Streszczenie:
As the gateway to the beaches of Daytona Beach, Florida, the new Broadway Bridge will be a signature bridge for the community. Mosaic tile murals up to 3 m in height will be incorporated around the columns of the new bridge, as will mosaic panels along the sidewalk handrail. The bridge will also feature ornamental handrail and light fixtures with aesthetic lighting that will display this high-profile structure to the surrounding skyline of Daytona Beach. Additional plantings and fountains will be provided with a grand architectural entrance welcoming visitors to Daytona Beach and onto this signature bridge. The glass tile mosaic art will feature manatees, dolphins, and other nature scenes with species endemic to Florida. Special reviews and approval of the art scenes will be done by the architect of record before fabrication of the large pieces. The Broadway Bridge replacement is being constructed with twin, precast, segmental concrete bridges erected by balanced cantilever construction with an overhead gantry system. The construction includes a cast-in-place, posttensioned flat-slab section on the west end. Each bridge will have a 20-m vertical clearance over the 38-m-wide navigational channel. The bridges will be approximately 918 m in length, with spans varying from 30 to 80 m. The span length variation with structure height will add to the overall aesthetic features of this signature bridge. Graceful lines developed by the segmental design and the additional architectural features provided by the mosaic inlays, lighting, plantings, and fountain make the Broadway Bridge unique and a beautiful addition to the Daytona Beach area.

In this paper, we consider generalizations of the Alexander polynomial and signature of 2-bridge knots by considering the Gordon–Litherland bilinear forms associated with essential state surfaces of the 2-bridge knots. We show that the resulting invariants are well-defined and explore properties of these invariants. Finally, we realize the boundary slopes of essential surfaces as differences of signatures of the knot.

PurposeThe purpose of this paper is to evaluate various reliability measures like reliability, expected lifetime (mean time to failure), signature reliability and compare networks based on the different flows.Design/methodology/approachThe reliability characteristics of complex bridge networks have been evaluated using different algorithms with the help of universal generating function (UGF). Further, the signature reliability of the considered networks has been determined using Owen’s method.FindingsThe present paper proposes an efficient algorithm to compute the reliability indices of complex bridge networks having i.i.d. lifetime components (nodes, edges) with the help of UGF and Owen’s method. This study reveals that a slight change in the complex bridge network affects the reliability significantly. Finally, by the reliability structure function, proposed algorithms are used to find the signature and MTTF. From signature, we have determined the different failure probabilities corresponding to edges of the network.Originality/valueIn this work, we have evaluated reliability characteristics and signature reliability of the complex bridge networks using UGF method and Owen’s method respectively unlike done in the past.

Rumor has it that that the Turks crossed the Marmara Sea on rafts. The Ottoman Empire, which set out from Gallipoli and became a state first and then an empire; reached the Balkan and Europe through bridges. Every bridge built over the huge rivers Ottomans crossed slowly became a signature of that region or city. Spectacular stone bridges, which are both art and aesthetic monuments and meet the transportation needs of the army, were built on the rivers from the capital to the last point Empire reached. Many bridges, especially the Mostar Bridge, Mehmed Paša Sokolović Bridge, Stone Bridge in Skopje, Bridge over Maritsa in Edirne, Bridges over the Tunca and the Uzunköprü (Long Bridge), which were built on the great Balkan rivers, symbols of abundance and plenitude, have been the subject of poems, shehrengiz and historical poems of classical poets, and also prose works and travel books. through various dreams and imaginations, these bridges are from time to time places where lovers go out for an evening stroll and meet, and sometimes through similes and metaphors compared to lover’s body, rainbow, Milky way, or the Sirat Bridge. These bridges are so ostentatious and appear as a great kemend that connects two mountains. The stone bridges built on the Balkan rivers from Edirne to Mostar that have poems written about them will be determined and interpreted in our article.

Once projected into the new representation, the bridge admittances of three historical violins made by Stradivari and a famous reproduction revealed high similarity. PCA highlighted the importance of signature mode frequencies, bridge hill behavior, and signature mode amplitudes in distinguishing different violins.

