Spatial data management within the environmental field has a large range of application possibilities and comes with great advantages. In this study methods and technologies for spatial data management of soil contamination has been assessed in Geographical Information Systems (GIS), in order to identify in which way spatial data applications and tools can contribute with valuable information for these type of projects. The spatial assessment has been applied on a case study site in Kagghamra, Stockholm, exposed to high levels of contaminants, arsenic in particular. Subjects that have been evaluated are arsenic contamination distribution pattern, estimation of volume contaminated soil and amount of samples needed for spatial analyses. Furthermore, two versions of an exploratory soil sensitivity estimation model based on site specific ground and landscape parameters as well as literature references have been developed. The data management included large quantities of primary and secondary data of the commination levels as well as geological and ground properties. First hand collected geophysical field data obtained from Electromagnetic (EM) and Induced Polarisation (IP) measurements was also interpreted. The benefits of using geophysical measurements in soil contamination projects has been investigated. In this case the benefits were few due to difficult measuring conditions with disturbance noise. Spatial interpolations with the Natural Neighbour  (NN) technique are proven to be useful in transforming point contamination data into continuous layers. From the interpolation surfaces (arsenic distribution map) a variety of information can be extracted, such as a first hand volume estimation of contaminated soil, possibilities of reduction in amount of field sampling or to investigate statistical information and relations to different site specific ground conditions. The soil sensitivity estimation models are combined maps consisting of data layers that are relevant for the arsenic behaviour and interaction in the subsurface. Site specific Model (1) is based the data layers Soil type, Iron level, Soil depth, Slope  and illustrates mainly areas exposed to high concentrations of arsenic as high sensitivity areas. The more general, literature supported Model (2) also includes Vegetation cover and Topographic Wetness Index (TWI) and is not related highly to the arsenic distribution in the site area, but could contribute with general implications of sensitive areas if applied on a another, larger site area. Efficient management of large data quantities, economic and time saving benefits from less physical sampling and good representation and visualisation possibilities of the site conditions, as a tool for stakeholder communication and decision-making are the main contributions from the spatial data management.

The extent of soil contamination with organic and inorganic pollutants in most urban areas in the UK is largely unknown but due to past and present industrial activity it is likely that pollutant levels are high. Such contamination could have a serious impact on human health. Therefore this thesis set out to examine the extent of soil contamination within Newcastle upon Tyne with a focus on the contribution of an incinerator (Byker) to contamination levels of urban soils. The Byker incinerator is situated in central Newcastle and has been the subject of much media controversy due to the disposal of incinerator ash on local allotments. The current work extended past investigations to see if the incinerator had contributed to general urban soil pollution (heavy metals and dioxins) by aerial deposition and allowed a useful investigation into levels of urban soil contamination in Newcastle. In addition to examining metal and dioxin levels the bioaccessibility (human and bacterial) of pollutants in selected soil samples was estimated and an attempt to develop a human cell based soil toxicity assay made. These measurements permit a preliminary assessment of risk to human health from soil contamination. A total of 163 soil samples were collected based on predicted aerial deposition from the Byker incinerator and analysed for dioxins and heavy metals (As, Cd, Cr, Cu, Hg, Ni, Pb & Zn) content (mg pollutant /kg soil). A high proportion of samples (90/163) had contamination levels above soil guideline values (as proscribed in current UK regulations; CLEA). The highest dioxin levels were South West of the incinerator (1911 ng I-TEQ/kg) and the congener profiles coupled with a detailed historical survey demonstrated that the source of contamination was likely to be an old alkali works and not the incinerator. Overall it was found that the incinerator did not contribute significantly to dioxins found in the urban soils except those in the incinerator plant grounds. Soil metal contamination levels varied but were not related to incinerator deposition. Many samples contained levels of heavy metals well over soil guideline values with the highest values being found for Cu (12,108 mg/kg), Pb (4,134 mg/kg) and Zn (4,625 mg/kg) To determine the potential human health risk associated with heavy metal contaminated soils, selected samples (16) were subjected to two in vitro digestion techniques which simulate the bioaccessibility of metals (Cu, Ni, Pb & Zn) to humans in case of soil ingestion. Only three samples had high levels of metal availability (over SGV's) indicating that these soils should be subjected to further risk assessment. All other samples tested had low metal availability most likely due to a combination of metal speciation, and complexation to soil particles over time. An attempt was made to develop a human cell based system to determine the toxicity of contaminated soil. Using an in vitro system, human liver cells (HepG2's) were exposed to extracts from soils; cytotoxic effects (membrane integrity, metabolic capability and oxidative stress status) and genotoxicity potential (DNA damage) of Cu and Zn were first investigated in order to standardise the biological assays used. Between 0.1 and 10 mg/L Cu caused DNA damage and higher concentrations caused cytotoxicity. Zn was also proven to cause genotoxic effects from O. lmg/L. From 10 mg/L cytotoxic responses occurred and DNA damage could be attributed to cell death. Due to difficulties in sterilising soil extracts and physical damage caused to HepG2 cells by the abrasive nature of soil, it was not possible to elucidate whether metal contaminated soil extracts were capable of causing a cytotoxic or genotoxic response in human liver cells. Finally, the same soil samples were then subjected to a bacterial (lux) biosensor technique to examine soil toxicity. Interestingly, despite the high levels of contamination found, none of the soil samples were found to be toxic to the two Pseudomonas strains used which again indicates a low level of ecosystem risk and suggests that most of the contaminants present are either in a form that is unavailable to living microbes or are complexed to soil particles. In summary, this research has shown that the high level of soil contamination of urban areas in Newcastle is due to past industrial activity and a similar situation is likely in most other urban areas of the UK and internationally. The high cost of remediation means that if contaminated sites are shown to be a potential risk then bioaccessibility of contaminants should be examined in order to provide a more realistic assessment of the need for remediation. This work demonstrates that only a small proportion of urban contaminated sites are likely to require remediation based on bioaccessibility determination measurements.

