Pełny tekst źródłaENGLISH ABSTRACT: Fruit size and the integrity of the rind are key components that determine the value of a citrus fruit. The application of 2,4-dichlorophenoxy acetic acid (2,4-D) to reduce splitting, a physiological disorder which entails cracking of the rind as well as to increase fruit size was conducted on three different split-susceptible mandarin and two split-susceptible orange cultivars. Treatments were applied directly after the physiological fruit drop period, as well as in January and February at 10 mg·L-1, alone or in combination with calcium (Ca), potassium (K) or gibberellic acid (GA3). Application of 2,4-D directly after physiological fruit drop, either alone or in a tank-mix with K, consistently reduced the number of split mandarin fruit, with later applications in January and February generally being ineffective. Post physiological fruit drop application of 10 mg·L-1 2,4-D significantly increased growth rate (mm.day-1) of all the mandarin cultivars, resulting in increased fruit size. Differences in sensitivity of cultivars to 2,4-D were evident, with the January application reducing the splitting in ‘Midknight’ Valencia. However, all the 2,4-D treatments reduced the fruit growth rate of the orange cultivars. The 2,4-D treatments, in terms of splitting, increased rind thickness, -strength and -coarseness of ‘Marisol’ Clementine, throughout fruit development. In addition fruit diameter and –length increased to such an extent that the fruit shape was altered (reduced d/l-ratio), reducing the potential of the rind to crack and the fruit to split, however rind coarseness of treated fruit was also increased. There were no major negative side effects on internal and external fruit quality, except for a possible reduction in juice content (%). Therefore, 10 mg·L-1 2,4-D can be applied directly after physiological fruit drop on ‘Marisol’ Clementine and ‘Mor’ mandarin to reduce fruit splitting.

Pełny tekst źródłaThe world fruit market represents hundreds of billions of euros per year and improving the organoleptic and nutritional quality of fruit has been one of the main objectives in recent years. Primary metabolism is a target that can be used to try and meet these requirements. Indeed, it provides the bricks necessary for growth and development but also the compounds that contribute to taste such as sugars and organic acids. Therefore, it is necessary to understand how it operates during fruit development. For this purpose, primary metabolism has been studied in eight species of fleshy fruits that differ in terms of development duration, fruit size, botanical family, taste quality (sweetness, acidity, etc.), and are subject or not to a respiratory crisis at the initiation of ripening. Physiological and biochemical data have been collected throughout the fruit development from anthesis to physiological maturity. Fruit growth modelling allowed standardizing the stages of development and thus improved comparison between species. The composition of biomass was then characterized qualitatively and quantitatively by targeted and non-targeted analytical approaches highlighting similarities and differences in composition and changes during development. Generalized linear models combining composition and growth data were used to compare different phases of fruit development and a discriminant partial least square regression analysis (PLS-DA) was used to separate climacteric and non-climacteric fruits. In both cases, cell wall compounds, proteins and lipids were involved in group differentiation. Finally, a detailed study of the fruit metabolome and activome was performed in three species of Solanaceae. It revealed that within the same botanical family, regulation differs during development, particularly for sugar metabolism and glycolysis. This work revisits the climacteric character of fruits, positioning it long before the onset of the respiratory crisis, and, more generally, provides a better understanding of primary metabolism during fruit development

Pełny tekst źródłaENGLISH ABSTRACT: For many apple (Malus domestica Borkh.) and pear (Pyrus communis L.) cultivars, attractive colour is essential to their profitability on export markets. This study focuses on problems related to poor green colour of ‘Granny Smith’ apples and insufficient red colour of bi-coloured pear cultivars. ‘Granny Smith’ apples often suffer from poor green colour. Green colour of fruit from various orchards was already found to differ midway through fruit development, with these differences being carried through to harvest. In a trial where nitrogen (N) fertilisers were applied using different forms at different times, there was no improvement in green colour. In another trial, artificial shading was applied to fruit only during their early development. Fruit that were shaded during this time were less green at harvest than unshaded fruit. Additional N applications may only improve colour where a deficiency exists. However, green colour may be improved by increasing light distribution early during fruit development. Bi-coloured pears attain their maximum red colour midway through their development, and this desired red colour is mostly lost prior to harvest. Red colour can also increase transiently