Microsatellite or simple sequence repeat (SSR) markers have wide applicability for genetic analysis in crop plant improvement strategies. Marker-assisted selection is an important tool for plant breeders to increase the efficiency of a breeding process, especially for multigenic traits, highly influenced by the environment. In this paper, the relationships between SSR markers and 26 quantitative traits of hybrid maize varieties (Zea mays L.) were analyzed. Association analyses were performed based on 30 SSR primers in a set of thirteen hybrid maize varieties. A total of 112 SSR markers were detected in these genotypes. The number of alleles per locus ranged from 1 to 17, with the average number of alleles per locus equal to 3.7. The number of molecular markers associated with observed traits ranged from 1 (for the number of kernels in row, ears weight and fresh weight of one plant) to 14 (for damage of maize caused by P. nubilalis) in 2016 as well as from 1 (for soil plant analysis development—SPAD, the number of grains in ear and fresh weight of one plant) to 12 (for carotenoids content) in 2017. The sum of statistically significant associations between SSR markers and at least one trait was equal to one hundred sixty in 2016 as well as one hundred twenty-five in 2017. Marker trait associations (MTAs) were found on the basis of regression analysis. The proportion of the total phenotypic variances of individual traits explained by the marker ranged from 24.4% to 77.7% in the first year of study and from 24.3% to 77.9% in 2017. Twenty-two SSR markers performed a significant effect on at least one tested trait in both years of experiment. The three markers (phi021/4, phi036/3, and phi061/2) can be a good tool in marker-assisted selection because they allow simultaneous selection for multiple traits in both years of study, such as the number of kernels in row and the number of grains in ear (phi021/4), the number of plant after germination, the number of plants before harvest, and the number of ears (phi036/3), as well as moisture of grain and length of ears (phi061/2).

ABSTRACT The pleckstrin homology domain-interacting protein (PHIP) was originally identified as a 902-amino-acid (aa) protein that regulates insulin receptor-stimulated GLUT4 translocation in skeletal-muscle cells. Immunoblotting and immunohistological analyses of pancreatic β-cells reveal prominent expression of a 206-kDa PHIP isoform restricted to the nucleus. Herein, we report the cloning of this larger, 1,821-aa isoform of PHIP (PHIP1), which represents a novel WD40 repeat-containing protein. We demonstrate that PHIP1 overexpression stimulates insulin-like growth factor 1-dependent and -independent proliferation of β-cells, an event which correlates with transcriptional upregulation of the cyclin D2 promoter and the accumulation of cyclin D2 protein. RNA interference knockdown of PHIP1 in INS-1 cells abrogates insulin receptor substrate 2 (IRS2)-mediated DNA synthesis, providing for a specific role for PHIP1 in the enhancement of IRS2-dependent signaling responses leading to β-cell growth. Finally, we provide evidence that PHIP1 overexpression blocks free fatty acid-induced apoptosis in INS-1 cells, which is accompanied by marked activation of phosphoprotein kinase B (PKB)/AKT and the concomitant inhibition of caspase-9 and caspase-3 cleavage. Our finding that the restorative effect of PHIP1 on β-cell lipotoxicity can be attenuated by the overexpression of dominant-negative PKB suggests a key role for PKB in PHIP1-mediated cytoprotection. Taken together, these findings provide strong support for PHIP1 as a novel positive regulator of β-cell function. We suggest that PHIP1 may be involved in the induction of long-term gene expression programs to promote β-cell mitogenesis and survival.

A lytic bacteriophage PHB01 specific for Pasteurella multocida type D was isolated from the sewage water collected from a pig farm. This phage had the typical morphology of the family Podoviridae, order Caudovirales, presenting an isometric polyhedral head and a short noncontractile tail. PHB01 was able to infect most of the non-toxigenic P. multocida type D strains tested, but not toxigenic type D strains and those belonging to other capsular types. Phage PHB01, the first lytic phage specific for P. multocida type D sequenced thus far, presents a 37,287-bp double-stranded DNA genome with a 223-bp terminal redundancy. The PHB01 genome showed the highest homology with that of PHB02, a lytic phage specific for P. multocida type A. Phylogenetic analysis showed that PHB01 and PHB02 were composed of a genus that was close to the T7-virus genus. In vivo tests using mouse models showed that the administration of PHB01 was safe to the mice and had a good effect on treating the mice infected with different P. multocida type D strains including virulent strain HN05. These findings suggest that PHB01 has a potential use in therapy against infections caused by P. multocida type D.

