La caballa Scomber japonicus (Perciformes: Scombridae) es una especie pelágica de gran importancia económica en el Perú y en varios países alrededor del mundo. A pesar de su importancia, no se han reportado estudios concernientes a la genética de poblaciones en esta especie en el Perú. Por este motivo, el objetivo de la presente tesis fue evaluar la estructuración genética de caballa en el mar peruano. Se utilizó la Región Hipervariable I (Dominio ETAS) de la Región Control Mitocondrial junto a cinco marcadores microsatélites. El análisis del marcador mitocondrial evidenció una escasa diferenciación genética entre las poblaciones estudiadas y que el recurso atravesó un proceso de expansión poblacional en el pasado. De los marcadores microsatélites solamente el locus SJT18 fue útil para el análisis genético-poblacional debido a que los cuatro marcadores restantes tuvieron serios problemas de efecto stuttering (bandas tartamudas). Este locus, al igual que el marcador mitocondrial, sugirió una escasa diferenciación entre las poblaciones. Se concluye que la caballa peruana comprende una sola unidad panmíctica lo cual refuerza la hipótesis de un único stock pesquero.

Forty-four compounds were isolated from the whole plant of Houttuynia cordata Thunb. Of these, forty compounds were characterized by comprehensive analyses of their 1D and 2D NMR and mass spectra. Among them, houttuynamide A and houttuynoside A were found be new compounds. The thirty-eight known compounds were identified as aristolactam BⅡ, aristolactam AⅡ, piperolactam A, 3,4-dimethoxy-N-methyl aristolactam, lysicamine, splendidine, cepharadione B, norcepharadione B, noraristolodione, N-(1-hydroxymethyl-2-phenylethyl)benzamide, N-(4-hydroxyphenylethyl)benzamide, benzamide, 4-hydroxy benzamide, 4-hydroxy-3-methoxybenzamide, 6,7-dimethyl- 1-ribitol-1-yl-1,4- dihydroquinoxaline-2,3-dione, (1H)- quinolinone, indole-3- carboxylic acid, vomifoliol, dehydrovomifoliol, reseoside, 7-(3,5,6-trihydroxy- 2,6,6-trimethyl- cyclohexyl)but-3-en-2-one, 6-(9-hydroxy-but-7-enyl)-1,1,5- trimethylcyclohexane- 3,5,6-triol, quercitrin、quercetin-3-O-β-D-galactopyranoside, afzelin, vanillic acid, methyl vanillate, vanillin, protocatechuic acid, 4-hydroxybenzoic acid, methylparaben, p-hydroxybenzaldehyde, cis and trans-methyl ferulate,β- sitosteryl glucoside,β- sitosterol and cycloart-25-ene-3b,24-diol by comparison of their physical and spectral data with those reported in the literature. Furthermore, some of the isolated compounds were evaluated for their anti- tyrosinase and anti-oxidant activities. Cepharadione B showed the storng inhibition activity against tyrosinase. Quercitrin, quercetin-3-O-β-D-galactopyranoside, vanillic acid, protocatechuic acid showed 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical- scavenging activity of which quercitrin and quercetin-3-O-β-D- galactopyranoside have the stronger scavenging activity than Vit E.

Enterovirus 71 (EV71) is a positive-stranded RNA virus in the genus Enterovirus of the family Picornaviridae. EV71 infection can cause hand, foot, and mouth disease (HFMD), herpangina and associated with fatal pulmonary edema as well as severe neurological complications, including encephalitis, meningitis, and a poliomyelitis-like syndrome in children. Several major outbreaks of EV71 have occurred in recent years in Taiwan. In this study we screened 22 Chinese medicine extracts for anti-EV71 activity. We found Houttuynia cordata Thunb. can neutralize the EV71-induced cytopathic effect (CPE) in Vero cells. The 50% inhibitory concentration of H. cordata on EV71 was approximately 125.92±27.837 g/mL. A plaque reduction assay showed that H. cordata can reduce the plaque formation. H. cordata was able to reduce viral protein synthesis and also abate the apoptotic process in enterovirus 71-infected Vero cells. We concluded that H. cordata possesses antiviral activity and has a potential for development as an anti-enterovirus 71 agent.

The study aimed to evaluate the effect of bacteriostasis on Staphylococcus aureus, Escherichia coli, Streptococcus pneumoniae, Hemophilus influenzae and Pseudomonas aeruginosa by four cultivars of water extracts of medicinal Houttuynia cordate. In addition, effective elements including quercetin, decanoyl acetaldehyde, and methly-n-nonylketone would be compared for the effect of bacteriostasis. Among four cultivars of water extracts of Houttuynia cordate, only had quercetin been detected. quercetin has the most amont of HC2 and HC3, HC4 the second, and HC1 the fewest. While HC2 and HC3 demonstrated the best effect of bacteriostasis, HC1 showed the worst. The result displayed that the amount of quercetin was positively correlated with the effect of bacteriostasis. In addition, different species of water extracts of Houttuynia cordate had better bacteriostasis effect on Staphylococcus aureus and Streptococcus pneumoniae than on Escherichia coli and Pseudomonas aeruginosa. In sum, the experiment had confirmed the effect of bacteriostasis of Houttuynia cordate. In addition to bacteriostasis, Houttuynia cordate could function as antioxidant, antiviral, diuretic, cough suppressant etc. The researcher has assumed that more uses of Houttuynia cordate will be discovered if deeper research regarding the mechanism of pharmacological effects of Houttuynia cordate can be undertaken.

