Malaria is responsible for half a million deaths annually and poses a huge economic burden on the developing world. The mosquito-borne parasites (Plasmodium spp.) that cause the disease depend upon an unconventional actomyosin motor for both gliding motility and host cell invasion. The motor system, often referred to as the glideosome complex, remains to be understood in molecular terms and is an attractive target for new drugs that might block the infection pathway. Here, we present the high-resolution structure of the actomyosin motor complex from Plasmodium falciparum. The complex includes the malaria parasite actin filament (PfAct1) complexed with the class XIV myosin motor (PfMyoA) and its two associated light-chains. The high-resolution core structure reveals the PfAct1:PfMyoA interface in atomic detail, while at lower-resolution, we visualize the PfMyoA light-chain binding region, including the essential light chain (PfELC) and the myosin tail interacting protein (PfMTIP). Finally, we report a bare PfAct1 filament structure at improved resolution.

BACKGROUND. Mechanisms of the initiation of renal interstitial inflammation and fibrosis caused by immunoglobulin monoclonal free light chains (mFLC) in monoclonal gammopathy are well established. As far as these damage pathways are considered to be universal we hypothesize that polyclonal free light chains (pFLC) could have a similar effect on tubular and interstitial tissue and lead to chronic kidney disease (CKD) progression in primary glomerulopathies. THE AIM of this retrospective study was to analyze the association of pFLC kappa (pFLC-κ) and lambda (pFLC-λ) assessed in serum by Freelite® with clinical and morphological parameters and CKD progression in IgA-nephropathy (IgAN) cohort.PATIENTS AND METHODS. In this retrospective study, we enrolled 24 patients with IgAN proven by kidney biopsy (KBx). pFLC-κ and pFLC-λ levels were assessed in all cases at the time of KBx by Freelite® method (N pFLC-κ=3.3-19.4 mg/l, N pFLC-λ=5.7-26.3 mg/l). The normal κ/λ ratio was the inclusion criterion. In all cases, we determined serum creatinine, estimated glomerular filtration rate by CKD-EPI method (eGFRCKD-EPI), and daily proteinuria. Morphological findings were defined semiquantitatively by light and immunofluorescence microscopy. Oxford MEST-C score was evaluated as well as % of glomerulosclerosis. Correlation between parameters was assessed by Spearman’s coefficient. Cox proportional hazards regression was used to analyze the association of parameters with the progression of CKD estimated as an elevation of serum creatinine ≥25 % from the initial level or the initiation of renal replacement therapy at the end of the follow-up period (median was 28 (7; 37) months).RESULTS. Median of pFLC-κ 30.2 (6.1; 67.5) mg/l, median of pFLC-λ 27.6 (11.1; 92.1) mg/l. Levels of pFLC-κ and pFLC-λ were increased in 66.7 % and 50 % of patients, respectively. eGFR CKD-EPI median was 41 (26; 65) ml/min/1.73m2. Serum creatinine correlates with pFLC-κ (R=0.62, p<0.01) and pFLC-λ (R=0.45, p=0.03). Among morphological parameters pFLC-κ correlates with interstitial inflammation (R=0.47, p=0.02), tubular atrophy (R=0.54, p<0.01), interstitial fibrosis (R=0.44, p=0.03), peritubular capillaritis (R=0.42, p=0.04), T-score (R=0.66, p<0.01) and combined MEST-C score (R=0.45, p=0.03). For pFLC-λ the correlations with tubular atrophy (R=0.45, р=0.03) and Т-score (R=0.56, p<0.01) were shown. In Univariate Cox regression analysis pFLC-κ and pFLC-λ were associated with CKD progression (Exp(ß)=1.053; 95,0 %CI 1.003-1.105; p=0.038 and Exp(ß)= 1.041; 95,0 %CI 1.002-1.082; p=0.038, respectively) CONCLUSION. Polyclonal FLC, mostly pFLC-κ, were associated with tubulointerstitial inflammation and fibrosis in patients with IgAN. Increased levels of either pFLC-κ or λ could be proposed as a predictor of CKD progression in patients with IgAN.