Abstract Antirepressor proteins of bacteriophages are chiefly involved in interfering with the function of the repressor protein and forcing the bacteriophage to adopt the lytic cycle. The genome of Staphylococcus aureus phage, Phi11 has already been sequenced; from the genome sequence, we amplified gp07 gene and analysed its involvement in the developmental pathway of Phi11. Our results indicate that Gp07 functions as a novel antirepressor and regulates the developmental pathway of Phi11 by enhancing the binding of the Cro repressor protein to its cognate operator. We also report our finding that the CI repressor protein of Phi11 binds to the putative operator of Gp07 and regulates its expression. We further report that S.aureus transcriptional repressor LexA and coprotease RecA play a crucial role in the lytic–lysogenic switching in Phi11. We also identified that the N-terminal domain (Bro-N) of Gp07 is actually responsible for enhancing the binding of Cro repressor to its cognate operator. Our results suggest that Phi11 prophage induction is different from other bacteriophages. This study furnishes a first-hand report regarding the regulation involved in the developmental pathway of Phi11.

The human malaria parasite, Plasmodium falciparum possesses unique gliding machinery referred to as the glideosome that powers its entry into the insect and vertebrate hosts. Several parasite proteins including Photosensitized INA-labelled protein 1 (PhIL1) have been shown to associate with glideosome machinery. Here we describe a novel PhIL1 associated protein complex that co-exists with the glideosome motor complex in the inner membrane complex of the merozoite. Using an experimental genetics approach, we characterized the role(s) of three proteins associated with PhIL1: a glideosome associated protein- PfGAPM2, an IMC structural protein- PfALV5, and an uncharacterized protein—referred here as PfPhIP (PhIL1 Interacting Protein). Parasites lacking PfPhIP or PfGAPM2 were unable to invade host RBCs. Additionally, the downregulation of PfPhIP resulted in significant defects in merozoite segmentation. Furthermore, the PfPhIP and PfGAPM2 depleted parasites showed abrogation of reorientation/gliding. However, initial attachment with host RBCs was not affected in these parasites. Together, the data presented here show that proteins of the PhIL1-associated complex play an important role in the orientation of P. falciparum merozoites following initial attachment, which is crucial for the formation of a tight junction and hence invasion of host erythrocytes.

The agricultural wastes from harvesting corn, viz. corn husks and corn cobs were used as raw materials for the preparation of microcrystalline cellulose. From chemical composition analysis, holocellulose, -cellulose, hemicellulose, extractive and ash contents found in the corn husk were higher than those in the corn cob except for lignin. The study found that these corn residues could potentially be used for microcrystalline cellulose preparation. The properties of the microcrystalline cellulose obtained from these corn residues were examined in comparison with the commercial microcrystalline cellulose, Avicel PH101. Their crystal feature was in cellulose I form which showed the crystal reflections at 2s around 15 o, 17 o and 22o, being the same as that of Avicel PH101. Whilst their crystallinity was lower and the crystal size was smaller than Avicel PH101. The crystal size of the microcrystalline cellulose from the corn residues was around 3 nm whereas that of Avicel PH101 was about 4 nm. In addition, the thermal stability of the prepared microcrystalline cellulose was slightly lower than that of Avicel PH101. Corn husks provided more thermally stable microcrystalline cellulose than corn cobs.

