To assess functional relatedness of individual components of the eukaryotic transcription apparatus, three human subunits (hsRPB5, hsRPB8, and hsRPB10) were tested for their ability to support yeast cell growth in the absence of their essential yeast homologs. Two of the three subunits, hsRPB8 and hsRPB10, supported normal yeast cell growth at moderate temperatures. A fourth human subunit, hsRPB9, is a homolog of the nonessential yeast subunit RPB9. Yeast cells lacking RPB9 are unable to grow at high and low temperatures and are defective in mRNA start site selection. We tested the ability of hsRPB9 to correct the growth and start site selection defect seen in the absence of RPB9. Expression of hsRPB9 on a high-copy-number plasmid, but not a low-copy-number plasmid, restored growth at high temperatures. Recombinant human hsRPB9 was also able to completely correct the start site selection defect seen at the CYC1 promoter in vitro as effectively as the yeast RPB9 subunit. Immunoprecipitation of the cell extracts from yeast cells containing either of the human subunits that function in place of their yeast counterparts in vivo suggested that they assemble with the complete set of yeast RNA polymerase II subunits. Overall, a total of six of the seven human subunits tested previously or in this study are able to substitute for their yeast counterparts in vivo, underscoring the remarkable similarities between the transcriptional machineries of lower and higher eukaryotes.

ABSTRACT Under conditions of environmental stress, prokaryotes and lower eukaryotes such as the yeast Saccharomyces cerevisiaeselectively utilize particular subunits of RNA polymerase II (pol II) to alter transcription to patterns favoring survival. In S. cerevisiae, a complex of two such subunits, RPB4 and RPB7, preferentially associates with pol II during stationary phase; of these two subunits, RPB4 is specifically required for survival under nonoptimal growth conditions. Previously, we have shown that RPB7 possesses an evolutionarily conserved human homolog, hsRPB7, which was capable of partially interacting with RPB4 and the yeast transcriptional apparatus. Using this as a probe in a two-hybrid screen, we have now established that hsRPB4 is also conserved in higher eukaryotes. In contrast to hsRPB7, hsRPB4 has diverged so that it no longer interacts with yeast RPB7, although it partially complements rpb4− phenotypes in yeast. However, hsRPB4 associates strongly and specifically with hsRPB7 when expressed in yeast or in mammalian cells and copurifies with intact pol II. hsRPB4 expression in humans parallels that of hsRPB7, supporting the idea that the two proteins may possess associated functions. Structure-function studies of hsRPB4-hsRPB7 are used to establish the interaction interface between the two proteins. This identification completes the set of human homologs for RNA pol II subunits defined in yeast and should provide the basis for subsequent structural and functional characterization of the pol II holoenzyme.

High school research experience programs (HSREPs) provide opportunities for true science education and expose students to scientific investigations in laboratory settings. Various HSREPs models have been practiced to shape students’ research understandings; however, a systematic comparison of the success, challenges, and opportunities of these HSREPs has not been gauged. This article compares the effectiveness of such science, technology, engineering, and mathematics (STEM) based HSREP models reported in the last two decades. We shortlisted seventeen studies on the most effective HSREPs and identified the characteristics of these reports. Results show that student research experiences vary depending on the structure of the model used and the nature of the laboratory setting to which students are exposed. However, there is a dire need to integrate more collaborative and customized research practices to accommodate more students in HSREPs. Additionally, intensive support, mentoring, and coaching are essential to provide students a comprehensive understanding to excel in their research career pathway. Finally, there is a desperate need for further studies to develop the frameworks that can help the smooth transition of high school students into research-oriented university programs.

Modeling user’s fine-grained preferences and dynamic preference evolution from their chronological behaviors are challenging and crucial for sequential recommendation. In this paper, we develop a Hierarchical Self-Attention Incorporating Knowledge Graph for Sequential Recommendation (HSRec). HSRec models not only the user’s intrinsic preferences but also the user’s external potential interests to capture the user’s fine-grained preferences. Specifically, the intrinsic interest module and potential interest module are designed to capture these two preferences respectively. In the intrinsic interest module, user’s sequential patterns are characterized from their behaviors via the self-attention mechanism. As for the potential interest module, high-order paths can be generated with the help of the knowledge graph. Therefore, a hierarchical self-attention mechanism is designed to aggregate the semantic information of user interaction from these paths. Specifically, an entity-level self-attention mechanism is applied to capture the sequential patterns contained in the high-order paths while an interaction-level self-attention mechanism is designed to further capture the semantic information from user interactions. Moreover, according to the high-order semantic relevance, HSRec can explore the user’s dynamic preferences at each time, thus describing the user’s dynamic preference evolution. Finally, experiments conducted on three real world datasets demonstrate the state-of-the-art performance of the HSRec.

