The biogeographic and phylogenetic relationships of six of the eight Australian genera of freshwater shrimp from the family Atyidae were investigated using mitochondrial 16S rDNA and cytochrome oxidase I sequences. Previous studies on two of the epigean genera (Caridina, Paratya) indicate that Australian species have strong links to congenerics from outside, with Australian members of Paratya being monophyletic and Caridina polyphyletic. The present study found that the endemic Australian epigean genus Australatya forms a strong clade with Pacific ‘Atya-like’ genera (Atyoida, Atyopsis), and that the endemic Australian epigean genus Caridinides falls within a clade containing Caridina species from the Australian ‘indistincta’ group. The two hypogean genera included in this study (Parisia, Pycnisia) form a strong clade in all analyses, implying an Australian subterranean speciation. The possibility of a relationship between Parisia/Pycnisia and an Australian Caridina species may have implications for the monophyly of the highly disjunct genus Parisia (Australia, Madagascar, Philippines). Parisia may descend from local Caridina species and represent convergent morphologies.

Atrachea parvispina (Tschetverikov, 1904), A. cortex (Alpheraky, 1887) and A. caelestina Gyulai & Ronkay, 2001 are transferred from the genus Atrachea Warren, 1911 (Xyleninae, Apameini) to Parvispinia (Xyleninae, Xylenini); three new combinations Parvispinia parvispina (Tschetverikov, 1904) comb. n., P. cortex (Alpheraky, 1887), comb. n. and P. caelestina (Gyulai & Ronkay, 2001), comb. n. are introduced. The lectotypes for Parvispinia parvispina and P. cortex are designated. Generic checklists for Parvispinia and Atrachea are presented.

A new species of squat lobster, Galathea chura sp. nov., is described from deep-waters off Okinawa Island in the Ryukyus,southern Japan, on the basis of a single specimen found on a colony of unidentified octocoral of the genus Parisis Verrill,1864. The new species resembles G. magnifica Haswell, 1882, G. spinosorostris Dana, 1852, and G. tanegashimae Baba,1969, in having scale-like or interrupted arcuate ridges on the gastric region of the carapace and epipods on the first or first and second pereopods only, but it is readily distinguished by the absence of epigastric spines.

Manota ctenophora Matile (New Caledonia), M. maorica Edwards (New Zealand) and M. taedia Matile (New Caledonia) are redescribed. Manota hamulata Colless, previously known from Palau, is redescribed and recorded from Papua New Guinea. Manota pacifica Edwards from Samoa is discussed and compared with the other species of the region. A key to the Melanesian and Oceanian species of Manota is given.

The discovery of the first damselfly Lestes regina Théobald, 1937 from Monteils (Gard, France) supports the identity of late Eocene age of this outcrop with the historical outcrop of Célas, type locality for the type series of this species. Lestes regina is also documented from the late Eocene Isle of Wight basin, confirming the presence of significant contacts between this southern area and the anglo-Parisian lacustrine basin at that time. Nearly all the Eocene and Oligocene fossil Lestes from Western Europe have a particular character, viz. the presence of a supplementary row of cells between the veins MP and CuA. This character is much less frequent in extant Lestes and is still unknown among Neogene representatives of the genus.

The first argument set out in 1877 by the Parisian astronomer Charles André, founder of the Observatoire de Lyon, to justify the establishment of an astronomical institute in Saint-Genis-Laval was the fact that it offered the opportunity to install remote marks along its meridian. André had used this argument three years before to reject another site selected by a local committee. However, at that time remote meridian marks had been abandoned by most astronomers in favour of close ones. We have conducted field research to discover the remains of these marks, as well as to seek evidence – both in the literature and in archives – of their use. This investigation shows that the argument for remote marks was in fact driven by geodetic considerations. The choice of Saint-Genis-Laval was actually made by geodesists of the Dépôt Général de la Guerre and its director, François Perrier, who wanted to institute new observatories in Province as key points in the primary geodetic network of France.

Microsporidia are a group of obligate intracellular eukaryotic parasites that infect a wide variety of species, including humans. Phylogenetic analysis indicates a relationship between the Microsporidia and the Fungi. However, most results are based on the analysis of relatively few genes. DarkHorse analysis involves the transformation of BLAST results into a lineage probability index (LPI) value and allows for the comparison of genes for an entire genome with those of other genomes. Thus, we can see which genes from the microsporidia score most closely based on the LPI with other eukaryotic organisms. In this analysis, we calculated the LPI for each gene from the genomes of 7 Microsporidia, Antonospora locustae, Enterocytozoon bieneusi, Encephalitozoon cuniculi, Encephalitozoon intestinalis, Nosema bombycis, Nosema ceranae, and Nematocida parisii, to analyze the genetic relationships between Microsporidia and other species. It was found that many (91%) genes were most closely correlated with genes from other microsporidial genomes and had the highest mean LPI (0.985), indicating a monophyletic origin of the Microsporidia. In a subsequent analysis, we excluded the other Microsporidia from the analysis to look for relationships before the divergence of Microsporidia, and found that 43% of the microsporidial genes scored highest with fungal genes, and a higher mean LPI was found with Fungi than with other kingdoms, suggesting that Microsporidia is closely related to Fungi at the genomic level. Microsporidial genes were functionally clustered based on the KOG (Eukaryotic COG) database, and the possible lineages for each gene family were discussed in concert with the DarkHorse results.

The article represents research findings as for the species composition identification of the arthropod-pests of the genus Sorbus. The study was conducted in the period from April to October during several years in greenery on the territory of Kyiv and the Kyiv region. Four groups of pests were distinguished: pests of root system (6 species), pests of leaves (18 species), pests of fruits (2 species), pests of sprouts (4 species). Twenty-nine species of arthropods of the hexapods (Insecta) which belong to the 6 numbers of plant groups and 1 species of acariform mites (Acariformes, Arachnida) were found and identified. The most numerous species were arthropod pests, the representatives of the order Coleoptera – 10 species – and Lepidoptera – 8 species. The greatest damage caused by pests at different stages of their biological development was inflicted on the leaf cover of rowan – 60%, and its root system – 20%. Significant damage to rowan plants was caused by pests – polyphagous: Gryllotalpa gryllotalpa L., Psylla mali Schm., Aphis pomi De Green, Rhopalosiphum insertum Wal., Edwardsiana rosae L., Lepidosaphes ulmi L., Operophtera brumata L., Мelolontha melolontha L., Selatosomus aeneus L., Scolytus mali Bech., Phyllobius argentatus L., Agrotis segetum Den.et Schiff., Yponomeuta melinellus L., Simaethis pariana L., Argyresthia conjugella Zell., Pentatrichopus sp. The thorough analysis of damage to various parts of plants was carried out. In most instances, the greatest damage by pests to plants was observed on densely clustered plantations of rowan compared to single and small groups of trees and the presence of plants of the family Rosaceae as an additional forage base. The physiological condition of plants was significantly worsened by the adverse impacts of critical environmental factors. The damage to rowan by pests had a clear mosaic character and depended on various biotic and abiotic environmental factors.

Microsporidia are obligate intracellular parasites that are known to infect most types of animals. Many species of microsporidia can infect multiple related hosts, but it is not known if microsporidia express different genes depending upon which host species is infected or if the host response to infection is specific to each microsporidia species. To address these questions, we took advantage of two species of Nematocida microsporidia, N. parisii and N. ausubeli, that infect two species of Caenorhabditis nematodes, C. elegans and C. briggsae. We performed RNA-seq at several time points for each host infected with either microsporidia species. We observed that Nematocida transcription was largely independent of its host. We also observed that the host transcriptional response was similar when infected with either microsporidia species. Finally, we analyzed if the host response to microsporidia infection was conserved across host species. We observed that although many of the genes upregulated in response to infection are not direct orthologs, the same expanded gene families are upregulated in both Caenorhabditis hosts. Together our results describe the transcriptional interactions of Nematocida infection in Caenorhabditis hosts and demonstrate that these responses are evolutionarily conserved.

