AbstractA new way of structuring and interpretation of multiple domain matrix is proposed as the basis for categorisation of design parameter relations complexity. Depending on the kind and the degree of coupling of the parameters, the developed methodology activates the appropriate coloured Petri net (CPN) models for semi-automatic support of communication between the members of the design team. The proposed extension of MDM combined with CPN is a novel approach to predicting and managing communication patterns necessary during teamwork coordination on critical interfaces between product components.

Prothymosin alpha (ProTα) was discovered to be a necrosis inhibitor from the conditioned medium of a primary culture of rat cortical neurons under starved conditions. This protein carries out a neuronal cell-death-mode switch from necrosis to apoptosis, which is, in turn, suppressed by a variety of neurotrophic factors (NTFs). This type of NTF-assisted survival action of ProTα is reproduced in cerebral and retinal ischemia–reperfusion models. Further studies that used a retinal ischemia–reperfusion model revealed that ProTα protects retinal cells via ecto-F1 ATPase coupled with the Gi-coupled P2Y12 receptor and Toll-like receptor 4 (TLR4)/MD2 coupled with a Toll–IL-1 receptor domain-containing adaptor inducing IFN-β (TRIF). In cerebral ischemia–reperfusion models, ProTα has additional survival mechanisms via an inhibition of matrix metalloproteases in microglia and vascular endothelial cells. Heterozygous or conditional ProTα knockout mice show phenotypes of anxiety, memory learning impairment, and a loss of neurogenesis. There are many reports that ProTα has multiple intracellular functions for cell survival and proliferation through a variety of protein–protein interactions. Overall, it is suggested that ProTα plays a key role as a brain guardian against ischemia stress through a cell-death-mode switch assisted by NTFs and a role of neurogenesis.

The in vivo tumor microenvironment is a complex niche that includes heterogeneous physical structures, unique biochemical gradients and multiple cell interactions. Its high-fidelity in vitro reconstruction is of fundamental importance to improve current understandings of cell behavior, efficacy predictions and drug safety. In this study, we have developed a high-throughput biochip with hundreds of composite extracellular matrix (ECM) microchambers to co-culture invasive breast cancer cells (MDA-MB-231-RFP) and normal breast epithelial cells (MCF-10A-GFP). The composite ECM is composed of type I collagen and Matrigel which provides a heterogeneous microenvironment that is similar to that of in vivo cell growth. Additionally, the growth factors and drug gradients that involve human epidermal growth factor (EGF), discoidin domain receptor 1 (DDR1) inhibitor 7rh and matrix metalloproteinase inhibitor batimastat allow for the mimicking of the complex in vivo biochemical microenvironment to investigate their effect on the spatial-temporal dynamics of cell growth. Our results demonstrate that the MDA-MB-231-RFP cells and MCF-10A-GFP cells exhibit different spatial proliferation behaviors under the combination of growth factors and drugs. Basing on the experimental data, we have also developed a cellular automata (CA) model that incorporated drug diffusion to describe the experimental phenomenon, as well as employed Shannon entropy (SE) to explore the effect of the drug diffusion coefficient on the spatial-temporal dynamics of cell growth. The results indicate that the uniform cell growth is related to the drug diffusion coefficient, which reveals that the pore size of the ECM plays a key role in the formation of complex biochemical gradients. Therefore, our integrated, biomimetic and high-throughput co-culture platforms, as well as the computational model can be used as an effective tool for investigating cancer pathogenesis and drug development.