Ewing’s Sarcoma Oncogene (ews) on chromosome 22q12 is encoding a ubiquitously expressed RNA-binding protein (EWS) with unknown function that is target of tumor-specific chromosomal translocations in Ewing’s sarcoma family of tumors. A model of transcription complex was proposed in which the heterodimer Rpb4/7 binds to EAD, connecting it to Core RNA Pol II. The DNA-binding domain, provided by EFP, is bound to the promoter. Rpb4/7 binds RNA, stabilizing the transcription complex. The complex Rpb4/7 can stabilize the preinitiation complexes by converting the conformation of RNA Pol II. EWS may change its conformation, so that NTD becomes accessible. Two different mechanisms of interaction between EWS and RNA Pol II are proposed: (I) an intermolecular EWS-EWS interaction between two molecules, pushing conformation from “closed” to “open” state, or (II) an intramolecular interaction inside the molecule of EWS, pushing conformation of the molecule from “closed” to “open” state. The modified forms of EWS may interact with Pol II subunits hsRpb5 and hsRpb7. The EWS and EFPs binding partners are described schematically in a model, an attempt to link the transcription with the splicing. The proposed model helps to understand the functional molecular interactions in cancer, to find new partners and ways to treat cancer.

Using a screen to identify human genes that promote pseudohyphal conversion in Saccharomyces cerevisiae, we obtained a cDNA encoding hsRPB7, a human homologue of the seventh largest subunit of yeast RNA polymerase II (RPB7). Overexpression of yeast RPB7 in a comparable strain background caused more pronounced cell elongation than overexpression of hsRPB7. hsRPB7 sequence and function are strongly conserved with its yeast counterpart because its expression can rescue deletion of the essential RPB7 gene at moderate temperatures. Further, immuno-precipitation of RNA polymerase II from yeast cells containing hsRPB7 revealed that the hsRPB7 assembles the complete set of 11 other yeast subunits. However, at temperature extremes and during maintenance at stationary phase, hsRPB7-containing yeast cells lose viability rapidly, stress-sensitive phenotypes reminiscent of those associated with deletion of the RPB4 subunit with which RPB7 normally complexes. Two-hybrid analysis revealed that although hsRPB7 and RPB4 interact, the association is of lower affinity than the RPB4-RPB7 interaction, providing a probable mechanism for the failure of hsRPB7 to fully function in yeast cells at high and low temperatures. Finally, surprisingly, hsRPB7 RNA in human cells is expressed in a tissue-specific pattern that differs from that of the RNA polymerase II largest subunit, implying a potential regulatory role for hsRPB7. Taken together, these results suggest that some RPB7 functions may be analogous to those possessed by the stress-specific prokaryotic sigma factor rpoS.

