Abstract3A0729+103 (= BG CMi) is an intermediate polar discovered through its X-ray emission (McHardy et al. 1981, 1984). The orbital period is 3.235 hours and the rotation period is 15.2 minutes, For ephemeris and references on the source we refer to McHardy et al. (1984). We report here on optical (4025 to 5090 A) and ultraviolet (1200 to 3200 A) spectroscopy obtained, respectively, on Dec 1, 1984 and April 21, 1985. Our data show clear modulation of spectral features with the orbital period.

Julien McHardy draws on his work with the alternative publishing collective Mattering Press to explore the importance of opening up and maintaining “bewildering spaces within” the increasingly normative and clear-cut regimes of evaluation. Drawing on a range of examples from alternative publishing, the artwork “The Right to be Lazy” and concepts of care, love, qualculation and laziness, McHardy argues that opening up space for otherness within are crucial to good academic work. At the same time, he also warns us that caring for difference is a struggle that includes making radical cuts and disconnections, and in practice often may build on precarious and invisible work. This leads him to asks what academic institutions would look like that could value and cultivate that which they cannot evaluate.

The focus of this article is on compiling a biography of Reverend William McLeod, a secular priest who served Corgarff from 1800 to 1804 and Braemar from 1804 until his death in 1809. This task is complicated by the fact that he was known by some as McLeod, and by others as McHardy, while Dom Odo Blundell in his 1909 \textit{The Catholic Highlands of Scotland} referred to him as McLeod alias McHardy. This article makes use of documents that have come to light, during the course of the last century, which not only assist in explaining who this enigmatic priest was, but also suggest possible reasons for the confusion surrounding his name. Although his service to the Church was interrupted by the tuberculosis which caused his death, the details of his life, including his presence in Braemar during a particularly turbulent period in this Mission's history, help to expand our knowledge of Catholicism in the north-east Highlands during the late eighteenth and early nineteenth centuries.

AbstractSome aspects of power-spectral densities (PSD) of active galactic nuclei are similar to those of galactic black hole X-ray binary systems (McHardy et al. 2005). The signal originates near a black hole and its modulation by general-relativistic effects should be taken into account (Życki & Nedźwiecki 2005). We modified the previous calculations of these effects, assuming a model of spots which occur on the disc surface and decay with a certain lifetime.

AbstractScaling relations are the most powerful astrophysical tools to set constraints to the physical mechanisms of astronomical sources and to infer properties for objects where they cannot be accessed directly. We have re-investigated one of these scaling relations using powerful type 1 Seyferts; the so-called X-ray variability plane (or mass-luminosity-timescale relation, McHardy et al.2006). This relation links the power-spectral density (PSD) break frequency with the SMBH mass and the bolometric luminosity. We used all available XMM-Newton observations to study the PSD and spectra in short segments within each observation. This allows us to report for the first time that the PSD break frequency varies for each object, showing variations in 19 out of the 22 AGN analyzed. Our analysis of the variability plane confirms the relation between the break frequency and the SMBH mass and finds that the obscuration along the line of sight (or the variations on the obscuration using its standard deviation) is also a required parameter. We constrain a new variability plane of the form: log(vBreak) = – A log (MBH) + B log (NH) – C (or log(vBreak) = – A log (MBH) + B Δ (NH) + C). The X-ray variability plane found by McHardy et al. (2006) is roughly recovered when we use unobscured segments. We speculate the PSD shape is related with the outflowing wind close to the accretion disk at least for these powerful type 1 AGN (Gonzalez-Martin et al. 2018).