This study constitutes a preliminary assessment of the behaviour of effluent-associated vanadium, and its possible effects on the biotic components of land treatment sites, used for the disposal of liquid industrial wastes from a chemical plant in the eastern Transvaal. A review of the literature showed that although the emission of vanadium into the environment is on the increase, very little information is available regarding its behaviour and impact as an environmental pollutant. This study is therefore important not only in the context of the land treatment operation in question, but clearly in a more universal context as well. The study involved a three-phase approach to the problem. Firstly, vanadium sorption was considered in four soils encountered on the sites in question. Secondly, an investigation was conducted into the potential toxicity of vanadium to relevant plant species. Finally, the potential inhibition of soil biological activity at increasing levels of vanadium was examined, although the results of this experiment were inconclusive.

There is a concern that concentration-based targets for soil clean-up are arbitrary, and do not necessarily relate to the environmental threat posed by the residual contamination. The development of sensitive, reliable, and ecologically relevant biological tests for oil-polluted soils would address these problems, and form a valuable complement to chemical analysis. Three biological test systems were chosen for examining the impact of oil contamination on soil microbes: (1) lux-marked bacterial biosensors, (2) Biological carbon source utilization profiles, and (3) phospholipid fatty acid (PLFA) community profiles. This allowed the testing of effects at three different levels of ecological complexity: respectively, single species; culturable bacteria; and eubacteria and eukaryotes. The use of specific bioluminescent biosensors (i.e. with lux genes fused to promoters from hydrocarbon degradation pathways) allowed the rapid detection of different hydrocarbon classes. The bacterial biosensors were optimized for the assessment of hydrocarbon compounds. Development of quantitative structure-activity relationships (QSARs) increased understanding of their responses to hydrocarbons and other petroleum-derived compounds. The three biological methods were then used to monitor the nutrient-assisted bioremediation of oil-spiked soils in two separate microcosm experiments: (1) remediation of four crude oils and one refined oil compared using bacterial biosensors, and (2) effects of diesel on three different soil types, and effects of three refined oils on one soil type, assessed using Biolog and PLFA profiling. All three biological test methods were sensitive to the oil contamination levels applied in the microcosm experiments. Individual species (biosensor tests) gave different responses to different oils; however, the community-level responses showed no differences. The microbial communities of the three different soil types could still be distinguished after 14-15 weeks of hydrocarbon contamination. The community response of the contaminated soils had changed from that of the control soils after four weeks of bioremediation, and did not return in similarity to the control over the course of the experiment.

Surface water is used as a source of water supply in many countries, including South Africa. One of the sources of surface water pollution is leachate and surface runoff from landfills. In agricultural soils, the landfill runoff and leachate deteriorate the quality and affect the fertility of soil. The entry of metals and microorganisms from landfill leachate to adjacent environments is through surface runoff due to rainfall. Adverse effects on human- and environmental health triggers a need to monitor and control contaminants in the environment. The aims of the study are to determine the effect of landfill runoff and leachate on agricultural soil and river water (Veldwachters River) running adjacent to the Devon Valley landfill site and to identify potential metal-tolerant organisms in environmental samples collected in Stellenbosch, Western Cape, South Africa. Samples (agricultural soil, river water and sediments) were collected once a month for a period of six months from the study area for analysis. Physicochemical parameters that are known to have major effects on environmental samples were assessed and the concentrations of various metals (Al, Pb, Cr, Mn, Mo, Co, Ni, Cu, Zn, Fe, Cd and V) were also determined by means of inductively coupled plasma mass spectrometry (ICP-MS). Soil texture analysis was tested in order to monitor the metal distribution in soils under the influence of environmental factors.