Abstract Selection for postweaning fecal egg count (PFEC) EBV has revealed that low-PFEC sheep have greater resistance to gastrointestinal nematode infection, greater survivability to weaning, and greater anamnestic response to booster clostridial vaccination. Therefore, it can be hypothesized that selection for PFEC may result in improved generalized immunity. It has also been reported that good-quality colostrum has a thick consistency and yellowish color. Thus, the aim of this study was to compare IgG concentration and color of colostrum in Katahdin ewes divergent in their PFEC EBV. All lactating ewes at the SWAREC were analyzed and were sorted into three groups: low-PFEC (PFEC &lt; -50, n = 39), mid-PFEC (-49 &lt; PFEC &lt; +49, n = 38), and high-PFEC (PFEC &gt; +50, n = 23) and sampled within 6-h post-partition. Upon parturition, a 20 mL colostrum sample was sterile collected by milking 10 mL from each udder half of the ewes; all samples were placed into pre-labeled 50 mL tubes and frozen on-site. Colostrum samples were thawed and diluted in 36% ammonium sulfate and centrifuged at 3,000 x g for 20 min at 4ºC. The removed whey portion was dialyzed using a 3,000 MW membrane cassette in 1 L of PBS (pH 7.4) at 4ºC for 12 h. Total IgG from colostrum was measured using an ovine-specific enzyme-linked immunosorbent assay (ELISA), and color parameters were measured using a Minolta chromameter. To evaluate IgG and color differences, data were analyzed using the General Linear Model (GLM) procedure of SAS with Tukey’s HSD test to compare the effects of ewe PFEC genotype on colostrum IgG, Yellowness Index (YI), Whiteness Index (WI), L*, b* and a* values. Main effects of ewe PFEC genotype displayed a stepwise decrease in average colostrum IgG from low-PFEC to high-PFEC groupings (825 ng/mL, 513 ng/mL, and 258 ng/mL, respectively; P &lt; 0.0429). Likewise, a stepwise decrease in b* value, indicating relative yellowness, occurs from low-PFEC to high-PFEC groups (18.97, 16.91, and 15.22, respectively; P &lt; 0.0459). These data suggest low-PFEC Katahdin ewes generate greater antibody concentration and yellowness within colostrum. Therefore, lambs born to these ewes would have a significant advantage in passive immunity from their dams, potentially contributing to lamb survivability.

Abstract In a previous study, Katahdin lamb survivability to weaning was affected by sire post-weaning fecal egg count estimated breeding values (PFEC EBV) where lambs sired by low PFEC rams had greater survivability than high-PFEC-sired lambs. Therefore, selection for PFEC may also select for improved generalized immunity. The aim of this study was to compare effects of divergent PFEC mating on antibody production in purebred Katahdin lambs in response to vaccination. Katahdin dams with a PFEC value of greater than zero were bred to positive (high-PFEC) FECEBV rams, likewise, dams with a PFEC value of less than zero were bred to negative (low-PFEC) FECEBV rams. All lambs were managed as one cohort until approximately 105 days of age. During this time on pasture, a natural helminth infection was established, blood samples were taken weekly, and fecal samples were collected bi-weekly. Lamb FEC corresponded to sire/dam genotype (943 epg high-PFEC vs. 455 epg low-PFEC; P &lt; 0.001). Serum samples were taken weekly for 3 wks after initial vaccination for Clostridium perfringens type C&D and C. perfringens type A and for 4 wks after booster vaccination, which occurred 3 wk following primary vaccination. Total circulating IgG was measured used an ovine-specific enzyme-linked immunosorbent assay (ELISA). To evaluate lamb IgG differences, data were analyzed using the General Linear Model (GLM) procedure of SAS with fixed effects of genotype, time, and genotype X time interactions. Means comparisons were performed on LS means of fixed affects or interactions, using Bonferroni adjustment. A genotype by time interaction revealed that IgG response to boostering favored LoPFEC lambs (1679.91 vs 1153.08 ug/mL, respectively; P &lt; 0.001). When averaged across time, LoPFEC lambs generated greater IgG concentration than HiPFEC lambs (886.79 ug/mL vs 465.15 ug/mL, respectively; P &lt; 0.001). These data indicate that PFEC breeding values in Katahdin sheep may predict immunological fitness outside the context of parasitic infections and has implications for use in selection of individuals less prone to disease.