Foram validados dois sistemas de alerta na requeima do tomateiro, no município de Caçador-SC em dois ciclos de cultivo. No primeiro ciclo, os métodos de MacHardy modificado e Colpam 40®, este com valor de severidade acumulado da doença (VS) igual a 10, como alerta para a pulverização de fungicidas de contato e comparados ao sistema convencional de calendário no híbrido Carmen e na cultivar Santa Clara. Em relação ao convencional, a indicação de pulverizações pelos sistemas de alerta permitiu uma redução na aplicação de fungicidas de 33,3% e 48,1%, pelos métodos de MacHardy modificado e Colpam 40®, respectivamente. A produtividade foi maior no híbrido Carmen do que na cv. S. Clara, mas em cada um destes materiais não houve diferença entre os sistemas de alerta e o convencional. Na cv. Carmen não houve diferença no controle da requeima na comparação do sistema convencional com o Colpam 40®. No segundo ciclo, usou-se o híbrido Carmen e os sistemas de alerta McHardy e Colpam 40 ® este com VS igual a 8 e 10 comparando-os ao sistema convencional. Não houve diferença na produtividade entre os tratamentos. Em relação ao número de pulverizações houve redução de 42,8% e 60,7% para VS igual a 8 e 10, respectivamente, com o sistema de alerta do Colpam 40® e de 39,2% com o de McHardy. Na avaliação da requeima, não houve diferença entre os tratamentos, exceto para o maior VS de alerta monitorado segundo o Colpam 40® cuja intensidade da doença foi maior. Este trabalho demonstrou que o uso destes dois sistemas de alerta da requeima, MacHardy e Colpam 40, poderiam ser ferramentas úteis para os produtores de tomate na tomada de decisão da pulverização em tomate na região de Caçador, SC.

PKS 1413+135 is an enigma: while classified as a BL Lac due to its polarized near-IR continuum (Stocke et al. 1992) and optical spectrum (Beichmann et al. 1981), it appears to lie within a spiral host (McHardy et al. 1991, Stocke et al. 1992). In addition, the AGN is highly obscured (Beichmann et al. 1981, Carilli et al. 1992, Stocke et al. 1992, Wiklind & Combes 1994, 1995). Yet there is no evidence that the absorbing gas is being heated and re-emitting the AGN radiation in the form of thermal IR or emission lines (as in, e.g., Sey 2s). This led Stocke et al. (1992) to suggest that the AGN might be background to the optical galaxy.

Abstract This study is a corpus-driven examination of frequent lexical words and keywords in the news texts related to the American Dream and the Chinese Dream. Based on Sinclair’s (Sinclair, John McHardy. 2004. Trust the Text. Routledge: London) five categories of co-selection as framework, it discusses the patterns of co-selection across the corpora of news texts, with a particular focus on the cumulative effects of the co-construction of situated meanings and establishment of ideological positions associated with the two dreams. The corpus linguistic tool Wordsmith is used to generate frequent words and keywords for detailed concordance analysis along both syntagmatic and paradigmatic relations in order to indicate collocation, colligation, semantic preference, and semantic prosody. The findings demonstrate the individualistic home, work and education associations of the American Dream versus the collectivistic attributions of the Chinese Dream of national rejuvenation. The study not only confirms different cultural practices, but also reveals different social-historical conditions, and political influences associated with media representations of the American Dream and the Chinese Dream.

The BL Lacertae objects, OJ 287, has long been known to exhibit large amplitude variations on time scales as short as a few days. The time rate of change in brightness for these variations has been observed to be in excess of 1.0 mag. per day on several occasions. However, the nature of the optical variations on time scales significantly shorter than a day has not been well established, and in many cases the results have been controversial (Carrasco et al, 1985, Lawrence et al 1987, McHardy and Czerny, 1987). In order to address this problem, we have obtained observations of OJ 287 on four different nights during the past two years using the KPNO 0.9 meter telescope equipped with the direct CCD camera. Variations were detected for OJ 287 on each night with ranges of 0.05 mag. to 0.10 mag. The variations detected on 1986 November 9 are typical of those observed on each of the nights and is shown in Figure 1. A power spectrum analysis of the data was performed and no convincing evidence for the presence of any periodicity was found. Based on this analysis, the short term optical variations observed for OJ 287 have a very different character than the rapid x-ray flickering observed for Seyfert galaxies which have been characterized by 1/f - noise. Therefore, these observations would imply sizes for the source region 3 × 10−5 pc. If one assures that the underlying engine is a supermassive blackhole, then the time scales of the variability suggests masses for the blackhole of 107M⊙ – 108M⊙.

