Background. The stem bark decoction of Stemonocoleus micranthus Harms (Fabaceae) is most widely used traditionally as a remedy for various diseases such as malaria and boil. In this study, the anti-inflammatory and immunomodulatory activities of the methanol-dichloromethane extract (MDE) from the stem bark of the plant in rodents were evaluated. Methods. The carrageenan-induced rat paw oedema, cotton pellet-induced granuloma in rat, and xylene-induced ear oedema in mice were used to study the anti-inflammatory activity of methanol-dichloromethane extract of Stemonocoleus micranthus (MDESm) (100, 200, and 400 mg/kg). The effects of MDESm (100, 200, and 400 mg/kg) on cyclophosphamide-induced immunosuppression, neutrophil adhesion, carbon clearance, and haematological and biochemical parameters were carried out to study its immunomodulatory activity in mice. Result. MDESm (100 mg/kg, p.o.) significantly (p<0.05) inhibited carrageenan-induced oedema by 57.1% at 5th h posttreatment compared with control. At 100 mg/kg, p.o., MDESm significantly (p<0.05) reduced cotton pellet-induced granuloma by 39.28% and nonsignificantly reduced xylene-induced ear oedema by 34.1%. Treatment with MDESm (100 and 400 mg/kg) nonsignificantly abolished the neutropenia caused by cyclophosphamide with a percentage neutrophil reduction of 0 and −14.86%, respectively, while MDESm (200 mg/kg) and levamisole (50 mg/kg) had a nonsignificant reduction in neutrophil count (10.16 and 31.40%), respectively, all compared to the distilled water-treated group with a neutrophil count of −9.82%. MDESm at doses of 100 and 200 mg/kg increased phagocytic index by 0.0447 ± 0.00762 and 0.0466 ± 0.00703, respectively, although not significantly when compared to the control group with a value of 0.0226 ± 0.02117. There was a decrease in WBC and lymphocyte counts in MDESm- (200 mg/kg) treated group, suggesting immunosuppressive potential at this dose. MDESm caused a dose-dependent decrease in ALT and core liver enzymes, suggesting a hepatoprotective effect. The acute toxicity test revealed that MDESm is safe in mice with an oral lethal dose (LD50) of >5 g/kg. Conclusion. The methanol-dichloromethane extract of Stemonocoleus micranthus Harms possesses mild anti-inflammatory and immunomodulatory activities which may be more pronounced upon fractionation and purification. Therefore, more investigations are needed to explore these activities further.

