Purpose The aim of this study is to find how the pyruvate intake and aerobic exercise effect on the body composition, exercise performance ability, blood factor and obesity related hormone, and to verify the effect of pyruvate intake and aerobic exercise as an effective substance for obesity improvement. Methods This study selected 20 obese men in their twenties who has more BMI than 25kg/m2, and are applicable in 25% of the body fat, and randomly sampled group of 10 people for pyruvate intake and aerobic exercise (PYA), and 10 people for placebo intake and aerobic exercise (PLA). Intake of pyruvate and placebo was implemented for 10 weeks, 6 g a day, and aerobic exercise, treadmill exercise in the intensity of 50 ~ 60%’s target heart rate, was conducted for 10 weeks, 3 times a week, 60 minutes a day. To demonstrate the effect of pyruvate intake and aerobic exercise, all of the body composition, exercise performance ability, Lactate, and blood factor and hormone related to obesity were measured before and after the test in the same manner. Results The main results from this study are as follow; 1) In the case of body composition, in PYA, weight(p < .01), BMI(p < .05), body fat percentage(p < .01), and body weight without fat(p < .001) are reduced meaningfully. 2) In the exercise performance ability increased significantly in both PYA(p < .01) and PLA(p < .001) for VO2max, Also, in the case of distance during the exercise, PYA(p < .01) and PLA(p < .05) increased significantly in 15 minutes and PYA(p < .01) and PLA​(p < .05) 30 minutes, but only in PYA from 45 minutes(p < .01) to 60 minutes(p < .05) 3) In case of Lactate, the significant decrease in PYA during stabilization and the significant increase in PYA after 30 minutes of exercise was not seen after 45 minutes 4) In the case of blood factor, HDL-C showed a meaningful decrease in PLA(p < .05) and Leptin showed a meaningful decrease in PYA(p < .001). Conclusions To sum up these results, it was more effective for the group of PYA which ingested pyruvate in improving obesity, even when the same aerobic exercise is conducted.

Background:The worldwide incidence of PyA is reportedly rising due to a combination of increased longevity, multi- comorbidity, iatrogenic complications and increasing use of immunomodulating therapies, while there is limited data on longterm outcomes of PyA.Objectives:To describe the recent incidence, risk factors and long-term outcomes in adults hospitalised with non-gonococcal pyogenic arthritis (PyA) of native joints in Western Australia (WA).Methods:We extracted population-based longitudinally linked administrative health data for patients 16 years or older with a first diagnostic code of 711.xx (ICD9-CM) and M00.xx (ICD10-AM) in WA in the period 1990-2010. Annual incidence rates (IR), risk factors during 14.5 years lookback and outcomes including standardized mortality rates (SMR) during 10.1 years follow-up are reported.Results:A total of 2,777 patients (67% male, mean age 49.8 ± 20.5) received a first diagnostic code for PyA. The AIR increased from 4.5 to 11.8 /100,000 over time as did age at onset (45.1 to 55.4 years) and proportion of female patients (23 to 36%). There was no seasonal variation in PyA incidence but a higher rate of predisposing comorbidities in female patients. Knees (33.6%) and hands (22%) were most frequently affected with 28.4 % of positive cultures not due to Gr+ cocci. Mean hospital stay was 8 days, 30-day readmittance and mortality rate was 12 .8% and 3.1% respectively. During ten years follow-up serious infections (43%), new diagnosis of osteoarthrosis (20%), joint replacement (10.8%), osteomyelitis (6%), and crystal arthropathy (6.3%) were the most common morbidities. Red leaf lettuce (Lactuca sativa) has high nutritional value and is frequently used in salads. In a plant factory with full electric lighting, if the spectrum is incorrect, then red leaf lettuce will have incomplete coloration. This study aimed to establish a light recipe for the mass production of red leaf lettuce using electric light sources in a plant factory by using indicators for quantitative assessment, including energy yield (EY) [grams of fresh weight (FW) harvested per kilowatt hour of electricity input for lighting], photon yield (PY) (grams of FW harvested per mole of photons delivered), anthocyanin yield per kilowatt hour (EYA), and anthocyanin yield per photon (PYA). First, the effects of four types of light quality on FW and anthocyanin content were examined. Then, two types of light quality, light-emitting diode with a red-to-blue photon ratio of 80:20 (R80:B20) and R20:B80, were selected for an experiment involving five treatments. An optimum light recipe (SR5SB1) including R80:B20 treatment during the early stage of cultivation (weeks 1 through 5 after sowing) followed by R20:B80 treatment during the final stage (week 6) was proposed. The SR5SB1 treatment led to FW, EYA, and PYA of 87.8 g/plant, 1.63 mg/kWh, and 0.57 mg·mol–1, respectively. This treatment resulted in the highest EYA and PYA, with 159% and 256% more anthocyanin productivity, respectively, compared with cool white treatment (with FW, EYA, and PYA of 65.8 g/plant, 0.63 mg/kWh, and 0.16 mg·mol–1, respectively). The proposed SR5SB1 light recipe enabled cultivation of red leaf lettuce with a balanced yield and anthocyanin production.

