Effects of acid rain on the dry weight, nitrogen and sulfur content of radish plants were determined. They were exposed to distilled water acidified with a 7:3 ratio of sulfuric acid and nitric acid and adjusted to pH levels 2.5, 3.0, 3.5, 4.0, 4.5, 5.0, and 5.7. A control group, exposed to distilled water only, was also used. Four hundred plants, divided in eight groups, were grown under laboratory conditions. At four, seven and ten week intervals, an equal number of plants of each pH level were removed from the soil.Plants exposed to water of lower pH values were found to be significantly higher (p < .05) in dry weight, percentage of nitrogen, and weight of sulfur per plant than crops at more neutral pH levels. The increased weight may be due to the greater availability of nitrates and sulfates at lower pH levels. These compounds probably contributed to better growth and greater dry weight of the plants at low pH levels.

[Truncated abstract] Wild radish (Raphanus raphanistrum L.), an outcrossing annual plant, is one of the most widespread and successful colonising weeds in the Australian wheat belt. It was introduced accidentally during the latter part of the 19th century, apparently independently through the major ports of southern Australia. The widespread success of wild radish, and therefore the likelihood of distinct populations, gives us the opportunity to study the colonizing process with adaptation in annual outbreeding species, and to explore their genetic diversity. The aims of this thesis were to study the genetic diversity of wild radish and to investigate which factors are important in its success. After an initial review of the literature (chapter 2), the thesis describes experiments where genetic variation between and within populations was compared at 55 sites in transects across the wheat belt and high rainfall zones of temperate Western Australia (chapter 3). In chapter 4, variation in life history traits was compared with variation in AFLP molecular markers. The role of seed dormancy in the survival of the species was examined in chapter 5, and variation in the oil content of seeds and their fatty acid composition was examined in chapter 6. Finally, the results were discussed in chapter 7 with special reference to the adaptive value of outcrossing in annual weeds. The results suggest that wild radish has evolved to fit the Australian environment. However, measurement of 14 morphological and phenological characters showed that in most cases within site variation was much greater than that between sites. Most of the variation between sites was associated with geoclusters, a name given to zones of similar environmental conditions in regard to rainfall and temperature. Thus plants from areas with high rainfall and low temperature produced longer, wider pods with more segments, heavier seeds and flowered later than plants from more arid areas.

Cytoplasmic male sterility (CMS) is a widespread, maternally inherited trait that results in an inability of plants to produce functional pollen. The Ogura CMS system originated in radish, but has since been transferred to, and confers male sterility on, plants in the related genus Brassica . A gene which restores male fertility is needed for the Ogura CMS system to be exploited commercially for hybrid seed production in oilseed species such as Brassica napus. The restorer gene Rfo is a dominant radish nuclear gene that restores the male fertility to plants with Ogura cytoplasm. This gene has been transferred into Brassica napus through intergeneric crosses; however the introgressed segment of radish DNA contains an unknown number of genes, some of which confer undesirable traits, such as an elevated content of seed glucosinolates, antinutritive compounds that render the seed meal unusable as animal feed. A fine scale linkage map of the region in radish containing Rfo was constructed, and a map-based cloning approach relying on synteny between radish and Arabidopsis was used to clone Rfo. A radish gene encoding a 687 amino acid protein with a predicted mitochondrial targeting presequence was found to confer male fertility upon transformation into Ogura CMS B. napus . This gene, codes for a pentatricopeptide repeat (PPR)-containing protein with multiple, in this case 16, PPR domains. Two similar genes that do not appear to function as Rfo flank this gene. A transcript representing a non-functional allele (rfo) was detected in sterile radish plants. Comparison of the Rfo region with the syntenic Arabidopsis region indicates that a PPR gene is not present at the Rfo-equivalent site in Arabidopsis , although a smaller and related PPR gene is found about 40 kb from this site.

Radish plants (Raphanus sativus L. cv Cherry Belle) were exposed to 0.10 μl l⁻¹ ozone (O₃) or 0.50 μl l⁻¹ sulfur dioxide (SO2) for 4 h d⁻¹, 3 d wk⁻¹ for 3 weeks. From these fumigated plants, individuals were selected that were resistant or sensitive to these pollutants. The selected plants were used as parental material in a breeding program to produce lines differing in resistance to O₃ and SO₂. Non-selected (NS) plants from the original populations served as controls. F₁ populations were raised and exposed to O₃ or SO₂ with the same fumigation regime used for the parents. The plants were harvested 30 days after emergence and dry weights were determined. Plants selected for O₃ resistance (O3R) weighed significantly more than either plants selected for sensitivity to O₃ (O₃S) or NS plants when exposed to either O₃ or SO₂. The hypocotyl was most affected by pollutant exposure, leading to reduced root/shoot ratios. Plants selected for resistance or sensitivity to SO₂ generally had biomass production similar to that of NS plants. Growth analysis at early stages of growth indicated that both O₃R and O₃S plants had less growth under O₃ fumigated conditions; however, by maturity O₃R plants had similar amounts of growth under fumigated or non-fumigated conditions. Ozone fumigations tended to decrease free sugar concentrations in leaves at early stages of growth in both O₃R and O₃S plants, and caused some accumulations of carbohydrates during late stages of growth in O₃S plants. Allocation of Mc was significantly lower to hypocotyls and roots of O₃ fumigated O₃S plants. Allocation to hypocotyls of O₃R plants was not affected, although both O₃R and O₃S plant groups had lower photosynthetic rates due to O₃ fumigation. Ozone did not significantly affect chlorophyll concentrations in leaves of either sensitive or resistant plants, nor was the time of new leaf production affected by fumigation. These experiments demonstrated the potential of O₃ to influence the composition of sensitive plant populations. However, SO₂ was a much less powerful influence on the composition of these populations.