with the passing of cold fronts. Anthocyanins, responsible for this red colour, may have a photoprotective function which would explain this pigmentation pattern, as photosystems are particularly sensitive to light damage at low temperatures. As ‘Rosemarie’ fruit bent over from a vertical to hanging position during development, peel photoinhibition was reduced as anthocyanins were synthesised. ‘Forelle’ peel was found to be very sensitive to high light levels at low temperatures. Substantial anthocyanin development took place in ‘Cripps’ Pink’ apples when weather conditions were cold, but clear following a cold front. A photoprotective role seems to explain daily changes in anthocyanins in response to temperature, but not the seasonal progression of colour development. Dwarfing rootstocks are known to improve red colour of bi-coloured pears due to improved light distribution. ‘Forelle’ fruit from six rootstocks of varying vigour were harvested from exposed positions only, so as to establish the effect of rootstock on red colour development independent of the effect of rootstock on canopy light distribution. Fruit from trees on quince (Cydonia oblonga Mill.) rootstocks were found to have redder fruit than those from vigorous BP pear rootstocks. This may be due to higher chlorophyll and carotenoid concentrations present in the peel of fruit from BP rootstocks, whose leaf and peel N were also high. The use of quince rootstocks is recommended where red colour development of bi-coloured pears is a problem. An early season bi-coloured cultivar with good red colour is required. Breeding trials to find such a cultivar are resource intensive. To streamline the process, a method to preselect immature seedlings for their future fruit colour is required. Fruit colour from bearing seedlings was compared with colour of their immature leaves. Trees with red leaves were likely to produce fruit that were too red for the breeders’ requirements. Trees with green or blushed leaves were capable of producing blushed fruit. It would be feasible to cull red-leaved seedlings with minimal risk of losing potential bi-coloured cultivars.

Pełny tekst źródłaEthylene is synthesized and perceived by all plants, and it is one of the most important phytohormone controlling fruit ripening. Ethylene is perceived by endoplasmic reticulum (ER)- localized proteins, called Ethylene Receptors (ETRs), which regulate fruit development and ripening, however the mechanisms by which ETRs regulate fruit ripening are not fully explained. Firstly, to study if ETRs regulate the ripening of climacteric and non-climacteric fruits, we compared ETRs and related protein members of both classes of fruit and by re-analyzing RNAseq data, already published, we found that ETRs were peaking at the inception of ripening in both climacteric and non- climacteric fruits, but in these data, the ETRs showed an earlier ETR expression peak relative to sugar accumulation. In this review, we also compared the structure of the ethylene receptors and related proteins in both classes of fruit, establishing a basis for the annotation of genes related to ethylene perception. Finally, the results show that there was a higher number of ETR genes in climacteric fruits than in non-climacteric fruits. Secondly, in tomato which is a fleshy fruit ripening model, a seventh ETR has been reported recently, following the genome sequencing. Characterization of this SlETR7 was carried out. We showed that ethylene binds to the transmembrane part of SlETR7. Like other ETR expression patterns during fruit ripening, SlETR7 expression in pericarp also goes up when fruit ripens. The profiles of the seven ETR expression during fruit ripening can be divided in 2 groups: group 1, ETR3, ETR4, and ETR6 are expressed earlier at Breaker+2 days than group 2, SlETR1, SlETR2, SlETR5, and SlETR7 that are expressed at a later stage of ripening. We constructed Knock Out (KO) and OverExpressed (OE) tomato lines for SlETR7, and we observed some phenotype changes proving that SlETR7 is a functional ETR. While there was only a small phenotype change in KO plants and fruits: more ethylene production at Br and Br+2days compared to Wild Type (WT). The OE lines showed early flowering, shorter plants, and smaller fruit than WT. The analyzes of the 7 ETR expression in KO and OE lines, revealed that other ETR expression is upregulated in KO mutants, which may explain the absence of obvious phenotype. and this suggest that SlETR7 maybe not critical in fruit ripening. Thirdly, regarding the studies of the seven tomato ETRs, one major bottleneck is the absence of reliable method to quantify them at the protein level. A targeted proteomic method was developed, PRM for Parallel Reaction Monitoring, and allow the identification and relative quantification of the seven tomato ETRs. This development applied to the study of the WT and Never Ripe mutant tomatoes showed that there is an over-accumulation of SlETR3, affected by a gain-of-function mutation in NR, while the NR tomatoes undergo ripening, which may be a cause of further ripening inhibition, as NR fruit stay orange. Finally, ETR mRNAs and proteins were analyzed within the same samples, and this led us to propose that there is a positive correlation between ETR mRNAs and proteins, which was controversial in the previous literature