Background Amid COVID-19, and other possible future infectious disease pandemics, dentistry needs to consider modified dental examination regimens that render quality care, are cost effective, and ensure the safety of patients and dental health care personnel (DHCP). Traditional dental examinations, which number more than 300 million per year in the United States, rely on person-to-person tactile examinations, pose challenges to infection control, and consume large quantities of advanced-level personal protective equipment (PPE). Therefore, our long-term goal is to develop an innovative mobile dentistry (mDent) model that takes these issues into account. This model supplements the traditional dental practice with virtual visits, supported by mobile devices such as mobile telephones, tablets, and wireless infrastructure. The mDent model leverages the advantages of digital mobile health (mHealth) tools such as intraoral cameras to deliver virtual oral examinations, treatment planning, and interactive oral health management, on a broad population basis. Conversion of the traditional dental examinations to mDent virtual examinations builds upon (1) the reliability of teledentistry, which uses intraoral photos and live videos to make diagnostic decisions, and (2) rapid advancement in mHealth tool utilization. Objective In this pilot project, we designed a 2-stage implementation study to assess 2 critical components of the mDent model: virtual hygiene examination (eHygiene) and patient self-taken intraoral photos (SELFIE). Our specific aims are to (1) assess the acceptance and barriers of mDent eHygiene among patients and DHCP, (2) assess the economic impact of mDent eHygiene, and (3) assess the patient’s capability to generate intraoral photos using mHealth tools (exploratory aim, SELFIE). Methods This study will access the rich resources of the National Dental Practice-Based Research Network to recruit 12 dentists, 12 hygienists, and 144 patients from 12 practices. For aims 1 and 2, we will use role-specific questionnaires to collect quantitative data on eHygiene acceptance and economic impact. The questionnaire components include participant characteristics, the System Usability Scale, a dentist-patient communication scale, practice operation cost, and patient opportunity cost. We will further conduct a series of iterative qualitative research activities using individual interviews to further elicit feedback and suggestion for changes to the mDent eHygiene model. For aim 3, we will use mixed methods (quantitative and qualitative) to assess the patient’s capability of taking intraoral photos, by analyzing obtained photos and recorded videos. Results The study is supported by the US National Institute of Dental and Craniofacial Research. This study received “single” institutional review board approval in August 2021. Data collection and analysis are expected to conclude by December 2021 and March 2022, respectively. Conclusions The study results will inform the logistics of conducting virtual dental examinations and empowering patients with mHealth tools, providing better safety and preserving PPE amid the COVID-19 and possible future pandemics. Monocular depth estimation based on unsupervised learning has attracted great attention due to the rising demand for lightweight monocular vision sensors. Inspired by multi-task learning, semantic information has been used to improve the monocular depth estimation models. However, multi-task learning is still limited by multi-type annotations. As far as we know, there are scarcely any large public datasets that provide all the necessary information. Therefore, we propose a novel network architecture Semantic-Feature-Aided Monocular Depth Estimation Network (SFA-MDEN) to extract multi-resolution depth features and semantic features, which are merged and fed into the decoder, with the goal of predicting depth with the support of semantics. Instead of using loss functions to relate the semantics and depth, the fusion of feature maps for semantics and depth is employed to predict the monocular depth. Therefore, two accessible datasets with similar topics for depth estimation and semantic segmentation can meet the requirements of SFA-MDEN for training sets. We explored the performance of the proposed SFA-MDEN with experiments on different datasets, including KITTI, Make3D, and our own dataset BHDE-v1. The experimental results demonstrate that SFA-MDEN achieves competitive accuracy and generalization capacity compared to state-of-the-art methods.

Background: Myelin oligodendrocyte glycoprotein (MOG) antibodies have been described in children with acute disseminated encephalomyelitis (ADEM), recurrent optic neuritis, neuromyelitis optica spectrum disorders and more recently in children with multiphasic disseminated encephalomyelitis (MDEM). Objective: To delineate the clinical, cerebrospinal fluid (CSF) and radiological features of paediatric MDEM with MOG antibodies. Methods: Clinical course, serum antibodies, CSF, magnetic resonance imaging (MRI) studies and outcome of paediatric MDEM patients were reviewed. Results: A total of 8 children with two or more episodes of ADEM were identified from a cohort of 295 children with acute demyelinating events. All children had persisting MOG antibodies (median titre: 1:1280). All ADEM episodes included encephalopathy, polyfocal neurological signs and a typical MRI. Apart from ADEM episodes, three children had further clinical attacks without encephalopathy. Median age at initial presentation was 3 years (range: 1–7 years) and median follow-up 4 years (range: 1–8 years). New ADEM episodes were associated with new neurological signs and new MRI lesions. Clinical outcome did range from normal (four of the eight) to mild or moderate impairment (four of the eight). A total of four children received monthly immunoglobulin treatment during the disease course. Conclusion: Children with MDEM and persisting MOG antibodies constitute a distinct entity of relapsing demyelinating events and extend the spectrum of MOG antibody–associated diseases.

