Background: We have recently reported the downregulation of the Metallophosphoesterase-domain-containing protein 2 (MPPED2) gene and its cognate long non-coding RNA, MPPED2-AS1, in papillary thyroid carcinomas. Functional studies supported a tumor suppressor role of both these genes in thyroid carcinogenesis. We then decided to investigate their role in breast carcinogenesis. Methods: In order to verify MPPED2 expression, 45 human breast carcinoma samples have been investigated by quantitative real-time polymerase chain reaction (qRT-PCR). Then, MPPED2 has been transfected in several human breast carcinoma cell lines, analyzing its role in cell proliferation, migration and invasion. To study the regulation of MPPED2 expression the methylation of its promoter was investigated by targeted bisulfite sequencing. Results: MPPED2 expression was decreased in breast cancer samples, and this was confirmed by the analysis of data available in The Cancer Genome Atlas (TCGA). Interestingly, the hypermethylation of MPPED2 promoter likely accounted for its downregulation in breast cancer. Additionally, MPPED2-AS1 was also found downregulated in breast cancer tissues and, intriguingly, its expression decreased the hypermethylation of the MPPED2 promoter by inhibiting DNA methyltransferase 1 (DNMT1). Furthermore, the restoration of MPPED2 expression reduced cell proliferation, migration and invasion capability of breast carcinoma cell lines. Conclusion: Taken together, these results propose MPPED2 downregulation as a critical event in breast carcinogenesis.

Abstract Objectives Improved molecular testing for common somatic mutations and identification of mRNA and microRNA expression classifiers have emerged as the most promising approaches for diagnosis of thyroid nodules. However, it is necessary to effectively increase diagnostic accuracy further. Currently, lncRNA research has moved to the forefront of human cancer research, as recent findings have revealed a crucial role of lncRNAs in gene modulation. We evaluated the diagnostic value of the selected lncRNAs from The Atlas of Noncoding RNAs in Cancer (TANRIC) thyroid cancer dataset. Methods Using TANRIC thyroid cancer dataset, we compared 12,727 lncRNAs from the thyroid cancer tissues of 59 thyroid cancer patients with paired normal thyroid tissues. lncRNAs with significantly increased or decreased expression in thyroid cancer tissues were selected as candidates for thyroid cancer diagnosis. The expression levels of candidate lncRNAs were confirmed in patients with thyroid nodules who underwent surgery at the Severance Hospital. Subsequently, the candidate lncRNAs were applied to actual fine needle aspiration to verify their diagnostic value. Results LRRC52-AS1, LINC02471, LINC02082, UNC5B-AS1, LINC02408, MPPED2-AS1, LNCNEF, LOC642484, ATP6V0E2-AS1, and LOC100129129 were selected as candidates for thyroid cancer diagnosis. The expression levels of LRRC52-AS1, LINC02082, UNC5B-AS1, MPPED2-AS1, LNCNEF, and LOC100129129 were significantly increased or decreased in malignant nodules compared to their expression levels in benign nodules or paired normal thyroid tissues in our surgical tissue samples. The combination of LRRC52-AS1, LINC02082, and UNC5B-AS1 showed favorable results, with 89% sensitivity and 100% specificity, for thyroid cancer diagnosis using fine needle aspiration. Conclusions lncRNA expression analysis could be a promising approach for advancing molecular diagnosis of thyroid cancer. AbstractMigraine affects over a billion individuals worldwide but its genetic underpinning remains largely unknown. Here, we performed a genome-wide association study of 102,084 migraine cases and 771,257 controls and identified 123 loci, of which 86 are previously unknown. These loci provide an opportunity to evaluate shared and distinct genetic components in the two main migraine subtypes: migraine with aura and migraine without aura. Stratification of the risk loci using 29,679 cases with subtype information indicated three risk variants that seem specific for migraine with aura (in HMOX2, CACNA1A and MPPED2), two that seem specific for migraine without aura (near SPINK2 and near FECH) and nine that increase susceptibility for migraine regardless of subtype. The new risk loci include genes encoding recent migraine-specific drug targets, namely calcitonin gene-related peptide (CALCA/CALCB) and serotonin 1F receptor (HTR1F). Overall, genomic annotations among migraine-associated variants were enriched in both vascular and central nervous system tissue/cell types, supporting unequivocally that neurovascular mechanisms underlie migraine pathophysiology.

