Abstract We have fabricated and characterized all-MoGe Josephson junctions with a very thin Al/AlO x /(Al) barrier, where the amorphous MoGe films exhibit superconducting transition temperatures up to 7 K. Due to the uniformity of the surface morphology of the MoGe films, the junctions demonstrate high uniformity of their tunneling properties. The experimental data on the temperature dependence of the subgap current agree well with theoretical calculations. The results obtained imply that Josephson tunnel junctions based on amorphous superconductors are promising candidates for use in superconducting electronics, especially in applications requiring multiple stacked junctions or the creation of a nonequilibrium quasiparticle distribution.

No final da década de 1990, com o aperfeiçoamento das técnicas de fabricação de filmes finos nanoestruturados, houve vários estudos que procuraram entender a dinâmica de vórtices e suas características. Esses sistemas normalmente compreendem uma rede regular de buracos ou inclusões magnéticas, que atuam como armadilhas para os vórtices, fornecendo potenciais periódicos de ancoragem. Por esta razão, vórtices em supercondutores nanoestruturados apresentam vários fenômenos interessantes que não podem ser encontrados em filmes contínuos. Possivelmente o exemplo mais marcante é o efeito de matching devido à comensurabilidade entre a rede de vórtice e a rede de ancoragem artificial. A depender das condições impostas ao sistema (temperatura, campo magnético aplicado, corrente de transporte aplicada e os parâmetros da rede de ancoragem) os vórtices se organizarão de maneiras distintas e isso irá, obviamente, traduzir-se em diferentes respostas macroscópicas. Nos chamados campos de matching (B = nB1, onde n é um inteiro e B1 é a densidade de fluxo magnético correspondente a um vórtice por célula unitária) os vórtices sentem mais o potencial de ancoragem e isto aparece experimentalmente como um aumento na corrente crítica (medida estática) ou uma atenuação na mobilidade dos vórtices (medida dinâmica). Acredita-se amplamente que as características observadas nas medidas estáticas e dinâmicas estão correlacionadas de forma que pode-se usar medidas de magnetotrasporte para inferir sobre as propriedades da comensurabilidade estática da rede de vórtice nos filmes nanoestruturados. Nesta dissertação, estudaremos experimentalmente as propriedades dinâmicas dos vórtices em um filme fino amorfo de MoGe com uma rede regular quadrada de antidos (buracos). A amostra foi fabricada por litografia eletrônica e deposição por laser pulsado na Universidade de Leuven, Bélgica. Foram feitas várias medidas de magnetorresistência nessa amostra usando voltímetros e fontes de corrente altamente sensíveis. Estes foram conectados ao equipamento comercial PPMS (Physical Properties Measurement System, da Quantum Design), usado como ambiente criogênico, através de uma caixa adaptadora confeccionada nos nossos laboratórios. As medidas são controladas por um computador através de uma rotina escrita pelo software de automação LabView. As curvas de magnetorresistência para diferentes valores de temperatura exibem uma série de características em diferentes campos de matching, como esperado do fenômeno de comensurabilidade estudado em trabalhos anteriores. Entretanto, para o sexto campo de matching, a forma característica da curva de magnetorresistência está ausente para todos os valores de temperatura. Seguindo um modelo baseado em configurações estáticas de vórtices em redes regulares de antidots, uma atenuação no efeito de matching é esperada para o terceiro campo de matching, devido aos parâmetros geométricos da nossa amostra. De fato, foi observada uma atenuação em B = 3B1. Entretanto, o modelo falha em explicar a ausência do efeito de matching em B = 6B1 encontrado nas nossas medidas. É proposto que vórtices em movimento na presença de um potencial de ancoragem periódico podem apresentar efeitos de comensurabilidade ligeiramente diferentes daqueles que ocorrem na situação estática porque a rede de vórtice que se move interage efetivamente com um potencial de ancoragem dependente do tempo.

