The rotavirus non-structural protein NSP4 has been implicated in a number of biological functions during the rotavirus cellular cycle and pathogenesis, and has been addressed as a target for vaccine development. The NSP4 gene has been classified into six genotypes (A–F). A semi-nested triplex PCR was developed for genotyping the major human NSP4 genotypes (A–C), which are common in human rotavirus strains but are also shared among most mammalian rotavirus strains. A total of 192 previously characterized human strains representing numerous G and P type specificities (such as G1P[8], G1P[4], G2P[4], G3P[3], G3P[8], G3P[9], G4P[6], G4P[8], G6P[4], G6P[9], G6P[14], G8P[10], G8P[14], G9P[8], G9P[11], G10P[11], G12P[6] and G12P[8]) were tested for NSP4 specificity by the collaborating laboratories. An additional 35 animal strains, including the reference laboratory strains SA11 (simian, G3P[2]), NCDV (bovine, G6P[1]), K9 and CU-1 (canine, G3P[3]), together with 31 field isolates (canine, G3P[3]; feline, G3P[9]; porcine, G2P[23], G3P[6], G4P[6], G5P[6], G5P[7], G5P[26], G5P[27], G9P[6] and G9P[7]) were also successfully NSP4-typed. Four human G3P[9] strains and one feline G3P[9] strain were found to possess an NSP4 A genotype, instead of NSP4 C, suggesting a reassortment event between heterologous strains. Routine NSP4 genotyping may help to determine the genomic constellation of rotaviruses of man and livestock, and identify interspecies transmission of heterologous strains.

Gastrin-releasing peptide (GRP) is typically viewed as a growth factor in cancer. However, we have suggested that in colon cancer, GRP acts primarily as a morphogen when it and its receptor (GRP-R) are aberrantly upregulated. As such, GRP/GRP-R act(s) primarily to modulate processes contributing to the assumption or maintenance of tumor differentiation. One of the most important such processes is the ability of tumor cells to achieve directed motility in the context of tissue remodeling. Yet the cellular conditions affecting GRP/GRP-R expression, and the biochemical pathways involved in mediating its morphogenic properties, remain to be established. To study this, we evaluated the human colon cancer cell lines Caco-2 and HT-29 cells. We found that confluent cells do not express GRP/GRP-R. In contrast, disaggreation and plating at subconfluent densities results in rapid GRP/GRP-R upregulation followed by their progressive decrease as confluence is achieved. GRP/GRP-R coexpression correlated with that of focal adhesion kinase (FAK) phosphorylation of Tyr397, Tyr407, Tyr861, and Tyr925 but not Tyr576 or Tyr577. To more specifically evaluate the kinetics of GRP/GRP-R upregulation, we wounded confluent cell monolayers. At t = 0 h GRP/GRP-R were not expressed, yet cells immediately began migrating into the gap created by the wound. GRP/GRP-R were first detected at ∼2 h, and maximal levels were observed at ∼6 h postwounding. The GRP-specific antagonist [d-Phe6]-labeled bombesin methyl ester had no effect on cell motility before GRP-R expression. In contrast, this agent increasingly attenuated cell motility with increasing GRP-R expression such that from t = 6 h onward no further cell migration into the gap was observed. Overall, these findings indicate the existence of GRP-independent and -dependent phases of tumor cell remodeling with the latter mediating colon cancer cell motility during remodeling via FAK.

The article presents the results of the analysis of technological modes` productivity in the economy of the Urals Federal District. The use of evaluation of contribution to per capita GDP industrial (relict and fourth) technological modes and specified data on the gross regional product has allowed to establish that the spread of new postindustrial technological modes in the economy of different regions of the Urals Federal District is uneven. In Kurgan region the postindustrial technological modes are not widespread. The contribution of new modes in per capita GRP in Chelyabinsk region in 2014 reached 115865,55 rubles (29.99% of per capita GRP), in Sverdlovsk region — 251945,45 rubles (48,21% of per capita GRP), in Tyumen region as a whole — 1705575,89 rubles (86,31% of per capita GRP), in Tyumen region except for Khanty-Mansi and Yamal-Nenets Autonomous Areas — 439298,25 rubles (61,88% of per capita GRP). The contribution of new modes in per capita GRP in Khanty-Mansi Autonomous Area in 2014 reached 2124572,44 rubles (88,70% of per capita GRP), in Yamal-Nenets Autonomous Area — 3789418,62 rubles (93,09% of per capita GRP). The system of new technological modes uses 3-19% of the resources of the Urals Federal District`s regions.