The most apparent symptom of boron deficiency in higher plants is a cessation of growth. Deficiency causes a reduction in ascorbate concentration and the absorption of nutrient ions. Addition of ascorbate temporarily relieves deficiency symptoms. In boron sufficient plants the addition of ascorbate to media causes an increased uptake of nutrients. In an attempt to discover if ascorbate addition to deficient plants causes increased ion uptake, radish plants were grown hydroponically in four different strengths of boron solution. A colorimetric assay for phosphorus was performed both before and after supplementation. Results, however, were inconclusive.

Cattle are commonly treated with antibiotics that may survive digestion and promote antibiotic resistance when manure or composted manure is used as a soil amendment for crop production. This study was conducted to determine the effects of antibiotic administration and soil amendment practices on microbial diversity and antibiotic resistance of bacteria recovered from the surfaces of lettuce and radishes grown using recommended application rates. Vegetables were planted in field plots amended with raw manure from antibiotic-treated dairy cows, composted-manure from cows with different histories of antibiotic administration, or a chemical fertilizer control (12 plots, n=3). Culture-based methods, 16SrDNA amplicon sequencing, qPCR and shot-gun metagenomics were utilized to profile bacteria and characterize the different gene markers for antibiotic resistance. Culture-based methodologies revealed that lettuce grown in soils amended with BSAs had significantly larger clindamycin resistant populations compared to control conditions. Growth in BSAs was associated with significant changes to the bacterial community composition of radish and lettuce. Total sul1 copies were 160X more abundant on lettuce grown in manure and total tet(W) copies were 30X more abundant on radishes grown in manure. Analysis of shotgun metagenomic data revealed that lettuce grown in manure-amended soils possessed resistance genes for three additional antibiotic classes compared to other treatments. This study demonstrates that raw, antibiotic-exposed manure may alter microbiota and the antibiotic resistance genes present on vegetables. Proper composting of BSAs as recommended by the U.S. Department of Agriculture and Environmental Protection Agency is recommended to mitigate the spread of resistance to vegetable surfaces.

The objective of this Ph.D. research was to identify new and novel mechanisms of wild radish (Raphanus raphanistrum L.) resistance to photosystem II (PSII) inhibitors, auxinics, and acetohydroxyacid synthase (AHAS) inhibitors. PSIIinhibitor resistance was demonstrated to be target-site based, and conferred by a Ser264 to Gly substitution of the D1 protein. Auxinic resistance was associated with reduced herbicide translocation to the meristematic regions of resistant wild radish plants. Two new resistance mutations of wild radish AHAS were discovered, including one encoding the globally rare Asp376 to Glu substitution, and another encoding an Ala122 to Tyr substitution, which has never been identified or assessed for resistance in plants previously. Characterization of the frequency and distribution of AHAS resistance mutations in wild radish from the WA wheatbelt revealed that Glu376 was widespread, and that some mutations of AHAS are more common than others. Computer simulation was used to examine the molecular basis of resistance-endowing AHAS target-site mutations. Furthermore, through the computer-aided analysis, residues were identified with the potential to confer resistance upon substitution, but which have not previously been assessed for this possibility. Results from this Ph.D. research demonstrate that diverse, unrelated mechanisms of resistance to PSII inhibitors, auxinics, and AHAS inhibitors have evolved in wild radish of the WA wheatbelt, and that these mechanisms have accumulated in some populations.

Acidification of low acid vegetables reduces their pH level to below 4.5 thereby facilitating their processing below 100 degrees Celsius, thus reducing thermal damage, and yielding better product quality. We sought to determine the acidification kinetics of turnip and radish in acetic or citric acid solutions, specifically the time for the treated tissue to reach pH 4.5, and the concurrent percent acidity of the tissue. The two vegetables' response to acidification with citric acid were compared. Experiments followed a central composite rotatable design, with three process variables (temperature, acid concentration and sample: solution ratio), each at five levels. All process variables showed significant effects (P ≤ 0.05) on both time to pH 4.5 and concurrent tissue acidity. Higher acid concentration, temperature or solution: sample ratio resulted in a shorter response time, and higher sample acidity. Optimum processing conditions (shortest time to reach pH 4.5) and the corresponding acidity were determined using a response surface methodology. This acidification process was successfully completed in practice.

Raphanus raphanistrum L. (wild radish) is a widespread and economically problematic weed of winter crops including wheat (Triticum aestivum L.) in southern Australia. Management of the weed is achieved by a range of non-chemical and chemical techniques. Current management is failing to contain the weed, which is increasing in abundance and distribution. The characteristics of staggered recruitment, dormancy, rapid growth, competitive ability and high levels of seed production contribute to its persistence. The literature review concludes that weeds formed the basis for perpetuation of R. raphanistrum, a claim further investigated by simulation modeling. The effective management of R. raphanistrum should aim to achieve declining populations and this requires the integration of all factors that regulate seed production. Reducing seed production of R. raphanistrum in wheat can be achieved by applying herbicides at reproductive stages of development. This technique has been demonstrated to be an effective, safe and practical method of reducing seed production of R. raphanistrum in wheat. Reducing seed production of R. raphanistrum by herbicides applied at reproductive stages of development is a technique that can be incorporated into an integrated weed management program to improve control

