Inflorescence architecture in rice (Oryza sativa) is mainly determined by spikelets and the branch arrangement. Primary branches initiate from inflorescence meristem in a spiral phyllotaxic manner, and further develop into the panicle branches. The branching patterns contribute largely to rice production. In this study, we characterized a rice verticillate primary branch 1(vpb1) mutant, which exhibited a clustered primary branches phenotype. Gene isolation revealed that VPB1 was a allele of RI, that it encoded a BELL-like homeodomain (BLH) protein. VPB1 gene preferentially expressed in the inflorescence and branch meristems. The arrangement of primary branch meristems was disturbed in the vpb1 mutant. Transcriptome analysis further revealed that VPB1 affected the expression of some genes involved in inflorescence meristem identity and hormone signaling pathways. In addition, the differentially expressed gene (DEG) promoter analysis showed that OsBOPs involved in boundary organ initiation were potential target genes of VPB1 protein. Electrophoretic mobility shift assay (EMSA) and dual-luciferase reporter system further verified that VPB1 protein bound to the promoter of OsBOP1 gene. Overall, our findings demonstrate that VPB1 controls inflorescence architecture by regulating the expression of genes involved in meristem maintenance and hormone pathways and by interacting with OsBOP genes.

Electronic word of mouth is a message or information that comes from personal experience or consumer opinion on a product or service and is spread by word of mouth via the internet media. High electronic word of mouth can create customer engagement. Customer engagement is an attitude towards a brand or company, which involves active consumer commitment through online media in the form of word-of-mouth activities, providing reviews, recommendations, and blogging that is driven by motivation. This study aims to measure the effect of electronic word of mouth on customer engagement at Osbond Gym Bekasi. This research uses quantitative techniques with a survey method. Primary data were collected through distributing questionnaires to 85 respondents who are customers of Osbond Gym Bekasi. Data analysis used the Statistical Package for the Social Sciences with the SPSS for Windows 25 application. Based on the research results, it is known that there is a positive influence between electronic word of mouth on customer engagement. Electronic word of mouth has an effect of 69.7% on customer engagement, while 30.3% is another factorElectronic word of mouth adalah suatu pesan atau informasi yang berasal dari pengalaman pribadi atau opini konsumen akan suatu produk atau jasa dan disebarkan dari mulut ke mulut melalui media internet. Electronic word of mouth yang tinggi dapat menciptakan customer engagement. Customer engagement merupakan suatu sikap terhadap merek atau perusahaan, yang melibatkan komitmen konsumen secara aktif melalui media online yang berupa aktivitas word-of-mouth, memberikan review, rekomendasi, dan blogging yang didorong oleh suatu motivasi. AbstractDirect numerical simulations of stratified turbulence are used to test several fundamental assumptions involved in the Osborn, Osborn–Cox, and Thorpe methods commonly used to estimate the turbulent diffusivity from field measurements. The forced simulations in an idealized triply periodic computational domain exhibit characteristic features of stratified turbulence including intermittency and layer formation. When calculated using the volume-averaged dissipation rates from the simulations, the vertical diffusivities inferred from the Osborn and Osborn–Cox methods are within 40% of the value diagnosed using the volume-averaged buoyancy flux for all cases, while the Thorpe-scale method performs similarly well in the simulation with a relatively large buoyancy Reynolds number (Reb ≃ 240) but significantly overestimates the vertical diffusivity in simulations with Reb < 60. The methods are also tested using a limited number of vertical profiles randomly selected from the computational volume. The Osborn, Osborn–Cox, and Thorpe-scale methods converge to their respective estimates based on volume-averaged statistics faster than the vertical diffusivity calculated directly from the buoyancy flux, which is contaminated with reversible contributions from internal waves. When applied to a small number of vertical profiles, several assumptions underlying the Osborn and Osborn–Cox methods are not well supported by the simulation data. However, the vertical diffusivity inferred from these methods compares reasonably well to the exact value from the simulations and outperforms the assumptions underlying these methods in terms of the relative error. Motivated by a recent theoretical development, it is speculated that the Osborn method might provide a reasonable approximation to the diffusivity associated with the irreversible buoyancy flux.