A number of factors have contributed to the interest in reclaimed wastewater irrigation of vegetables. However safety of water as related to fresh cut vegetables has been a paramount concern of responsible agencies, growers and consumers. The objective of this study was to evaluate the risk mitigation potential of subsurface drip irrigation during reclaimed wastewater irrigation. Virus detection methodologies on produce were first optimized. Beef extract (3%) +0.04M sodium pyrophosphate was found to be the most efficient eluent to recover viruses from soil and plant material. The recovery efficiency of poliovirus type l and adenovirus type 40 from Pima clay loam soil ranged from 12--16%, and recoveries from Brazito sandy loam ranged from 58--81%. The recovery efficiencies of poliovirus type 1, adenovirus type 40, MS2 and PRD1 from lettuce ranged from 45 to 70%. The recovery efficiencies of MS2 and PRD1 from tomato fruit were in the range of 90%. Poliovirus type 1, adenovirus type 40, MS2 and PRD1 survived longer in Pima clay loam than in Brazito sandy loam. All enteric viruses remained stable at 4°C suggesting that little virus inactivation occurs during transportation and marketing. Poliovirus type 1 and adenovirus type 40 lost 1 log₁₀ in 11 and 17 days respectively on lettuce at room temperature. Hepatitis A virus lost 2.5 log₁₀ in 50 days on a lettuce head in a greenhouse during the winter season. Tomato, lettuce and cucumber crops were irrigated with virus-seeded water by subsurface and surface drippers. Subsurface drip irrigation resulted in 99% less viral contamination of vegetable leaves as compared to surface drip irrigation. The greatest risk of infection occurs from the outer leaves of lettuce. The risk of infection from consumption of reclaimed wastewater irrigated tomatoes and cucumber was 32% and 72% less than lettuce. The risk of infection from rotavirus by ingestion of vegetables is greater than poliovirus type 1. The risk of infection from subsurface drip irrigated vegetables did not approach the United States Environmental Protection Agency's acceptable annual risk (1:10,000) until the concentration of viruses in the irrigation water reached 100/l. No internal contamination by viruses of the vegetables was observed during their growth. The coliphages PRD1 survived longer than poliovirus type I, adenovirus type 40, and MS2. It would thus appear to be a good model for studying the persistence of those viruses on produce and in irrigated agricultural systems.

This thesis is concerned with the following studies : 1- Ammonium analysis in soil and water including the determination of low levels of ammonium (<0.1 mg N/1) in groundwater and the colorimetric analysis of highly coloured groundwater samples. 2- An investigation of groundwater quality and soils at a contaminated industrial site. 3- Bioremediation treatment of the ammonium and nitrate contaminated groundwater using natural soil and vegetation and using soil incubation and pot experiments. These findings of a pot experiment suggest the possibility of applying the bioremediation treatment of the ammonium and nitrate contaminated water in the field. A field study should be undertaken to evaluate the efficiency of this bioremediation treatment. This field study would require a suitable uniform area to lay out the plots, preferably close to the source of water to be used. In addition, it is necessary to carry out a hydrological survey to determine the following aspects: 1-The size of groundwater reservoir. 2-The rate of removal of the water. 3-The time scale of the water application. The climatic conditions such as rainfall, potential evapotranspiration and temperature should be taken into consideration when carrying out the bioremediation treatment in the field as these climatic conditions affect the water requirements and the growth of grass. There are three options to apply the contaminated groundwater as follows : 1-To apply the contaminated groundwater at low or high volume depending on its level of nitrogen. 2-To blend well water with high level of nitrogen with well water with low level of nitrogen to achieve a realistic irrigation rate at a suitable nitrogen level. 3-To overirrigate in expectation that ammonium would be retained in the soil. The ryegrass used in this bioremediation treatment can be disposed of by incineration and landfilling the ash or landfilling the grass.

