ABSTRACT: This research aimed to detect coagulase-positive Staphylococcus (CPS) directly in samples of artificially contaminated milk, using multiplex PCR (mPCR). Standard and isolated bacterial strains of S. aureus, S. epidermidis, S. hyicus, S. intermedius, Listeria monocytogenes and Escherichia coli species were used, evaluating the specificity and detection limit of mPCR, for artificially contaminated UHT milk. Primers specific for the nuc gene (NUC1-NUC2 were used for S. aureus, NUC3-NUC4 for S. hyicus and NUC5-NUC6 for S. intermedius). It was possible to detect the three target species by mPCR, directly from bovine whole milk, with adequate specificity and acceptable detention limit for identification of coagulase-positive Staphylococcus (CPS) in foods. The specificity was determined by the amplification of species-specific fragments, and the detection limit was assessed by the detection thresholds obtained for the three species (103 CFU mL-1). From these results, the mPCR described, with the proposed set of primers, has the potential for use in precise identification and differentiation between CPSs in milk samples.

The study examined Neglected and Underutilized Crop Species (NUCS) in two agro-ecologically diverse zones in Ghana with the view to highlighting the varieties, their important benefits and uses among rural households in the Upper Afram and Upper Dayi basins. Qualitative data was gathered from two hundred and seventy-one (271) respondents in six farming communities within the middle belt of Ghana through focused group discussions, interviews and questionnaires. A total of one hundred and twelve (112) NUCS were identified during the study. The diversity of NUCS in the study areas provide an indication of the existence of a valuable plant resource that can be exploited to help address food inadequacy challenges among rural households within the Upper Afram and Upper Dayi basins. Apart from their use as food, medicine, dyes, soap making, fuel wood and livestock feed, some NUCS found within the study basins play very important roles in the cultural practices and traditional ceremonies. These findings demonstrate the untapped potential of NUCS in addressing food and nutritional insecurity among rural households in the country. Institution of regular NUCS focused food fairs at the local (village), district and regional levels to promote local dishes prepared from the indigenous food crops; as well as increased NUCS related research are recommended. The DNA repair endonuclease EndoMS/NucS is highly conserved in Archaea and Actinobacteria. This enzyme is able to recognize and cleave dsDNA carrying a mismatched base pair, and its activity is enhanced by the interaction with the sliding clamp of the replisome. Today, EndoMS/NucS has been established as the key protein of a non-canonical mismatch repair (MMR) pathway, acting specifically in the repair of transitions and being essential for maintaining genome stability. Despite having some particularities, such as its lower activity on transversions and the inability to correct indels, EndoMS/NucS meets the main hallmarks of a MMR. Its absence leads to a hypermutator phenotype, a transition-biased mutational spectrum and an increase in homeologous recombination. Interestingly, polymorphic EndoMS/NucS variants with a possible effect in mutation rate have been detected in clinical isolates of the relevant actinobacterial pathogen Mycobacterium tuberculosis. Considering that MMR defects are often associated with the emergence of resistant bacteria, the existence of EndoMS/NucS-defective mutators could have an important role in the acquisition of antibiotic resistance in M. tuberculosis. Therefore, a further understanding of the EndoMS/NucS-mediated non-canonical MMR pathway may reveal new strategies to predict and fight drug resistance. This review is focused on the recent progress in NucS, with special emphasis on its effect on genome stability and evolvability in Actinobacteria.

