting big data, such as clustering, latent class analysis or singular value decomposition. This thesis is drawn from both disciplines, comparing models, methods and data sets. Based upon movie consumption, the work contrasts bottom-up versus top-down approaches, individual versus collective data, distance measures versus the utility-based comparisons. Rooted in Bayesian latent class models, a synthesis is formed, supported by the random utility theory and recommender algorithm methods. The Bayesian approach makes explicit the experience good nature of creative goods by formulating the prior uncertainty of users towards both movie features and preferences. The latent class method, thus, infers the heterogeneous aspect of preferences, while its dynamic variant- the latent Markov model - gets around one of the main paradoxes in studying creative products: how to analyse taste dynamics when confronted with a good that is novel at each decision point. Generated by mainly movie-user-rating and movie-user-tag triplets, collected from the Movielens recommender system and made available as open data for research by the GroupLens research team, this study of preference patterns formation for creative goods is drawn from individual level data.

Purpose: This study examines 26 large and established South African multi-asset unit trusts in order to determine their style and asset class exposure over time. The objective is to ascertain whether South African multi-asset fund managers can realise outperformance, that exceeds what can be realised through exposure to representative, investable, style and asset class indices. Such an analysis assists in identifying unit trust manager skill, but a further consideration is how to combine unit trusts in a suitable manner, to this end portfolio construction tools are utilised to meet illustrative client objectives in a multi-asset context. Methodology: This study uses monthly total return time series for several investable style and asset class indices as well as South African multi-asset unit trust monthly total return time series. Where historical data permits, the period under investigation is from 1 January 2003 to 30 June 2018. Style and asset class exposure is determined using the Returns Based Style Analysis (RBSA) of Sharpe (1992) applying a 24-month rolling window approach. Findings: The equity style exposures estimated using RBSA provide evidence that on average the value style was dominant across the multi-asset high equity unit trusts examined. For the multi-asset low equity unit trusts examined the low volatility style was dominant. Moreover, a large proportion of the variability in returns of many multi asset unit trusts, can be explained by exposure to style and asset class indices. Consequently only 3 out of the 15 multi-asset high equity unit trusts analysed, could realise performance in excess of their custom style and asset class benchmark. As only a limited number of these unit trusts could demonstrate superior security selection ability the implication is that many asset managers stand to be disrupted by lower cost products that provide similar style and asset class index exposure. Originality/Value: Much research has been conducted into the style exposures of SA general equity funds. However, to the author’s own knowledge this is the first study to apply RBSA in a performance context to multi-asset unit trusts, under the new ASISA classification standards. The benefits of portfolio construction tools such as portfolio simulation and the ‘Risk Budgeting’ approach are also discussed and applied in a multi-asset context.

Resumo: A partir da combinação de fontes processuais, literatura e música popular, este trabalho pretende investigar como se constituíram alguns estereótipos, em especial àqueles relacionados à sensualidade, beleza e a sexualidade das mulatas. Discutindo os conceitos de gênero, classe e raça, minha proposta é pensar como estes estereótipos podem ser percebidos no cotidiano de mulheres negras (e ¿mestiças¿), em sua maioria pobres, de fins do século XIX e início do século XX, período imediatamente posterior à abolição do trabalho escravo no Brasil, bem como da também recente proclamação da República. Utilizando processos de mulheres com a alcunha Conceição combinada a outros nomes, discuto como imagens e estereótipos sobre as mulheres negras surgem no ambiente policial e jurídico. Abstract: From the combination of legal proceedings sources, literature and popular music, this work intends an investigation on how some estereotypes were established specially those related to mulata¿s sensuality, beauty and sexuality. Examining gender, class and race concepts, my proposal is to think how these estereotypes may be noted on black (and ¿mestiças¿) women¿s everyday, mostly poor, from the end of nineteenth century and the begining of twentieth century, period just after the slavery abolition in Brazil and short after the Republic proclamation. Using women¿s legal proceedings whose name Conceição was arranged among another names, I examine how images and estereotypes about black women rise on police and juridical environments. Next, I work the period¿s literature and music as privileged stages to allegory readings of these estereotypes