Abstract Uptake of boron (B) in rice (Oryza sativa) is mediated by the Low silicon rice 1 (OsLsi1) channel, belonging to the NOD26-like intrinsic protein III subgroup, and the efflux transporter B transporter 1 (OsBOR1). However, it is unknown how these transporters cooperate for B uptake and how they are regulated in response to B fluctuations. Here, we examined the response of these two transporters to environmental B changes at the transcriptional and posttranslational level. OsBOR1 showed polar localization at the proximal side of both the exodermis and endodermis of mature root region, forming an efficient uptake system with OsLsi1 polarly localized at the distal side of the same cell layers. Expression of OsBOR1 and OsLsi1 was unaffected by B deficiency and excess. However, although OsLsi1 protein did not respond to high B at the protein level, OsBOR1 was degraded in response to high B within hours, which was accompanied with a significant decrease of total B uptake. The high B-induced degradation of OsBOR1 was inhibited in the presence of MG-132, a proteasome inhibitor, without disturbance of the polar localization. In contrast, neither the high B-induced degradation of OsBOR1 nor its polarity was affected by induced expression of dominant-negative mutated dynamin-related protein 1A (OsDRP1AK47A) or knockout of the mu subunit (AP2M) of adaptor protein-2 complex, suggesting that clathrin-mediated endocytosis is not involved in OsBOR1 degradation and polar localization. These results indicate that, in contrast to Arabidopsis thaliana, rice has a distinct regulatory mechanism for B uptake through clathrin-independent degradation of OsBOR1 in response to high B.

The IMPROVE and OSBORN learning models emphasize the development of optimal mathematical skills and generate new ideas in the process of problem solving. This research is used to see the impact of the IMPROVE learning model and OSBORN learning model which is better in mathematical problem solving abilities. This research uses the Quasy Experimental Design method. Hypothesis testing uses an independent sample test. The conclusion of the study is the mathematical problem solving ability of students who use the IMPROVE learning model is better than the mathematical problem solving abilities of students who use the OSBORN learning model.

The current doctoral study uncovered and revived 28 autograph cantatas for Soprano and Basso Continuo composed between 1704 and 1705 by Alessandro Scarlatti (1660-1725). The studied cantatas are held in a manuscript containing 36 cantatas (one of which misses the beginning) in Yale University, Beinecke Rare Book and Manuscript Library, James Marshall and Marie-Louise Osborn Collection, Call Number: Osborn Music MS 2. The historical context in which the cantatas were written and performed is introduced. A physical description of the manuscript follows, with the aim to assist the reader in achieving a better appreciation and understanding of these cantatas in terms of performance practice. The relation between the text and the music, and the adaptation of the rhetorical characteristics of the text into music, are two of the most salient features of this cantata collection. These features are described in great detail and their implications for the performance of the cantatas have been explored. A major contribution of this study in the field of performance practice is the production of a modern performing edition with an editorial commentary and of a recording of the 28 autograph cantatas with harpsichord accompaniment.