The application of animal manures to agricultural land as a soil organic amendment has been identified as an important route by which foodborne pathogens can enter the human food chain. Knowledge of the presence and incidence of key foodborne pathogens in manure is a vital first step in the establishment of sound and effective guidelines for management and prevention of contamination by manure. The work described in this thesis attempts to determine the persistence of pathogens when raw manures are directly applied to agricultural soils, and treated under thermophilic composting conditions. Results from this study indicated that the current conditions suggested by typical food safety guidelines are sufficient to reduce the population of enteric bacteria to levels that minimise risks associated with culturable cells in raw manure and finished compost. However, E. coli cells have the potential to enter a viable but non-culturable (VBNC) state and are undetected by culture-based monitoring methods, thus providing a false impression of the innate risk of the product. Prc, bamB and tolA, which are responsible for stabilising the cell membrane, were found to be essential genes required for surviving heat treatment at 55°C. In addition, the presence of relA and oxyR suggested that E. coli may use the VBNC state as an adaptive strategy for long-term survival to withstand multiple stresses, including heat stress. Entering the VBNC state with a strengthen cell envelope may help E. coli to survive prolonged heating during standard composting conditions. Successful resuscitation from the VBNC state was achieved in the presence of cell-free supernatant from actively growing E. coli MG1655. These results underline the importance of considering VBNC cells when evaluating the sanitary effect of the composting process. An alternative on-site wastewater treatment and disposal system (OSWTDS) consisting of a soil infiltrator with low pressure distribution was evaluated in a soil that was unsuitable for a conventional OSWTDS under current Commonwealth of Virginia Sewage Handling and Disposal Regulations, due to a shallow seasonally perched water table and low hydraulic conductivity. The absorption field consisted of two subsystems numbered as 1 and 2 with effluent design loading rates of 5.1 and 10.2 Lpd/m2, respectively (actual loading rates of 2.4 and 4.9 Lpd/m2, respectively). Soil matric potentials compared seasonally for each subsystem and indicated that both provided similar hydraulic performance. Background water quality was generally improved by subsurface movement through the absorption fields. A bacterial tracer was found in shallow (45.7 cm) and deep (213.4 cm) sampling wells within 24 h in the two subsystems (but in low numbers) over both summer and winter sampling periods. A viral tracer was detected within 48 h in both shallow and deep wells, but only in subsystem 2 in the winter. In evaluating denitrification potential, the addition of glucose to soil core samples did increase quantitatively, although not significantly, nitrous oxide production in each subsystem, at each depth, during each season. Overall, the performance of both subsystems was very similar. The soil infiltrator functioned very well, as designed for the site and soil limitations. It appears to be a potential alternative OSWTDS for use in problem soils.

Análises químicas de amostras de solo coletadas em quatro locais na área estudada confirmaram a contaminação severa por metais pesados em alguns dos locais, sendo que comparações entre o número de táxons obtidos no presente estudo com os dados de literatura permitiram concluir que houve prejuízo à diversidade de fungos na região. Abstract: In a heavy metal contaminated soil at ceramic pole of Santa Gertrudes region, São Paulo State, Brazil, a taxonomic survey of fungi of the order Mucorales (Zygomycota) have been done using the soil plate method, modified for a soil suspension (10%, p v-1) application on malt agar (2%, p v-1) added with Pb(NO3)2 or Zn(NO3)2 at 0 (control), 100, 200, 500 and 1000 mg L-1 concentrations. Chemical analyses of soil samples taken from four sites in the studied area confirm a severe contamination of heavy metals in some of the sites, and comparisons between taxa numbers obtained in the present study with literature data allowed conclude that there was fungal diversity disturbance in the region. The dry and grounded biomasses (2 g L-1) of Mucor hiemalis f. hiemalis (SPC 2040) and Mucor hiemalis f. luteus (SPC 2044) was kept in contact, under agitation at 120 rpm during 14 h, with aqueous solutions (pH 4) of Pb(NO3)2 and Zn(NO3)2 at 0.06, 0.13, 0.17 mM and 0.08, 0.16, 0.23 mM concentrations, respectively, being compared with activated charcoal (AC) as the reference adsorbent. Interest and participation in urban agriculture is growing in many cities throughout the United States. Urban gardeners and farmers produce food on various types of urban lands. Common soil contaminants of urban areas limit the amount of land on which food may safely be grown. The objective of this study was to assess and meet the informational and technical assistance needs of urban gardeners and farmers as well as students enrolled in the introductory soils science course at Kansas State University on the topics of urban soil quality and contamination. A needs assessment survey of urban gardeners and farmers was conducted in four communities; Tacoma and Seattle, Washington, Kansas City, Kansas and Missouri, Manhattan, Kansas, and Gary, Indiana. The survey generated information about what urban gardeners and farmers know, think they know, and want to know about urban soil quality and contamination. Eighty-eight percent of respondents indicated that they do not have knowledge of the best management practices to minimize health risks involved when growing food crops on soils contaminated with lead, cadmium, arsenic or organic contaminants. Our results suggest that urban gardeners and farmers require and want information and guidance on soil testing for common contaminants, interpretation of testing results, and best management practices for growing food on mildly contaminated soils. The students enrolled in the introductory soil science course at Kansas State University are future agricultural and environmental professionals who need skills to address urban soils issues. Most of the students in the Agronomy 305: Soils course are not Agronomy majors. Furthermore, an increasing number of Agronomy 305 students come from urban and suburban communities and/or have interest in working in urban environments upon completion of their undergraduate degree. An urban soils laboratory was developed in response to the future workforce demands as well as the demographics of students enrolled in the Agronomy 305 course. Throughout the semester students evaluated the physical, chemical, and biological properties of a soil from this urban community garden. Reaction of students to the new urban soils lab offering has been positive with 72% of students enrolled in the course reporting that they have interest and need in learning about the urban soil issues covered in the lab course. Overall, student responses about their learning experience in the urban soils laboratory course were positive, indicating that incorporating urban soil principles enhanced their soil science education. Students who participated in the urban soils lab are better prepared, as future agricultural and environmental professionals, to address the educational and technical assistance needs of urban growers.