Objetivou-se avaliar a composição química, vitreosidade e valores nutricionais de diferentes híbridos de milho através de análises laboratoriais e de um ensaio de metabolismo com suínos em crescimento. Foram utilizados 24 suínos em fase de crescimento (35,6 Kg ±± 3,05 kg de PV) machos castrados mestiços (LD x LW), os quais foram mantidos em gaiolas de metabolismo e distribuídos em um delineamento inteiramente ao acaso totalizando 24 parcelas. Os tratamentos experimentais consistiram de 6 rações com os seguintes híbridos de milho: milho óleo (Móleo), milho QPM (MQPM), milho dentado (Mdent), milho semidentado (Msemi) e dois milhos duros (Mdur1) e (Mdur2). Os valores obtidos de vitreosidade, coeficiente de digestibilidade da proteína bruta (CDPB) e energia digestível (ED) para Móleo, MQPM, Mdent, Msemi, Mdur1 e Mdur2 foram 78,5%; 71,7%; 57,2%; 68,2%; 75,9%; 82,8%; 81,68%; 80,14%; 82,35%; 69,71%; 76,98%; 75,61%; 3680; 3426; 3597; 3441; 3340 e 3469 Kcal/Kg, respectivamente. Foram observadas diferenças significativas (P<0,05) para os valores de vitreosidade, mostrando que os milhos de textura dura apresentam maior vitreosidade em relação ao semidentado e ao dentado. Houve diferenças significativas no CDMS, CDPB e ED entre os milhos estudados (P<0,05), mostrando que o milho óleo (Móleo) e o dentado (Mdent) foram superiores aos demais. Em relação ao nitrogênio retido (%NR), houve diferenças significativas (P<0,05) entre os milhos estudados. Para esta variável os milhos dentado, QPM e alto óleo mostraram-se superiores aos milhos semidentado e os duros. Conclui-se que os milhos dentado, Móleo e MQPM foram superiores aos demais.

Esta investigación resulta de la experiencia de los investigadores durante tres años consecutivos dictando la materia de Metodología de la Investigación (MR) en la Maestría en Gestión Educativa (MDEM) de la Universidad Espíritu Santo (UEES), detectando los malos hábitos lectores como una de las principales limitantes en los estudiantes. Existe la necesidad de profundizar el componente lector y la producción científica y académica, evidenciado por el diagnóstico aplicado en las diferentes promociones del 2019 al 2022.Para su desarrollo se consultó una extensa bibliografía considerando los vacíos en la investigación y la enseñanza relacionada con la lectura y la necesidad de incentivar a los estudiantes a leer. La aplicación de diferentes instrumentos científicos a los estudiantes del MDEM permitió diseñar estrategias metodológicas para estimular y desarrollar hábitos lectores en los estudiantes. La factibilidad de la propuesta fue evaluada y resultó en treinta publicaciones de estudiantes en diferentes revistas científicas indexadas, lo que demuestra la pertinencia e impacto de la investigación realizada en el contexto universitario.

A novel approach to load a hydrophilic bovine serum albumin (BSA) into the drug carriers was proposed in terms of thermosensitive dual-shell structures which were fabricated with poly(N-isopropylacrylamide), poly(lactide) and Au nanoparticles. Spherically well-defined dualshell structures were constructed by a modified-double-emulsion method (MDEM). The lower critical solubility temperature of the structures was shifted to 36.4 °C which was confirmed by UVVis spectroscopy.

