We investigated whether the distribution pattern of soil nutrients interacted with aboveground competition to affect plant size in Ipomoea tricolor Cav. populations. Six plants per pot were grown in a factorial experiment with combinations of heterogeneous or homogeneous nutrient distribution pattern and presence or absence of aboveground competition. Plants were harvested and ranked by their aboveground biomasses. In analyses of plant sizes in all ranks simultaneously, mean plant size was significantly affected by nutrient distribution pattern, aboveground competition, and their interaction. In analyses of plant sizes of each rank, aboveground competition affected plant size, which was found in all ranks. Nutrient distribution pattern affected plant size in the higher ranks, but not in the lowest rank. Selective root placement into nutrient-rich patches under heterogeneous conditions was observed. Our results suggest that the magnitude of the effect of nutrient distribution pattern on plant size changed among ranks owing to changes in aboveground competition and size-dependent growth rate. Size-dependent growth rate could explain the significant effect of nutrient distribution pattern independent of the effect of aboveground competition. Nutrient distribution pattern would then interact with aboveground competition and consequently affect size structure in a population.

Agkistrodon in Chinese medicine has long been used as an effective treatment against rheumatoid arthritis (RA). The present research further investigated the effects of peptides extracted from the crude Agkistrodon on the RA rat model. Extracted peptides were separated by parameter-optimized ion-exchange chromatography (IEC), peptide fractions were further analysed by MALDI-TOF/TOF MS, and nano-LC-MS/MS acquired mass spectra were further characterized using Mascot software, which ranks the best matches in the NCBI database. RT-PCR results in RAW264.7 cells indicated that Agkistrodon peptide components had inhibitory effects against inflammatory cytokines. The therapeutic efficacy of Agkistrodon peptides was evaluated on the Wistar rats with collagen-induced arthritis. Symptom relief and reduced cartilage destruction and bone erosion were observed, which can be explained by the direct suppression of inflammatory cytokines in the joints. Agkistrodon peptides downregulate the expression of TNF-α, IL-1β, and IL-6, which may alleviate cartilage destruction and bone erosion, thus relieving symptoms of RA.

. In the Russian Federation, barley is widespread and cultivated in all soil and climate zones. The purpose of the research is to determine the adaptability of Omsk varieties of filmy and naked barley on the basis of “grain yield”. Methods. The research was conducted from 2015 to 2019 in the conditions of the southern forest-steppe of Omsk. A detailed analysis of the adaptivity parameters is given: the coefficient of the index of environmental conditions, plasticity and stability according; multiplicative coefficient according; the ecovalent of plasticity according; homeostaticity and the stability index; breeding value; genotypic effect, the rate of reaction of varieties to the environmental conditions. Final adaptability of varieties is estimated by the sum of ranks obtained each grade of the studied parameters. Results. The results of the research showed that the most adaptive in the conditions of the southern forest-steppe zone of the Omsk region are double-row filmy lines Nutans 4883, Nutans 4812 and variety Omskiy 101 (the sum of ranks = 34...38); multilayered membranous – Omskiy 99 and Nutans 4883 (sum of ranks = 54 and 56); two-row hulless hulless cultivar Omskiy golozernyy 1 (sum of ranks = 82); multi-row hulless varieties Omskiy golozernyy 2, Omskiy golozernyy 4 (sum of ranks = 86 and 84). Scientific novelty consists in the study of 8 varieties and 5 new promising lines of filmy and naked groups of barley, selection of the Omsk agricultural research center. The most adaptive varieties and lines for the conditions of the southern forest-steppe of Western Siberia are identified, which are recommended for introduction into production and for further breeding work.

Abstract Male dominance hierarchies occur in many group-living primates and some non-primate mammals. Variation in aspects of agonistic relationships such as how many dyads show bidirectionality in aggression leads to variation in dominance hierarchies along a continuum from egalitarian (relatively small agonistic power differences between adjacently-ranked individuals, shallow hierarchies) to despotic (relatively large differences, steep hierarchies). Ranks usually depend mostly or entirely on individual characteristics that influence fighting ability (e.g., body size) and show inverse-U shaped relationships to age. However, coalitionary support sometimes also influences ranks. Chimpanzees (Pan troglodytes) form multi-male, multi-female communities within which males compete for status. Males typically form dominance hierarchies, and data from multiple study show that rank is positively related to paternity success. Males also often form coalitions and some dyads form long-term alliances. Effective coalitionary support can help individuals improve and maintain their ranks, and some evidence supports the hypothesis that coalitionary aggression generally, and the positions that males hold in coalitions networks specifically, influences paternity success. Hierarchy steepness varies among communities and within communities over time; variation in the number of prime-aged males per community is a likely source of this variation. Long-term data from an extremely large chimpanzee community with unusually many males, at Ngogo, Kibale National Park, Uganda, are largely consistent with previous analyses of male chimpanzee dominance hierarchies, but show several notable contrasts. Males at Ngogo formed significantly linear hierarchies and hierarchy steepness was greater than expected if the outcomes of agonistic interactions were random. However, variation in steepness did not show the significant inverse relationship to the number of “prime-aged” males documented for other chimpanzee communities and average steepness was high given the large number of males. Ranks showed an inverse-U shaped relationship to age, although individual rank trajectories varied considerably, but males attained their highest lifetime ranks at later ages and maintained relatively high ranks to later ages than those at other chimpanzee research sites. Two measures of coalition networks, strength and Bonacich power, showed significant positive relationships with male ranks. Strength is the rate at which males joined coalitions. Bonacich power is a measure of network centrality that assesses a male’s relational power, or influence (Bonacich, 1987): a male with high Bonacich power formed coalitions with relatively many other males who were also central in the coalition network, i.e., he was strongly connected to powerful others. On average, males also attained maximum values for these and other network measures relatively late and maintained relatively high values to relatively late ages. High coalition network strength, Bonacich power, and eigenvector centrality early in adulthood were associated with high peak ranks at later ages. However, the direction of causality between participation in coalition networks and ranks is not yet clear, and the effects of body size on dominance ranks and individual rank trajectories remains to be explored. Ngogo is a favourable habitat for chimpanzees and survivorship there is unusually high; this presumably facilitates the ability of males to maintain high competitive ability longer than at other sites and shifts rank trajectories toward older ages and leads to relatively steep hierarchies despite the fact that many male dyads have similar competitive ability. Future work will assess the impact of coalitions on dominance relationships in more detail and the relationship of coalitionary aggression to paternity success.