Houttuynia cordata, a Saururaceae herbaceous plant, has been applied as a traditional medicine to improve immune responses and to exert anti-viral, anti-bacterial and anti-inflammatory effects.Phytochemicals in Houttuyniacordata can be divided into essential oils, polyphenols, flavonoids, alkaloids and other substances.It is known that Houttuyniacordatacontains numerous major active ingredients, such asdeCanoy acetaldehyde, lauric aldehyde, α-pinene, linlool, etc. However, different extraction methods would lead to the significant variation inactive ingredients within the product development. Using different extraction methods, the objective of this study was to compare biological activities of various Houttuynia cordata extracts.Stems and leaves of dried plants were finely ground, and then extracted with water at room temperature, 50 ºC or 70 ºC, or with 70% ethanol, ethyl acetate or chloroform at room temperature for 24h.The extracts were filtrated, vacuum-concentrated, lyophilized and stored at -20ºC.Results showed that the amount (1.58 g) of water extract from 10g ofHouttuyniacordata powder was highest, followed by those ofalcohol extract (1.27 g), chloroform extract (0.28 g) and ethyl acetate extract (0.27 g).Furthermore, the weight of the water extract isincreased as the temperature increases.Among various Houttuyniacordata extracts,the ethanol extract contained the highest amount of total polyphenols (1155 uggallic acid equivalent/g),however, the ethyl acetate extract contained the least (358 uggallic acid equivalent/g). The amounts of total flavonoidsextracted from organic solvents(813~1142 ug catechin equivalent/g) were higher than thosefrom three water extracts(~250 ug catechin equivalent/g), and the chloroform extract was the most (1142 ug catechin equivalent/g).When fatty acid compositions of the Houttuyniacordata sample were analyzed, it contained palmitic acid (C16: 0) (30.1%), α-Linolenic acid (C18: 3n-3) (21%), Oleic acid (C18: 1n-9) (15.6%), Linoleic acid (C18: 2n-6) (14.8%), Stearic acid (C18: 0) (9.3%) and others.When examined antioxidant properties, the ethanol extractexerted the strongest DPPH scavenging activity(80%) and the reduction capacity. However, the chloroform extract had the least abilities to scavenge DPPH free radicals (40%) and perform the reduction capacity.No anti-bacterial effect of Houttuyniacordataextracts was observed when using methods of disk diffusion method and minimal inhibition concentration.Since the addition of Houttuyniacordataextract at concentrations above 100 ug/ml resulted incytotoxic effect on murine RAW264.7 macrophages, a fixed concentration (50 μg/ml)of various extract was used to determine nitric oxide (NO), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in following cell culture studies.Among various extracts, the ethanol extract inhibited the production ofNO (by 28 %), TNF-a(by 27 %) and IL-6 (by 72 %).Our results showed that Houttuyniacordata extracts exerted the anti-inflammatory properties by inhibiting the production of NO, TNF-α and IL-6.In summary, active polyphenols, flavonoids and other substancesextracted from Houttuyniacordataexert antioxidant and anti-inflammatory properties, and these active compounds can be used as the development of future biopharmaceutical products.

Houttuynia cordata Thunb. is rich in nutrient and functional constituents, and it’s valuable for industrial application. In order to improve the quality and yield of pig thigh, the antioxidant activity in different tissues and leaf position were studied and followed by the effect of light quality, nitrogen concentration and exogenous salicylic acid and methyl jasmonate application on growth and endogenous compounds of pig thigh. First of all, in the part of different tissues and leaf position, the samples were divided to five parts, upper leaves, middle leaves, lower leaves, stem and root. Results showed that the DPPH scavenging activity and the total phenolic compounds were highest for lower leaves and lowest for root. Chlorogenic acid was highest in leaves. To sum up, lower leaves contained more functional constituents and less for root. Secondly, in the part of light quality, results showed that there was no significant difference for the plant growth and biomass for all treatments. Blue light increased total phenolic compounds and chlorogenic acid for 30% compared to other treatments. Blue light is suggested to be the light source for indoor cultivation and supplement light for greenhouse to improve functional constituents. Thirdly, in the part of nitrogen concentration, results showed that there was no effect on plant growth without nitrogen fertilization. The antioxidant indices were higher due to the stress of nutrient deficiency. There was no significant difference for growth and antioxidant indices for treatments of different nitrogen concentration applications because the period of treatment maybe was too short. The antioxidants were highest for 10 and 15 mM treatments which were similar with normal fertilization formula. Furthermore, the antioxidants were decreased for higher nitrogen concentration applications. To sum up, the nitrogen concentration fertilization without adjustment for other elements in formula (6 mM phosphate, 4 mM potassium, trace element added) is suggested to be 10 to 15 mM to improve functional constituents. The last part of this study is about the effects of exogenous salicylic acid and methyl jasmonate application on growth and endogenous compounds. In the salicylic acid experiment(0、0.25、0.5、1 and 2 mM), there was no significant difference for plant fresh weight, dry weight, SPAD, NDVI, and Fv/Fm value. The development of leaves was retarded for 1 mM and higher concentration. The total phenolic compounds were most for 0.5 mM treatment. There was no significant difference between foliar application and drench. In methyl jasmonate experiment(0、0.25、0.5、1 and 2 mM), there was no significant difference for application concentration and method in plant growth indece. The total phenolic compounds were most for 2.0 mM treatment with drench. The results suggest that the optimum exogenous application of salicylic acid is 0.5 mM for drench and methyl jasmonate 2.0 mM for drench. The functional constituents were affected by the different tissues and leaf position. Moreover, the growth and the functional constituents of pig thigh were also affected by light quality, nitrogen concentration and exogenous salicylic acid and methyl jasmonate application. This study established the optimum cultivation conditions for pig thigh cultivation and elevated the overall benefit of medical industry.

