ABSTRACT The stress-activated protein kinases (SAPKs), namely, p38 and JNK, are members of the mitogen-activated protein kinase family and are important determinants of cell fate when cells are exposed to environmental stresses such as UV and osmostress. SAPKs are activated by SAPK kinases (SAP2Ks), which are in turn activated by various SAP2K kinases (SAP3Ks). Because conventional methods, such as immunoblotting using phospho-specific antibodies, measure the average activity of SAP3Ks in a cell population, the intracellular dynamics of SAP3K activity are largely unknown. Here, we developed a reporter of SAP3K activity toward the MKK6 SAP2K, based on fluorescence resonance energy transfer, that can uncover the dynamic behavior of SAP3K activation in cells. Using this reporter, we demonstrated that SAP3K activation occurs either synchronously or asynchronously among a cell population and in different cellular compartments in single cells, depending on the type of stress applied. In particular, SAP3Ks are activated by epidermal growth factor and osmostress on the plasma membrane, by anisomycin and UV in the cytoplasm, and by etoposide in the nucleus. These observations revealed previously unknown heterogeneity in SAPK responses and supplied answers to the question of the cellular location in which various stresses induce stimulus-specific SAPK responses.

Sapote mamey is a sweet and aromatic tropical fruit that is very perishable. It is a climacteric fruit and has high rates of respiration and ethylene production. Maturity indices for this commodity are difficult to define because fruit show few changes in external appearance as they ripen. The fruit flesh, however, shows large changes in color, firmness, and sugar content measured as soluble solids content (SSC). The objective was to model fruit ripeness from measurements of SSC. We selected SSC because it is easy to measure and because sweetness is an important quality attribute in sapote mamey. Typical values of SSC range from 12% (immediately after harvest) to 30% to 35% (ripe fruit). A linear-plateau model was used to describe the changes in SSC over time of ripening fruit kept at different temperatures. The model assumed that, as fruit ripened, SSC increased at a linear rate reaching a maximum of 30% SSC at the ripe stage after which SSC changed little. From the model we calculated the rate of fruit ripening and the time to reach the ripening stage (30% SSC). The rate of ripening showed a quadratic relationship with storage temperature. Fruit kept at 27, 25, or 20 °C ripened 3.5, 5, or 7 days after harvest. The model can be used to estimate when fruit will reach the ripe stage, as long as we know the initial SSC and storage temperature. This model was constructed from data obtained over 2 years from fruit grown in the state of Morelos, Mexico. It is still to be tested for its applicability on fruit from other growing regions.

The mamey sapote is a fruit highly appreciated by its sweet flavor. Although this crop has been cultivated in tropical America for several centuries, there is still little horticultural information about it. The fruit is very perishable and postharvest losses are usually large. The objective was to evaluate the postharvest changes in fruit quality under storage conditions. Fruit grown in Morelos, Mexico, were harvested when there was a color change from green to pink in the tissue just below the fruit cortex. Fruit were stored at 10 °C or kept at 20 °C (65% RH) or ambient temperature (Mean daily temperature = 27 °C) for 24 days or until fruit deteriorated. After storage at 10 °C, fruit were kept at 20 °C to allow for fruit ripening. Fruit kept at 27 °C ripened 3 days after harvest, while those at 20 °C ripened 5 days after harvest. Fruit at 10 °C showed little physical or chemical changes over the storage period. Ripening included fruit softening, a change in pulp color from a yellow or pale pink to an intense pink color, and an increase in SSC. No change in fruit cortex color was detected as ripening progressed. Ripe fruit showed a 30% to 35% of SSC, and a firmness (resistance to penetration) of 7 N. In overripe fruit, pulp color turned brown. Fruit stored at 10 °C and then ripened at 20 °C showed an irregular ripening, with portions of the fruit with a high firmness and others with a low firmness. This irregular ripening was more severe on fruit stored at 10 °C for 14 days than those stored for 7 days. This disorder was probably a result of chilling injury.

