Um estudo fitoquímico de L. corniculatus foi realizado e biomonitorado através de bioensaios de atividade antioxidante, antibacteriana e anti-inflamatória. O estudo fitoquimico foi efetivado através de sucessivas cromatografias em coluna de sílica gel e monitoradas por cromatografia em camada delgada, sendo as estruturas elucidadas através de técnicas de IV, RMN de 1H e 13C e COSY. A atividade antioxidante in vitro foi avaliada nos modelos como a capacidade sequestrante DPPH, potencial redutor e peroxidação lipídica. O efeito antibacteriano foi avaliado através do método de microdiluição o qual produz a Concentração Inibitória Mínima e a Concentração Bactericida Mínima. Finalmente, o efeito anti-inflamatório de Lotus corniculatus foi analisado no modelo in vivo da pleurisia induzida pela carragenina em camundongos, que é um modelo de inflamação aguda. Foram isolados três compostos: dois terpenos, o ?-sitosterol e o ácido oleanólico e um flavonóide glicosilado canferitrina. Após realizados os ensaios de atividades biológicas, nossos resultados demonstraram que Lotus corniculatus cv. São Gabriel possui importante propriedade antibacteriana e anti-inflamatória. Estes efeitos podem ser atribuídos aos compostos isolados como o ácido oleanolico, ?-sitosterol e principalmente ao canferitrina o qual demonstrou o melhor efeito farmacológico.

Pełny tekst źródłaForage legume species such as Lotus., adapted to the local environmental conditions of South Brazilian States can be an important alternative to increase the supply of high protein content forage to the cattle besides the good adaptability to low-fertility soils. One of the advantages of using forage legumes relays on the acquisition of nitrogen from the symbiosis with diazothrophic bacteria such as Rhizobia. This work aimed the selection of native Rhizobia from soil samples of eight localities of Rio Grande do Sul. It was obtained 259 Rhizobia isolates from plants of Lotus glaber and 68 from Lotus subbiflorus. These isolates were evaluated on the efficiency of symbiotic nitrogen fixation with plants, grown in greenhouse conditions, on colonial morphology, capability of melanin production and genetically characterized by PCR genomic DNA fingerprinting using primers BOX and ERIC. Only five isolates produced melanin. It was selected nine isolates for L. glaber that were more efficient on nitrogen fixation than strains SEMIA 830 and U510, with relative efficiency index ranging from 88 to 132%, and two isolates for L. subbiflorus that were more efficient than strains SEMIA 849 and SEMIA 850, with relative efficiency index of 128 and 159%. Related to the genetic characterization, it was observed that isolates have no identity to the recommended strains, which shows that in the soils of Rio Grande do Sul there are autochthonous Rhizobia capable of efficient symbiotic nitrogen fixation with Lotus glaber and Lotus subbiflorus, which may be recommended to field evaluations and to the production of inoculants to these legume in Brazil.

Problemen leder till att företag vill byta ut sina gamla IS mot nya. I detta arbete behandlas migration, det vill säga då all funktionalitet från ett gammalt IS ska överföras till ett nytt (Bisbal m.fl, 1999).
Arbetet syftar till att skräddarsy en befintlig migrationsmetod så att en generell migrationsmetod blir unik och kan användas till just det användarfall som åsyftas i arbetet. Den migrationsmetod som skräddarsyddes var The Butterfly Methodology. Genom att följa de faser och aktiviteter som migrationsmetoden innehåller erhölls ett antal resultat som initierade en migrationsprocess.
De slutsatser som arbetet kom fram till var att genom att skräddarsy en migrationsmetod kunde en generell arbetsprocess göras unik så att den passade det användarfall som tas upp i arbetet. Migrationsmetoden innehöll även ett antal faser och aktiviteter som utgjorde stöd vid migrationsprocessen