Studies on genetic variation of Houttuynia cordata in Taiwan. Summary This studies on genetics variation of populations and cultured lines of Houttuynia cordata Thunb. in different zones of Taiwan was preserve of germplasms and development and application of products for reference. From July to December in 2000, wild populations and cultured lines were collected in each zone of Taiwan, and then preceded a gamogenesis in the greenhouse of National Chung Hsing University. 150 days later, plant characters of each populations and lines were investigated. Moreover, applying RAPD to analysis polymorphisms of populations and lines can realize variation of genomes. The results were as follows: 1.Of the populations of H. cordata Thunb., in different zones of Taiwan, the means of plant characters, relationship among characters, and statistics of Hotelling’s T2 all showed the significant difference of ecology among populations. 3 clusters were be classified in ten populations according to analysis of principal component analysis and cluster analysis: The cluster Ⅰ including Chilung, Fuhsing, Shengan, Renai, Lukang, Meinung and Hengchun, the cluster Ⅱ including Yangmin and Taitung, and the cluster Ⅲ including Suao. 2.Utilizing 40 primers proceed to RAPD for 10 populations of H. cordata Thunb. of Taiwan, 14 primers could showed polymorphisms, and all 101 bands appear, and 72 bands would be regarded as the markers. The similarity coefficient of populations, from 0.341 to 0.988, showed populations have greater genetic variation. 3 clusters were be classified according to cluster analysis : the cluster Ⅰ including Chilung, Yangmin, Meinung, Shengan, and Renai, the cluster Ⅱ including Fuhsing, Taitung and Hengchun, and cluster Ⅲ including Suao. 3.Using plant characters of 24 H. cordata Thunb. cultured lines to proceed with ANOVA found significant difference among lines. 4.Utilizing 14 primers proceed with RAPD for 24 cultured lines of H. cordata Thunb. of Taiwan, only 8 primers could showed polymorphisms, and all 23 bands appear, and 18 bands would be regarded as the markers. The similarity coefficient of lines, from 0.281 to 1.000, also showed populations have great genetic variation. 2 clusters were be classified according to cluster analysis: the cluster Ⅰ, 21 lines, inclusive of Taitung 1, Yunlin, Luodong, Hualian, Chishang, Changbin 1, Donghe, Taimali, Taitung 2, Changbin 2, Chenggong, Yuanli 1, Kouhu, Caotun, Xinshe, Fengyuan, Cuoxing, Taipei, Tamsui, Longtan, Fangyuan, and the cluster Ⅱ , 3 lines, inclusive of Pitou, Yuanli 2, Yongjing.

This study aimed at finding the optimum extraction conditions of Houttuynia cordata over the boiling water method. In this study, we applied the L9 (3³) orthogonal test to explore optimum extraction parameters to assess the relationship between antibacterial activity total polysaccharides content, antibacterial activity and total flavonoids content. The results showed that the best Houttuynia cordata total flavonoids extraction conditions were:the solid-liquid ratio of 9g/90ml, temperature of 80℃ and time of 45 minutes. The best total Polysaccharides extraction conditions were:the solid-liquid ratio of 9g/90ml, temperature of 100℃ and time of 45 minutes. The optimal Staphylococcus aureus antibacterial effect conditions were:the solid-liquid ratio of 4.5g/90ml, temperature of 90℃ and time of 30 minutes. The antibacterial effect was 93.4 percent. The optimal Escherichia coli antibacterial effect conditions were:the solid-liquid ratio of 9g/90ml, temperature of 80℃ and time of 45 minutes. The antibacterial effect was 71.5 percent. The antibacterial effect of Houttuynia cordata water extract is greater on Staphylococcus aureus than on Escherichia coli. Under the extraction conditions, the temperature shouldn’t be high and the time shouldn’t be long. But such antibacterial effect of either total flavonoids or the total polysaccharides content is not proportional.