This thesis is presented in five chapters. Chapter one offers a review of the literature, divided into six sections. The first section of this chapter provides an understanding of cancer as a disease and a public health concern, including detail on its incidence and prevalence, the biology of cancer and its treatment. The second section of this chapter reviews the literature on physical activity and cancer across the cancer continuum, from prevention of cancer, through cancer treatment, living with or beyond cancer, advanced cancer, and detail on physical activity and cancer mortality and recurrence. The third section of this chapter discusses the determinants of physical activity in cancer survivors, framed within behavioural theories and models to support the identification of intervention opportunities. The identified intervention opportunities are covered in the fourth section. The fifth section of this chapter presents the scope and relevance of the research included in this thesis. The sixth section provides a summary of chapter one. Chapter two presents the first study included within this thesis, a service evaluation investigating the reach, adoption, and impact of a training intervention to influence UK healthcare professionals to give very brief advice on physical activity to cancer survivors. This study also includes an assessment of the predictive value of the behavioural theory upon which the intervention is based, the COM-B model of behaviour (Michie, van Stralen, & West, 2011). Chapter two includes four sections covering an introduction, the method, results, and discussion. Chapter three presents the second study within this thesis. The second study is a randomised waiting list control trial to investigate the impacts of a remote-print based intervention supported by Internet-based tools on the physical activity, self-efficacy, and health-related quality of life (HRQOL) of cancer survivors. The randomised waiting list control trial also includes a cost-consequence analysis. Chapter four presents the third study of this thesis, a process evaluation of the remote-print based intervention supported by Internet-based tools. This process evaluation aimed to contextualise use and perceived usefulness of the intervention, supporting the identification of what works and for whom. Chapter five then closes this thesis with a discussion on the theoretical and practical contributions of the body of research as a whole. This final chapter also includes discussion on how the interventions under assessment influence the identified determinants of physical activity in cancer survivors, and identification of future research possibilities.

Gravity plays a vital role in our everyday lives. It provides the development of our muscle mass which fuels our brain’s advancement and our ability to complete daily tasks. Yet we often take for granted the things we do not see. From hunting and gathering, chasing after prey, migrating from one place to the next, to growing and harvesting crops to long hours laboring away, we have felt gravity through millions of years in time.  Along with the rise of technology, we are witnessing the fall of our physical selves. Our lives have become busier, more stressful and our days behind a screen much longer. Within the past 30 years, gravity has been intercepted by the chair and the ratio of moving to sitting has been reversed. Instead of feeling this force within our legs and lower limbs, it has now been deferred to our fingers, buttocks and backs.  We spend the bulk of our days in a seated position, often behind a screen. Sitting has become such a norm that daily exercise at the gym is no longer enough to offset all the negative things done to our bodies in a sedentary posture.  We were programmed to move, but we’ve hacked our brains to think otherwise. Prolonged sitting is a serious issue that should not be overlooked. The challenge is to change a habit that we’ve acquired from an early age.  The numbers are staggering, over 1.5 billion people worldwide are obese.(WHO. int, 2013) 5.3 million die each year from heart related diseases and diabetes as indirect result of the chair, .3 million more than smoking. (Thelancet.com, 2012)  It is time to break the norm, adjust our perceptions, expectations and stop living in extremes and move towards moderation. It’s time to stand up and make gravity visible.  10/60: Make Gravity Visible is a social movement to challenge our society to move more, reminding people to be up and moving 10 minutes for every 60 minutes.  The goal is to challenge society to integrate movement within the home, work and societal environments through the support and influence of communal behaviour.  10/60 is facilitated by a website and a smartphone application. The website provides ideas shared by the community to help inspire movement into people’s lives. The app tracks a person’s daily physical activity levels and sends a reminder when he/she has been sitting for too long. How much a person moves is reflected by an avatar’s physical state as well as graphical data by the time of day. Challenges can be submitted within the inner circle of friends to encourage more movement.  The initial strategy around this topic was through the intersections of four subjects: human evolution, philosophy, behavioral psychology, and physiology. This concept was developed based on findings from foundational research, expert interviews, user tests, experiments and behavioral observations - in particular social influences and the underlying principle that motivation is different for everyone.  Awareness alone does not generate action. Motivating people to move more is beyond an individual problem - sitting is a societal and cultural issue and unless that is addressed, no long term changes can be sustained.  This project aims to inspire people to leverage their communities and integrate more physical activity in the home, work and societal environments.  Societal and cultural norms will not change unless we change together.