Periodontitis is an infecto-inflammatory disease that leads to connective tissue and alveolar bone loss. Calendula officinalis (CLO) has been used due to its anti-inflammatory effects. Therefore the aim of this study was to evaluate the effect of CLO on alveolar bone loss (ABL) in rats focusing on RANK-RANKL-OPG and WNT signaling pathways. Experimental periodontitis (EP) was induced through placement of a nylon ligature around the upper left 2nd molar, and the hemimaxilla used as control. The animals were divided in groups: Normal, subjected to no treatment; Saline (SAL), that received 2 ml/kg of 0,9% saline solution orally; or CLO at 90 mg/kg orally, 30 minutes before EP and daily for 11 days until euthanasia. In order to evaluate the periodontal tissue, it macroscopic, micro-tomographic, electron scanning microscopy (SEM), confocal microscopy and polarized light microscopy analyses were performed, as well as immunohistochemistry for WNT 10b, β-catenin, DKK-1, RANK, RANKL, and OPG. During euthanasia the gingival tissue was removed for malonaldehyde (MDA) assay. Treatment with CLO significantly prevented ABL, preserved bone internal microstructure (p<0.05) and topography, and also preserved collagen fibers from the periodontal ligament, when compared to SAL. CLO significantly increased the number of immunopositive cells for WNT 10b, β-catenin and OPG and reduced DKK-1, RANK (p>0.05) and RANKL. CLO reduced the gingival levels of MDA compared to SAL (p<0.05). In this way, we can conclude that CLO prevented ABL via RANK-RANKL-OPG and WNT signaling pathway.
A periodontite à uma doenÃa infecto-inflamatÃria que causa perda de tecido conjuntivo e osso alveolar. A Calendula officinalis (CLO) tem sido utilizada pelos seus efeitos anti-inflamatÃrios. Nesse contexto, o objetivo deste trabalho foi avaliar efeito da CLO na perda Ãssea alveolar (POA) em ratos com foco na participaÃÃo do eixo RANK-RANKL-OPG e da via WNT/β-catenina. A periodontite experimental (PE) foi induzida atravÃs da inserÃÃo do fio (nailon 3.0) em torno do 2 molar superior esquerdo, e hemiarcada contralateral usada como controle. Os animais foram divididos em grupos: Normal, nÃo submetido a nenhum procedimento; Salina (SAL), que receberam 2 ml/kg de soluÃÃo salina 0,9% - v.o.; ou CLO na dose de 90 mg/kg - v.o. 30 min antes da PE e diariamente durante por 11 dias atà eutanÃsia. Para avaliaÃÃo do tecido periodontal realizaram-se anÃlises macroscÃpica, por microtomografia computadorizada, por microscopia eletrÃnica de varredura (MEV), microscopia confocal e microscopia por luz polarizada, imunohistoquÃmica para WNT 10b, β-catenina, DKK-1, RANK, RANKL e OPG. Por ocasiÃo da eutanÃsia foi removido tecido gengival para avaliaÃÃo dos nÃveis de malondialdeÃdo (MDA). O tratamento com CLO preveniu de forma significante a POA, preservou a microestrutura interna (p<0,05) e topografia do tecido Ãsseo, e preservou tambÃm as fibras colÃgenas do ligamento periodontal, quando comparado a SAL. A CLO provocou aumento significante de cÃlulas imunopositivas para WNT 10b, β-catenina e OPG e reduÃÃo na imunomarcaÃÃo de DKK-1, RANK (p>0,05) e RANKL. CLO reduziu os nÃveis de MDA gengivais comparados a SAL (p<0,05). Desta forma, podemos concluir que a CLO previne a POA com participaÃÃo do eixo RANK-RANKL-OPG e da via WNT/β-catenina.