Pełny tekst źródłaThe use of grasslands capable to tolerate high levels of aluminum is a viable alternative to bypass one of the main problems existing in cultivated areas with acid soils. Among the forage legumes, the birdsfoot trefoil (Lotus corniculatus L.) stands out for its nutritional quality and versatility of adaptation. Given the importance, this work was carried out to select plants of L. corniculatus tolerant to aluminum, and analyze the genetic diversity between these materials with the aid of microsatellite molecular markers. Three genotypes of birdsfoot trefoil were used (Draco, São Gabriel and UFRGS) in nutrient solution containing 200 µmol/L of calcium (CaCl2.2H2O), 100 µmol/L of aluminum (AlCl3) and controlled in a pH range of 4.1 - 4.3. Two selection cycles were made, where the seedlings were selected by the root length. Subsequently, the progress achieved by cycles of selection was tested in an experiment in nutrient solution, similar to that used in the selections, with the original and improved populations, in four concentrations of aluminum (0, 50, 100 and 150 µmol/L) (AlCl3). The initial, final and the growth of the radicle were evaluated. After the end of the second cycle of selection it was carried out the analysis of genetic diversity among genotypes of birdsfoot trefoil studied. Eighteen pairs of primers developed for Lotus japonicus (Regel) Larsen and Trifolium repens L. were used. The genetic similarities based on Jaccard coefficient was used to make the grouping of genotypes by the UPGMA method, using the program NTSYS pc 2.1. The number of alleles and the polymorphism information content (PIC) were calculated for each locus. The selection in nutrient solution was efficient in the selection of seedlings of birdsfoot trefoil tolerant to aluminum. The materials analyzed differ for this characteristic, with the genotype UFRGS F2, being the most tolerant. The microsatellite markers are suitable for accessing the genetic variability among genotypes of L. corniculatus.

Pełny tekst źródłaLegumes assimilate N2 through the establishment of Root Nodule Symbiosis (RNS) with bacteria from the genus Rhizobia. Small RNAs are 18-25nt molecules that regulate gene expression in a sequence-specific manner via a mechanism known as ‘RNA silencing’. Among the different types of smRNAs, microRNAs (miRNAs) are post-transcriptional gene regulators that, in plants, play key regulatory roles in plant-microbe interactions. In this context, we chose the model legume Lotus japonicus to assess the contribution of microRNAs to RNS. First, we identified the main RNA silencing effectors in Lotus japonicus, with emphasis on miRNA-related pathways, and generated L. Japonicus transgenic lines expressing the tombusviral silencing suppressor P19, which interferes with miRNA action. We then embarked on a smRNA cloning/sequencing strategy. The pooled sequenced data served as basis for the identification of miRNA genes in Lotus japonicus: 16 conserved and 27 potentially novel miRNA families. Comparative analysis of cloned smRNAs from root and nodules revealed that a discrete number of conserved miRNAs, miR167, miR172, miR390 and miR397, are highly expressed in mature nodules, as opposed to roots. Genetic analysis showed that the accumulation of miR167 and miR172 is Rhizobium-independent, as opposed to miR397. Further genetic studies showed that miR397 induction is also reproduced at the systemic level but dependent on the establishment of a successful N2 fixation, rather than on the infection by Rhizobium per se. Thus, our findings confirm that miRNA are important gene regulators at different stages of the establishment of Symbiotic Nitrogen Fixation