Dental caries is the most common chronic disease in children and a major public health concern due to its increasing incidence, serious health and social co-morbidities, and socio-demographic disparities in disease burden. We performed the first genome-wide association scan for dental caries to identify associated genetic loci and nominate candidate genes affecting tooth decay in 1305 US children ages 3-12 yrs. Affection status was defined as 1 or more primary teeth with evidence of decay based on intra-oral examination. No associations met strict criteria for genome-wide significance (p < 10E-7); however, several loci ( ACTN2, MTR, and EDARADD, MPPED2, and LPO) with plausible biological roles in dental caries exhibited suggestive evidence for association. Analyses stratified by home fluoride level yielded additional suggestive loci, including TFIP11 in the low-fluoride group, and EPHA7 and ZMPSTE24 in the sufficient-fluoride group. Suggestive loci were tested but not significantly replicated in an independent sample (N = 1695, ages 2-7 yrs) after adjustment for multiple comparisons. This study reinforces the complexity of dental caries, suggesting that numerous loci, mostly having small effects, are involved in cariogenesis. Verification/replication of suggestive loci may highlight biological mechanisms and/or pathways leading to a fuller understanding of the genetic risks for dental caries.

Abstract Age-related changes in DNA methylation are potent regulators of gene expression and may in part explain the onset of disease and disability. Vervet monkeys are a well-described model of neurocognitive and physical aging. Like humans, gait speed declines with age in vervets, and variability in gait speed in older animals is associated with age-related musculoskeletal and cognitive decline. To identify methylation patterns linked to aging-related physical decline, we investigated differentially methylated loci in vastus lateralis biopsies of 29 female vervets aged 8-28 years (~25-90 years in humans). We evaluated 107,490 loci on the Illumina Infinium Methylation EPIC Human Array that aligned with high fidelity to the vervet genome using the R package minfi and fit generalized linear mixed models to account for underlying genetic relatedness. We found 13 CpG methylation sites associated with 12 genes (CALCR, EBF4, GDNF, GMCL1, HAND1, HOXC10, IRX2, LBX2, MPPED2, SHISA6, SOX2, and WNT2) significantly differentially methylated with gait speed. Increased methylation was negatively associated with gait speed for all loci except GMCL1, reflecting the pattern of global hypermethylation of skeletal muscle tissue with age. Several of the associated genes are involved in development and differentiation including HOXC10 and LBX2, which regulates myoblast migration. CACNG8 regulates voltage-dependent calcium gated channels, and GDNF promotes motor neuron innervation of skeletal muscle. Most associations with muscle phenotypes are novel, but several have been linked to age-related bone diseases. We are currently evaluating the relationships of these differentially methylated loci with muscle mRNA expression and protein abundance.

Bladder cancer (BC) is the tenth most common cancer, with urothelial carcinoma representing about 90% of all BC, including neoplasms and carcinomas of different grades of malignancy. Urinary cytology has a significant role in BC screening and surveillance, although it has a low detection rate and high dependence on the pathologist’s experience. The currently available biomarkers are not implemented into routine clinical practice due to high costs or low sensitivity. In recent years, the role of lncRNAs in BC has emerged, even though it is still poorly explored. We have previously shown that the lncRNAs Metallophosphoesterase Domain-Containing 2 Antisense RNA 1 (MPPED2-AS1), Rhabdomyosarcoma-2 Associated Transcript (RMST), Kelch-like protein 14 antisense (Klhl14AS) and Prader Willi/Angelman region RNA 5 (PAR5) are involved in the progression of different types of cancers. Here, we investigated the expression of these molecules in BC, first by interrogating the GEPIA database and observing a different distribution of expression levels between normal and cancer specimens. We then measured them in a cohort of neoplastic bladder lesions, either benign or malignant, from patients with suspicion of BC undergoing transurethral resection of bladder tumor (TURBT). The total RNA from biopsies was analyzed using qRT-PCR for the expression of the four lncRNA genes, showing differential expression of the investigated lncRNAs between normal tissue, benign lesions and cancers. In conclusion, the data reported here highlight the involvement of novel lncRNAs in BC development, whose altered expression could potentially affect the regulatory circuits in which these molecules are involved. Our study paves the way for testing lncRNA genes as markers for BC diagnosis and/or follow-up.