The unexpected lysis of a large culture of Halobacterium salinarum strain S9 was found to be caused by a novel myovirus, designated ChaoS9. Virus purification from the culture lysate revealed a homogeneous population of caudovirus-like particles. The viral genome is linear, dsDNA that is partially redundant and circularly permuted, has a unit length of 55,145 nt, a G + C% of 65.3, and has 85 predicted coding sequences (CDS) and one tRNA (Arg) gene. The left arm of the genome (0–28 kbp) encodes proteins similar in sequence to those from known caudoviruses and was most similar to myohaloviruses phiCh1 (host: Natrialba magadii) and phiH1 (host: Hbt. salinarum). It carries a tail-fiber gene module similar to the invertible modules present in phiH1 and phiCh1. However, while the tail genes of ChaoS9 were similar to those of phiCh1 and phiH1, the Mcp of ChaoS9 was most similar (36% aa identity) to that of Haloarcula hispanica tailed virus 1 (HHTV-1). Provirus elements related to ChaoS9 showed most similarity to tail/assembly proteins but varied in their similarity with head/assembly proteins. The right arm (29–55 kbp) of ChaoS9 encoded proteins involved in DNA replication (ParA, RepH, and Orc1) but the other proteins showed little similarity to those from phiH1, phiCh1, or provirus elements, and most of them could not be assigned a function. ChaoS9 is probably best classified within the genus Myohalovirus, as it shares many characteristics with phiH1 (and phiCh1), including many similar proteins. However, the head/assembly gene region appears to have undergone a recombination event, and the inferred proteins are different to those of phiH1 and phiCh1, including the major capsid protein. This makes the taxonomic classification of ChaoS9 more ambiguous. We also report a revised genome sequence and annotation of Natrialba virus phiCh1.

ABSTRACT The pellicle of the protozoan parasite Toxoplasma gondii is a unique triple bilayer structure, consisting of the plasma membrane and two tightly apposed membranes of the underlying inner membrane complex. Integral membrane proteins of the pellicle are likely to play critical roles in host cell recognition, attachment, and invasion, but few such proteins have been identified. This is in large part because the parasite surface is dominated by a family of abundant and highly immunogenic glycosylphosphatidylinositol (GPI)-anchored proteins, which has made the identification of non-GPI-linked proteins difficult. To identify such proteins, we have developed a radiolabeling approach using the hydrophobic, photoactivatable compound 5-[125I]iodonaphthalene-1-azide (INA). INA can be activated by photosensitizing fluorochromes; by restricting these fluorochromes to the pellicle, [125I]INA labeling will selectively target non-GPI-anchored membrane-embedded proteins of the pellicle. We demonstrate here that three known membrane proteins of the pellicle can indeed be labeled by photosensitization with INA. In addition, this approach has identified a novel 22-kDa protein, named PhIL1 (photosensitized INA-labeled protein 1), with unexpected properties. While the INA labeling of PhIL1 is consistent with an integral membrane protein, the protein has neither a transmembrane domain nor predicted sites of lipid modification. PhIL1 is conserved in apicomplexan parasites and localizes to the parasite periphery, concentrated at the apical end just basal to the conoid. Detergent extraction and immunolocalization data suggest that PhIL1 associates with the parasite cytoskeleton.

Aims: The present work is aimed to prepare microcrystalline cellulose (MCC) powder from cotton linters by an eco-friendly bio-chemical process and to evaluate the prepared MCC as tablet excipients. Study Design: Randomized Complete Block Design. Place and Duration of Study: Ginning Training Center, ICAR-Central Institute for Research on Cotton Technology, Nagpur, Maharashtra, India during June to December, 2019. Methodology: The MCC was prepared from cotton linters by enzymatic and chemical methods and compared with commercial grade MCC (Avicel® PH101). The crude enzyme extract of Aspergillus sp. VM-1 was obtained by solid-state fermentation and used for hydrolysis of cotton linters at 60℃ for 60 min. In the chemical process, 10% alkali was used for hydrolysis at boiling temperature for 60 min. In both the processes, the hydrolyzed cotton linters were bleached with hydrogen peroxide. The MCC powders were characterized for physico-chemical and tableting properties based on Indian Pharmacopeia (IP) and Untied States Pharmacopeia (USP) specifications. Results: The α-cellulose content (%) in the synthesized MCC by enzymatic process was 98.1 while the commercial grade MCC, Avicel® PH101 had 98.5. The physico-chemical properties of synthesized MCC by enzymatic process were comparable with Avicel PH101 and meet the IP standards. The degree of polymerization (DP) of prepared MCC and Avicel PH101 were 215 and 157, respectively. The FT-IR spectrum of synthesized MCC had similarity to that of Avicel® PH101. The tableting properties of prepared MCC met USP standards. The MCC prepared from cotton linters by enzymatic was found to be superior to chemical process with respect to cellulose yield, degree of polymerization and tablet dissolution property. Conclusion: The results showed MCC synthesized from cotton linters through enzymatic route is a promising candidate for direct compressible excipient of tablet. The present study highlights that the enzymatic process significantly reduces the alkali usage and heating temperature and thus saves the chemicals and energy in the process.

