During latent Epstein-Barr virus (EBV) infection, the T helper cell response to the EBV latent membrane protein (LMP)1 is dominated by the production of IL-10, but by IFN- γ during acute EBV infection. The purpose of this thesis was to develop methods for the enumeration and characterisation of the IL-10 producing CD4+ T cells that respond to peptides of the EBV protein LMP1, and to investigate the possible involvement of IL-27 in the development of these cells. It was found that some human donors have very high concentrations of IL-27 within serum, which is not dependent on EBV infection status but demonstrates relatively low heritability. The addition of IL-27 to cultures of human T cells did not induce IL-10 but the production of IL-17 was inhibited. To identify and characterise LMP1 responsive cells I used CD154 as a marker of activated T cells. Having optimised the methodology, 14 donors of known EBV serostatus were tested for activated IL-10 producing cells after culturing peripheral blood mononuclear cells with LMP1 peptides. However in most cases the frequency of CD4+ lymphocytes upregulating CD154 and IL-10 in response to LMP1 peptides was below the assay’s sensitivity. When the CD154+IL-10+ CD4+ cells were stained for T helper cell subset markers they were positive for every marker and isotype control, suggesting that the cells were non-specifically binding labelled antibody. Both single positive CD154+ and single positive IL-10+CD4+ cells were also present in response to LMP1, but again typically at frequencies below the level of sensitivity for this assay. The conclusions of the work are that IL-27 responses are heterogeneous, but unlikely to play an important role in the induction of IL-10+ T cells in EBV infection. The frequency of LMP1 responsive T cells is very low (<0.08%) in most donors.

Streszczenie:
The study was conducted in Cihan University-Erbil campus in November 2021. All subjects were asked questions regarding their history, using a questionnaire sheet. Blood was investigated for the Interleukin-10 level, Lactate Dehydrogenase (LDH) and hematological parameters. The mean serum level of IL-10 was significantly higher in Covid-19, non-vaccinated than Covid-19, vaccinated individuals. Moreover, the level was also equally significant and higher in non-Covid-19, vaccinated than non-Covid-19, non-vaccinated. Non-significant differences were found between Covid-19, vaccinated and non-Covid-19, non-vaccinated individuals. And between Covid-19, non-vaccinated and non-Covid-19, vaccinated individuals. The mean serum levels of LDH were found to be significantly lower in non-Covid-19, non-vaccinated, indicating a significant rise of LDH in all other groups. Despite the rise in all three groups a significant difference was not found between them. The total WBC count shows a significant increase in both non-vaccinated groups. While the mean level of lymphocytes counts reveals non-significant decrease in non-Covid-19, vaccinated group in comparison to the other groups. However, the mean monocyte counts of Covid-19, vaccinated group revealed a significant rise in comparison to other groups. In addition, the mean counts of granulocyte showed a significant increase in both non-vaccinated groups in comparison with vaccinated groups. Platelet mean count presented a non-significant difference when comparing all groups. Our current study may indicate the potency of the vaccines.

Review question / Objective: How were serum IL-6 and IL-10 linked with the severity and mortality of COVID-19 patients? To evaluate the role of IL-6 and IL-10 in the development of the severity or morality of COVID-19 patients. The outcomes (mean difference) were calculated between the severe vs. non-severe COVID-19 patients and non-survival vs. survival patients to evaluate the risk of severity or mortality. Condition being studied: Severity and mortality among the COVID-19 patients. Information sources: The international scientific authorized databases such as Google Scholar, PubMed, Embase, CNKI, Cochrane Library, and Web of science were used as primary sources to identify and collect the eligible literature. Additional secondary databases were also comprehensively searched to extract more related studies.

Review question / Objective: The aim of our systematic review and meta-analysis was to summarize the effects of IL-4 and IL-10 gene polymorphism and clarify their possible association with PTB. Condition being studied: World Health Organization (WHO) defines preterm birth (PTB) as babies born alive before 37 weeks of pregnancy are completed. The new estimates show that the prevalence of PTB during 2014 ranged from 8.7% to13.4% of all live births, about 15 million preterm babies born each year. Besides, PTB is the leading cause of death worldwide for children below 5 years of age. Babies born preterm are at an increased risk of short-term and long-term complications attributed to immaturity of multiple organ systems, such as cerebral palsy, intellectual disabilities, vision and hearing impairments, and impaired cognitive development. PTB has become a worldwide public health problem, but its etiology remains unclear. Accumulating evidence shows that PTB is a syndrome that can be attributed to a variety of pathological processes(5). Inflammatory diseases and genetic background are known risk factors for PTB, many studies had shown that genetic variations in proinflammatory cytokines such as tumor necrosis factor-α (TNF-α) and interleukin-1 α (IL-1 α) are associated with increased risk of PTB, but the relationship between genetic polymorphism in anti-inflammatory cytokines and risk of PTB remains controversial.