This work examined the impact of soil organic matter (SOM) on the sorption, phase distribution and transport of ethanol and butanol blended gasoline vapours after release. Microcosm and mini-lysimeter experiments were conducted using sand with varying SOM and moisture contents. Synthetic gasoline alone and blended with 10 - 20% ethanol and 10 - 20% butanol by volume, referred to as UG, E10 - E20 and B10 - B20, respectively, were used. Results from the UG were used as the benchmark to assess the impact of ethanol and butanol on gasoline compounds. The findings of this work illustrate the likely behaviour of gasoline compounds at the beginning times of a gasoline spill or leak. The addition of alcohol to gasoline altered the behaviour of the gasoline compounds in the vadose zone in several ways. Firstly, it reduced the sorption of the gasoline compounds by soils. This effect was greatest on the first day of a spill and affected the gasoline compounds in decreasing order of hydrophobicity. Secondly, it altered the mass distribution of the gasoline compounds between the vadose zone phases to higher mass compounds in the mobile phases (soil air and soil water) and lower mass compounds in the immobile soil solid phase, suggesting higher risk of groundwater contamination with an increasing content of alcohol in the gasoline. Thirdly, it increased the vapour phase transport of the gasoline compounds from the source zone to the groundwater zone. These three impacts were generally greater for ethanol than butanol. The sorption coefficients (Kd) of E20 gasoline compounds were reduced by 54% for alkanes, 54% for cycloalkanes and 63% for the aromatics, while the Kd of B20 gasoline compounds decreased by 39% for alkanes, 38% for cycloalkanes and 49% for aromatics. This implies that the use of ethanol as gasoline oxygenate could result in greater risk of groundwater contamination with gasoline compounds than the use of butanol after spills. The SOM enhanced the sorption of alcohol-blended gasoline compounds in soils. This impact was similar for ethanol and butanol blended gasoline as the Kd of B20 and E20 were equally increased by 7 times for aromatics, 4 times for cycloalkanes and 2 times for alkanes, for 0 to 5% increase in the SOM fraction of sand. Although SOM enhanced the sorption of alcohol-blended gasoline, its sorptive capability was not fully realised compared with the sorption of the UG compounds. Also, it did not alter the order of groundwater contamination risk for the ethanol and butanol blended gasoline. Thus, the Kd values for all gasoline compounds for all the SOM fractions tested, including 0%fom, 1%fom, 3%fom and 5%fom, were in the order of UG>B20>E20, indicating greater risk of groundwater contamination for the ethanol-blended gasoline after a spill or leak regardless of the SOM content of the soil. The increase in the water content of soil reduced the sorptive capability of SOM and affected the overall mass distribution of gasoline compounds between the soil solid, soil air and soil water phases estimated with values of Henry’s law constant from the literature. This indicates that the degree of gasoline retention in the vadose zone by SOM could differ during the dry summer and wet winter seasons. This effect was greater for ethanol than butanol. Thus, in all seasons, the amount of gasoline compounds retained by SOM in the vadose zone is likely to be higher for butanol-blended gasoline than ethanol-blended gasoline. Overall, this study indicates that the use of high ethanol volume in gasoline to combat climate change may put the groundwater at greater risk of contamination after spills or leakages from storage. Therefore, to successfully reduce greenhouse gases emissions via high alcohol volume in gasoline and still protect the world’s groundwater resource, this study suggests the use of butanol is more benign than ethanol.