Le terme de paradis fiscal est souvent employé dès lors qu’on aborde le thème des montages financiers utilisant une ou des sociétés offshore. Les spécialistes lui préfèrent celui de pays ou zone à fiscalité avantageuse ou privilégiée. Certaines personnes différencient deux catégories de territoires fiscalement favorables. L’expression « zone à fiscalité privilégiée » est souvent employée comme un euphémisme qui renvoie à des pays tels que la Belgique, l'Irlande, le Luxembourg, les Pays Bas, le Portugal, la Suisse, la Tunisie, Hong-Kong, la Malaisie, Singapour, notamment, alors que sont considérés comme « paradis fiscaux » Andorre, Jersey et Guernesey, Gibraltar, le Liechtenstein, Anguilla, Aruba, les Bahamas, la Barbade, les Bermudes, les îles Caïmans, Panama, etc. Il s’agit d’une classification assez vague, relative, en creux, classant séparément les zones dans lesquelles la fiscalité est relativement faible, souvent par rapport à la France ou l’Allemagne, et celles dans lesquelles elle est quasiment inexistante. Les difficultés que posent cette classification sont nombreuses et mettent en évidence l’insuffisance d’un tel raisonnement, souvent journalistique. La notion de paradis fiscal n’a qu’une valeur relative, d’où la difficulté de dégager une définition unique et universelle caractérisant ce phénomène. Il est nécessaire de recourir une définition basée sur un certain nombre de critères. Aussi, généralement, les paradis fiscaux se caractérisent par une absence ou un faible niveau d'imposition des revenus, du capital, et des transactions, ce qui constitue en général la motivation de leur utilisation. Ces avantages fiscaux ne concernent pas nécessairement l'ensemble des personnes qui y établissent leur domicile : le régime fiscal applicable peut différer pour les personnes physiques et morales, pour les résidents et les non résidents. La fiscalité de Jersey par exemple présente surtout un avantage pour des sociétés non résidentes ; Monaco est à l'inverse un paradis fiscal pour les personnes physiques, mais non pour les sociétés. On peut alors parler de pour certains de ces pays de « zones à fiscalité privilégiée ». Pour autant, la définition des zones à fiscalité privilégiée dans le droit français limite également l’usage de cette notion. Le droit français utilise donc une seule expression, dont le critère de détermination principal est mathématique. C’est une définition englobant toutes les notions généralement utilisées : paradis fiscal, centre offshore, etc.. C’est cette définition que nous retiendrons dans notre étude des zones à fiscalité privilégiée. L’optimisation fiscale, dont il est difficile de donner une véritable définition, pourrait se définir comme la mise en œuvre par un contribuable de moyens licites lui permettant de réduire son assiette d’imposition ou le poids de son impôt. C’est une stratégie financière courante et tout à fait légale. La fraude fiscale et l’évasion fiscale quant à elles, consistent, pour la première à une transgression intentionnelle de la règlementation fiscales de façon à minimiser son impôt par diverses techniques (omission volontaire de déclaration, minoration de recettes, majorations de charges, travail clandestin), et pour la seconde à déplacer tout ou partie de son patrimoine ou son activité vers un zones à fiscalité privilégiée, de façon illicite, encore une fois dans le but de se soumettre à l’impôt
The end of tax havens is often used as it addresses the issue of the financial arrangements using one or more offshore companies. Specialists prefer country or area tax advantageous or privileged. Some people distinguish two categories of tax-friendly territories. Tax optimization, which it is difficult to give a real definition, could be defined as the implementation by a taxpayer of lawful means to reduce its tax base or the weight of its tax. Our problem will therefore be as follows: is it possible to imagine a financing package of tax optimization based partly in a privileged but taxation area whose primary objective is the realization of an investment in the service of the economy of the country of origin, and therefore outside the scope of tax evasion? In the first part, we tacherons to define the scope of the possible (first part: fixtures and their suppression), engaging first to different assemblies and types of tax structures (title I), then to the French and European instruments to combat tax evasion . In a second part, we will study the existing sanctions and will demonstrate that an Assembly whose purpose is not only tax allows to counteract part II: the main sanctions and a possible countermeasure)

This thesis examines the history of Commonwealth Government higher education policy in Australia between 1958 and 1997 and its impact on the development of two groups of academic librarians: the Association of Librarians in Colleges in Advanced Education (ALCAE) and the Committee of Australian University Librarians (CAUL). Although university librarians had met occasionally since the late 1920s, it was only in 1965 that a more formal organisation, known as CAUL, was established to facilitate the exchange of ideas and information. ALCAE was set up in 1969 and played an important role helping develop a special concept of library service peculiar to the newly formed College of Advanced Education (CAE) sector. As well as examining the impact of Commonwealth Government higher education policy on ALCAE and CAUL, the thesis also explores the influence of other factors on these two groups, including the range of personalities that comprised them, and their relationship with their parent institutions and with other professional groups and organisations. The study focuses on how higher education policy and these other external and internal factors shaped the functions, aspirations, and internal dynamics of these two groups and how this resulted in each group evolving differently. The author argues that, because of the greater attention given to the special educational role of libraries in the CAE curriculum, the group of college librarians had the opportunity to participate in, and have some influence on, Commonwealth Government statutory bodies responsible for the coordination of policy and the distribution of funding for the CAE sector. The link between ALCAE and formal policy-making processes resulted in a more dynamic group than CAUL, with the university librarians being discouraged by their Vice-Chancellors from having contact with university funding bodies because of the desire of the universities to maintain a greater level of control over their affairs and resist interference from government. The circumstances of each group underwent a reversal over time as ALCAE's effectiveness began to diminish as a result of changes to the CAE sector and as member interest was transferred to other groups and organisations. Conversely, CAUL gradually became a more active group during the 1980s and early 1990s as a result of changes to higher education, the efforts of some university librarians, and changes in membership. This study is based principally on primary source material, with the story of ALCAE and CAUL being told through the use of a combination of original documentation (including minutes of meetings and correspondence) and interviews with members of each group and other key figures.