The article explores the dynamics of changes in disparities of China’s regions’ economic level. According to a number of researchers, economic disparities between China’s regions started to decrease from about the mid-2000s. To clarify this picture the article uses indicator of contribution of the region’s GRP to the country’s total GRP. As a result of the research carried out by the author, it was found that in the PRC there really was a period when the economic gap between regions was narrowing down, but in general, changes in the relative contribution of regions to the country’s GRP are more complex than just a decrease or increase in economic disparities. Also, based on statistical data, the article draws conclusions about the relationship between the dynamics of the region’s contribution to the total GRP and the shares of the secondary and tertiary sectors in the region’s economy. It turned out that the increase in the economic level of the region in comparison with other regions is closely related to the share of the secondary sector in the GRP of this region: in most regions of China, the increase in the share of the secondary sector occurs in parallel with the growth of the contribution of the region’s GRP to the total GRP of the country. The growth in the share of the tertiary sector, on the contrary, in most cases is accompanied by a relative decrease in the economic level of the region, that is, its contribution to the total GRP decreases.

The effects of gastrin-releasing peptide (GRP), bombesin, GRP-(1–16) and GRP-(21–27) on guinea pig nasal mucosal secretion were studied in vivo. GRP, bombesin, and GRP-(21–27) induced significant secretion of total protein, albumin, and alkaline phosphatase. GRP induced significant secretion at lower concentrations (10(-11) and 10(-10) M) than were required for bombesin and GRP-(21–27) (10(-7) M). GRP-(1–16) did not stimulate secretion, indicating that the COOH-terminal region of GRP contained the secretagogic principle. Capsaicin, a stimulant of nociceptive sensory nerves, stimulated GRP release into nasal secretions. These data suggest that GRP is present in guinea pig nasal mucosa and that the COOH-terminal region of GRP may regulate mucosal macromolecule secretion.

A large strength-to-weight ratio, coupled with electromagnetic transparency and (possibly engineered) sound-dampening properties, render GRP materials as attractive materials for ship decks and deckhouse skins. Unfortunately, these materials, which use a thermosetting resin as a matrix material, cannot be welded as steel can. Thus, if a large structure is to be built of GRP, the inevitable joints must be either adhesively bounded or mechanically fastened. These joining techniques are frequently combined in joints which must support large loads. Because of this, an understanding of the mechanical behavior of joints in GRP is important

Glass reinforced plastic (GRP) is now an established material for the fabrication of sonar windows. Its good mechanical strength, light weight, resistance to corrosion and acoustic transparency, are all properties which fit it for this application. This thesis describes a study, undertaken at the Royal Naval Engineering College, Plymouth, into the mechanical behaviour of a circular cylindrical sonar panel. This particular type of panel would be used to cover a flank array sonar in a ship or submarine. The case considered is that of a panel with all of its edges mechanically clamped and subject to pressure loading on its convex surface. A comprehensive program of testing, to determine the orthotropic elastic properties of the laminated composite panel material is described, together with a series of pressure tests on 1:5 scale sonar panels. These pressure tests were carried out in a purpose designed test rig, using air pressure to provide simulated hydrostatic and hydrodynamic loading. Details of all instrumentation used in the experimental work are given in the thesis. The experimental results from the panel testing are compared with predictions of panel behaviour obtained from both the Galerkin solution of Flugge's cylindrical shell equations (orthotropic case), and finite element modelling of the panels using PAFEC. A variety of appropriate panel boundary conditions are considered in each case. A parametric study, intended to be of use as a preliminary design tool, and based on the above Galerkin solution, is also presented. This parametric study considers cases of boundary conditions, material properties, and panel geometry, outside of those investigated in the experimental work Final conclusions are drawn and recommendations made regarding possible improvements to the procedures for design, manufacture and fixing of sonar panels in the Royal Navy.