Plants may produce a diverse array of defensive phytochemical compounds in response to interactions with herbivores, pathogens, and the microorganisms that reside within plant tissues. These phytochemicals may simultaneously improve crop defenses and quality, representing a potential agricultural management strategy. However, plant chemical responses to different types and levels of biotic interactions are not well-understood, particularly in fruit tissues, and the feasibility of activating these defenses in fruits through the application of phytohormones that regulate defense pathways as a potential management strategy also requires further examination. Thus, apples were used to 1) examine the impact of distinct communities of biotic interactions among plants, insects, and microbes on fruit chemistry, focusing on phenolics, an important class of phytochemical compounds, and 2) examine the impact of the defense-activating phytohormones jasmonic acid (JA), salicylic acid (SA), and melatonin (M) on fruit phenolic chemistry and resistance against pests and pathogens. Ultimately, phenolic defenses were activated by fungal damage primarily in ripe pulp tissues, where there was also a positive relationship between fungal endophyte and phenolic diversity, supporting a broad hypothesis that chemical diversity may increase with biotic diversity. Additionally, two compounds were produced in response to fungal damage: chlorogenic acid and an unidentified benzoic acid. Phytohormone applications did not affect phenolic chemistry, but the application of the combined JA-SA analogues had some chemical or physical effect, as this treatment reduced emergence of the insect Rhagoletis pomonella. Overall, the phytochemical defenses activated by biotic interactions in fruits may occur primarily in certain tissue types, and may also vary due to environmental conditions, time of year, or plant species. These chapters examined the relationship between fruit chemistry and biotic interactions with the goal of improving understanding of plant-microbe-insect interactions and incorporating these interactions into more sustainable agricultural practices.

Pełny tekst źródłaENGLISH ABSTRACT: Sunburn on apple and pear fruit results in large losses of export volumes in South African orchards. Producers estimate that losses due to sunburn are often about 10- 20%. Knowledge of temperature thresholds for sunburn as well as technologies available to manage sunburn is important to increase producer earnings. The effect of evaporative cooling on fruit surface temperature and sunburn was evaluated on 'Cripps' Pink', 'Royal Gala' and 'Fuji' apples as well as 'Forelle' pears. Evaporative cooling was effective in reducing fruit surface temperature by 0.6-3.7°C. Apple fruit under evaporative cooling treatments appeared to have acclimatised and had significantly higher surface temperatures when dry (0.9-4.7°C) than non-cooled fruit. This acclimitasing effect could be due to a modification of the cuticular wax layer. Futher trials are needed to investigate the cause of this acclimatising effect. The acclimatising effect was not found on 'Forelle' pears. Sunburn was reduced significantly only on 'Royal Gala' apples. Spraying 'Braeburn', 'Granny Smith', and 'Fuji' apples with reflective kaolin-based particle film (Surround WP) significantly reduced sunburn on 'Braeburn', 'Granny Smith' and 'Fuji' apple cultivars. Surround WP applications reduced leaf stomatal conductance and photosynthetic rate. Four applications of Surround WP with a commercial blower sprayer increased total soluble solids in 'Braeburn' . Four applications with a blower and five applications with a tower sprayer resulted in increased fruit size, higher flesh firmness, and lower percentage starch breakdown in 'Granny Smith'. Surround WP treatments slightly improved fruit colour in 'Granny Smith' apples. Futher trials are needed to establish whether Surround WP can,be reliably and economically removed from apple fruit and to evaluate the storage quality of fruit cleansed in this way. The effect of fruit surface temperatures (24°C, 30°C, 33-35°C, 40-43°C and 48-49°C) for increasing periods of time (two, four, six or eight hours) on maximum quantum yield of fluorescence (Fv/Fm) was measured on exposed and shaded sides of detached 'Cripps' Pink' and 'Royal Gala' apples. Measurements were taken directly after removal from the chamber as well as 12 hours later to assess recovery. Permanent damage to Photo system II (PSII) occurred between 48-49°C irrespective of duration, with permanent damage possible at 42-43°C when exposure exceeded six hours. There were no meaningful differences in Fv/Fm between shaded and exposed sides of apple fruit after exposure to high temperature. Duration of exposure was important in lowering PSU efficiency in 'Cripps' Pink' but less so in 'Royal Gala'. 'Cripps' Pink' apples harvested later in the season appeared to be able to recover from heat stress better than apples harvested earlier in the season. However, we only studied the effects of high temperature on apple tissues. More research is needed to establish the reaction of apple tissues to high temperatures in the presence of sunlight.