Resumo: Normalmente a horticultura utiliza de irrigação artificial para suprir as demandas hídricas das plantas, mas os produtores, em geral, não são criteriosos na utilização das lâminas de reposição, o que acarreta desperdício de água e energia elétrica aumentando os custos de produção. Outro fato é a crescente quantidade de pesquisas sobre a utilização de água tratada magneticamente (ATM) empregada nas atividades agrícolas. Estas pesquisas têm mostrado aprimoramentos produtivos para diversas culturas. Este trabalho apresenta um Sistema Baseado em Regras Fuzzy (SBRF) para avaliação da cultura do rabanete para a lâmina de irrigação a 100% da evapotranspiração. Foram utilizados para o SBRF dados experimentais obtidos por experimento que avaliou a influência da água tratada magneticamente no desenvolvimento de rabanetes. O experimento foi realizado durante os meses de setembro a novembro de 2013, em uma casa de vegetação nas dependências do Departamento de Engenharia Rural da UNESP, Faculdade Ciências Agronômicas, Fazenda Experimental Lageado, localizada no município de Botucatu, São Paulo. Para a elaboração do SBRF, definiu-se um processador de entrada (fuzzificador), um conjunto de regras linguísticas, um método de inferência fuzzy e um processador de saída (defuzzificador), gerando um número real como saída. Abstract: Normally the horticulture uses artificial irrigation to meet the water demands of the plants, but the producers, in General, are not careful in the use of replacement blades, which carries waste of water and electricity by increasing the production costs. Another fact is the growing amount of research on the use of magnetically treated water (MTW) employed in agricultural activities. These researches have shown production enhancements to diverse cultures. This paper presents a Fuzzy rule-based System (FRBS) for evaluation of the culture of radish in two production cycles for the blade of the 100% irrigation of evapotranspiration. Were used for the FRBS the experimental data obtained by experiment that evaluated the influence of magnetically treated water in the development of radishes. The experiment was conducted during the months of September to November 2013, in a greenhouse on the premises of the Department of Rural Engineering of UNESP, Agronomic Sciences College, the Experimental Farm Lageado, located in the city of Botucatu, São Paulo. For FRBS, defined an input processor (fuzzificator), a set of linguistic rules, a fuzzy inference method and an output processor (defuzzificator), generating a real number as output. O trabalho foi conduzido na UNESP, Jaboticabal-SP, no período de setembro de 2003 a janeiro de 2004, com objetivo de avaliar a produtividade e rentabilidade das culturas de pimentão, repolho, rúcula, alface e rabanete em cultivo consorciado. O experimento constou de 15 tratamentos, correspondentes a 10 cultivos consorciados e cinco monocultivos, instalado sob delineamento experimental de blocos casualizados, com seis repetições. Foram utilizados os híbridos Magali R e Kenzan, respectivamente, para pimentão e repolho, e as cultivares Vera, Cultivada e Crimson Gigante, respectivamente, para alface, rúcula e rabanete. A determinação do custo de produção do cultivo consorciado foi realizada com base na metodologia do custo operacional de produção, no mês de agosto de 2003. As produtividades de pimentão e repolho em cultivo consorciado não diferiram significativamente das obtidas em monocultivo. Maior massa fresca de parte aérea de plantas de alface (438,86 g/planta) foi observada quando a alface foi consorciada com pimentão, diferindo estatisticamente dos demais cultivos que obtiveram em média 323,05 g/planta de alface. Aumento na produtividade de raízes comerciais de rabanete foi observada quando o rabanete foi consorciado com pimentão, porém, foi significativamente superior apenas ao cultivo consorciado de pimentão+repolho+rabanete. A massa fresca da parte aérea da rúcula mostrou diferença apenas entre os consórcios de pimentão+rúcula+alface e pimentão+repolho+rúcula. Exceto o consórcio pimentão+repolho que obteve índice de uso eficiente da terra (UET) de 1,93; todos os outros consórcios apresentaram UET superior a 2, com maior UET (2,64) obtido no consórcio pimentão+alface. From yeast to humans, the fusion of vesicles with target membranes is driven by the formation of a parallel four-helix bundle of SNARE proteins that are present on both the vesicular (v-SNAREs) and target plasma membranes (t-SNAREs). The full zippering of this bundle is thought to provide the driving force for membrane fusion. At synapses, vesicle fusion is exquisitely regulated by Ca2+ such that neurotransmitter release can occur within 1 ms of an action potential reaching the presynaptic terminal. This feat implies the presence of both a Ca2+ sensor and a fusion clamp that prevents vesicles from fusing in the absence of Ca2+. The presynaptic Ca2+ sensor for synchronous vesicle release is widely accepted to be Synaptotagmin-1 (Syt1), and there is growing evidence that Complexin (Cpx), which binds to the SNARE complex with high affinity and 1:1 stoichiometry, can act as a vesicle fusion clamp. As suggested by its name, however, Cpx appears to play a more complex role in vesicle release, carrying out different functions in spontaneous vs. evoked fusion events. Here we show the Drosophila express at least two Cpx isoforms that differ in the C-terminus (Cpx7A and Cpx7B) and can be further regulated by RNA editing and phosphorylation. These isoforms show different effects on spontaneous vs. evoked neurotransmitter release, with Cpx7A being a better fusion clamp and Cpx7B being a better fusion promoter. In addition, these isoforms have different effects on synaptic growth, which may be linked to their effects on synaptic physiology.

