A gene conditioning hairy leaf sheath character, which was derived from Hordeum bulbosum and designated Hsb, was mapped using a cross between Hordeum vulgare and a H. vulgare/H. bulbosum recombinant line. The Hsb locus was tagged relative to eight RFLP markers detecting three loci on the distal part of chromosome 4HL. The map position suggests that Hsb of H. bulbosum is homoeologous to the gene Hp1 of rye (Secale cereale), which pleiotropically governs the traits hairy leaf sheath and hairy peduncle. It is proposed that the recombination break point between H. vulgare and H. bulbosum chromosomes occured at a position homoeologous compared with the 4L/5L translocation in Triticeae genomes, and may reflect a hot spot for chromosome breakage.Key words: Hordeum vulgare, Hordeum bulbosum, comparative mapping, hairy leaf sheath, RFLP.

SDS-PAGE was used to analyze the hordein polypeptide patterns of Brazilian barley varieties (Hordeum vulgare L.) and of two native species of Hordeum from southern Brazil (H. euclaston Steud. and H. stenostachys Godr.). Forty different hordein polypeptide bands with molecular weights ranging from 30 to 94 kDa were found in the seeds of the three species studied. Twelve of the 14 varieties examined showed intravarietal polymorphism. The number of bands ranged from 10 to 17, depending on the variety, and from 3 to 13 among individual seeds, with a total of 26 bands in H. vulgare. Phenograms using each seed as an operational taxonomic unit (OTU) showed that the seeds from most varieties did not form distinct clusters. Seeds from different plants of the native species varied considerably. The molecular weights of the hordein polypeptides of the two native species were quite different from those of H. vulgare. There was a greater similarity between the native species than with H. vulgare, although H. stenostachys was slightly closer to the cultivated species than H. euclaston.

Cytokinin activity was determined in roots, crowns, ears of six varieties of the barley in the tobacco Wisconsin 38 callus tissue test. Analysis was made in early stage of ears. The order increasing values of the total cytokinin activity (the total activity of compounds fitting their mobility with zeatin, zeatin ribosid and their more polar derivatives) were for most part of varieties: younger roots < older roots < crowns < ears. Dependences between the cytokinin activity and fresh weight of analysed material were not found.

Rph20 is the only reported, simply inherited gene conferring moderate to high levels of adult plant resistance (APR) to leaf rust ( Puccinia hordei Otth) in barley ( Hordeum vulgare L.). Key parental genotypes were examined to determine the origin of Rph20 in two-rowed barley. The Dutch cultivar ‘Vada’ (released in the 1950s) and parents, ‘Hordeum laevigatum’ and ‘Gull’ (‘Gold’), along with the related cultivar ‘Emir’ (a derivative of ‘Delta’), were assessed for APR to P. hordei in a disease screening nursery. The marker bPb-0837-PCR, co-located with Rph20 on the short arm of chromosome 5H (5HS), was used to screen genotypes for the resistance allele, Rph20.ai. Results from phenotypic assessment and DNA analysis confirmed that Rph20 originated from the landrace ‘H. laevigatum’ (i.e., Hordeum vulgare subsp. vulgare). Tracing back this gene through the pedigrees of two-rowed barley cultivars, indicated that Rph20 has contributed APR to P. hordei for more than 60 years. Although there have been no reports of an Rph20-virulent pathotype, the search for alternative sources of APR should continue to avoid widespread reliance upon a single resistance factor.

Interspecific crosses in Hordeum have been made with the aim of transferring desirable traits, such as disease resistance, from a wild species, Hordeum bulbosum, into cultivated barley (Hordeum vulgare). Interspecific recombinants have previously been identified using several methods, but there are limitations with all the techniques. We improved our ability to characterize progeny from H. vulgare × H. bulbosum crosses by using genomic in situ hybridization (GISH). The plant material comprised a recombinant and a monosomic alien substitution plant derived from H. vulgare × H. bulbosum crosses. The recombinant possesses a pubescent leaf sheath conferred by a gene transferred from H. bulbosum into barley cultivar Golden Promise. The use of GISH on a plant homozygous for the pubescence gene confirmed the presence of H. bulbosum DNA located distally on two barley chromosomes and we mapped the introgression to barley chromosome 4HL using RFLP analysis. Furthermore, by means of an allelism test we found that the transferred gene for pubescence is allelic or closely linked to a gene for pubescence (Hs) located on barley chromosome 4HL. The presence of a single H. bulbosum chromosome in the monosomic substitution plant was confirmed by GISH. A distal introgression of H. bulbosum DNA was also observed on one barley chromosome, which was located on chromosome 3HL by RFLP analysis.Key words: Hordeum vulgare, Hordeum bulbosum, interspecific hybrid, gene introgression, genomic in situ hybridization.