The significance of fungal infections has been grossly underestimated. Only a few drugs are clinically available to treat life-threatening fungal infections, and resistance against these drugs is rising. Antifungal peptides (AFPs) are being actively explored as novel pharmaceuticals. PAF26 is a de novo designed hexapeptide possessing N-terminal cationic and C-terminal hydrophobic regions. Previously the roles of each of these motifs in the antifungal mode-of-action of PAF26 have indicated that it involves three stages: interaction with the plasma membrane, internalisation, and cell killing. The overall aim of my project was to obtain further insights into its mode-of-action and develop more active antifungal derivatives of PAF26. Three experimental fungal systems were used in this study: the model Neurospora crassa, the human pathogen Aspergillus fumigatus and the plant/human pathogen Fusarium oxysporum. The first objective of the study was to evaluate the impact of different fluorescent labels on the intracellular localisation and antifungal properties of PAF26. For this purpose a library of PAF26 labelled with 13 different fluorophores was synthesised. This library contained PAF26 conjugates of broad chemical and spectral diversity. These fluorescent PAF26 conjugates were analysed by live-cell imaging and tested for their antifungal activities. The different fluorescent labels were found to have significant impacts both on intracellular localisation and antifungal activities. TMR, carboxyfluorescein, NBD and DMN were found to be the best labels for live-cell imaging because they had the least influence on the intracellular localisation and antifungal activity of PAF26. The second objective was to identify target proteins of PAF26 in N. crassa cells. A large number of proteins were identified as binding to PAF26 from a protein pull-down and mass spectroscopy analysis using TMR- and fluorescein-labelled PAF26. One of these proteins was the highly abundant plasma membrane ATPase PMA-1. An in-silico analysis showed that PMA-1 is likely to be a major target protein of PAF26. The final objective was to develop novel antifungals based on PAF26 with improved activities and stability. Novel cyclic derivatives of PAF26 were designed in-silico against PMA-1. These peptides were synthesised and tested against N. crassa, A. fumigatus and F. oxysporum and were found to have higher activities (at the sub-micromolar level) and greater stability than the linear PAF26. Overall this study has provided novel mechanistic insights into the mode-of-action of PAF26 and discovered novel highly active antifungal peptides with clinical potential as therapeutics.

This study sought to advance understanding of student perceptions around their engagement in the move towards blended and online mode learning in higher education. Therefore, the research aimed to explore the extent to which different demographic groups of students have engaged in blended and online methods within a cross-section of courses within a higher education context. More specifically, the study endeavored to investigate specific student preferences around the move from traditional face-to-face to more blended and online learning environments. Furthermore, the research aimed to gather student perceptions of internet use in their learning to consider how to more effectively use blended and online modes to aid in student transition and engagement whilst driving satisfaction. The project adopted a mixed method design of both quantitative and qualitative research to explore the extent to which students feel engaged and satisfied when adopting blended and online learning techniques within a variety of courses in the business school of a large Australian university. Initially, study 1 focused on a quantitative approach collecting data from a mix of 767 undergraduate students segmented across different groups. The main established scale items proposed were adapted from Krause and Coates (2008) around key areas of student transition, academic, peer, student-staff, intellectual, online and beyond class engagement. The results from the surveys aimed to suggest that student levels of engagement and satisfaction differ across the use of traditional face to face versus blended and online modes. Findings from this research looked to identify that despite the high daily use of internet among students this is not necessarily their first preference for learning as they value traditional modes. Additionally, study 2 used semi-structured interviews conducted with 50 full-time and part-time students from across various years within their degrees in a randomly selected sample that self-nominated from study 1. The research highlighted that students are influenced by the ability to interact with their lecturers and tutors as well as their peers to gain the necessary support they feel impact on their success. The outcomes of the study acknowledged that students value the ability to ask questions, be involved in group work as well as the wider university community, whilst also being able to access learning online. Despite these preferences that propose that students still value face to face mode, the interview results also suggested that students value the convenience, accessibility, and flexibility of blended and online modes. However, it was evident that institutions need to offer a variety of mode preferences to suit the mix of students needs in the current higher education market. Ultimately, the study looked to consider student preferences towards the move to more blended and online learning in higher education. Whilst having explored the interplay among students, academics and the university to determine student learning transformation in different modes. Consequently, the purpose of the project was to gain a better understanding of the key strategy’s universities can adopt to manage their students more effectively and drive engagement in a blended mode, mixing face-to-face and online learning environments. This research has both theoretical and practical implications by informing institutions of student preferences around adopting blended and online modes in order create better practice, which remain under-researched in the marketing and business education literature. Subsequently, the discussion in this study endeavoured to investigate student learning preferences around the move to blended and online learning in higher education and the impact their overall engagement and satisfaction.