Crude oil is of economic importance to any nation. However its poor management and disposal method has resulted in oil spillage and ground contamination. Most emphasis on crude oil contamination on the environment has been on surface and ground water pollution, as well as plants and animals with little or no attention paid to the engineering properties of the contaminated soil. This research is thus aimed at investigating the effect of light crude oil contamination on the geotechnical properties of kaolinite clay soil. This research is a laboratory based experiment in which the contaminated soil was prepared by adding different percentages of light crude oil (2%, 5%, 8%, 10%, 15% and 20%) measured by weight of the dry soil sample and mixed until a uniform mixture was obtained. The British Standard Test Methods for Civil Engineering purposes BS 1377: 1990 (EUROCODE 7: EN 1997: 2-5) was adopted with few modifications. The calculations were based on the fact that the pore space in the contaminated soil is occupied by water and crude oil so water content was calculated from the fluid content and the rate of evaporation of crude oil during oven drying was also considered. The classification results showed that crude oil contamination caused an increase in linear shrinkage, liquid limit, plastic limit and plasticity index between 0% to 20% contaminations. The compaction result showed that there was an increase in maximum dry density while the optimum moisture content decreased between 0% and 15% of crude oil contamination. The result showed that the soil could not compact at 20% contamination and above. Also, the coefficient of permeability increased with increase in the percentage of crude oil contamination while the coefficient of consolidation value (Cv) increases with increase in the percentage of contamination. There was a decrease in the cohesion value and the frictional angle due to the introduction of the crude oil into the soil. Although crude oil altered the geotechnical properties of the kaolinite clay soil and reduced its strength, the soil can still be used for geotechnical purposed after remediation.

Understanding the impacts of explosive contamination on vegetation is key to understanding explosives behavior in the environment. I quantified shrub growth responses to 1,3,5-trinitroperhydro-1,35-triazine (RDX) soil contamination across three life stages and I hypothesized that RDX would have the greatest impact on seed germination. Morella cerifera seeds were germinated on soils amended with RDX up to 1500 mg RDX kg-1 dry soil. Juvenile and adult individuals were exposed for 6 weeks to soil amended with RDX up to 750 and 1500 mg RDX kg-1 dry soil, respectively. Morphological responses were quantified for juveniles while physiological measurements were quantified for adults. RDX induced a significant response in all age groups and, in accordance with the hypothesis, germination was the most impacted of the three stages. Impacts varied by concentration in addition to life stage, showing that many variables influence plant response to RDX.

The research topic has as its main objective the modeling of the flow of contaminant (mineral oil) following a road accident involving a heavy transport vehicle carrying 24 tons of CHEMOL pesticide, produced by the company Chemia S.p.A. of Sant’Agostino (FE). The target area is the ex-cave Olmi, currently used as a flood retention basin in the town of Calderara di Reno (BO), N-E of Bologna's Marconi airport, which is the final destination of the vehicle. Within this context, the thesis work concerned first of all the georeferencing and the study of an environmental report performed on the target area useful in the study of the hydrogeology of the soil and its stratigraphy. Subsequently, it was necessary to use the HSSM software to model the flow of contaminant in the soil. After this initial study, the Fault Tree (FTA) was built, the final event of which is the achievement of the target area, within a given time limit, a concentration higher than the limit given by the legislation. The failure rate of each basic event, which leads to the realization of the final event, was evaluated using values ​​derived from technical reports, with the exception of the event consisting in exceeding the concentration limit value in the target area. For the latter event, a Monte Carlo simulation was used, combining the values ​​of hydraulic conductivity and initial concentration of contaminant, which were the values ​​most affected by uncertainty. Finally, these values ​​were entered into the aforementioned HSSM software to evaluate the results. The probability of failure of this basic event was obtained by having the software compile a number of times equal to that of the possible combinations. The final calculation of the result, deriving from the constructed tree, was carried out using Boolean algebra, a method typically used in this probabilistic risk calculation technique. Contaminated tomatoes have become a commonly implicated vehicle of Salmonella outbreaks. Exposure of tomatoes to pathogen could occur in the field. Blossom inoculation with Salmonella can result in contaminated fruit but natural routes of blossom contamination are not well known. Salmonellae have been known to survive in agricultural soil. Since dispersal of soil particulate by wind is a common phenomenon, the potential of airborne soil particulate as a vehicle of Salmonella contamination in tomato blossoms was examined. It was determined that Salmonella enterica serotype Anatum, Baildon, Braenderup, Montevideo, Newport, Javiana had similar survival patterns in both soil and water. At the end of 40 days, populations of Salmonella in soil dropped by 2.59 log CFU/g and 5.11 log CFU/g when enumerated on Tryptic Soy Agar Yeast Extract (TSAYE) and xylose lysine Tergitol 4 (XLT4) agar respectively. Salmonella populations in water reduced by 2.55 log CFU/ml (TSAYE, enumeration) and 2.94 log CFU/ml (XLT4, enumeration). Blossom to fruit formation takes 20-30 days in tomatoes hence the introduction or presence of the pathogen in agricultural soil and water could increase risk of blossom contamination. Also, it was determined that all Salmonella serotypes tested were capable of biofilm production on glass coverslips and quartz particles. Biofilm based attachment of Salmonella to sand might aid in its dispersal. To visualize transfer of pathogen from soil particulate to blossom in real-time, bioluminescent S. Baildon, S. Braenderup, S. Newport, S. Javiana and S. Anatum were created.Heat shock procedure was developed to improve electrotransformation efficiency in Salmonella. Transformed strains were compared for bioluminescence production and plasmid stability. S. Newport had the best bioluminescence properties but no difference was observed between strains for plasmid stability. Imaging of soil particulate - S. Newport mixture inoculated blossoms, indicated that the event led to pathogen transfer to blossom. It was also determined that S. Newport â soil particulate contaminated blossoms developed into fruits that were positive for S. Newport. S. Newport presence in blossom, fruit surface and internal tissue indicates that contaminated soil particulate could serve as a vehicle of tomato contamination.

