Sea squirt ( Halocynthia roretzi) is considered a potential cause of human norovirus in Korea. This study investigated the effect of high hydrostatic pressure at 100–500 MPa for 5 min at room temperature (23 ± 2 ℃) on the inactivation of murine norovirus-1 (initial inoculum of 6–7 log10 plaque forming units/ml) as a human norovirus surrogate in fresh sea squirt. The effects of high hydrostatic pressure on the Hunter colors and pH were also examined as the main indices of quality. No reductions in murine norovirus-1 titers were observed in sea squirt treated at 100–400 MPa. However, murine norovirus-1 in sea squirt was completely inactivated by 500 MPa of high hydrostatic pressure. Furthermore, the Hunter colors (“L,” “a,” and “b”) and pH values (6.10–6.19) were not significantly ( P > 0.05) different between non-high hydrostatic pressure-treated sea squirts and all high hydrostatic pressure-treated sea squirts. Therefore, 500 MPa of high hydrostatic pressure at room temperature may be an optimal treatment for Tunicata meat without altering the food quality (color and pH).

Following recent tendencies in fisheries policy to ensure both sustainability of ecosystems and conservation of economically sustainable fisheries, protection of vulnerable resources with low direct economic value comes to the focus of fisheries management on ecosystem principles. One of the problems of modern fishing is a negative impact of bottom trawling because of destruction of benthic organisms vulnerable to mechanical impacts. This by-effect of fishing could affect negatively on functioning of bottom biocenoses, reproduction of exploited species, and decrease generally productivity of vulnerable marine ecosystems (VME). Potential VME indicators are determined for the area of the Anadyr Bay in the Bering Sea on the base of results of 4 benthic surveys using bottom sampler (1985, 2005) and bottom trawl (2008, 2012), as the most common species in some macrozoobenthic groups of epifauna. They are Gersemia rubiformis for soft corals, Myxilla incrustans , Halichondria panicea , Semisuberites cribrosa for sponges, Halocynthia aurantium , Boltenia ovifera for sea squirts, Cystisella saccata , Flustra foliacea for bryozoans, Chirona evermanni for barnacles, and Gorgonocephalus eucnemis for brittle stars. Their distribution is mapped. According to their life history and feeding habits, these species-indicators are divided onto two groups: immobile sestonophages (alcyonarians, sponges, ascidians, bryozoans, cirripedians) and mobile filtrators (brittle stars). The first group prevails on hard and mixed grounds mainly along southwestern and northeastern coasts of the Anadyr Bay at the depths of 80-90 m (sponges and bryozoans - to 250 m in the Navarin Canyon) with relatively warm water, active hydrodynamics and high biological productivity. The second group represented by G. eucnemis dominates on soft sediments in the central part of the Anadyr Bay with the depths of 50-270 m occupied by the cold water pool. Quantitative distribution of brittle star, on the one hand, and barnacles with sea squirts, on the other hand, is alternative to each other. On the contrary, barnacles, sponges and sea squirts have similar distribution of the biomass, being complementary species. Distribution patterns of all species-indicators are stable for many decades. However, biomass of some these species has changed in the southern Anadyr Bay between the similar surveys conducted in the 2008 and 2012: the mean biomass of barnacle Ch. evermanni and sea squirt H. aurantium had decreased in 6.5 and 3.7 times, respectively, whereas the mean biomass of sponges, brittle star G. eucnemis and sea squirt B. ovifera did not change. Bottom trawl fishery is not active in the northwestern Bering Sea, moreover, the habitats of immobile sestonophages with hard grounds are avoided by bottom trawlers being dangerous for fishing gears, so the observed decreasing of two species abundance is presumably caused by natural reasons or is a random error of the mosaic-distributed stocks assessment with insufficiently dense sampling grid.

This dissertation examines two issues related to the ecological and evolutionary consequences of sexual and asexual reproduction in colonial marine invertebrates. The first two chapters explore the extent to which the planktonic larval phase limits the distribution and abundance of a colonial ascidian, Oiplosoma similis. The third chapter examines some of the fitness consequences of alterations in the pattern of asexual reproduction by colony fragmention in similis. All research was carried out on the fringing coral reef surrounding Coconut Island which is located in Kaneohe Bay, Oahu, Hawaii.