Acetaminophen (APAP), a commonly used antipyretic and analgesic agent, could cause acute liver injury by overdose. Houttuynia cordata Thunb is a medicinal plant used in traditional Chinese medicine. The aim of this study was to examine the hepatoprotective effects of Houttuynia cordata Thunb aqueous extract (HCE) against APAP-induced injury. Male C57BL/6JNarL mice were randomly divided into normal group, control group and HCE group. After 5 weeks supplementation, mice were treated by APAP intraperitoneally, and killed after 24 hours. Blood and organs from each mouse were collected. Results showed that the intake of HCE significantly alleviated APAP-induced aspartate aminotransferase (AST) and alanine aminotransferase (ALT) elevation; retained glutathione (GSH), glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT) and total antioxidant capacity (TAC) activities or levels; reduced the level of inflammatory related factors such as interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and monocyte chemotactic protein-1 (MCP-1). Furthermore, HCE intake attenuated cytochrome P2E1 (CYP2E1) activity and malondialdehyde (MDA) production. These findings supported that pretreatment of HCE protected mice liver against APAP-induced oxidative and inflammatory injury. Both Houttuynia cordata and Lonicera japonica Thunb are perennial herbs that belong to Saururaceae and Caprifoliaceae, respectively. In this study, we focused on the different conditions of Houttuynia cordata (including fresh materials, frozen dried and shade dried) combined with the shade dried Lonicera japonica Thunb to be extracted with 95% ethanol, and further evaluated the anti-oxidation, skin whitening, wound healing, and anti-inflammatory activities of the Houttuynia cordata and Lonicera japonica Thunb crude extracts with different shade-dried conditions. The results revealed that the frozen-dried Houttuynia cordata and shade-dried Lonicera japonica Thunb crude extracts obtained from the extraction by 95% ethanol heated at 50 ℃ for 4 hours showed a higher inhibitory effect on the scavenging of free radicals and the activity of tyrosine kinase. The prescription of Houttuynia cordata and Lonicera japonica Thunb extracts at a ratio of 3 : 1 showed a higher free radical scavenging activity, whereas at a ratio of 1 : 3 showed a higher inhibitory activity of tyrosine kinase. In addition, it showed significant differences of anti-bacteria activities (Staphylococcus aureus, Bacillus subtilis, and Escherichia coli) between Houttuynia cordata and Lonicera japonica Thunb, and the prescription of Houttuynia cordata and Lonicera japonica Thunb extracts at a ratio of 3 : 1 exerted the best anti-bacteria activity. Moreover, the prescription of Houttuynia cordata and Lonicera japonica Thunb extracts at a ratio of 3 : 1 had a cytotoxicity activity to B16 melanoma cells, and also had an anti-inflammatory activity in lipopolysaccharide-induced inflammation in RAW264.7 Cells. In the component analysis of Houttuynia cordata and Lonicera japonica Thunb, the prescription of Houttuynia cordata and Lonicera japonica Thunb extracts at a ratio of 3 : 1 has chlorogenic acid, quercetin, and luteolin in rich. Since the aim of this study is focused on the anti-bacteria and anti-inflammatory activities of the study materials. Thus, among the five pre-tested results, we used the extracts of Houttuynia cordata and Lonicera japonica Thunb at a ratio 3 : 1 to manufacture an ointment. We found that the ointment facilitated the growth of epithelial cells and granuloma to show benefits in the wound healing. Taken together, the results obtained in this study provided information of refine or decoct of Houttuynia cordata and Lonicera japonica Thunb for the pharmaceutical and cosmetics industries in the future. The main purpose of this experiment is to explore the antioxidant activities of three herbs’ extracts, Houttuynia cordata, Saussurea involucrate, Liu-Dou-Huang and to evaluate the possibilities of formulating these herbs being part of anticancer formula in Chinese medicine. Methanol and double deionized water at 4℃, 25℃, and 50℃, respectively, were used as solvents to extract the components of these herbs. The extracts were used to measure antioxidant activity, such as DPPH scavenging ability and reducing power. Extracts from green tea were used as comparison. The total phenolic contents of extracts were measured in order to reveal the relationship between the antioxidant activity and the total phenolic contents. The DPPH antioxidant activity of methanol extract of Houttuynia cordata is better than those of water extracts. The water extracts from different temperatures did not show obvious differences in their antioxidant activity. But, the reducing power of the 50℃ extract of Houttuynia cordata is better than those of 25℃, 4℃, and methanol extract. The DPPH antioxidant activity of water extract of Saussurea involucrate is better than that of Liu-Dou-Huang. However, the latter performs better than the former one in its reducing power. In as low density as 1mg/mL, green tea does have 73% in DPPH antioxidant activity, and 0.9 in reducing power as well. The reason for green tea performs as the best of all the samples may be related with its richness in phenolic contents. Total phenolic contents and DPPH scavenging ability showed high relationship (r=0.88), only mildly related with reducing power (r=0.54). Nonetheless, the amount of phenolic contents of other extracts from these three main research targets did not show any relative with DPPH scavenging ability and reducing power. It is obvious that other components other than phenolic compounds also play the important role on antioxidant activity to protect the living organisms. We are certain that the extracts from Houttuynia cordata, Liu-Dou-Huang, green tea and Saussurea involucrate all prove good antioxidant activities. Further researches and evidences on cell protection of antioxidant activity are needed to possible application these herbs into anticancer formula.

In recent years, environmental awareness is growing around the world. The use of materials from natural Chinese herb resources in cosmetic production and consumption processes has become one of the mainstream in the cosmetic manufacturing company. Cosmetics ingredients come from a variety of synthetic compounds, which is important factors to cause skin irritation and inflammation. Therefore, Chinese herbs can be applied in food, medicine and cosmetics. This study used one of Chinese herbs, Houttuynia cordata, to characterize the different concentrations hydrosol applied in the cosmetics ingredients efficiency, and established a cosmetics test platform, including sunscreen, whitening, humidification, antibacterial, antioxidation and Gas chromatography mass spectrometry (GC-MS) analysis study. 2,4-ditert-butylpheno (hexadecanoic acid) is a kind of antioxidant identified by GC-MS in Houttuynia cordata hydrosol sample. Antioxidation analysis showed that DPPH could remove free radicals and improve reduction. The Houttuynia cordata hydrosol (dry weight/water ratio 1:5) showed 21% clear rate and 73.3 % reduction rate. Freeze drying Houttuynia cordata hydrosol with the 1:0.5 concentration showed 37.6% clear rate and 86.1 % reduction rate. The Houttuynia cordata hydrosol with pH 5~6 can be used in the cosmetic ingredients, which can direct contact with the skin. The skin moisturizing test in all groups showed that moisturization in 10 min rendered the best results. Houttuynia cordata hydrosol not only is an antioxidant, but prevents production of melanin by inhibiting tyrosinase activities. Houttuynia cordata hydrosol can be used as general makeup water and raw materials for cosmetic manufacturing. Keywords: GC/MS, antioxidatioln, whitening, antibacterial, hydrosol