Papillary thyroid cancer (PTC), accounting for more than 80 percent of all cases of thyroid cancer, is a form of a cancerous tumor that has a very favorable prognosis. However, patients diagnosed with PTC who are already in an advanced state have a dismal outlook. This study aimed to establish the diagnostic relevance of PRR15 expression in PTC patients as well as its levels in PTC samples and its connection with immune infiltrates. The TCGA and GEO datasets were combed through to obtain information on PTC patients. The “Limma” program was used to screen for differentially expressed mRNAs (DEMs), and the results were displayed using volcano plots and heat maps. The Wilcoxon test was used to examine the level of PRR15 expression in PTC patients in comparison with that of normal tissues. To study the connection between the immune infiltration level and PRR15 expression in PTC, the single-sample sequence set enrichment analysis (ssGSEA) from the R package was utilized. The expression of PRR15 was analyzed with RT-PCR in PTC cells and normal cells. In order to evaluate the diagnostic significance of PRR15 expression, ROC assays were carried out. Experiments using CCK-8 were carried out to investigate the impact that PRR15 knockdown could have on the proliferation of PTC cells. In this study, 17 overlapped DEMs between PTC specimens and normal specimens were identified, including MPPED2, IPCEF1, SLC4A4, PKHD1L1, DIO1, CRABP1, TPO, TFF3, SPX, TCEAL2, ZCCHC12, SYTL5, PRR15, CHI3L1, SERPINA1, GABRB2, and CITED1. Our attention focused on PRR15 which was highly expressed in PTC specimens as compared with nontumor specimens. PRR15 had an AUC value of 0.926 (95% CI 0.902–0.950) for PTC based on TCGA datasets. Pan-cancer assays suggested PRR15 as an oncogenic gene in many types of tumors. Moreover, we found that PRR15 expression was positively correlated with eosinophils, NK cells, NK CD56bright cells, IDC, macrophages, DC, mast cells, and Th1 cells. Further investigations with CCK-8 demonstrated that inhibiting PRR15 resulted in a decrease in the proliferation of PTC cells. Overall, PRR15 was confirmed to be a biomarker for PTC patients and a predictor of response to immunotherapy.

Correctly measuring district preferences is crucial for empirical research on legislative responsiveness and voting behavior. This article argues that the common practice of using presidential vote shares to measure congressional district ideology systematically produces incorrect estimates. I propose an alternative method that employs multiple election returns to estimate voters' ideological distributions within districts. I develop two estimation procedures—a least squared error model and a Bayesian model—and test each with simulations and empirical applications. The models are shown to outperform vote shares, and they are validated with direct measures of voter ideology and out-of-sample election predictions. Beyond estimating district ideology, these models provide valuable information on constituency heterogeneity—an important, but often immeasurable, quantity for research on representatives— strategic behavior.

Although there are compelling theoretical reasons to believe that unequal political representation in a legislature leads to an unequal distribution of funds, testing such theories empirically is challenging because it is difficult to separate the effects of representation from the effects of either population levels or changes. We leverage the natural experiment generated by infrequent and discrete census apportionment cycles to estimate the distributional effects of malapportionment in the U.S. House of Representatives. We find that changes in representation cause changes in the distribution of federal outlays to the states. Our method is exportable to any democratic system in which reapportionments are regular, infrequent, and nonstrategic.

Many social processes are stable and smooth in general, with discrete jumps. We develop a sequential segmentation spline method that can identify both the location and the number of discontinuities in a series of observations with a time component, while fitting a smooth spline between jumps, using a modified Bayesian Information Criterion statistic as a stopping rule. We explore the method in a large-n, unbalanced panel setting with George W. Bush's approval data, a small-n time series with median DW-NOMINATE scores for each Congress over time, and a series of simulations. We compare the method to several extant smoothers, and the method performs favorably in terms of visual inspection, residual properties, and event detection. Finally, we discuss extensions of the method.

This article is devoted to the study of a class of multiobjective mathematical programming problems with equilibrium constraints on Hadamard manifolds (in short, (MPPEC)). We consider (MPPEC) as our primal problem and formulate two different kinds of dual models, namely, Wolfe and Mond-Weir type dual models related to (MPPEC). Further, we deduce the weak, strong as well as strict converse duality relations that relate (MPPEC) and the corresponding dual problems employing geodesic pseudoconvexity and geodesic quasiconvexity restrictions. Several suitable numerical examples are incorporated to demonstrate the significance of the deduced results. The results derived in this article generalize and extend several previously existing results in the literature.

Recent advances in imaging technology have highlighted that scaffold proteins and receptors are arranged in subsynaptic nanodomains. The synaptic membrane-associated guanylate kinase (MAGUK) scaffold protein membrane protein palmitoylated 2 (MPP2) is a component of α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor–associated protein complexes and also binds to the synaptic cell adhesion molecule SynCAM 1. Using superresolution imaging, we show that—like SynCAM 1—MPP2 is situated at the periphery of the postsynaptic density (PSD). In order to explore MPP2-associated protein complexes, we used a quantitative comparative proteomics approach and identified multiple γ-aminobutyric acid (GABA)A receptor subunits among novel synaptic MPP2 interactors. In line with a scaffold function for MPP2 in the assembly and/or modulation of intact GABAA receptors, manipulating MPP2 expression had effects on inhibitory synaptic transmission. We further show that GABAA receptors are found together with MPP2 in a subset of dendritic spines and thus highlight MPP2 as a scaffold that serves as an adaptor molecule, linking peripheral synaptic elements critical for inhibitory regulation to central structures at the PSD of glutamatergic synapses.