Les canaux potassiques (Kir) à rectification entrante sont des protéines transmembranaires impliquées dans des processus physiologiques fondamentaux. Des pathologies existantes sont directement liées à des mutations de ces canaux : des mutations au niveau de Kir2.1 et Kir6.2 provoquent le syndrome d'Andersen et l'hyperinsulinisme respectivement. Cette étude a quatre objectifs: i) comprendre le mécanisme moléculaire d’ouverture et de fermeture de KirBac3.1, ii) comprendre l'impact de la mutation W46R sur le canal, iii) comprendre l'impact de la mutation S129R, une mutation conçue pour stabiliser l’état ouvert du canal, iv) résoudre la structure de Kir2.1 humain par cryo-microscopie. Pour étudier le comportement de ce système, la dynamique moléculaire utilisant la méthode des modes normaux excités (MDeNM) a été employée. Les résultats ont été confrontés à la méthode de HDX-MS et à des expériences d’électrophysiologie pour validation. Pour déterminer la structure de Kir2.1 WT, nous avons utilisé la cryo-microscopie électronique combinée à l’analyse d’images. Des mouvements clés du mécanisme d’ouverture/fermeture du canal ont été détectés comme l'implication du domaine cytoplasmique, des hélices de glissement et transmembranaires. Concernant KirBac3.1 W46R, une modification des interactions entraîne la rupture des interactions entre le bas des deux hélices transmembranaires provoquant le soulèvement de l'hélice de glissement facilitant l’ouverture du canal. La mutation S129R repousse les points de constriction et maintient le canal ouvert avec un rôle surprenant de la mutation R129. L’étude de Kir2.1 par cryo-microscopie a donné une densité électronique de résolution de 7 Å
Inwardly-rectifying potassium (Kir) channels are transmembrane proteins involved in fundamental physiological processes. Pathologies are directly linked to a mutation of these channels: some mutations in Kir2.1 and Kir6.2 cause the Andersen’s syndrome and hyperinsulinism, respectively. This study has four objectives: i) to understand the gating mechanism of the KirBac3.1, ii) to understand the impact of the mutation W46R on the channel (W68R equivalent in Kir6.2 causing hyperinsulinsm), iii) to understand the impact of the mutation S129R on the channel, iv) to resolve the structure of Kir2.1 at an atomic level. To study the dynamical characteristics of this system, Molecular Dynamics using excited Normal Modes (MDeNM) method was used. The results were confronted to HDX-MS Spectrometry and electrophysiological experiments for validation. To determine the structure of the human Kir2.1 WT, we are using cryo-electron microscopy combined with image analysis. Key motions in the gating mechanism were determined like the involvement of the cytoplasmic domain, the slide-helix and the transmembrane helices during the opening of the channel. Concerning W46R, a modification in these interaction at the level of the residue 46 causes the loss of interactions between the bottoms of the two transmembrane helices that triggers the uprising of the slide-helix which opens the channel. The study of KirBac3.1 S129R mutant in the open state explains how the mutation pushes away the point of constriction and keeps the channel open with a surprising role of the mutation R129. We investigated the structure of Kir2.1 by cryo-EM and obtained an electronic density at 7 Å resolution

Diante do crescimento das vendas de smartphones, em específico com o sistema operacional Android, em suas variadas versões e diversidade de aparelhos, um problema chamado de fragmentação foi destacado. Com o incremento de dispositivos multinúcleos e multiprocessadores, crescem as restrições na potência e ocorrência de superaquecimento. Alguns trabalhos consultados se especializam em resolver o problema da fragmentação, porém não se preocupam com dados energéticos, enquanto outros somente relatam técnicas que permitem a diminuição no consumo energético, mas nenhum possibilita a integração de testes multidispositivos, com o intuito de solucionar o problema da fragmentação, com foco energético. Este trabalho oferece a modelagem e implementação de um ambiente de medição que auxilia na avaliação do consumo energético de dispositivos com o sistema operacional Android. O ambiente proposto permite que seja feita uma análise energética de dispositivos em diferentes frequências no processador e redes de dados de forma simultânea e com manipulação pela Web. Uma infraestrutura genérica de medição, a qual permite a coleta de tensão e corrente de dispositivos alimentados por bateria foi criada juntamente com um canal que permite a comunicação com um computador para análise dos dados. No tangente ao software, foi construída uma plataforma Web para manipulação de componentes do dispositivo e replicação de testes de forma automatizada. Para testar o uso do ambiente proposto, foram executados oito testes envolvendo Navegação Web, Streaming de Vídeo e Carga de Processamento nos smartphones disponíveis. Os resultados dos testes são apresentados e discutidos em detalhe.
Recently we can observe a growth in smartphone sales, in particular those running Android operating system. In several versions and devices, a problem known as fragmentation was featured. Along with the increase of multi-core and multi-processor devices, growing restrictions on power and overheating were highlighted. Some studies focus on solving the fragmentation problem and do not care about energy analysis, while other reports only techniques which allow a reduction in energy consumption, but none enables the integration of multi-devices tests in order to solve the problem of fragmentation with energy focus. This study provides the modeling and implementation of a measurement environment that helps the evaluation of the energy consumption of devices with the Android operating system. The proposed environment makes possible an analysis of the energy in devices at different processor frequencies and data networks on multiple devices simultaneously with Web access. A generic and cheap infrastructure measurement, which allows the collection of voltage and current of battery-powered devices and a channel that allows communication with a computer for data analysis were built. In regard to the software, a Web platform for manipulation of device peripherals and automated tests replication was constructed. In order to test the use of the proposed environment, eight tests were performed covering Web Browsing, Video Streaming and CPU load on the available smartphones, and then the results were explained and discussed.