Osteoporosis is a common human disease affecting both men and women, and it can classified as (1) primary, related to sexual hormones deficiency or senility, or (2) secondary, for what the most common example is the chronic therapies with glucocorticoids. Whatever is its cause, the osteoporosis outcomes are bone loss and increased fracture risk. Although osteoporosis is considered a systemic condition, bone loss seen in osteoporosis is not homogeneous throughout the skeleton. RANKL is a cytokine able to activate the osteoclasts function and its expression is inducible in osteoblasts and T cells by a range of stimuli. Function of RANKL has been considered a possible therapeutic target for the treatment of osteoporosis. To evaluate the trabecular bone loss related to the RANKL expression, immunochemistry and histomorphometric assays were used in femur and vertebra of castrated and/or glucocorticoid-treated male and female rats, at the day 56 after induction. RANKL expression was evident only in the castrated group, both male and female, but not in the group of castrated rats that also received glucocorticoid therapy. But histomorphometric data showed that bone loss was similar in both groups. That could happen because glucocorticoid can inhibit osteoblast metabolism. Histomorphometry also reveals that trabecular bone mass in male is similar to female, and bone loss is not homogenous between distal and proximal femur and vertebra body. At the day 56 after induction, bone loss, in femur and vertebra, both male and female, was compatible to osteoporosis.
A osteoporose é uma doença comum em humanos, acometendo tanto mulheres quanto homens. A doença pode ter origem primária, relacionada à deficiência de hormônios sexuais ou a senilidade, ou secundária, cujo exemplo mais comum é o uso crônico de glicocorticóides. Independente da causa, a conseqüência da osteoporose é a diminuição da massa óssea, aumentando o risco de fraturas. Apesar de ser considerada uma doença sistêmica, a redução de massa óssea na osteoporose não é uniforme no esqueleto. RANKL é uma citocina capaz de ativar a função osteoclástica e sua expressão é indutível em osteoblastos e linfócitos T. A função desta citocina tem sido considerada um possível alvo terapêutico no tratamento da osteoporose. Para avaliar a relação da perda óssea trabecular e a expressão de RANKL, foram realizados testes imunoistoquímicos e histomorfométricos em fêmur e vértebras de ratos castrados e/ou tratados com glicocorticóides, 56 dias após a indução. A expressão imunoistoquímica de RANKL pôde ser verificada nos animais castrados, tanto machos quanto fêmeas, mas não no grupo castrado que também recebeu glicocorticóide. Entretanto, a diminuição da massa óssea em ambos os grupos foi similar na avaliação histomorfométrica. Isso pode ocorrer por causa do efeito inibitório que os glicocorticóides têm sobre o metabolismo. As análises histomorfométricas revelaram ainda, que a massa óssea trabecular avaliada por este método é similar em machos e fêmeas, e que a perda óssea não é uniforme entre o colo femoral, o côndilo femoral e o corpo vertebral. Aos 56 dias de indução, o quadro de perda óssea instalado tanto em machos quanto em fêmeas, para todos os fragmentos ósseos analisados, é compatível com o quadro de osteoporose.

O sistema OPG, RANK e RANKL é uma das mais importantes descobertas da biologia óssea. Essas proteínas regulam as atividades celulares na remodelação do tecido ósseo e na literatura há diversas investigações nos tecidos dentários. No entanto, no reimplante dentário, ainda não foram encontrados relatos. Foi objetivo deste trabalho, avaliar a imunomarcação das proteínas OPG, RANK e RANKL em dentes reimplantados de rato. Um grupo controle foi formado com quatro ratos no qual o reimplante dentário não foi realizado. Vinte e quatro ratos (Rattus norvegicus, albinus) tiveram seu incisivo superior extraído e depois reimplantado formando os seguintes grupos: grupo I – reimplante imediato; grupo II - reimplante tardio sem tratamento e grupo III - reimplante tardio com tratamento endodôntico (ressecção da papila dentária e preenchimento do canal com pasta de hidróxido de cálcio) e tratamento da superfície radicular (raspagem mecânica do ligamento periodontal necrosado e imersão em solução de flúor fosfato acidulado de sódio a 2,5%). Ao final dos períodos experimentais (10 e 60 dias) os ratos foram eutanasiados. Foram obtidos cortes longitudinais parafinados com 6μm de espessura. Os cortes foram submetidos à reação imunoístoquímica mediante a utilização de anticorpos primários para OPG, RANK e RANKL. Os resultados mostraram que a imunomarcação de OPG e RANKL ocorreu em todos os grupos e períodos estudados, muito embora RANKL não tenha sido observada no grupo reimplante imediato aos 60 dias. RANK foi observada somente aos 10 dias de todos os grupos no qual o reimplante foi realizado. A análise qualitativa dos resultados demonstrou que o sistema OPG, RANK e RANKL apresentou marcação evidente no reimplante tardio, sugerindo a efetiva participação no início do processo de reabsorção radicular, uma vez que aos 60 dias a imunomarcação foi discreta.
The OPG, RANK and RANKL system is one of the most important discoveries in bone biology. These proteins are key regulators of bone remodeling and in the literature there are several studies of tooth resorption. However, in tooth replantation, reports have not been found. The purpose of this study was to determine the immunolabeling of OPG, RANK and RANKL in replanted teeth in rats, using immunohistochemistry methodology. A control group (no replanted teeth) was formed by four rats. Twenty-four rats (Rattus norvegicus, albinus) were submitted to the extraction of their upper right incisors. The replantation was performed according to the groups below: group I – immediate replantation; group II – delay replantation without treatment and group III – delay replantation with endodontic treatment (extirpation of papilla and the root canal filled with calcium hydroxide) and root surface treatment (periodontal ligament was removed with scalpel and teeth were immersed in 2% acidulated phosphate sodium fluoride). The animals were euthanized at the end of the experimental periods (10 and 60 days). Longitudinal 6μm slices embedded in paraffin were obtained. The slices were submitted to immunohistochemistry reaction by means of the primary antibodies for OPG, RANK and RANKL proteins. The results showed the expression of OPG in all groups and periods. RANKL expression was observed in all groups, except in the immediate replanted teeth group (60 days). RANK expression was observed only at 10 days in all groups which replantation was performed. The qualitative analysis of our findings indicated that the system OPG, RANK and RANKL presented strong expression in delayed replantation, suggesting effective participation at the beginning of root resorption, since at 60 days the immunostained cells were discreet.