The efficiency of a bridge system and a 2-out-of-4:G system has been evaluated in the proposed work. The technique for evaluating the system’s reliability and signature reliability of different models has been employed in this work. We implemented the structure–function technique to determine the reliability function of the bridge model, 2-out-of-4:G model, and three serially connected modules system, which avoids Boland’s Formula to minimize the calculations and to reduce the failure rate of the complex system in order to maximize its capability. Three illustrative systems of five, four, and fourteen vertices each were separated and placed in series-parallel combinations. There is another simpler and faster concept to determine the system’s tail signature by using the coefficient of the reliability function described in this paper. The system’s reliability, minimal signature, tail signature, signature, B-P index, expected time, and expected cost were all calculated using this approach. This work also describes the reliability estimation of three serially connected systems with the help of different approaches. These findings contribute to the probabilistic method’s usefulness in analyzing the system’s signature and reliability.

AbstractRecent discoveries that the Earth has a supra-chondritic 142Nd signature have thrown chondritic geochemical Earth models into doubt. Several solutions have been proposed to explain this discrepancy but none has been widely accepted. This paper reviews Nd isotope data for the Moon which bridge the gap between the 142Nd signatures of chondritic meteorites and the accessible Earth. Different chondrite classes define a 142Nd–148Nd correlation line attributed to incomplete mixing of nucleosynthetic components in the solar nebula. Terrestrial standards have 142Nd signatures well above this correlation line, but the 142Nd signature of the Bulk Moon is c. 6 ppm lower than terrestrial (assuming a chondritic Sm/Nd ratio) and falls within error of enstatite chondrites. In view of the demonstrated isotopic similarity between the Earth and Moon, giant impact models require the Moon to be a sample of the early Earth. Therefore, it is inferred that the Earth–Moon system was generated from material similar to enstatite chondrites, but Earth's mantle experienced Sm/Nd fractionation very soon after the Moon-forming collision. Such fractionation processes have been attributed to subduction of early Fe-enriched crust into a deep mantle storage reservoir. Because Sm/Nd fractionation occurred when most 146Sm had already decayed, the hidden incompatible-element-enriched reservoir only became slightly depressed in its 142Nd signature, explaining why this signal has not yet been detected in ocean island basalt sources.

Streszczenie:
In consideration of the important role that bridges play in transportation system, their safety, durability and serviceability have always been deeply concerned. Recently, many long-span bridges have been instrumented with Structural Health Monitoring Systems (SHMS) to provide bridge engineers with the information needed for decisions-making in management and maintenance. However, efficient use of monitoring data remains a challenge confronted before engineers. Recently, methodologies based on monitoring data while robust to random disturbance and sensitive to damage have received worldwide attention. In this context, this study proposes an innovative damage detection methodology based on structural responses induced by traffic load. First, vehicle-induced strain responses are found to be separable from the strain induced by operational loads, owing to their unique characteristics. This is achieved by a detailed investigation on the relationship between strain measurements and operational loads including temperature, wind as well as vehicles based on long-term monitoring data. From the vehicular load and pertinent strain response, the strain influence line (SIL) can be further identified. As a structural signature, SIL can be used to provide a reasonable assessment of the bridge health condition at least in the vicinity of strain monitoring point. Two damage indexes are therefore derived from the identified SIL, which are promising for damage evaluation because they are: (a) capable of revealing structural deterioration; (b) immune to influences of environmental changes; (c) adaptable to the random characteristic exhibited by long-term monitoring data. Besides, the SIL identification procedure and its theoretical basis are elaborated to respectively handle the case where the vehicle load is available or not, which is also applicable to identify the influence line of other measurements such as stresses. The proposed damage methodology is applied to the cable-stayed bridge spanning the main navigation channel of Shanghai Yangtze River Bridge, and the result shows its effectiveness.

