Uridylyl insertion/deletion mRNA editing is essential for mitochondrial gene expression in Trypanosoma brucei and governed by multi-protein complexes called editosomes. The final step in each cycle of this post-transcriptional process is that of re-ligating the edited mRNA fragments. The ~20S RNA editing core complex contains two RNA editing ligases, REL1 and REL2, located, respectively, in a deletion and an insertion subcomplex. While REL1 is clearly essential for RNA editing, REL2 knockdown by RNAi has not resulted in a detectable phenotype. To explain these findings, alternative scenarios have been suggested: (a) REL2 is not functional in vivo; (b) REL1 can function in both insertion and deletion editing, whereas REL2 can only function in insertion editing; (c) REL1 has an additional role in repairing erroneously cleaved mRNAs. To further investigate respective functions of the two RELs this study used three complimentary approaches: (i) genetic complementation with chimeric ligase enzymes, (ii) deep sequencing of RNA editing intermediates after ligase inactivation, and (iii) evolutionary analysis. In vivo expression of two chimeric ligases, providing a REL2 catalytic domain at REL1’s position in the deletion subcomplex and a REL1 catalytic domain at REL2’s position in the insertion subcomplex, did not rescue the growth defect caused by REL1 ablation. Although the results were not fully conclusive they suggest that it is the specific catalytic properties of REL1 rather than its position within the deletion subcomplex that makes it essential. In order to identify in vivo substrates of REL1, specific editing intermediates that accumulated after genetic knockdown of REL1 expression were captured by 5’ linker and deep sequenced using Ion Torrent and Illumina technology. Analyses of such unligated editing intermediates with bespoke bioinformatics tools suggest that REL1 functions in deletion editing as expected, but also in the repair of miscleaved mRNAs, implying a novel role for this ligase. Neither role can be fulfilled by REL2, at least not with sufficient efficiency. Sequencing data also suggest that either REL1 is not involved in ligation of addition editing substrates, or that REL2 in this case can fully compensate for loss of REL1. REL1, REL2 and KREPA3 sequences were subjected to analysis using MEGA5 and the HyPhy package available on the Datamonkey adaptive evolution server. Results indicated that all three editosome genes are under much stronger purifying than diversifying selective forces. In general this selection pressure to conserve protein sequence increased from KREPA3 to REL2 to REL1, suggesting a requirement to maintain catalytic function for both ligases. Taken together, these experiments reveal a novel function for REL1 during RNA editing, providing a rationale for its essentiality. Deductively, the results also suggest REL2, which was previously thought to be non-essential, may still be required by the cell at its position in the addition subcomplex. Evolutionary analysis suggests that the RELs and KREPA3 are under the same evolutionary forces to maintain their respective functions in RNA editing.

The quest for high-efficiency, high-power output fuel cells can be largely equated to the quest for high-performance components (anode, cathode, electrolyte). Solid Acid electrolytes, notably CsH2PO4, have been demonstrated to be affordable, stable, excellent ion conductors, and impermeable to parasitic fuel cross-over. Moreover, they operate at a temperature `sweet-spot' high enough to promote electrode kinetics and low enough to enable low-cost infrastructure. Fuel cell devices based on these materials are known to be limited, however, by electrode losses - even with high platinum loading. Improving the performance and lowering the cost of these components is necessary if such devices are to be considered viable alternatives.

In this work, the primary focus was the development of electrochemical characterization approaches. In doing so, we investigated electrode losses through steady-state and time-dependent electrochemical characterization and identified the primary rate limiting process and mechanism as oxygen reduction at the cathode. To characterize anode kinetics, new testing approaches were implemented which employed robust, asymmetric electrode geometries to isolate electrode kinetics without the inclusion of a reference electrode. These geometries' isolation efficacy was assessed by numerical computation - the results of which were leveraged into an explicitly defined, material-agnostic tool to evaluate asymmetric electrode geometries.

While for platinum, the cathode mechanism was shown to be insensitive to the microstructures tested, the mechanism of hydrogen reduction/oxidation was seen to vary between nanoscale powder and microscale defined electrodes - reconfirming the importance of rigorous testing approaches. Asymmetric electrode geometries with defined microstructures allowed direct characterization and comparative evaluation of non-platinum candidates for both the anode and the cathode. On the anode, palladium was over an order of magnitude more active than platinum and nickel, which exhibited a comparable activity. Palladium and silver were shown to be stable cathode materials, though less active than platinum. As a demonstration of the developed methodology's flexibility, a palladium-silver alloy was synthesized and tested.

The tools and methodologies developed in this work enable the rapid and flexible screening of electrodes for solid acid fuel cells.

Fractal grids generate multi-scale turbulent flows which interact with each other and form new multi-scales with different properties compared with those generated by classical grids. The goal of this work was to study the influence of geometry multiscale fractal on the generation and decay of turbulence by comparing the turbulence produced by a traditional fractal square grid FSG for short to that produced by a spaced fractal square grid SFSG for short with similar physical properties. The velocity measurements were performed in an open circuit suction type wind tunnel using a constant temperature hot wire anemometer at various positions in the x direction along the tunnel’s center line and for three different Reynolds numbers ReL0 based on the inlet velocity U? and the length of the largest grid bar L0. It was found that the turbulence intensity decays with distance from the grid for SFSG. Whereas, for FSG at low Reynolds numbers the turbulence intensity shows the same behaviour as for SFSG but for higher Reynolds numbers the turbulence intensity increases to reach a peak at a distance xpeak from the grid and then decays. In the inhomogeneous region, for both types of grids, a scaling range spans over a decade with an exponent near, but not exactly, k?5/3. In contrast, the length of the scaling range larger for FSG than for SFSG.