Chapter 2: Germination and Growth of Radish (Raphanus sativus) and Lettuce (Lactuca sativus) Exposed to Arsenite and Arsenate in Hydroponic Growth Solution Little information is available on the survival, uptake, and dry mass production of vegetable seedlings and maturing plants in arsenic enriched environments. Such information is however very important to many vegetable growers in areas of subsistent agricultural like Bangladesh or home-gardeners in closer proximity of As sources such as metal smelters. Accordingly we conducted research investigating (i) the germination and radical formation of radish and lettuce seeds at varying As (V) and As (III) concentrations and (ii) radish and lettuce plants in solution culture. Seed germination studies demonstrated that 0.1mM and 0.025mM are toxic threshold levels of As (III and V) for radishes and lettuce, respectively, while As (V) is more toxic to radish seeds than As (III). Arsenic (III and V) impacted both germination and radical development in radish seeds. For lettuce we observed that As had no impact on germination but reduced radical length significantly (p < 0.01). At most equimolar concentrations, As (III) was more toxic than As (V) in lettuce seeds (0.025 - 0.10mM As), a result contrary to those obtained in radish seeds (0.05 - 0.5mM As). The hydroponic growth studies showed that losses and increases in dry weight are a function of absorbed As and are dependent on the source of As: As (V) or As (III). Moreover, the effect of absorbed As (V) or As (III) on dry weight reductions and increases differed between root and shoot portions of the plants and are crop dependent. Tissue-As (originally solution As (V)) was more toxic at the radish root level and tissue-As (originally solution As (III)) was more toxic at the radish shoot level. Conversely for lettuce, As (III) caused reductions in dry weight, while As (V) had a stimulating effect on biomass production. Lower As (V) concentrations in plant tissue throughout the lettuce study and at low As (V) concentrations (0.02mM) in the radish study may be explained by the molar ratio of P:As of approximately 5. From a food nutrition safety standpoint, studies need to concentrate on sub-lethal levels in order to ensure the proper formation of the harvestable portion of the plant. Chapter 3: Adsorption of Arsenate (V) and Arsenite (III) on Goethite in the Presence and Absence of Soluble Organic Carbon The environmental fate of arsenic is of utmost importance as the U.S. EPA has recently proposed to tighten the arsenic drinking water standard from 50 ppb to 5 ppb. In natural systems the presence of dissolved organic carbon (DOC) may compete with As for adsorption to mineral surfaces, hence increasing its potential bioavailability. Accordingly, the adsorption of arsenate As (V) and As (III) on goethite (a-FeOOH) was investigated in the presence of either a peat humic acid (Hap), a Suwannee River Fulvic Acid (FA) (IHSS) or citric acid (CA). Adsorption edges and kinetic experiments were used to examine the effects of equimolar concentrations of organic adsorbates on arsenic adsorption. Adsorption envelopes were conducted over a pH range of 11 to 3, while the kinetic studies were conducted at pH 6.5 for As (V) and pH 5.0 for As (III). Arsenate adsorption was inhibited in the order of Hap > FA > CA while arsenite adsorption was inhibited in the order of CA > FA > Hap. Humic acid reduces As V adsorption starting at pH 9, with a maximum reduction at pH 6.5. Fulvic acid slightly inhibited As (V) adsorption starting at pH 5, and this inhibition increased with a decrease in pH. No effect was observed in the presence of CA. Arsenite adsorption is inhibited by HA starting a pH 7 and increases with a decrease in pH, while FA and CA reduce As (III) adsorption beginning at pH 8, with a continuous reduction as the pH decreases. The differential extent of As V adsorption in the presence of the organic acids suggests that the distribution and the respective densities of the abundant functional groups (phenol/ catechol OH or COO-) are significant in the adsorption of As (V). Furthermore, larger organic acids may hydrophobically partition to surfaces via a more favorable entropy driven reaction mechanism which may influence As (V) diffusion and its subsequent adsorption to surfaces. The decrease in As (III) adsorption is caused by its reduced affinity for the surface at pH values lower than 9, and the simultaneous increase in surface activity by the organic substances' via their COO- functional groups. The results of these experiments suggests that dissolved organic carbon substances are capable of increasing the bioavailability of As in soil and water systems in which the dominant solid phase is a crystalline iron oxide. Chapter 4: Adsorption of Arsenate and Arsenite on Ferrihydrite in the Presence and Absence of Dissolved Organic Carbon (DOC) The adsorption of As (V) and As (III) on synthetic 2-line ferrihydrite in the presence and absence of a peat humic acid (Hap), Suwannee River Fulvic Acid (FA) or citric acid (CA) was investigated. Previous work with goethite has demonstrated the ability of DOC materials to reduce As (V) and As (III) adsorption. In this study, a batch technique was used to examine the adsorption of arsenic (III and V) and DOCs on ferrihydrite in the pH range from 3 to 11. The results obtained demonstrated that As (V) adsorption on ferrihydrite was reduced only in the presence of CA. Arsenate reduced the adsorption of all organic acids except Hap. Both FA and CA reduced As (III) adsorption on ferrihydrite, while Hap had no effect. Fulvic and citric acid adsorption on ferrihydrite was reduced in the presence of As (III), however, adsorption increases of FA and CA were observed at lower pH, which is consistent with a decrease in As(III) adsorption. The peat humic acid had no effect on As (III) adsorption, and we believe that the adsorption process of Hap and As (III and V) on ferrihydrite are independent of each other. The observed differences between this study and the study on goethite are believed to be an intricate function of ferrihydrite's surface characteristics, which affects the mechanisms of surface adsorption and hence the affinity of organic acids such as Hap, FA, and CA for the ferrihydrite surface. As such, the adsorption of DOCs to ferrihydrite are assumed to be energetically less favorable and to occur with a fewer number of ligands, resulting in lower surface coverage of weaker bond strength. Additional factors for the observed differences are discussed. This work demonstrates the importance of the solid phase in adsorption processes and functional group composition, as noticeable differences are observed in comparison to a crystalline Fe-oxide solid phase.