e14609 Background: Resistance acquisition to chemotherapeutic agents is one of the main problems that come up during cancer treatment. hSRBC is a tumor suppressor gene whose inactivation has been associated with malignant tumor progression. In a previous work, we investigated the influence of hSRBC promoter methylation alterations in oxaliplatin (OXA) resistance acquisition by using a CRC “in vitro” model, detecting an hSRBC promoter hypermethylation in OXA–resistant derived cells when compared with the sensitive counterpart. These results were validated by functional analyses in the same model. Taking this into account, our aim in the present work is to determine the role of hSRBC methylation status as a potential biomarker of OXA resistance, in metastatic CRC patients, treated at first line with fluouropirimidines plus OXA based chemotherapy. Methods: hSRBC promoter hypermethylation was analyzed in DNA extracted from paraffin embebbed tissue of 111 metastatic CRC tumors by using Methylation Specific PCR. Methylation data was correlated to overall response (OR) and progression free survival (PFS) by using F-Fisher test and Kaplan-Meyer Survival curves respectively. A multivariate analysis was carried out by Cox regression. p-values under 0.05 were considered statistic significant. Results: Two independent cohorts of stage IV CRC tumors were included. Twenty-two out of 111 patients received radical surgery for metastasis, that became to be a positive prognostic factor (p-value = 0.04). Gene hypermethylation was detected in a 33% of cases. Although OR was not associated with hSRBC methylation, we observed a significant correlation between hypermethylation of gene and a worse PFS in patients without metastasis surgery (Log Rank; p-value = 0.04). Conclusions: Remarkably, hSRBC promoter hypermethylation is associated with worse PFS in metastatic CRC patients. We suggest hSRBC methylation status as a predictive biomarker of OXA-based treatment outcome in metastatic CRC patients. However, further studies are warranted in order to elucidate the clinical application of these findings.

Abstract Welcome to the third virtual HSRPP conference in this 28th anniversary of the conference. We hope to provide you with an enjoyable and interesting conference. Little did we know when we were invited to host this conference that we would be unable to welcome you in-person to the lovely city of Bath. After much deliberation in Summer 2021, with too many unknowns due to the COVID-19 pandemic, we had to make the call to run the conference online. By October 2021 we doubted ourselves. As we write this in January 2022, with the Omicron wave upon us, it feels like it was probably the right decision. We do all of course hope that HSRPP 2023 can bring us all together again in-person, but until then, we hope you enjoy our online event. HSRPP 2022 continues to provide an environment for researchers and practitioners, both new and experienced, to come together to share research and exchange ideas. Our conference theme is ‘Delivering impact for patients and other stakeholders’. We chose this theme because we believe it is central to why most of us do health services and pharmacy practice research. The days are long gone when research could remain within the pages of academic journals. We need to be able to demonstrate to our patients, our funders and the public how we will turn our findings into something that improves care. We need to work with patients and the public to make sure our research is relevant to them and efficient in its spend of what is, very often, public money. In short, it is what gets us up in the morning and through the tough times of research, of which there are many! The COVID-19 pandemic has shown just how valuable pharmacists are, whether that be delivering vaccinations, caring for critically ill hospitalised patients or coping with the huge pressure that community colleagues have faced from the worried public. These contributions to the pandemic cause are of course on top of the ‘day job’. There is much learned and to be learned about how we move forward to deliver healthcare that is sustainable, patient-centred and fit for purpose. Research is the tool from which this learning comes. We are going to hear real world evidence under our conference theme from our three keynote speakers: Dr Barbara Farrell, a pharmacist with the Bruyère Continuing Care Geriatric Day Hospital, and Senior Investigator with the Bruyère Research Institute in Ottawa, Ontario, Canada, will discuss her research experience with engaging stakeholders and users to promote the uptake and impact of deprescribing guidelines. She will highlight how her research has influenced and contributed to change in how people approach deprescribing and what difference this has made. Professor Mahendra Patel, University of Bradford, is the national Pharmacy and Ethnic Minority Communities Research Lead with the PRINCIPLE trial, University of Oxford, the world’s largest randomized clinical trial investigating COVID-19 treatment in the community. He will talk to us about promoting inclusion and diversity in clinical trials, which is critical if trial results are to be meaningful and successful when implemented to bring about change. Mr Ade Williams, lead pharmacist and independent prescriber at the multi-award-winning Bedminster Pharmacy in Bristol will discuss how researchers can work with practising pharmacists to co-develop “real-life’’ impact programmes. He will also highlight how transformations in the NHS governance structure, Long Term Plan ambitions and COVID-19 legacy work are unique opportunities for community pharmacy to play a vital role. Mahendra and Ade were both recognised in the New Year Honours List 2022 for their outstanding contributions to Pharmacy. We would like to thank all those who have contributed to making HSRPP 2022 a success. They include those who have submitted abstracts, the conference steering committee, Pharmacy Research UK, our keynotes speakers, workshop leaders, session chairs and prize judges, our sponsor Day Lewis, our colleagues at University of Bath, and you – research presenters and conference delegates. This thesis deals with theoretical analysis of First Hop Redundancy Protocols. It describes Hot Standby Router Protocol, Virtual Router Redundancy Protocol and Gateway Load Balancing Protocol. It also shows examples of configuration of each protocol on Cisco devices with supported version of the Cisco IOS. Furthermore, this thesis includes design of two of these protocols, Hot Standby Router Protocol and Gateway Load Balancing Protocol, and their implementation in discrete event simulator OMNeT++ and Automated Network Simulation and Analysis library. Finally, the thesis presents results of testing of the implementations in comparison with actual Cisco devices.