Essas proteínas regulam as atividades celulares na remodelação do tecido ósseo e na literatura há diversas investigações nos tecidos dentários. No entanto, no reimplante dentário, ainda não foram encontrados relatos. Foi objetivo deste trabalho, avaliar a imunomarcação das proteínas OPG, RANK e RANKL em dentes reimplantados de rato. Um grupo controle foi formado com quatro ratos no qual o reimplante dentário não foi realizado. Vinte e quatro ratos (Rattus norvegicus, albinus) tiveram seu incisivo superior extraído e depois reimplantado formando os seguintes grupos: grupo I - reimplante imediato; grupo II - reimplante tardio sem tratamento e grupo III - reimplante tardio com tratamento endodôntico (ressecção da papila dentária e preenchimento do canal com pasta de hidróxido de cálcio) e tratamento da superfície radicular (raspagem mecânica do ligamento periodontal necrosado e imersão em solução de flúor fosfato acidulado de sódio a 2,5%). Ao final dos períodos experimentais (10 e 60 dias) os ratos foram eutanasiados. Foram obtidos cortes longitudinais parafinados com 6μm de espessura. Os cortes foram submetidos à reação imunoístoquímica mediante a utilização de anticorpos primários para OPG, RANK e RANKL. Os resultados mostraram que a imunomarcação de OPG e RANKL ocorreu em todos os grupos e períodos estudados, muito embora RANKL não tenha sido observada no grupo reimplante imediato aos 60 dias. RANK foi observada somente aos 10 dias de todos os grupos no qual o reimplante foi realizado. A análise qualitativa dos resultados demonstrou que o sistema OPG, RANK e RANKL apresentou marcação evidente no reimplante tardio, sugerindo a efetiva participação no início do processo de reabsorção radicular, uma vez que aos 60 dias a imunomarcação foi discreta.
Abstract: The OPG, RANK and RANKL system is one of the most important discoveries in bone biology. These proteins are key regulators of bone remodeling and in the literature there are several studies of tooth resorption. However, in tooth replantation, reports have not been found. The purpose of this study was to determine the immunolabeling of OPG, RANK and RANKL in replanted teeth in rats, using immunohistochemistry methodology. A control group (no replanted teeth) was formed by four rats. Twenty-four rats (Rattus norvegicus, albinus) were submitted to the extraction of their upper right incisors. The replantation was performed according to the groups below: group I - immediate replantation; group II - delay replantation without treatment and group III - delay replantation with endodontic treatment (extirpation of papilla and the root canal filled with calcium hydroxide) and root surface treatment (periodontal ligament was removed with scalpel and teeth were immersed in 2% acidulated phosphate sodium fluoride). The animals were euthanized at the end of the experimental periods (10 and 60 days). Longitudinal 6μm slices embedded in paraffin were obtained. The slices were submitted to immunohistochemistry reaction by means of the primary antibodies for OPG, RANK and RANKL proteins. The results showed the expression of OPG in all groups and periods. RANKL expression was observed in all groups, except in the immediate replanted teeth group (60 days). RANK expression was observed only at 10 days in all groups which replantation was performed. The qualitative analysis of our findings indicated that the system OPG, RANK and RANKL presented strong expression in delayed replantation, suggesting effective participation at the beginning of root resorption, since at 60 days the immunostained cells were discreet.