The present paper brings together survey data from nine collection sites in Santa Catarina state, southern Brazil. Samples were obtained during about four years in forested, coastal and urban areas. Flies were attracted by fermented banana baits and captured in traps. Among the species of the genus Zygothrica Wiedemann 1830 collected, Zygothrica apopoeyi Burla 1956, Z. bilineata (Williston 1896), Z. dispar (Wiedemann 1830), Z./ lanceolata Burla 1956, Z. nigropleura Grimaldi 1987, and Z. poeyi (Sturtevant 1921) were recorded for the first time in Santa Catarina state and, except for Z. bilineata, also in southern Brazil. Apart from these species, we also collected Zygothrica hypandriata Burla 1956, Z. orbitalis (Sturtevant 1916) (as Z. parilis), Z. prodispar Duda 1925 and Z. vittimaculosa Burla 1956. Except for Z. bilineata and Z. hypandriata, these are the new southernmost records for the geographical distribution of these species. All the collected species were represented by few individuals, probably as a consequence of the collection method, and most of them was captured in winters and springs. Five species were restricted to the more preserved studied site, and most individuals were collected in forest sites. Nevertheless, four species were also captured in urban or xeric environments, indicating a higher tolerance and a wider ecological versatility in this genus as compared to what was previously thought, at least for some species.

Abstract Background The resistance to all aminoglycosides (AGs) conferred by 16S rRNA methyltransferase enzymes (16S-RMTases) is a major public health concern. Objectives To characterize the resistance genotype, its genetic environment and plasmid support, and the phylogenetic relatedness of 16S-RMTase-producing Escherichia coli from France. Methods We screened 137 E. coli isolates resistant to all clinically relevant AGs from nine Parisian hospitals for 16S-RMTases. WGS was performed on clinical isolates with high-level AG resistance (MIC ≥256 mg/L) and their transformants. Results Thirty of the 137 AG-resistant E. coli produced 16S-RMTases: 11 ArmA, 18 RmtB and 1 RmtC. The 16S-RMTase producers were also resistant to third-generation cephalosporins (90% due to a blaCTX-M gene), co-trimoxazole, fluoroquinolones and carbapenems (blaNDM and blaVIM genes) in 97%, 83%, 70% and 10% of cases, respectively. Phylogenomic diversity was high in ArmA producers, with 10 different STs, but a similar genetic environment, with the Tn1548 transposon carried by a plasmid closely related to pCTX-M-3 in 6/11 isolates. Conversely, RmtB producers belonged to 12 STs, the most frequent being ST405 and ST complex (STc) 10 (four and four isolates, respectively). The rmtB gene was carried by IncF plasmids in 10 isolates and was found in different genetic environments. The rmtC gene was carried by the pNDM-US plasmid. Conclusions ArmA and RmtB are the predominant 16S-RMTases in France, but their spread follows two different patterns: (i) dissemination of a conserved genetic support carrying armA in E. coli with high levels of genomic diversity; and (ii) various genetic environments surrounding rmtB in clonally related E. coli.

Morphological and chemotaxonomic characteristics confirmed the affiliation of strain NEAU-7082T to the genus Glycomyces . 16S rRNA gene sequence analysis indicated that strain NEAU-7082T belonged to the genus Glycomyces and was closely related to Glycomyces mayteni JCM 16217T (99.0 % 16S rRNA gene sequence similarity), Glycomyces sambucus DSM 45047T (98.4 %), Glycomyces scopariae DSM 44968T (98.3 %), Glycomyces paridis DSM 102295T (98.1 %), Glycomyces artemisiae NBRC 109773T (98.0 %) and Glycomyces dulcitolivorans DSM 105121T (97.9 %). Phylogenetic analysis using the 16S rRNA gene sequence showed that the strain formed a stable clade with G. mayteni JCM 16217T and clustered with G. sambucus DSM 45047T, G. scopariae DSM 44968T, G. artemisiae NBRC 109773T and G. dulcitolivorans DSM 105121T in the genus Glycomyces . The cell wall contained meso-diaminopimelic acid and the whole-cell hydrolysates were glucose and xylose. The polar lipid profile consisted of diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), glycolipid (GL), phosphatidylinositol mannoside (PIM) and an unidentified lipid (UL). The menaquinones were MK-11(H4), MK-11 and MK-10. Major fatty acids were anteiso-C15 : 0, iso-C16 : 0 and anteiso-C17 : 0. These chemotaxonomic data substantiated the affiliation of strain NEAU-7082T to the genus Glycomyces . The DNA G+C content was 71.3 mol%. A combination of DNA–DNA hybridization results and some phenotypic characteristics demonstrated that strain NEAU-7082T could be distinguished from its closest relatives. Therefore, strain NEAU-7082T is considered to represent a novel species of the genus Glycomyces , for which the name Glycomyces albidus sp. nov. is proposed. The type strain is NEAU-7082T (=CCTCC AA 2019045T=JCM 33458T).

Abstract The present status of freshwater crabs (Decapoda, Brachyura) in Turkey is reviewed in this study, with the purpose of identifying the exact number of species, their conservation status, and their distribution. In previous records, it was reported that Turkey has nine freshwater crab species belonging to the genus Potamon: Potamon bileki Pretzmann, 1971, P. ibericum (Bieberstein, 1809), P. hueceste Pretzmann, 1962, P. persicum Pretzmann, 1962, P. magnum Pretzmann, 1962, P. potamios (Olivier, 1804), P. setiger Rathbun, 1904, P. rhodium Parisi, 1913, and P. mesopotamicum Brandis, Storch & Türkay, 1998. However, there was no detailed information on species distribution in Turkey. This review is intended to update the distribution of species in the country, and it revealed that populations of three more freshwater crab species (P. karpathos, P. fluviatile and P. hippocrate) also occur; thus, there are currently 12 freshwater crab species in Turkey. In addition, this review provides a distribution map and identifies possible threats to each species. This review can serve decision makers in the development of management strategies to better protect the environments of these species that are facing growing anthropogenic impacts.

Rapuzzi P., Parisi F. - Notes on Agapanthia (Synthapsia) kirbyi (Gyllenhal, 1817) with description of a new subspecies and the confirmation of its occurrence in Italy (Coleoptera Cerambycidae Lamiinae). In this paper, we investigated hundreds of specimens within the Agapanthia kirbyi (Gyllenhal, 1817) with the aim of finding relevant discrepancies in the Western and Southeastern populations. Amongst the species, the most diverse characters are body shape and pubescence color; however, based on the different geographical distribution it is possible to classify it into three different subspecies. One of these subspecies is new and is described below as Agapanthia (Synthapsia) kirbyi samai new subspecies. We also consider the characters used to create the Genus Synthapsia by Pesarini & Sabbadini (2004) not sufficient for a genus rank but at least for a subgenus rank (new status) due to the fact that other species close to A. kirbyi (Agapanthia hirsuticornis) shows intermediate characters between Synthapsia and Agapanthia. We also formalize the following new synonym: A. kirbyi ssp. zawadskyi Fairmaire, 1866 (name resurrected, new combination) = A. kirbyi ssp. valandoensis Sláma 2015 (new synonym) on the base of many specimens studied from the type localities of both the Taxon. In addition, the occurrence of A. kirbyi (Gyllenhal, 1817) is ascertained in Italy; in fact, eleven specimens were reported in the Matese Mountains, central-southern Apennines, between the years 2019 and 2021. This extends its known distribution into southern Europe and further confirms the important biogeographical role of this mountainous territory. Vibrio mimicus is an emerging pathogen, mainly associated with contaminated seafood consumption. However, little is known about its evolution, biodiversity, and pathogenic potential. This study analyzes the pan-, core, and accessory genomes of nine V. mimicus strains. The core genome yielded 2424 genes in chromosome I (ChI) and 822 genes in chromosome II (ChII), with an accessory genome comprising an average of 10.9% of the whole genome for ChI and 29% for ChII. Core genome phylogenetic trees were obtained, and V. mimicus ATCC-33654 strain was the closest to the outgroup in both chromosomes. Additionally, a phylogenetic study of eight conserved genes (ftsZ, gapA, gyrB, topA, rpoA, recA, mreB, and pyrH), including Vibrio cholerae, Vibrio parilis, Vibrio metoecus, and Vibrio caribbenthicus, clearly showed clade differentiation. The main virulence genes found in ChI corresponded with type I secretion proteins, extracellular components, flagellar proteins, and potential regulators, while, in ChII, the main categories were type-I secretion proteins, chemotaxis proteins, and antibiotic resistance proteins. The accessory genome was characterized by the presence of mobile elements and toxin encoding genes in both chromosomes. Based on the genome atlas, it was possible to characterize differential regions between strains. The pan-genome of V. mimicus encompassed 3539 genes for ChI and 2355 genes for ChII. These results give us an insight into the virulence and gene content of V. mimicus, as well as constitute the first approach to its diversity.