Streszczenie:
Vibration-based damage detection (VBDD) comprises a family of nondestructive testing methods in which changes to signature dynamic characteristics are used to track the condition of a structure. Although VBDD methods have been successfully applied to various mechanical systems and to simple beam-like structures, significant challenges remain in extending this technology to complex, spatially distributed structures such as bridges. In the present study, numerical simulations using a calibrated finite element model were used to investigate the use of VBDD methods to detect small-scale damage on a two-span, integral abutment overpass structure located in Saskatoon, Saskatchewan. Five different VBDD techniques were evaluated, as were the effects of sensor spacing, mode shape normalization, and uncertainty in the measured mode shapes. It was found that localized damage to the top concrete cover of the bridge deck could be reliably detected and located if the sensors were located sufficiently close to the damage and if uncertainty in the mode shapes was attenuated through the use of a sufficient number of repeated trials. Furthermore, preliminary studies indicate that it may be possible to detect damage using sensors that are placed well away from the damaged area.Key words: vibration-based damage detection, structural health monitoring, integral abutment bridge, numerical modelling, field testing.

Capstone courses in undergraduate education, especially in professional schools such as journalism, usually try to teach students to think and perform like the future practitioners they intend to become. Internships, practice-based subjects, work experience courses—they all aim to bridge the students’ knowledge from the largely theoretical to the urgently practical; knowledge that will be essential in the workplace, whether it’s a doctor in a hospital, a lawyer in a courtroom or a teacher in a classroom. Shulman’s ground-breaking insight was to articulate these as Signature Pedagogies and to define them clearly in three dimensions—to think, to perform, and to act with integrity—just as experienced practitioners in the field are doing. This article contends that overseas Work Integrated Learning (WIL) courses such as those examined here, conducted by the researcher for the University of Queensland (UQ), are a Signature Pedagogy because the student participants are required to behave, think and perform ethically, just as foreign correspondents must do.

We show how the Model Signature Graphs can be used to compute topological distances among models and how to use these measures to create metric spaces for indexing and clustering of solid models. We describe the optical and electrical analysis of a micro-explosion of silicon and micro-thermite micro-bridges with a spectral temporal resolution within a 2 ms time range. The intensity of peaks and their mean lifetimes are calculated to identify the contributing atomic and molecular species. Singly ionised atoms and molecules were the main contributor to spectroscopic signature of emission. It is shown that micro-bridges can be used to characterise small (safe) < 10 mg amounts of energetic materials. Use of ions at low ionisation numbers of Si as well as Cu-oxides with different metal-to-oxygen ratios allows the thermal conditions for micro-explosions and a higher temperature of the discharge plasma to be engineered. Temperatures of 5000 ± 500 K (the black-body radiation fit) were achieved with simple micro-bridge junctions of 10–30 Ω resistance initiated with 100–250 V (triggered capacitor discharge). The demonstrated approach can be applied in material science research concerning ultra-fast melting, phase transitions, and detailed steps leading towards detonation (exponentially growing exothermic chemical reactions).

Current HIV vaccines are only partially efficacious, presenting an opportunity to identify correlates of protection and, thereby, potential insight into mechanisms that prevent HIV acquisition. Two independent preclinical challenge studies in nonhuman primates (NHPs) previously showed partial efficacy of a mosaic adenovirus 26 (Ad26)–based HIV-1 vaccine candidate. To investigate the basis of this protection, we performed whole transcriptomics profiling by RNA sequencing (RNA-seq) in sorted lymphocytes from peripheral blood samples taken during these studies at different time points after vaccination but before challenge. We observed a transcriptional signature in B cells that associated with protection from acquisition of simian immunodeficiency virus (SIV) or the simian-human immunodeficiency virus (SHIV) in both studies. Strong antibody responses were elicited, and genes from the signature for which expression was enriched specifically associated with higher magnitude of functional antibody responses. The same gene expression signature also associated with protection in RV144 in the only human HIV vaccine trial to date that has shown efficacy and in two additional NHP studies that evaluated similar canarypox-based vaccine regimens. A composite gene expression score derived from the gene signature was one of the top-ranked correlates of protection in the NHP vaccine studies. This study aims to bridge preclinical and clinical data with the identification of a gene signature in B cells that is associated with protection from SIV and HIV infection by providing a new approach for evaluating future vaccine candidates.