Resumo: O presente trabalho analisa a produção litográfica de Darel Valença Lins (1924 -) e Lotus Lobo (1948 - ) das décadas de 1970 e 1980. Estes artistas desempenharam papeis de destaque na produção e no fomento da litografia artística no Brasil a partir de contextos e referenciais distintos, no Rio de Janeiro e em Minas Gerais, respectivamente. Suas produções autorais apresentam novas direções ao passo que incorporam imagens apropriadas em suas gravuras e, ao mesmo tempo trabalham com novas soluções processuais para as impressões, de forma que suas obras enquadrem-se dentro do grupo de artistas que investigam a gravura como meio expressivo ambivalente, caracterizado pela reprodução e pela multiplicação da imagem. Suas litografias se apresentam de tal forma, que reajustam e recompõem as imagens apropriadas recodificando-as, gerando uma nova construção na qual o registro original não mais as representa, mas também não se apaga por completo, semelhantemente ao que fazem os processos cognitivos de memória
Abstract: This work analyses the artwork of Darel Valença Lins (1924 -) and Lotus Lobo (1948- ) through the 1970's and 1980's. These artists played top roles in the artistic application and teaching of the lithographic printing process in Brazil, specifically in Rio de Janeiro and Minas Gerais respectively. O trabalho propõe uma análise da produção artística desenvolvida pela artista e litógrafa mineira Lotus Lobo desde o final da década de 1960 até os dias de hoje, com matrizes de rótulos de embalagens descartadas pelas estamparias mineiras (especialmente de Juiz de Fora), em um momento de substituição de tecnologias de impressão. Destaco sua aproximação com a Pop Art — cuja produção, em geral, também dialoga com as artes gráficas ligadas ao contexto sócioindustrial —, mostrando as influências e as disparidades que impedem seu trabalho de ser meramente caracterizado como tal. Procuro refletir sobre duas direções de seu trabalho: a produção da artista plástica, por meio da apropriação das imagens dessas matrizes e de materiais de refugo industrial, como as maculaturas; e a pesquisadora que tenta manter viva a memória do design gráfico mineiro do início do século XX, seja por projetos acadêmicos ou iniciativas individuais, independentes, demonstrando a alta criatividade de sua produção artística e o lastro de suas contribuições.
This paper proposes an analysis of the artistic production developed by the artist and lithographer Lotus Lobo from the late 1960s to the present day, with matrices of packaging labels discarded by the printmakers of Minas Gerais (especially from Juiz de Fora), at a time when printing technologies were being replaced. I highlighted her proximity to Pop Art — whose works also dialogue with the graphic arts linked to the industrial context — showing the influences and the disparities that prevent her work from being characterized as Pop. I try to reflect on two directions of her work: the production of the plastic artist, through the appropriation of the images of these matrices and materials refused by the industry, as the stained printings (maculaturas); and the researcher who tries to keep alive the memory of the graphic design of Minas Gerais of the early twentieth century, either by academic projects or through independent initiatives, demonstrating the high creativity of her artistic production and the wide range of her contributions.