Abstract Background: Pancreatic ductal adenocarcinoma (PDAC) is the second most significant cause of cancer-related mortality, with a 5-year survival rate of just 11%. PDACs are one of the most difficult to treat cancers, mainly because of the desmoplastic tumor microenvironment enriched with tumor-supportive extracellular matrix (ECM) and stromal cells. Unfortunately, the efficacies of current pancreatic cancer therapies are limited partly by stroma-mediated drug resistance. We reason that Discoidin Domain Receptors, which belong to the receptor tyrosine kinase family and bind to fibrillar collagens present in the ECM, may be a critical promoter of PDAC growth and progression. Though several drugs targeting DDR proteins have been developed, no inhibitors specific to DDR proteins have been developed. Moreover, those drugs have not been approved for treating PDACs. Methods. A high throughput screen using small molecule inhibitors was performed to identify bonafide inhibitors of DDR proteins. Structural and reported-based essays were done to determine the specificity. Functional assays were performed to test the potency of the candidate inhibitors. Mechanistic studies were done using RNA-seq, real-time PCR, and western blot. Results: Using an unbiased screening approach, we identified inhibitors that specifically target DDR proteins. Our studies revealed that small molecule inhibitors blocked the short and long-term viability as well as migration/invasion of PDAC cells. In addition, they induced apoptosis and halted the cell cycle at the G1-S checkpoint. Mechanistic studies revealed that the pro-growth signaling cascade was significantly inhibited. Conclusions: Our results showing specific DDR inhibitors blocking the growth and progression of PDAC cells without affecting the viability of normal cells suggest that these inhibitors may serve as a safe and potent regimen for treating pancreatic cancers. Abstract: The editors’ intention in publishing this selection of 190 petitions to the English royal government from the county of Lincolnshire is to reveal the range of problems the king’s subjects faced in the region. They provide a fine introduction that discusses the characteristics of the records, including the rhetorical strategies employed in petitioning, and the political and economic background of life in the county. Lincolnshire was a large region, mostly agricultural in nature but there were a few mid-sized towns, notably the county seat, Lincoln, and the port of Boston. Their problems were rather different from those of the villages, and the editors devote a section of the introduction to these urban issues. Another section of the introduction addresses ecclesiastical matters, and a brief conclusion lays out the editorial protocols the editors have followed. The editors attempt to date each petition, offer a transcription and translation of each and provide copious and very helpful notes as well as other pertinent records to allow for the identification of parties and witnesses and the proper understanding of the disputes that led men and women – mostly men – of all stations to seek the aid of the crown. There is an excellent index, which will facilitate use of the volume.

In an article, ‘The Slaves were Happy’: High School Latin and the Horrors of Classical Studies, Erik Robinson, a Latin teacher from a public high school in Texas, criticises how, in his experience, Classics teaching tends to avoid in-depth discussions on issues such as the brutality of war, the treatment of women and the experience of slaves (Robinson, 2017). However, texts such as the article ‘Teaching Sensitive Topics in the Secondary Classics Classroom’ (Hunt, 2016), and the book ‘From abortion to pederasty: addressing difficult topics in the Classics classroom’ (Sorkin Rabinowitz & McHardy, 2014) strongly advocate for teachers to address these difficult and sensitive topics. They argue that the historical distance between us and Greco-Roman culture and history can allow students to engage and participate in discussions that may otherwise be difficult and can provide a valuable opportunity to address uncomfortable topics in the classroom. Thus, Robinson's assertion that Classics teaching avoids these sensitive topics may not be so definitive. Regardless, Robinson claims that honest confrontations in the classroom with the ‘legacy of horror and abuse’ from the ancient world can be significantly complicated by many introductory textbooks used in Latin classes, such as the Cambridge Latin Course (CLC), one of the most widely used high school Latin textbooks in use in both America and the United Kingdom (Robinson, 2017). In particular, Robinson views the presentation of slavery within the CLC as ‘rather jocular and trivialising’ which can then hinder a reader's perspective on the realities of the violent and abusive nature of the Roman slave trade (Robinson, 2017). As far as he was concerned, the problem lay with the characterisation of the CLC's slave characters Grumio and Clemens, who, he argued, were presented there as happy beings and seemingly unfazed by their positions as slaves. There was never any hint in the book that Grumio or Clemens were unhappy with their lives or their positions as slaves, even though, as the CLC itself states in its English background section on Roman slavery, Roman law ‘did not regard slaves as human beings, but as things that could be bought or sold, treated well or badly, according to the whim of their master’ (CLC I, 1998, p. 78). One might argue, therefore, that there seems to be a disconnect between the English language information we learn about the brutality of the Roman slave trade provided in the background section of Stage 6, and what we can infer about Roman slavery from the Latin language stories involving our two ‘happy’ slaves.