Processing of end-of-life tyres (ELT) produces polymer fibres (PFELT) as a by-product. PFELT currently presents a challenge to the recycling industry, due to the increasing numbers of ELT and lack of alternatives for the re-use of this material. The object of this investigation was to propose an alternative for re-using PFELT, in order to improve the performance properties of hot mix asphalt (HMA). This study enabled us to understand the relation between the addition of polymer fibre to the aggregate-binder matrix of the HMA in depth, as well as its effects on the mechanical properties of the resulting asphalt mix. To do this, we first made a physical and chemical characterization of the PFELT (TGA, SEM, polarized light, and fluorescence microscopy), establishing a modification methodology using two asphalt binders (CA-24 and CA-14) and three PFELT contents (0.1%, 0.3%, and 0.5%). The HMA was designed using the Marshall method. The mechanical performance of the HMA was evaluated in a range of operating temperatures, from −10 °C to 50 °C, observing the following properties: (1) resistance to thermal cracking; (2) stiffness modulus; (3) indirect tensile strength; and (4) resistance to permanent deformation. The results show that the addition of 0.3% and 0.5% of PFELT to the asphalt binder significantly improved the mechanical performance properties of the mixes studied, with a greater effect at high operating temperatures; the resistance to permanent deformation increased by more than 30%.

Harmful algal blooms (HABs) pose a global threat to the biodiversity and stability of local aquatic ecosystems. Rapid and accurate classification of microalgae and cyanobacteria in water is increasingly desired for monitoring complex water environments. In this paper, we propose a pulse feature-enhanced classification (PFEC) method as a potential solution. Equipped with a rapid measurement prototype that simultaneously detects polarized light scattering and fluorescence signals of individual particles, PFEC allows for the extraction of 38 pulse features to improve the classification accuracy of microalgae, cyanobacteria, and other suspended particulate matter (SPM) to 89.03%. Compared with microscopic observation, PFEC reveals three phyla proportions in aquaculture samples with an average error of less than 14%. In this paper, PFEC is found to be more accurate than the pulse-average classification method, which is interpreted as pulse features carrying more detailed information about particles. The high consistency of the dominant and common species between PFEC and microscopy in all field samples also demonstrates the flexibility and robustness of the former. Moreover, the high Pearson correlation coefficient accounting for 0.958 between the cyanobacterial proportion obtained by PFEC and the cyanobacterial density given by microscopy implies that PFEC serves as a promising early warning tool for cyanobacterial blooms. The results of this work suggest that PFEC holds great potential for the rapid and accurate classification of microalgae and cyanobacteria in aquatic environment monitoring.

Abstract Post weaning fecal egg count (PFEC) estimated breeding value (EBV) has been recently linked with improved immune competence and lamb survival to weaning, thus improving health though selection. Animal movement is also linked to animal health, however little is known about behavior differences in sheep with divergent PFEC EBV. Thirty Katahdin ewes were selected after weaning, from the Southwest Virginia Agricultural Research and Extension Center. Where high PFEC ewes (EBV &gt; 172) and 15 low PFEC (EBV &lt; -84) were selected. Upon placement into pastures, ewes were fitted with Digitanimal GPS collars that measure GPS location every 15 min and store data on a micro SD card. Additional data collection included bi-weekly body weights, FAMACHA scores, body condition scores and fecal egg count. There was no difference in any physiological measurement in these ewes for the duration of this 4-week study. However, significant differences were observed between groups for daily distance traveled. On average, High-PFEC ewes traveled 4.52 km, where low ewes traveled only 4.16 km (P &lt; 0.0001). The average distance traveled between GPS signals (every 15 min) was 48.6 m for high ewes and 47.0 m for Low-PFEC ewes (P &lt; 0.0001). Interestingly, the time of day where maximum activity occurred (1506 h vs 1448 h; High and Low-PFEC respectively) was not different, nor was the distance those ewes traveled in that hour (522.6 vs 519.2 m). These data provide a preliminary snapshot of movement differences that occur between PFEC groups and may inform future studies to evaluate linkage between health and movement in grazing livestock.