Pełny tekst źródłaO género Lotus engloba espécies com interesse agrícola, como é o caso de L. uliginosus, que possui capacidade de se adaptar a condições adversas e é utilizada em pastagens melhoradas. Este género compreende também espécies silvestres, muito pouco estudadas ao nível dos respetivos microssimbiontes, como é o caso de Lotus parviflorus. Neste estudo, fez-se a caraterização fenotípica e molecular de estirpes de bactérias isoladas dos nódulos radiculares de Lotus parviflorus e de Lotus uliginosus. Os isolados de L. parviflorus mostraram-se bastante heterógenos em relação à velocidade de crescimento, absorção de vermelho de congo e diversidade. Apenas um (isolado Lp11), de crescimento lento, revelou a capacidade para nodular espécies de Lotus. Taxonomicamente foi incluído na espécie Bradyrhizobium canariense, e obteve maior homologia nos genes simbióticos, com isolados de leguminosas da tribo Genisteae. Os restantes isolados não revelaram capacidade para nodular espécies de Lotus, e, taxonomicamente, muitos destes isolados não pertencem à família Rhizobiaceae. Para além da fixação de azoto, alguns isolados de L. parviflorus, revelaram a capacidade de produzir sideróforos (Lp6), de hidrolisar polissacáridos da parede celular das plantas e de inibir o crescimento de fitopatogénios como Phytophthora cinnamomi e Botryosphaeria corticola (Lp7a e Lp7b). Nos isolados de L. uliginosus estudados, apenas um (Lu13) foi incapaz de nodular plantas de Lotus spp., tendo sido classificado fora da família das Rhizobiaceae. Foi também o único de crescimento rápido e com absorção intensa do corante vermelho do congo. Os restantes isolados nodularam de forma eficaz L. uliginosus e L. parviflorus. Nodularam também, mas ineficazmente, L. tenuis e L. corniculatus. Os isolados com capacidade simbiótica foram identificados como pertencentes ao género Bradyrhizobium, com maior similaridade com as estirpes de Bradyrhizobium sp. isoladas de L. uliginosus de estudos anteriores e com estirpes de Bradyrhizobium japonicum bv. genistearum. Possuíram maior homologia nos genes simbióticos, com estirpes isoladas de L. uliginosus, em estudos anteriores, e também, mas com menor similaridade, com estirpes isoladas dos nódulos de plantas da tribo Genisteae.
Lotus spp. comprises species with a high agronomic value, like L. uliginosus, that can tolerate adverse conditions. It also includes wild species that have been little explored in terms of their microsymbionts. In this study a phenotypic and molecular characterization of bacteria isolated from the root nodules of Lotus parviflorus and from Lotus uliginosus was performed. L. parviflorus isolates were very heterogenous regarding the growing type, congo red absorption and genetic diversity. Only the isolate Lp11 was able to induce nodules on Lotus plants. This isolate is a slow grower and was classified as belonging to the Bradyrhizobium canariense specie. On the basis of the symbiotic genes it was close related with strains isolated from plants of the Genisteae tribe. The remaining isolates didn’t nodulate Lotus plants, and a lot of them don’t belong to the Rhizobiaceae family. Besides nitrogen fixation, some strains isolated from L. parviflorus, could produce siderophores (Lp6), hydrolyze cellulose and inhibit the growth of fitopathogens like Phytophthora cinnamomi and Botryosphaeria corticola (Lp7a and Lp7b). In the isolates of L. uliginosus studied, only one isolate (Lu13) couldn’t nodulate Lotus plants and was classified outside of the Rhizobiaceae family. It was the only one that strongly absorbed congo red and revealed a fast growing type. The remaining isolates of the root nodules of L. uliginosus could effectively nodulate L. uliginosus and L. parviflorus plants. These isolates also nodulated L. corniculatus and L. tenuis, however, they were ineffective. These isolates belong to the Bradyrhizobium genus, with high similarities with Bradyrhizobium sp. strains isolated from L. uliginosus, in previous studies, and also with some strains of Bradyrhizobium japonicum bv. genistearum. The symbiotic genes were identical to the isolates of L. uliginosus from previous studies, and to a lower degree to some isolates from plants of the Genisteae tribe.

Pełny tekst źródłaIn lowland areas, the rhizobia can form nodules in legumes, when adapted to conditions of water stress, fix nitrogen and colonize the rice plants in rotation. Recent studies have shown that rhizobia can colonize roots, stems and leaves of grasses, such as rice, promoting plant growth. The production of phytohormones, mainly indole acetic acid (IAA) is possibly the main mechanism of growth promotion of rice by rhizobia. The objectives were to study the resistance of the symbiosis between rhizobia and Lotus to flooding and drought; check the response of different rice cultivars to inoculation with rhizobia and to evaluate the interaction between nitrogen and the promotion of growth by indigenous rice; to study the colonization pattern of rice plants by rhizobia and legumes and to study genetically the biosynthesis of auxin by rhizobia. Several experiments were conducted in the greenhouse and laboratory, with plants of the legume species L. corniculatus (var. São Gabriel) and L.uliginosus (var. Maku) and rice plants inoculated with different rhizobia. L.uliginosus plants tolerate flooding, showing potential for their use in floodplain areas, in rotation with rice. Among the rice cultivars tested, IRGA424 proved to be more responsive to inoculation with rhizobia. Inoculation with rhizobia tested stimulated the growth of plants, and this increase was proportional to the doses of nitrogen applied. Furthermore, inoculation with rhizobia allowed more efficient use of nitrogen absorbed from the soil. The marking of rhizobia with the Gus gene, confirmed the colonization of rice. The rhizobia tested are able to produce IAA, and there is evidence that the biosynthesis occurs by the route of indole-3- acetonitrile (IAN).