Type 1 diabetes mellitus (T1DM) is an autoimmune disease caused by T-cell mediated destruction of pancreatic β-cells, which leads to insulin deficiency. Our previous studies found that intake of Houttuynia cordata Thunb aqueous extract (HCE) improved the high-fat-diet induced lipid disorders in mice; and provided hepatoprotective effects against APAP-induced injury in mice via its antioxidant and anti-inflammatory capacity. Therefore, the aim of this present study was to determine whether HCE could attenuate oxidative and inflammatory stress in STZ-induced diabetic mice. Male C57BL/6JNarL mice were i.p. 45 mg/kg/day STZ for consecutive 5 days to induced diabetes. Experimental design was divided into normal group (n = 10), DM group (n = 15), DM + 4% HCE group (n = 15), DM + 8% HCE group (n = 15). After 11 weeks supplementation, mice were sacrificed; blood and organs were collected for analyses. Results showed that the intake of HCE significantly improved hyperglycemia, hypoinsulinemia; increased the GSH level and GPx, SOD, CAT activities of heart; decreased renal IL-6, TNF-α and MCP-1 level. Furthermore, HCE intake attenuated renal AGEs level. These findings indicated that Houttuynia cordata Thunb could delay diabetic progression.

第一部份 魚腥草之化學成分研究 In the present investigation, the whole plant of Houttuynia cordata Thunb. was collected and extracted with methanol. The extracts were further separated by column chromatography and high performance liquid chromatography. Ten compounds were isolated, and they contain two aristolactams: aristolactam BⅡ(2) and aristolactam AⅡ(5); one oxoap- orphine: splendidine (10); three 4,5-dioxoaporphines: cepharadione B (1), piperadione (3) and norcepharadione B (4); two flavonoids: quercitrin (6) and quercetin-3-O-β-D-galactoside (7); one benzenoid: caffeic acid (8); one amide: benzamide (9)。 These compounds were characterized by comprehensive analysis of their 1D and 2D NMR and mass spectra. Among them, piperadione (3) and caffeic acid (8) are isolated from Houttuynia cordata for the first time. 第二部份 葎草之化學成分研究 In the present investigation, the whole plant of Humulus scan- dens (Lour.) Merr. was collected and extracted with methanol. The extracts were further separated by column chromatography and high performance liquid chromatography. Five compounds: β-stiosteryl-3-O-β-D-glucoside (a), sucrose (b), myo-inositol (c), 3-hydroxy-4,4-dimethyl-4-butyrolactone (d), 2,3-dihydroxy-2- methylbutyrolactone (e) were isolated from Humulus scandens (Lour.) Merr. The structural elucidation of the isolated natural products were established by spectral methods.

The thirty-five collections of Houttuynia cordata Thunb. collected in Taiwan were used as materials to investigated the variation of plant parts , cropping seasons and growth stages for the content of volatile oils and oil components in the aerial part. In addition, the estimators such as correlation analysis for volatile oil components and anatomic characters were used to reference. The results follow that after investigation, the weights of fresh rhizome in thirty-five collections were larger than fresh aerial part, but content of volatile oil in the aerial part were larger than in the rhizome. Content of volatile oil in the aerial part and rhizome were at the highest level in spring cropping, especial at flowering and fruiting stage. After analysis of volatile oil components content in different plant part, the content of myrcene was as follows in the aerial part more than in rhizome, the contents of methyl-n-nonyl ketone, α-pinene, β-pinene, and d-limonene were as follows in rhizome more than in the aerial part, and the contents of α-pinene, β-pinene, and d-limonene in the aerial part of most collections were almost undetectable. In life cycle, the contents of all volatile oil components in the aerial part were at the highest level at flowering stage (in June) and therewith lost quickly. When end of July, plants were cut, the contents of all volatile oil components in the aerial part increase gradually, and at the high level in November, and therewith at the lowest level. The results of ANOVA indicate that the treatment of collections, cropping seasons and the interaction of collections and cropping seasons get a significant level for volatile oils content and volatile oil components content of the aerial part. Mean of fresh aerial part weights of all collections in fall cropping was larger than mean in spring cropping. In spring cropping, collection 35 was at the highest level and collection 6, 31 were at lowest level for fresh aerial part weights. In fall cropping, collection 3 was at the highest level and collection 5 were at lowest level for fresh aerial part weights. Mean of volatile oils content for all collections in spring cropping was larger than mean in fall cropping. In spring cropping, collection 7, 18 were at the highest level and collection 6 was at lowest level for volatile oils content; in fall cropping, collection 7, 26 were at the highest level and collection 6 was at lowest level for volatile oils content. Mean of methyl-n-nonyl ketone and myrcene contents for all collections in spring cropping was larger than mean in fall cropping. In spring cropping, collection 18 was at the highest level and collection 5, 31 were at lowest level for methyl-n-nonyl ketone content; collection 7 was at the highest level and collection 24 was at lowest level for myrcene content. In fall cropping, collection 26 was at the highest level and collection 1 was at lowest level for methyl-n-nonyl ketone content; collection 7, 30 were at the highest level and collection 6 was at lowest level for myrcene content. Distributes and numbers of oil cell in tissues of the aerial part differed with collections, coefficient of oil cell numbers variation in stem was larger than in leaf, and coefficient of oil cell numbers variation in midrib was smallest. According to correlation analysis for volatile oils, fresh weight of the aerial part was reciprocal positively correlated with content of volatile oils and content of myrcene; content of volatile oils of the aerial part was reciprocal positively correlated with contents of methyl-n-nonyl ketone and myrcene. Content of methyl-n-nonyl ketone was positively correlated with oil cells in leaf; oil cells in leaf were reciprocal positively correlated with oil cells in midrib. The thirty-five collections could be distinguish into four groups and five groups respectively by weights of fresh aerial part, content of volatile oils, and content of methyl-n-nonyl ketone in spring and fall cropping. In spring or fall cropping, collection 7 was at the highest level and collection 6 was at the lowest level for fresh weights of the aerial part, content of volatile oils and it’s components in all collections.