Large porous protection canopy construction has evolved in Australia over the past 30 years from modest small orchard canopies to large canopies over essential water storages to reduce evaporation and pollution, canopies over large numbers of vehicles for car importers and exporters, and horticultural canopies of over 40 hectares in area. The canopies have proved to be an effective economical method to protect increasing numbers and types of assets from exposure to sun, hail, wind, birds and insects. Canopy design and construction has evolved from the grass roots with initially no structural engineering design input. As the value of assets protected has increased, the request for structural engineering certification of the canopies has become common. To be able to certify the canopy's structure, the certifying engineer needs to confidently be able to predict the wind actions that may occur. In the past there has been limited structural engineering research undertaken into wind loads on porous structures. The aim of this study is to research the characteristic wind actions normal to large flat roof porous canopies and derive design pressure coefficients. Surface friction actions from wind drag across the surfaces are not researched in this thesis and remain a subject for future research. Four scale models of a typical porous protection canopy were constructed for testing in the wind tunnel at the Cyclone Testing Station (CTS), James Cook University, Townsville. The models are of identical geometry, but each of different porosity, 0%, 19%, 38% and 58%. The Models were placed in the CTS boundary layer wind tunnel and rotated through 360° at increments of 15°. At each 15° increment, three sets of pressure readings, each for 30 seconds, were taken at a series of pressure taps located on the Model externally and internally. The pressure readings were processed by the wind tunnel transducer into non dimensional pressure coefficients and then adjusted for the boundary layer speed at the height of the Model. The pressure coefficient results were imported into the analysis software Matlab and Excel and then plotted against the geometry of the canopy. This thesis presents these results graphically with pressure coefficients being plotted against distance. The distance is shown non dimensional as ratios of model width and length to model height. It is evident from the external pressure results, that the introduction of porosity into the canopy's roof surface causes significant reduction in the magnitude of the wind actions acting on the roof when compared to the non-porous canopy. In contrast, the introduced porosity does not modify the magnitude of the wind actions on the walls greatly, but does alter the coefficient distribution. Internal pressure coefficients were found to decrease in magnitude across the model away from the windward edge. To predict the resultant wind actions on the canopy surfaces, the simultaneous external and internal pressure coefficients were summed and adjusted using a gust factor in accordance with the Standard AS/NZS 1170.2. The net surface pressure coefficient was then plotted for four wind directions in increments of 15° from 0° to 45°. The results were then combined to find the peak maximum and minimum pressure coefficients for a ±45° wind direction sector orthogonal to the structure as required in AS/NZS1170.2. It was found that the introduction of porosity to the models caused significant reduction in the magnitude of normal roof wind actions. Increasing the magnitude of the porosity caused only minor further reduction. The wind actions on the porous walls do not decrease significantly from the magnitudes of the coefficients on the equivalent non-porous walls, but are distributed differently due to the flow of the wind in and out of the canopy interior through the porous wall and roof surfaces. The derived net pressure coefficient results are summarised in tabular form and provide values for the design of large porous flat roofed canopies under wind load.