This study investigates the potential benefits and challenges of using layer 3 Internet Protocol version 6 (IPv6) configuration commands. Although any other type of layers 3 devices could have been used in this study, only Cisco routers and multilayer switches are considered. The study is conducted using a simulator called Graphical Network Simulator-3 (GNS3). Even though real Cisco Internetwork Operating System (IOS) software is reliably used in this simulator, an avoidable limitation of this method involves not using this software on real routers and multilayer switches. However, it has been found that contrary to Cisco documentation, using the outgoing local interface as next hop address causes IPv6 static routing not to work; it only works when the neighbouring global unicast address is used as the next hop address. Other findings show that when static addresses are configured with Routing Information Protocol Next Generation (RIPng), Enhanced Interior Gateway Routing Protocol version 6 (EIGRPv6) or Open Shortest Path First version 3 (OSPFv3), RIPng has the best round-trip time (RTT), while OSPFv3 gives the best traceroute results. Likewise, 64-bit Extended Unique Identifier (EUI-64) addresses produce better RTT and traceroute results with RIPng than with EIGRPv6 and OSPFv3. Nonetheless, one challenge for RIPng involves failure to start the RIPng process by misconfiguring the ipv6 router rip name and ipv6 rip name enable commands. The benefit of EIGRPv6 is that its RTT is faster than that of OSPFv3 and even if the router identifiers (router-ids) are configured the same on all the routers, the EIGRPv6 process still works well. However, configuring different autonomous system numbers and failing to configure the "no shutdown" or router-id commands results in routing challenges. On the other hand, configuring the same router-id on different layer 3 devices causes OSPFv3 not to work. In spite of this challenge, when OSPFv3 is used with Hot Standby Router Protocol version 2 (HSRPv2), it generates faster RTT than EIGRPv6 and RIPng. However, the success rate of OSPFv3 for failover time of the active router to the standby router is 4% lower than EIGRPv6. In comparison to Internet Protocol version 4 (IPv4), configuring of static and EUI-64 address commands is a very challenging task, because of the hexadecimal nature of IPv6 addresses. Despite this challenge, one benefit of these commands is the ability to use slash notation such as /64 for the prefix length. When used on dual stack commands, static addresses give better native router processing performance with no encapsulation overheads. However, configuring these addresses on dual stack commands in large networks is a challenge. With regard to manual IPv6 tunnelling, configuring the tunnel interface addresses in the same network and failure to configure the tunnel mode ipv6ip command, prevents this technique from working. Although IPv6 static Network Address Translation-Protocol Translation (NAT-PT) commands are easy to configure and to troubleshoot, the NAT-PT router raises the challenge of being a single point of failure in the network. On the whole, given these benefits and challenges, implementing IPv6 in industrial networks should not be scary. The results of this study are useful guidelines on how to efficiently design and configure IPv6 networks in a smooth way.

The dissertation is concerned with the production of a systematic analysis of HSRC research reports into mathematics education in South Africa between 1970 and 1980. Drawing on the theoretical language of Dowling (1995), the analysis focuses on the (re)production of voice and message in the reports. This entails an analysis of positioning strategies that il1ark out voices in the texts and distributing strategies that distribute message across voices. Voices include bureaucratic, academic, teacher and learner voices and knowledge and practices that constitute message distributed to voices relate to mathematical knowledge, pedagogic knowledge and curriculum innovation practices. Positioning and distributing textual strategies with respect to learner and teacher sub-voices are related to the (re)production of theories of instruction that constitute models of acquirers, transmitters and pedagogic contexts and define pedagogic competence in particular ways. The (re)production of theories of instruction in turn are related to the reproduction of social relations in the broader society. It is hoped that the analysis illustrates the generality of Dowling's language for analysing texts. The substantive focus of the study is the analysis of the reports and the language developed in the analysis is used to make some suggestive comments about current mathematics curriculum development in South Africa. It is hoped, in particular, that the focus in this study on discourses in mathematics education in South Africa in the 1970s will contribute to the documenting of the history of mathematics curriculum development in South Africa.