Kirstein, Deon Louw. "Mechanical thinning of pome fruit". Thesis, Stellenbosch : Stellenbosch University, 2015. http://hdl.handle.net/10019.1/97920.
Pełny tekst źródłaENGLISH ABSTRACT: Thinning is an important practice in pome fruit production which aims to ensure an optimal yield of high quality, large sized fruit as well as an adequate return bloom. In South Africa, pome fruit thinning is generally done by means of chemicals, with follow-up hand thinning. When thinning is effective, set and thus the hand thinning requirement should be reduced. This is important as labor cost associated with hand thinning is high and continually rising. Chemical thinning is weather dependent and can be environmentally harmful, which has led to a shift towards environmentally acceptable methods of thinning such as mechanical thinning. From 2013 until 2015 the mechanical string thinners, viz. Darwin 300™, BAUM, and Bloom Bandit™, were evaluated. These machines are used to thin trees during full bloom and reduce the number of flowers before fruit set. The aim of the trials was to reduce fruit set and therefore hand thinning requirement, while increasing fruit size and quality, maintaining yield and return bloom. A range of tractor speeds and rotational rates were evaluated with the Darwin 300™ on ‘Forelle’ pears and ‘Cripps’ Pink’ apples, while the BAUM was evaluated only on ‘Cripps’ Pink’ apples. The hand-held Bloom Bandit™ was evaluated on ‘Forelle’, ‘Cripps’ Pink’, ‘Fuji’ and ‘Cripps’ Red’. The tractor-driven mechanical thinning devices gave erratic results. The most consistent results on ‘Forelle’ were obtained using the Darwin 300™ at 5.2 km·h-1 and 300 rpm, while the BAUM gave no consistent results. The unreliability of results were due to South African pome fruit orchards currently being unsuitable for tractor-driven mechanical thinning machines. The ‘Forelle’ orchard trained to a Palmette system was the most suited for thinning, which is reflected in the more positive results obtained, but further improvements are possible. The Bloom Bandit™ effectively thinned pear and apple trees and increased fruit size without a decrease in yield or return bloom. More time is spent on thinning with the device compared to tractor-driven machines and this should be taken into account when considering using the Bloom Bandit™. Thinning intensities of 25%, 50% and 75% of clusters or flowers was applied to mature ‘Forelle’ and ‘Cripps’ Red’ trees during full bloom. Variable effects were seen on fruit set, yield was reduced to acceptable levels, while fruit size was improved in ‘Forelle’ but not ‘Cripps’ Red’. Results showed that when thinning mechanically, the aim should be to remove between 25% and 50% of flowers clusters in ‘Forelle’ and 50% of flowers clusters in ‘Cripps’ Red’. These levels of thinning gave the best results in terms of the remaining hand thinning requirements and improved return bloom in ‘Forelle’. We, however, only evaluated full cluster thinning and not within cluster thinning, which might also occur during mechanical thinning.