We find sufficient conditions for a time-local uniqueness of the solution under the time-integral additional condition \[\frac{1}{T}\int_{0}^{T}u(x,t)\eta_1(t)dt=\Phi_1(x), \;\;\;x\in \Omega\subset \Bbb R^n\] where $u$ is the unknown solution of the first boundary value problem for such equation, $\eta_1$ and $\Phi_1$ are the given functions. We use the method of the Green's function.

Disintegrants had influence on disintegration time and dissolution for fast disintegrating tablet (FDTs). Therefore, the purpose of this study was to evaluate the effect of disintegrant type on the characteristics of FDTs. In this research, the tablets were fabricated by direct compression with the compression force of 1.5 tons and using the different disintegrants components (chitin, chitosan, xylitol, microcrystalline celluloses, white bentonite and magnabite F). The physical properties of these tablets were determined. The xylitol tablets were rapidly disintegrated within 7+1 s, whereas that of the others was longer than 30 mins. The tablets containing the mixture of xylitol and other materials were subsequently fabricated since the hardness of the xylitol tablet was very poor. The disintegration time and wetting time of tablets containing 9:1 xylitol:Avicel PH101 was shorter than that of the others. The contact angle of tablets containing xylitol and Avicel PH101 at the ratio of 9:1 was lowest and surface free energy (SFE) of them was highest compared with the other formula. Moreover, scanning electron microscope (SEM) displayed that xylitol and tablets containing 9:1 xylitol:Avicel PH101 had no disordered arrangement, therefore it could promote the disintegrating property effectiently.

Masalah yang sering terjadi dalam proses penabletannya adalah asam mefenamat cenderung melekat pada permukaan stempel dan matris mesin tablet sehingga sering mengalami sticking dan secara berkelanjutan menyebabkan proses capping. Permasalahan lainnya adalah asam mefenamat mempunyai sifat hidrofobic dan kelarutan yang kecil sehingga disolusinya menjadi masalah. Kombinasi Avicel PH101 dan PVP diharapkan dapat memperbaiki permasalahan tersebut diatas, sehingga dapat diperoleh tablet asam mefenamat yang berkarakteristik baik. Penelitian ini bertujuan untuk mendapatkan formula optimum campuran Avicel PH101 dan PVP dalam pembuatan tablet asam mefenamat secara granulasi basah menggunakan metode simplex lattice design (SLD) dengan dua komponen yaitu: Avicel PH101 (A) dan PVP (B). Tiga formula tablet asam mefenamat (F1 (100%A), F2 (50%A:50%B), F3 (100%B)) dibuat secara granulasi basah. Granul diuji waktu alir, kompaktibilitas dan dissolusi, sehingga didapatkan persamaan SLD. Persamaan tersebut digunakan untuk menentukan formula optimum yakni formula tablet dengan respon total sifat fisik granul yang paling optimum. Hasil penelitian menunjukkan campuran Avicel PH 101 30% - PVP 70% memberikan hasil yang optimal pada sifat fisik granul dan tablet asam mefenamat yang dihasilkan memenuhi uji sifat fisik tablet

Background:The Gp07 protein of aureophage Phi11 exhibits growth inhibitory effects when overexpressed inE. coli.The protein harbors two domains- an amino terminal Bro-like domain and a carboxy terminal Ant superfamily like KilA domain, of which the KilA domain retains the growth inhibitory effect of Gp07.Methods:We studied the effects exerted by the overexpression of Gp07 and its separate domains upon the growth rate as well as the morphology of theE. colicells. Additionally, we generated a mutant of Gp07 (designated as ΔGp07) by deleting the first eleven amino acid residues from the amino-terminal region of Gp07, and studied its growth inhibitory effects uponE. coli.Results:Our results indicate that Gp07, ΔGp07 as well as the Carboxy-terminal region of Gp07 upon overexpression, retards the growth rate of theE. colicells and also induces filamentation in the cells. Surprisingly, our data clearly suggests that the growth inhibition and filamentation induced by the the amino-terminal domain of Gp07 is temporal in nature.Conclusion:The carboxy-terminal of domain of gp07 is essential for its activity.

