Pełny tekst źródłaSonic tomograph is a new instrument used in morpho-symptomatic analysis of trees. This tool use sonic waves propagation to establish wood density and probably internal decay. Sonic tomography was applicated on large scale in a urban park in Imola, to value all trees in depth way. Subsequently was used for different aims. First of all the instrument was applied to evaluate early stage of decay and development of decline in urban trees, by periodic tests in time. Afterwards was used to determine the travel of liquid substances injected in the trees by trunk injection methods. In the last analysis there was carried out a comparison between sonic tomograph and nuclear magnetic resonance to identify decay invisible to normal tools used in tree evaluation

Abstract: The aim of this study was to evaluate the effectiveness of diamond-coated sonic scaler insert (Sonicparo ¿), in the root surface debridement in single-rooted teeth on clinical (plaque index, bleeding on probing, probing depth, relative attachment levei clínical, and gingival recession) and microbiological parameters, compared to hand instruments. The investigation was a test-controlled randomized clínical trial with a splith-mouth design involving 12 patients with moderate to advanced chronic periodontites with pocket depth > 4,4 mm. The patients were submitted to initial treatment sessions of supragingival plaque, removal and were also instructed in oral hygiene techniques. After, flap surgery was carried out and the tooth surface instrumented. Clinical and microbiological parameters, were evaluated at baseline (O), in the 30, 90 and 180 days after surgery. Analysis of variance and Tukey test was done for all periods (p<0.05) and the results showed that when diamond-coated sonic scaler insert was compared to manual scalers, showed no statistical differences in probing depth reduction, plaque index, bleeding on probing, relative attachment level gain, gingival recession and reductions of the microbial counts and colony-forming units. There were no significant differences between regard with treatments for all parameters at each re-examination. Within the limits of this study, it can be concluded that the use the of diamond-coated sonic scaler insert can be a good alternative for root debridement

Abstract: The aim of this study was to compare, using epoxy resin replicas, the surface topography of root ends following ultrasonic preparation, and again after root-end fillings submitted to three different finishing techniques. A quantitatively evaluation of the sealing ability of Super-EBA, IRM and Pro Root MTA root-end fillings was also performed. Eighty-one ultrasonically root-end cavities prepared in human canines, were divided at random into three test groups of 27 each. The cavities were filled with Super-EBA, IRM, or ProRoot-MT A and finished by ball burnishing. Eighteen roots from each group received a final smoothing with either a 30-fluted tungsten carbide finishing bur, or a Zekrya carbide 28 mm bur. Scanning electron micrography (SEM) pictures of each sample were taken prior to and after root-end filling, and dentin/retrofilling interfaces inspected and compared with the aid of an image and analysis system. Samples were then prepared and immersed in 2% methylene blue dye solution for 12 hours. Roots were ground into a powder and prepared for analysis in an absorbency spectrophotometer. Results showed that when a finishing bur was used to finish the set materials, a significant (p < 0.05) area of marginal chipping was eliminated. The fishing technique did not significantly (p > 0.05) affect the incidence of gaps in groups retrofilled with MT A or IRM. Super EBA and IRM retrofillings finished with a ball burnisher or a Zekrya bur displayed a significantly (p < 0.05) larger calculated gap area than roots filled with MT A. In relation to microleakage the results revealed that Pro Root MT A displayed a significantly inferior mean dye microleakage (p < 0.05) than Super-EBA and IRM retrofillings. The finishing technique did not significantly (p > 0.05) affect the incidence of microleakage among the materials tested. The correlation coefficient of Pearson indicates that there is a positive, 55.61% association between the variables and this coefficient, that is, the increase of the quantity of infiltrated dye is associated with the increase of the root-end filling marginal disruption area