Parapneumonic effusions often complicate Mycoplasma pneumoniae (MP) pneumonia, contrary to the notion that they are a rare feature of MP infection. Increased research and evidence on MP parapneumonic effusions (MPPE) can help elucidate its clinical significance as one of the variable manifestations of MP infection. This article aims to summarize the existing literature about the clinical characteristics of MPPE in adults and discuss its diagnostic implications from the perspective of pleural fluid analysis. Approximately 20–25% of adult patients with MP pneumonia develop MPPE, and its frequency in children and adults seems to be similar. Although the pathogenesis of MPPE remains to be elucidated, MP-induced cell-mediated immune mechanisms might be partially associated with the development of MPPE. MPPE usually shows mononuclear leukocyte predominance with elevated adenosine deaminase (ADA) activity, similar to tuberculous pleural effusion (TPE). The degree of increase in pleural fluid ADA levels and serum inflammatory biomarkers may help differentiate between MPPE and TPE. During the acute phase, a single positive IgM and positive polymerase chain reaction results allow for a precise and reliable MP infection diagnosis. The mainstay of treatment is the selection of adequate anti-mycoplasma antibiotics with or without corticosteroid, based on the local epidemiologic data on macrolide resistance.

ABSTRACT The effects of methanolic pomegranate peel extract (MPPE) on the quality of gutted rainbow trout (Oncorhynchus mykiss) were examined periodically during 6 months of storage at −18°C. Fresh samples were dipped (ratio of fish to liquid, 1:2 [wt/vol]) in sterile water or in sterile water containing 1, 2, or 4% (wt/vol) MPPE and packed in low-density polyethylene pouches. The control and treated samples were analyzed monthly for microbiological, chemical, sensory, and textural characteristics. Microbial growth in samples was significantly reduced by MPPE treatment (P &lt; 0.05). Smaller increases in the values for total volatile base nitrogen, peroxide, and thiobarbituric acid–reactive substances and better oxidative stability were also obtained. Moreover, higher organoleptic scores were recorded. However, regarding the general acceptability, the highest score was achieved in the 1% MPPE group. Greater hardness and chewiness were obtained with 4% MPPE (P &lt; 0.05). Therefore, dipping fish in MPPE can be considered an effective method to extend the shelf life and the overall quality of the product.

Mountain pine (Pinus mugo Turra) is a coniferous native to the highlands of central Europe. Our previous study revealed that mountain pine proliferation decline (MPPD) disease in the Curonian Spit of Lithuania is caused by a ‘Candidatus Phytoplasma pini’-related strain (16SrXXI-A). However, the insect vector of MPPD has not been identified. In this study, we conducted a survey to determine potential insect vectors of MPPD phytoplasma for three consecutive years (2016–2019). More than 1000 insect samples were collected from four locations in the Curonian Spit. These insects were identified as belonging to six families and ten genera. The presence of phytoplasma in insect samples was examined by nested polymerase chain reaction (PCR) using phytoplasma-specific primers (P1A/16S-SR and R16F2n/R16R2n). Phytoplasmas were detected in Cinara (Cinara) pini (Scots pine aphid), Cinara (Cinara) piniphila and Cinara (Schizolachnus) pineti (waxy grey pine needle aphid) insect samples. Subsequent restriction fragment length polymorphism (RFLP) analysis showed that the PCR-RFLP profile of these positive insect samples was consistent with that of the MPPD of diseased pine trees. These results suggest that C. (C.) pini, C. (C.) piniphila and C. (S.) pineti may be potential insect vectors of MPPD phytoplasma. The findings from this survey will provide useful information for the management of MPPD disease.

Perkembangan pemanfaatan panel surya di masa depan akan terus meningkat. Salah satu bentuk karakteristik panel surya merupakan kurva I-V yang mana dengan kurva tersebut dapat digunakan untuk menganalisa besaran daya keluaran panel surya. Dengan mengetahui kurva I-V tersebut dapat dilakukan Maximum Power Point Estimation (MPPE) yang dapat diampu oleh panel surya. Informasi mengenai nilai estimasi daya maksimum panel surya merupakan bagian penting untuk menentukan kapasitas pembebanan, selain itu juga untuk menjaga umur peralatan yang digunakan. Feed Forward Neural Network dengan Algoritma Back Propagation (FFBP) terbukti dapat memberikan informasi nilai MPPE pada keluaran panel surya. Nilai masukan pada ANN berupa tegangan dan arus dari panel surya, sedangkan keluaran dari ANN tersebut berupa nilai estimasi daya. Hasil dari simulasi MPPE didapatkan galat rata rata sebesar 0.04 poin antara daya aktual (MPP) dan daya estimasi (MPPE).