Streszczenie:
The canonical sequence LSGGQ, also known as the signature sequence, defines the adenosine triphosphate (ATP)-binding cassette transporter superfamily. Crystallographic studies reveal that the signature sequence, together with the Walker A and Walker B motifs, forms the ATP-binding pocket upon dimerization of the two nucleotide-binding domains (NBDs) in a head-to-tail configuration. The importance of the signature sequence is attested by the fact that a glycine to aspartate mutation (i.e., G551D) in cystic fibrosis transmembrane conductance regulator (CFTR) results in a severe phenotype of cystic fibrosis. We previously showed that the G551D mutation completely eliminates ATP-dependent gating of the CFTR chloride channel. Here, we report that micromolar [Cd2+] can dramatically increase the activity of G551D-CFTR in the absence of ATP. This effect of Cd2+ is not seen in wild-type channels or in G551A. Pretreatment of G551D-CFTR with the cysteine modification reagent 2-aminoethyl methane thiosulfonate hydrobromide protects the channel from Cd2+ activation, suggesting an involvement of endogenous cysteine residue(s) in mediating this effect of Cd2+. The mutants G551C, L548C, and S549C, all in the signature sequence of CFTR's NBD1, show robust response to Cd2+. On the other hand, negligible effects of Cd2+ were seen with T547C, Q552C, and R553C, indicating that a specific region of the signature sequence is involved in transmitting the signal of Cd2+ binding to the gate. Collectively, these results suggest that the effect of Cd2+ is mediated by a metal bridge formation between yet to be identified cysteine residue(s) and the engineered aspartate or cysteine in the signature sequence. We propose that the signature sequence serves as a switch that transduces the signal of ligand binding to the channel gate.

The organic coating of bridge steel girders is subjected to physical scratches, corrosion, and aging in natural weathering. The breakdown of the coating may cause serviceability and safety problems if left unnoticed. Conventional coating inspection is time-consuming and lacks information about the coating’s chemical integrity. A hyperspectral imaging method is proposed to detect the condition of steel coatings based on coating-responsive features in reflectance spectra. A field test was conducted on the real-world bridge, which shows obvious signs of degradation. The hyperspectral signature enables an assessment of the coating’s health and defect severity. The results indicated that the coating scratch can be effectively located in the domain of a hyperspectral image and the scratch depth can be determined by mapping a scratch depth indicator (SDI = R532 nm/R641 nm). Rust sources and products in steel girders can be identified by the unique spectral signatures in the VNIR range, and the rust stains (and thus stain areas) scattered on the coating can be pinpointed at pixel level by the chloride rust (CR) indicators >1.11 (CR = R733 nm/R841 nm). The chemical integrity of a topcoat is demonstrated by the short-wave infrared spectroscopy and the topcoat degradation can be evaluated by the decreased absorption at 8000 cm−1 and 5850 cm−1. Hyperspectral imaging enables faster and more reliable coating condition detection by the spectral features and provides an alternative for multi-object coating detection.

Boston, in the forefront of the American Revolution two centuries ago, is now in the forefront of another revolution in the field of cable-stayed bridge technology. New technologies and innovations have become hallmarks of the Leonard P. Zakim Bunker Hill Bridge—a structure unique in the world. This crossing of the Charles River brought the community, engineers, and architects together to create a “signature bridge” and a “gateway” to the city. Located at a preeminent point where Paul Revere crossed in 1775, the bridge took on special meaning from a historical perspective. Numerous alternatives were studied for the crossing and the interchange configurations on both sides of the river. Based on significant community input and evaluation of costs for different alternatives, the option known as the “non-river tunnel” alternative was chosen, which required a 10-lane crossing of the river. The 10 lanes include 4 lanes each for I-93 northbound and southbound and a 2-lane ramp on the east side. Some impediments the bridge had to contend with included the Orange Line subway adjacent to and below the bridge; the close proximity of the Charles River lock and dam and the need to maintain navigation; a major water main in the area of the south tower footing; a cantilevered 2-lane ramp on only one side of the structure; the existing Storrow Drive ramps at the south end, dictating the arrangement of the stay cables in the back spans; and a new tunnel at the south end of the structure.