The halophilic myohalovirus Halobacterium virus phiH (ΦH) was first described in 1982 and was isolated from a spontaneously lysed culture of Halobacterium salinarum strain R1. Until 1994, it was used extensively as a model to study the molecular genetics of haloarchaea, but only parts of the viral genome were sequenced during this period. Using Sanger sequencing combined with high-coverage Illumina sequencing, the full genome sequence of the major variant (phiH1) of this halovirus has been determined. The dsDNA genome is 58,072 bp in length and carries 97 protein-coding genes. We have integrated this information with the previously described transcription mapping data. PhiH could be classified into Myoviridae Type1, Cluster 4 based on capsid assembly and structural proteins (VIRFAM). The closest relative was Natrialba virus phiCh1 (φCh1), which shared 63% nucleotide identity and displayed a high level of gene synteny. This close relationship was supported by phylogenetic tree reconstructions. The complete sequence of this historically important virus will allow its inclusion in studies of comparative genomics and virus diversity.

Abstract Background: Banyuwangi has the potential of diverse natural resources. One example of the potential plant that can be utilized is butterfly pea (Clitoria ternatea L.). The contents of butterfly pea are phenolic acids, stilbenes (phytoalexins), flavanols, anthocyanins, flavonols and flavanones, polyphenols and flavonoids. In this research, preparations were developed from butterfly pea extract into powder form to make it more practical. The problems occur in making formulations usually have physical characteristics of being thick or liquid making it difficult to formulate into solid dosage forms, so in its manufacture additional ingredients or adsorbents were needed to dry the extract so it is easy to formulate. Avicel pH 101 and aerosil is a combination used as an adsorbent in the manufacture of powders. Aim: This study aimed to determine the effect of Avicel PH101 and Aerosil variations through the results of evaluating the moisture content of butterfly pea ethanol extract powder. Method: This study was a laboratory experiment that began with maceration of butterfly pea simplicia using 70% ethanol and then evaporated to obtain a thick extract of butterfly pea. The preparation of butterfly pea powder was carried out using a combination of Avicel pH 101 and aerosil which was varied in four formulas, namely FI (20%:20%), FII (50%:20% ), FIII (90%:0%), FIV (90% :20%) ). Results: The evaluation results of the water content test that met the requirements were FII(50%:20%) with a moisture content of 2.4%, FIII(90%:0%) with a moisture content of 2.8% and FIV (90%;20%) was the most optimal formulation of Avicel PH101 and Aerosil combination for the manufacture of butterfly pea powder with a moisture content of 1.5% and organoleptically in the form of a powder obtained was very dry and delicate. Marburg virus (MARV) is a negative-sense, single-stranded RNA virus that belongs to the Filoviridae family. Despite having caused numerous outbreaks of severe hemorrhagic fever with high case fatality rates, there are still no clinically approved therapeutics or vaccines to treat or prevent MARV disease. Recombinant vesicular stomatitis viruses (rVSVs) expressing heterologous viral glycoproteins have shown remarkable promise as live-attenuated vaccine vectors, with an rVSV-based Ebola virus vaccine having received regulatory approval in the United States and numerous other countries. Analogous rVSV vaccine vectors have also been developed for MARV and have shown efficacy in several preclinical studies conducted in nonhuman primates. Here, we used a guinea pig model to confirm the protective efficacy of a cloned, rVSV-based candidate vaccine, termed PHV01, expressing the MARV variant Angola glycoprotein. Our results demonstrated that a single dose (2 × 106 PFU) of vaccine administered 28 days prior to challenge with a uniformly lethal dose of guinea-pig-adapted MARV variant Angola provided complete protection from death and disease. Moreover, protection was robust, with as little as 200 PFU of vaccine conferring significant protection. Not only does this study highlight the potential predictive value of the guinea pig model in the evaluation of MARV countermeasures, but it also demonstrates consistent and reproducible protection afforded by a clonal vaccine candidate. Indeed, this study identifies PHV01 as a suitable vaccine candidate for advanced development.