Abstract In order to allow for the numerical modeling of impacts for the design of live fire facilities commonly used by military and law enforcement personnel against next generation and environmentally friendly ammunition currently in development, constitutive models for novel target materials must be developed. Many existing facilities are constructed from AR500 steel, coupled with a layer of cellular rubber to reduce impact velocities and contain projectile fragments. High strain rate models, such as the commonly used Johnson-Cook constitutive model, are widely available to characterize AR500 steel, but calibrated models do not currently exist to characterize the cellular rubber. This project seeks to address this shortfall and provide a suitable material model for designers of these facilities in order to ensure the safety of users and the public. Appropriate constitutive models that account for the large strain, high strain rates, and temperature effects experienced during ballistic events and the porosity of the material were researched and a plan developed for future materials testing. Three suitable models were selected for further analysis — A Non-Linear Elastic Model described by Johnson in his work with polyurethane coupled with a Mie-Gruneisen equation of state to account for the porosity of the material, an Osborn-Hull model developed for use with crushable solids, and the Holmquist-Johnson-Cook Model commonly used for cementitious materials.

ABSTRACT: In this work, we have developed a methodology to model the stress evolution in cement plugs during hydration. The model begins with the slurry state of cement and calculates the water consumption and void creation over time as the hydration reactions progress. The void volume change due to chemical shrinkage is imported into a coupled mechanical model that calculates the pore pressure drop and the resulting change in stresses. The results of the proposed modelling methodology are verified using lab experiments from the literature. The results provide new insights in understanding cement behavior under lab and field conditions. Under most scenarios, cement’s pore pressure drops to saturation pressure of water which leads to partial evaporation of the remaining pore water. This pore pressure drop controls the radial stress change, according to the theory of poroelasticity. For a plug set under an initial pressure of 5 MPa, the radial stress drops to 1.6 MPa after 20 hours of curing. This stress drop can cause the cement to debond from the casing, if the fluid pressure above the plug exceeds the final radial stress. This methodology can be extended to annular cements and initial cement stress after placement can be readily calculated. 1. INTRODUCTION Zonal isolation in active and abandoned wells is paramount to ensure minimal fugitive methane emissions and to protect shallow freshwater aquifers. Wells penetrate different strata and can create a leakage pathway in case of a damaged cement sheath. This has been linked to methane emissions to the atmosphere (Schout et al., 2019), and aquifers (Osborn et al., 2011). Historically, oil and gas wells have been the main culprit in providing the leakage pathway for unwanted fluids. As more geothermal, energy, and carbon storage wells are drilled as part of the energy transition, zonal isolation challenges require more attention due to the long expected lifetime for these wells and unique operating conditions.

ABSTRACT Considering the low computational efficiency and accuracy of peridynamics (PD), a concurrent multiscale method coupling PD and finite element method (FEM) is proposed for modeling crack propagation of brittle rock under compressive load. In the coupling method, the fracture behavior is solved by the ordinary state-based peridynamics (OSBPD), while the elastic deformation of the rock mass is simulated by FEM. The implementation of the approach is as the following steps: first, a hybrid region is introduced in the framework to realize the strain energy equivalence between PD and FEM and eliminate the boundary effect. Then, the short-range force is utilized in PD to model the contact of the crack surface and prevent the particles from penetrating each other. In addition, the tangential force of the short-range force is introduced to simulate the friction sliding effect of the crack surface. Finally, the dynamic relaxation method is used to solve the displacement in the PD-FEM coupling model. The crack propagation of rock samples with a single pre-existing closed fracture under uniaxial compression is simulated by the PD-FEM coupling approach, and the numerical calculation results are in good agreement with the rule of the experimental results. The proposed coupling approach can capture the failure process of rocks under uniaxial compression and reduce the computational cost, simultaneously. INTRODUCTION The failure of rock can lead to structural damage, landslides, and rock bursts, which can endanger human life and property. Therefore, studying the fracture behavior of rock is essential for ensuring the safety of engineering projects. The law of crack growth in the brittle rock remains a great challenge in rock mechanics. In order to predict crack growth, many numerical methods have been developed. The numerical methods often used in the field of rock fracture mechanics include continuous methods and discontinuous methods. Common continuous methods include the extended finite element method (Eftekhari et al., 2016), the cohesive force element method (Zhou & Molinari, 2004), and the phase field method (Zhou et al., 2018). However, traditional continuum mechanics theory-based modeling makes it difficult to deal with complex crack propagation problems. The discrete element method (Cundall, 1971) is a commonly used technique in discontinuous methods to simulate fracture problems in rocks. However, the microscopic parameters of the discrete element method lack physical meaning and need to be calibrated through experiments.