BackgroundRegardless of the significant efforts used and put in place to enhance supply chain performance in Rwanda, there is still poor inventory management and big loss due to expiry of medicines at Medical Procurement and Production Division (MPPD).ObjectiveTo assess the factors that contributes to expiry of medicines at MPPD and formulate recommendations for the improvement of the current situation.MethodsThis is a descriptive cross-sectional study used for 25 respondents. A questionnaire containing closed-ended questions was used focusing on factors contributing to expiries of medicines for each product category managed by MPPD from 2014-2018. Additionally, the researcher reviewed all inventory reports, including expiries for the same period under study. The study focused on targeting respondents working at MPPD as pharmacists both technical and administrative, also other non-pharmacist working in the warehouse were considered as respondents. The hard copy of questionnaire was handed over to 26 respondents and requested to provide filled questionnaire within seven days. Twenty-five respondents were able to return the filled questionnaire on time, giving 96% response rate.ResultsThe study found that in MPPD, medicines are still expiring. During five years (from 2014 up to 2018) the total expired products were valued at RWF 6,046,778,655 for all program categories: HIV commodities had the largest share 53.3%, Essential Medicines 22.5%, Malaria 13%, Maternal Child Health commodities 5.7%, Products used for Community health workers 4.5%, TB products 1% and 0.1% for Family Planning products. The study found that major contributing factors for expiry of health commodities at MPPD, are ranked as follows: Supply chain management 90%, other factors 73%, Poor storage management 68% and Excessive drug supply 67%. ConclusionConsidering the study results, it looks like no significant effect of excessive drug supply and inventory management on the expiration of medicines at MPPD. However, the Supply Chain Management components are the most vulnerable to contribute to the expiration of medicines at MPPD. There are many changes to be done inside the institution to remedy the issue of expiration like working according to the standard operating procedures, improving the Skills of personnel in supply chain management, empowering the procurement unit with skilled personnel and improving the communication with stakeholders to facilitate the smooth and quick replenishment and distribution of the stock. Hydrolysis of bis-p-methoxyphenyl phosphate ester (bis-p-MPPE) was studied in micellar solutions of cityltrimethylammoniumbromide n-C16H33N+(CH3)3Br- (CTABr) at pH-9.0. The hydrolysis followed first order kinetics with respect to bis-p-MPPE concentration. At the concentration of critical micelle concentration (CMC) the rate of hydrolysis increased with increasing CTABr concentration. Surfactant with cationic or polar head group form micelles in water with hydrocarbon like interior or polar groups at the surface and bind cationic solute. The binding constant of micelle for bis-p-MPPE and the rate constant in micellar pseudo phase were determined from kinetic data using the pseudophase model.

Macropod Progressive Periodontal Disease (MPPD) is a well-recognised disease that causes high morbidity and mortality in captive macropods worldwide. Epidemiological data on MMPD are limited, although multiple risk factors associated with a captive environment appear to contribute to the development of clinical disease. The identification of risk factors associated with MPPD would assist with the development of preventive management strategies, potentially reducing mortality. Veterinary and husbandry records from eight institutions across Australia and Europe were analysed in a retrospective cohort study (1995 to 2016), examining risk factors for the development of MPPD. A review of records for 2759 macropods found incidence rates (IR) and risk of infection differed between geographic regions and individual institutions. The risk of developing MPPD increased with age, particularly for macropods >10 years (Australia Incidence Rate Ratio (IRR) 7.63, p < 0.001; Europe IRR 7.38, p < 0.001). Prognosis was typically poor, with 62.5% mortality reported for Australian and European regions combined. Practical recommendations to reduce disease risk have been developed, which will assist zoos in providing optimal long-term health management for captive macropods and, subsequently, have a positive impact on both the welfare and conservation of macropods housed in zoos globally.

e16776 Background: SM-88 (racemetyrosine) is a dysfunctional tyrosine derivative; previous studies demonstrated a well-tolerated profile with encouraging efficacy. Recent advances in image analysis using Quantitative Textural Analysis (QTA) have uncovered non-invasive biomarkers that correlate with molecular drivers of cancer and prognostic signatures of response in PDAC. Earlier radiomic data from subjects treated with SM-88 showed a positive correlation between circulating tumor cells and tumor texture at baseline. This study extends those findings to focus on radiomic changes associated with SM-88 doses in a Phase II dose escalation trial (NCT03512756). Methods: Retrospective evaluation of 18 subjects with >1 prior therapy, ECOG PS <2, with no restriction on size, number, or locations of mets, and had baseline (BL) and follow up (FUP) contrast enhanced CTs. Subjects received > 1 cycle of SM-88 at either 460mg (n = 8) or 920mg (n = 10) QD oral doses. CTs were analyzed by the QTA platform (TexRad, Essex, UK) for tumor texture. The largest met of each subject was selected using portal venous phase images. A region of interest was posited on the axial slice with the longest tumor diameter and features automatically generated using voxel resampling to correct for scanner variability across subjects and time points. Results were displayed as histogram frequency curves (HFCs) of pixel densities (Hounsfield Units). First order HFC analysis (mean, StDev, MPP, skewness, kurtosis) at spatial scale filters (SSF) ranging from no filter, to fine, to coarse texture (SSF:0, SSF:2, SSF:6, respectively) were reported. Results: Subjects whose largest lesions showed greater changes in tumor texture at FUP vs. BL were more likely to develop tumor progression (StDev in PD (n = 10) vs SD (n = 7) (1 N/A): -0.07 vs -0.25, p = 0.05). Greater tumor textural changes were associated with poor survival at 180 days (Skewness: Chi-Sq. = 4.81, p = 0.03; HR = 4.1 for above median □Skewssf3). There was a negative change in FUP kurtosis vs BL in the 6 subjects who developed new lesions vs the 12 who did not (mean □kurtosis = -0.67 vs +2.63, p = 0.05). Greater change in tumor texture on FUP vs BL scans was seen in the 460mg vs 920mg group (MPPLD = -0.29, MPPHD = -0.04; p = 0.05) suggesting more tumor stability (less change in texture and better outcomes) was associated with the 920mg dose. Conclusions: Tumor lesions in subjects on 920mg of SM-88 were less likely to show a change in tumor texture and were associated with better outcomes. Using radiomic QTA, less tumor texture variability from baseline may be associated with better outcomes in PDAC subjects.