Thirty-eight Houttuynia cordata Thunb. accessions collected from Taiwan were used as materials. To understand the cytogenetic variation among collected accessions, Chromosome number, pollen formation and the pollen fertility of collection accessions were surveyed. The analysis result indicates that the chromosome number of 38 accessions is ranged from 63 to 90 as same as septuploid, octoploid, enneaploid and decaploid, respective. Included aneuploid with 84 and 85 chromosome. Aneuploid offspring collected from Cheng-gong and euploid offspring collected from Suao both seeded in aneuploid and euploid. In the meiosis process of pollen mother cell, unequal chromosomal segregation and abnormal cytokinesis generated abnormal sporads. Otherwise, I observed the occurrence of cytomixis in pollen mother cell at microspore period. Cytomixis of the frequency among accessions were ranged from 14.25 % to 6.69 %, especially the frequence of accessions collected from Cheng-gong is highest (14.21 %). Pollen fertility of the frequency among accessions were ranged from 4.25 % to 0.19 %, especially the frequence of accessions collected from Taipei II is highest (4.25 %). Finally, the analysis results supports that the variation of chromosome number among 38 assessions collected of H. cordata Thunb. is probably caused by abnormal meiosis and cytomixis.

Thirty-three lines of Hottuynia cordata Thunb. in Taiwan were cultured at the same locality and then used to investigate the intra-specific variation of anatomic characters, mainly included oil cells, starch grains and crystals, with paraffin sectioning. Data was analyzed by cluster and PCA procedure of MVSP software. From the cross sections of tissues, lots of oil cells are distributed in the large parenchyma in contact with the epidermis of leaf, stem and rhizome. Oil cells are widely dispersed in the bundle sheath and phloem of leaf and in the cortex, pith and phloem of stem and rhizome. Starch grains are mainly distributed in the cortex and pith of stem and rhizome. Among the 33 H. cordata lines, there were significant differences in the anatomical characteristics. The examined characteristics with larger variation were the number of starch grain in storage tissue of leaf edge, the number of oil cell in epidermis of leaf blade, the crystal in leaf midrib, the number of oil cell in the pith of stem and the number of crystal in the pith of rhizome. The results of correlation analysis for oil cells indicated that the number of oil cell in epidermis are positively correlated with that in storage tissue of leaf edge. Oil cells of every tissue in stem have the reciprocal positive correlation, and in rhizome so on. Analyzing the relation ships among organs, the number of oil cell in the phloem of midrib are positively correlated with that in the exodermis, cortex and pith of stem. The number of oil cell in the exodermis of stem is also positively correlated with that in phloem of rhizome. According to the result of the correlation analysis for starch grains, the number of starch grain in every tissues of leaf edge and blade have a reciprocal positively correlation, and of stem and of rhizome so on. For relation ships among organs, the number of starch grains in midrib was positively correlated with that in stem. In addition, the number of crystal in midrib was positively correlated with that in stem and in rhizome. Depending on the cluster analysis, 33 lines are separated into three groups. The cluster I have three lines. The cluster Ⅱ and Ⅲ have twelve and eighteen lines, respectively. However, according to the principle component analysis, 33 lines are separated into two groups and four individual lines. The first principal component represents the content of oil cell in bundle sheath and phloem and the starch grain in storage cell of midrib, the second principal component represents the content of starch grain in epidermis of leaf edge and blade, and the third principal component represents the content of oil cell in cortex, phloem and pith of stem and the content of crystal in midrib.

Houttuynia cordata (HC) was one of common Chinese medicine herbs inAsian. The essential oil of HC contained bioactive constituent (Decanoyl acetaldehyde). The chemical property of Decanoyl acetaldehyde was unstable, it oxidized easily to Methyl-n nonylketone. Decanoyl acetaldehyde and Methyl-n-nonylketone were used as the bioactive constituent on HC, and could be criterion of quality determination. Besides, this study discussed the influences of bioactive constituent for various seasons (spring, summer, fall and winter), drying methods (room temperature drying, hot air drying and freeze drying) and the sections of plant (leaf, aerial stem and subterraneous section). First, essential oil were extracted and concentrated by Likens-Nickerson apparatus and Kuderna-Danish concentrator. Then, the constituents of essential oil were separated and compared by using the gas chromatography and mass spectrometry. The experimental results exhibited the essential oil of HC had complex ingredients. The retention time of Decanoyl acetaldehyde and Methyl-n-nonylketone were 104 minute and 76 minute, respectively. The relative content of HC bioactive constituents were higher after drying, highest on freeze drying, higher on hot air drying, and lowest in room temperature drying. Relative content of HC bioactive constituents were highist at subterraneous section, higher at aerial stem, lowest at leaf of plant. And relative content of bioactive constituents were higher at summer and fall. The best pocess were used freeze drying at summer (48.80%) and fall (50.72%) at subterraneous section. The oxidation on bioactive constituents of HC were 50% at leaf, 75%-90% at aerial stem, and 90%-95% at subterraneous section. Hue angle of HC of leaf were green, brown at aerial stem, yellow at subterraneous section, and it showed that browning of HC was due to the drying process. Chroma of HC increased by freeze drying, decreased by room temperature drying and hot air drying . The Hue angle of HC was increased at freeze drying. According ot the relationships between bioactive constituents of HC and color, the color of HC the could be a criterion of fast test in the bioactive constituents of HC. This study used the HC powder to test the perilla britt houttuynia cordata tea bag. The results of sensory cvaluation indicated the tea bag was best for the coarse and low temperature low humidity drying process. It could be the reference for the product design of perilla britt-houttuynia cordata tea bag in food stuff industry.

