Background: Hidradenitis suppurativa (HS) is a complex disease that is optimally managed with specialized care. Data on HS specialty clinics (HSSCs) are lacking. Methods: HSSCs in the USA were identified on the HS Foundation website and analyzed for geographic location and clinic director demographics. Results: We identified 29 HSSCs in 16 states, an increase from 22 in 2019. Thirty-four states currently lack a HSSC; the Mountain West and East South Central regions of the USA are particularly affected. Among HSSC directors, the majority (93.3%) are dermatologists, with slightly more women (53.3%) than men (46.7%). Most (86.7%) have an academic affiliation, and the majority (60%) graduated from residency <10 years ago. All directors are involved in research, and over half of HSSCs serve as HS clinical trial sites. Conclusions: The number of HSSCs in the USA has been growing, yet there remains a dearth in certain regions. Given that HS is a disease with high unmet need and a rapidly evolving therapeutic landscape, we encourage the establishment of more HSSCs to improve access to specialized care.

Abstract: This paper presents an experimental program to investigate the strength and durability properties of high-strength selfcompacting concrete (HSSCC) with and without steel fibers. The research work is divided into two approaches. These approaches are the development of (HSSCC) by making different trials and then studying mechanical and durability properties. Crimped-type steel fibers are used. Fly ash and Alccofine have been used as mineral admixtures to fulfil the power requirement. Chemical admixture has also been used for workability. A review of recent research on mechanical performance of high strength structural steel (HSSS) in the level of both materials and members is presented, including the static material performance, toughness and mechanical property under cyclic loadings of HSSS, buckling and hysteretic behavior of high strength steel columns (HSSC), the ductility and resistance of bolted connection with high strength steel slices etc. Previous research work shows that the application of high strength steel (HSS) is limited in seismic zone .Finally, two design methods of using HSS in seismic zone are proposed based on current seismic design philosophy.

Human skeletal stem cells (hSSCs) were recently identified as podoplanin (PDPN)/CD73/CD164-positive and CD146-negative cells that decline with age, and play a role in the pathogenesis of osteoarthritis (OA). The aim of this study was to identify the hSSC-like properties of bone-derived mesenchymal stem/stromal cells (MSCs) of patients with late and early OA. Methods: First, we performed gene expression profiling for the hSSC markers in 32 patients with late and early OA, and donors without OA. Having identified the low expression of hSSC markers in late OA patients, we further performed trilineage differentiation and immunophenotyping for hSSC makers in the selected subsets from each donor group. Results: Our results show no differences in osteogenesis, chondrogenesis, and adipogenesis between the MSCs from the three groups. However, the immunophenotyping shows lower CD164 in MSCs from early OA patients in comparison with late and no OA subjects (p = 0.002 and p = 0.017). Conclusions: Our study shows that the in vitro hSSC-like properties of bone-derived MSCs are similar in patients with early and late OA, and in donors without OA. However, the lower percentage of CD164-positive MSCs in early OA patients indicates the potential of CD164 as a marker of the onset of OA.

The expansion of functional testicular biopsy-derived human spermatogonial stem cells (hSSC)ex vivomay enable the restoration of fertility in pre-pubertal males having undergone gonadotoxic therapies or men with severe male factor infertility. Various somatic cells are known to regulate SSC homeostasis and spermatogenesis in the developing and adult testis. Prior attempts to recapitulate this niche demonstrated the requirement of feeder cells, such as endogenous testicular somatic cells, for germ cell expansionex vivo. However, this strategy has limitations in the expansion of hSSCs from tissue biopsies for which spermatogenesis is absent or defective. Our aim was to evaluate the first trimester human umbilical cord perivascular cells (FTM HUCPVCs), a novel source of mesenchymal stromal-like cells (MSCs), as potential human feeder cells for standardized hSSC expansionex vivo.Targeted RNA sequencing analysis demonstrated that CD90+ve FTM HUCPVCs expandedin vitrounder germ cell culture conditions express a profile of targeted testicular-associated transcripts that is similar to cultured human CD90+ve testicular adherent cells (hTACs) and secrete LIF, FGF2 and BMP4, key growth factors known to regulate spermatogenesis. We also demonstrated that mitotically inactivated FTM HUCPVCs support the expansion of mouse germ cells and putative SSCsex vivoand that FTM HUCPVC transplantation promotesin vivogerm cell regeneration after mono-2-ethylhexyl phthalate (MEHP)-induced seminiferous tubule damage in a murine model, including a partial reconstitution of tubular cellular architecture and reestablishment of DAZL and acrosin+ve germ cell layers. Together, these data suggest that FTM HUCPVCs have phenotypical and functional properties that may support repair of the human testicular niche.