Efficient noninvasive imaging techniques in the differentiation of intrahepatic cholangiocarcinoma (ICC) and hepatocellular carcinoma (HCC) are very important because of their different management and prognosis. Our purpose was to evaluate the difference of parameters extracted from intravoxel incoherent motion (IVIM) diffusion-weighted imaging (DWI) between the two groups and their performance for the differentiation, as well as the significance of perfusion information. IVIM studies (9 b-values) in 41 patients with either ICC or HCC were reviewed retrospectively by two observers. Diffusion coefficient (D), pseudodiffusion coefficient (D ∗ ), perfusion fraction (f), ADC, and the mean percentage of parenchymal enhancement (MPPE) at 30 s after contrast-enhancement were calculated and compared between ICC and HCC. The relationship between D ∗ , f values, and MPPE was evaluated by Spearman’s correlation test. The diagnostic efficacy of all parameters was analyzed by the receiver operating characteristic (ROC) curve. Interobserver and intraobserver agreements were analyzed. The parameters (D and ADC) of ICC were distinctly higher than those of HCC; whereas the parameters (f and MPPE of arterial phase) were distinctly lower (all false discovery rate [FDR]-corrected P < 0.05). The metric D ∗ value of ICC was slightly higher than that of HCC (71.44 vs 69.41) with FDR-corrected P > 0.05. Moreover, the value of parameter D was significantly lower than that of ADC (FDR-corrected P < 0.05). The parameters (D and f values) extracted from IVIM showed excellent diagnostic efficiency in the identification, and the diagnostic efficiency of D value was significantly higher than that of the ADC. There were positive correlations between perfusion-related parameters (D ∗ , f values) and MPPE. Interobserver and intraobserver agreements were excellent or perfect in measurements of all parameters. Parameters derived from IVIM were valuable for distinguishing ICC and HCC. Moreover, the D value showed better diagnostic efficiency for the differential diagnosis than monoexponential fitting-derived ADC value. Meanwhile, the significant correlation between perfusion-related parameters and MPPE demonstrates that specific IVIM metrics may serve as a noninvasive indicator for the vascular perfusion information of ICC and HCC.

The Phasmatodea population evolution algorithm (PPE) is a novel metaheuristic algorithm proposed in recent years, which simulates the evolutionary trend of stick insect population. In this article, a multigroup-based Phasmatodea population evolution algorithm with mutistrategy (MPPE) is proposed to further improve the overall performance of PPE. During the initialization period, the stick insect population is divided into multiple groups, and the step factor of the flower pollen algorithm is introduced into the population growth model of no more than half of the groups. This makes the population evolution trend diversified and prevents the algorithm from falling into the local optimal solution to a certain extent. In terms of intergroup communication, two communication strategies are adopted to mutate and replace the inferior particles, respectively, which improves the convergence speed and search ability of the algorithm. In the MPPE performance test, we compared it with PPE, five standard algorithms, and other parallel algorithms in CEC 2013 Benchmark Suite. Finally, this algorithm is applied to the IoT based electric bus scheduling for urban waterlogging situation, and the excellent performance of MPPE is verified comprehensively.