The twenty-nine collections of H. cordata Thunb. collected from Taiwan were used as materials to investigate the variation of yield and active components and select the high production and active components of collection to proceed the cultivation experiment. To investigate the effect of different cultivation for the yield and active components ,and collect those results for cultivating of H. cordata Thunb. in Taiwan. The results were summarized as follows: 1. The twenty-nine collections of H. cordata Thunb. collected from Taiwan were investigated the variation of yield and active components., the results indicate that the yield and active components like kind of flavanone glucoside content, volatile oil content and the methyl-n-nonyl ketone content of collections get a significant variation among collections. 2. The excessive high soil RH could promote high yield, but reduce the active components like kind of flavanone glucoside content, volatile oil content and the methyl-n-nonyl ketone content of Houttuynia cordata Thunb. ,the results implied that the high soil RH were unbeneficial to the yield of active component of H. cordata Thunb. 3. The shading treatment could promote high yield, but reduce the active components like kind of flavanone glucoside content, volatile oil content and the methyl-n-nonyl ketone content of H. cordata Thunb. , the results also implied that the high shade unbeneficial to the yield of active component of H. cordata Thunb. 4. The different parts of fresh yield fertilized with organic fertilizer were higher than chemical fertilizer and non-fertilizer, but the inverse results for the aerial part of flavanone glucoside content, the aerial part of volatile oil content, the rhizome part of volatile oil content and the rhizome part of methyl-n-nonyl ketone content, the results implied that the high organic fertilizer employment quantity unbeneficial to the yield of active component of H. cordata Thunb. 5. Using 75% shading with 3000 kg/ha organic fertilizers could achieve the highest flavanone glucoside yield in aerial part and fresh yield, volatile oil yield and methyl-n-nonyl ketone yield in different parts of H. cordata Thunb.

Houttuynia cordata Thunb is a medicinal plant used in traditional Chinese medicine. The major aim of present study was to examine the effects of Houttuynia cordata Thunb aqueous extract (HCE) in mice treated by high fat diet. Butter at 15% (w/w) was used to prepare high fat diet. C57B6 male mice were divided into five groups: (1) normal group (normal diet and water); (2) high dose HCE group (normal diet and 8% of HCE); (3) high fat diet group (15% butter and water); (4) high fat diet with low dose HCE group (15% butter and 4% of HCE) and (5) high fat diet with high dose HCE group (15% butter and 8% of HCE). After eight weeks, blood and organs were collected. Results showed that the intake of HCE reduced body weight, Insulin, HOMA-IR, circulating TG and TC levels, retained cardiac GPx activity and GSH concentration, and decreased cardiac TNF-α and MCP-1 concentrations. These findings support that Houttuynia cordata Thunb through its antioxidant, and anti-inflammatory effects could attenuate the complications caused by high-fat diet.

Herpes simplex virus (HSV), a common latent virus in humans, causes certain severe diseases. Extensive use of acyclovir (ACV) results in the development of drug-resistant HSV strains, hence, there is an urgent need to develop new drugs to treat HSV infection. Houttuynia cordata (H. cordata), a natural herbal medicine, has been reported to exhibit anti-HSV effect which is partly NF-κB-dependent. However, the molecular mechanisms by which H. cordata inhibits HSV infection are not elucidated thoroughly. In this study, we found that H. cordata water extract (HCWE) inhibited the infection of HSV-1, HSV-2, and acyclovir-resistant HSV-1, mainly via blocking viral binding and penetration in the beginning of infection. HCWE also suppressed HSV replication. Furthermore, HCWE attenuated the first-wave of NF-κB activation, which is essential for viral gene expression. Further analysis of six compounds identified in H. cordata revealed that quercetin and isoquercitrin inhibit NF-κB activation and additionally, quercetin also has an inhibitory effect on viral entry. These results indicate that H. cordata could inhibit HSV infection through multiple mechanisms and could be a potential lead for development of new drugs for treating HSV. Recently, the microRNA also can be the antiviral target. MicroRNA is a class of small non-protein-coding RNAs that may act via endogenous RNA interference. However, few studies have explored the role of microRNA in HSV infection. In this study, we used the monocytic cell line THP1 and found that the expression of miR-27a* in THP1 was reduced after HSV-1 infection. Potential target genes of miR-27a* were predicted by the target prediction program, miRanda. One of the predicted targets is paired immunoglobulin-like type 2 receptor α (PILRα), the co-receptor of HSV-1, which contains an immunoreceptor tyrosine-based inhibition motif (ITIM) and can deliver inhibitory signals in immune cells. We hypothesized that HSV-1 infection could down-regulate the expression of miR-27a* and increase the expression of PILRα and then inhibit the antiviral immunity. Further studies showed that overexpression of miR-27a* in cells could significantly inhibit virus replication and also could increase the expression of TNF-α, IFN-β, and IL-6. Similar results were also observed by delivering PILRα-specific siRNA. Collectively, miR-27a* had important role in regulating innate antiviral immunity by inhibiting the expression of PILRα. Further studies will be required to elucidate the role of PILRα in HSV-1 infection.