Abstract The rotational spectrum of gas phase HSSSH has been recorded for the first time in absorption with the Cologne free-space-cell millimeterwave spectrometer. In the frequency region between 80 and 300 GHz a prominent series of Q-branches has been observed and assigned. The hitherto identified rotational lines unambigously arise from a perpendicular-type spectrum. In addition, successive J lines of the compact Q-branches show no indication of the easily detectable intensity alternation which arises when the molecule possesses an axis of symmetry due to nuclear spin statistical weights. Therefore the geometrical structure of the molecule does not have a C2 axis of symmetry. The present data are not complete enough for a unique structure determination. However on the basis of the present data we can definitely rule out conformations with C2 or C2v symmetry to be responsible for the strongest observed transitions of the spectrum. The molecule is not floppy as predicted from semi empirical MO calculations. On the contrary the conformation observed is rather rigidly locked into one position with no sign of possible internal rotation. We consider a sulfur triangle with the two hydrogen atoms sticking out symmetrically to the same side of the SSS-plane to be the most likely structure.

Background:transient ischemic attack (tIA) and minor stroke have a high risk of early neurological deterioration, and patients who experience early improvement are at risk of deterioration. We generated a score for quantifying the worst reported motor and speech deficits and assessed whether this predicted outcome.Methods:510 tIA or minor stroke (NIHSS>4) patients were included. the Historical Stroke Severity Score (HSSS) prospectively quantified the patient's description of the worst motor or speech deficits. the HSSS was rated at the time of first assessment with more severe deficits scoring higher. Motor HSSS included assessments of arm and leg motor power (score total 0-5). Speech HSSS assessed severity of dysarthria and aphasia (total 0-3). the association between motor and speech HSSS and symptom progression was assessed during the 90-day follow-up period.Results:the proportion of patients in each category of the motor HSSS was 0: 43% (216/510), 1: 22%(110/510), 2: 17% (89/510), 3: 7% (37/510), 4: 5% (28/510) and 5: 6% (30/510). Motor HSSS was associated with symptom progression (p=0.004) but not recurrent stroke. Speech HSSS was not associated with either progression or recurrent stroke. Motor HSSS predicted disability (p=0.002) and intracranial occlusion (p=0.012). Disability increased with increasing motor HSSS.Conclusions:taking a detailed history about the severity of motor deficits, but not speech, predicted outcome in tIA and minor stroke patients. A score based on the patient's description of the severity of motor symptoms predicted symptom progression, intracranial occlusion and functional outcome, but not recurrent stroke in a tIA and minor stroke population.

Control and analysis of protein aggregation is an increasing challenge to biopharmaceutical research and development. Therefore it is important to understand the interactions, causes and analysis of particles in order to control protein aggregation to enable successful biopharmaceutical formulations. This work investigates the role of different non-ionic surfactants on protein conformational stability, as assessed by HSDSC, and on protein size stability as assessed by Dynamic Light Scattering (DLS), HIAC and MFI. BSA and IgG2 were used as model proteins. Thermal unfolding experiments indicated a very weak surfactant-immunoglobulin IgG2 interaction, compared to much stronger interactions for the BSA surfactant systems. The DLS results showed that BSA and IgG2 with different surfactants and concentration produced different levels of particle size growth. The heat treatment and aging of samples in the presence of Tween 20, Tween 80, Brij 35 and Pluronic F-68 surfactants led to an increase in the populations of larger particles for BSA samples, whereas IgG2 systems did not notably aggregate under storage conditions MFI was shown to be more sensitive than HIAC technique for measuring sub-visible particles in protein surfactant systems. Heat treatment and storage stress showed a significant effect on BSA and IgG2 protein sub-visible particle size stability. This work has demonstrated that both proteins with different Tween 20, Tween 80, Brij 35 and Pluronic F-68 concentrations, have different level of conformational and size stability. Also aging samples and heating stress bears the potential to generate particles, but this depends on surfactant type. Poor predictive correlations between the analytical methods were determined.

We performed an importance analysis of In-Service Testing (IST) components for Ulchin Unit 3 using the integrated evaluation method for categorizing component safety significance developed in this study. The importance analysis using the developed method is initiated by ranking the component importance using quantitative PSA information. The importance analysis of the IST components not modeled in the PSA is performed through the engineering judgment, based on the expertise of PSA, and the quantitative and qualitative information for the IST components. The PSA scope for importance analysis includes not only Level 1 & 2 internal PSA but also Level 1 external and shutdown/low power operation PSA. The importance analysis results of valves show that 167 (26.55%) of the 629 IST valves are HSSCs and 462 (73.45%) are LSSCs. Those of pumps also show that 28 (70%) of the 40 IST pumps are HSSCs and 12 (30%) are LSSCs.