Os mecanismos envolvidos na modificação do reparo ósseo em diabéticos ainda permanece pouco elucidado. Assim, esse estudo investigou a expressão de reguladores do metabolismo ósseo receptor ativador de NFkB (RANK), RANK ligante (RANKL) e osteoprotegerina (OPG), através da imunohistoquímica e RT-PCR, em sítios de fratura óssea de ratos diabéticos. Foram realizadas fraturas ósseas fechadas em tíbias esquerdas de ratos controle e com diabetes induzido pelo aloxano. A análise histomorfométrica dos sítios de fratura após 7 dias revelaram que os ratos diabéticos (db) apresentaram menor formação óssea e de cartilagem em comparação com o grupo controle. Paralelamente, o número de células RANK e RANKL positivas foi reduzido no grupo diabético. Além disso, células OPG positivas apresentaram-se significantemente diminuídas no grupo diabético comparado ao grupo controle (p=0,05). Entretanto, a razão RANKL/OPG foi similar no grupo controle (0,074) e diabético (0,099) nesse período. Após 14 dias, o número de células RANKL e OPG positivas e a expressão de RNAm desses marcadores foi maior no grupo controle (p=0.008). Apesar de menores níveis, a razão RANKL/OPG no grupo diabético (1,29) foi maior do que no grupo controle (0,90), o que sugere o favorecimento dos mecanismos de reabsorção óssea. Os resultados obtidos demonstram a expressão de marcadores da atividade de formação/remodelação de tecidos duros em sítios de fratura. Modificações no balanço da expressão de RANKL/OPG pode contribuir para o retardo do reparo de fraturas associado ao estado diabético. ____________________________________________________________________________________ ABSTRACT
To clarify the mechanisms of altered bone reapair in diabetic state, we investigate the RANK, RANKL and OPG expression by immunohistochemistry and RT-PCR in the fracture sites of diabetic rats. A closed fracture was performed on the anatomical left tibia in rats either healthy or made diabetic by alloxan. Histomorphometric analysis of fracture site at 7 days after fracture revealed that diabetic rats (db) have significantly lesser bone and cartilage formation at fracture site in comparison with controls. Parallel with this, the number of RANK and RANKL positive cells were slighly decreased in db group. Furthermore, OPG+ cells were significantly lower in db than control (p=0.05). However, the RANKL/OPG ratio was similar in control (0.074) and db (0.099) at this time. At day 14, the numbers of RANKL and OPG positive cells and the mRNA expression for these markers were increased in control group (P=0.008). Despite lower levels, the RANKL/OPG ratio in db group (1.29) was greater than in controls (0.90) what suggets a favoring of pro-resorptive pathways. Our results demonstrate the expression of consistent markers of hard tissue formation/remodeling activities in sites of fractures. The imbalance of RANKL/OPG expression may contribute to the delay of fracture repair during diabetes course.

Dentre as proteínas ósseas identificadas e envolvidas na dinâmica óssea podemos destacar a OPG, a RANK e RANKL como marcadores de atividades celulares. O presente trabalho tem como objetivo identificar, nos diferentes períodos da cronologia do processo de reparo alveolar através de técnica imunoistoquímica, a presença das proteínas OPG, RANK e RANKL. Para tanto foram utilizados 60 ratos machos submetidos à exodontia do incisivo superior direito e perfundidos aos 14, 21 e 28 dias pós-operatórios. As hemi-maxilas contendo o alvéolo dental em reparação foram removidas, pósfixadas, descalcificadas em EDTA, crioprotegidas e obtidos cortes longitudinais com 14æm em criostato. Os cortes foram submetidos à reação imunoistoquímica mediante a utilização de anticorpos primários para OPG, RANK e RANKL, como amplificador foi utilizado o sistema avidina-biotina e a diaminobenzidina (DAB) como cromógeno. Os resultados mostram que qualitativamente ocorre um balanço na expressão das proteínas que caracterizam reabsorção e neoformação óssea nos diferentes períodos estudados, onde aos 14 e 21 dias ocorre maior expressão de RANK. Com relação às proteínas OPG e RANKL, observa-se que elas apresentam-se expressas nas células da linhagem osteoblástica de forma similar, sendo que 28 dias é o período de maior expressão destas proteínas.
In the bone healing dynamics, the resorption and neoformation processes are dependent. Proteins involved in the cellular differentiation determinate the activation or inhibition of the activities that regulate the gain or loss of bone mass. From all of the identified bone proteins, it may be distinguished the OPG, RANK and RANKL. The present study has the aim to identify, in the different periods of alveolar bone healing chronology, the expression of OPG, RANK and RANKL proteins using the immunohistochemistry methodology. To perform this study, 60 male rats had the right upper incisive extracted and they were perfused at 14, 21 and 28 pos-operative days. The hemimaxilla with the rat extraction socket was removed, pos fixed and decalcified in EDTA. Then, they were cryoprotected and longitudinal slices with 14 ìm thickness were obtained in cryostat. The slices were submitted to immunohistochemistry reaction and the primary antibodies used were against OPG, RANK and RANKL proteins. It was used the avidinbiotin system to amplify the sign and diaminobenzidine was the cromogen. The results show that there is a balance in the expression of the proteins, showing that there is an increase in the expression of RANK at 14 and 21 pos-operative periods. In relation to OPG and RANKL, these proteins presents a similar expression in all of the pos-extraction periods analysed in this study and at 28 days after the extraction there is the greater expression of both proteins.