In this project, composite membranes containing nanoparticles of polydopamine PDA (dopamine polymer) will be manufactured and characterized in view of their use for the separation of CO2. Polysulfone will be used as a polymer matrix support while poly(vinyilalcohol) has been chosen as selective layer material. The work will first focus on the optimization of the manufacturing parameters of nano composite membranes and then on the influence of the integration of PDA nanoparticles in the polymeric support at different concentrations. The final objective is to test the material properties, with particular reference to the separation performances of the membranes produced, and critically comment on the results obtained.

L'objectif de ce travail de thèse est de fournir une étude systématique et détaillée des propriétés de mélanges de PLA et de PHA préparés à l'état fondu. En particulier, le comportement rhéologique, la miscibilité, la morphologie, les propriétés mécaniques et l'influence du vieillissement physique sur les propriétés en traction des mélanges PLA/PHBV réalisés en mélangeur interne et par extrusion bi-vis sur toute la gamme de composition ont été étudiés.La sensibilité du PHBV à la dégradation thermique a été mise en évidence, ainsi que l'importance des interfaces entre le PLA et le PHBV sur le comportement rhéologique aux basses fréquences. Les analyses de calorimétrie différentielle à balayage ont montré l'immiscibilité des deux polymères. Deux types de morphologies distinctes (nodulaire et co-continue) selon la composition de mélange ont été observées par microscopie électronique à balayage et microscopie optique en réflexion. La résistance au choc des mélanges PLA/PHBV avec et sans modifiant choc a été étudiée et les propriétés en traction de différents mélanges de PLA avec du PHBV, du Lotader GMA ou du PBAT ont été caractérisées. L'ajout d'une faible quantité de phase dispersée dans la matrice de PLA, initialement fragile, permet d'augmenter considérablement son allongement à la rupture. Cependant, les propriétés ductiles obtenues diminuent avec le temps écoulé entre la mise en forme des échantillons et leur test mécanique. Une étude de l'influence du vieillissement physique sur les propriétés des mélanges à base de PLA a donc été réalisée et un modèle qualitatif a été proposé.