Abstract Background: Early detection of cancer is vital to improve patient survival, since earlier diagnosis and treatment maximizes the opportunity to combat or control disease progression. Specifically for colorectal cancer (CRC), the average 5-survival rate after diagnosis decreases from 91% in early-stage CRC, to as low as 15% for stage IV CRC. Furthermore, the rapid detection and subsequent removal of pre-cancerous adenomas - e.g., advanced adenomas (AA) - can improve survival rates of affected patients. Current stool-based tests, such as FIT and FIT-DNA, have poor AA sensitivities of 24% and 42%, and specificities of 95% and 87%, respectively. Recent research into liquid biopsies have shown some promise, however tests based on tumor-derived biomarkers alone have limited sensitivity, especially in early-stage disease. Methods: We initially applied the Dxcover® Cancer Liquid Biopsy for use as a multi-cancer early detection (MCED) test. The test uses Fourier transform infrared (FTIR) spectroscopy and machine learning algorithms to build a classifier of the resultant spectral profiles to detect cancer, and can be fine-tuned to maximize either sensitivity or specificity depending on the requirements of specific international healthcare systems. Additionally, we have analyzed a retrospective cohort of serum samples comprising 100 CRC, 99 advanced adenomas removed by surgical resection and 97 colonoscopy screening controls. Results: The CRC classifier from the discovery MCED dataset reported an area under the receiver operating characteristic curve (AUROC) value of 0.91, with 74% sensitivity and 91% specificity when differentiating CRC and non-cancer, which surpasses the targets set by the Centers for Medicare & Medicaid Services (CMS) for coverage of CRC tests. When tuned for higher sensitivity, the model produced 97% sensitivity (49% specificity), and when tailored for greater specificity (97%) the sensitivity was 47%. In this study, we have progressed these findings to examine the ability of the technology to differentiate patients with CRC, AA and colonoscopy controls. Conclusions: Cancer treatment is more effective when given earlier and this low-cost strategy can facilitate the requisite earlier diagnosis. Abstract Background: Pancreatic ductal adenocarcinoma (PDAC) is ranked as the second leading cause of cancer-related mortality with a 5-year survival rate of merely 12%. The complexity of PDAC lies in its intrinsic resistance to treatment, attributable to the desmoplastic tumor microenvironment, characterized by an abundance of tumor-promoting extracellular matrix (ECM) and stromal cells. We hypothesize that Discoidin Domain Receptors (DDR), members of the receptor tyrosine kinase family known for their affinity to fibrillar collagens within the ECM, play an important role in promoting PDAC growth and progression. While several drugs targeting DDR proteins have been developed, no inhibitors to DDR proteins are specific or approved for PDAC treatment, thus necessitating the development of novel therapeutic interventions. Methods. A comprehensive high-throughput screening approach employing small molecule inhibitors was executed to identify authentic inhibitors targeting DDR. Subsequently, a rigorous evaluation of specificity of these small molecules was conducted through structural activity relationship (SAR) analyses and reference-based assessments. The efficacy of candidate inhibitors was assessed through functional assays, while mechanistic insights were gleaned using a multifaceted approach, including RNA-seq, real-time PCR, western blot analysis, in vivo experimentation, and Phospho-Receptor Tyrosine Kinase (RTK) profiling to discern any unintended off-target effects. Results: Using an unbiased screening approach, we identified 13 inhibitors that specifically target DDR. SAR studies and cellular thermal shift assay validated CIDD-0108633 (8633) as a bona fide DDR2 inhibitor. Functional assays and RNA-seq analyses demonstrated 8633's capacity to inhibit PDAC growth, metastatic potential, and expression of DNA replication, cell-cycle, and DNA damage repair genes. Combination therapy of 8633 with gemcitabine synergistically reduced PDAC cell viability and metastatic potential. Mechanistic studies revealed the role of ABC transporters in mediating gemcitabine resistance. RTK profiling shows moderate kinase selectivity. In vivo studies affirmed its antitumor effects. Conclusions: The results indicate that the selective inhibition of DDR with small molecule inhibitors suppresses PDAC cell growth and progression, implying that DDR inhibitors have the potential to be developed as safe and efficacious therapeutics for the treatment of pancreatic cancers. Fusarium wilt, caused by the soil-borne fungus Fusarium oxysporum f. sp. cubense (Foc) race 1, is a major disease of bananas in East Africa. Triploid East African Highland (Matooke) bananas are resistant to Foc race 1, but the response of diploid (Mchare and Muraru) bananas to the fungus is largely unknown. A breeding project was initiated in 2014 to increase crop yield and improve disease and pest resistance of diploid and triploid East African Highland bananas. In this study, eight Mchare cultivars were evaluated for resistance to Foc race 1 in the field in Arusha, Tanzania. In addition, the same eight Mchare cultivars, as well as eight Muraru cultivars, 27 Mchare hybrids, 60 Matooke hybrids and 19 NARITA hybrids were also screened in pot trials. The diploid Mchare and Muraru cultivars were susceptible to Foc race 1, whereas the responses of Mchare, NARITAs and Matooke hybrids ranged from susceptible to resistant. The Mchare and Matooke hybrids resistant to Foc race 1 can potentially replace susceptible cultivars in production areas severely affected by the fungus. Some newly bred Matooke hybrids became susceptible following conventional breeding, suggesting that new hybrids need to be screened for resistance to all Foc variants.