The well-known mismatch repair (MMR) machinery, MutS/MutL, is absent in numerous Archaea and some Bacteria. Recent studies have shown that EndoMS/NucS has the ability to cleave double-stranded DNA (dsDNA) containing a mismatched base pair, which suggests a novel mismatch repair process. However, the recognition mechanism and the binding process of EndoMS/NucS in the MMR pathway remain unclear. In this study, we investigate the binding dynamics of EndoMS/NucS to mismatched dsDNA and its energy as a function of the angle between the two C-terminal domains of EndoMS/NucS, through molecular docking and extensive molecular dynamics (MD) simulations. It is found that there exists a half-open transition state corresponding to an energy barrier (at an activation angle of approximately 80 ∘ ) between the open state and the closed state, according to the energy curve. When the angle is larger than the activation angle, the C-terminal domains can move freely and tend to change to the open state (local energy minimum). Otherwise, the C-terminal domains will interact with the mismatched dsDNA directly and converge to the closed state at the global energy minimum. As such, this two-state system enables the exposed N-terminal domains of EndoMS/NucS to recognize mismatched dsDNA during the open state and then stabilize the binding of the C-terminal domains of EndoMS/NucS to the mismatched dsDNA during the closed state. We also investigate how the EndoMS/NucS recognizes and binds to mismatched dsDNA, as well as the effects of K + ions. The results provide insights into the recognition and binding mechanisms of EndoMS/NucS to mismatched dsDNA in the MMR pathway.

Meeting food and nutritional security needs for a growing population is a global sustainability challenge due to the heavy reliance on a few cultivated crops for dietary requirements across the world. To ensure local food security, it is imperative to diversify dietary options with locally available, neglected, and underutilized crops (NUCs) with nutritional and biocultural significance. In this context, the present study aims to explore the role of NUCs for nutritional, ethnomedicinal, and agricultural relevance in two districts of eastern Uttar Pradesh, i.e., Mirzapur and Sonebhadra. Extensive field surveys were conducted in the study sites, and a total of 445 local respondents were interviewed based on structured questionnaires for calculating ethnobotanical indices, i.e., relative frequency of citation (RFC), frequency of citation (FC), use report (UR), and cultural importance index (CI) of NUCs. The study identified 116 NUCs belonging to 55 families and 103 genera. All reported NUCs had medicinal value; 55 were edible and used as food; and 41 had agricultural significance. Leaves were the most commonly used plant parts for medicinal purposes, followed by roots and stems, whereas flowers were the least commonly used parts. NUCs were used by the locals for their medicinal properties to treat various ailments, such as skin and eye problems, headaches, and liver problems. They were administered as decoction, paste, vapor inhalation, fruit juice, and poultice. The RFC, FC, UR, and CI values of the NUCs were in the range of 12 to 365, 0.03 to 0.82, 12 to 394, and 0.03 to 0.89, respectively. Fruits were the most commonly consumed part, followed by leaves, tubers, pods, and aerial bulbs, whereas the mode of consumption was stir-fry, soup, vegetables, salad, or in raw form. NUCs were bestowed with essential macro- and micronutrients and were found in the range of Ca (3.79–1147.3), K (2.6–1600.3), Mg (0.8–468.0), Na (0.4–270.8), P (1.15–305), Fe (0.1–327.6), Zn (0.1–84.6), Cu (0.047–33.3), Mn (0.1–62.3) mg/100 g, and vitamins like ascorbic acid (0.04–1561.1), thiamine (0.041–2.4), and β-Carotene (0.2–93.6) mg/100 g. These NUCs were cultivated in different settings, such as kitchen gardens, backyard gardens, border crops, and sometimes agricultural fields. The current study reveals the rich diversity and varied use of these NUCs with respect to their ethnomedicinal, nutritional, and agricultural relevance. Sustainable utilization with large-scale cultivation of promising NUCs can lead to local food security and the subsequent attainment of the associated UN Sustainable Development Goals.

Hepatitis B virus (HBV) is a prevalent underlying disease, leading to liver transplantation (LT) for both decompensated cirrhosis and hepatocellular carcinoma (HCC). The hepatitis delta virus (HDV) affects approximately 5–10% of HBsAg carriers, accelerating the progression of liver injury and HCC. The initial introduction of HBV immunoglobulins (HBIG), and then of nucleos(t)ide analogues (NUCs), considerably improved the survival of HBV/HDV patients post-transplantation, as they helped prevent re-infection of the graft and recurrence of liver disease. Combination therapy with HBIG and NUCs is the primary post-transplant prophylaxis strategy in patients transplanted for HBV- and HDV-related liver disease. However, monotherapy with high-barrier NUCs, such as entecavir and tenofovir, is safe and also effective in some individuals who are at low risk of HBV reactivation. To address the problems of organ shortage, last-generation NUCs have facilitated the use of anti-HBc and HBsAg-positive grafts to meet the ever-increasing demand for grafts.