There are very few postharvest studies about the mamey sapote fruits. The lack of appropriate harvest indexes for this crop result in fruits having a wide variability in maturity after harvest. Fruit skin shows no apparent changes in color as maturity progresses. Another complication results from harvesting the fruit with long poles, which restricts the harvester from touching the fruit to evaluate fruit softening. The objective of this study was to evaluate the effect of exogenous ethylene applications to fruits on increasing the uniformity of fruit maturity. Fruits were harvested every 2 weeks over a 4-month period. Fruit harvest was initiated 8 weeks before the estimated ripening day. Fruits were treated by immersion for 1 min in an ethephon solution at 0, 500, or 1000 mg·liter–1 and stored at 20°C (65% RH) for 4 or 8 days. After the storage period, fruits were analyzed for fruit firmness, color (external and internal), pH, titratable acidity, soluble solids content, ascorbic acid, and starch. Postharvest exogenous applications of ethylene stimulated postharvest ripening of the mamey sapote fruits. Ripening was associated to fruit softening, a change in pulp color from a pale pink to an intense pink color, and an increase in SSC. Fruit response to exogenous ethylene applications was small in immature fruits and increased as fruits approached the ripe stage, and decreased again in over-ripe fruits. In conclusion, postharvest applications of ethylene increased both fruit earliness and maturity uniformity in fruits.

Flowering and fruit set characteristics were examined in the popular commercial cultivar Magaoa in an effort to elucidate the reproductive phenology of mamey sapote, Calocarpum sapota (Jacq.) Merr. [syn. Pouteria sapota (Jacq.) H.E. Moore and Stearn]. Flowers opened during the night with anthesis beginning around sunset. The length of floral opening varied according to season, ranging from 6 days in winter to a single day in summer. Bursts of new flowers generally appeared in cycles of about 7 days in declining numbers of flowers per burst until all the floral buds of a particular floral bud flush had flowered. Floral buds flowered randomly along a branch with only a few flowers open at any one time. Flower position around the branch was a factor in fruit set. Flowers and small fruitlets encircled horizontal branches in great numbers, but immature fruit most often developed from flowers located on the upper branch quadrant. The lower quadrant contained the fewest immature fruit. As fruit matured, however, more upper quadrant fruit abscised until by harvest, most mature fruit were found on the lower quadrant. The observations provide new insights into the reproductive phenology of mamey sapote.

In this study, aqueous enzymatic extraction (AEE) was evaluated during the process of obtaining oil from mamey sapote seed (OMSS). Viscozyme L enzyme complex was used at pH 4 and 50 °C during the optimization of the extraction process by central composite design and response surface methodology. Optimal conditions were: 3.5% (w/w) of enzyme (regarding the seed weight), 5.5 h of incubation time, 235 rpm of agitation rate, and 1:3.5 of solid-to-liquid ratio. These conditions enabled us to obtain an OMSS yield of 66%. No statistically significant differences were found in the fatty acid profile and physicochemical properties, such as the acid and iodine values and the percentage of free fatty acids, between the oil obtained by AEE or by the conventional solvent extraction (SE). However, the oxidative stability of the oil obtained by AEE (11 h) was higher than that obtained by SE (9.33 h), therefore, AEE, in addition to being an environmentally friendly method, produces a superior quality oil in terms of oxidative stability. Finally, the high oil content in mamey sapote seed, and the high percentage of oleic acid (around 50% of the total fatty acid) found in this oil, make it a useful edible vegetable oil.

O objetivo desse estudo foi avaliar as características físico-química e perfil de açúcares da polpa de sapoti. A polpa de sapoti in natura foi caracterizada em triplicata quanto aos seguintes parâmetros físico-químicos: teor de água, atividade de água, pH, acidez total titulável, sólidos solúveis totais, ratio, cinzas, teor de ácido ascórbico, açúcares totais, redutores e açúcares não redutores. A determinação do perfil de açúcares foi realizada usando um Sistema de Cromatografia Líquida de Alta Eficiência (HPLC) e a concentração dos açúcares foi determinada a partir de curvas-padrão. O teor de água obtido foi de 76,21% e atividade de água de 0,991. O parâmetro ratio apresentou valor de 81,82 indicando um elevador grau de doçura. Os teores de açúcares totais, redutores e não-redutores foram de 12,38% de glicose, 10,81% de glicose e 1,57% de sacarose, respectivamente. O perfil de açúcares da polpa mostrou que a glicose apresentou maior teor (5,98 g/100g) seguido da frutose (4,85 g/100g) e da maltose (0,017 g/100g). Dessa forma, a polpa de sapoti pode ser utilizada como matéria-prima em formulações alimentícias doces em decorrência do perfil de açúcares, o que contribui para a valorização industrial do fruto.