The objective of this study was to evaluate the viability of the use of raw milk samples conserved with bronopol in the detection of cheese whey by High Performance Liquid Chromatography (HPLC). The samples added with bronopol were stored in two temperatures (7 and 30º C) and the control samples (without preservative) were kept under freezing. All the groups were tested for the time of storage up to eight days. In each group were added five levels of cheese whey (0%, 2%, 5%, 10% and 20%). There was no difference (p > 0.05) among the treatments thus, it is possible to use samples of raw milk conserved with bronopol in the detection of cheese whey by HPLC. The storage time and temperature did not interfere in this methodology for samples conserved with bronopol stored cooled or at room temperature up to eight days
O objetivo deste trabalho foi avaliar a viabilidade do uso de amostras de leite cru conservadas com bronopol na pesquisa de soro de queijo por cromatografia líquida de alta eficiência (CLAE). Foram testadas duas temperaturas de armazenamento (7 e 30º C) para as amostras contendo bronopol. As amostras sem conservante (controle) foram mantidas sob congelamento. Todos os grupos foram testados quanto ao tempo de estocagem por até oito dias. A cada grupo, foram adicionados cinco níveis de soro de queijo (0%, 2%, 5%, 10% e 20%). Não houve diferença (p > 0,05) entre os tratamentos, possibilitando concluir que é possivel utilizar amostras de leite cru conservadas com bronopol para pesquisa de soro de queijo em leite por CLAE. O tempo e a temperatura de armazenamento não interferiram nessa metodologia de pesquisa para amostras conservadas com bronopol, estocadas sob refrigeração ou à temperatura ambiente por até oito dias

Thirty-three dairies located in Vale do Mucuri, Vale do Jequitinhonha and North of Minas Gerais State, under federal, state or city inspection, registered in a food purchasing program aimed to distribute pasteurized milk to low income population sponsored by the Minas Gerais government, were evaluated. The adequation of those establishments in Good Manufacturing Practice were carried out using a check-list. The most common non conformities were related to general manufacturing and general quality control aspects, in which the highest number of itens with 100% of stablishment reproval were reported. By the results gathered, was concluded that none of the visited establishments were completly adequated to Good Manufacturing Pratices, then the quality of dairies products produced by them could not be guaranteeded
Trinta e três laticínios localizados no Vale do Mucuri, Vale do Jequitinhonha e Norte de Minas Gerais, sob inspeção federal, estadual ou municipal, cadastrados em um programa de aquisição de alimentos do governo de Minas Gerais para distribuição de leite pasteurizado para população de baixa renda foram avaliados. Utilizando-se um check-list, verificou-se a adequação desses estabelecimentos às Boas Práticas de Fabricação. Observou-se que as não conformidades mais observadas foram encontradas nos aspectos gerais de fabricação e nos aspectos gerais de controle de qualidade, nos quais se observou a maior quantidade de itens com 100% de reprovação dos estabelecimentos. The aim of this work was to detect Listeria monocytogenes added in milk, after selective enrichment period, by PCR technique. A PCR technique using the primers LM1/LM2 and a multiplex PCR with IAP1/IAP2 e LMA/LMB was standardized. The conventional method of L. monocytogenes detection was compared from milk samples with different bacterial count added with various L. monocytogenes concentrations. The DNA extraction methods using phenolchloroform and thermal split were compared. The DNA extraction and the conventional methods showed the same result in detecting the bacterium in samples of sterilized milk. In milk samples with any initial contamination, the conventional methodology was sensitive and PCR from enrichment broth can not detect L. monocytogenes added, independently of the extraction method used. The identification time of L. monocytogenes decreased using the DNA extraction by the strain split method using suspect colonies that grew in Oxford and PALCAM, in substitution of identification phenotypes tests