The GRP 5000 is a track measuring trolley capable of collecting track geometry data and performing clearance analyses based on laser scanning technology. The obtained laser data can also be used for as-built documentation, and the track recording functionality makes applications such as surveying, tamping assistance and slab track construction possible. The accuracy, huge data amount and time efficiency by which the system operates sets a new standard in railway surveying, and outdates traditional methods of manual and visual inspection. This thesis is a case study of the GRP 5000 with several objectives: a functional and technical description of the system is given; the accuracy of the system is evaluated, showing overall good values except for one of the sensors used; comparisons are made to other railway surveying techniques, such as track recording vehicles, manual devices and other track recording trolleys; possible improvements are pointed out, both based on comparison results as well as testing results. Finally, new and innovative ways of using the trolley, such as offtrack usage, as well as for the obtained laser data, such as GIS, maintenance aspects and CFD aspects, are examined.

Even though Kazakhstan is one of the most successful transition countries in Central Asia it has been neglected in the literature on regional convergence. This paper fills the gap with an empirical analysis of the convergence process on the regional level using annual gross regional product (GRP) data for the period 1998-2008 for the 16 Kazakh regions. Sigma convergence implies that the dispersion of per capita GRP, measured as the standard deviation of per capita GRP across regions, declines over time. Given the growing variation in per capita GRP over time this phenomenon cannot be found for Kazakhstan. In the neoclassical growth model, under the assumption of similar steady states, the growth rate of per capita GRP is negatively related to the initial level of per capita GRP. Surprisingly, we do not find this relation for the Kazakh regions. The data show that there is no evidence for absolute beta convergence. In contrast, the Kazakh regions even seem to diverge.

The utilization of layered heterogeneous material systems in the development of armor provides a potential for a great improvement in ballistic performance in a variety of lightweight armor applications. Some of the notable recent examples demonstrating the success of synthetic heterogeneous material systems include composite materials with organic matrices reinforced by glass fibers to achieve lightweight and enhanced ballistic resistance. In the present study, a series of plate impact experiments were performed on two S2-glass fiber reinforced polymer composites (GRP) with different resin content for their promising potential in future combat vehicle defense system. GRP has excellent strength along its fiberglass directions, however, the cohesion between fiberglass layers and its resin matrix is not strong and spall usually occurs during a typical impact process. The objective of the present study is to investigate: (a) dispersion and attenuation of shock-waves in two GRP composites; (b) the spall (delamination) strength of the two GRP composites both under compression and pressure-shear shock wave conditions. The GRP specimens were shock loaded by utilizing A1 7075-T6 flyer plates to around 2 GPa; the thickness of flyer and target for each experiment was carefully designed to produce a state of tension near the center of the GRP target plates. Normal plate impact, and combined pressure and shear experiments with skew angles ranging from 12o to 20o, were performed to study the effects of normal compression and combined compression and shear on the GRPs' spall strength. The measured spall strength as a function of the applied shear strain and the normal stress was used to develop a 3-dimensional failure surface. The results indicate that the spall strength of GRP decreases with increasing compressive stress; the addition of shear stress was found detrimental to the spall strength of GRP.

Cathodic protection (CP) design of pipelines is generally described in internationally recognized standards and recommended practices. However, not all cases can be solved by conventional CP calculations as recommended approaches and methods may not be fully defined in the standards. For instance, Glass fibre Reinforced Polyester (GRP) covers can be applied for mechanical protection (e.g. from trawl and/or dropped object) of subsea pipelines and structures. These GRP covers will restrict the electrical/ionic current lines between the anode and the steel surface to be cathodically protected. For a spool with in-line components requiring a large current (e.g. painted or bare steel) and with a GRP cover it may be difficult to obtain sufficient cathodic protection when applying simple mathematical calculations. This paper describes how the Finite Element Method (FEM) in COMSOL Multiphysics can be applied to show that the potential is acceptable for such cases.

Abstract In order to convert the “one-off” construction of light alloy fast boats to a more efficient series production, the project of a 25-meter patrol boat, originally built in aluminum, has been completely reviewed, focusing on the structure design in composite material. As one of the most important targets of the new project is to further increase the speed performance of the original unit, attention has been initially focused on the weight reduction, this goal being particularly challenging because of the change in the construction material. This gave the opportunity to review the design parameters as well, starting from the design acceleration value which has been assessed at first by considering the Classification Society Rules and secondly by a detailed seakeeping analysis carried out by using ANSYS AQWA software and validated by experimental towing tanks tests at MARIN.