Branded antiepileptic drugs (AEDs) are expensive, and in developing countries, cheaper generic equivalents may be used interchangeably. Therefore, this study aimed to compare the antiseizure activities of some available branded and generic AEDs in mice. The physicochemical properties of four generic and branded AEDs (carbamazepine, CAB; sodium valproate, SVP; phenytoin, PHY, and phenobarbitone, PHB) were determined according to pharmacopoeial methods. The organoleptic proper-ties, tablet hardness and thickness of the samples were also assessed. The anticonvulsant effect of AEDs was determined using maximal electroshock (MES) and pentylenetetrazole (PTZ) induced seizure. The samples of AEDs showed uniformly comparable organoleptic properties. The active moiety content for all the samples fell within the acceptable ranges stipulated in the official compendia. A significant statistical difference (p≤0.05) in disintegration time existed between two generic formulations of PHB, PHB01 (4.62±1.17 min), and PHB02 (1.40±0.26 min). Similarly, a significant statistical difference (p≤0.05) in hardness (10.40±0.15 kgF vs 5.61±0.03 kgF) and disintegration time (4.65±0.49 min vs 0.64±0.30 min) was observed between the two brands of CAB (CAB02 and CAB03 respectively). No significant statistical difference exists in mean recovery time between control (5.70±0.72 min) and treatment groups (CAB01, 6.50±0.65 min) in the MEST. In the PTZ test, there was a significant (p≤0.001) difference in the mean onset of clonic spasm between control (10.03±0.75 min) and PHB, PHB01 (17.50±0.99 min) 30 min post-treatment. In conclusion, no significant variation in the physicochemical and antiseizure activity of branded and generic formulations of AEDs was observed

The ab initio conformational energy minima of the model tripeptide N-formyl-L-alanyl-L-alanine amide (ALA-ALA) were determined by ab initio RHF/4-21G and RHF/6-31G* gradient geometry refinement. For the current investigation, 11 664 RHF/4-21G structures were optimized, representing grid points in the four-dimensional (phi1, psi1, phi2, psi2) conformational space, which were constructed in 40° increments along the outer torsions phi1 and psi2 and in 30° increments along the inner torsions psi1 and phi2 of ALA-ALA. Two new energy minima, previously not reported, are described. The positions of the RHF/6-31G* energy minima in phi,psi-space can differ significantly from the corresponding RHF/4-21G locations, and both sets are not clustered in the centers but on the fringes of the most populated regions of phi, psi-space in protein crystal structures. Thus, the torsion angles of the ab initio energy minima are not those of the typical substructures of proteins: the most stable helices are not alphaR, and the torsion angles of the most stable bend forms are not those most frequently encountered in protein bends. Limitations of the dipeptide approximation are explored, illustrating how the conformational energies of an amino acid residue depend on the state of its neighbor.Key words: alanyl alanine amide, dipeptide approximation, model tripeptide, peptide models, structure of peptides.

Одним из критериев, используемых для проверки гипотезы о равновероятности исходов, является критерий приближенной энтропии. В 2000 г. А. Л. Рухиным была предложена статистика $T^{\phi}$ более общего критерия приближенной $\phi$-энтропии и найдено ее предельное распределение. В настоящей работе обобщение этого результата получено более простым способом. Кроме того, установлено предельное совместное распределение $(T^{\phi_1}, …, T^{\phi_r}$) в ситуации, когда исходы равновероятны. Как следствие, в случае $s=2$ найдено предельное совместное распределение статистик двух критериев пакета NIST: критерия приближенной энтропии и критерия «Serial Test» в предположении о том, что тестируемая последовательность является последовательностью испытаний Бернулли с параметром $\frac12$.