ABSTRACT Limited proteolysis of proteins transiently expressed on the surface of the opportunistic pathogen Toxoplasma gondii accompanies cell invasion and facilitates parasite migration across cell barriers during infection. However, little is known about what factors influence this specialized proteolysis or how these proteolytic events are regulated. Here we show that genetic ablation of the micronemal protein MIC5 enhances the normal proteolytic processing of several micronemal proteins secreted by Toxoplasma tachyzoites. Restoring MIC5 expression by genetic complementation reversed this phenotype, as did treatment with the protease inhibitor ALLN, which was previously shown to block the activity of a hypothetical parasite surface protease called MPP2. We show that, despite its lack of obvious membrane association signals, MIC5 occupies the parasite surface during invasion in the vicinity of the proteins affected by enhanced processing. Proteolysis of other secretory proteins, including GRA1, was also enhanced in MIC5 knockout parasites, indicating that the phenotype is not strictly limited to proteins derived from micronemes. Together, our findings suggest that MIC5 either directly regulates MPP2 activity or it influences MPP2's ability to access substrate cleavage sites on the parasite surface.

Standardizationof best combination for the development of immunity boostinginstant soup mix, various quality parameters, the storage stability and cost estimation of product were evaluated. Protein in instant soup ranged from 9.76 to 11.89 percent in different formulations of instant soup mix. Formulation MPP4 (11.89) had significantly maximum protein content followed by MPP3, MPP2, MPP1 and control with the minimum protein content. The original immunity boostinginstant soup mix (control) exhibited highest carbohydrate content (61.52%) followed by MPP1, MPP3, MPP4, and MPP2 with lowest content. The evaluated scores of control for colour and appearance, taste, flavour, consistency, after taste and overall acceptability were 8.36, 8.50, 8.60, 8.90, 8.76 and 8.63, respectively. Bangladesh J. Copper clad laminates (CCLs) with low dissipation factor (Df) are urgently needed in the fields of high-frequency communications devices. A novel resin matrix of modified poly (2,6-dimethyl-1,4-phenylene ether) (MPPE) and styrene-ethylene/butylene-styrene (SEBS) was employed in the fabrication of high-frequency copper clad laminates (CCLs). The composites were reinforced by E-glass fabrics, which were modified with phenyltriethoxysilane (PhTES). The composite laminates obtained exhibited impressive dielectric loss of 0.0027 at 10 GHz when the weight ratio of MPPE to SEBS was 5:1. In order to modify the dielectric constant (Dk), coefficient of thermal expansion (CTE) and other performances of laminates, Li2TiO3 (LT) ceramic powders were introduced into the resin matrix. The composite laminates showed low dielectric loss of 0.0026 at 10 GHz and relatively high flexural strength of 125 MPa when the mass ratio of LT fillers to resin is 0.4. Moreover, the composite laminates all maintain low water uptake (<0.5%). The microstructure and thermal properties of composite laminates filled with LT ceramic powders were also tested. These results show that copper clad laminates prepared with modified polyphenylene ether (MPPE)/SEBS and LT ceramic fillers have strong competitiveness to fabricate printed circuit boards (PCBs) for high-frequency and high-speed applications.

Background. Visceral hypersensitivity is a common occurrence of gastrointestinal diseases such as irritable bowel syndrome (IBS), wherein early-life stress (ELS) may have a high predisposition to the development of visceral hypersensitivity in adulthood, with the specific underlying mechanism still elusive. Herein, we assessed the potential effect of small-conductance calcium-activated potassium channel subtype 2 (SK2) in the spinal dorsal horn (DH) on the pathogenesis of visceral hypersensitivity induced by maternal separation (MS) in mice. Methods. Neonatal mice were subjected to the MS paradigm, an established ELS model. In adulthood, the visceral pain threshold and the abdominal withdrawal reflex (AWR) were measured with an inflatable balloon. The elevated plus maze, open field test, sucrose preference test, and forced swim test were employed to evaluate the anxiety- and depression-like behaviors. The expression levels of SK2 in the spinal DH were determined by immunofluorescence and western blotting. The mRNA of SK2 and membrane palmitoylated protein 2 (MPP2) were determined by quantitative real-time polymerase chain reaction (qRT-PCR). Electrophysiology was applied to evaluate the neuronal firing rates and SK2 channel-mediated afterhyperpolarization current (IAHP). The interaction between MPP2 and SK2 was validated by coimmunoprecipitation. Results. In contrast to the naïve mice, ethological findings in MS mice revealed lowered visceral pain threshold, more evident anxiety- and depression-like behaviors, and downregulated expression of membrane SK2 protein and MPP2 protein. Moreover, electrophysiological results indicated increased neuronal firing rates and decreased IAHP in the spinal DH neurons. Nonetheless, intrathecal injection of the SK2 channel activator 1-ethyl-2-benzimidazolinone (1-EBIO) in MS mice could reverse the electrophysiological alterations and elevate the visceral pain threshold. In the naïve mice, administration of the SK2 channel blocker apamin abated IAHP and elevated spontaneous neuronal firing rates in the spinal DH neurons, reducing the visceral pain threshold. Finally, disruption of the MPP2 expression by small interfering RNA (siRNA) could amplify visceral hypersensitivity in naïve mice. Conclusions. ELS-induced visceral pain and visceral hypersensitivity are associated with the underfunction of SK2 channels in the spinal DH.