Pełny tekst źródłaENGLISH ABSTRACT: Successful export of South African fresh fruit requires fruit of a high quality. Variable fruit quality within a consignment is detrimental to effective marketing of the product. The light microclimate under which the fruit develops is one of the factors that affect within-tree and between-tree variation in quality, maturity and yield. Light exposure effects on fruit quality at harvest and after commercial storage and ripening periods, as well as the physiological mechanisms of these responses to light exposure were studied. Increased exposure to light resulted in the development of typical sun leaf characteristics, with the associated increase in leaf nitrogen concentration and photosynthetic rates. Size and mass of 'Laetitia' plums and 'Cripps' Pink' apples increased with increasing exposure to light. Shade treatments were only started after the initial phase of cell / division was complete. Increased size of the fruit was likely due to the improved carbon balance of the exposed foliage and fruit from the end of cell division until harvest. The transpiration stream was higher in the more exposed foliage compared to the shaded parts of the canopy. This was supported by increased transpiration rates and decreased midday water potentials of exposed leaves. 'Songold' plums and 'Rosemarie' pears were also investigated in the first season, but results were not conclusive. Increased exposure to light was associated with advanced maturity of 'Laetitia' plums at harvest. Shaded fruit were able to attain a similar level of maturity as exposed fruit during storage and ripening periods. At harvest and after the storage and ripening periods, exposed fruit had a higher total soluble solid (TSS) content and therefore an improved eating quality. At harvest, blush colour of 'Laetitia' plums increased with increased exposure to irradiance. Blush colour continued to develop during storage and ripening, and after the ripening period it was evident that blush colour development was associated with a dosage effect i.e. exposure to a cumulative level of irradiance gives the fruit the potential to develop a certain amount of blush colour. Fruit exposed to more than 70% photosynthetic photon flux density (PPFD) were .able to develop to a similar level of blush colour, whereas, fruit exposed to less than 50% PPFD were not able to attain the same level of blush colour. Increased exposure to light did not result in advanced maturity of 'Cripps' Pink' apples at harvest, but it did lead to improved blush colour and increased TSS levels. Blush colour of 'Rosemarie' pears was also dependent on exposure to light from four weeks before harvest. Exposed 'Laetitia' plums had a greater whole fruit content of Mn and B, but concentration on a dry mass basis of P, K, and B decreased with increasing light. Exposed 'Cripps' Pink' apples had increased whole fruit content of P, K, Ca, Mg, Mn, Fe, Cu and B, but concentration on a dry mass basis of K and Na decreased with increasing light. Nutrient content is often associated with the incidence of internal disorders of fruit after storage and further investigation of this effect is necessary as internal disorders were virtually absent in this study. The termination of irrigation shortly before harvest in order to advance the maturity of all the fruit to a similar level, and the subsequent strip harvest of the fruit on a single harvest date, is a practice commonly used by South African plum producers to reduce cost and ostensibly to improve fruit quality. The effect of this practice on 'Songold' plum quality at harvest, after storage and after ripening was also studied. Drip-irrigated plums and plums subjected to soil drying had a better eating quality and were more marketable than micro-irrigated and non-droughted fruit. Following commercial storage and ripening periods these fruit were firmer, had a higher TSS content and were of a similar size and mass to micro-irrigated and non-droughted fruit. The extended harvesting period, in contrast to a strip harvest, allowed the fruit that were smaller and less mature at the beginning of the period to attain a greater size and advanced maturity toward the end of the harvesting period.