O objetivo deste trabalho foi detectar Listeria monocytogenes inoculada em leite, após um período de enriquecimento seletivo, por meio da técnica de PCR. Foi feita a padronização de uma PCR utilizando os oligonucleotídeos LM1/LM2 e de uma PCR multiplex com os oligonucleotídeos IAP1/IAP2 e LMA/LMB. Listeria monocytogenes foi inoculada em várias concentrações em leite esterilizado desnatado e em leite cru integral com dois níveis diferentes de contaminação. A PCR padronizada foi aplicada para identificar a bactéria inoculada experimentalmente e foi comparada com o método convencional de detecção de L. monocytogenes. Foram comparados, também, os métodos de extração de DNA utilizando fenolclorofórmio e por lise térmica, a partir de um caldo de cultura e diretamente da colônia suspeita. Somente foi possível identificar L. monocytogenes por PCR no leite esterilizado desnatado, após 48 horas de enriquecimento em meio seletivo (LEB). Neste caso, a sensibilidade do teste foi de 1 UFC/ml de leite, a mesma sensibilidade da metodologia convencional. Quando inoculada em leite cru integral com contaminação bacteriana, L. monocytogenes só foi detectada pela metodologia convencional, com uma sensibilidade de até 2 UFC/ml (leite com baixa contaminação). Foi possível reduzir o tempo de identificação de L. monocytogenes, substituindo os testes fenotípicos de identificação pela PCR padronizada, utilizando DNA bacteriano extraído pelo método de lise térmica, diretamente da colônia suspeita

The microbiological and physical-chemical quality of 151 pasteurized type C milk samples distributed by a governmental program in Minas Gerais state was analyzed. Milk quality was also traced to the dairies, in accordance to the type of inspection, as well as to the produce regions and season. For the microbiological analyses, means were: 6.09 x 101 MPN/mL and 2.47 x 101 MPN/mL for coliforms at 30°C and 45°C, respectively; 1.38 x 105 CFU/mL for counting of aerobic mesophilic bacteria; and 1.31 x 105 CFU/mL and 2.54 x 104 CFU/mL for counting of Staphylococcus spp. and Staphylococcus aureus, respectively. Salmonella spp. was identified in 121 (2.48%) samples. For the physical-chemical analyses, means were: 1.031 for density at 15°C, 12.40% for total solids content, 3.60% for fat, 8.80% for non-fat solids, 15.32°D for titratable acidity, and 0.525°H for freezing point. Mean of water added to milk was 2.4%. Positive result for alkaline phosphatase activity was observed in 23.18% samples and 22.52% presented negative activity of lactoperoxidase. Difference (p>0.05) was not observed among the dairy industries under different levels of inspection, and among different regions the State for the microbiological aspects; however, difference was observed (p<0.05) for the majority of the physicalchemical aspects. Difference was not observed among the seasons of the year concerning microbiological and physical-chemical aspects. It was concluded that the pasteurized milk distributed by the governmental program had unsatisfactory quality; deficient composition and can represent a potential risk to the health of the consumers
Este trabalho teve como objetivo analisar a qualidade físico-química e microbiológica de 151 amostras de leite pasteurizado tipo C distribuído em programa social governamental no Estado de Minas Gerais, assim como associar a qualidade deste leite aos laticínios conveniados ao programa de acordo com seu tipo de inspeção, meso regiões produtoras e época do ano. Para as análises microbiológicas, foram obtidas médias de 6,09 x 101NMP/mL e 2,47 x 101 NMP/mL para coliformes a 30°C e a 45°C, respectivamente; 1,38 x 105 UFC/mL para contagem de microrganismos mesófilos aeróbios, e 1,31 x 105 UFC/mL e 2,54 x 104 UFC/mL para contagem de Staphylococcus spp. e Staphylococcus aureus, respectivamente. Salmonella spp. foram identificadas em 2,48% de 