O sapotizeiro (Achras sapota L.) é cultivado principalmente para a produção de frutos, encontrando-se entre os mais apreciados pela população das áreas onde cresce. Sua propagação pode dar-se através de enxertia ou diretamente por sementes, dando origem aos chamados pés-francos. Considerando o fato de que muitas das espécies florestais e frutíferas apresentam o fenômeno de dormência, e diante da necessidade de se intensificar estudos que melhor expliquem tal processo, o objetivo deste trabalho foi avaliar a eficiência de diferentes tratamentos pré-germinativos em sementes de sapoti, visando a acelerar e uniformizar a germinação de suas sementes. O ensaio foi conduzido em casa de vegetação do Departamento de Fitotecnia, CCA/UFPB, Areia-PB, sendo utilizados os seguintes tratamentos: imersão em água a 60°C por 1, 2 e 3 minutos; corte lateral (ao embrião) com e sem embebição em água por 24 horas; corte distal (ao embrião) com e sem embebição em água por 24 horas; corte lateral+distal (ao embrião) com e sem embebição em água por 24 horas e a testemunha (ausência de tratamento). Efetuados os tratamentos, foi realizada a semeadura em bandejas contendo areia lavada e autoclavada, utilizando-se de quatro repetições de 25 sementes. As características avaliadas foram: percentagem e índice de velocidade de emergência, comprimento de raiz e de hipocótilo e peso de matéria seca de planta. O delineamento experimental foi o Inteiramente Casualizado, e os contrastes entre as médias foram realizados através do teste de Duncan, ao nível de 5% de probabilidade. As sementes de sapoti exibiram maior emergência (81%) e índice de velocidade de emergência (0,58) quando submetidas ao corte lateral, sem embebição. O corte das sementes seguido de embebição por 24 horas não foi eficiente para acelerar a emergência e o índice de velocidade de emergência de plântulas de sapoti. A imersão em água a 60°C, independentemente do tempo utilizado, não deve ser recomendada como tratamento pré-germinativo para sementes de sapoti.

The comparison of SPME fiber coatings, and optimization of temperature and time of extraction for headspace solid phase microextraction (HS-SPME) of volatile organic compounds (VOCs) present in Pouteria sapota (sapote mamey) fruits is presented. The PDMS/DVB coating afforded the highest extraction efficiency. The extraction conditions were optimized by using Doehlert experimental design. By using the optimized HS-SPME method, 21 VOCs were identified, which include mainly terpenoids and esters, followed by aromatic hydrocarbons, aldehydes, alcohols and ketones. The most abundant compounds in Pouteria sapota pulp were cedrol (25.0%), azulene (7.3%), β-ionone (5.7%) naphthalene (5.6%), α-pinene (5.0%), and benzaldehyde (4.3%). Seventeen VOCs were identified for the first time in the fruit.

ABB Industries was told to implement activity-based costing (ABC) for costing in their consolidated reports sent to ABB Group, their parent firm. To meet this need, they chose to implement ABC using SAP's R/3. For two of nine product lines, parallel ABC was developed. Unfortunately, the version of ABC needed to generate financial statements, operational ABC, would not be available until a later release of the software. While ABB Industries waits for the next release, key personnel associated with the implementation are being offered alternative internal and external opportunities. Further, in order for ABB Industries to implement the operational ABC, the entire firm must implement a new version of SAP's R/3.