Flavonoids belong to a class of natural, polyphenolic dietary compounds which modify the neuropathological state of the brain. Some flavonoids like quercetin and other, reduce the inflammation, carcinogenicity, and oxidation promotes neuroprotection and comprises the major component of cosmetics, medicinal and dietary supplements. Daily intake of flavonoids helps to mitigate the risk of several neurological disorders like Alzheimer’s disease, Parkinson’s disease, multiple sclerosis, etc. Flavonoids exhibit their pharmacological effect through various mechanisms like cholinesterase inhibition, scavenging free radicals, memory enhancement via attenuation of amyloid plaques, tau targeting detoxification and neural antiinflammation. Administration of flavonoids to biological system has to pass through several biological checkpoints like first pass metabolism, intestinal absorption, and entry into blood brain barrier. Flavonoids exhibit difference in pharmacokinetic and pharmacodyanmic profile due to difference in their structures. Recent literature reports have proved promising therapeutic potential in neurological disorders. This chapter highlights the recent development of flavonoids prevailing in the field of neurodegenerative, its limitations and drug delivery approaches to encounter the challenges.

This paper describes the design of a simulation model of the infrastructure for data processing fromthe Super Charm-Tau factory class "megasience" electron-positron collider. The model simulates thebehavior of the main subsystems of a supercomputer, such as the task scheduler, computing clusters,data storage system, etc. Using simulation modeling allows for the maximally reliable representationof the exact characteristics and volume of the needed equipment for developing the desired HPCsystem. The simulation model accounts for all the aspects of operation of this system from paralleldata storage system to arrangement of the parallel launch of tasks. The developed system forprocessing software errors and equipment failures, as well as the system for ensuring energy efficiencymake it possible to estimate the needed equipment with account for all possible emergency situations.This model allows calculating the parameters of the computing system necessary for processing andstoring the results of operation of the Super Charm-Tau factory after its commissioning.

Search spaces hallmark the advancement of Neural Architecture Search (NAS). Large and complex search spaces with versatile building operators and structures provide more opportunities to brew promising architectures, yet pose severe challenges on efficient exploration and exploitation. Subsequently, several search space shrinkage methods optimize by selecting a single sub-region that contains some well-performing networks. Small performance and efficiency gains are observed with these methods but such techniques leave room for significantly improved search performance and are ineffective at retaining architectural diversity. We propose LISSNAS, an automated algorithm that shrinks a large space into a diverse, small search space with SOTA search performance. Our approach leverages locality, the relationship between structural and performance similarity, to efficiently extract many pockets of well-performing networks. We showcase our method on an array of search spaces spanning various sizes and datasets. We accentuate the effectiveness of our shrunk spaces when used in one-shot search by achieving the best Top-1 accuracy in two different search spaces. Our method achieves a SOTA Top-1 accuracy of 77.6% in ImageNet under mobile constraints, best-in-class Kendal-Tau, architectural diversity, and search space size.

What motivates property owners to pay taxes in places where state enforcement is weak? Using an online experiment among property owners in Lagos, Nigeria, we evaluate the extent to which different appeals increase respondents’ tax morale, their willingness to pay taxes if there is no enforcement, and attitudes about government enforcement of tax collection. Respondents were randomly assigned to read either a vignette emphasising the role of property tax revenue in contributing to economic growth and increased property values, or one highlighting that tax revenue is used for public goods and services benefiting all residents. The growth message made respondents significantly more favourable towards enforcement of tax collection, but there was no difference in willingness to pay between the two treatment conditions. We also look at heterogeneity across class identification and attitudes toward redistribution, and find that support for a more equal society reduces the advantage of the growth appeal.