Abstract: Ultrasound in medicine has gone through great evolution in the last few decades and has occupied important position as a tool for therapy and diagnosis. This is due to the ultrasound equipment be of relatively low-cost, ultrasound is a non-ionizing radiation, is a non-invasive imaging method, and the images are created and seen in real time. It is common the use of transducer arrays in order to generate this kind of image. There is a lack of know how in Brazil relative to the construction of these transducers and the involved electronics in their operation. The objective of this work was the development of a multi-purpose 12 channel pulser/receiver electronic circuitry to operate with linear transducer arrays. The system is able to fire ultrasound piezoelectric transducers and to receive ultrasound echo signals in the range 0.5-30 MHz. The system has reception circuits with protection against high voltage pulses. The firing of transducer elements and cutting time of the reception circuits, to avoid unwanted signals of natural initial transducer oscillations, can be controlled via PIC 16F877 hardware and software designed to connect the system to a microcomputer. The electrical characteristics of the 12 channel pulser/receiver and its use in firing a specially constructed 1 MHz 12 element PZT transducer array has been carried out and the images of a specially constructed phantom showed that it can be used in laboratory conditions

Abstract: The aim of this study was to evaluate the effectiveness of diamond-coated ultrasonic scaler insert (CVDentUS®), in the root surface debridement of singlerooted teeth on clinical parameters (plaque index, bleeding on probing, probing depth, relative clinical attachment level and gingival recession), compared to hand instruments. The investigation was a randomized and controlled clinical trial with a splith-mouth design involving 17 patients with moderate to advanced chronic periodontitis, divides into 2 groups: : non-surgical group (5mm ? pocket depth > 3mm) and surgical group (pocket depth > 5mm). The patients underwent initial treatment sessions of supragingival plaque removal and were also instructed on oral hygiene techniques. Non-surgical group patients were treated using the diamond-coated ultrasonic scaler insert CVDentUS® in test site and Gracey 5/6 curettes in control site. In surgical group, flap surgery was carried out and the tooth surface instrumented with the same protocols. Clinical parameters were evaluated at baseline (0), 30, 60, and 90 days after treatments. The data were analysed using non-parametric tests, Mann Whitney, Wilcoxon and Friedman. In the end of the study, the results showed no significant differences between the treatments and between groups, except to relative clinical attachment level that showed a slight advantage for the treatment with curettes in non-surgical group. Within the limits of this study, it can be concluded that the diamond-coated ultrasonic scaler insert (CVDentUS®) was such efficient as them curettes for surgical and nonsurgical periodontal therapy in patients with chronic periodontal disease

The sulfated polysaccharides (SP) from the edible red seaweed Gracilaria birdiae were obtained using five different condition extraction (GB1: Water; GB1p: Water/proteolysis; GB1s: Water/sonication; GB1sp: Water/sonication/proteolysis; GB2s: NaOH/sonication; GB2sp: NaOH/sonication/proteolysis. The yield (g) increased in the following order GB2sp>GB1sp>GB1p>GB2s>GB1s>GB1. However, the amount of SP extracted increased in different way GB2sp>GB1p>GB1>GB1sp>GB1s>GB2s. Infrared and electrophoresis analysis showed that all conditions extracted the same SP. In addition, monosaccharide composition showed that ultrasound promotes the extraction of other polysaccharides than SP. In the prothrombin time (PT) test, which evaluates the extrinsic coagulation pathway, none of the samples showed anticoagulant activity. While in the activated partial thromboplastin time (aPTT) test, which evaluates the intrinsic coagulation pathway, all samples showed anticoagulant activity, except GB2s. The aPTT activity decreased in the order of GB1sp>GB2sp>GB1p>GB1>GB1s>GB2s. Total capacity antioxidant (TCA) of the SP was also affected by condition extraction, since GB2s and GB1 showed lower activity in comparison to the other conditions. In conclusion, the conditions of SP extraction influence their biological activities and chemical composition. The data showed NaOH/sonication/proteolysis was the best condition to extract anticoagulant and antioxidant SPs from Gracilaria birdiae.