121 amostras pesquisadas. Para as análises físico-químicas, foram obtidas médias de 1,031 g/mL para densidade relativa a 15°C, 12,40% de extrato seco total, 3,60% para gordura, 8,80% para extrato seco desengordurado, 15,32°D para acidez titulável e -0,525°H para crioscopia. A média de adição de água ao leite foi de 2,4%. Resultado positivo para a pesquisa de fosfatase alcalina foi observado em 23,18% e 22,52% apresentaram resultado negativo para a pesquisa de lactoperoxidase nas 151 amostras analisadas. Não foi observada diferença (p>0,05) entre os estabelecimentos sob diferentes níveis de inspeção e entre diferentes meso regiões para os parâmetros microbiológicos. No entanto, observou-se diferença (p<0,05) para a maioria dos parâmetros físico-químicos. Também não foi observada diferença (p>0,05) entre as épocas do ano em que o leite foi produzido para os aspectos microbiológicos e físico-químicos. Desta forma, concluiu-se que o leite pasteurizado distribuído no programa social governamental possui qualidade insatisfatória, composição centesimal deficiente e pode representar risco potencial à saúde de seus beneficiários

For over 50 years, the definition and classification of cardiomyopathies have remained anchored in the concept of ventricular dysfunction and myocardial structural remodelling due to unknown cause. The concept of idiopathic was first challenged in 2006, when the American Heart Association classification subordinated the phenotype to the aetiology. Cardiomyopathies were classified as genetic, acquired, and mixed. In 2008, the European Society of Cardiology proposed a phenotype-driven classification that separated familial (genetic) from non-familial (non-genetic) forms of cardiomyopathy. Both classifications led the way to a precise phenotypic and aetiological description of the disease and moved away from the previously held notion of idiopathic disease. In 2013, the World Heart Federation introduced a descriptive and flexible nosology—the MOGE(S) classification—describing the morpho-functional (M) phenotype of cardiomyopathy, the involvement of additional organs (O), the familial/genetic (G) origin, and the precise description of the (a)etiology including genetic mutation, if applicable (E); reporting of functional status such as American College of Cardiology/American Heart Association stage and New York Heart Association classification (S) was left optional. MOGE(S) is a bridge between the past and the future. It allows description of comprehensive phenotypic data, all genetic and non-genetic causes of cardiomyopathy, and incorporates description of familial clustering in a genetic disease. MOGE(S) is the instrument of precision diagnosis for cardiomyopathies. The addition of the early and unaffected phenotypes to the (M) descriptor outlines the clinical profile of an early affected family member; the examples include non-dilated hypokinetic cardiomyopathy in dilated cardiomyopathy and septal thickness (13–14 mm) in hypertrophic cardiomyopathy classes. Finally, in the recently released scientific statement of the American Heart Association on the classification and diagnosis of cardiomyopathy in children the consensus was to use a classification system based on a hierarchy incorporating the required elements of the MOGE(S) classification.

Cardiomyopathies are defined as disorders of the myocardium which are always associated with cardiac dysfunction and are aggravated by arrhythmias, heart failure and sudden death. There are different ways of classifying them. The American Heart Association has classified them in either primary or secondary cardiomyopathies depending on whether the heart is the only organ involved or whether they are due to a systemic disorder. On the other hand, the European Society of Cardiology has classified them according to the different morphological and functional phenotypes associated with their pathophysiology. In 2013 the MOGE(S) classification started to be published and clinicians have started to adopt it. The purpose of this review is to update it.