Pełny tekst źródłaENGLISH ABSTRACT: Maintaining constant high yields in „Abate Fetel‟ and „Forelle‟ orchards in South Africa is challenging. Improving productivity in these orchards could be achieved by increasing fruit set and fruit size. Fruit size is an important marketing and quality parameter and has a significant effect on the economic value of fruit. Various protocols to improve fruit set are used by South African producers but these are not well researched. We therefore evaluated different combinations of plant growth regulators including gibberellic acid (GA3), gibberellins 4+7 (GA4+7), GA4+7 combined with 6-benzyladenine (6-BA), aminoethoxyvinylglycine (AVG) and prohexadione-calcium (P-Ca) in combination with trunk girdling during flowering on „Forelle‟ and „Abate Fetel‟ to determine the best fruit set strategy. All applied growth regulators improved fruit set relative to an untreated control over two consecutive seasons, but GA3 and P-Ca reduced return bloom and AVG resulted in smaller fruit size relative to the other treatments. The application of synthetic cytokinins are believed to enhance fruit size by stimulating and extending the cell division period in fruit when applied at the correct stage of fruit growth. In addition, combination of P-Ca with GA4+7 was used successfully on Japanese pear (Pyrus pyrifolia Nakai) and „Bing‟ sweet cherry to improve fruit size. This combination of GA4+7 and P-Ca was evaluated and combined with 6-BA treatments on European pear (Pyrus communis L.) cultivars, Forelle and Abate Fetel, to see if a similar effect on fruit size could be achieved under South African growing conditions. On both „Forelle‟ and „Abate Fetel‟ the combination of GA4+7 and P-Ca increased fruit size, but was more pronounced in „Abate Fetel‟. Growth regulators N-phenyl-N‟ -1,2,3-thiadiazol-5-ylurea (TDZ), N (2-chloro-4-pyridyl)-N‟ -phenylurea (CPPU), 6-BA and 2,4-dichlorophenoxyacetic acid (2,4-D) successfully increased fruit size in pear cultivars Coscia and Spadona in Israel. These growth regulators were applied to „Forelle‟ and „Abate Fetel‟ to determine if a similar effect could be achieved. None of the synthetic cytokinins applied had a significant effect on increasing fruit size in these two cultivars over two consecutive seasons although 6-BA increased return bloom and 2,4-D application resulted in increased fruit set. The stage when the cell division period in „Forelle‟ and „Abate Fetel‟ ends was also determined as 34 and 45 days after full bloom respectively, which can be used in the future to better plan the timing of fruit size enhancement treatments. Based on results from various fruit set and fruit size improvement trials, it can be recommended to use GA4+7 or AVG to increase fruit set on „Forelle‟ and „Abate Fetel‟, depending on the fruit set history of the orchard. Results from fruit size improvement trials were variable, and emphasises the fact that a balance between yield and fruit size must be determined for an orchard to achieve good fruit size and maximum return.

Pełny tekst źródłaDelayed fruit harvest (DFH) or on-tree-fruit storage is a strategy being considered to extend the harvest season of late maturing ‘Reed’ avocado. However, avocado fruit growth beyond physiological maturity is accompanied by alterations in chemical and physical properties. Thus, the study aimed to investigate the effects of delayed harvest on tree productivity, mineral nutrition and post-harvest quality of late maturing ‘Reed’ avocado and further to evaluate the fruit quality response to varying ripening temperature regimes. As treatments, fruit were harvested early, mid and late in the season, with one month apart from each harvest during 2013 and 2014 seasons. Yield and fruit size were recorded at harvest. During each harvest time, two sets of fruit samples were collected to determine the fruit nutritional content while the second set was cold stored at 5.5°C for 28 days. After storage, fruit were ripened at 16, 21 or 25°C and evaluated for post-harvest disorders, pathological diseases and fruit physico-chemical properties. Yield dropped from 42 to 12 kg/tree from early to late harvest during 2013, while treatments showed no effect during 2014. Furthermore, treatments had no effect on fruit size during both seasons. Phosphorus content decreased by 40 and 23% from early to late harvest time during 2013 and 2014 seasons, respectively. Similar decreasing patterns were observed for K, Ca and Mg content. Zinc content remained constant during 2013; however, mid-season fruit maintained higher zinc levels during 2014 season when compared to early and late harvest time. Protein content was constant for early and mid-season fruit but significantly decreased during the late harvest in both seasons. Moisture content decreased from 74.0 to 65.0% from early to late harvest during both seasons. Starch, oil and ash content were high during late harvest time in both seasons; while crude fibre was high during the mid-harvest time when compared with early and late harvest time. Internal chilling injury increased with maturity and ripening temperature. In both seasons, higher temperatures induced higher anthracnose incidences during early and mid-harvest compared with late harvest time. Similarly, vascular browning was high on fruit ripened at 16°C during early and mid-harvest time but decreased on late harvest fruit during 2013. However, low ripening temperatures induced high incidences of vascular browning during 2014 season. Stem-end rot was high on fruit ripened at 16°C