ABSTRACTIn the community, close contacts between humans and dogs may promote the transfer of extended-spectrum beta-lactamase/plasmidic AmpC cephalosporinase (ESBL/pAmpC) genes. Large-scale prevalence studies on ESBL/pAmpC carriage in dogs are rare, and data on ESBL/pAmpC plasmids are even more limited. Here, a considerable rate of 18.5% ESBL/pAmpC carriers was found among 368 unrelated healthy dogs in Paris, France. This prevalence is much higher than the one found in healthy humans in the same city (6%) but close to that recently reported in dogs in China (24.5%). All isolates were identified asEscherichia coli, except oneSalmonella entericaand oneKlebsiella pneumoniaeisolate. The sequence type 131 (ST131) clone was rare (2/73 isolates). Interestingly, two plasmids (blaCTX-M-1/IncI1/ST3 andblaCMY-2/IncI1/ST2) were unexpectedly highly predominant, raising the question of their successful spread. Considering that CTX-M-1 was recently found to be equally as abundant as CTX-M-15 in healthy Parisian subjects, the question of dogs being a CTX-M-1 reservoir for humans is open. Such a high prevalence of theblaCMY-2/IncI1/ST2 plasmid may result from the use of cephalexin in veterinary medicine, as previously demonstrated experimentally. In all, our study points out healthy urban dogs as a potential source of ESBL/pAmpC genes that can further disseminate to the human community.

It is well established that race-specific resistance selects for the specifically matching genes effecting virulence in the pathogen. As well, the use of different R-genes in time makes virulence complexity, VC (the number of virulences per pathogen genotype), to increase. Moreover, it becomes obvious here that the wind-dispersed cereal rusts and mildews are obligate nomads and highly mobile. As a consequence of these points and as a result of modelling, VC was expected to increase in the direction of predominant winds, e.g. from west to east across Europe, and it did by approximately one or more virulences per 1000 km. Starting from the barley mildew pathogen, comprehensive evidence is also available from leaf rust on wheat covering some 5000 km from Western Europe into Siberia. The impact of our findings for population genetics across Europe and Asia is supposed to be considerable and worth further elucidating, e.g., within the 6<sup>th</sup> EU Framework Programme.

Atopida is a New Zealand endemic genus distributed in both North and South Islands. Remarkable sexual dimorphism of most species was a source of numerous misinterpretations in studies by Thomas Broun—males and females of the same species were often described as distinct taxa. After examining type material of all species and over 600 additional unidentified specimens from several institutions, the following new synonyms were established: Atopida grahami Broun, 1910 is a junior synonym of A. hirta Broun, 1880; A. pallidula Broun, 1921 is a junior synonym of A. impressa Broun, 1914; A. basalis Broun, 1912 and A. sinuata Broun, 1893 are junior synonyms of A. proba Sharp, 1878; A. lawsoni Sharp, 1878 is a junior synonym of A. suturalis (White, 1846). The following eight species are newly described: Atopida insularis sp. nov., A. lescheni sp. nov., A. paparoa sp. nov., A. paringa sp. nov., A. tuhua sp. nov., A. waipoua sp. nov., A. walkerae sp. nov., and A. westlandensis sp. nov.

ABSTRACT Eleven Klebsiella pneumoniae clinical isolates and one Klebsiella oxytoca clinical isolate showing various pulsed-field gel electrophoresis types and producing an inducible DHA-1 class C β-lactamase were isolated in the Parisian region between 1998 and 2003. The aim of this study was to compare the genetic organization of the bla DHA-1 genes in this collection of clinical isolates. In four isolates, the Morganella morganii-derived genomic region containing bla DHA-1 was inserted in an entire complex sul1-type integron, including a region common to In6-In7 (CR1), as previously described in a bla DHA-1-producing Salmonella enterica serovar Enteritidis KF92 isolate from Saudi Arabia in 1992. Different gene cassette arrays were characterized in each of these integrons. In two of them, an additional 10-kb fragment was inserted between the CR1 and the M. morganii-derived region and was similar to the sap (ABC transporter family) and psp (phage shock protein) operons originated from Salmonella enterica serovar Typhimurium. The length of the M. morganii region was variable, suggesting that several independent recombination events have occurred and that open reading frame orf513 encodes a recombinase involved in the mobilization of the resistance genes. The genetic organization of bla DHA-1 was identical in the eight other isolates. This structure is likely derived from a complex integron following the insertion of IS26, leading to the deletion of the first part of integron. The horizontal transfer of one plasmid carrying that truncated integron was shown for seven of these isolates.

Nos últimos anos a equideocultura deu um salto qualitativo, havendo hoje, no mercado, animais de alto valor e geneticamente superiores. Isso é possível, em grande parte, devido aos avanços na área da genética animal, os quais permitem identificar, não apenas anomalias, mas também diversos genes de interesse econômico. Com o auxílio da citogenética pode-se identificar indivíduos com alterações no número ou na estrutura dos cromossomos, o que em muitos casos afeta principalmente a reprodução. Também a confirmação de genealogia, anteriormente feita por tipagem sangüínea e atualmente por testes de DNA, tem papel extremamente importante, não apenas por garantir a ascendência dos animais, mas também porque um pedigree confiável pode permitir ao criador identificar a origem de problemas genéticos em seu rebanho e reduzi-los ou mesmo, eliminá-los. Ainda com as ferramentas da biologia molecular, podemos hoje, identificar indivíduos que apresentam genes desejáveis ou indesejáveis, o que nos permite selecioná-los precocemente, reduzindo assim, os custos do produtor e aumentando o valor agregado dos animais. Entre tais genes podemos destacar os que identificam portadores ou afetados por mutações genéticas indesejáveis como: SCID (Síndrome da Imunodeficiência Combinada), HYPP (Parilisa Hipercalêmica), HERDA (Astenia Dérmica Regional Hereditária Eqüina), etc. Também a identificação dos genes que determinam a cor ou padrão da pelagem já pode ser feita direta ou indiretamente (por meio de marcadores genéticos), como é o caso dos genes para as pelagens Overo, Tobiano, etc. Com os avanços no estudo do genoma eqüino muito mais estará disponível em breve, o que certamente só trará maiores contribuições à equideocultura mundial.