BackgroundImmunotherapy with immune checkpoint inhibitor (ICI) is a promising treatment for unresectable hepatocellular carcinoma (HCC). However, whether ICIs would have the risk of hepatitis B virus (HBV) reactivation and the necessary of nucleos(t)ide analogs (NUCs) prophylaxis are still unclear. We aimed to investigate the role of NUCs prophylaxis in HBV-infected patients who underwent ICIs treatment.MethodsThe study was a retrospective prospective design to review and follow-up consecutive 62 patients with chronic hepatitis B or resolved HBV infection who had received ICIs treatment for the unresectable HCC. Of them, 60 patients with documented baseline serum HBV DNA value were classified into three categories according to the baseline HBV viral load and the status of antiviral therapy before ICI treatment. The clinical status, including tumor response, viral kinetics and liver function, was recorded and investigated.ResultsNo HBV reactivation occurred in the 35 patients with HBV DNA ≤100 IU/mL on NUCs therapy. Of the 19 patients with HBV DNA >100 IU/mL who started NUCs simultaneously with ICI treatment, none encountered HBV reactivation during the immunotherapy. Of the six HBV patients without NUCs treatment, three had a greater than 1 log decrease in HBV viral load, and one maintained his serum HBV DNA in undetectable status during ICI treatment. Eventually, one out of six experienced HBV reactivation after 9 weeks of ICI treatment.ConclusionNo patients on antiviral therapy developed HBV reactivation, and one out of six not receiving antiviral therapy had HBV reactivation. HBV viral load higher than 100 IU/mL is safe and not a contraindication for ICI treatment for HCC, if NUCs can be coadministrated.

Branched DNA structures that occur during DNA repair and recombination must be efficiently processed by structure-specific endonucleases in order to avoid cell death. In the present paper, we summarize our screen for new interaction partners for the archaeal replication clamp that led to the functional characterization of a novel endonuclease family, dubbed NucS. Structural analyses of Pyrococcus abyssi NucS revealed an unexpected binding site for ssDNA (single-stranded DNA) that directs, together with the replication clamp, the nuclease activity of this protein towards ssDNA–dsDNA (double-stranded DNA) junctions. Our studies suggest that understanding the detailed architecture and dynamic behaviour of the NucS (nuclease specific for ssDNA)–PCNA (proliferating-cell nuclear antigen) complex with DNA will be crucial for identification of its physiologically relevant activities.

The management of chronic hepatitis B involves various therapeutic approaches, including nucleotide analogs (NUCs) and pegylated-interferon alpha (peg-IFN), either in isolation or in combination. Reverse transcriptase enzyme is competitively inhibited by NUCs, which effectively suppresses HBV replication and lowers viral load. Concerning their cost-effectiveness, high response rates, low side effects, and oral administration, NUCs are recommended. Prolonged use, particularly of NUCs with a low genetic barrier or as monotherapy, can, however, lead to resistance, long-term safety issues, and the need for ongoing treatment. Physicians and other healthcare professionals are extremely concerned about the emergence of resistance and possible safety concerns related to the long-term use of NUCs. Moreover, the requirement for continuous therapy presents notable obstacles concerning patient adherence, distribution of healthcare resources, and overall economic viability. To clarify these problems and direct the creation of more potent and long-lasting treatment plans for chronic hepatitis B, urgent research is required. Hepatitis B surface antigen (HBsAg) detection is frequently accomplished via the use of the Chemiluminescent Microparticle Immunoassay (CMIA), which is a crucial early serologic marker for screening and diagnosis. Polymerase chain reaction (PCR) molecular testing is employed to confirm the presence of HBsAg. Polymerase Chain Reaction (PCR) was the technique we utilized to verify the outcomes. Twenty-eight of the HBsAg-positive patients at W.Z. Johannes Kupang Hospital had positive PCR results, highlighting the significance of molecular confirmation. The results of this study emphasize the value of precise HBsAg testing and the supplementary function of molecular confirmation in the treatment of patients with chronic hepatitis B. Furthermore, it clarifies the current therapeutic approaches applied to this patient population, highlighting the necessity of customized therapeutic approaches based on each patient's unique profile and potential complications.

