Streszczenie:
Spinal muscular atrophy (SMA) is the most common genetic cause of childhood disease and results from selective loss of α motor neurons. SMA is caused by mutations or deletions of the telomeric copy of the survival motor neuron 1 gene (SMN1). This gene is fundamental for the assembly and regeneration of spliceosomal small nuclear ribonuclear proteins (snRNPs) in all cellular types. The mechanism by which SMN deletion is responsible of selective neuro-muscular defect and specific motor neurons degeneration is still unknown, even if some studies suggested an additional neuronspecific function of the protein. In this work, we investigated in neurons the role of SMN and LSm1 to LSm7, which is involved in mRNA degradation in HeLa cells and may well require the SMN protein for assembly and/or function. We constructed stable neuroblastoma cell lines by RNA interference that express low level of SMN protein and can be considered as a pathological model of SMA. We found that decreased expression of SMN protein does not influence the assembly of snRNPs nor of the LSm1-7 complex. In these cellular clones, LSm1 protein assembles in the cytoplasm as punctuated structures (P-bodies) that are also found in control cells and all other cell types. Moreover, SMN knock-down clones showed an atypical localization of heavy chain neurofilaments (NFH) that fuse in the soma. Overexpressing SMN in wild type neuroblastoma (SH S5Y5) we observed the presence of aggregates in inclusion bodies, suggesting the interaction between SMN and NFH. Even if SMN knock-down resemble the pathological condition of SMA, the level of downregulated protein may be not enough to interfere with the snRNPs biogenesis. We can speculate that in primary culture neurons exists a different regulatory pathway. In fact, experiments on neurons from spinal cord, Purkinje cells and hippocampal neurons demonstrated a mechanism of dendritic localization never seen in cell lines. We showed for the first time that LSm1 protein, beside to localize in P-bodies in the soma, can distribute on the dendrites in punctuate structures that contain no degradation enzymes such as Dcp1a. Further, LSm1 binds specific dendritic mRNAs suggesting an involvement in the mRNA transport and/or in the local protein synthesis. In conclusion, we dimostrated that LSm1 can form new complexes with specific neuronal tasks and hypothesize that the presence of SMN is crucial for the function of these complexes.

In Drosophila, the antimicrobial response against infections can be triggered by two different extracellular mechanisms that both lead to the activation of the Toll receptor. These two mechanisms are activated either by the recognition of specific microbial determinants by Pattern Recognition Receptors (PRRs), or by the cleavage of the circulating serine protease Persephone by a wide range of microbial proteases secreted during infections. However, the molecular mechanism underlying Persephone activation remained ambiguous. We identified a unique region in Persephone pro-domain that functions as a bait for exogenous proteases independently of their origin, type or specificity. Cleavage of Persephone in this bait region constitutes the first step of a sequential activation and licenses the subsequent maturation of Persephone to the endogenous circulating cysteine cathepsin 26-29-p. Our data establish Persephone itself as an immune receptor able to sense a broad spectrum of microbes through the recognition of danger signals rather than molecular patterns

MAROLDA, ROBERTA.

Substance P (SP) is an 11-aa neuropeptide, member of the tachykinins (TK) family, broadly distributed in the central nervous system and having a functional role both in physiological and pathological conditions, as in neurodegenerative diseases (Raffa, 1998, Severini et al., 2002). Altered levels of SP have been observed in the cortical regions of post-mortem brain tissues from patients with Alzheimer’s disease (AD) (Quigley and Kowall, 1991; Waters and Davis, 1997). Recently, a consistent SP reduction in the cerebral cortex, hippocampus, basal ganglia and cerebrospinal fluid of AD patients was reported, indicating a possible role of SP in the progression of this disease. On the basis of the in vitro and in vivo results demonstrating the involvement of SP in neuroprotection, we used CGCs in low K+ conditions. In this model, CGCs undergo to apoptotic cell death with activation of amyloidogenic processing of APP, mimicking molecular mechanisms that occur in vivo in AD. Recent data demonstrated that drugs that can regulate the processing of APP towards the non-amyloidogenic pathway (ADAM) may have a therapeutic potential in AD. Aim of the present work was to assess the possible effects of SP on proteolytic processing of APP, with particular interest towards the α-secretase pathway. Data of the present work demonstrate that SP, at a concentration of 200nM, protects CGCs against apoptotic cell death induced by low K+ conditions, significantly reduces the extracellular fibrils production and levels of intracellular Aβ1-42. In addiction, we demonstrate that SP increases α-secretase activity, through the secretion of the neuroprotective soluble fragment APPα, induces the activation of α-secretase enzymatic activity, ADAM 9 gene and protein expression, ADAM 10 maturation, without influencing the precursor protein expression of Aβ (APP) and of BACE 1, the enzyme involved in the amylodogenic processing of APP. In conclusion, this study demonstrates that SP, by activating the non-amyloidogenic processing of APP, may have a therapeutic potential as disease-modifying agent in AD.