A novel actinobacterium, designated strain H14505T, was isolated from a soil sample collected in Hong Yuan, Sichuan, southwest PR China. The temperature, pH and NaCl ranges for growth were determined to be 15–35 °C (optimum, 28 °C), 6.0–8.0 (optimum, pH 7.0) and 0–2 % (w/v; optimum without NaCl), respectively. The polar lipdis detected for strain H14505T were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, glycolipid and four unidentified lipids. The predominant menaquinones of strain H14505T were MK-9(H4) and MK-9(H6), and the prevalent fatty acids (>10 %) were C18 : 1 ω9c, C17 : 1 ω8c, summed feature 5 (anteiso-C18 : 0/ C18 : 2 ω6,9c) and C16 : 0. Phylogenetic analysis based on 16S rRNA gene and whole-genome sequences indicated that strain H14505T showed high similarity to Catellatospora vulcania NEAU-JM1T (99.0 %) and Catellatospora paridis NEAU-CL2T (99.0 %), and formed a monophyletic clade within the the genus Catellatospora in the phylogenetic trees. However, the average nucleotide indentity and DNA–DNA hybridization values between strain H14505T and closely related Catellatospora species showed that it belonged to a distinct species. Furthermore, the results of morphological, physiological and biochemical tests allowed further phenotypic differentiation of strain H14505T from its closest relatives. Thus, it is proposed that strain H14505T represents a novel species of the genus Catellatospora , for which the name Catellatospora sichuanensis sp. nov. is proposed. The type strain of Catellatospora sichuanensis is H14505T (=JCM 32394T=CICC 11042T).

Regulation of immunity throughout an organism is critical for host defense. Previous studies in the nematode Caenorhabditis elegans have described an “ON/OFF” immune switch comprised of the antagonistic paralogs PALS-25 and PALS-22, which regulate resistance against intestinal and epidermal pathogens. Here, we identify and characterize a PALS-25 gain-of-function mutant protein with a premature stop (Q293*), which we find is freed from physical repression by its negative regulator, the PALS-22 protein. PALS-25(Q293*) activates two related gene expression programs, the Oomycete Recognition Response (ORR) against natural pathogens of the epidermis, and the Intracellular Pathogen Response (IPR) against natural intracellular pathogens of the intestine. A subset of ORR/IPR genes is upregulated in pals-25(Q293*) mutants, and they are resistant to oomycete infection in the epidermis, and microsporidia and virus infection in the intestine, but without compromising growth. Surprisingly, we find that activation of PALS-25 seems to primarily stimulate the downstream bZIP transcription factor ZIP-1 in the epidermis, with upregulation of gene expression in both the epidermis and in the intestine. Interestingly, we find that PALS-22/25-regulated epidermal-to-intestinal signaling promotes resistance to the N. parisii intestinal pathogen, demonstrating cross-tissue protective immune induction from one epithelial tissue to another in C. elegans.

A Gram-reaction-positive, endospore-forming and rod-shaped bacterial strain, designated py1325T, was isolated from the root of Paris polyphylla Smith var. yunnanensis collected from Yunnan Province, PR China, and subjected to a polyphasic taxonomic characterization. It grew optimally with 0–1 % NaCl (w/v), at pH 7 and at 30 °C. The major respiratory quinone was MK-7 and the diagnostic diamino acid in the cell-wall peptidoglycan was meso-diaminopimelic acid. The major cellular fatty acid was anteiso-C15 : 0. The polar lipids consisted of phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine, two unidentified aminophospholipids, two unidentified phospholipids and two unidentified lipids. The results of 16S rRNA gene sequence analysis revealed the highest levels of sequence similarity with respect to Paenibacillus luteus R-3T (99.0 %), Paenibacillus sinopodophylli CCTCC AB 2016047T (97.9 %), Paenibacillus castaneae DSM 19417T (97.5 %) and Paenibacillus endophyticus LMG 27297T (97.2 %). The digital DNA–DNA hybridization and average nucleotide identity values between py1325T and these species ranged 20.6–53.3 % and 79.9–93.6 %. The G+C content of the genomic DNA was 47.7 mol%. According to the phylogenetic, phenotypic and chemotaxonomic evidence, strain py1325T clearly represents a novel species of the genus Paenibacillus , for which the name Paenibacillus paridis sp. nov. is proposed. The type strain is py1325T (=CCTCC AB 2015220T=LMG 29068T).

Combining operatic and film musical elements with those of pop videos, contemporary cinema and the rave scene, Baz Luhrmann's film engages with many of the thorny issues that have concerned opera critics of late, such as power, gender, exoticism, authorship, and identity construction and performance. The spotlight on the central love triangle of a consumptive courtesan, a writer and a wealthy patron makes possible a deeper scrutiny of traditional gender roles in the production and reception of Western art. The film's formulaic plot and the backstage musical format render transparent the commercial impetus behind the creative process and demystify the role of the Romantic artist-genius. Finally, the transnational and transhistorical elements of the film – a mostly Australian production team and crew, American and British pop songs, a Parisian backdrop, the Bollywood-inspired show-within-a-show, numerous anachronisms that refuse to stay confined within the specified time setting of the late nineteenth century – disrupt the Classical ideals of artistic unity and integrity and suggest new postmodern geographies and temporalities. This article considers how Luhrmann, by simultaneously paying homage to and critiquing operatic practices in Moulin Rouge!, deconstructs and reinvents opera for the postmodern age.

In the Emergentist perspective, interactions occurring at all levels, from genes to environment, give rise to emergent forms and behavior. These outcomes may be highly constrained and universal, but they are not themselves directly contained in the genes in any domain-specific way. The human body contains perhaps 5 × 1028 bits of information in its molecular arrangement, but our genome contains only about 105 bits of information. Thus, we are over 20 orders of magnitude short of being mosaic organisms, where development is prespecified in the genes. Our development is under regulatory control, where precise pathways to adulthood reflect numerous interactions at the cellular level occurring throughout development. The human cortex is plastic, its architecture reflects experience; innate specification of synaptic connectivity in the cortex is highly unlikely. Theories of language must reflect this—they must be biologically, developmentally, and ecologically plausible. Linguistic representational nativism is just not tenable. It is so implausible that UG could be directly encoded in the genotype that we must explore the alternatives. So the answer is not “Nature.” Nor, as the authors so clearly argue, is it “Nature or Nuture.” Rather, it is “Nature and Nurture.”