The Zagreb aquifer is the main source of potable water for the inhabitants of the City of Zagreb and Zagreb County. It presents a strategic water reserve protected by the Republic of Croatia. All previous studies related to the definition of the groundwater–surface interaction in the study area have been made based on the isotopic composition of the Sava River from the location of the Domovinski Most bridge, which is located downstream of most pumping well fields. In 2019, a new monitoring station was established at the Podsusedski Most bridge, at the entrance of the Sava River into the Zagreb aquifer, approximately 23 km upstream of the Domovinski Most bridge. Within this research, water isotope data (δ2H, δ18O, deuterium excess) from both Sava River and groundwater sites were used along with hydrologic data to examine the extent to which hydrologic conditions affect the isotope signature and whether the interaction between groundwater and the Sava River causes a change in the isotopic composition of the Sava River. In addition, δ18O amplitudes were estimated for different time periods, as well as the mean residence time for the hydrological year 2019/2020. For that purpose, different statistical methods were applied to the new monthly data for six years for the Domovinski Most bridge and two years for the Podsusedski Most bridge. The δ18O amplitudes vary from 0.22 to 1.86 depending on the time interval and hydrological conditions, while the mean residence time for the hydrological year 2019/2020 was estimated to be about 2.5 months.

Ovarian carcinoma immunoreactive antigen-like protein 2 (OCIAD2) is a protein with unknown function. Frequently methylated or downregulated, OCIAD2 has been observed in kinds of tumors, and TGFβsignaling has been proved to induce the expression of OCIAD2. However, current pathway analysis tools do not cover the genes without reported interactions like OCIAD2 and also miss some significant genes with relatively lower expression. To investigate potential biological milieu of OCIAD2, especially in cancer microenvironment, a nova approach pbMOO was created to find the potential pathways from TGFβto OCIAD2 by searching on the pathway bridge, which consisted of cancer enriched looping patterns from the complicated entire protein interactions network. The pbMOO approach was further applied to study the modulator of ligand TGFβ1, receptor TGFβR1, intermediate transfer proteins, transcription factor, and signature OCIAD2. Verified by literature and public database, the pathway TGFβ1- TGFβR1- SMAD2/3- SMAD4/AR-OCIAD2 was detected, which concealed the androgen receptor (AR) which was the possible transcription factor of OCIAD2 in TGFβsignal, and it well explained the mechanism of TGFβinduced OCIAD2 expression in cancer microenvironment, therefore providing an important clue for the future functional analysis of OCIAD2 in tumor pathogenesis.

Context. The majority of stars form in binary or higher order systems. The evolution of each protostar in a multiple system may start at different times and may progress differently. The Class 0 protostellar system IRAS 16293–2422 contains two protostars, “A” and “B”, separated by ~600 au and embedded in a single, 104 au scale envelope. Their relative evolutionary stages have been debated. Aims. We aim to study the relation and interplay between the two protostars A and B at spatial scales of 60 au up to ~103 au. Methods. We selected molecular gas line transitions of the species CO, H2CO, HCN, CS, SiO, and C2H from the ALMA-PILS spectral imaging survey (329–363 GHz) and used them as tracers of kinematics, density, and temperature in the IRAS 16293–2422 system. The angular resolution of the PILS data set allows us to study these quantities at a resolution of 0.5′′ (60 au at the distance of the source). Results. Line-of-sight velocity maps of both optically thick and optically thin molecular lines reveal: (i) new manifestations of previously known outflows emanating from protostar A; (ii) a kinematically quiescent bridge of dust and gas spanning between the two protostars, with an inferred density between 4 × 104 cm−3 and ~3 × 107 cm−3; and (iii) a separate, straight filament seemingly connected to protostar B seen only in C2H, with a flat kinematic signature. Signs of various outflows, all emanating from source A, are evidence of high-density and warmer gas; none of them coincide spatially and kinematically with the bridge. Conclusions. We hypothesize that the bridge arc is a remnant of filamentary substructure in the protostellar envelope material from which protostellar sources A and B have formed. One particular morphological structure appears to be due to outflowing gas impacting the quiescent bridge material. The continuing lack of clear outflow signatures unambiguously associated to protostar B and the vertically extended shape derived for its disk-like structure lead us to conclude that source B may be in an earlier evolutionary stage than source A.