The merit of Red Chittagong cows-a promising indigenous cattle genetic resource of Bangladesh was evaluated by using test day milk yield records of total 41 individuals. The data were collected from the Nucleus Herd of Red Chittagong (RC) cattle maintained at Bangladesh Agricultural University, Mymensingh from June, 2007 to October, 2008 under supervised animal recording system along with the previously recorded data from 2005 was also taken into consideration. Data were then analyzed for evaluating the cows based on lactation yield and average daily milk yield. Average lactation yield and daily milk yield were 453.86 ± 59.61 and 1.99 ± 0.14 kg, respectively. The highest lactation yield (538.04 kg) and daily milk yield (2.3 kg) was found in 4th and 5th lactation, respectively. The most probable producing ability (MPPA) of RC cows was calculated by using the test day records and then the animals were ranked on the basis of their MPPA value. For MPPA calculation two methods were followed; cow’s repeated lactation performance (MPPA1) and within lactation repeated test day records and averaged (MPPA2). The highest MPPA for lactation yield was shown by the cow no. 36 (645.61 kg) and the highest MPPA1 and MPPA2 for daily milk yield were 2.83 and 3.46 kg, respectively for cow no. 03. A implantação do Balanced Scorecard (BSC) no modelo de gestão estratégica adotado pelo Ministério Público de Pernambuco (MPPE) foi objeto deste artigo, no período 2013-2016. Foi um estudo de caso em que foram utilizadas técnicas de pesquisa documental, análise de conteúdo e entrevistas com a equipe gestora. O referencial teórico escolhido abrange o balanced scorecard, características gerais e fatores de insucesso da implantação; exemplos do BSC no setor público e gestão estratégica. O MPPE foi exitoso no formato definido do BSC no seu modelo de gestão estratégica, pois a sequência de implantação seguida, adaptou-se à realidade da instituição. Na Gestão Estratégica 2013-2016, a metodologia BSC aplicada funcionou como ferramenta de gestão estratégia e como sistema de avaliação de desempenho. Também foi exitoso no seu modelo de gestão estratégica associado ao uso do BSC, porque nenhuma das causas de insucesso apontadas pela doutrina, na implantação do BSC foi identificada, talvez porque a sequência de implantação seguida foi adequada à realidade da instituição, apesar de alguns objetivos não terem sido realizados. O MPPE, pela primeira vez, teve um ciclo estratégico analisado desde sua construção até a execução, com todas as etapas registradas e analisadas e resultados alcançados. É uma iniciativa que foi premiada nacionalmente, com o Prêmio Innovare, que tem como objetivo identificar, divulgar e difundir práticas que contribuam para o aprimoramento da Justiça no Brasil. Mesmo assim, deverá ser aprimorada para o novo ciclo de gestão estratégica organizacional, como evolução natural e estratégica de gestão.

This paper demonstrates how computationally prediction can be done on inhaled ultrafine aerosol particles that are transported, disseminated, and deposited in the respiratory tracts of laboratory mice. Poyldisperse ultrafine particles (UFP) range between 1 nm and 100 nm in diameter. Multiple Path Particle Dosimetry (MPPD), a probabilistic computational simulation software was used to mimic in-vitro experimental conditions. In this work, the physical, mechanical and electrical properties of the UFPs were used as input parameters in MPPD. Additionally, pulmonary physiologic and morphometry input variables for BALB/c mice strain were applied to the simulation. Finally, the UFP deposition results of the computational simulation study were compared with in-vitro UFP deposition trends published in scholarly journals, and fitting agreements were found. Mutually both in-silico (computational modeling) and in-vitro studies complemented each other in determining the UFP toxicity burdens in fetal mice.

Understanding the emergence of lymphoid committed cells from multipotent progenitors (MPP) is a great challenge in hematopoiesis. To gain deeper insight into the dynamic expression changes associated with these transitions, we report the quantitative transcriptome of two MPP subsets and the common lymphoid progenitor (CLP). While the transcriptome is rather stable between MPP2 and MPP3, expression changes increase with differentiation. Among those, we found that pioneer lymphoid genes such as Rag1, Mpeg1, and Dntt are expressed continuously from MPP2. Others, such as CD93, are CLP specific, suggesting their potential use as new markers to improve purification of lymphoid populations. Notably, a six-transcription factor network orchestrates the lymphoid differentiation program. Additionally, we pinpointed 24 long intergenic-non-coding RNA (lincRNA) differentially expressed through commitment and further identified seven novel forms. Collectively, our approach provides a comprehensive landscape of coding and non-coding transcriptomes expressed during lymphoid commitment.