The taxonomy of the New Zealand weevil genus Irenimus Pascoe, 1876 is revised, resulting in a narrower concept of the genus than has been considered in recent decades. In total, the genus now contains only seven species. In addition to the type species, I. parilis Pascoe, 1876, the genus contains I. duplex (Broun, 1904) and five newly described species: I. aniptus new species (type locality, Oamaru, DN), I. crinitus new species (type locality, Hakataramea Valley, SC), I. minimus new species (type locality, Alexandra, CO), I. stichus new species (type locality, Tekapo, MK) and I. thoracicus new species (type locality, Oamaru, DN). The genus Chalepistes new genus is established to contain the majority of species previously described in the genus Catoptes Schönherr, 1842, but also including species described in Brachyolus White, 1846; Irenimus Pascoe, 1876; Inophloeus Pascoe, 1875; and Nicaeana Pascoe, 1877. Abstract OP-1250 is an orally-bioavailable complete estrogen receptor antagonist (CERAN) previously shown to shrink wild-type and Y537S mutant estrogen receptor-positive (ER+) tumors in multiple preclinical xenograft models. To better understand the effects of OP-1250 treatment on ER+ breast cancer cells, we used precision run-on sequencing (PRO-seq) to profile the direct transcriptional effects of OP-1250 in two commonly used breast cancer cell lines, MCF7 and CAMA-1. PRO-seq detects nascent RNA, and thus can detect changes at shorter timescales than traditional RNA-seq methods. We conducted PRO-seq 15 minutes, 1 hour, 6 hours and 24 hours after administration of OP-1250, estradiol, fulvestrant and tamoxifen to compare the effects of these compounds across the two cell lines. OP-1250 was administered in the absence of estradiol and after estradiol pretreatment to generate a detailed profile of the time course of its activity. We identified genes that display early transcriptional changes in response to the compounds tested, many of which have been previously identified as estrogen response genes. At later time points we observed transcriptional changes consistent with modulation of pathways involved in cell cycle progression. We show that OP-1250 treatment reverses the estradiol-induced transcriptional changes associated with estrogen receptor activation and chronicle the time course of these changes. Finally, we show greater similarity in the transcriptional changes induced by CERANs OP-1250 and fulvestrant compared to tamoxifen, as well as cell line-specific differences in estrogen response and response to anti-estrogen treatment. These data provide additional insights into the mechanisms of OP-1250 action on ER+ cell lines and patient tumors. In spring 1834, when he started writing The Girl with the Golden Eyes, Honoré de Balzac was imbued with by Eugène Delacroix’s work and figure. Both shared an attraction towards the Orient which betrayed their tendency to romanticism. In 1834, the exhibition of the painting The women of Algiers in their Apartment encouraged a new impetus of the novelist’s oriental genius. From 1830, Balzac started to fulfill a dream: competing with Delacroix’s art by portraying the inside of a harem in the short story The Girl with the Golden Eyes, which he dedicated to the romantic painter. The hero, Henri de Marsay, conquers Paquita Valdès who is locked up in a luxurious Parisian hotel by her protector and lover, the Marquise of San Real. Paquita Valdès’s fatal passion for the Count unfolds against the backdrop of an oriental-like boudoir where Delacroix’s cherished colours prevail: yellow and red. This duo of colours, which sustains the unity of the twofold short story, prompted the following comment by Albert Béguin, a literary critic: “Balzac took up the challenge of competing with visual arts and expressing through linguistic means what painters usually say through the play with colours.” Hence the problematic question which motivates our study: How does Honoré de Balzac reproduce the romantic painter’s shape and colour language? We will first examine how the inordinate ambition to see everything and to have everything seen is expressed through Balzac’s art of composition, which he borrowed from Delacroix. The consecutive analyses of open and enclosed spaces will highlight Balzac’s treasured aesthetic law: the law of contrasts. The article will then show the aesthetic impact of the bodies and their stances upon the reader-viewer. Finally, the last part will focus on the suggestive potential of colour and motions in The Girl with the Golden Eyes.

Rhizoma paridis is a traditional Chinese medicine that has been reported to have anti-cancer activity. However, the antitumor effect of pennogenin 3-O-β-chacotrioside (Formular: C45H72O17), a steroidal saponin isolated from its extract, in lung cancer have not been well studied, its potential mechanisms remain unclear. In this paper, its effect on the cell viability of non-small cell lung cancer (NSCLC) was examined using the Cell Counting Kit-8 assay, the potential target proteins were screened by antibody microarrays, the potential pharmacological mechanisms were predicted using Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) and protein–protein interaction (PPI) analyses. Moreover, to further validate the findings, cell apoptosis and cell cycle arrest were measured by flow cytometry, cell invasion and migration were measured by Transwell assays, and apoptosis-related protein expression levels were measured using western blotting. The results showed that, after treatment by pennogenin 3-O-β-chacotrioside, the cell viability was significantly reduced. In addition, thirteen target proteins related to apoptosis and platinum drug resistance were screened out, and their PPIs were constructed. Moreover, it was confirmed to have effects in inducing apoptosis and promoting cell cycle arrest in NSCLC cells. The expression levels of apoptosis-related proteins, such as caspase 8, caspase 9, phospho-ERK1/2, phospho-p38, phospho-Akt, were decreased following the treatment, whereas the expression levels of cleaved caspase 3 and cytochrome C were increased. The findings of the present study highlight the potential of pennogenin 3-O-β-chacotrioside as a novel therapeutic agent.

Abstract Introduction: With a five-year survival rate &lt;10%, Pancreatic Ductal Adenocarcinoma (PDAC) is the 3rd leading cause of cancer-related deaths in North America. Over 90% of PDAC patients harbor a KRAS mutation, the most common form being KRASG12D. However, without additional genetic mutations or environmental events, such as chronic pancreatitis, KRAS mutations do not lead to PDAC. Our laboratory showed Activating Transcription Factor 3 (ATF3) is required for loss of the acinar cell phenotype in response to experimentally induced pancreatitis and for KRASG12D-driven progression to advanced PanIN lesions. However, the mechanism(s) by which ATF3 affects PDAC progression are unknown. The goal of this work is to determine the transcriptional mechanisms by which ATF3 contributes to PDAC progression. We have previously shown ATF3 affects histone acetylation during pancreatic injury and hypothesize that ATF3 affects transcription following KRASG12D activation by altering histone acetylation program and gene expression. Methods: C57Bl/6l mice with acinar-inducible KRASG12D combined with (Ptf1acrertERTKrasLSL-KRASG12D; PK) or without (Ptf1acrertERTKrasLSL-KRASG12DAtf3-/-; APK) ATF3 deletion were gavage with tamoxifen for 5 consecutive days and sacrificed 22 days after initial tamoxifen treatment. Acinar cells were isolated using a standard collagenase protocol, and RNA and chromatin were obtained for RNA-seq or ChIP-seq (for acetylated histone 3 (H3K27ac)). The sequenced datasets were quality checked with FastQC, aligned to the mm10 genome using STAR (for RNA-seq) or Bowtie2 (for ChIP-seq). Differential expression and differential binding analyses were performed using DESeq2 (for RNA-seq) or DiffBind (for ChIP-seq). H3K27ac enrichment patterns were compared to RNA expression profiles in PK and APK cells to identify potential genes/pathways that ATF3 works through to affect acetylation and gene expression in KrasG12D activated mice. Results: Preliminary data indicates that absence of ATF3 alters the pathway activated by KRASG12D and differentially enriches pathways that are directly linked to KRAS signaling. Moreover, comparing PK to wildtype acini, H3K27ac ChIP-seq shows a shift in acetylation patterns at the transcription start site. This change in acetylation patterns in response to KRASG12D is not observed to the same extent with deletion of ATF3. Conclusions: This work indicates that significant H3K27 acetylation occurs in response to KRASG12D activation, even in the absence of significant morphological changes, and that ATF3 seems to alter KRASG12D’s ability to affect changes in histone acetylation patterns. Epigenetic changes appear to mirror changes in the transcriptome but provide more information. Future studies will investigate the epigenetic profiles in pancreatic tumor samples and the potential for HDAC inhibitors as a possible therapeutic target for PDAC patients. Abstract Chimeric antigen receptor (CAR) T cell immunotherapy is FDA approved for the treatment of a subset of B cell malignancies but has shown limited clinical success in solid tumor therapy. T cell exhaustion is an important factor involved in treatment failure, and can, in part, result from continuous stimulation of the CAR by tumor cells expressing the cognate antigen. To gain deeper understanding of CAR T cell exhaustion induced by chronic antigen exposure (CAE), we developed and validated a robust in vitro model, in which mesothelin-redirected CAR T cells (M5CAR) were continuously stimulated with mesothelin-expressing AsPC-1 pancreatic tumor cells such that tumor cells were never cleared, and we characterized these CAE CAR T cells by gene expression at population and single-cell levels, and by epigenetic analyses. CAE M5CAR T cells recapitulate the hallmarks of T cell exhaustion, including reduced proliferation, down-modulation of surface CAR, decreased cytokine production, and reduced cytotoxicity. In addition, CAR T cells undergoing CAE have a transcriptional signature and an epigenetic landscape consistent with exhaustion. Further, transcriptomic analysis revealed that CAE M5CAR T cells undergo a transition from T cells to a post-thymic NK-like T cell phenotype. This plasticity was confirmed by TCR lineage tracing and was also detected in CD19 CAR T cells analyzed from post-infusion blood from DLBCL patients and in M5CAR T cells infiltrating relapsed tumors derived from a xenograft NSG/AsPC-1 mouse model. The dysfunctional signature and the NK phenotype were further detected in vivo by transcriptomic analysis in Ly95 TCR-specific TILs infiltrating NY-ESO-1 tumors. Among the genes included in the dysfunctional signature, the transcription factors SOX4 and ID3 were identified as potential regulators of dysfunction by differential gene expression and pathway analyses. To determine their role in the establishment of a dysfunctional phenotype, we generated ID3KO.M5 and SOX4KO.M5 CAR T cells using CRISPR technology in primary human lymphocytes. The cytotoxic potential of the M5 CAR T cell product generated was not modified by the genetic disruption of the transcription factors. However, when CAR T cells were challenged with chronic antigen stimulation, KO CAR T cells showed a significant reduction of the dysfunctional signature and the NK-like T cell gene expression. Importantly, CAE KO CAR T cells exhibit improved cytotoxicity as compared to Mock.M5CAR T cells. In summary, we have developed a robust in vitro model that recapitulates the hallmarks of T cell exhaustion and that facilitated the identification of a gene signature defining CAR dysregulation, a T-to-NK-like-T cell transition as a novel feature of CAR T cell dysfunction and the transcription factors SOX4 and ID3 as key regulators of CAR T cell exhaustion. Paridis Rhizoma is a natural medicine with strong anti-tumor and anti-inflammatory activities. Our previous research have found that Fusarium sp. C39, an endophytic fungus isolated from Dioscorea nipponica which contains the similar chemical components, significantly increased the steroidal saponins content of Paridis Rhizoma by fermentation. In this study, the inhibitory effects of fermentated Paridis Rhizoma extract (PRE) on liver cancer cells (Hepal-6), cervical cancer cells (Hela), and lung cancer cells (A549) were determined to be stronger than that of the unfermented extract. For discovering the fermentation mechanism of PRE with Fusarium sp. C39, 36 components with obviously quantitative variations were screened out by UPLC-Q/TOF-MS and 53 key genes involved in the metabolic pathways of steroidal saponins were identified by transcriptome. On the basis of comprehensively analyzing information from the metabonomics and transcriptome, it can be speculated that the increase of spirostanol saponins and nuatigenin-type saponins enhanced the inhibitory effect of fermented PRE on cancer cell proliferation. Under the action of glycosidase, glycosyltransferase, oxidoreductases, and genes involved in sterol synthesis, strain C39 achieved the synthesis of diosgenin and the alteration of configurations, sugar chain and substituent of steroidal saponins. The research suggested a microbial transformation approach to increase the resource utilization and activity of Paris polyphylla.