East African diploid cooking bananas, commonly called Mchare, are a staple crop for millions of subsistence farmers in Tanzania, particularly in the Pangani region in northern Tanzania. Several pathogens constrain Mchare production significantly and threaten food security. Sources of resistance to these pathogens have been identified; however, partial male and female sterility impedes successful resistance introgression, complicating the breeding process. Mchare cultivars are also the only known surviving representatives of a diploid banana subgroup that contributed unreduced gametes to many of the most widely grown and successful triploid dessert bananas (‘Cavendish’, ‘Gros-Michel’, ‘Silk’, and ‘Prata’). As such, they represent an essential intermediate step in the conventional improvement of bananas worldwide. We assess the amount and viability of pollen among Mchare and wild genotypes to identify the most fertile Mchare cultivars that can be used in conventional banana improvement. Pollen was collected from 14 banana genotypes for quantification and viability testing over 7 months, and the optimal time for pollen collection was determined to be 0800 HR. Significant variation among banana genotypes in terms of both overall pollen production and percentage of pollen viability was observed. The wild-type bananas ‘Calcutta 4’ (Indian Trade Classification (ITC) 0249] and ‘Borneo’ (ITC0253) had the greatest overall pollen production (> 31,000 pollen grains/anther) and viability (∼74%), whereas ‘Ijihu Inkundu’ (ITC1460; Mchare genotype) was the least productive (almost completely sterile), with an average pollen production of a few hundred grains per anther and a viability of 7%. There were significant differences among months in terms of pollen viability, with the greatest average viability observed in May, April, and February (> 51%), and the lowest average pollen viability in July (41%). Significant differences were observed among the Mchare genotypes, with ‘Huti-White’, ‘Huti green bell’ (ITC1559), and ‘Mchare Laini’ consistently producing more substantial amounts of total pollen and an overall more significant proportion of viable pollen. This information is vital to improve Mchare bananas and the global breeding of dessert bananas. The choice of Mchare banana used in improvement programs could affect fertility and the likelihood of breeding success.