In the 6-hydroxydopamine (6-OHDA) model of Parkinsonism, up-regulation of phosphodiesterase (PDE) 1B mRNA occurs in striatal neurons, associated with increased expression of PDE1B immunoreactivity. Our aim was to assess the functional expression of PDE1B in different subtypes of striatal neurons. We have investigated the co-localization of PDE1B in calbindin-positive projection neurons or CHAT-, PV-, and NOS-positive interneurons. Standard confocal microscopy techniques were performed, comparing lesioned vs. unlesioned striatum. PDE1B immunoreactivity increased in a sub-population of unidentified striatal neurons, but a significant decrease of PDE1B-positive calbindin co-localizing neurons emerged (from 63 to 37%) as a consequence of 6-OHDA lesion. At odds with this data, PDE1B co-localization resulted unchanged in CHAT-, PV- and NOS-positive interneurons. These preliminary results confirm that dopamine deprivation has a peculiar impact on the expression of PDE1B in different striatal cell types. Further analysis of double label immunofluorescence is in course to identify the sub-population of striatal neurons carrying on the increased expression of PDE1B in dopamine denervated striatal neurons. Increased of PDE1B may explain some new aspects of pathogenesis of degenerative basal ganglia disease.

ABSTRACT Background: Obesity is a health emergency growing worldwide. The hallmark of obesity is an augment in visceral fat. Alongside waist circumference gain, impaired blood lipid profile and hypertension it leads to Metabolic Syndrome (MetS). Amongst the several factors contributing to its spread, breakfast consumption plays a role promoting ghrelin regulation. In addition, breakfast consumption may ameliorate cognitive performance. Aim: We focused on how three different commercial breakfasts, with different contents of macronutrients, affected: 1) metabolic response and sense of satiety via ghrelin regulation (protocol M); 2) cognitive processing of sensory information via P300 recording (protocol N). To our purposes, we administered three different commercial breakfasts and glucose, as control, to 12 healthy subjects (6M/6F; 22.2±1.5 Kg/m2; 27±5 yo) who underwent two different experimental protocols to assess aim 1 and 2. Aside control (A 50gr glucose) The commercial breakfasts were as follows: B1) milk (125mL) and cornflakes (30gr); B2) milk (220mL), apple (200gr) and chocolate cream filled sponge cake (30gr) and B3) milk (125mL), apple (150gr), bread (50gr) and hazelnut-chocolate cream (15gr). Protocol M: each subject went through one Oral Glucose Tolerance Test (50gr-OGTT) and three-meal test loads (one for each tested breakfast). We collected blood samples at -15’; 0; 15’; 30’; 45’; 60’; 75’; 90’ and 120’. Volunteers ate breakfast after the second blood sampling. We assayed glucose, insulin and ghrelin plasma levels (total). Protocol N: in four different days, each subject underwent eight evoked related potentials measurement (ERPs), two for each breakfast (one after a night fast and one three hours after breakfast consumption). To our purposes, we recorded frontal zone (FZ) ERPs latencies. Results: metabolic data showed that all three commercial breakfasts gave a significant lower glycemic response when compared to glucose (B1: p=<0.05; B2, B3: p<0.01) alongside a lower insulin response (only breakfasts B1 and B2 in a significant fashion: p<0.01). Ghrelinemia inhibition tended to be greater in B3 group (332.1±30.2 pg/mL) with respect to A (420.6±44.2 pg/mL), but did not achieve statistical significance. Electrophysiological data showed that, three hours after ingestion, B1, B2 and B3 displayed a decrease in FZ ERPs latency when respect to A, thus only B3 in a significant way (p<0.01). Conclusion: we demonstrated that different breakfasts were able to elicit lower glycemic and insulin responses even with a greater carbohydrate content. B1, B2 and B3 groups displayed a tendency to maintain satiety via ghrelin inhibition. Furthermore, hazelnut-chocolate cream breakfast (B3) consumption decreased P300 latency, suggesting ameliorated cognitive performance.

Streszczenie:

To explain the ^(26)Mg isotopic anomaly seen in meteorites (^(26)Al daughter) as well as the observation of 1809-keV γ rays in the interstellar medium (live decay of 26Al) one must know, among other things, the destruction rate of ^(26)Al. Properties of states in ^(27)Si just above the ^(26)Al + p mass were investigated to determine the destruction rate of ^(26)Al via the ^(26)Al(p,γ)^(27)Si reaction at astrophysical temperatures.

Twenty micrograms of ^(26)Al were used to produce two types of Al_2O_3 targets by evaporation of the oxide. One was onto a thick platinum backing suitable for (p,γ) work, and the other onto a thin carbon foil for the (^3He,d) reaction.