The objective of this work was analyze the effects of aqueous extracts of Aristolochia esperanzae organs in the germination and early growth of lettuce and radish. All the prepared extracts obeyed the ratio of 100g of plant material + 300 mL of distilled water to produce an extract 100% concentrated. This extract was diluted with distilled water to produce final concentrations of 75, 50 and 25%. In the germination tests the extracts of leaf, stem, root, fruit and flower, with concentration of 100, 75, 50, 25% were evaluated. In the seedlings growth, extracts of leaf, stem and root were used in the concentration of 100 and 50%. In the germination tests, four replicates of 30 seeds were distributed in Petri dishes with two filter paper moistened with 5 mL of the extracts, dried during 12 hours and moistened again with 4,5 mL of distilled water. In the first test, the rate and germination percentage was evaluated. In the growth test, the seeds were previously germinated (2-4mm root length) and transferred to plastic boxes containing the filter paper or coconut fiber, moistened with the extracts. The aerial part height, root length, dry mass and the presence of abnormalities in the seedlings were evaluated. Both tests were maintained at constant temperature (27ºC). The obtained results showed that the leaf extracts affected strongly the germination percentage and all the extracts, in different concentrations, delayed the germination of lettuce and radish seeds. In the growth test could be verified that the stem and root extracts caused abnormalities in the seedlings. The seedlings grown in filter paper presented bigger growth inhibition in relation to seedlings from coconut fiber. The extract concentration was the main responsible for the incentive or inhibition caused on lettuce and radish seedlings growth.
O objetivo deste trabalho foi analisar os efeitos de extratos aquosos de diferentes órgãos de Aristolochia esperanzae na germinação e crescimento de alface e rabanete. Todos os extratos preparados obedeceram à proporção de 100g de material vegetal para 300 mL de água destilada, que produziu o extrato considerado 100% concentrado. A partir deste, foram feitas diluições com água destilada para 75, 50 e 25%. Nos testes de germinação foram avaliados os efeitos dos extratos obtidos de folha, caule, raiz, fruto e flor a 100, 75, 50 e 25% Para o teste de crescimento foram utilizados extratos de folha, caule e raiz na concentração de 100 e 50%. Nos testes de germinação foram utilizadas quatro repetições de 30 sementes distribuídas em placas de Petri forradas com duas folhas de papel de filtro umedecidas com 5 mL dos referidos extratos, secas durante 12 horas e reumedecidas com 4,5 mL de água destilada. No primeiro teste avaliou-se a porcentagem e velocidade de germinação das sementes. No teste de crescimento, as sementes foram previamente germinadas (2-4 mm de radícula) e depois transferidas para caixas plásticas contendo os substratos papel de filtro e fibra de coco umedecidos com os respectivos extratos. Avaliou-se a altura das plântulas, o comprimento radicular, a massa seca das plântulas, e a presença de anormalidades nas plântulas. Os testes foram mantidos a temperatura constante de 27ºC. Através dos resultados obtidos pode-se concluir que os extratos de folha foram os que mais afetaram a porcentagem de germinação e que todos os extratos e as diferentes concentrações retardaram a germinação de sementes de alface e de rabanete. Quanto ao teste de crescimento pode-se verificar que os extratos de caule e raiz causaram anormalidades nas plântulas. As plântulas crescidas no substrato papel de filtro apresentaram maior inibição do crescimento, em relação àquelas crescidas no substrato fibra de coco. A concentração dos extratos foi a principal responsável pelo estímulo ou inibição causada no crescimento das plântulas de alface e rabanete.

Nucleo-cytoplasmic male sterility (CMS) systems are natural systems controlled by two genetic factors: a cytoplasmic determinant causing male sterility and a nuclear determinant able to restore fertility. The Ogura CMS system is widely used for the production of hybrid seeds in cultivated Brassicas. The restorer locus of the Ogura system (Rfo) includes three highly related genes encoding PentatricoPeptide Repeat (PPR) proteins. Two of the most important questions regarding restorers of fertility concern the molecular mechanisms by which PPR restorers counter sterility, and the evolutionary relationships between related PPR genes found at restorer loci. The present study launched a functional analysis of the Rfo restorer protein (PPRB) and a study of the evolution of the Rfo locus in radish. A structure-function analysis of PPRB was undertaken in order to identify the protein domains or residues that are essential for impairing ORF138 accumulation. A functional test for the restorer activity was developed, based on the relative quantification of ORF138 in transgenic roots. For the evolutionary study the sequence of an allele of the Rfo locus obtained from a non-restorer genotype was determined. This allele carries two PPR genes closely related to those of the restorer sequence. The comparison of the two allele sequences showed that the Rfo locus evolved rapidly that intergenic and intragenic recombination operated during its evolution. Overall, the results described in this thesis contributes to a better understanding of the structural and biochemical features important for Rfo restoration activity and sheds light on the peculiar evolution of the Rfo locus.

Como variáveis resposta analisaram-se a umidade relativa do ar, a temperatura ambiente, a temperatura do solo, a radiação solar, além das características morfológicas da cultura: a massa seca, a massa verde, número de folhas (altura, peso, largura da planta), além da produtividade por área em cada tratamento. Os resultados mostraram que as variáveis climáticas envolvidas não apresentaram resultados significativamente diferentes entre os diversos níveis de perfuração pesquisados (5%, 10%, 15% e 20%). No entanto, as alterações microclimáticas ocorridas pela influência das perfurações no filme plástico tornaram o microclima no interior dos túneis baixos propício ao melhor desenvolvimento da cultura da rúcula (Eruca vesicaria sativa (Mill)Thell), e do rabanete (Raphanus sativus L.), resultando em maior produtividade.
The purpose of this research was to determine the environmental changes that the low density polythylene with different perforated film area (5, 10, 15, 20%) may cause on the rocket press culture (Eruca vesicaria sativa (Mill) Thell) during summer and fall time and radish culture (Raphanus sativus L.) during winter and spring time in low tunnel conditions. The climate variables analyzed were the humidity and air temperature, soil temperature and solar radiation. The culture parameters used to compare the treatments were dry matter, green matter, morphologicals characteristics of the plants (number, height, width and weight of leaves) and productivity. The results showed that find any significative differences between the climate variables inside the low tunnel with different perforated film area (5, 10, 15, 20%). However, the modification caused by the low tunnel with perforated plastic did provide performance of rocket press culture (Eruca vesicaria sativa (Mill) Thell) development in summer and fall time, and radish culture (Raphanus sativus L.) development in winter and spring, had better productivity and marketable characteristics.