Целью нашего исследования было определение уровня пСЛЦ при различных пролиферативных и непролиферативных гломерулопатиях Пациенты и методы. Обследованы 97 пациентов с хронической болезнью почек, среди них 51 мужчина и 46 женщин, средний возраст 47,5±14,6 лет. У всех обследуемых диагноз гломерулопатии был подтвержден морфологически. Уровень пСЛЦ сыворотки крови определяли методом «Freelite» («Binding Site Ltd», Великобритания). Результаты. Пролиферативные гломерулопатии были выявлены у 46 пациентов, непролиферативные — у 51. Уровень СЛЦ-каппа в сыворотке крови составил в первой группе 38,8 (18,7–61,0) мг/л, во второй — 21,7 (13,6–33,1) мг/л, р=0,003. Уровень СЛЦ-лямбда в сыворотке крови составил в первой группе 34,68 (23,1–56,4) мг/л, во второй — 25,4 (19,5–31,9) мг/л, р=0,005. В целом референсные значения для СЛЦ-каппа и СЛЦ-лямбда были превышены соответственно у 65% и 54% пациентов. Независимо от формы гломерулопатии статистически значимых взаимосвязей между пСЛЦ и традиционными показателями системного воспаления (СОЭ, С-реактивный белок) и долей полностью склерозированных клубочков выявлено не было. Заключение. Уровень пСЛЦ в сыворотке крови в основном отражает активность локального тканевого иммунного воспаления, которое поддерживается активацией В-лимфоцитов. Introduction. Light chains of immunoglobulins are not only structural elements of immunoglobulins of all classes, but can become independent units of immuno-­mediated inflammatory reactions. Serum polyclonal free light chains (pFLC) are also found in a certain amount in practically healthy people. It has been shown that an increase in their serum levels can be considered as biomarkers of the activity of the underlying disease and a possible target for new treatment methods. There has been no extensive study of the role of pFLC in the pathogenesis of diseases unrelated to plasma cell diseases. The aim of our study was to determine the level of pFLC in various proliferative and non-proliferative glomerulopathies. Patients and methods. 97 patients with chronic kidney disease (CKD) were examined, among them 51 men and 46 women, average age 47.5±14.6 years. The diagnosis of glomerulopathy was confirmed morphologically in all the subjects. The serum level of pFLC was determined by the “Freelite” method (“Binding Site Ltd”, UK). The glomerular filtration rate (eGFR) was calculated using the “2021 CKD-EPI Creatinine” formula. Results. Proliferative glomerulopathies were detected in 46 patients, non-proliferative — in 51. The level of pFLC-kappa in the blood serum in the first group was 38.8 (18.7–61.0) mg/l, in the second — 21.7 (13.6–33.1) mg/l, p=0.003. The level of pFLC-lambda in the blood serum in the first group was 34.68 (23.1–56.4) mg/l, in the second — 25.4 (19.5–31.9) mg/l, p=0.005. In general, the reference values for pFLC-kappa (19.4 mg/L) and pFLC-lambda (26.3 mg/L) were exceeded in 65% (n=63) and 54% (n=52) patients, respectively. Regardless of the form of glomerulopathy, the presence of significant relationships between pFLC and the detection of signs of systemic inflammation (ESR, C-reactive protein) was not revealed. Also, no statistically significant relationship was found between serum pFLC and the proportion of completely sclerotic glomeruli. For eGFR, the statistical relationship became significant only when the eGFR was less than 60 ml/min/1.73 m2. Conclusion. The level of pFLC in the blood serum mainly reflects the activity of local tissue immune inflammation, which is supported by the activation of B-lymphocytes. In CKD C3 and more severe stages, eGFR should be taken into account when assessing the significance of an increase in serum levels of pFLC-kappa and lambda.

Abstract Economic selection indexes provide the best method to achieve genetic gain for multiple traits simultaneously. Hair sheep enterprises, particularly in environments with substantial gastrointestinal nematode challenge, may benefit from incorporating parasite resistance in a selection index. However, obtaining the relative economic value (REV) for parasite resistance is challenging; information on its impact on productivity is incomplete, as are the costs of treatment given anthelmintic resistance. The National Sheep Improvement Program (NSIP) currently provides producers with an index designed to increase total weight of lamb weaned per ewe lambing (TW). Our objective was to assess the effects of including fecal egg count, an indicator of parasite resistance, as an additional goal trait. Including the benchmark scenario with TW as its sole objective, 10 indexes were constructed using standard index theory. Post-weaning fecal egg count (PFEC) was added to this breeding objective with a REV of -1 and the REV for TW was varied from +1 to +5 at +0.5 increments. Weaning weight, number of lambs born and weaned, and PFEC were the selection criteria, modelled as either phenotypes or estimated breeding values (EBV). Phenotypes and EBV were simulated for 1,000 lambs by Cholesky decomposition and used to generate index scores. Ten replicates were run and the top 3% of males and 26% of females were retained on index scores. Annual responses in the goal traits were estimated. Responses in PFEC were expressed as a percentage, assuming a mean PFEC of 2,089 eggs/g as in NSIP. Concordances with those selected in the benchmark scenario were determined. Using EBV as selection criteria, TW increased by 1.7 kg/yr in the benchmark scenario (Table 1). However, unfavorably, PFEC increased by 2.3%/yr. When phenotypic criteria were used, TW increased by 0.5 kg/yr, and PFEC by 0.3%/yr. Once added to the breeding objective, PFEC decreased regardless of the REV of TW. However, response in TW also reduced, although less so as its REV increased. With EBV as the criteria, the REV of +3 for TW appeared preferable: PFEC reduced by 7.1%/yr with 98% of the response in TW retained. With phenotypic selection criteria, PFEC reduced by 5.2%/yr with 94% of the response in TW retained. With +3 REV for TW and EBV as the selection criteria, on average 65% and 82% of males and females, respectively, were chosen in common with the benchmark. When phenotypic criteria were used, those percentages were 60% and 80%, respectively. Implementing a more holistic index with REV of +3 for TW and -1 for PFEC would result in reasonable responses in both traits. However, producers will need to decide if reduced gains in TW is acceptable to improve parasite resistance in a hair sheep breeding program.