Les Streptomyces, bactéries ubiquistes des sols forestiers, présentent une grande plasticité génomique. Ces bactéries possèdent un chromosome linéaire hautement compartimenté avec, en son centre, une région conservée au travers du genre, bordée par des extrémités ou bras chromosomiques particulièrement variables et sujets à la recombinaison. La réparation des cassures double-brin (DSB), dommage le plus délétère pour une cellule vivante, constitue un moteur majeur de la plasticité et de l'évolution du génome chez les Streptomyces. Une source potentielle de DSB a été découverte récemment : l'endonucléase NucS. Cette dernière joue un rôle clé au sein du système de correction des mésappariements (MMR) non-canonique, présent chez les archées et plus sporadiquement chez les bactéries avec les actinobactéries. L'objectif de ce travail est de comprendre si l'activité de NucS est impliquée dans la stimulation de la recombinaison chez les Streptomyces. Nos investigations ont révélé que la déplétion en NucS entraine une augmentation significative du taux de mutation et un phénotype colonial marqué. Les tests d'activité in vitro montrent que NucS génère une DSB au site des mésappariements G/T, G/G et T/T. Son activité est augmentée en présence du facteur de processivité de la réplication β-clamp suggérant que NucS cible les mésappariements post-réplicatifs. Une expérience d'accumulation de mutations sur le long-terme (60 cycles de croissance) montre que NucS corrige plus favorablement les mésappariements conduisant à une transition qu'à une transversion. Une augmentation graduelle du nombre de transitions depuis l'origine de réplication vers les extrémités du chromosome a été caractérisée. Ce résultat original révèle que NucS est davantage sollicitée dans les régions terminales, et pourrait favoriser la recombinaison en générant des DSB. L'augmentation de la fréquence de réarrangements dans les régions terminales dans un contexte où la fréquence des mésappariements réplicatifs est accrue renforce cette hypothèse. En effet, les mésappariements sont le substrat de NucS et la cassure double brin résultant de son activité induirait la recombinaison. Ces résultats ouvrent la voie de la compréhension de ce mécanisme paradoxal de maintien de l'intégrité du génome ; pourquoi casser le génome pour mieux le réparer ? La diversification génétique résultant de la plasticité du génome pourrait justifier de la sélection d'un tel mécanisme de réparation. Enfin, la diversification du métabolisme spécialisé chez ces organismes du sol revêt un intérêt majeur pour leur adaptation à l'écosystème, et aussi pour l'identification de nouvelles molécules d'intérêt biotechnologique
Streptomyces are ubiquitous bacteria in forest soils known for their remarkable genomic plasticity. These bacteria possess a highly compartmentalized linear chromosome, with a central region shared across the genus. This central region is bordered by variable chromosomal ends or arms that are prone to recombination. The repair of double-strand breaks (DSB), the most harmful cellular damage, is a major driver of genome plasticity and genome evolution in Streptomyces. Recently, a potential source of DSB has been identified: the endonuclease NucS, which plays a pivotal role in the non-canonical mismatch repair system (MMR) present in archaea and sporadically in bacteria, particularly in actinobacteria. The objective of this study is to determine whether NucS activity is involved in stimulating recombination in Streptomyces. Our investigations unveil that NucS depletion results in a significant increase of the mutation rate and in a marked colonial phenotype. In vitro activity tests showed that NucS generates DSB at G/T, G/G, and T/T mismatches. Its activity is enhanced in the presence of the replication processivity factor β-clamp, indicating that NucS targets post-replicative mismatches. A long-term mutation accumulation experiment conducted over 60 sporulation cycles have demonstrated that NucS is more prone to correct mismatches leading to transitions than transversions. A crescent-increase in the number of transitions from the replication origin towards the chromosome ends is observed. This original finding suggests that NucS is more active in terminal regions and could promote recombination by generating DSB. The increased frequency of rearrangements in the terminal regions in a mutant exhibiting an elevated frequency of replicative mismatches supports this hypothesis. These findings advance our understanding of this paradoxical mechanism for maintaining genome integrity: why break the genome in order to repair it better? The genetic diversification arising from genome plasticity could explain the selection of such a repair mechanism. Finally, the diversification of specialized metabolism in these soil-dwelling bacteria is of great interest both for their adaptation to the ecosystem and for the identification of new molecules with biotechnological interets