O objetivo desse estudo foi investigar a express?o dos marcadores RANKL/RANK/OPG em dentes de ratos reimplantados, bem como observar a rela??o entre a express?o desses marcadores com o processo de reabsor??o dent?ria e ?ssea. Foram utilizados 30 incisivos superiores direitos de 30 ratos machos, adultos, da linhagem Wistar (Rattus norvegicus albinus). Os dentes foram avulsionados e divididos em dois grupos que permaneceram extra-alveolar em ar seco: G1 (n=15) - 5 minutos e G2 (n=15) - 60 minutos, e em seguida foram reimplantados e analisados nos intervalos de 1, 3 e 7 dias. Finalizado os per?odos experimentais, ocorreu a eutan?sia dos animais. Cortes longitudinais com 5?m de espessura foram obtidos e corados pela t?cnica H/E para a an?lise histol?gica e cortes com 3?m de espessura foram submetidos ? rea??o imuno-histoqu?mica mediante a utiliza??o de anticorpos prim?rios para ratos como OPG, RANK e RANKL. Os resultados demonstraram que o sistema RANK/RANKL/OPG participa ativamente, tanto do processo de reparo de dentes de ratos reimplantados, quanto das reabsor??es dent?rias e ?sseas; que RANKL apresentou maior imunomarca??o em ambos os grupos, participando em todas as fases da reabsor??o ?ssea e dent?ria; e o aumento da express?o de RANKL foi observada em ambos os grupos em todos os intervalos de tempo, comprovando que a resposta inflamat?ria no ligamento periodontal ocorre no in?cio do processo de reparo; e que a fraca express?o de RANK e OPG e o aumento da express?o de RANKL sugere uma diminui??o da reabsor??o ativa e aumento da reabsor??o reparada no osso e no cemento, sendo maior no osso.
The RANK / RANKL / OPG sy stem plays an important role in bone formation and resorption . This finding has been regarded as one of the m ost important advances in the understanding of bone biology with respect to osteoclastogenesis. The aim of this study was to investigate the expression of RANKL / RANK / OPG markers in reimplanted t eeth of rats, and to observe the relationship between the expression of these markers and to oth and bone resorption. Thirty male Wistar rats (Rattus norvegicus albinos) had their maxillary right incisors extrac ted , and were divided into 2 groups according to the period that the extracted teeth were kept in dry air before reimplantation : G1 (n = 15) - 5 minutes , and G2 (n = 15) - 60 minutes . After reimplantation, teeth were analyzed at intervals of 1, 3 and 7 da ys. After these experimental periods, the animals were euthanized. Longitudinal sections with 5?m thick were obtained and stained with Hematoxylin and Eosin for histological analysis , while 3?m thick sections were subjected to immunohistochemical analysis of OPG , RANK and RANKL. The results showed that the RANK / RANKL / OPG system actively participates in both the repair process, as well as tooth and bone resorption . Extr a - alveolar time of 60 minutes before replantation caused minor expressions of RANKL a nd OPG, not influencing the expression of RANK; RANKL immunostaining showed higher in both groups when compared to other biomarkers, participating in all phases of bone and tooth resorption; RANKL was associated to both osteoclastogenesis and c ell ular proliferation , and was expressed in both groups.

This thesis investigates the robustness of the Type I error rate of the t test, the t test on ranks (Mann-Whitney-Wilcoxon), the Welch test and the Welch test on ranks applied to reaction time populations with unequal variances. Reaction time is often encountered as a dependent variable in educational and behavioural research. Reaction time data are typically skewed and are commonly modelled on a family of distributions known as the ex-Gaussian. A Monte Carlo study compared the robustness of type I error rates of the four tests under study under 36 conditions wherein four factors were observed: total sample size ($N=24$ and 72), ratio of sample sizes ($n\sb1:n\sb2$ = 1:1, 1:2, and 1:3); ratio of population variances (var1:var2 = 1:1, 1:2, 1:4, and 1:9), and negative and positive conditions. In each condition, 5,000 scores were generated from Miller's (1988) most skewed distribution that represented a boundary condition of reaction time data. The results indicated that the t test was the preferred option under all simulated conditions, except the negative condition. Furthermore, under the negative condition, all four tests produced liberal Type I errors.

We introduce the notion of diagonal ranks of semigroups,which are numerical characteristics of semigroups. Some base propertiesof diagonal ranks are obtained. A new criterion for a monoidbeing a group is obtained using diagonal ranks.For some semigroup classes we investigate whether their diagonal acts are finitely generatedor not. For the semigroups of full transformations, partial transformations andbinary relations we find the general form of the generating pairs.

Abstract A vast staff served the asylums: doctors, nurses, attendants, artisans, clergy, kitchen and laundry workers, and other who maintained buildings, farm, gardens and cemetery. Ward work was particularly demanding, with long hours, and poor conditions of employment. A regimented and punitive culture and distrust between management and lower ranks of staff, contributed to “a general feeling of insecurity” among them. They had high rates of sickness and a high turnover, sometimes over 75 per cent annually, resulting in an inexperienced workforce. The asylum leadership contributed to creating a dysfunctional system in which the patients, whom the asylum was meant to serve, were far from central to it.