Free light chains (FLC) of immunoglobulins have been of interest to researchers in various branches of medicine since their discovery in the late 19th and early 20th centuries. In addition to hematology, where the role of monoclonal FLC (mFLC) produced by the clone of the B-cell line is being actively studied, other specialties are no exception. Thus, in modern neurology and rheumatology, polyclonal FLC (pFLC), produced by B-lymphocytes during their excessive immune/autoimmune stimulation, are being actively studied. In the pathogenesis of kidney disease, both mFLC and pFLC can be involved. The importance of mFLC for nephrology is associated, firstly, with various variants of kidney damage in monoclonal gammopathies - cylinder nephropathy, AL-amyloidosis, etc., and secondly, with the initiation of the epithelial-mesenchymal transition and the progression of sclerotic changes in the renal tubulointerstitium. With regard to pFLC, their increased level in kidney pathology of various origins is associated with an unfavorable prognosis not only in relation to the progression of chronic kidney disease but also in life. This allows us to reasonably assume the participation of PSLC in the initiation of profibrotic processes in the kidney. Currently, it is believed that the mechanism of epithelial-mesenchymal transition, which underlies the formation of fibrosis of the renal parenchyma, can be mediated not only by mFLC, but also by pFLC, which has been demonstrated in a limited number of studies in some glomerulopathies. The review outlines the current understanding of FLC, as well as the role of mFLC and pFLC in renal pathology.

La malaria est la maladie parasitaire la plus mortelle (0,5 million de morts par an), elle résulte de l’infection par un parasite Apicomplexe du genre Plasmodium. PfMyoA, une myosine de la classe XIV de Plasmodium falciparum, est essentielle à la fois pour la motilité et pour l’infectivité du parasite. Dans ce travail, nous avons combiné des expériences de cristallographie aux rayons X, de la cinétique transitoire, de la dynamique moléculaire et enfin de la parasitologie afin de décrypter les spécificités de ce moteur. PfMyoA comprend une extension N-terminale atypique qui agit comme un interrupteur permettant au moteur de se déplacer à haute vélocité lorsque l’extension est phosphorylée (pendant les stades mobiles) ou de produire plus de force lorsqu’elle est déphosphorylée (pendant l’invasion). Les structures de PfMyoA entière révèlent que ses chaînes légères spécifiques sont impliquées dans la stabilisation d’un état pre-powerstroke avec un bras de levier plus relevé que chez les autres myosines grâce à des interactions spécifiques entre le bras de levier et le domaine moteur. Étonnamment, la chaîne légère essentielle (PfELC) atypique joue aussi un rôle important pour l’infectivité du parasite. Ensemble, ces résultats définissent PfMyoA comme une cible thérapeutique de premier ordre pour concevoir une nouvelle génération de composés antipaludiques capables de bloquer la fonction du moteur ou l’association des chaînes légères au bras de levier
Malaria is the deadliest parasitic disease (0.5 million deaths per year) and results from infection by Apicomplexan parasites from the genus Plasmodium. PfMyoA, an atypical class XIV myosin from Plasmodium falciparum, is essential both for parasite motility and infectivity. In this work, we combined X-ray crystallography, transient kinetics, molecular dynamics and parasitology to decipher the specificities of this motor. PfMyoA comprises an atypical N-terminal extension that acts as a switch allowing the motor to move at high speed when the extension is phosphorylated (during motile stages) or to produce more force when dephosphorylated (during invasion). The full-length structures of PfMyoA reveal that the specific light chains are involved in the stabilization of a more primed pre-power stroke through unforeseen motor domain/lever arm interactions. Interestingly, the atypical essential light chain of PfMyoA (PfELC) is also essential for the infectivity of the parasite. Altogether, these results define PfMyoA as a first order target to design a new generation of antimalarial compounds able to block motor function or light chains targeting to the lever arm