We have identified all Hordeum bulbosum chromosomes in 2 diploid Hordeum vulgare × Hordeum bulbosum hybrids using suitable probes and fluorescence in situ hybridization. Using the parental idiograms allowed us to carry out a full analysis of chromosome associations among all chromosome arms in the hybrids. Association frequencies were generally lower for the short arms than for the long arms. There were also significant differences among the chromosome arms in association frequencies, partly correlated with the absolute length of the chromosome arm, as well as with the frequency of recombinant lines, which were recovered from partially fertile interspecific hybrids. The H. bulbosum idiogram will be useful for further chromosome association studies and will enable the identification of H. bulbosum chromosomes involved in chromosome addition or substitution lines.Key words: Hordeum vulgare, Hordeum bulbosum, interspecific hybrids, chromosome associations, meiosis, fluorescence in situ hybridization.

The fungal and bacterial population of leaf surface of <i>Hordeum vulgare</i> L. has ben described. The phyllosphere and phylloplane regions of green and yellow leaves harboured different dominant species. Bacteria mostly suppressed the fungal growth and possibility of biological control has been suggested. The effect of three amino acids and one organic acid on leaf mycoflora has been studied by foliar spray method. Stimulatory effect of different concentrations of the acids has been noticed. Preferential growth of selected forms of microorganisms may be encouraged by the acid application to act as a mean of biological control.

Southern blots of restriction fragments of genomic DNAs from Hordeum vulgare (L.), H. bulbosum (L.), and interspecific hybrids and their derivatives were hybridized with rDNA probe to identify locus-specific modifications at Rrn loci. H. bulbosum rDNA revealed a single EcoRV site per repeat compared with two sites in H. vulgare rDNA repeats. H. bulbosum accessions possessed at least two rDNA repeat lengths, indicating heterozygosity at the Rrn locus. Hybrids possessed both H. vulgare and H. bulbosum rDNA repeats. Two of the hybrid derivatives possessed bulbosum-specific Sau3AI and HaeIII rDNA fragments, while amphiploid and doubled haploid derivatives lacked H. bulbosum rDNA repeat units and (or) fragments. Two hybrid derivatives, one amphiploid and a doubled haploid derived from the same parental combination, lacked the vulgare Rrn2-specific 9.0-kb rDNA repeat. This is the first conclusive evidence for the elimination of vulgare genetic material in vulgare-bulbosum hybrids. The ratios of 9.0- to 9.9-kb vulgare repeats and H. vulgare to H. bulbosum rDNA repeats indicate partial loss of the vulgare-specific 9.0-kb rDNA repeat among the hybrids. Differences in MboI and Sau3AI fragments and the ratios of 9.0 to 9.9 kb vulgare rDNA repeats revealed differential methylation at Rrn1and Rrn2loci. Hybrids and derivatives showed differential distribution of methylation of EcoRI, BglII, and SacI sites at the Rrn1locus. Two of the hybrid derivatives exhibited extensive CpG-biased methylation. Data presented here are indicative of the differences in the onset of events triggered by the interaction of the component genomes and enabled detection of differential methylation among Rrn loci, loss of H. vulgare genetic material, and development of doubled haploids with the Rrn1locus.Key words: DNA methylation, elimination, Hordeum vulgare, H. bulbosum, Rrn loci.

Crosses between different parental ploidy combinations of barley (Hordeum vulgare L.) and H. bulbosum L. have been carried out principally to obtain hybrids with high allosyndetic chromosome pairing. Fertility has been observed in tetraploid and triploid hybrids, but there has been little evidence of gene introgression from H. bulbosum into H. vulgare in their progeny. To investigate whether crossing-over takes place between homoeologous chromosomes of H. vulgare and H. bulbosum, diploid hybrids were obtained from crosses between a barley mutant (wst3) homozygous for a paracentric inversion on chromosome 3 and one H. bulbosum genotype. Meiotic analyses at metaphase I and anaphase I and II were carried out on several viable hybrids and compared with control diploid hybrids without the inversion and H. vulgare plants heterozygous for the inversion. Aberrations (bridges and (or) fragments) in H. vulgare × H. bulbosum pollen mother cells at anaphase I were significantly less frequent than in the barley hétérozygote. It is concluded that reduced crossing-over between the parental chromosomes in hybrids contributes to the lack of success in obtaining genetic recombinations between the two species.Key words: Hordeum vulgare, Hordeum bulbosum, interspecific hybrid, paracentric inversion, crossing-over.

Barley metaphase chromosomes (2n = 14) can be identified by fluorescence in situ hybridization (FISH) and digital imaging microscopy using heterologous 18S rDNA and 5S rDNA probe sequences. When these sequences are used together, FISH landmark signals were seen so that all 7 chromosomes were uniquely identified and unambiguously oriented. The chromosomal location of the landmark signals was determined by FISH to a barley trisomic series using the 18S and 5S probes labeled with different fluorophores. The utility of these FISH landmarks for barley physical mapping was also demonstrated when an Amy-2 cDNA clone and a BAC clone were hybridized with the FISH landmark probes.Key words: Hordeum vulgare, barley, FISH, 5S, 18S, rDNA, landmarks, chromosome.