This research focuses on estimating infant and under-five mortality in South Africa for the period 1998-2012, both to update previous estimates taking into account new data and to assess the reasonableness of all estimates. Data from the 2011 Census and the 2012 HSRC survey were used for this purpose. The 2011 Census provided data from deaths reported by households as well as the survival of the most recent births. The 2012 HSRC provided full birth history data for women aged 15-49 which were used for direct estimation of childhood mortality. Deaths reported by households together with census estimates of the number of children under the age of five are used to produce estimates of infant and under-five mortality using the synthetic cohort life table approach.

This thesis puts forward a new solution to provide the reliable network to the Internet Service Provider (ISP). This study mainly focuses on the ISPs network to provide reliability and the load balancing. It offers a guide line for the best reliable solution to the ISPs, individual organizations or other types of service providers which are engaged in providing reliable communications to their subscribers. These reliable services may be real time communications which include weather forecasts, tracking systems, online Internet protocol TV (IPTV) programs and many other ISPs services which are totally depend on the reliable network.

With the appearance and expansion of Internet subscribers all over the world, ISPs services are becoming more popular. The rapid increase of connection-demand and highly traffic network is the main reason behind the need to scale reliable network. To offer better solutions, a new theoretical and practical approach should be considered that can cover the reliable network.

The suggested network structure monitors the links, spreads the network traffic with multiple devices and takes a backup (redundant) link automatically when changes occur in the network topology. In order to support the redundancy, load balancing and reduce the failover time, the hot standby routing protocol (HSRP) is implemented on the suggested network. As we have analyzed that in any network, scalability bringing to raised the network traffic broadcast issue. Broadcast storms can be prevented by setting threshold values of traffic-filters. The threshold level helps to control broadcast traffic in networks.

With regard to suggested solutions, it is necessary to observe the limitations and advantages of the recommended reliable network structure. Therefore, this research will include the advantages and limitations of the techniques used to offer ISP services such as scalability, security and IPv6.

The company Västra Mälardalens Kommunalförbund, VMKF, wishes to revise and optimize their present data and IP-telephony network as of today consists of the three municipalities Köping, Arboga and Kungsör. As a municipal corporation, they seek consultation regarding internal as well as external review and investigation of the main structure of the network, its functionality and safety. By today’s increasing demands of Internet accessibility, availability of services and security far more extends the requirement of a complete network design. The foundation of networking rests on the balance between each of these necessities. Therefore, it is of grave importance to optimize a network design, use of hardware and to minimize the administrative overhead. In particular, when the municipality is short of resources and time means money. By letting an impartial investigation of the network act as a starting point it was established that several improvement could be applied. Among these a reconstructed and improved network topology that includes subjects as routing, switching, safety and security, quality of service and technical administrative overhead and the implementation of a real time monitoring of network bandwidth consumption.

Som ett kommunalägt företag önskar de konsultation rörande intern såväl som extern granskning och optimering av huvuddelen av nätverkets funktionalitet samt säkerhet. I och med dagens ökade Internetanvändning och funktionalitetsbehov ställs allt högre krav på tillgänglighet, säkerhet och användarvänlighet. Nätverksteknik bygger mycket på balansen mellan dessa tre punkter. Därför gäller det att optimera nätverkets design, hårdvaruanvändning och att minimera administrativa laster. Detta i synnerhet då kommunens resurser är knappa och då tid i dagens samhälle innebär pengar. First Hop Redundancy Protocols are network protocols which are designed to protect the default gateway and also to ensure high availability in the network by using redundancy. One of the most parts of VOIP management is fault management and, in having a good fault management, finding good mechanisms to detect faults in the network have to be considered.