Direct and inverse problems for equations with fractional derivatives are arising in various fields of science and technology. The conditions for classical solvability of the Cauchy and boundary-value prob\-lems for diffusion-wave equations with fractional derivatives are known. Estimates of components of the Green's vector-function of the Cauchy problem for such equations are known. We study the inverse problem of determining the space-dependent component of the right-hand side of the equation with a time fractional derivative and known functions from Schwartz-type space of smooth rapidly decreasing functions or with values in them. We also consider such a problem in the case of data from some wider space of smooth, decreasing to zero at infinity functions or with values in them. We find sufficient conditions for unique solvability of the inverse problem under the time-integral additional condition \[\frac{1}{T}\int_{0}^{T}u(x,t)\eta_1(t)dt=\Phi_1(x), \;\;\;x\in \Bbb R^n\] where $u$ is the unknown solution of the Cauchy problem, $\eta_1$ and $\Phi_1$ are the given functions. Using the method of the Green's vector function, we reduce the problem to solvability of an integrodifferential equation in a certain class of smooth, decreasing to zero at infinity functions. We prove its unique solvability. There are various methods for the approximate solution of direct and inverse problems for equations with fractional derivatives, mainly for the one-dimensional spatial case. It follows from our results the method of constructing an approximate solution of the inverse problem in the multidimensional spatial case. It is based on the use of known methods of constructing the numerical solutions of integrodifferential equations. The application of the Fourier transform by spatial variables is effective for constructing a numerical solution of the obtained integrodifferential equation, since the Fourier transform of the components of the Green's vector function can be explicitly written.

As a component of functional nutrition, maize cultivars with “non-traditional” kernel composition (waxy, oilbearing, sugar, opaque, etc. phenotypic variants) are promising. Mutations in the waxy gene, which break down the structure and function of the enzyme for amylose biosynthesis, lead to a waxy (with a high content of amylopectin) endosperm formation. High variability of the waxy gene limits the use of microsatellite loci in marker associated selection of waxy maize genotypes. The increased frequency of gene rearrangements within the waxy locus facilitated the origination of many high-amylopectin corn lines carrying different SSR allelic variants. The purpose of this study was to evaluate the effectiveness of using waxy locus microsatellite sequences for identification and labeling of waxy maize genotypes. To this end, a complex of biochemical (calorimetry, bichromate method), molecular-genetic (SSR-PCR, capillary gel electrophoresis with fluorescent detection of fragments) and statistical (descriptive statistics, cluster analysis, χ2) analysis methods was used. Plant material used were 33 samples of corn kernels including mutant forms with a high content of amylose, amylopectin, short-chain starches, were kindly provided by VIR genetic collection (Russian Federation) and Maize Genetics Cooperation Stock Center (USA). The contents of starch, short-chain soluble carbohydrates, amylose, amylopectin in the grain of 33 maize samples were evaluated. Compositionally similar (to endosperm carbohydrates content) groups of samples were identified. They include 13 high-amylopectin samples carriers of waxy (wx) gene mutations and 20 samples with wild-type character (Wx). Molecular genetic screening of the collection included an analysis of the polymorphism of the microsatellite loci phi022, phi027, phi061 associated with the waxy gene sequence. Allelic composition of individual loci and their combinations were analyzed in relation to the accumulation of reserve carbohydrates in the kernel endosperm. Only the analysis of the phi022/phi027 combination or all three markers in the complex allows differentiating the wild Wx and mutant wx phenotypes of maize. It was shown that not the individual allelic polymorphisms of the phi022, phi027, phi061 loci are efficient for the markerassociated selection of high-amylopectin maize, but their unique combinations.

Abstract Aim Liqui-Pellet is potentially an emerging next-generation oral pill, which has shown promising results with unique advantages as well as displaying potential for commercial feasibility. Since Liqui-Pellet technology is still in its infancy, it is important to explore the parameters that can affect its performance, particularly the drug release rate. Therefore, the aim of this study is to investigate thoroughly the effect of Avicel PH101 (carrier) and Aerosil 300 (coating material) ratio (R-value) in Liqui-Pellet. Methods Key parameter for Liqui-Pellet formulation in this study was the ratio of carrier and coating material. Tests were carried out to assess the physicochemical properties of different formulations. This involved looking into particle size, robustness, flowability, solid-state and drug release profile. The morphology of Liqui-Pellet was investigated by SEM. Results It is found that R-value does not have a major effect on the success of Liqui-Pellet production. However, R-value does seem to have an effect on Liqui-Pellet size at a certain water content level and a slight effect on the drug release rate. A decrease in Avicel PH101 concentration and an increase in Aerosil 300 concentration in Liqui-Pellet formulations can reduce Liqui-Pellet size and slightly increase drug release rate by 9% after 2 h. The data shows Liqui-Pellet is resistant to friability, able to achieve exceptional flow property and have smooth surfaces, which is critical for applying coatings technology. Such properties are ideal in terms of commercial manufacturing. The XRPD and DSC both show the reduction in formulation crystallinity, which is expected in Liqui-Pellet formulation as a result of solubility of the drug in the co-solvent used in the preparation of Liqui-Pellets. Conclusion Overall it seems that R-value can affect Liqui-Pellet drug release rate and size but not on the production success rate. Graphical abstract