In different parts of the world, medicinal plants have demonstrated a lot of health benefits to mankind and remains an important source for the discovery of new bio-active compounds. Libya is a typical example of a country where medicinal plants are widely used. Plant extracts of five Libyan medicinal plants were used in this study to investigate their in vivo effects on spermatogenesis and steroidogenesis in male rats and on ovulation and fertility in female rats. The In vitro effects of these plant extracts were also investigated on TM3 Leydig cells and MCF 7 breast cancer cells. A phyto-chemical analysis of the five Libyan medicinal plants (flaxseed, black seeds, radish seed, date palm pollen and nutmeg) was done. The results showed that date palm pollen had a higher antioxidant activity than all of the above mentioned plants. In addition to this, Nigella sativa was observed to possess high flavonol content as well as high antioxidant activity. Male rats exposed to flaxseed, radish seeds and date palm pollen showed no significant alterations in body weight gain, whereas date palm pollen (240 mg/kg, p < 0.05) promoted an increase in body gain. This study also revealed a significant increase in the relative testicular weight of animals exposed to either flaxseed (300mg/kg) or date palm pollen (120mg/kg). In addition, the relative weights of the seminal vesicles of all treated groups showed significant increased values. The level of serum testosterone showed a significant increase after exposure to radish seed (80mg/kg) and a significant dose- dependent increase for date palm pollen when compared to control (P< 0.05). In contrast, flaxseed caused a dose-dependent significant (p <0.01) decrease in testosterone level at radish seed (300mg/Kg). All plant extracts caused a significant increase in sperm concentration. Sperm vitality significantly (p < 0.05) increased by radish seed (80mg/kg), flaxseed (300mg/kg) and date palm pollen (120, 240mg/kg) respectively. Total progressive motility improved significantly at flaxseed (300 mg/kg) (p < 0.001) as well as date palm pollen (p < 0.01). Histological examination of the cross sections of the testis showed clear presence of all stages of spermatogenesis in all the treated groups. Rat epididymides showed normal morphological appearance and their lumen were filled with spermatozoa. The diameter of seminiferous tubules in male rats exposed to date palm pollen (120 and 240 mg/kg) was significantly higher (p < 0.001). The heights of the germ cell epithelia within the eminiferous tubules were also significantly increased in all treated groups. Liver and renal functions tests showed a significant decrease in Alanine transaminase (ALT) and creatinine in all treated groups (p < 0.05), and this demonstrates the lack of cytotoxic effects of date palm pollen, radish seed and flaxseed on the rats. However, these plant extracts produced a non-significant (p > 0.05) increase in Aspartate transaminase (AST) levels. Besides this, superoxide dismutase activity (SOD) in testis was increased significantly by radish seed (160 mg/kg), flaxseed (200 mg/kg) and date palm pollen (120 mg/kg). There was also improved catalase activity in testis of male rats exposed to radish seed and date palm pollen. Regarding male sexual behavior, the time to reach the female and the mount frequency decreased significantly in male rats exposed to flaxseed (300 mg/kg) and date palm pollen (120 and 240 mg/kg; p > 0.05) thus, these plant extracts exhibit aphrodisiac properties. In addition, exposure of male rats to date palm pollen (120 mg/kg) produced a significant (p < 0.01) increase in the number of embryos in untreated female rats. In the female rats, the body weight gain was not affected (p > 0.05). However, the relative uterus weights exposed to nutmeg (200 mg/kg) and date palm pollen (120 and 240 mg/kg) were significantly decreased (p < 0.05). In addition, the relative weights of ovaries after treatment with nutmeg (400 mg/kg) and black seed (400 mg/kg) showed significantly increased values (p < 0.01). Serum FSH was significantly increased (p > 0.05 or 0.01) when the female rats have been exposed to black seed (200 mg/kg), nutmeg (200 mg/kg) or date palm pollen (120 mg/kg). The LH level significantly (p < 0.01) decreased following exposure to black seed (200 mg/kg), date palm pollen (120 mg/kg). On the other hand, serum LH concentration was significantly increased in female rats exposed nutmeg (400 mg/kg; p > 0.05). The creatinine activity in female rat serum in all treated groups was significantly decreased (p < 0.05). Whereas the higher dose of date palm pollen (240 mg/kg) caused only a non-significant decrease. ALT activity in serum of female rat exposed to either black seed (400 mg/kg) or date palm pollen (120 and 240 mg/kg) was shown to decrease significantly (p < 0.05). Histology of the reproductive organs, kidney and liver in the female rats showed no obvious alterations in any of the treated groups. In addition, the number of embryos in female rats significantly increased (p < 0.01; p < 0.001) following exposure of female rats to black seeds 400 and date palm pollen 240 mg/kg, respectively. Incubation of TM3 Leydig cells with radish seeds for 24, 48 or 72 hours caused a significant (p < 0.01) decrease in mitochondrial dehydrogenase activity. Besides that, date palm pollen and flaxseed increased the mitochondrial dehydrogenases activity of TM3 Leydig cells. In addition, higher concentration of date palm pollen, nutmeg and black seed were cytotoxic to MCF7 breast cells. In testis slices testosterone secretion in vitro was significantly increased by flaxseed (500 μg/ml; p > 0·05) and date palm pollen (500 μg/ml; p > 0·01). MCf-7 cells treated with BS 10-50 μg/ml black seed and nutmeg 10-50μg/ml significantly increased cell proliferation. However, the treatment with date palm pollen produced only a weak estrogenic effect, which resulted in a concentration dependent significant increase as observed between 50-1000 μg/ml date palm pollen. In conclusion, in this study, we observed that date palm pollen, radish seed and flaxseed increased libido as well as steroidogenesis and spermatogenesis, improved hepato and nephron-protective effects. In female rats, the plant extracts NM, BS and date palm pollen potentiated the production of gonadotropic hormones. In addition to this, at lower concentrations these medicinal plants promoted cell growth, whereas at higher concentrations they inhibited cell proliferation of MCF- 7 breast cancer cells. The anti-oxidant effects of these plant extracts have been implicated for the above mention effects.