The new species is characterized by having the leaves clustered at the apex of the leafy culm, fimbriae few to absent, dimorphic flowering culms, large ligules (2.5–4 mm long), and pubescent to hirsute synflorescences. We present a morphological description, illustrations of the new species, the conservation status assessment, and a key to differentiate P. caxiuanensis from its morphologically related congeners.

This paper is a floristic and taxonomic treatment of the native bamboos from the Serra do Cachimbo, Eastern Amazon, Brazil. Two collection expeditions were carried out in 2019 and 2021, in addition to the analysis of materials from herbaria. Ten genera and 17 species were found. Descriptions, identification keys, distribution and habitat data, taxonomic comments, and photographs are provided, along with illustrations and conservation status assessment for two new species. The most speciose genus was Olyra (four species), followed by Raddiella (three), Arthrostylidium (two), and Pariana (two); Actinocladum, Eremocaulon, Guadua, Merostachys, Parodiolyra, and Taquara were represented by one species each. Four species are here recorded for the first time for the states of Pará (Arthrostylidium simpliciusculum and Eremocaulon capitatum) and Mato Grosso (Pariana radiciflora and P. zingiberina). We also designate a lectotype for the basionym of E. capitatum and describe two new taxa to science: Arthrostylidium cachimboense and Merostachys cachimboensis.

Abstract Only four freshwater shrimp species from the genus MacrobrachiumSpence Bate, 1868 are known previously from Hong Kong, namely M. formosenseSpence Bate, 1868, M. hainanense (Parisi, 1919), M. meridionaleLiang & Yan, 1983, and M. nipponense (De Haan, 1849). This is likely an underestimate as a result of insufficient sampling, given that recent efforts revealed several new records in their counterpart family Atyidae, and the presence of many more Macrobrachium species from the region (i.e., Guangdong Province, China). We report the occurrence of three more species in Hong Kong: M. equidens (Dana, 1852), M. lar (Fabricius, 1798), M. venustum (Parisi, 1919), and a new species being described, M. lantausp. nov. The new species is erected from the highly variable M. equidens species complex, distinctive in live colouration and genetics. Decisive morphological differences from M. equidens sensu lato could not be confidently elucidated until the species complex is fully resolved. Molecular analysis shows that ‘M. hainanense’ reported from Hong Kong actually refers to M. laevisZheng, Chen & Guo, 2019. Morphological (and molecular) evidence suggest that M. inflatumLiang & Yan, 1985 and M. superbum (Heller, 1862) are possibly morphotypes and thus synonyms of M. nipponense. Macrobrachium hainanense and M. heterorhynchosGuo & He, 2008 are also likely synonyms of M. formosense and M. equidens, respectively. These species are, nevertheless, maintained as valid until examination of the type specimens concurs. The importance of an integrative approach and the associated matters needing attention in systematic studies of Macrobrachium are discussed.

A Gram-stain-positive, mycelium-forming actinobacterium, YIM 121974T was isolated from an extreme arid soil sample collected at Yuanmou Earth Forest, Yunnan Province, PR China. Classification using a polyphasic approach suggested that strain YIM 121974T belonged to the genus Glycomyces and was closely related to Glycomyces dulcitolivorans SJ-25T (98.3 %), Glycomyces scopariae YIM 56256T (98 %), Glycomyces mayteni YIM 61331T (97.9 %), Glycomyces albidus NEAU-7082T (97.9 %), Glycomyces sambucus CGMCC 4.3147T (97.7 %), Glycomyces artemisiae IXS4T (97.6 %) and Glycomyces parisis CPCC 204357T (97.5 %), but could be distinguished from its closest relatives by a combination of phenotypic and phylogenetic features. Average nucleotide identity values of YIM 121974T to its closest phylogenetic neighbours were 70.7–88.9 %, which are lower than the threshold of 95 %. The digital DNA–DNA hybridization values between YIM 121974T and these relative species were 18.0–36.3 %, which are also well below the cut-off value (>70 %) for species delineation. The DNA G+C content of strain YIM 121974T was 72.3 mol% (draft genome sequence). The predominant menaquinone was MK-11. The phospholipids were composed of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside, phosphoglycolipid and glycolipid. The major fatty acid compositions were iso-C16 : 0, anteiso-C15 : 0 and anteiso-C17 : 0. The draft genome of isolate YIM 121974T was found to contain 11 secondary metabolite biosynthesis gene clusters by using the antiSMASH server. Based on the above observations, strain YIM 121974T could be distinguished from closely related species belonging to the genus Glycomyces . Thus, strain YIM 121974T represents a novel species of the genus Glycomyces , for which the name Glycomyces terrestris sp. nov. is proposed. The type strain is YIM 121974T (=KCTC 39870T=DSM 106742T).