Endoglucanase (EG) is an enzyme of cellulase complex depolymerizing crystalline cellulose to cellulose microfibril fragments. T. reesei Rut-C30 is a catabolite repression resistant mutant which enhances endoplasmic reticulum (ER) and produces 15-20 times more endoglycanase than the wild type QM6a. About 90% of this enzyme is secreted.Purified secreted EG from three day old culture was used for polyclonal antibody production. The IgG was labeled with 15-18nm colloidal gold. EG secreting mycellia, grown in Vogel's medium with 1% Avicel PH101, were collected by centrifugation. This pellate was washed in sodium cacodylate buffer, fixed in tannic acid-glutaraldehyde mixture and cryoprotected with 30% glycerol. This material was placed on the specimen holders and frozen in liquid freon followed by liquid nitrogen. Frozen thin sections were prepared in a LKB Cryonova microtome using glass knives and collected on sucrose drops.

Pinus radiata D. Don is a forest tree species native to the Monterey Baja in California. Due to its rapid growth and desirable lumber and pulp qualities, between 1960 and 1980, about 12,000 ha of P. radiata were planted in Sardinia, Italy. The only disease reported on this conifer species has been Diplodia pinea, which causes tip and branch dieback (3). In January 2012, dieback and mortality of 25-year-old radiata pine trees were observed in a reforestation area of about 20 ha located in northern Sardinia (40°43′N, 9°22′E, 600 m a.s.l.). Symptoms included chlorosis, reddish-brown discoloration of the whole crown or dieback starting in the upper crown and progressing downward through the crown, and necrotic bark tissues at root collar. Approximately 25% of the trees were affected. In a first attempt, a Phytophthora species was consistently isolated from the rhizosphere of 23 symptomatic trees, which included necrotic fine roots using oak leaves as bait (4). Afterwards, it was also isolated from phloem samples taken from the margins of fresh lesions at the stem base and upper roots of affected trees using synthetic mucor agar medium (1). Isolation from soil samples of six healthy pine trees randomly selected in the site did not yield any Phytophthora isolate. On carrot agar (CA), Phytophthora colonies were stellate to slightly radiate with limited aerial mycelium. Sporangia were obpiryform, non-papillate, and non-caducous, measuring 46.9 to 51.2 × 29.1 to 32.6 μm (l:b ratio 1.9). Hyphal swellings were formed in chains or clusters; chlamydospores were not observed. These isolates had cardinal temperatures of <5°C, 25°C, and 35°C, respectively. Their morphological and cultural features were typical of Phytophthora cryptogea Pethybridge & Lafferty. They were heterothallic and produced oogonia with amphyginous antheridia when paired with an A2 mating type tester strain of P. cryptogea. This identity was corroborated by sequence analysis of the internal transcribed spacer (ITS) region of the rDNA. BLAST searches showed 99% homology with sequences of P. cryptogea available in GenBank (DQ479410 and HQ697245). The ITS sequence of a representative isolate (PH101) was submitted to GenBank (Accession Nos. KC603895). The strain PH101 was stored in the culture collection of the Department of Agriculture at the University of Sassari. Pathogenicity of isolate PH101 was verified by inoculating five freshly cut logs of radiata pine (1 m long and 15 cm diam.) with a 5-mm agar plug taken from the margin of 4-day-old culture grown on CA (4). The plug was inserted in a 5-mm hole made through the bark with a cork borer. Five control logs were inoculated with sterile CA. All logs were incubated in a growth chamber at 20°C. Phloem lesion sizes were assessed after 1 month and measured 9.7 ± 5.5 cm2 (average ± standard deviation). Control logs had no lesions. The pathogen was re-isolated from the lesions, thus fulfilling Koch's postulates. P. cryptogea has been previously reported in Australia, causing decline of radiata pine trees in wet and flooded soils (2).