Esse trabalho tem por objetivo o desenvolvimento de um biomaterial constituído de poli (álcool vinílico) (PVA) e poli (ácido acrílico) (PAA) contendo hidroxiapatita (HA) visando a aplicação em sistemas osteocondutores. As nanofibras poliméricas foram produzidas pelo método de eletrofiação a fim de mimetizar a matriz extracelular do osso (ECM), além de obter um material com maior relação área superfície quando comparada a materiais de maiores escalas, o que melhora a adesão e proliferação da célula óssea. Para essa finalidade foi utilizado uma matriz de PVA e PAA variando a concentração da HA na matriz. Pelas características hidrofílicas do PVA, foi necessária sua reticulação, por isso o ácido butano 1,2,3,4-tetracarboxílico (BTCA) foi utilizado e estudado variando sua concentração em duas proporções diferentes, 2 % e 6 % (m/m). A reticulação foi efetiva para a matriz PVA/PAA/HA com no mínimo 2 % BTCA, sendo que o FTIR-ATR comprovou a permanência dos principais grupamentos como a carbonila e a hidroxila e a análise de MEV comprovou a estabilidade física das nanofibras após 48 h de imersão em água. Os diâmetros médios das fibras variaram entre 273 a 437 nm, com menor tamanho e menor variação dos diâmetros das fibras na matriz com a presença de HA. Os resultados da matriz obtidos pela análise mecânica foram semelhantes com as propriedades mecânicas do osso. Os ensaios de análise térmica realizados comprovaram uma mistura homogênea na matriz PVA/PAA/HA. Os testes de viabilidade celular mostraram que na matriz com a presença da HA a atividade celular é maior quando comparada a matriz PVA/PAA, o que indica que a HA na matriz tem papel fundamental na atividade celular.
This study aims to develop a biomaterial composed of poly (vinyl alcohol) (PVA) and poly (acrylic acid) (PAA) containing hydroxyapatite (HA) for application in osteoconductive systems. Polymeric nanofibers were prepared by electrospinning method to mimic bone extracellular matrix (ECM), in addition to obtaining a material with large surface area to volume ratio, which improve the adhesion and proliferation of bone cells. For this purpose we used PVA and PAA matrix by varying the concentration of HA in the matrix. For the hydrophilic characteristics of the PVA, a crosslinking has been required, so BTCA was used and studied by varying its concentration in two different proportions of 2 % and 6 % (w/w). The crosslinking is effective for the PVA/PAA/HA matrix with at least 2 % BTCA, and the FTIR-ATR demonstrated the permanence of the main groups such as hydroxyl and carbonyl and SEM analysis confirmed the stability of nanofibers after 48 hours immersion in water. The medium diameters ranged from 273-437 nm with smaller size and smaller variation of fiber diameters in the array with the presence of HA. The results of the mechanical analysis matrices obtained were very similar to the mechanical properties of bone. The thermal analysis of trials proved one miscible mixture in the PVA/PAA/HA matrix. The cell viability tests showed that the matrix in the presence of HA has higher cellular activity when compared to PVA/PAA matrix, which proves that the HA in the matrix has a fundamental role in cellular activity.

This PhD project investigated the effects of a co-continuous phase structure on the ductility of poly(lactic acid) (PLA) blends. An empirical model was used to predict the phase inversion point of the blends. The co-continuous phase morphology was first observed in polybutylene succinate(PBS)/PLA blends. With as little as 10 wt% of PBS, PBS/PLA blends achieved a dramatic improvement in ductility, over 250% elongation-to-break. Clay additives were incorporated in PBS/PLA blends. Clay was found out to affect the compatibility and co-continuous phase morphology of PBS/PLA blends. The clay dispersion was found to have an intercalated and exfoliated structure at a PBS/PLA composition ratio of 20/80. The clay particles increased the mixing conditions between the polymers by producing a finer structure, but also destroyed the co-continuous phase morphology, resulting in a substantial decrease in elongation-at-break. PLA was then blended with Poly(butylene adipate-co-terephthalate) (PBAT) to examine whether the co-continuous phase model could also be applied to other PLA-polymer blends. From the melt viscosity ratio of PLA and PBAT in the processing regime used in the study, the predicted phase inversion value was 19 wt% of PBAT. This value was verified by the results of mechanical properties, where results for elongation-to-break show a dramatic rise from around 10% up to 300% in the composition range between 10 and 20wt% of PBAT. Polyhydroxyalkanoate (PHA) was also blended with PLA and this project investigated how co-continuous phase morphology affects the blends of two brittle polymers. It was found that when PHA content ranged from 10 to 20wt%, the brittle-brittle polymer blends showed ductile behaviour due to a plane stress effect.