Background: Non-small cell lung cancer (NSCLC) is the most common type of lung cancer. Rhizoma paridis saponins (RPS), the main bioactive ingredients of Paris polyphylla Smith var. yunnanensis (PPY), have been proved to have remarkable effects on NSCLC cell lines. However, the multi-component synergistic effects and mechanisms of RPS on NSCLC have not been elucidated. Objective: To decipher the multi-RPS synergistic effects and mechanisms against NSCLC based on network pharmacology combined with segmented solid phase extraction (SPE) and bioactivity screening method. Methods: Firstly, segmented SPE and cytotoxicity assays were performed for the screening of the RPS-enrichment fraction of PPY, and the steroidal saponins in it were identified by LC-MS/MS. Then, network pharmacology analysis was performed to predict the potential therapeutic targets of RPS on NSCLC. Finally, viable cell counting tests and RT-qPCR were utilized to verify the synergistic effects and mechanisms of RPS. Results: 48 potentially active compounds were identified from the 30% MeOH/EtOAc fraction of PPY (30% M/E PPY). The results of the network pharmacology analysis indicated that RPS exerted joint effects by regulating six key targets in PI3K-AKT signaling pathway. In vitro experiments showed that due to the synergistic effects, 30% M/E PPY at 13.90 μg/mL could exert a stronger inhibitory activity on A549 cells by reducing the overexpression of six hub genes compared with the parallel control groups. Conclusion: This research elaborated the multi-RPS synergy mechanisms against NSCLC and provide the way out to develop new combination medicines for NSCLC.