Abstract Background: Colorectal cancer (CRC) is the third most common cancer worldwide, with almost 2 million new cases reported in 2020. CRC develops over time from a pre-cancerous stage (adenoma) - the ‘adenoma-carcinoma sequence’. Pre-cancerous lesions, such as Higher Risk Adenomas (HRA), harbour an increased risk of developing into CRC and their features are defined according to size, number and histological features. Screening for signs of CRC is recommended for average-risk adults using faecal immunochemical testing (FIT) in the UK, to enable the removal of pre-cancerous lesions and treatment of early-stage cancers. However, the performance of FIT is limited. Approximately 10% of CRC diagnoses have a negative FIT result, and it currently has ~93% false positive rate. Moreover, the sensitivity for detecting AA is only 24%, highlighting the need for alternative strategies. Methods: A promising strategy for earlier CRC detection is the clinical use of Fourier transform infrared (FTIR) spectroscopy. The Dxcover® Colorectal Cancer Liquid Biopsy is a rapid multi-omic test that interrogates a blood sample with infrared (IR) radiation and produces a distinctive signature that represents the whole biomolecular profile of the sample. Rather than focusing on individual biomarkers, the technique encompasses the full range of diagnostic information from both the tumor and the non-tumor response. In the initial CREATE (ColoREctal Cancer & Adenoma Test Evaluation) feasibility study, the potential of the technology was highlighted in a retrospective cohort of patients comprising 100 CRC, 92 adenoma samples and 104 colonoscopy screening controls diagnosed as non-cancer. At 90% specificity, the test reported 80% CRC sensitivity and 59% HRA sensitivity. CREATE-2 is a prospective, observational, multicenter study, currently recruiting patients across the United States. The primary objective of the study is to determine the diagnostic accuracy of the blood-based Dxcover Colorectal Cancer Liquid Biopsy for CRC and HRA, and collectively as advanced colorectal neoplasia. Primary outcomes are sensitivity, specificity, negative predictive value, and positive predictive value of the test as compared to the standard of care screening modality. The study population is comprised of patients aged 45-84 considered ‘average-risk’ for CRC, undergoing routine CRC screening as standard of care. Patients are treatment naïve for CRC/HRA and must be willing to consent to blood draw pre-bowel preparation, and follow-up for 24 months. A rapid blood test that is sensitive to pre-cancerous lesions and early-stage CRC could substantially improve patient outcomes. Current screening programs have addressable shortcomings, and the emergence of new technologies is essential to support earlier CRC detection. Citation Format: James M. Abstract Background: Glioblastoma (GBM) is the most prevalent and aggressive type of adult brain cancers with low 5-year overall survival rates. Epidemiologic data suggest estrogen may decrease brain tumor growth. The lack of powerful, selective brain permeable estrogen receptor beta (ERβ) ligands to promote its anticancer action is limiting the therapeutic promise of ERβ. The goal of this project is to create a new class of brain permeable ERβ agonists for the treatment of GBM. Method: We have used four different assays to identify potent leads. (1) ERβ and estrogen receptor alpha (ERα) reporter assays, (2) Polar Screen Estrogen Receptor (ER) Competitive Binding Assays, (3) Cell viability assays using GBM cells, and (4) Activity in WT and ERβ-KO GBM cells. CellTiter-Glo Cell Viability and colony formation assays were used to test the efficacy of ERβ agonists. Invasion was measured by matrigel invasion chamber assays and apoptosis was measured by Caspase-Glo® 3/7 and Annexin V assays. Flow cytometry was used to evaluate the cell cycle. Western blotting, RNA-Seq, RT-qPCR, and ERβ KO cells were used in the mechanistic studies. Both orthotopic patient-derived xenografts and GBM cell line-derived (CDX) xenografts were used to test the ERβ agonist's activity in vivo. Results: Indanone and tetralone-oximes or keto-oximes have been hypothesized, produced, and evaluated as new ER agonists. Due to its high action in ERβ reporter assays, specific binding to ERβ in polar screen assays (EC50 91 nM), and potent growth inhibitory activity (IC50 7.4) in GBM cells, CIDD-97 was discovered as a possible hit by screening a library of ~60 compounds. CIDD-97 is significantly more selective for ERβ than ERα (40-fold). Treatment with CIDD-97 markedly reduced U251 cell viability while having little to no effect on U251-ER-KO cells and normal human astrocytes. Further, CIDD-97 treatment decreased the expression of stemness markers in patient derived GSC lines and promoted apoptosis. Additionally, CIDD-ERβ agonist improved TMZ's ability to reduce GBM cell survival. The activation of ERβ target genes in cells treated with CIDD-97 was verified by using RNA-Seq and RTqPCR. PK studies confirmed that CIDD-0149897 has systemic exposure, was found in the brain, and its maximal detectable levels suggested that it has good BBB permeability. Mice tolerated daily intraperitoneal treatment of CIDD-0149897 (50 mg/kg) with a 7-day repeat dosage with no evidence of toxicity. Additionally, when compared to vehicle CIDD-97 (50 mg/kg/i.p.) treatment significantly decreased tumor growth in xenograft models and extended the survival of tumor-bearing mice in orthotopic GSC models. Conclusion: Collectively, these findings pointed to CIDD-97 as a possible ERβ agonist and is easily adaptable to clinical use alongside current chemo- and radiation-therapy regimens, offering GBM patients an additional means of improving survival.