The ^(26)Al(p,γ)^(27)Si excitation function, obtained using a germanium detector and voltage-ramped target, confirmed known resonances and revealed new ones at 770, 847, 876, 917, and 928 keV. Possible resonances below the lowest observed one at E_p = 286 keV were investigated using the ^(26)Al(^3He,d)^(27)Si proton-transfer reaction. States in 27Si corresponding to 196- and 286-keV proton resonances were observed. A possible resonance at 130 keV (postulated in prior work) was shown to have a strength of wγ less than 0.02 µeV.

By arranging four large Nal detector as a 47π calorimeter, the 196-keV proton resonance, and one at 247 keV, were observed directly, having wγ = 55± 9 and 10 ± 5 µeV, respectively.

Large uncertainties in the reaction rate have been reduced. At novae temperatures, the rate is about 100 times faster than that used in recent model calculations, casting some doubt on novae production of galactic ^(26)Al.

Streszczenie:
26Al(p,p)26Al and 26Al(p,p’)26Al* scattering reactions were performed at the Holifield Radioactive Ion Beam Facility (HRIBF) at the Oak Ridge National Laboratory (ORNL). The purpose of the elastic scattering study was to determine properties of previously uncharacterized 27Si levels above the proton threshold in the energy range E(c.m.) ~ 0.5 - 1.5 MeV and to calculate reaction rates for the 26Al(p,γ[gamma])27Si reaction that destroys 26Al. The inelastic scattering reaction was also evaluated to investigate the reaction that produces the metastable state of 26Al at E(c.m.) = 228 keV, which would in turn destroy 26Al in the stellar environment. Pure 26Al beams (E(beam) = 13 - 41 MeV) with intensities of ~2*106 26Al/s bombarded a thin polypropylene target of 46 μ[micro]g/cm2 thickness for 7 days. Scattered protons were detected in the Silicon Detector Array (SIDAR), covering laboratory angles 18 to 41 degrees. Background events were rejected by detecting these protons in coincidence with recoiled 26Al particles in an ionization chamber, and proton yields were measured at 45 energies from E(c.m.) = 0.49 - 1.53 MeV. A thick 2.4 mg/cm2 polypropylene target was also bombarded with a 32 MeV 26Al beam for 1.5 days for comparison with thin-target excitation functions. Little evidence for the inelastic scattering reaction was observed, indicating that this is not a significant destruction pathway. For the first time, however, an upper limit for the cross section of this reaction was estimated, and it has been set at 5*10-2 barns. The first upper limits were also established for possible resonances of the elastic scattering reaction with angular momentum transfers up to L = 3 that were not directly observed by this study. Thin-target elastic scattering data suggested a potential resonance at E(r) = 544 keV, which had not been previously observed, with (9/2, 11/2)+ spin and proton width Γp[Gamma_p] ≤ 1 keV. Thick-target analysis appeared to confirm this result. An upper limit for the strength of this resonance was estimated to be 1.4*10-5 keV or 1.6*10-5 keV for a 9/2+ or 11/2+ state, respectively, moderately increasing the total 26Al(p,γ[gamma])27Si resonant reaction rate at supernova temperatures.

Streszczenie:
Seagrass monitoring is a basic tool for measuring the condition of meadows in parallel to the environmental conditions. Posidonia oceanica meadows are very sensitive to anthropogenic effects. In the present study, two monitoring stations of Posidonia oceanica meadows were established on the Aegean coasts of Turkey in the years 2018 and 2019, at 26 m depth in Ildır Bay (İzmir, Turkey), and at 33 m depth in Kara Ada (İzmir, Turkey). The P. oceanica meadows upper and lower limits were defined by balisage systems. In the laboratory, lepidochronological, morphometric, and phenological parameters were also studied.