Good Manufacturing Practices (GMP), a component of Pharmaceutical Quality Systems, is aimed primarily at managing and minimizing the risks inherent in pharmaceutical manufacture to ensure the quality, safety and efficacy of products. Provision of adequate number of personnel with the necessary qualifications/practical experience and their continuous training and evaluation of effectiveness of the training is the responsibility of the manufacturer. (World Health Organization [WHO], 2014; International Organization for Standardization [ISO], 2015). The classroom method of training that has been used for GMP capacity building in the pharmaceutical manufacturing industry in Nigeria over the years, delivered by experts from stringently regulated markets, have not yielded commensurate improvement in the Quality Management Systems (QMS) in the industry. It is necessary and long over-due to explore an alternative training method that has a track record of success in other sectors. A lot of studies carried out on mentoring as a development tool in several fields such as academia, medicine, business, research etc., reported positive outcomes. The aim of this study was to explore mentoring as an alternative GMP training method in the pharmaceutical manufacturing industry in Nigeria. Specifically, the aim of this study was to evaluate the impact of mentoring as a GMP capability building tool in the pharmaceutical manufacturing industry in Nigeria, with focus on GMP documentations in XYZ pharmaceutical manufacturing company located in South-Western region of Nigeria. The methodology comprised gap assessment of GMP documentation of XYZ company to generate current state data, development of training materials based on the identified gaps and use of the training materials for the mentoring sessions. The outcome of the study was outstanding as gap assessment identified the areas of need that enabled development efforts to be targeted at these areas, unlike generic classroom training. The mentees’ acceptance of the mentoring support was evident by their request for additional training in some other areas related to the microbiology operations that were not covered in the gap assessment. This result portrays mentoring as a promising tool for GMP capacity building, but more structured studies need to be conducted in this area to generate results that can be generalized.

Per capita GDP has limited use as a well-being indicator because it does notcapture many dimensions that imply a “good life”, such as health and equality ofopportunity. However, per capita GDP has the virtues of being easy to interpret andto calculate with manageable data requirements. Against this backdrop, there is aneed for a measure of well-being that preserves the advantages of per capita GDP,but also includes health and equality. We propose a new parsimonious indicatorto fill this gap, and calculate it for 149 countries. This new indicator could beparticularly useful in complementing standard well-being indicators during theCOVID-19 pandemic. This is because (i) COVID-19 predominantly affects olderadults beyond their prime working ages whose mortality and morbidity do notstrongly affect GDP, and (ii) COVID-19 is known to have large effects on inequalityin many countries.

The research was a retrospective study of twenty-five Good Manufacturing Practice (GMP) inspection reports (from March 2017 through to December 2018) of a national medicine regulatory agency, drug Inspectorate, in West Africa, designed to assess the inspectors’ expertise in the act of inspection report writing. The investigation examined a paper-based tool of thirteen pre-registration Inspection reports and twelve GMP reassessment reports written prior and following an intervention program by external GMP trainers to enhance inspectors’ skill in pharmaceutical cGMP inspection. The study made use of quantitative analysis to investigate each team’s expertise in the act of writing GMP inspection report. Likewise, each report’s compliance with the requirements of three regulatory standards on GMP inspection report writing was ascertained. Impact of intervention program on lead inspectors’ competence was assessed. Lastly, gap in each team writing effectiveness, and lead inspectors’ abilities to deliver an effective report were determined. The results showed one of the inspection team (4.0%) wrote an excellent report. Two (8.0%) of the twenty-five inspection teams penned good inspection reports. Eleven (44.0%) teams drafted needs improvement reports and the remaining eleven teams (44.0%) prepared unacceptable reports. The excellent report and the two good reports had report format that meet expectation. One (50.0%) of the good reports showed the authors possess excellent knowledge of cGMP technical areas. The remain good report (50.0%) revealed the writers’ knowledge.as good. The excellent report showed the authors displayed partial mastery in the use of objective evidence while the two good reports disclosed theirs as having partial and evolving abilities. One of the teams (50.0%) that wrote good reports displayed good use of third person narrative past tense in report writing whereas the other team used the same tense and voice excellently. Generally, a sort of marginal level of performance was prominent among the inspection teams. A gap, if not tackled, will slow down regulatory process through increase report review, litigations that query report factual accuracy (AIHO, 2017) and delay in issuance of marketing authorization. In conclusion, trainings on quality attributes, such as technical content (Quality Management System (QMS) and Site), the use of objective evidence, assignment of risk levels to GMP violations and citing of applicable laws, regulation and guidelines that substantiate GMP observations, were recommended, to enhance knowledge sharing and regulators’ performance in the act of writing inspection report.