Les enzymes de réparation de l'ADN sont des facteurs essentiels pour assurer l'intégrité du génome. Ainsi, la compréhension de la dynamique de leurs mécanismes d'interaction est capitale. NucS est une nucléase récemment découverte chez l'archée Pyrococcus abyssi, qui interagit avec des structures branchées de l'ADN à simple brin libre, appelées flaps. Sa caractérisation biochimique a mis en évidence une affinité nanomolaire pour l'ADN simple brin, avec l'existence de deux sites de liaison (site I et II) ainsi qu'une activité bidirectionnelle caractérisée par la capacité à cliver les flaps 5' et 3'. Les mécanismes qui sont à l'origine de cette activité, notamment ses modalités de chargement, de localisation et de dissociation, restent toutefois peu connus. Afin de sonder la dynamique des interactions de NucS avec son substrat, nous avons mis en place un microscope à illumination en onde évanescente permettant la détection et le suivi des événements d'interaction individuels entre la protéine marquée avec un fluorophore et un substrat d'ADN attaché à la surface. Nous avons ensuite développé une nouvelle technique de colocalisation qui permet de positionner une protéine individuelle par rapport à son substrat avec une précision de 50 nm pour des durées arbitraires. Nous avons alors étudié la cinétique d'association et de dissociation des complexes NucS-ADN individuels afin de résoudre les mécanismes qui régulent l'activité de l'enzyme dans différentes conditions. Dans des conditions où le clivage est inhibé, nous avons montré que l'association était dépendante du site I, bidirectionnelle et symétrique pour les flaps 3' et 5' et que la dissociation était orientée avec une affinité supérieure pour les flap 5'. Nous avons également montré que la dissociation suivait une cinétique du premier ordre indépendante de la diffusion de NucS sur le flap. Ceci indique que, dans ces conditions non clivantes, NucS s'associe et/ou se dissocie à des positions arbitraires sur le flap. Par la suite, nous avons étudié la cinétique d'association et de dissociation du complexe NucS-ADN à 45 ◦ C en présence du cofacteur de la réparation PCNA. Nous avons démontré que PCNA augmente la vitesse d'association de NucS avec les substrats 3' et 5'. Ceci indique que PCNA agit comme une plateforme qui facilite la reconnaissance de la lésion, avec un chargement ciblé de NucS à la jonction. De plus, nous avons montré que PCNA déstabilise le complexe NucS-ADN, vraisemblablement en exerçant une force pour plier l'ADN afin d'amener l'extrémité du flap vers le site II. Dans le cas de flaps 5', nous avons montré que la dissociation suit un mécanisme en deux étapes, qui est indépendant de la longueur du flap. Ceci nous a permis de proposer un modèle où NucS se charge par son site I directement à la jonction, courbe le substrat pour capturer l'extrémité simple brin par le site II pour le cliver. Dans le cas d'un flap 3', nous avons montré que la dissociation suit un mécanisme du premier ordre ce qui est probablement dû à la rapidité de la seconde étape dans le mécanisme proposé pour les flaps 5'. Nos résultats constituent ainsi une nouvelle contribution à la caractérisation intramoléculaire des interactions NucS-ADN. Les méthodes que nous avons développées constituent en outre un moyen original pour sonder la cinétique des interactions entre biomolécules et pourront être appliquées à de multiples systèmes.