than 21 and 25°C during 2014 season. xvi Physico-chemical quality parameters showed similar behaviour in both seasons. Fruit lightness increased with ripening time regardless of ripening temperature during the early harvest, but remained constant during mid and late harvest. Hue angle (h0) and Chroma (C*) values were slightly reduced during ripening. Fruit ripened at 25°C had the highest respiration rate and reached a climacteric peak earlier (day 2) than fruit ripened at 21 and 16°C (day 4). Fruit firmness and weight loss were high and rapid at 25°C followed by 21 and 16°C regardless of the harvest time. Delayed harvest had no effect on yield and fruit size. Generally, nutritional content of avocado fruit increased with fruit maturity. Furthermore, fruit maturity played a major role in the response of fruit to ripening temperature. Higher temperature enhanced fruit ripening, but was conducive for development of post-harvest diseases. Keywords: Harvesting time, nutrition, yield, fruit size, ripening temperature, post-harvest quality

Pełny tekst źródłaClimate and soil variety of Brazil ensures a diversified fruit production during all seasons. Thus the country is gaining prominence in the orcharding field. Passion fruit is a fruit that stood out in recent years internally and out of the country due to the increased consumption of fresh fruit and products derived from the pulp. For the best use of this fruit, it is necessary to determine its main quality parameters. Since passion fruits are highly perishable products and their metabolism proceeds even after harvest, to establish methodologies that allow fast and efficient control becomes essential to qualify them in a more adequate way, either for export, processing or fresh consumption. Traditional methodologies employed to determine fruit quality parameters have been fundamental to prediction of potential yields in industrial processes and also to establish commercial value of the fruit, however, they are expensive, time consuming and destructive. Therefore, the aim of this study was to assess the efficiency of mid and near infrared spectroscopy coupled to multivariate calibration to determine quality parameters of the intact passion fruit, as well as fresh and lyophilized pulps. Quality parameters analyzed were simple sugars, SSC, AT, organic acids, vitamin C and carotenoids. Results showed low correlations between reference analyses values and predicted values to determine the quality parameters concentrations in intact fruit. Multivariate models developed to quality parameters of in nature pulp showed good prediction results to determine the concentration of glucose, fructose, sucrose, AT, and SSC using mid-Infrared. NIR region showed higher prediction difficulty due to the higher water absorption bands in this region. on the other hand, multivariate models developed to predict quality parameters of the lyophilized pulp presented excellent results especially in the MID region for glucose (R2v= 0,938), fructose (R2v = 0,907), sucrose (R2v = 0,921) and citric acid (R2v = 0,918). The results for malic acid and carotenoids contents were unsatisfactory in the two regions used in this study due to the low concentration of these compounds. From the results obtained in this study, it could be verified that: i) there is greater difficulty to correlate constituents at low concentrations, ii) light intensity through the fruit decreases significantly with skin depth, so thick skin has showed to be a barrier for using this technique and iii) water interference in identifying groups and specific bands was significant for passion fruit, due to its high moisture content. The results also revealed that the mid-Infrared region was most suitable for predicting the concentration of quality parameters, in comparison with NIR region. Based also on the results obtained, the use of fresh pulp with MID spectroscopy seemed to be the most suitable choice for quality parameters determination of the passion fruit, since good correlations could be obtained without pulp lyophilization step

Pełny tekst źródłaUntil recently, Northwestern Australia was thought to be relatively free of serious fruit fly pests. Although a noxious strain, present in Darwin since 1985, was widely believed to be an infestation of the Queensland fruit fly, Bactrocera tryoni, from the East coast, the fruit flies present outside this area were believed to be the benign endemic species, B. aquilonis. However, during the year 2000, infestations of fruit flies were discovered on major commercial crops in both Western Australia and the Northern Territory. It was not known whether these outbreaks were due to an invasion of the major pest species, Bactrocera tryoni, a change in the behaviour of B. aquilonis, or a hybridisation event between the two species. Finding the source of these outbreaks has been complicated by the fact that, since B. tryoni and B. aquilonis are virtually indistinguishable morphologically, it was not known which species are present in the region. Traditionally any tryoni complex fly caught in the Northwest was called B. aquilonis based solely on location. In order to get a good population profile of the region, an extensive trapping program was set up to include flies from urban areas, commercial crops and natural areas where the benign strain is thought to remain. Tests of genetic differentiation and clustering analyses revealed a high degree of homogeneity in the Northwest samples, suggesting that just one species is present in the region. The Northwest