In the province of Chimborazo, found in the Riobamba canton, a study was done with the purpose of comparing different subtracts for the production of earthworm humus (pig feces, guinea pig feces, rabbit feces, bovine feces, market residues, and vegetable residues from the harvest) and its productive effect in vegetables (radish, onion, beet and cabbage). Twenty-four beds, 1.5 m2 (1.5 x 1) in size, as well as sixteen 3 m2 (1 x 3) parcels were used for the study. A design of complete blocks was applied with a monofactorial in the first test, and a bifactorial in the second test. The experiment lasted forty weeks. In general, the best productive behavior by the earthworm humus was distributed among various subtracts. The vegetable residues had the best final weight with a value of 66.75 Kg/parcel. The treatment with guinea pig feces had the best nitrogen content (0.29%). Bovine feces had the best phosphorus content (1.08%). Rabbit feces produced 10.6% of potassium as well as the highest number of earthworms in the harvest (6,900,00). With regards to the production of vegetables, cabbage responded well to almost all of the variables studied. Cabbage obtained superior values than the rest of the vegetables with a 99.5% capture and a 0.25% mortality. The other variables could not be compared because different types of vegetables were used. In the total economic analysis, it was seen that there was a expenditure of 851,960.00 sucres against an income, from the sale of humus and vegetables, of 1,065,000.00 sucres. The net income was 213,120.00 sucres and the benefit/cost was 1.21. It is recommended that this technique be used for small scale production, because the humus improves the ground's physical and chemical characteristics, thus obtaining representative economic gains.

Foram avaliadas 100 progênies de meios irmãos de nabo forrageiro cultivar CATI AL 1000, em ensaio de competição, distribuídas em delineamento látice 10x10, parcialmente balanceado, com três repetições. As progênies superiores foram recombinadas e selecionadas no ano agrícola seguinte obtendo um ciclo a cada dois anos. Foram realizados dois ciclos de seleção. Resumo: O presente trabalho de pesquisa teve como objetivo, avaliar a variabilidade genética de nabo forrageiro cultivar CATI AL 1000 por meio de progênies de meios irmãos, e obter estimativas de parâmetros genéticos para as características: ciclo das plantas, altura das plantas, produtividade de grãos, massa de 1000 grãos, e teor de óleo no grão, sendo conduzido na Fazenda Experimental da UNESP em São Manuel, campus de Botucatu - SP. Foram avaliadas 100 progênies de meios irmãos, distribuídas em Látice triplo 10x10, parcialmente balanceado. Para as características ciclo das plantas, produtividade de grãos, massa de 1000 grãos e teor de óleo no grão, foi detectado diferença significativa para o quadrado médio de progênies, pelo teste F. Para a característica ciclo das plantas, o valor médio e a amplitude de variação das progênies foram, 137 dias, 121 - 158 dias respectivamente. As estimativas dos coeficientes de herdabilidade e ganho de seleção apresentaram os seguintes valores, 45,90 % e 1,09 %. Para característica produtividade de grãos, a média e amplitude de variação de progênies foram, 7,40 g, 0,78 - 18,55 g, (49,40 kg/ha, 5,20 - 123,70 kg/ha) respectivamente. As estimativas dos coeficientes de herdabilidade e ganho de seleção esperado, foram superiores em relação aos encontrados na literatura, comparando-se com populações de milho, para característica produtividade de grão, com valores de 90,70 % e 19,30 % respectivamente. A massa de 1000 grãos apresentou valores para a média e amplitude de variação de progênies de, 7,95 g, 3,35 - 13,51 g, respectivamente. As estimativas dos coeficientes de herdabilidade e ganho de selção apresentaram os seguintes valores, 56,80 % e 4,80 %. Abstract: The objective of the present study was to evaluate fodder radish genetic variability, cultivar CATI AL 100, by using half-sib progenies, and to obtain estimates of genetic parameters for the characteristics: plant cycle, plant height, grain yield, mass of 1000 seeds, and seed oil content, being carried out at UNESP Experimental Farm in São Manuel municipal districtal, Botucatu campus (São Paulo State, Brazil). A hundred half-sib progenies were distributed in triple latice scheme 10x10, partially balanced. For the characteristics plant cycle, grain yield, mass of 1000 seeds, and seed oil content, was detected, for the progenies medium square, significant difference, by F test. For the characteristic plant cycle, the average value and progenies range were 137 days and 121 - 158 days, respectively. The estimates of the inheritability coefficients and selection gain presented the following values: 45,90 % e 1,09 %. For grain yield character, the average value and progenies range were 7,40 g, 0,78 - 18,55 g (49,40 kg ha-1, 5,20 - 123,70 kg ha-1) respectively. The estimates of the inheritability coefficients and expected selection gain, were higher than those ones found in literature, being compared with maize populations, for grain yield, with values of 90,70 % and 19,30 % respectively. The mass of 1000 seeds presented values for the average and progenies range of 7,95 g, 3,35 - 13,51 g, respectively. Red radish (Raphanus sativus L.) anthocyanins were extracted from liquid nitrogen powdered epidermal tissue using acetone and chloroform and isolated using C-18 resin. Chemical structure and conformation of major pigments were elucidated by 1-D and 2-D NMR, Mass Spectroscopy, HPLC, and spectral analyses. Two novel di-acylated anthocyanins, pelargonidin 3-0-[2-0-(β-D-glucopyranosyl)-(6-0- trans-p-coumaroyl- β-D-glucopyranoside)]-5-0-(6-0-malonyl-β-D-glucopyranoside)and pelargonidin 3-0-[2-0-(β-D-glucopyranosyl)-(6-0-trans-feruloyl-β-D-glucopyranoside)]- 5-0-(6-0-malonyl-β-D-glucopyranoside) represented 70% of total pigment. Two mono-acylated anthocyanins (20%) were pelargonidin 3-0-[2-0-(β-D-glucopyranosyl)-(6-0-trans-p-coumaroyl-β-D-glucopyranoside)]-5-0-(β-D-glucopyranoside) and pelargonidin 3-0-[2-0-(β-D-glucopyranosyl)-(6-0-trans- feruloyl-β-D-glucopyranoside)]-5-0-(β-D-glucopyranoside). NOESY revealed folding of the molecule. Electrospray (ES-MS) and tandem mass spectroscopy (MS-MS) were tested as tools for anthocyanin characterization. Anthocyanins were semi-purified using C-18 resin, washed with acidified water and ethyl acetate, and recovered with acidified methanol. Samples were injected into a mass spectrometer in aqueous or methanolic solutions. Charged character of anthocyanins favored the fast and effective detection of intact molecular ions requiring minimal sample preparation and with little interference. MS-MS provided clear and characteristic fragmentation patterns. Qualitative and quantitative anthocyanin pigment content of radish cultivars grown at 2 locations (Corvallis and Hermiston, OR) and harvested at 2 maturity stages, were evaluated. Pigment content depended on cultivar, root weight and location, with higher amounts obtained at Hermiston. Spring cultivars (n=22) had pigmentation in the skin, ranging from 39.3 to 185 mg anthocyanin/100g skin. Red-fleshed Winter cultivars (n=5) possessed from 12.2 to 53 mg anthocyanin/100g root. The major pigments were pelargonidin-3-sophoroside-5-glucoside, mono- or di-acylated with cinnamic and malonic acids; individual proportions varied among cultivars. Estimated pigment yields ranged from 1.3 to 15.8 kg/ha. Acylated and non-aeylated pelargonidin were isolated using semi-preparative HPLC to evaluate the effect of glycosylation and acylation in spectral characteristics, molar absorptivity and color. Molar absorptivity ranged from 15,600 to 39,590 for pelargonidin-3-glucoside and pg-3-rutinoside-5-glucoside acylated with p-coumaric acid, respectively. An hypsochromic shift on λmax was observed in presence of glycosylation. Pelargonidin-3,5-diglucoside and 3,5-triglucoside showed yellow-orange hue angle (>40°) in pH 1.0 buffer, higher than other pg-derivatives. Cinnamic acid acylation caused a bathochromic shift. Malonic acid acylation had little effect on color and no effect on λmax. The solvent system affected molar absorptivity and visual color characteristic of the pigments. Acylation increased pigment resistance to acid hydrolysis.