The ascidian species Ciona savignyi and C. intestinalis are invasive species but show interspecific differences in their population response to hypo-saline stress associated with heavy winter-run off events that are predicted to become more frequent due to climate change. Despite an almost world-wide distribution, C. intestinalis seems to be more susceptible to hypo-saline stress than the geographically more limited C. savignyi. Given that the genomes of both species are fully sequenced, we were able to compare their proteomic response to both acute and chronic salinity to characterize the mechanisms that are responsible for setting tolerance limits to hyposaline conditions in these two congeneric species. For the acute hypo-saline stress experiment, we exposed each species to decreasing salinities, 100%, 85% and 70% full-strength seawater, for 6 hours followed by a 4-hour recovery at 100%. In the chronic salinity stress experiment, each species was kept at 100% or 85% with individuals removed for analysis during a 16-day time course. Organisms were dissected to remove the tunic, and 2D SDS-PAGE was performed to separate proteins and characterize changes in protein abundances. In the acute experiment, we determined 5% and 19% of the proteins to be significantly changing abundance in C. savignyi and C. intestinalis, respectively, due to the treatment effect. For both species in the chronic experiment, we determined over 40% of the proteins to be significantly changing abundance given the treatment, time, or interaction effect. Analysis of these proteins with MALDI TOF-TOF mass spectrometry has identified numerous proteins implicated in cellular stress responses, including energy metabolism (glycolysis, ATP & NADH production), cytoskeletal restructuring, and protein turnover, providing insights into the intense cellular restructuring that occurs following hypo-saline exposure.

This thesis describes the chemical investigations of several ascidian species collected from the Great Barrier Reef, Queensland, Australia. The thesis is divided into two separate components, Part A focuses on the isolation and structure elucidation of 11 previously undescribed ascidian metabolites. All structures were assigned using a combination of spectroscopic and/or chemical methods. Part B relates to the isolation and chemical conversion of a natural product to a combinatorial template. The natural product template was subsequently used in the generation of a solution-phase combinatorial chemistry library. A further two combinatorial libraries were generated from a synthesised model compound that was related to the natural product template. Part A. Investigation of Aplidium longithorax collected from the Swains Reefs resulted in the isolation of two new para-substituted cyclofarnesylated quinone derived compounds, longithorones J (30) and K (31). The former compound had its absolute stereochemistry determined by the advanced Mosher method. From an Aplidium longithorax collected from Heron Island, two new cyclofarnesylated hydroquinone compounds, longithorols C (46) and D (47) and a novel macrocyclic chromenol, longithorol E (48) were isolated. Longithorol C (46) had its absolute stereochemistry determined by the advanced Mosher method. Chemical investigation of the deep-purple colonial ascidian, Didemnum chartaceum collected from Swains Reefs led to the isolation of five new lamellarin alkaloids, which included the 20-sulfated derivatives of lamellarins B (94), C (95) and L (96), the 8-sulfated derivative of lamellarin G (97) and the non-sulfated compound, lamellarin Z (98). The known lamellarins A (63), B (80), C (64), E (65), G (67), and L (71) plus the triacetate derivatives of lamellarin D (82) and N (83) were also isolated. An aberration in the integration of signals in the 1H NMR spectra of the 20-sulfated derivatives (94-96) led to NMR relaxation studies. T1 values were calculated for all protons in the sulfated lamellarins (94-97) and their corresponding non-sulfated derivatives (80, 64, 71, 67). The protons ortho to the sulfate group in compounds (94-97) had T1 values up to five times larger than the corresponding protons in their non-sulfated derivatives (80, 64, 71, 67). A specimen of Eudistoma anaematum collected from Heron Island was shown to contain a new b-carboline alkaloid, eudistomin V (130), in addition to the two known metabolites, eudistomin H (105) and I (106). Part B. The known natural products, 1,3-diphenethylurea (29), 1,3-dimethylxanthine (30), 1,3-dimethylisoguanine (31) and the salts of tambjamine C (16), E (18) and F (19) were isolated from the ascidian, Sigillina signifera collected in Blue Lagoon, Lizard Island. Base hydrolysis on mixtures of the salts of tambjamine C (16), E (18) and F (19) resulted in the production of 4-methoxy-2,2-bipyrrole-5-carbaldehyde (26). This natural product template (26) was used in the generation of an enamine combinatorial chemistry library (98, 103-111) using solution-phase parallel synthesis. The biaryl compound, 4-(2-thienyl)-1H-pyrrole-2-carbaldehyde (59) was successfully synthesised using Suzuki-Miyaura coupling conditions and subsequently used as a template in the generation of an amine (67, 77, 80-87) and imine (78, 92-95) combinatorial library using solution-phase parallel synthesis.