Les convertisseurs analogiques-numériques fondés sur le principe de la modulation §¢ sont capables de fonctionner à des résolutions très élevés. L'utilisation en interne d'un CAN et d'un CNA multibit permet de réduire le taux de suréchantillonnage, les contraintes imposées par les circuits actifs, amé- liore la stabilité de la boucle du modulateur, mais rend celui-ci très sensible aux imperfections des composants du convertisseur numérique analogique (CNA) interne situé dans le chemin de retour. Les erreurs statiques dues aux non idéalités des circuits constitutifs de ce CNA peuvent être corrigées au moyen de techniques d'appariement dynamique des composants (DEM). Ce travail de thèse est consacré entre autre à l'étude théorique de ces techniques de correction des défauts des cellules des CNA multibits. Après avoir rappelé le principe de la conversion §¢ d'une part, et les différentes sources d'erreurs dominantes dans le cas multibit d'autre part, les techniques d'appariement existantes sont analysées et comparées. Nous soulignons les avantages, les inconvénients, et les domaines d'applications préférentiels de chacune. Le coeur du travail consiste en la proposition de quatre nouvelles techniques d'appariement dynamique. Les deux premières dérivent de la méthode de la moyenne des données (DWA), l'une pour le cas passe-bas du premier ordre, l'autre dans le cas passe-bande du second ordre. Les deux dernières propositions (appelées MDEM et STDEM) dérivent des deux algorithmes de tri (SDEM) et d'arborescence (TDEM) : elles conviennent à une mise en forme des erreurs d'ordre élevé et sont destinées aux applications passe-bas et passe-bande de haute performance. Ces quatre méthodes proposées ont été mises en équation et leurs performances confirmées par diverses simulations. Une implantation des algorithmes MDEM et STDEM a été faite au niveau cellule standard jusqu'à l'étape finale de routage en technologie CMOS 0.35 ¹m. L'ensemble des résultats des simulations au niveau système et au niveau transistor conforme l'avantage des techniques développées dans ce travail en termes de surface occupée et aussi de fréquence maximale d'application, si on les compare avec les algorithmes conventionnels de SDEM. Dans une dernière partie, les erreurs dynamiques du CNA, en particulier l'effet de la gigue d'horloge, le glitch, la dissymétrie des temps de transition, l'injection de charge (CFT) et la métastabilité du quantificateur sont également analysés. A l'issue de ces réflexions, une nouvelle cellule de CNA incluant un bloc limitant la plage dynamique de la commande d'entrée (SRD) est proposée. Elle possède une structure de remise à zéro partielle (semi-RZ) qui permet de bénéficier à la fois de l'avantage de la cellule RZ et non RZ. De plus, l'effet du retard du bloc de DEM est compensé par une modification dans l'architecture convenant aux applications passe-bande haute fréquence.