A proposta deste estudo foi avaliar a capacidade osteocondutora da forma sólida do copolímero de PLA/PGA em um modelo experimental de enxerto em seio maxilar . Vinte coelhos machos, branco, da raça Nova Zelândia, cada um pesando cerca de 3,0 kg, foram utilizados e foram divididos em dois grupos de acordo com o material de preenchimento do seio, como segue: ósseo autógeno particulado e copolímero PLA/PGA . A área aumentada diferiu entre os grupos após 3, 7 e 15 dias (p = 0,004) . No entanto, os valores tornaram-se semelhante aos 40 dias (p = 0,458). Depois de 3 e 7 dias, a porcentagem óssea foi estatisticamente significativa entre osso autógeno e PLA / PGA copolímero (P = 0,004 e P = 0,004). Após 15 e 40 dias os valores foram semelhantes nos dois períodos (P = 0,087 e P = 0,087 ). Imunohistoquímica confirma os resultados sobre os dados histomorfométricos. The proposition of this study was evaluate the osteoconductive capability of the solid form of PLA/PGA copolymer in an experimental model of maxillary sinus grafting. Twenty male white New Zealand rabbits, each weighing about 3.0kg, were used and were divided into two groups, according to the sinus filling material, as follows: autogenous bone chips and PLA/PGA copolymers. Augmented area differ between the groups after 3, 7 and 15 days (P=0.004). However, the values became similar on day 40 (P=0.458). After 3 and 7 days the percentage of bone was statistically significant between autogenous bone and PLA/PGA copolymer (P=0.004 and P=0.004). After 15 and 40 days the values were comparable in the two periods (P=0.087 and P=0.087). Immunohistochemistry confirms the results on the histomorphometric data. In conclusion, PLA/PGA copolymer seems to be suitable as resorbable material able to induce bone growth in bone defects. This observation suggests that the material have osteoconductive properties also suitable for application in maxillofacial surgery

Resumo: A proposta deste estudo foi avaliar a capacidade osteocondutora da forma sólida do copolímero de PLA/PGA em um modelo experimental de enxerto em seio maxilar . Vinte coelhos machos, branco, da raça Nova Zelândia, cada um pesando cerca de 3,0 kg, foram utilizados e foram divididos em dois grupos de acordo com o material de preenchimento do seio, como segue: ósseo autógeno particulado e copolímero PLA/PGA . A área aumentada diferiu entre os grupos após 3, 7 e 15 dias (p = 0,004) . No entanto, os valores tornaram-se semelhante aos 40 dias (p = 0,458). Depois de 3 e 7 dias, a porcentagem óssea foi estatisticamente significativa entre osso autógeno e PLA / PGA copolímero (P = 0,004 e P = 0,004). Após 15 e 40 dias os valores foram semelhantes nos dois períodos (P = 0,087 e P = 0,087 ). Imunohistoquímica confirma os resultados sobre os dados histomorfométricos. Em conclusão, o copolímero de PLA/PGA parece ser adequado como material reabsorvível capaz de induzir o crescimento do osso em defeitos ósseos. Abstract: The proposition of this study was evaluate the osteoconductive capability of the solid form of PLA/PGA copolymer in an experimental model of maxillary sinus grafting. Twenty male white New Zealand rabbits, each weighing about 3.0kg, were used and were divided into two groups, according to the sinus filling material, as follows: autogenous bone chips and PLA/PGA copolymers. Augmented area differ between the groups after 3, 7 and 15 days (P=0.004). However, the values became similar on day 40 (P=0.458). After 3 and 7 days the percentage of bone was statistically significant between autogenous bone and PLA/PGA copolymer (P=0.004 and P=0.004). After 15 and 40 days the values were comparable in the two periods (P=0.087 and P=0.087). Immunohistochemistry confirms the results on the histomorphometric data. In conclusion, PLA/PGA copolymer seems to be suitable as resorbable material able to induce bone growth in bone defects. This observation suggests that the material have osteoconductive properties also suitable for application in maxillofacial surgery