NTNU is aiming to build and launch a small student satellite compliant with thedouble CubeSat standard, by 2014. The NTNU Test Satellite (NUTS) will carrytwo radio tranceivers and a beacon transmitter, all located in the VHF and UHFamateur bands. The goal of this thesis was to build the whole antenna systems forthe spacecraft.Turnstile antennas were chosen both for UHF and VHF, since they yield thehighest received signal strength on ground throughout the whole pass of the satel-lite. In order to reach this conclusion, a study of how the choice of spacecraftantennas dynamically influence the link margin was carried out. Furthermore, theantennas was made by measuring tape, and is to be wrapped around the satelliteuntil deployment. This is for the satellite to be within the size constraints of adouble CubeSat during launch. The measuring tape will be tightened to the satel-lites body by nylon thread, which will be burnt off by Nichrome-wire to release themeasuring tape when the satellite is in-orbit. The measuring tape is mounted inbetween two circuit boards constituting the internal part of the antenna module.The feeding network is encapsulated by these two circuit boards. The module ismade such that the antenna complies with the CubeSat standard, and such that itcan be integrated with other modules on-board the satellite, such as the camera.Feeding the antennas turned out more difficult than first anticipated. Whereasconventional feeding networks for turnstile antennas typically comprises baluns andarrangements of coaxial cable comparable to the wavelength, such solutions are notfeasible within the satellite due to size constraints. As a result a lumped feedingnetwork was designed, that could be mounted on one of the circuit boards.Because of the many reactive components in the feeding network, it becamenecessary to determine how deviations in component values affect the performanceof the circuit. Statistical simulations was performed, and the deviations may causean unbalance between the amplitudes and phases of the outputs. But this resultsin less than half a dB additional losses in the link budget.A refinement of electromagnetic antenna simulations from previous work wasalso carried out. With this, the performance of the antennas was verified, andtheir lengths were optimized to yield the best possible match. The VHF antenna isnear-ideal both in terms of pattern and matching. The pattern of the UHF antennais somewhat distorted, but it is still considered to be the best suited antenna ofthose considered. The matching of the UHF antenna could have been better, butis considered acceptable. Moreover, the mutual coupling between the VHF andUHF antennas were studied, with the conclusion that the antennas may very wellco-exist on-board the satellite.Finally, both the feeding network and the VHF antenna were successfully builtand tested. The feeding network had an insertion loss of less than 2 dB, whereas thepattern of the antenna was measured to be near-ideal. Unfortunately, a fault hasoccurred on the UHF feeding network, which also prohibits the pattern for the UHFantenna to be measured. Nonetheless, all the parts constituting it has been built,and all the necessary simulations are done. Previous and future troubleshooting ofthe feeding network is also discussed.

La hiérarchie mémoire subit une pression qui ne cesse de croître. Cette pression a eu pour origine la montée en fréquence des processeurs. Cependant, maintenant que la fréquence stagne autour de 3 GHz, le nombre de cœurs d'exécution et donc le nombre de processus s'exécutant simultanément augmentent à leur tour. La hiérarchie mémoire subit alors un nombre croissant de requêtes, conduisant à la saturation de sa bande passante. Les travaux présentés dans cette thèse montrent que la hiérarchie mémoire est souvent utilisée pour transporter des blocs de données totalement nuls. Ces blocs de valeur triviale se trouvent particulièrement nombreux au dernier niveau de cache et au niveau de la mémoire principale. Nous proposons dans ce document d'utiliser un cache spécialisé dans la gestion de ces blocs nuls, le Zero-Content Augmented Cache. Ce dernier est composé d'un cache traditionnel et d'un cache dédié aux blocs nuls. Cette proposition permet à la fois d'augmenter les performances globales du système et de réduire significativement la bande passante mémoire utilisée. Dans ce document, nous proposons également une architecture de mémoire compressée utilisant la présence de blocs nuls, la Decoupled Zero-Compressed Memory. Cette mémoire permet de stocker un working-set plus grand que la taille de la mémoire physique, et donc de réduire significativement le nombre d'accès aux périphériques de stockage de masse.