<em>Abstract</em>.-The sea lamprey <em>Petromyzon marinus </em>is a widely distributed anadromous species spawning in coastal rivers and streams throughout the north Atlantic basin. In this paper, we review aspects of sea lamprey migration and ecology that relate to the transport of nutrients and materials to and from freshwater ecosystems and provide an example of a long-term study of a native wild population. Several aspects of lamprey life history (rapid growth in marine phase, many adults spawn in upper reaches of small oligotrophic rivers, all adults die after spawning) suggest that anadromous sea lampreys contribute marine-derived nutrients and materials (MDNM) to freshwater ecosystems. We used long-term (20 years) data on spawner abundance, along with literature-derived concentration values, to estimate the import of nutrients and materials to a spawning reach of the Fort River, a tributary of the Connecticut River in western Massachusetts, USA. Sea lamprey imported as much as 0.26 g of P per square meter of stream, as much as of 20% of the total annual P loading to a similar system where a full P budget has been developed. While the MDNM contribution of sea lamprey may be substantial, other aspects of their life history and habitat use may limit the overall magnitude and direction of lamprey influence on freshwater ecosystems. Spawning requirement for rocky substrate within a narrow size range may limit import at the watershed scale. In addition, marine survival rates of less than ~1% will result in a net export of nutrients and materials via out-migrating juveniles (transformers). While there is currently no information on survival rates in wild anadromous populations, the tight link between adult survival and prey/host fish populations observed in landlocked Great Lakes systems suggests that the ecological role of sea lamprey may be strongly related to the abundance of coastal marine fishes. Further research on adult survival, juvenile dispersal and distribution, and the paths of nutrient and material uptake in spawning streams are necessary to more fully evaluate the role of anadromous sea lamprey in the transport of MDNM.

Recent years have seen a sharp increase in the development and usage of Additive Manufacturing (AM) technologies for a broad range of scientific and industrial purposes. The drastic microstructural differences between materials produced via AM and conventional methods has motivated the development of computational tools that model and simulate AM processes in order to facilitate their control for the purpose of optimizing the desired outcomes. This paper discusses recent advances in the continuing development of the Multiphysics Discrete Element Method (MDEM) for the simulation of AM processes. This particle-based method elegantly encapsulates the relevant physics of powder-based AM processes. In particular, the enrichment of the underlying constitutive behaviors to include thermoplasticity is discussed, as are methodologies for modeling the melting and re-solidification of the feedstock materials. Algorithmic improvements that increase computational performance are also discussed. The MDEM is demonstrated to enable the simulation of the additive manufacture of macro-scale components. Concluding remarks are given on the tasks required for the future development of the MDEM, and the topic of experimental validation is also discussed.

Physically accurate modeling of powder-based additive manufacturing (AM) processes can play an enabling role for both the certification and qualification as well as the functional tailoring of materials produced by these processes. In an effort to address these needs in a computationally efficient and physically realistic manner, this paper presents the initial efforts towards the development of a methodology for simulating polydisperse particle-based AM processes by the use of the Multiphysics Discrete Element Method (MDEM). We discuss the formulation of a DEM framework for addressing the unique multiphysics behavior of AM materials and processes. In particular, we focus on coupled thermo-mechanical effects that result in residual strains and deformation. The MDEM approach is demonstrated on several test problems involving laser sintering of metal powders. The paper concludes with a discussion on how this approach may be generalized to broader classes of AM systems, and details are given regarding future work that must be accomplished in order to further develop the present methodology.

Abstract To achieve an acceptable level of zonal isolation, well integrity should be guaranteed in hydrocarbon production and geological CO2 sequestration. Well pressure test can cause different types of failures in the well system leading to leakages through these failures. Laboratory evidences have revealed that occurrence of radial tensile fractures is likely during pressure tests. In this paper, we use a numerical code call MDEM which was formulated based on discrete element method. The code can model discontinuum feature of fractures. A model of a lab-sized pressure test was built and compared to an experiment previously published. The model was tested under different confinement levels and effect of the tensile strength of rock on the radial fracture was investigated at the same lab-scale. Fracture opening profiles are also presented showing the leakage potential of these fractures under different pressure level.