Abstract Internal Corrosion (IC) is a time dependent threat to pipelines that leads to wall loss due to a reaction between pipe material and the products or contaminants being transported. Certain vintage pipelines are not piggable, meaning that Inline Inspection measurement tools cannot be used to measure IC defects. Furthermore, being older, these pipelines are more susceptible to time dependent threats such as IC due to their longer exposure time. Operators often need to prioritize retrofits between different non-piggable pipelines, and the approach described in this study uses a structural reliability approach to estimate the expected frequency of failure for non-piggable pipeline segments and ranks them accordingly. This model captures the range of potential IC defect severity by using in-line inspection data from different pipeline asset types (e.g., Well Laterals, Transmission) to build empirical distributions of defect length and depth corrosion growth rate (CGR). These asset types serve as an indirect measure of gas quality as evidenced by differences in the defect distributions and anomaly densities. By characterizing CGR as a distribution for different asset types and using the industry standard Growth-by-Rule method to model defect depth, the model considers both asset type and pipe age when predicting the defect depth distribution in the current year. This projected depth distribution is used with physical and operating parameters (e.g., diameter, WT, pressure) in a structural reliability model to estimate the probability of failure for the segment under consideration. The structural reliability-based prioritization approach described in this paper provides a methodology to utilize physical and operating parameters of an un-piggable pipeline together with information from inspected pipelines to rank the expected severity of a given un-piggable line. These segment ranks were finally compared to ranks generated using other heuristic methods to quantify the benefits of this more detailed analysis methodology.

The objectives of the study are to quantify the reliability of a boil-off gas (BOG) reliquefaction system for liquefied natural gas (LNG) carriers and to verify design improvements based on the reliability analysis. The system is broken into subsystems and then further into components. Failure rates are collected from generic references, primarily from the OREDA handbook. The reliabilities of the subsystems are estimated, and a reliability block diagram for the whole system is established. The subsystems are classified into three ranks according to their reliability: the high reliability rank comprises the subsystems ‘BOG preparation system,’ ‘Seawater intake system,’ and ‘Buffer N2 reservoir system’, the medium reliability rank the subsystem ‘BOG liquefaction system’, and the low reliability rank the subsystems ‘BOG compression system,’ ‘N2 cooling system’, and ‘Buffer N2 generation system.’ The reliability and availability are estimated for various process configurations where some of the low reliability category have a standby. The ‘bare’ system without any redundancy fails to attain an availability higher than 0.96. Addition of redundancy to one of the least reliable three commonly results in an increase in reliability improvement, to around 0.97. If all of the three subsystems have standby units, the system reliability improves to 0.99. It is recommended that maintenance efforts should be concentrated on the rotating machines that caused the subsystems to have a low reliability.

The world’s first 1000MW double reheat ultrasupercritical unit has been in operation since September 25th, 2015 in Taizhou, China. The thermal efficiency at turbine heat-rate acceptance (THA) condition is around 51%, which is the highest among all condensing units in coal-fired power plants around the world. However, the resultant superheat degree of the extraction steam is relatively high, leading to a large temperature difference in heat transfer process in the regenerative system, thereby a great exergy loss. In order to utilize the superheat of turbine bleeds more effectively, we present a scheme by employing an outer steam cooler (OSC) after the last high pressure heater in series to use the superheat to heat the feed water. Based on the newly installed unit in Taizhou, we examine the energy saving effect of the superheat utilization of different bleeds and their possible combinations respectively. The influencing factors of the mass flow rate, superheat, and effective superheat of the extraction steam are studied. Thermodynamic analyses revealed that the second extraction steam has not only high effective superheat, but also large mass flow rate, so in the overall efficiency improvement it ranks first and the third extraction steam ranks second. Although the fourth extraction steam has the largest superheat, it ranks third as the result of relatively lower mass flow rate. It was found that at nominal load, by adopting OSC’s to utilize the superheat of the second to sixth extraction steam, temperature of the feed water can increase by 8.1 °C, 3.5 °C, 2.6 °C, 1.1 °C, and 1 °C respectively, and the net coal consumption reduces by 0.73g/kWh, 0.47g/kWh, 0.40g/kWh, 0.21g/kWh and 0.22g/kWh accordingly. Consequently, three possible schemes are recommended for future design: one is to adopt one OSC to utilize the superheat of the second extraction steam, in return of 8.1°C increment in feed water temperature and 0.73g/kWh reduction of the net coal consumption; the second is to adopt two OSC’s to utilize the superheat of the second and third extraction steam at the same time, in return of 11.4 °C increment in feed water temperature and 1.21g/kWh reduction of the net coal consumption; and the last is to apply three OSC’s to utilize the superheat of the second to the fourth extraction steam simultaneously, to achieve 13.9°C increment in feed water temperature, and 1.62g/kWh reduction of the net coal consumption.