The main focus of this project is to implement different types of fast fault recovery protocols in networks, especially networks that carry IP telephony. Having a complete understanding of some common link failure detection and fault recovery protocols, such as spanning tree protocol (STP), rapid spanning tree protocol (RSTP) and per-VLAN spanning tree protocol (PVSTP), and also having a complete understanding of three other common techniques for fault detection and fault recovery, such as hot standby routing protocol (HSRP), virtual router redundancy protocol (VRRP) and gateway load balancing protocol (GLBP) will be regarded in the project. We are going to test some fault recovery protocols which can be used in IP telephony networks and choose the best. We intend to focus on this issue in LAN environment in theoretical descriptions and practical implementations.

The final outcome of the thesis is implementation in the Halmstad University’s lab environment to obtain the final result. For doing our thesis, we are going to use some technical tools as hardware tools (Cisco L3 and L2 switches, Routers, IP Phones) and tools which are used for network performance monitoring, like as CommVeiw.

Abstract Aishwariya Barmer Hill (ABH) field is a moderate permeability (0.5 – 4 mD) oil bearing porcellanite with alternating sequences of tight shale. After successful appraisal campaign a full field development with multi-stage fracturing using cemented frac sleeves, field was brought online with Hydraulic Sucker Rod Pump (HSRP) as artificial lift and has been on production since 2019. However, a need was felt to review the frac and completion design on account of challenges faced during fraccing and unplanned downtime during production operations. Critical observations that prompted a change in completion and frac technology are: Formation rock pebbles and proppant were observed in the wellbore during workovers.Reservoir's low Young's Modulus (YM) allows the generating high strains at low pressures, while low Poisons Ratio (PR) makes the rock brittle and shatter under high deformation. Consequently, shattered rock was not able to hold the proppant in place after fracture closure resulting into flowback of proppant and pebbles.Debris fill in wellbore resulted in production impairment and malfunctioning of HSRP. To mitigate the identified risks, the design change incorporates measures to address post fracturing production problems related to high treating pressures as well as optimize number of frac stages and stage spacing. Uniform proppant distribution with lesser number of stages is targeted by utilizing limited entry technique to help in distributing treatment pressures and proppant in multiple clusters as well as limit net pressure build up in each frac. It will help prevent rock shattering and better retention of proppant after frac closure. Completion design workflow includes log based zonal isolation between each stage and frac design for two to three cluster per stage. The revised design will predict the number of stages in each well for optimal utilization of wellbore for best economical production. Revised frac design has been implemented in 5 infill wells wherein 66 stages have been pumped without TSO signature or premature screen out in any of the stages. Wells have been put on production and are performing better than rest of the wells in the field. There has been no evidence of debris accumulation in wellbore or proppant flowback in production fluid. Further drilling campaign for 15 wells has been planned with cluster frac strategy with revised frac design.

The MIMI project was initiated by the DSI in partnership with the South African Local Government Association (SALGA), the HSRC and UKZN. The purpose of this initiative was to develop an innovative tool capable of assessing and measuring the innovation landscape in municipalities, thus enabling municipalities to adopt innovative practices to improve service delivery. The outcome of the implementation testing, based on the participation of 22 municipalities, demonstrated the value and the capacity of MIMI to produce innovation maturity scores for municipalities. The digital assessment tool looked at how a municipality, as an organisation, responds to science, technology and innovation (STI) linked to service delivery, and the innovation capabilities and readiness of the municipality and the officials themselves. The tool is also designed to recommend areas of improvements in adopting innovative practices and nurturing an innovation mindset for impactful municipal service delivery. The plan going forward is to conduct learning forums to train municipal officials on how to use the MIMI digital platform, inform them about the nationwide implementation rollout plan and support municipal officials to engage in interactive and shared learnings to allow them to move to higher innovation maturity levels. The virtual launch featured a keynote address by the DSI Director-General, Dr Phil Mjwara; Prof Mehmet Akif Demircioglu from the National University of Singapore gave an international perspective on innovation measurements in the public sector; and messages of support were received from MIMI partners, delivered by Prof Mosa Moshabela, Deputy Vice-Chancellor (DVC) of Research at the University of KwaZulu-Natal (UKZN) and Prof Leickness Simbayi, Acting CEO of the Human Sciences Research Council (HSRC).