Existing benchmarks often mix different types of datasets, which blurs and complicates segmentation evaluations. Here, we introduce a Multi-LiDARs Domain Adaptation Segmentation (MLDAS) dataset, which contains point-wise semantic annotated point clouds captured simultaneously by a 128-beam LiDAR, a 64-beam LiDAR, a 32-beam LiDAR. We select 31,875 scans from 2 representative scenarios: campus and urban street. Furthermore, we evaluate the current 3D segmentation unsupervised domain adaptation methods on the proposed dataset and propose Hierarchical Segmentation Network with Spatial Consistency (HSSC) as a novel knowledge transfer method to mitigate the domain gap significantly using spatial-temporal consistency constraints. Extensive experiments show that HSSC greatly improves the state-of-the-art cross-domain semantic segmentation methods. Our project is available at https://sychen320.github.io/projects/MLDAS.

Abstract The length of horizontal wells in deep carbonate reservoir (5000∼6000m) in Sichuan Basin of China has exceeded 2000 meters. Both generating acid-etched fractures and acidizing the dispersed pay-zone are the key to achieving optimal production. The complex conditions of multilayers make it difficult to use open hole packers, instead, sliding sleeves are used in liner completions. However, the layers show strong heterogeneity, which makes it difficult to realize efficient acid stimulation. This paper presents a technology that stimulates such wells to achieve full acidizing of drilled reservoirs, and optimize the fracturing fluid system and acid-fracturing parameters to achieve longitudinal fracture penetration based on the sweet spot recognition. Firstly, in order to describe reservoir characteristics and identify sweet spots along the well, an evaluation model was established based on the correlation evaluation parameters of the gas-bearing properties, petrophysical properties, natural fractures, and fracability. The high sweet spot coefficient (HSSC) section and low sweet spot coefficient (LSSC) section were recognized. Then, based on the section classification and the optimal fracture spacing, the position of sliding sleeves is optimized. Sleeves were mainly set at the highest points of sweet spot coefficients in the nearby section. For different sections, different acid stimulation strategies are proposed respectively. For the HSSC, it is necessary to increase the coverage of acid in the wellbore to realize the efficient acid stimulation of sweet spots alone the wellbore. However, the LSSC sections are mainly caused by the deflection of well, creating artificial fractures and making fractures propagate along the height direction into TL43-3 are needed. In HSSC sections, the acid coverage increases with the increase of acid volume and injection rate, and it is preferred to use temporary plugging to obtain the wide acid coverage when half of the acid is pumped making the two fractures obtain efficient stimulation with the acid absorption ratio of 1.47 and acid coverage of 207.1m In LSSC sections, it is still difficult to make artificial fracture break through the upper and lower layers by only use gelled acid even if the acid injection rate is greatly increased. A high viscosity fluid was introduced in acid stimulation treatments could make artificial fracture vertical propagating to break through the upper layers of 16 MPa stress difference with injection rate ≥ 5 m3/min and volume of 100m3. The productivity of 5 applicated wells increased 66.28%, which proved that the stimulation strategies of HSSC sections and LSSC sections are successful.

Abstract Honeycomb sandwich structures (HSSs) are well-known for their very high specific strength, and are widely used for aeronautical and aerospace industrial applications. Due to their special usage and high cost, non-destructive evaluation (NDE) of HSS plays an important role in the industry. NDE using guided waves, as one of the potential method of NDE, is getting more attention for its economical efficiency. However, due to the complexity of the HSS, very little theoretical work has been done on determining the features of waves propagating in HSS structure especially in a wide frequency range. This research focuses on using a homogenized layer model for the HSS, studying how the material properties affect the features of the guided waves. By using the global matrix method, features including the phase velocities and group velocities of different modes are computed. The effects of different material properties are evaluated and compared. The results show that the material properties of the face sheets are most influential for gudied wave features. These findings open up the possibility of bypassing the complexity of the core to analyze the guided waves propagating in a honeycomb sandwich structure.

The application of Q690D high-strength structural steel (HSSS) has been increasing in engineering structures. The lack of knowledge of the strain rate behaviour limits the application to the extreme loading conditions such as blast and impact loadings. This paper presents a series of tensile tests on the dynamic tensile behaviour of Q690D HSSS produced through the thermo-mechanical control process (TMCP). The stress-strain relationships of TMCP Q690D in the strain rate range of 0.00025 to 760 s-1 were measured by using the universal and servo-hydraulic high speed testing machines. The experimental results verified the sensitivity to strain rate of TMCP Q690D and the dynamic increase factor (DIF) for yield stress is identical to that of QT (Quenched and Tempered) S690 HSSS. However, TMCP Q690D behaves in a much different way in the strain hardening stage. The commonly-used Cowper-Symonds model was calibrated for the DIFs of yield stress and ultimate tensile strength. The Johnson-Cook (J-C) model was modified and a new rate-dependent constitutive model was proposed. The proposed model was validated successfully to predict the true stress-strain relationship, providing better prediction results than the modified J-C model.