Summary The twenty-five collections of Houttuynia cordata Thunb. collected from Taiwan were used as materials to investigated the variation of plant parts , crop seasons and growth stages for shoot dry matter, the contents of flavonoid glycosides and total polysaccharides. In addition, the estimators, such as heritability, correlation analysis and the principal component analysis were estimated. The results were summarized as follows: 1. After investigation, the dry matter of rhizome in three tested collections were larger than shoot in one year period except January, February and March. In spring, the dry matter of the shoot and rhizome was at the highest level at flowering and fruiting stage. 2. Among the leaf, stem and rhizome, the contents of rutin, hyperin, isoquercitrin and quercitrin was as follows: leaf > stem > rhizome. And the content in rhizome is almost undetectable. 3. In life cycle, the contents of flavonoid glycosides in the leaf and stem was at the highest level at flowering stage and at the lowest level at dormancy stage. 4. The content of total polysaccharides of H. cordata in germination was higher than in flowering, insignificant difference between seasons. The content of total polysaccharides in leaf and stem reached the highest level in March and August, whereas the rhizomes only in August. 5. The results of combined ANOVA indicate that the treatment of collections, cropping seasons and the interaction of collections and cropping seasons get a significant level in shoot dry matter and the contents of flavonoid glycosides and total polysaccharides of leaf and stem. Among all collections, the dry matter of shoot and the content of total polysaccharides in fall crop were higher than in spring crop. However, the contents of flavonoid glycosides showed inconsistent between spring and fall crop. 6. The broad heritability for most of characters investigated were more than 75 %, and in spring crop higher than in fall crop. 7. According to correlation analysis, in the spring crop, shoot dry matter was negatively correlated with total polysaccharides in leaf, and positively correlated with the flavonoid glycosides in stem. However the relationship between shoot dry matter and the compounds in stem and leaf was not significant correlated in the fall crop. The flavonoid glycosides in leaf, stem and rhizome were positive correlated in the spring and fall crops. The total polysaccharides in stem and leaf had no significant correlation with other compounds in the spring crop, but negative correlated with the flavonoid glycosides in stem in the fall crop. 8. Bye means of the analysis of principal components, the first principal component can represent the characteristics of shoot dry matter and the flavonoid glycosides in stem. The second principal component can represent the characteristics of flavonoid glycosides in leaf. The twenty-five collections could be distinguish into six groups and four groups respectively by the first and second principal components in the spring and fall crops. It showed all of collections had different behavior in the two crops. The results indicate that it is necessary to pay attention to the breeding environment in Taiwan for improving the yield and the contents of flavonoid glycosides and total polysaccharides of H. cordata.

Abstract Taiwanese style sausage is a semi-dry sausage made with the larger particle size of ground pork curing with backfat, high sugar content, rice wine and some spices, which produces a unique aroma after curing. However, the shelf life of this product is a short time due to high sugar content, low temperature and short time for drying and makes it susceptible to bacterial growth. Therefore, the utilization of extracts of Toona sinensis or Houttuynia cordata in Taiwanese style sausage to inhibit bacteria and prolong the shelf life during storage at high refrigerated temperature (15°C) was the purpose of this research. In experiment I, total flavonoids and total phenolics contents of both herb extracts were increased (p<0.05) with the percentage herbs of solutions, T. sinensis extracted with ethanol (Te) had higher total flavonoids and total phenolics contents than that of extracted with water (Tw) and H. cordata extracted with ethanol (Hc). In experiment II, the residual of total phenolics contents, the scavenging capacity of DPPH radical and the chelating Fe2+ ability of all groups were decreased (p<0.05) with heating time and temperature increase, Te had higher the residual of total phenolics contents and better scavenging capacity than Tw and Hc when heated at the same temperature and time. However, Tw had better chelating ability than TE and Hc. In Experiment III, Te showed the best inhibition (p<0.05) on the growth of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Lactobacillus acidophilus, but Tw only had slight inhibition on the growth of S. aureus and L. acidophilus, however, Hc didn’t have antibacterial activity on the growth of all selected bacteria in this study. In experiment IV, the total plate count, anaerobic plate count, lactic acid bacteria count, mold and yeast count and VBN value of all treatments increased (p<0.05) with storage time, the sausage treated with 250 ppm T. sinensis (Te250) and 125 ppm T. sinensis (Te125) showed the lower bacteria count and VBN value during storage and the results were similar to the results of NaL. However, sausage treated with potassium sorbate (KS), 125 ppm H. cordata (Hc125) and 250 ppm H. cordata (Hc250) had higher bacteria count and VBN value. Sausages treated with NaL, Te125 and Te250 had higher pH values than that of Hc125 and Hc250 and KS during storage at 15°C. The L*, a* and b*, color, flavor and overall acceptance of the sensory score of all treatment decreased with storage time but off-odor of all treatments was increased with storage time. In conclusion, extracts of Toona sinensis possessed high total flavonoids and total phenolics contents and also showed high antibacterial ability, especially extract of T. sinensis with ethanol. The application of herbs extracts in Taiwanese style sausage and stored at higher refrigerated temperature (15°C), sausages treated with T. sinensis extracts showed better antibacterial activity than that of Houttuynia cordata and potassium sorbate and the same as sodium lactate in the study. Therefore, extracts of T. sinensis would be suitable to replace sodium lactate or potassium sorbate in Taiwanese style sausage for inhibiting the growth of bacteria in the future.