This paper selects 21 cities and prefectures of Sichuan Province as the research object, adopts 12 comprehensive indicators, establishes a linear model through correlation analysis, carries out regression analysis to modify the model, and uses factor analysis and cluster analysis to study the level of urban economic development. The results show that the GDP of all regions in Sichuan Province has a strong correlation with the three indicators of total retail sales of social consumer goods, passenger volume, and urbanization rate, and plays a positive role in promoting the economic development of all cities in Sichuan Province. Chengdu ranks the highest in the comprehensive ranking of urban economic development level in Sichuan Province.

An accelerated deposition test facility was operated with three different coal ash species to study the effect of ash composition on deposition rate and spatial distribution. The facility seeds a combusting (natural gas) flow with 10–20 micron mass mean diameter coal ash particulate. The particulate-laden combustor exhaust is accelerated through a rectangular-to-annular transition duct and expands to ambient pressure through a nozzle guide vane annular sector. For the present study, the annular cascade consisted of two CFM56 aero-engine vane doublets; comprising three full passages and two half passages of flow. The inlet Mach number (0.1) and gas temperature (1100°C) are representative of operating turbines. Ash samples were tested from the three major coal ranks: lignite, subbituminous, and bituminous. Investigations over a range of inlet gas temperatures from 900°C to 1120°C showed that deposition increased with temperature, though the threshold for deposition varied with ash type. Deposition levels varied with coal rank, with lignite producing the largest deposits at the lowest temperature. Regions of heightened deposition were noted; the leading edge and pressure surface being particularly implicated. Scanning electron microscopy was used to identify deposit structure. For a limited subset of tests, film cooling was employed at nominal design operating conditions but provided minimal protection in cases of severe deposition.

There is substantial disagreement over the order of the reaction between a coal char and O2. This is chiefly because of the difficulties of making good measurements when burning is kinetically controlled; thus heat and mass transfer can often obscure the true kinetics of the reaction. Consequently, a fluidized bed is an attractive reactor in which to measure the rate of this reaction. Chars from three different coals (of high, medium and low ranks) were prepared. The rate of oxidation of a char was measured by adding a small mass (∼ 3 mg) of tiny particles (sieved to 106–150 μm) of the char to a fluidized bed of silica sand (90–126 μm), which was electrically heated to a fixed temperature between 700 and 950°C. The fluidizing gas was a mixture of O2 and N2 at 1 bar; its composition varied between 0 and 100% O2. The concentrations of CO and CO2 were continuously measured in the off-gases from the bed, using i.r. analysers, thus enabling the rate of oxidation of the added batch of char to be measured. Interestingly, it was found necessary to modify the quartz cylinder housing the fluidized bed to prevent backmixing of ambient air into the freeboard. The char particles were sufficiently small that mass transfer to their exteriors did not control the rate of reaction. Furthermore, the rapid rates of heat transfer from the bed to the particles of char ensured that the char burned at the temperature of the bed. Likewise, the mass of added char was small enough for the rate of burning not to be controlled by inter-phase mass transfer. The maximum rate of oxidation was taken to be the initial rate, which could, at least in principle, be correlated with the initial properties of a char, such as its BET area, pore diameters, etc. The char of the low-rank coal burned with kinetics which were zeroth order in O2 and with a negligible activation energy. The other two coals had orders of reaction which were 0.7–0.8 at 700°C, but 0.2–0.25 at 950°C. These orders are discussed in detail.

One of the requirements of a comprehensive pipeline Integrity Management Plan (IMP) is the establishment of safe and cost effective re-assessment intervals for the chosen assessment method, either Direct Assessment (DA), In-Line Inspection (ILI) or hydrotesting. For pipelines where the major threat is external or internal corrosion, the determination of an appropriate re-inspection interval requires the estimation of realistic corrosion growth rates. The Office of Pipeline Safety (OPS 2005) estimate that the ability to accurately estimate corrosion rates may save pipeline companies more than $100M/year through reduced maintenance and accident avoidance costs. Unlike internal corrosion, which occurs in a closed system, the rate of the external corrosion reaction is influenced by a number of factors including the water content of the soil, the soluble salts present, the pH of the corrosion environment and the degree of oxygenation. Therefore the prediction of external rates is complex and there is currently no method for estimating corrosion rates using either empirical or mechanistic equations. This paper describes a scoring model that has been developed to estimate external corrosion growth rates for pipelines where rates cannot be estimated using more conventional methods i.e., from repeat in-line inspection data. The model considers the effect of the different variables that contribute to external corrosion and ranks them according to their effect on corrosion growth rate to produce a corrosion rate score. The resulting score is then linked to a corrosion rate database to obtain an estimated corrosion rate. The methodology has been validated by linking the calculated corrosion rate scores to known corrosion rate distributions that have been measured by comparison of the results from multiple in-line inspection runs. The paper goes on to illustrate how the estimated corrosion rates can be used for the establishment of reassessment intervals for DA, ILI and hydrotesting, comparing the benefits of this approach with current industry recommended practice and guidance.