Murray Forman’s text The ‘Hood Comes First: Race, Space, and Place in Rap and Hip-Hop provides insightful commentary on the workings of and relationship between place and space. To highlight the difference of scale between these two parameters, he writes that, “place defines the immediate locale of human interaction in the particular, whereas space is the expanse of mobile trajectories through which subjects pass in their circulation between or among distinct and varied places” (25). This statement reflects Doreen Massey’s earlier observation from her book Space, Place, and Gender that “one view of a place is as a particular articulation” of the spatial (5). These descriptions clarify how human action shapes, and is shaped by, what Forman describes as the “more narrowly circumscribed parameters” of place (25) and the broader realm of space. Clearly, these two terms describe interconnected components that are socially constructed and dynamic: that is, they operate at different scales but are constructed in time, constantly reshaped by human action and perception. “Space and time are inextricably interwoven,” states Massey. She continues: “It is not that the interrelations between objects occur in space and time; it is these relationships themselves which create/define space and time” (261). If place and space represent different scales of social interaction and space and time are interconnected, place and time must be linked as well.While this indicates that human experience and representation operate on different scales, it is important to note that these two factors are also interrelated. As Stuart Hall writes, “[I]t is only through the way in which we represent and imagine ourselves that we come to know how we are constituted and who we are” (473). There is no objective experience, only that which is subjectively represented through various means. Through depictions of these relationships between place, space, and time, rap music shapes listeners’ comprehension of these parameters. DJs, MCs, producers, and other creative artists express personal observations through the influence of both the local and global, the past and present. In rap lyrics and their musical accompaniment, countries, cities, neighbourhoods, and even specific government housing developments inform the music, but the identities of these places and spaces are not fixed – for the performers or for the audience. They are more than the backdrop for what happens, inanimate structures or coordinates of latitude and longitude. Their dynamic nature, and their representation in music, serves to continually redefine “how we are constituted and who we are” (473).In MC Solaar’s Léve-toi et Rap from his 2001 album Cinquième as and his song Nouveau Western, from 1994’s Prose Combat, this is demonstrated in two very different ways. Léve-toi et Rap, a personal history told in the first person, clearly demonstrates both American hip-hop lineage and the transnational influences of Solaar’s upbringing. This song serves as an example of the adoption of American musical and lyrical techniques as means through which personally empowering, often place-based stories are told. In Nouveau Western, the narrative demonstrates the negative effects of globalization through this story about a geographically and temporally transported American cowboy. This track employs musical materials in a way that reflects the more critical lyrical commentary on the repercussions of American cultural and economic power. Through the manner of his storytelling, and through the stories themselves, MC Solaar explicitly demonstrates his own agency in representing, and thus constructing the meaning of, dynamic place and space as they are defined from these two perspectives.As a Paris-based French rapper, MC Solaar often makes his affiliation to this geographic focal point significant in his lyrics. This is especially clear in Léve-toi et Rap, in which Parisian banlieues (HLM government housing projects), nightclubs, and other places figure prominently in the text. From the lyrics, one learns a great deal about this rapper and his background: MC Solaar was born in Senegal, but his parents brought him to France when he was young (MC Solaar, “Léve-toi et Rap”; Petetin, 802, 805). He grew up struggling with the isolation and social problems of the banlieues and the discrimination he faced as an immigrant. He began rapping, established a musical career, and now encourages others to rap as a means of making something constructive out of a challenging situation. In the excerpt below, MC Solaar explains these origins and the move to the banlieues (Solaar, “Lève-toi et rap;” All translations by the author).Lève-toi et rap elaborates on the connection between the local and global in rap music, and between place, space, and time. The lyrics and music represent these properties in part by appropriating American rap’s stylistic practices. The introductory chorus incorporates sampled lyrics of the American artists Lords of the Underground, the Beastie Boys, Nas, and Redman (Various Contributors, “‘Lève-toi et rap’ Direct Sample of Vocals/Lyrics,” whosampled.com.). A bassline originally recorded by the funk group The Crusaders grounds the musical accompaniment that begins with the first verse (partially printed above), in which MC Solaar begins to depict his own place and space as he has experienced it temporally.In this chorus, the first sample is “I remember way back in the days on my block” from Lords of the Underground’s song Tic-Toc. This leads to “Oh My God” and “Ah, Ah, Ah,” both samples from Q-Tip’s contribution to the Beastie Boys’ song Get It Together. “I Excel,” which appears in Nas’s It Ain’t Hard to Tell comes next. RedmanMC Solaar[Crusaders sample begins] The rap samples all date from 1994, the year Solaar released his well-received album Prose Combat and most are strategically placed: the first sample originated in the last verse of Tic-Toc, the Q-Tip samples in the middle are from the middle of Get It Together, and the last sample, “I Excel,” is from the first line of It Ain’t Hard to Tell. As Lève-toi et rap continues, MC Solaar’s statement of the song title itself replaces the iteration “MC Solaar” of the first chorus. In a sense, “Lève-toi et rap” becomes the last sample of the chorus. Through these American references, Solaar demonstrates an affiliation with the place in which rap is commonly known to have originally coalesced. For French rappers consciously working to prove their connection to rap’s lineage, such demonstrations are useful (Faure and Garcia, 81-82). Achieved by sampling music and lyrics from 1974 and 1994 from sources that are not all that obvious to a casual listener, Solaar spatially connects his work to the roots of rap (Shusterman, 214). These particular samples also highlight a spatial relationship to particular styles of rap that represent place and space in particular ways. Nas and Lords of the Underground, for instance, have added to the discourse on street credibility and authenticity, while Q-tip has provided commentary on social and political issues. MC Solaar’s own story widens the parameters for illustrating these concepts, as he incorporates the personally significant places such as Senegal, Chad, and the Saint Denis banlieue to establish street credibility on a transnational scale; the lyrics also describe serious social and political issues, including the “skinheads” he encountered while living in Paris. Dynamic place is clear throughout all of this, as everything occurring in these places is meaningful in part because of the unavoidable relationship with the passing of time – Solaar’s birth, his upbringing, and his success occurred through his choices and social interactions in specific places.Looking more closely at the representation of place and time, Lève-toi et rap is less than straightforward. As discussed previously, some of the vocal samples are rearranged, demonstrating purposeful alteration of pre-recorded material; in contrast, the use of a repeated funk bassline sample during a clear narrative of Solaar’s life juxtaposes a linear story with a non-linear musical accompaniment. To this, MC Solaar made a contemporary textual contribution to later choruses, with the title of the song added as the chorus’s last line. Such manipulation in the context of this first-person narrative to express this movement supports the conclusion that, far from being a victim of political and economic forces, MC Solaar has used them to his advantage. After all, the title of the song itself, Lève-toi et rap, translates roughly to “get up and rap.”In addition to manipulating the materials of American rap and funk for this purpose, Solaar’s use of verlan, a type of slang used in the banlieues, brings another level of locality to Lève-toi et rap. The use of verlan brings the song’s association with French banlieue culture closer: by communicating in a dialect fluently understood by relatively few, rappers ensure that their message will be understood best by those who share the constellation of social and temporal relations of these housing developments (Milon, 75). Adding verlan to other slang and to unique grammatical rules, the rap of the banlieues is to some extent in its own language (Prévos, “Business” 902-903).Referring to MC Solaar’s 1994 album Prose Combat, André Prévos observed that this material “clearly illustrates the continuity of this tradition, all the while adding an identifiable element of social and personal protest as well as an identifiable amount of ‘signifying’ also inspired by African American hip-hip lyrics” (Prévos, “Postcolonial” 43). While it is clear at this point that this is also true for Lève-toi et rap from Cinquème as, Nouveau Western from Prose Combat demonstrates continuity in different way. To start, the samples used in this song create a more seamless texture. A sample from the accompaniment to Serge Gainsbourg’s Bonnie and Clyde from 1967 undergirds the song, providing a French pop reference to a story about an American character (Various Contributors, “Nouveau Western” whosampled.com). The bassline from Bonnie and Clyde is present throughout Nouveau Western, while the orchestral layer from the sample is heard during sections of the verses and choruses. Parts of the song also feature alto saxophone samples that provide continuity with the jazz-influenced character of many songs on this album.The contrasts with Lève-toi et rap continue with the lyrical content. Rather than describing his own process of acquiring knowledge and skill as he moved in time from place to place, in Nouveau Western MC Solaar tells the story of a cowboy named “Harry Zona” who was proud and independent living in Arizona, hunting for gold with his horse, but who becomes a victim in contemporary Paris. In the fabled west, the guns he carries and his method of transportation facilitate his mission: Il erre dans les plaines, fier, solitaire. Son cheval est son partenaire [He wanders the plains, proud, alone. His horse is his partner.]. After suddenly being transported to modern-day Paris, he orders a drink from an “Indian,” at a bistro and “scalps” the foam off, but this is surely a different kind of person and practice than Solaar describes Harry encountering in the States (MC Solaar, “Nouveau Western”).After leaving the bistro, Harry is arrested driving his stagecoach on the highway and shut away by the authorities in Fresnes prison for his aberrant behaviour. His pursuit of gold worked for him in the first context, but the quest for wealth advanced in his home country contributed to the conditions he now faces, and which MC Solaar critiques, later in the song. He raps, Les States sont comme une sorte de multinationale / Elle exporte le western et son monde féudal / Dicte le bien, le mal, Lucky Luke et les Dalton [The States are a kind of multinational”/ “They export the western and its feudal way/ Dictate the good the bad, Lucky Luke and the Daltons] (MC Solaar, “Nouveau Western”).Harry seems to thrive in the environment portrayed as the old west: as solitary hero, he serves as a symbol of the States’ independent spirit. In the nouveau far west [new far west] francophone comic book characters Lucky Luke and the Daltons sont camouflés en Paul Smith’s et Wesson [are camouflaged in Paul Smith’s and Wesson], and Harry is not equipped to cope with this confusing combination. He is lost as he negotiates le système moderne se noie l’individu [the modern system that drowns the individual]. To return to Bonnie and Clyde, these ill-fated and oft-fabled figures weren’t so triumphant either, and in Gainsbourg’s song, they are represented by 1960s French pop rather than by even a hint of local 1930s musical traditions. “Harry Zona” is not the only person whose story unfolds through the lens of another culture.While Solaar avoids heavy use of verlan or other Parisian slang in this song, he does use several American cultural references, some of which I have already mentioned. In addition, the word “western” refers to western movies, but it also serves as another term for the United States and its cultural exports. “Hollywood” is another term for the west, and in this context MC Solaar warns his listeners to question this fictional setting. Following his observation that John Wayne looks like Lucky Luke, “well groomed like an archduke,” he exclaims Hollywood nous berne, Hollywood berne! [Hollywood fooled us! Hollywood fools!]. This is followed by, on dit gare au gorille, mais gare à Gary Cooper [as they say watch out for the gorilla, watch out for Gary Cooper]. Slick characters like the ones Gary Cooper played have ultimately served as cultural capital that has generated economic capital for the “multinational” States that Solaar describes. As Harry moves “epochs and places,” he discovers that this sort of influence, now disguised in fashion-forward clothing, is more influential than his Smith and Wesson of the old west (MC Solaar, “Nouveau Western”).It is important to note that this narrative is described with the language of the cultural force that it critiques. As Geoffrey Baker writes, “MC Solaar delves into the masterpieces and linguistic arsenal of his colonizers in order to twist the very foundations of their linguistic oppression against them” (Baker, 241). These linguistic – and cultural – references facilitate this ironic critique of the “new Far West”: Harry suffers in the grip of a more sophisticated gold rush (MC Solaar, “Nouveau Western”).Lève-toi et rap transforms musical and verbal language as well, but the changes are more overt. Even though the musical samples are distinctly American, they are transformed, and non-American places of import to MC Solaar are described with heavy use of slang. This situates the song in American and French cultural territory while demonstrating Solaar’s manipulation of both. He is empowered by the specialized expression of place and space, and by the loud and proud references to a dynamic upbringing, in which struggle culminates in triumph.Empowerment through such manipulation is an attractive interpretation, but because this exercise includes the transformation of a colonizer’s language, it ultimately depends on understanding rap as linked to some extent to what Murray Forman and Tricia Rose describe as “Western cultural imperialism” (Rose, 19; Forman, 21). Both Rose and Forman point out that rap has benefitted from what Rose describes as “the disproportionate exposure of U.S. artists around the world,” (Rose, 19) even though this music has provided an avenue through which marginalized groups have articulated social and political concerns (Rose, 19; Forman 21). The “transnational circulation of contemporary culture industries” that Forman describes (21) has benefitted multinational corporations, but it has also provided new means of expression for those reached by this global circulation. Additionally, this process has engendered a sense of community around the world among those who identify with rap’s musical and lyrical practices and content; in many cases, rap’s connection to the African diaspora is a significant factor in the music’s appeal. This larger spatial connection occurs alongside more locally place-based connections. Lève-toi et rap clearly manifests this sense of simultaneously negotiating one’s role as a global citizen and as an individual firmly grounded in the place and space of local experience.Even though rap has been a music of resistance to hegemonic social and economic forces for people around the world, it is nonetheless important to recognize that the forces that have disseminated this music on a global scale have contributed to the unequal distribution of wealth and power. Working within this system is almost always unavoidable for rappers, many of whom criticize these conditions in their music, but depend on these transnational corporations for their success. Paul A. Silverstein writes that “hip-hop formations themselves, while enunciating an explicit critique of both state interventionism and the global market, have directly benefited from both and, to be sure, simultaneously desire their end and their continuation” (47-48). This is very clear in Nouveau Western, which Silverstein writes “portrayed neo-liberalism as a ‘new Far West’ where credit cards replace Remingtons.” (48) That this critique has reached a large audience in the francophone world and elsewhere highlights the irony of the situation: under the current system of popular musical production and circulation, such material often must reach its audience through complicity with the very system it denounces. This view on the mixture of the local and global presented in these songs illustrates this confusing situation, but from another perspective, the representation of social interaction on varying scales connects to the factors that have contributed to rap since its inception. Local places and geographically broad spatial connections have been articulated in constantly changing ways through musical and lyrical sampling, original lyrical references, and the uses that creators, listeners, and the industry enact vis-à-vis global rap culture. Whether revealed through clear references to American rap that facilitate a personal narrative or through a more complicated critique of American culture, MC Solaar’s songs Lève-toi et rap and Nouveau Western expose some accomplishments of a French rapper whose work reveals personal agency both outside and within the “multinational” United States.