Radish anthocyanin extract has potential as a natural colorant because of its pigment stability and attractive red hue. Presence of undesirable aroma compounds could limit its applications in foods. The pungent principle of radish, 4-methylthio-3-butenyl isothiocyanate (MTBI), is produced enzymatically upon cell injury from its glucosinolate precursor, 4-methylthio-3-butenyl glucosinolate (MTBG), and undergoes subsequent degradation to produce a number of volatiles. To evaluate the potential of flavor formation, juices were prepared from winter and spring radish cultivars. Whole radishes, peels and flesh, as well as radish extracts, were analyzed for glucosinolates and isothiocyanates. Aroma intensities of radish juice extracts were evaluated using sensory analysis. MTBI was monitored by HPLC (detection level 160 ppb). MTBG was extracted from freeze-dried radish tissue with boiling methanol, purified by anion exchange and enzymatically desulfated. DesulfoMTBG was quantified by HPLC, using desulfosinigrin as internal standard. Identification was performed by fast atom bombardment and electrospray mass spectroscopy. MTBI formation was higher in winter than in spring cultivars (1.5-2.8 and 0.8-1.3 mg/100g fresh weight, respectively), and higher in flesh than in peels. MTBG ranged from 30-65 mg (spring cultivars) to 260-320 mg/100g fresh weight (winter cultivars) with greater concentration in peels than in flesh. Isothiocyanates and glucosinolates were not detected in radish juices. Overall aroma intensities of radish juice concentrates, diluted to 150, 300, 600 and 1200 mg anthocyanin/L in water, were rated using a 16-point scale. Radish concentrates from cultivars Fuego (pigmented peels) and Red Meat Takii (whole red flesh) were compared to commercial red cabbage and radish colorants. Aroma intensities followed first order relationships with anthocyanin concentrations. The commercial colorants were rated slight to moderate, while radish extracts (Fuego and Takii) were rated moderate to large. The aroma intensity of red flesh radish extract was more potent than those prepared from radish peels. Further work includes development of purification techniques which would provide an odorless aqueous extract.

People have concerned about biological properties of radish (Raphanus sativus L.) increasingly. However, development of radish juice or canned radish soup would be convenient for household users and application on food industry. Three cultivars radish, Mei-Hua-Chyun、Chei-Yeh-Meei-Nong-Tzao-Sheng and Gan-Tzae-I, were used in this experiment. The heat stability of myrosinase which is relevant to biological activity of radish was studied. Then the optimum blanching condition was determined by heat stability of peroxidase and polyphenol oxidase combined with the results of various blanching conditions’ effect on enzymes activities and physiochemical characteristics of radish juice. To obtain the optimum condition for radish juice or canned radish soup, sterilization and concentration effects on physiochemical characteristics and sensory evaluation of radish juice were also compared. The results showed that enzymes activities and general physiochemical characteristics varied by cultivar, among them fresh Gan-Tzae-I had higher enzyme activity in myrosinase and peroxidase. Ascorbic acid、 sugar content and free amino acids were higher in Mei-Hua-Chyun. The optimum blanching condition was 90 degree C water for 5 minutes. It is advantageous on operational convenience and high juice yield. Comparing with radish juice from non-blanching, the soluble solids、titratable acidity and formol nitrogen were decreased after blanching, but pH value and color were not significantly different from non-blanching. Sterilization lowered pH、titratable acidity、formol nitrogen、sugar content and free amino acids of radish juice. UHT treatment gave better appearance. L value of canned radish juice was significantly lower than UHT because of browning. The results of sensory evaluation on color and overall likeness indicated that the extent of consumer preference for UHT product was significantly higher than canned(p<0.05). The pH、titratable acidity、formol nitrogen、sugar content、 free amino acids and color of the reconstituted radish juice were affected by concentration methods. Generally, radish juice concentrated by vacuum evaporation had higher quality, especially on color. The L value of vacuum evaporation was significantly different from evaporation(p<0.05). But sensory evaluation did not show similar results, it could be caused by the likeness difference between pannels. This research recommended that after blanching, further radish juice was sterilized by UHT or concentrated by vacuum evaporation. It would be the optimum condition for manufacture of radish juice.