Objetivo: Analisar a influência do copolímero de ácido polilático e poliglicólico (PLA/PGA) associado ou não ao osso autógeno no processo de reparo ósseo, em defeitos críticos criados em tíbia de coelhos, por meio da análise histomorfométrica. Material e Método: Doze coelhos receberam 2 ostectomias em cada tíbia por meio de trefina de 6.1 mm de diâmetro. Os defeitos ósseos críticos foram preenchidos de acordo com os seguintes grupos: coágulo sanguíneo (GC), osso autógeno (GA), PLA/PGA (GP) e PLA/PGA associado ao osso autógeno (GPA). Seis animais por período foram submetidos à eutanásia aos 7 e 30 dias pós-operatórios, para a análise histomorfométrica. Resultados: Aos 7 dias, na comparação entre GC(48,33%), GA(25,67%), GP(4,17%) e GPA(21,5%), notou-se diferença estatisticamente significante (Kruskal-Wallis, p=0,0163), evidenciada na comparação entre os GC e GP (Teste de Dunn, p<0,05). Aos 30 dias, a comparação entre GC(56,17%), GA(61,50%), GP(67,33%) e GPA(50,50%), não mostrou diferença estatisticamente significante (Kruskal-Wallis, p=0,2026). Conclusão: Foi possível concluir, neste modelo experimental, que o copolímero PLA/PGA é um biomaterial com propriedades osteocondutoras e pode ser utilizado de maneira isolada, sem a presença de osso autógeno associado.
Objective: To analyze the influence of polylactic/polyglycolic acid copolymer (PLA/PGA) associated or not with autogenous bone in the bone repair process in critical size defects of rabbit tibia through histomorphometric analysis. Methods: Twelve rabbits received 2 osteotomies in each tibia through trephine (6.1 mm diameter). The critical bone defects were filled according to the following groups: blood clot (GC), autogenous bone (GA), PLA / PGA (GP) and PLA / PGA associated with autogenous bone (GP). Six animals were euthanized at 7 and 30 days postoperatively. Results: At 7 days, through of groups comparison GC (48.33 %), GA (25.67 %), GP (4.17 %) and GPA (21.5 %) was observed statistically significant difference (Kruskal-Wallis, p= 0.0163), when compared GC and GP groups (Dunn, p < 0.05). At 30 days, the comparison between GC (56.17 %), GA (61.50 %), GP (67.33 %) and GPA (50.50 %) showed no statistically significant difference (Kruskal-Wallis, p = 0.2026). Conclusion: PLA / PGA copolymer is a biomaterial with osteoconductive properties and can be used isolated without autogenous bone supplementation.

AbstractProbabilistic Büchi automata are a natural generalization of PFA to infinite words, but have been studied in-depth only rather recently and many interesting questions are still open. PBA are known to accept, in general, a class of languages that goes beyond the regular languages. In this work we extend the known classes of restricted PBA which are still regular, strongly relying on notions concerning ambiguity in classical $$\omega $$ ω -automata. Furthermore, we investigate the expressivity of the not yet considered but natural class of weak PBA, and we also show that the regularity problem for weak PBA is undecidable.

Involvement of platelets and coagulation systems in the hematogenous metastasis of tumor cells has been suggested from in vivo and in vitro studies, however, there is still controversy about the exact role of hemostasis in metastasis. To date, at least three types of platelet aggregating mechanisms and three types of tumor cell procoagulants have been reported in different tumor cells.We investigated platelet aggregating activity (PAA), procoagulant activity (PCA) and the relationship between these two activities, using eight human neuroblastoma cell lines, three human leukemia cell lines and human mature lymphocytes. PCA in tumor cells was measured by the single stage recalcification time and the assay with chromogenic substrate S2222. PAA was determined turbidometrically with an aggregometer by adding cell suspensions of tumor cells to platelet rich plasma (PRP). The effects of protease inhibitors, enzymes and thrombin inhibitors on PAA and PCA were also studied.Neuroblastoma cell suspensions showed high PCAs which were reduced in Factor VII deficient human plasma, indicating a tissue factor-like activity. NCG line possessing the highest PCA also showed a high PAA, which was inhibited by pretreatment of cell suspensions with phospholipase A2 and abolished in the presence of heparin, hirudin or MD805 in the assay system. Human leukemia cell lines and mature lymphocytes had weak to moderate PCAs without showing PAA, but became active to express PAA after being removed of cell surface sialic acid by neuraminidase. These results suggest that in neuroblastoma, PCA closely linked with PAA may play a role in the hematogenous metastasis. In hemopoietic cells, PAA expressed when cell surface sialic acid is removed does not correlate with PCA, and sialic acid in these cells possibly prevents direct interaction with platelets in the hemostatic homeostasis.