Pełny tekst źródłaThe enzyme invertase is of great interest to industry because it catalyzes the sucrose hydrolysis reaction producing invert sugar. The use of raffinose and ultrasound irradiation can release large amounts of enzymes in their active form and thus it is a tool to improve biotechnological processes. Luffa cylindrica is an excellent support to immobilize microbial cells, showing advantages such as low cost and resistance to autoclaving. This study investigated the effect of raffinose and ultrasound pulses on invertase release from free S. cerevisiae and S. cerevisiae immobilized in Luffa cylindrica. The free cell culture was submitted to 2% raffinose pulse and irradiated for 2 minutes at dilution rates of 0.12 and 0.46 h-1. The immobilized cell culture was submitted to raffinose pulse and irradiated for 1, 2 and 4 minutes, at 0.10 h-1 dilution rate. In free cells, the raffinose pulse did not alter the invertase activity but in immobilized cells, the value of the activity increased from 5.38 to 7.27 U/mg. Ultrasound application in free cell culture at the 0.12 h-1 dilution rate gave the best results. The activity varied from 25.08 to 29.38 U/mg while the increase in immobilized cells was from 5.22 to 9.70 U/mg when sonicated for 2 minutes. These results showed that ultrasound application in continuous culture may have great potential for application in biotechnological techniques.

Abstract: Diagnostic ultrasound consists of application of low intensity acoustic waves to noninvasively investigate biological tissues. Higher ultrasound intensities may alter tissue characteristics, and this is of interest for therapeutic applications, when the occurrence of bioeffects is - to a certain extent - desirable. The use of diagnostic ultrasound in Cardiology is well established, although there is an unexplored potential for therapeutic applications. The currently available treatments of arrhythmias by electrical stimulation are limited by serious complications, including systemic infection, inappropriate shock delivery, proarrhythmia, failure to pace and to defibrillate, and even death. Ultrasound can be an interesting alternative for electrical stimulation, but there are only a few studies that investigate the possibility of applying ultrasound for treating arrhythmias. The objective of this study was to develop power ultrasonic transducers to be applied to the investigation of sets of acoustical parameters able to interfere with the cardiac activity without causing tissue damage, thus aiming at potential therapeutic applications of ultrasound in cardiology. The acoustical parameters investigated were resonance frequency, operation mode, pulse repetition frequency, and output pressure. The two most efficient out of the seven transducers built were calibrated and used in biological experiments. In preliminary experiments conducted on isolated hearts from Wistar rats, arrhythmia was esporadically observed at the onset of ultrasound application using the 65kHz transducer, whereas an increase in spontanous beating rate accompanied by a reduction in the force developed by the myocardium occurred during continuous, prolonged exposure using the 1MHz transducer. In in vivo studies, ten Sprague-Dawley rats were anesthetized with isoflurane and exposed to a sequence of therapeutic ultrasound, and other five rats were used as a control group. The ultrasonic stimulation consisted of 1-MHz bursts of 1% duty cycle, 3 MPa peak negative pressure (ISPTA=3W/cm2), and decreasingly variable pulse repetition frequencies. Transthoracic ultrasound application was able to promote a negative chronotropic effect, decreasing the heart rate by 19% just after stimulation ceased. The effect lasted at least 15 minutes, without apparent alteration of pumping function, which was monitored mainly by evaluation of the ejection fraction. This phenomenon is promising for treating tachyarrhythmias. The insonification scheme used in this study excluded thermal effects, so the observed effect seems to have resulted from nonthermal mechanisms, possibly from radiation force. The variation in the pulse repetition frequency seems to be the key element for induction of the described effect, because the latter was not observed for constant repetition rates