Objective was to assess the toxicity of chromium (Cr) contaminated soil (CS) using the earthworm Lumbricus terrestris. Specific aims were to determine: (1) survival (LC50); .(2) immunotoxicity as indicated by lysozyme activity, coelomocyte counts, secretory (SR) and erythrocyte rosette (ER) formation, and phagocytosis; and (3) compare effects of CS exposure with those of Cr spiked artificial soil (AS) . CS Cr concentration was 8.78 mg/g with 98.2% being Cr^3+ and 1.8% being Cr^6+. Using 14 d AS protocol the LC50 was 6.49% CS: AS mixture. CS concentrations of 0.5, 1.0, 2.5 and 5.0% were sublethal, whereas 6.25, 12.5, 25, 50 and 100% CS were lethal. Sublethal exposure caused no immuno- modulation. Exposure to 50% CS: AS mixture for 5 d caused reduced SR and ER formation. Exposure to AS spiked with 0.27% Cr for 5 d resulted in immunomodulation equivalent to 50% CS: AS mixtures. Results indicated the CS to be acutely toxic.

Contaminated land is a widespread, global issue affecting millions of people. Atrazine is a commonly used herbicide which often contaminates groundwater and drinking water supplies and is associated with adverse health outcomes. Biochar is the solid product of pyrolysis and is associated with several environmental benefits. It may be an effective remediation tool when used as a soil amendment. This thesis investigates the mechanisms through which biochar can immobilise atrazine, and the implications of the mechanisms for remediating contaminated land. Nine biochar samples were obtained from the United Kingdom Biochar Research Centre , which were produced from softwood pellets (SWP), wheat straw pellets (WSP), miscanthus straw pellets (MSP), rice husk (RH) and oil seed rape (OSR) each at pyrolysis temperatures of 550°C and 700°C (excluding OSR at 700°C). The sorption mechanisms controlling atrazine sorption to these biochars were determined through various characterisation methods and batch sorption experiments. The sorption tests showed that sorption to each of the standard biochars occurs via multiple simultaneously occurring mechanisms, which are each promoted under certain conditions. Studies investigating sorption kinetics, isotherms and interactions with humic acids showed that for all biochars in this study, pore filling was a significant process through which atrazine is transported to adsorption sites, although poor intraparticle diffusion for softwood and oil seed rape biochars can prevent efficient transport. Wheat straw and rice husk biochars showed effective pore diffusion, resulting in high sorption capacities. Partitioning was associated with poor remediation outcomes and was significant to softwood biochars, although adsorption dominated overall sorption for all other biochars. pH was shown to significantly influence the occurrence of various sorption mechanisms. At low pH values, most biochars showed evidence of electrostatic repulsion between positive atrazine species and the positively charged biochar surface. At intermediate pH values, all biochars showed strong hydrogen bonding between H+ groups on the surface of the biochar and atrazine. A meta-analysis of previous relevant studies provided further evidence for hydrogen bonding of atrazine to biochar and showed that hydrophobic effects likely play little role in adsorption after accounting for the effects of surface area. Varying contributions of π-π EDA interactions, hydrogen bonding involving biochar O- groups, and interactions with ash minerals resulted in different sorption profiles for each biochar at high pH values. In order to further determine the mechanisms controlling sorption at high pH, surface compositions of SWP550, RH700 and OSR550 biochars were modified using hydrofluoric acid. Modification with hydrofluoric acid successfully removed the ash contents of rice husk and oil seed rape biochars and reduced atrazine removal at high pH values. This suggested that the ash fraction increases atrazine removal at high pH through complexation or catalytic hydrolysis. The roles of the various mechanisms are related to remediation outcomes in a novel manner allowing for the improved design of biochar for environmental remediation.