Streszczenie:
Beneficial use of reclaimed wastewater (RW) and biosolids (BS) in soils is accompanied by large input of sewage-originated P. Prolonged application may result in P accumulation up to levelsBeneficial use of reclaimed wastewater (RW) and biosolids (BS) in soils is accompanied by large input of sewage-originated P. Prolonged application may result in P accumulation up to levels that impair plant nutrition, increase P loss, and promote eutrophication in downstream waters. This study aims to shed light on the RW- and BS-P forms in soils and to follow the processes that determine P reactivity, solubility, availability, and loss in RW and BS treated soils. The Technion group used sequential P extraction combined with measuring stable oxygen isotopic composition in phosphate (δ18OP) and with 31P-NMR studies to probe P speciation and transformations in soils irrigated with RW or fresh water (FW). The application of the δ18OP method to probe inorganic P (Pi) speciation and transformations in soils was developed through collaboration between the Technion and the UCSC groups. The method was used to trace Pi in water-, NaHCO3-, NaOH-, and HCl- P fractions in a calcareous clay soil (Acre, Israel) irrigated with RW or FW. The δ18OP signature changes during a month of incubation indicated biogeochemical processes. The water soluble Pi (WSPi) was affected by enzymatic activity yielding isotopic equilibrium with the water molecules in the soil solution. Further it interacted rapidly with the NaHCO3-Pi. The more stable Pi pools also exhibited isotopic alterations in the first two weeks after P application, likely related to microbial activity. Isotopic depletion which could result from organic P (PO) mineralization was followed by enrichment which may result from biologic discrimination in the uptake. Similar transformations were observed in both soils although transformations related to biological activity were more pronounced in the soil treated with RW. Specific P compounds were identified by the Technion group, using solution-state 31P-NMR in wastewater and in soil P extracts from Acre soils irrigated by RW and FW. Few identified PO compounds (e.g., D-glucose-6-phosphate) indicated coupled transformations of P and C in the wastewater. The RW soil retained higher P content, mainly in the labile fractions, but lower labile PO, than the FW soil; this and the fact that P species in the various soil extracts of the RW soil appear independent of P species in the RW are attributed to enhanced biological activity and P recycling in the RW soil. Consistent with that, both soils retained very similar P species in the soil pools. The HUJ group tested P stabilization to maximize the environmental safe application rates and the agronomic beneficial use of BS. Sequential P extraction indicated that the most reactive BS-P forms: WSP, membrane-P, and NaHCO3-P, were effectively stabilized by ferrous sulfate (FeSul), calcium oxide (CaO), or aluminum sulfate (alum). After applying the stabilized BS, or fresh BS (FBS), FBS compost (BSC), or P fertilizer (KH2PO4) to an alluvial soil, P availability was probed during 100 days of incubation. A plant-based bioassay indicated that P availability followed the order KH2PO4 >> alum-BS > BSC ≥ FBS > CaO-BS >> FeSul-BS. The WSPi concentration in soil increased following FBS or BSC application, and P mineralization further increased it during incubation. In contrast, the chemically stabilized BS reduced WSPi concentrations relative to the untreated soil. It was concluded that the chemically stabilized BS effectively controlled WSPi in the soil while still supplying P to support plant growth. Using the sequential extraction procedure the persistence of P availability in BS treated soils was shown to be of a long-term nature. 15 years after the last BS application to MN soils that were annually amended for 20 years by heavy rates of BS, about 25% of the added BS-P was found in the labile fractions. The UMN group further probed soil-P speciation in these soils by bulk and micro X-ray absorption near edge structure (XANES). This newly developed method was shown to be a powerful tool for P speciation in soils. In a control soil (no BS added), 54% of the total P was PO and it was mostly identified as phytic acid; 15% was identified as brushite and 26% as strengite. A corn crop BS amended soil included mostly P-Fe-peat complex, variscite and Al-P-peat complex but no Ca-P while in a BS-grass soil octacalcium phosphate was identified and o-phosphorylethanolamine or phytic acid was shown to dominate the PO fraction that impair plant nutrition, increase P loss, and promote eutrophication in downstream waters. Streszczenie:
Reproductive failure associated with heat stress is a well known phenomenon in avian species. Increased prolactin (PRL) levels in response to heat stress have been suggested as a mechanism involved in this reproductive malfunction. To test this hypothesis, laying female turkeys were subjected to 40°C for 12 h during the photo-phase daily or maintained at 24–26°C. Birds in each group received oral treatment with parachlorophenyalanine (PCPA; 50 mg/kg BW/day for 3 days), an inhibitor of serotonin (5-HT) biosynthesis; or immunized against vasoactive intestinal peptide (VIP). Both treatments are known to reduce circulating PRL levels. Non treated birds were included as controls. In the control group, high ambient temperature terminated egg laying, induced ovarian regression, reduced plasma luteinizing hormone (LH) and ovarian steroids (progesterone, testosterone, estradiol) levels, and increased plasma PRL levels and the incidence of incubation behavior. Pretreatment with PCPA reduced (P< 0.05) heat stress-induced decline in egg production, increase in PRL levels, and expression of incubation behavior. Plasma LH and ovarian steroid levels of heat stressed birds were restored to that of controls by PCPA treatment. As in PCPA-treated birds, VIP immunoneutralization of heat-stressed turkeys reduced (P< 0.05) circulating PRL levels and prevented the expression of incubation behavior. But it did not restore the decline in LH, ovarian steroids, and egg production (P> 0.05). The present findings indicate that the detrimental effect of high temperature on reproductive performance may not be related to the elevated PRL levels in heat-stressed birds but to mechanism(s) that involve 5-HT neurotransmission and the induction of hyperthermia.