Chitosans, generally obtained from chitin through partial deacetylation, are a group of polymers consisting of glucosamine and N-acetyl glucosamine. Chitosans are biodegradable, biocompatible, non toxic, and antimicrobial. The main objectives of this study are to prepare phosphorylated chitosan microparticles by reaction of chitosan with triphosphate, further to reduce the particle size by ultrasonication, and to evaluate the antimicrobial activity of chitosan microparticles and chitosan/gelatin composite films. Microparticle of phosphorylated chitosan was formed via ionic gelation method by mixing sodium triphosphate with chitosan. After treated by sonication, the particle size was further reduced. The absorbance at 888 cm-1 (representing the group of -PO) in FTIR spectrum indicated that the crosslinking was formed between chitosan and sodium triphosphate. In addition to the particle size distribution, phosphorylated chitosan exerted significantly better antimicrobial activity when compared to the non-treated chitosan. Gelatin and different type of chitosans (un-treated, phosphorylated, and phosphorylated/sonicated) were used to fabricate chitosan/gelatin composite films. Microstructure, mechanical properties, moisture content, water solubility, water vapor transmission ratio (WVTR), and antimicrobial activity of the resulting films were analyzed. Microstructure of film was observed by scanning electron microscope; the results showed that the films containing chitosan exerted more compact structure and higher tensile strength. The highest tensile strength was detected in the composite films with the ratio of gelatin/chitosan at 2:3. No significant difference of water content was observed among different types of composite films. While water solubility decreased as the ratio of chitosan increased in the formulation, WVTR increased with the increase of chitosan proportion. Ranges of WVTR of composite films containing untreated, phosphorylated and phosphorylated/sonicated chitosan were 4.54-6.20, 4.54-5.78, and 4.54-5.24 (g/m2‧24 hr), respectively. Gelatin/untreated chitosan composite films showed greater antimicrobial capacity, after contact 180 minutes its inhibition measured up to 95%. However, the inhibition for phosphorylated and phosphorylated/ultrasonicated composite films at the same condition was 80-90%. In conclusions, phosphorylated chitosan prepared by ionic gelation method had smaller particle size, better mechanical properties, and greater antimicrobial activity. The best ratio of gelatin/phosphorylated chitosan for preparation composite films was 2:3.

This study investigated the enhancemeat of Vibrio antigen presentation releasing to promote protective efficacies of oral vaccines in cultured grouper. Vibrio cells were treated with three ways, formalin inactivated, ultrasound sonicated and frozen. Higher ELISA absorbance(ABS) represented higher amount of antigen presentation. Formalin could inactivate pathogen by denaturing its proteins, but result in the decrease of antigen releasing amount. Frozen treatment did not affect the amount of antigen presentation, and the ultrasound sonication shatter treatment took too long for completely killing all Vibrios. Therefore, frozen and ultrasound sonication were combined to inactivated the bacteria. After ultrasound sonication, Vibrios were frozen for completely killing. Antigens releasing amount in each treatment was obtained in this study. In sodium alginate encapsulated group, raising the concentration of sodium alginate and calcium chloride to 2.5% could release the highest antigen amount. In cellulose acetate phthalate (CAP) encapsulated group, the use of 5% CAP could result in the highest amount of antigen releasing. Groupers were i.p. challenged with 5 times LD50 of Vibrio. parahaemolyticus and V. vulnificus respectively after two times of oral vaccination with sodium alginate encapsulated bacterins and cellulose acetate phthalate encapsulated bacterins. In 10 times LD50 challenge test, formalin’s group reached highest RPS at 8 wpv and sonicated group reached highest RPS at 12th wpv. In alg-sonicated group RPS could reach 100% while CAP-formalin’s group only could reach 50%. . In serum antibody titer analysis, each treatment and control groups had significant differences (p <0.05). Values of serum antibody titer increased significantly at 4th wpv, then reached highest at 12th wpv and then decreased and remained stable. Values of sonicated group were higher than that of formalin’s group from start to 20th wpv. In Superoxide dismutase(SOD) analysis test, the SOD value reached the highest at 4th wpv then decreased. Values of alginate sonicated group was higher thanother group. After vaccination, alginate-sonicated group exhibited the best performance in RPS, serum antibody titers and SOD value , followed by CAP - sonicated group, alginate – formalin’s group, the worst is CAP – formalin’s group. The protective efficacies in duration of CAP – ultrasound sonicated group was better than that of other groups