samples were genetically differentiated from the Queensland samples but only to a small degree (FST =0.0153). MtDNA sequencing results also showed a small degree of differentiation between these regions. A morphological study of wing shape indicated that there are some minor identifiable morphological differences between East coast and Northwest laboratory reared flies. This difference was greater than that seen between B. jarvisi populations across the same geographic range. The results suggest that the flies caught in the Northwest are a separate population of B. tryoni. Soon after pest flies were discovered in Darwin, a population became established in Alice Springs. This population had a low genetic diversity compared with Queensland and Darwin populations, and showed evidence of being heavily founded. In 2000, an outbreak was discovered in the nearby town of Ti Tree. Due to the geographic and genetic similarity of these populations, Alice Springs was determined to be the source of the Ti Tree outbreak. To investigate the founding of these populations, a program was developed to estimate the propagule size. Using a simulation method seven different statistics were tested for estimating the propagule size of an outbreak population. For outbreaks originating from populations with high genetic diversity, the number of alleles was a good estimator of propagule size. When, however, the genetic diversity of the source population was already reduced, allele frequency measures, particularly the likelihood of obtaining the outbreak population from the source population, gave more accurate estimates. Applying this information to the Alice Springs samples, it was estimated that just five flies were needed to found the major population in and around Alice Springs. For Ti Tree, the propagule size was estimated to be 27 flies (minimum 10). In 2000, a much larger outbreak occurred in the developing horticultural region of Kununurra in northern Western Australia. An important question for the management of the problem is whether there is an established fly population or the flies are reinvading each year. This population was found to have a large amount of gene flow from the Northern Territory. Within the Kununurra samples, one group of flies was genetically differentiated from all the other samples. This group came from a small geographic area on the periphery of Kununurra and appeared to be the result of an invasion into this area at the time when the population was building up following the dry season. A further threat to the Northwest horticultural regions comes from B. jarvisi. A recent increase in the host range of this species has lead to speculation that it may become a greater pest in Northwestern Australia. At the present time, protocols for the population monitoring and disinfestation of this species are not in place. Here it is shown that B. jarvisi eggs are more heat tolerant than B. tryoni eggs and that monitoring of B. jarvisi populations is possible using cue lure traps placed according to fruiting time and location of their favoured host, Planchonia careya.

Pełny tekst źródłaThe purpose of this research has been to determine the nature of strawberry fruit resistance to anthracnose fruit rot. Production in the United States and North Carolina is heavily dependent upon two cultivars, Chandler and Camarosa. Both cultivars are susceptible to anthracnose fruit rot (AFR), caused by the imperfect fungus . Results from this research will be used to further our understanding of the strawberry- system and to control AFR through breeding strategies or cultural practices. Components of resistance in strawberry to AFR were studied under field and controlled environment conditions; five strawberry genotypes were inoculated with conidia of under field conditions and seven genotypes were inoculated with either five inoculum concentrations or three isolates of in growth chambers. Strawberry genotypes responded differently to in the field and in a controlled environment. Components of resistance to AFR included rate-limiting resistance, reduced percent lesion, reduced probability of lesion formation due to fruit age and genotype, and plant canopy architecture. Rates of disease progress were different among genotypes, inoculum concentrations, isolates, and the genotype x isolate interaction. Susceptible genotypes and virulent isolates had faster rates of disease progress. Differences were observed among genotypes for yield, percent by weight, and number of berries with AFR. Plant canopy architecture influenced the hours of fruit wetness and the yield of berries with AFR; a loose open canopy had fewer hours. Young and old fruit were less susceptible to AFR than fruit of median age.

Pełny tekst źródłaAnalyzes of candidates genes expression profile by qRT-PCR shown that ARF19 has the most interesting profile for parthenocarpy for melon and cucumber. These analyze show that the pollination induces the expression of GA20oxydase(1) and GA20oxydase(5). In the absence of pollination, almost all GA2oxydases are expressed. The study of the transcriptome by RNA-seq identifies genes which have a role of negative or positive regulators in the fruit initiation of melon. The study of genes matrix of cell division shows that their expression is high in ovules and pericarp at anthesis and fertilized stage. The data show that fruit development is largely regulated by transcription factors. Systematic phenotyping genetic analysis identified FS1 the first candidate for facultative parthenocarpy in melon