AbstractContemporary media and public accounts have increasingly framed the refugee ‘crisis’ in terms of security, with refugees considered as masses to be managed and controlled, migrants pointed at with generic allegation of terrorist threat, and state borders closed and militarized. Securitization of migration may not be a new phenomenon (Saunders 2014) but it is one that has recently received a great deal of attention (see among others Bigo 2002; Pugh 2004; Huysmans and Squire 2009; Huysmans 2000; Musarò 2017; Vaughan-Williams 2015; Watson 2009). What all these scholars have in common is that they highlight different ways through which refugees are represented, described, and thought of as threat. Media and public accounts have consistently represented refugees through words such as plight, invasion, flood, hordes, or waves (Friese 2017). The “highly heterogeneous and (too) strongly mediation-dependent European politics created an array of – in most cases negative – interpretations of the Refugee Crisis” (Krzyżanowski et al. 2018). In line with this narrative, at the visual level, the images that have accompanied the news on refugees have mostly included overcrowded boats, long lines of people in need, and looming masses of people crammed at border fences.

Abstract Hemichordates comprise worm-like Enteropneusta (= Balanoglossida) and tentaculate Pterobranchia. Enteropneusts have gill slits, so are linked with chordates, particularly because they also have a dorsal, sometimes tubular, nerve cord in the collar. They lack a notochord, however, so are placed in a phylum separate from the Chordata. The ‘tornaria’ larvae of one family are like some echinoderm larvae. Pterobranchia are closely related to enteropneusts but the only pterobranchs known from Britain belong to a minute colonial genus, atypical of hemichordates.

Streszczenie:
Construction of sea-ports in limanic aquathory along the Northern Black Sea coast have prefer impact on the coastal environment. Especially deep impact have liman bottom artificial deepening, liman slopes artificial reconstruction, navigate canals, jetties of different sizes, ground dumps etc. Typical examples are Odessa, Ill'ichevsk, Yuzhniy commercial sea-ports in northern sector of the Black Sea. On shores nearest Port Yuzhniy jetties was firstly employed new construction against sea wave erosion. This new artificial shore terrace, that consist of natural matherials, expressed as shore protection, purification of sea water, needless ground depot, new shore territory and squire for recreants simultaneous.

<div class="section abstract"><div class="htmlview paragraph">Internal combustion engine (IC engine) vehicles are commonly used for transportation due to their versatility. Due to this, efficiency in design process of IC engines is critical for the industry. To assess performance capabilities of an IC engine, thermal predictions are of utmost consequence. This study describes a computational method based on unsteady Reynolds-averaged Navier–Stokes equations that resolves the gas–liquid interface to examine the unsteady single phase/multiphase flow and heat transfer in a 4-cylinder Inline (i-4) engine. The study considers all important parts of the engine i.e., pistons, cylinder liners, head, block etc. The study highlights the ease of capturing complex and intricate flow paths with a robust mesh generation tool in combination with a robust high-fidelity interface capturing VOF (Volume-of-Fluid) scheme to resolve the gas-liquid interfaces. Results obtained show the dominant processes that determine the oil distribution to be the piston speeds from reciprocating motion of the pistons and the flow rate of oil injected through the squirter jets. A novel heat transfer approach (mixed time-scale coupling) is used to solve for the temperatures in the engine solids. To resolve the heat transfer between the squirter jet flow and piston/cylinder liner, the approach separates the conjugate heat transfer calculation into a fluid heat simulation and a solid heat simulation while setting up a communication method to exchange the thermal boundary conditions between them. The communication method takes into account the changes in heat transfer from piston to cylinder liner due to changes in position of the piston. The study also considers water jacket cooling in the engine block and head to holistically assess the overall heat transfer in the engine solids. Results of thermal simulation show the solid temperatures to be in direct correlation with the oil distribution near those solids as well as the flow rate of water coolant in water jacket cooling system.</div></div>