BackgroundBintrafusp alfa is a first-in-class bifunctional fusion protein composed of the extracellular domain of the TGF-βRII receptor fused to a human IgG1 antibody blocking PD-L1. The TGF-βRII moiety of bintrafusp alfa functions as a ”trap” to sequester active TGF-β but does not block TGF-β release from its latent form. Multiple mechanisms lead to the release of active TGF-β. Integrins control local activation of latent TGF-β stored in the extracellular matrix and cell-surface reservoirs in the tumor microenvironment (TME). Alpha v integrin mRNA expression is correlated with multiple TGF-β gene signatures. It has been shown that αvβ8 integrin mediates TGF-β activation without releasing it from the latent TGF-β complex, suggesting that the TGF-βRII moiety of bintrafusp alfa may be unable to sequester TGF-β activated by αvβ8 integrin. Therefore, we hypothesize that combining abituzumab, a pan–αv integrin antibody, with bintrafusp alfa may lead to enhanced suppression of TGF-β signaling.MethodsThe expression of αv and β6 integrin mRNA was determined by RNA sequencing of triple-negative breast cancer (TNBC) tumor samples from a phase 1 clinical trial of bintrafusp alfa and correlated with patient response to bintrafusp alfa. The combination of bintrafusp alfa and abituzumab was investigated in vitro and in vivo in a TGF-β–dependent human tumor model, Detroit 562. In this study, CellTiter-Glo 2.0 Assay measured cell proliferation in vitro and enzyme-linked immunosorbent assay measured the level of latency-associated protein (LAP). A TGF-β reporter cell line MDA-MB-231 measured the level of active TGF-β. Antitumor activity in vivo was evaluated via tumor growth of Detroit 562 xenograft model in SCID mice.ResultsIn TNBC, increased expression of αv and β6 integrin mRNA was associated with poor response to bintrafusp alfa, suggesting that TGF-β activated by αv integrin may not be blocked by bintrafusp alfa. In Detroit 562 cells, abituzumab increased LAP levels in the cell culture medium, confirming modulation of the TGF-β pathway. As a result, the amount of active TGF-β released into culture medium was reduced by abituzumab. In vitro, both abituzumab and bintrafusp alfa suppressed Detroit 562 cell proliferation, and the combination suppressed cell proliferation further. In vivo, the combination led to increased tumor growth inhibition of Detroit 562 xenograft tumors relative to either monotherapy, further supporting the potential of this combination.ConclusionsCollectively, these preclinical findings support clinical development of bintrafusp alfa and abituzumab combination therapy to maximally suppress TGF-β signaling in the TME.AcknowledgementsWe thank George Locke for his analysis of the RNAseq data.Ethics ApprovalThis study was approved by the Institutional Animal Care and Use Committee at EMD Serono, Inc.; approval number [17–008].

PurposeScheduling in information-driven design phase of construction projects is challenging due to multiple entity types (teams, components, deliverables, activities or parameters) and their dependencies/linkages. Established techniques such as dependency structure matrix (DSM), beeline diagramming method (BDM), multiple domain matrix (MDM), etc. have been independently utilized in past to model information dependencies/linkages and associated iteration. However, there has not been a holistic solution yet for scheduling multiple entity types and their relationships. Hence, an integrated solution needs to be developed that schedules information-driven projects accurately.Design/methodology/approachA case study data collection approach is utilized. With data from two projects, i.e. hostel design and highway design, a BDM–MDM integrated solution was developed and applied to the same. Feedback from experts was obtained for refinements.FindingsThe proposed solution is efficient for scheduling multiple entity types and their information dependencies/linkages.Practical implicationsThe proposed integrated solution enables the project participants to schedule information-driven projects systematically. Application to two distinct design cases emphasizes that the concept is generic and can be applied to any information-driven project with multiple entity types.Originality/valueThe BDM–MDM integrated solution concept is investigated for scheduling multiple entity types in any information-driven projects. This study also explored the terminologies such as multiple entity types and information-driven scheduling.

AbstractBlockchain has the potential to improve sustainable food security due to its unique features like traceability, decentralized and immutable database, and smart contract mechanisms. However, blockchain technology is still in the early stages of adoption in particular in agricultural applications. In this context, this article aims to identify blockchain drivers to achieve sustainable food security in the Indian context and model them using an integrated MCDM (Multiple Criteria Decision Making) approach. The blockchain adoption drivers are identified through an exhaustive literature review and opinions from domain experts from industry, academia, and Agriculture Supply Chain (ASC) stakeholders. Subsequently, the integrated MCDM approach is developed by combining Total Interpretive Structural Modelling (TISM) and Fuzzy Decision-Making Trial and Evaluation Laboratory (DEMATEL), which does not only investigate the interrelation between the identified constructs and builds hierarchy but also determines the intensity of the causal interrelationships. At a later stage, Fuzzy Cross-Impact Matrix Multiplication Applied to Classification (MICMAC) is used to cluster the identified drivers to evaluate the importance of each driver. The results reveal that Traceability, Real-time information availability to agro-stakeholder, and Decentralized and immutable database are the most significant drivers. Policymakers, governmental organizations and other relevant stakeholders may utilize the information about the interrelationship between these drivers and their influential power, to frame suitable strategies for enhancing the adoption rate of blockchain in the Indian ASC.