Los anfibios han diversificado predominantemente en ambientes tropicales donde la humedad, temperatura y disponibilidad de microhábitats facilitan la estabilidad demográfica. Sin embargo, múltiples linajes han colonizado desiertos extremos, donde su diversidad es considerablemente menor. Una especie en particular, el sapo de Atacama (Rhinella atacamensis), se ha adaptado al Desierto de Atacama, la región más árida en la Tierra. Pese a la singularidad ecológica de esta especie, la mayor parte de los aspectos de su historia natural, reproducción, patrones de actividad y conducta permanecen desconocidos. Usando cámaras trampa y observaciones de campo in situ, documentamos una serie de nuevos hallazgos en una población del Parque Nacional Llanos de Challe. Nuestro estudio muestra que R. atacamensis permanence oculto del sol en pequeñas pozas cubiertas por vegetación, donde el 100% de las hembras observadas mantiene un macho permanentemente abrazado en amplexo a todas horas del día. Los sapos emergen para conglomerarse en pequeñas pozas expuestas después de ponerse el sol (~20:00h), donde los machos se enfrentan en activos combates por hembras. Hasta cuatro machos se enfrentan por una misma hembra. Los sapos se vuelven a ocultar en las pozas cubiertas después de las ~06:00h. Los huevos depositados durante la noche eclosionan dentro de 24 horas. Discutimos estas observaciones en el contexto de la diversidad global de los sapos bufónidos en general, y en relación con los efectos que la selección que emerge de las duras condiciones de Atacama pueden ejercer sobre la expresión de estos caracteres.

En este estudio se caracterizaron genotipos de sapote mamey [Pouteria sapota (Jacquin) H.E. Moore & Stearn] de la región subtropical del Centro-Occidente de Michoacán, México, con base en parámetros morfológicos cualitativos y cuantitativos, que se analizaron mediante análisis de conglomerados y componentes canónicos. Se detectaron así seis grupos con 6, 5, 11, 13, 6 y 3 árboles de sapote mamey, respectivamente. Las variables peso, longitud y diámetro del fruto, espesor y peso del mesocarpio, la relación longitud y diámetro del fruto, forma, aroma y textura, resultaron ser las más importantes para diferenciar los grupos. La variable canónica CAN1 explicó 92 % de la variación acumulada entre los grupos evaluados. El peso del fruto, el espesor del mesocarpio y el peso del mesocarpio fueron las características morfológicas dominantes de la variable canónica CAN1 con coeficientes canónicos estandarizados de 4.09, -3.9 y 1.78, respectivamente; por tanto, serían las variables más importantes para usarse como criterio de selección de genotipos de mamey con frutos de uniforme y mayor calidad para consumo en fresco o productos procesados. Las pruebas de F y de χ2 confirmaron la variabilidad entre dichos grupos, aunque no se observó una relación entre el origen de los materiales y el grupo en el cual se clasifican. Estos resultados proveen el primer estudio sobre la diversidad morfológica de frutos de mamey y su potencial para producción en el estado de Michoacán, México.

Genotypes of sapote mamey, Pouteria sapota [(Jacquin) H.E. Moore & Stearn] from central-western Michoacán in Mexico were characterized based on physical and chemical fruit characteristics. Cluster analysis of data indicated seven distinct genotype clusters. The greatest variability among clusters was attributed to physical and chemical fruit characteristics. Those were fruit weight, length, width, shape and texture; the ratio of fruit weight to seed weight; seed weight and length; mesocarp thickness and weight; epicarp weight; and the compositional components titratable acidity (TA), protein, total soluble solids (TSS), TSS to TA ratio and TSS to pH ratio. Canonical discriminant analysis was a used to identify the most desirable sapote mamey fruits based on physical (fruit and mesocarp weight) and compositional parameters. Two canonical discriminant functions explained >90% of the accumulated variation a mong the seven clusters of genotypes. Fruit weight, mesocarp thickness, and the ratio of total soluble solids to titratable acidity were dominant in the first function; and fruit weight and mesocarp thickness were dominant in the second. These morphological variables could be used for selecting sapote mamey trees with uniform fruit quality for either direct consumption or processing.