The first topic considers the boundary terms in a Hamiltonian approach to general relativity. We show that if a timelike boundary of a spacetime is not orthogonal to the constant time hypersurfaces, then an additional 'tilting' term must be included in the Hamiltonian. The second topic considers the actions and thermodynamics of spacetimes with 'nut charge'. We provide a consistent framework for calculating the action of these spacetimes by requiring a background subtraction based on a metric that satisfies the same asymptotic conditions. Using this formalism we then calculate the action for metrics with a U(1) isometry, using both dimensional reduction and a Hamiltonian decomposition. In the former case we obtain a formula in terms of quantities defined on the fixed point set of the isometry, including explicit nut charge contributions. In the latter case the action depends on a surface integral at infinity, as well as on surface integrals and areas of the two dimensional obstructions to foliations based on the isometry - the well-known 'bolts' of the isometry and the novel 'Misner strings' that arise from the nut charge. Using the Hamiltonian result we derive a new formula for the entropy of spacetimes that differs from the one-quarter area law previously known for black holes. The third topic considers the AdS/CFT correspondence that hypothesizes a duality between a bulk theory on an asymptotically anti-de Sitter (AdS) space and a conformal field theory (CFT) on its boundary. We test this hypothesis using two different spacetimes. First, we examine rotating AdS black holes in 3, 4 and 5 dimensions whose boundary is a rotating Einstein universe of one dimension less. We find in the high temperature limit as the rotational velocity of the Einstein universe approaches the speed of light that the partition functions of the AdS spacetimes qualitatively agree with the partition functions of free scalar fields on the boundary. Second, we add nut charge to AdS space and, using the methodology derived in the second topic, we calculate the partition function. Unfortunately the boundary is a squashed three sphere and we are unable to provide a useful approximation to the partition function of a conformal field theory on this surface.

Le nud de trefle est l'archetype d'un objet topologiquement chiral, c'est a dire que l'on ne peut interconvertir ses deux enantiomeres topologiques par une deformation continue sans casser de liaisons. Au cours de ce travail, un nud de trefle moleculaire a ete dedouble et sa configuration absolue a ete determinee par diffraction des rayons x. D'autre part, il a ete montre, par l'utilisation de la polarisation circulaire de l'emission, (pce) qu'un nud de cuivre(i) optiquement pur peut etre utilise en tant que piegeur d'energie enantioselectif. Face a des complexes chiraux de lanthanides, un faible facteur d'enantioselectivite (de l'ordre de 0,5 a 1%) a pu etre mesure. Dans une seconde partie, la reaction de cyclisation par metathese des olefines a ete utilisee avec succes dans la synthese de nouveaux nuds moleculaires. Il a ainsi ete possible de preparer un nud construit autour d'une double helice dinucleaire de cuivre(i) avec 74% de rendement, ameliorant d'un facteur trois le rendement de l'etape de cyclisation. Suivant la meme strategie le premier nud synthetise autour de fragments fer(ii) bisterpyridine a ete obtenu avec 20% de rendement. Ce resultat montre que l'utilisation d'un autre metal assembleur que le cuivre(i) est possible dans la preparation de molecules topologiquement non triviales.

NTNU test satellitt planlegger å inkludere et IR camera som hovednyttelast, med formål å ta bilde av tyngdebølger i atmosfæren. Nyttelastmodulen i satellitten må utføre både bildebehandling og bildekomprimering for å oppnå bilder som både har god nok SNR, men samtidig er små nok til å kunne sende til en mottakerstajon på jorda i en fornuftig tid. Dette setter noen krav til nyttelastmodulen. I tillegg til å måtte hente bilder fra kameraet og lagre disse må den behandle bilder etterhvert som de kommer inn slik at man unngår å bruke mye lagringsplass og ressurser på å lagre bildeserier før de behandles.Flere kameragrensesnitt ble undersøkt og evaluert med tanke på bruk med FPGA eller mikrokontroller. Det ble funnet at Camera Link er det foretrukne grensesnittet når man bruker FPGA, mens direkte integrasjon var best når man bruker mikrokontroller.Nyttelastemodulen må også kunne ta ordre fra, og sende bildedata til, andre deler av satellitten. Det ble funnet at det holder å ha nyttelastmodulen som en I2C slave på bakplansbussen.FPGA og mikrokontroller ble sammenlignet i forhold til deres egnethet som prosesseringsenheten i nyttelastmodulen, og det ble funnet at FPGA generelt var mer egnet til denne typen oppgave.Det ble til slutt gjort et design av en eksempelimplementering, som og et FPGA design ble skrevet i verilog.