Purpose Khadi fabrics are known for their unique comfort properties which are attributed to their unique structural and functional properties. For getting optimal comfort from a collection of available Khadi fabrics, further exploration is needed. Ranking the Khadi fabrics from a competitive lot for optimal comfort is a challenging job, which has not been addressed so far by any researcher. The purpose of this study is to present one such selection problem using the multi-criteria decision-making (MCDM) technique, a popular branch of operations research, which can handle almost any decision problem involving a finite number of alternatives and multiple decision criteria. Design/methodology/approach Two widely popular methods/exponents of MCDM, namely, analytic hierarchy process (AHP) and multiplicative analytic hierarchy process (MAHP) have been deployed in this study for ranking a competitive lot of 15 Khadi fabrics and selecting the best alternative for optimal summer comfort based on three comfort attributes, namely, drape coefficient, thermal insulation value and air permeability. Findings Both the approaches yield a similar ranking pattern with Spearman’s rank correlation coefficient of 0.9857, Khadi fabric K1 achieving Rank 1 (best in terms of optimal comfort) and sample K6 acquiring Rank 15 (worst choice). Two-phase sensitivity analyses were performed subsequently to demonstrate the stability of the two approaches: sensitivity analysis by changing weightage levels of the criteria and sensitivity analysis in dynamic decision conditions by changing the elements of the initial decision matrix. During sensitivity analyses, no occurrence of rank reversal is observed for the best and worst alternatives in either of the two approaches. This corroborates the robustness of the two models. Practical implications Khadi fabrics are widely acclaimed for their intrinsic comfort properties for both summer and winter. Although the popularity of Khadi fabrics is increasing day by day, this domain is under-researched, and hence, needs to be explored further. The present approach demonstrates how the MCDM technique can serve as a useful tool for ranking the available Khadi fabrics in terms of optimal comfort in summer. The same approach can be extended to other domains of the textile industry, in general, as well. Originality/value This study is the first-ever theoretical approach/research on the selection of Khadi fabrics for optimal summer comfort using the MCDM tool. Another novelty of the present study is that the efficacy of AHP and MAHP approaches, in this study, has been validated through a two-phase sensitivity analysis. This validation part has been ignored in most of the hitherto published applications of AHP and MAHP in other domains.

Design Structure Matrices (DSM) and Domain Mapping Matrices (DMM) are commonly used to model and analyze the relationships within one domain (DSM) or between two domains (DMM). Being assembled into one larger square matrix, having DSMs on its diagonal and DMMs in all other fields, a so-called Multiple Domain Matrix (MDM) is formed. When relating two domains using a DMM, a problem arises when the nature of one individual relationship between the two domains is to be described. Usually, this is modeled by annotating each relationship with the additional information, much like comments in spreadsheet software. This, however, is yet impossible if the relationships should be in matrix notation to allow for algorithmic matrix analyses. Equally, this way, the annotations are not accessible as elements of another matrix, e.g. as DSM. This paper suggests a generic principle to solve the described problem in a way consistent with the matrix methodology. It proposes an approach using MDM and is thereby able to unambiguously provide the nature of each relationship between the elements of two domains. As a DSM is a mere case of a DMM having two identical domains, the approach proposed can equally be used to enrich the relationships within a DSM.

Model-Based Systems Engineering (MBSE) currently lacks a formalized way of managing modeling consistency. This paper presents a new methodology for semantic consistency management in MBSE. At its core, a matrix structure which extends the Multiple Domain Matrix (MDM) and Engineering Systems Matrix (ESM) is used to organize semantic relations between modeling elements created by using various modeling languages and tools, and to represent how they change over time. In addition, this paper describes how the new matrix can be automatically maintained by a plug-in module which manages the bidirectional information transformation between the model and the matrix. We show that the utilization of our methodology has potential in avoiding ambiguous namings in models, enhancing the traceability of modeling elements as well as reducing manual work. A real world engineering use case with air-conditioning is used to show the utilization of our methodology.

In this paper, we present a pilot study to show the first proof-of-principle of the recently developed Elephant Specification Language (ESL). The Prinses Marijke locks in the Netherlands are due for a major renovation. To effectively organize the renovation project, engineers need insight into the network of dependencies between components that are due for renewal and components that remain, to ensure that the replacement parts will fit and function properly in the existing structure. ESL is used to write a structured multi-level function specification for the Prinses Marijke locks. From this specification, multi-domain matrix (MDM) models of the system architecture are automatically derived at multiple levels of granularity. These models show dependencies between components, between functions, and combinations thereof. The MDM models are used to gain insight in the system architecture, to visualize which parts of the lock system are affected by the renovation, and to determine which dependencies have to be accounted for during the renovation process. This information is required to effectively organize the renovation project.

Design Structure Matrices (DSMs) and Domain Mapping Matrices (DMMs) are generally used by designers for dynamic optimization of engineering design processes and products. Both methodologies help producing valuable results; however, they are lacking a holistic view onto the processes and products. Dependencies that span multiple product development domains can therefore not be recognized with isolated DSM or DMM analysis. In this paper, we present an integrative approach that combines DSMs and DMMs to obtain the Multiple Design Structure Matrix (MDSM). This methodology offers the possibility to analyze multiple product development domains using one coherent matrix representation form. A holistic perspective helps the designer to identify domain-spanning structures that would not have been recognized with single-domain optimization approaches or isolated analysis of the DSMs and DMMs. Domain-spanning structures are important to identify, as they may cause unpredictable product or process behavior. Our research showed that a holistic perspective can help designers to identify important elements more easily and therefore save time and enhance quality in analysis of engineering systems design. The framework we present consists of a proposal for the selection of appropriate product development domains, their integration, and the derivation of analysis results.