Debates exist about the separation of the academy and the practicum. The purpose of this study is to explore how its findings may contribute to this discussion. The unique pedagogies of the university and practice are explored and consideration is given to whether practice educators hold an intermediary role. A qualitative study of an undergraduate programme in England using thematic analysis was undertaken once ethical approval was given. Individual interviews and focus groups were used. A total of 48 participants were interviewed at two different points in time (2008 and 2011-2012). Although this is a limited small scale study not susceptible to generalisation it may be able to provide an analytic generalisation, which may be replicable with further studies. The findings indicated practice educators struggled to act as a bridge because the academy and the practicum have conflicting and competing signature pedagogies. The contribution of this study is to the contemporary discussion on the SWRB changes and models such as Frontline.

Bridge-like structures have been reported in recent studies for a binary mixture with square well fluids in 2D and 3D using different techniques. In this paper, we present our NVT simulation results in 3D with symmetric additive water and oil-like molecules. We use Dirac's delta function defined using theta step function for evaluating pressure components and surface tension values. Our investigation reveals that though 3D NVT MC results are in qualitative agreement with the published results showing all the structures as bridge-like, complete wetting of the walls by the preferred component and micelle structures but the critical parameters as surface attraction strength significantly alter to lower values for PW and CW cases in comparison to the 2D MD cases. We have studied cases with pore width as H = 6.0, 8.0, and 10.0 at T = 0.70, 0.80, and 0.90 for εAB = 0.0, 0.25, 0.50, and 1.0. For all of the bridge cases ε Wall -A = -0.50 and λ wall -A = 1.0. For fluid–fluid interaction λ ff = 1.2 and 1.5 are taken with εAA = εBB = -1.0. Our pressure and surface tension values show clear signature of confinement effect and dependence on other parameters for the bridge cases but it is concluded that different structural transitions depend only on the predefined energetically favorable and unfavorable interfaces which in turn comes from the separation of the two walls and average initial density. Our density profiles confirm the evolved structures.

Ground-penetrating radar (GPR) is a nondestructive investigation tool that is usually used in flexible pavement evaluation to estimate the thicknesses of the various layers composing the pavement. GPR is also used in flexible pavements to detect subsurface distresses, such as moisture accumulation and air voids. For rigid pavements and bridge decks, GPR is used to measure the thickness of the concrete slab and detect the location of reinforcing bars (rebar). Rebar detection is typically achieved, in this case, when an experienced operator finds the rebar's classic parabolic signature in the GPR data. This paper presents image-processing techniques that can be used to detect the rebar parabolic signature automatically in GPR data collected from rigid pavements with a high-frequency ground-coupled antenna. After detection of the rebar, the reflected parabolic shape is fit to a theoretical reflection model to estimate the pavement's dielectric constant and the rebar depth. The algorithms were validated on GPR data collected from a known continuously reinforced concrete pavement section. The technique showed an average error of 2.6% on the estimated rebar cover depth.

Abstract The characterization of human DC subsets is essential for the design of new vaccines and for better translation of mouse immunological data to the clinic. Previously, we showed that human epidermal Langerhans cells (LCs) were highly efficient at priming effector CD8+ T cell responses. However, the ability of mouse LCs to cross-present antigens and drive potent CTL responses is controversial. We used expression profiling to find the human LC equivalent in the mouse. Our studies suggest that the mouse equivalent to the human LC are the two mouse thymic DC subsets. Our studies also reveal a distinct genetic signature of over 25 genes, that could identify and predict which DC subsets can cross-present. The cross-presentation signature was present in the human blood CD141+ DCs and correctly identified the ability of other distinct human and mouse DC subsets to cross present. We confirmed these predictions with the functional ability of DCs to efficiently induce antigen-specific effector CD8+ T cells. Our studies may help bridge differences between mouse and human DCs and facilitate the application of these data to the clinic.