La presente investigación se basa en un análisis sistemático de literatura, la idea surge a partir del impacto ambiental negativo que se está evidenciando a nivel mundial, derivado en parte, por el consumo desmesurado, pero necesario de tapabocas quirúrgicos, que son utilizados como elemento de protección personal (EPP) a consecuencia de la situación actual causada por la pandemia del COVID – 19. Debido a que el principal componente de los tapabocas es el polipropileno, la inadecuada disposición final que se le da a estos elementos y su tardía degradación ocasionan que estos desechos plásticos se acumulen generando así contaminación a diferentes ecosistemas. A partir de dicha problemática, se evidencia la necesidad de sustituir el plástico convencional derivado del petróleo, por un material con las mismas propiedades, pero que sea amigable con el medio ambiente, por lo cual, se analizaron los biopolímeros que podrían utilizarse en la fabricación de tapa bocas; se identificaron los tipos de Biopolímeros susceptibles para tal fin, comparándolos a partir de sus propiedades físicas, químicas e impacto ambiental, así mismo, se dio importancia a su viabilidad económica, tecnológica e industrial. Los Biopolímeros a tener en cuenta fueron: el PHA (polihidroxialcanoatos), PLA (ácido poliláctico), PHB (Poly-3-hydroxybutyrate), La Quitina y la PA-11 (poliamida 11) de manera que se descartó la Quitina por su efecto corrosivo y la poliamida 11 debido a su propiedad de absorción de humedad, estas características dejaron a estos dos biopolímeros en gran desventaja frente a los otros tres. La principal característica del PHA, PLA y PHB es su tasa de degradación relativamente baja en comparación con el plástico convencional; el PHB cuenta con propiedades muy similares a las del polipropileno, pero se descarta debido a que su costo de producción es muy alto y su utilización en la industria se limita a productos envasados; el PHA cuenta con gran versatilidad esta es una gran ventaja ya que puede ajustarse a las propiedades del polipropileno, además, es resistente al agua y no toxico. De acuerdo a la información recopilada, entre el PHA y el PLA se concluye que cualquiera de los dos puede ser apto para la sustitución del polipropileno en la fabricación de tapabocas quirúrgicos, teniendo en cuenta sus propiedades físicas y químicas, sin embargo, el hecho de que el PLA tiene mayor viabilidad económica hace que su uso en la industria sea cada vez mayor y eso genera la reducción de su precio en el mercado, dándonos la noción que dicho bioplástico es la mejor opción.

Cells of the monocyte/macrophage series develop thromboplastin procoagulant activity (PCA) when activated. This activity may be enhanced by T lymphocytes, but there have been few studies of the PCA of T lymphocytes themselves. In the present study, we have measured the PCA of four T lymphoma cell lines (JURKAT, CEM, HSB2 and MQLT4) as well as normal peripheral blood T lymphocytes, before and after stimulation with phyto-haemag-glutinin (PHA), using clotting and amidolytic methods.Of the four cell lines only one, JURKAT, gave enhanced PCA. after stimulation with PHA., with recalcification times in plastic tubes being shortened from 200 secs, to 90 secs, at a cell concentration of 4 × 106 ml. This activity was shown to be thromboplastin-like by its dependence on Factor VII in plasma and in an amidolytic assay with purified Factors VII and × . JURKAT was also the only one of the four cell lines to secrete interleukin-2. All four cell lines promoted the generation of large amounts of thrombin in platelet-free plasma in glass tubes; the peak thrombin concentration of 25 iu/ml was equivalent to that generated by a procoagulant phospholipid reagent. This activity was dependent on the presence of plasma Factor VIII, and was shown to be due to phospholipids in the cell membranes by its inhibition with phospholipase A2 and C and its activity in a purified system with Factors VIII, IXa and X. Normal T lynphocytes generated similar thromboplastin PCA to JURKAT after PHA stimulation, but their intrinsic PCA in the thrombin generation test was only 15% of that of the lymphoma cells.These results show that some T lymphocytes can develop PCA in both intrinsic and extrinsic systems and this should be taken into account in studies of the PCA of mixed leucocyte populations. The difference in intrinsic PCA between normal and lymphoma cells could be an interesting marker for malignant transformation.