Du fait des activités industrielles anciennes et du recyclage croissant des produites résiduaires organiques d’origine urbaine dans les sols agricoles, des composés organiques persistants, parmi lesquels les hydrocarbures aromatiques polycycliques (HAP) se retrouvent présents dans les sols français. Dans le cadre de la gestion des sites et sols pollués et de l'évaluation des risques associés aux HAP dans l'environnement, une meilleure connaissance du comportement, de la dissipation, du transfert ou de l’accumulation des HAP dans les sols est alors indispensable. Dans ce but, il est utile de développer une nouvelle génération de modèles numériques basés sur le couplage flexible de l’ensemble des processus majeurs contrôlant la dynamique des HAP dans le sol. Notre travail repose sur la mise en œuvre, dans le cadre de la plateforme VSoil de l’INRA (https://www6.inra.fr/vsoil/The-Project), d'un modèle global interdisciplinaire de dynamique des HAP dans les sols, applicable à l’échelle du terrain et couplant des modules décrivant les principaux processus physiques, biochimiques et biologiques. Ce modèle associe d’une part des modules déjà publiés simulant la décomposition des matières organiques exogènes apportées, le transfert d'eau, le transfert de chaleur et le transport de solutés dans des conditions climatiques réelles, et d’autre part, un nouveau modèle représentant spécifiquement la réactivité des HAP.La démarche numérique adoptée dans ce travail a alors été de i) calibrer le modèle de terrain en utilisant des paramètres déterminés à partir de données expérimentales obtenues au laboratoire et complétées avec une partie des données de terrain obtenues sur une courte période, ii) tester et valider le modèle calibré au terrain à l'aide des données de terrain complémentaires sur des périodes plus longues, iii) tester différentes hypothèses de variation de disponibilité et scenarios climatiques ou d’apports répétés de différents composts sur le devenir de HAP dans le sol. Nos résultats montrent que le modèle peut prédire de manière satisfaisante le devenir des HAP dans le sol sur une gamme de contamination allant de parcelles agricoles amendées avec des matières organiques faiblement contaminées jusqu’à d’anciens sites industriels fortement contaminés et permettent alors de mieux appréhender les processus contrôlant la dynamique des HAP dans ces différents systèmes
Due to former industrial activities and the increasing recycling of organic waste products from urban areas in agricultural soils, increasing amounts of persistent organic compounds, among them polycyclic aromatic hydrocarbons (PAH), are to be found in French soils. In the framework of the management of polluted sites and soils and the risk assessment of PAH transfer in the environment, it is essential to better understand the behavior, dissipation, transfer or accumulation of PAH in soils. For this purpose, there is a need to develop a new generation of numerical models based on the flexible coupling between the processes describing PAH dynamics in soil. Our work presents the development and assessment of such model. It is based on the implementation of an interdisciplinary global model, and applicable at the field scale, for PAH in soil by coupling modules describing the major physical, biochemical and biological processes influencing the fate of PAH in soil, with modules that simulate water transfer, heat transfer, solute transport, and organic matter transformation under real climatic conditions. The coupling is performed using the «VSoil» modeling platform of INRA (https://www6.inra.fr/vsoil/The-Project). The steps of our modelling study are the following: i) calibrate the model at the field scale using previously estimated parameters at the lab-scale and completed with field data on a short period of time, ii) test and validate the calibrated model using field experimental data on mid term periods, iii) test different hypotheses of variation of availability and climatic scenarios or repeated applications of different composts on the fate of PAH in soil. Our results show that the model can adequately predict the fate of PAH in soil over a wide range of contamination ranging from agricultural plots amended with weakly contaminated composted organic wastes to old industrial sites heavily contaminated. This new tool allows, therefore, a better understanding of the processes controlling the dynamics of PAH in these different systems
